Selected AbstractsDedifferentiation of intrinsic response properties of motoneurons in organotypic cultures of the spinal cord of the adult turtleEUROPEAN JOURNAL OF NEUROSCIENCE, Issue 7 2000Jean-François Perrier Abstract Explant cultures from the spinal cord of adult turtles were established and used to study the sensitivity of the intrinsic response properties of motoneurons to the changes in connectivity and milieu imposed by isolation in culture. Transverse sections 700 ,m thick were explanted on cover slips and maintained in roller-tube cultures in medium containing serum and the growth factors brain-derived neurotrophin factor (BDNF), neurotrophin-3 (NT3), glial cell line-derived neurotrophic factor (GDNF) and ciliary neurotrophic factor (CNTF). The gross morphology of acute sections was maintained after 4 weeks in culture. Cell bodies of motoneurons remained stainable in fixed cultures with an antibody against choline acetyltransferase (ChAT) throughout the culture period. During culture, motoneurons maintained stable resting membrane potentials and were contacted by functional synapses. The ability to generate action potentials was also preserved as was delayed inward rectification and generation of calcium spikes in the presence of tetra-ethyl ammonium (TEA). In response to depolarization, however, motoneurons presented strong outward rectification, and only 41% of the cells recorded from maintained the ability to fire repetitively. By the second week in culture, a fraction of motoneurons displayed fast and slow transient outward rectification and low-threshold calcium spikes, features not seen in turtle motoneurons in acute slices. On the other hand, properties mediated by L-type Ca2+ channels disappeared during the first few days in culture. Our observations show that the phenotypical intrinsic response properties of mature spinal motoneurons are modified in explant cultures. The properties acquired resemble the properties in juvenile motoneurons in several species of terrestrial vertebrates. [source] Changes in vertebral structure during growth of reared rainbow trout, Oncorhynchus mykiss (Walbaum): a new approach using modelling of vertebral bone profilesJOURNAL OF FISH DISEASES, Issue 3 2009M-H Deschamps Abstract Severe bone resorption of the vertebral body in reared rainbow trout was thought to be a dysfunction in mineral balance induced by increased growth rate in unfavourable rearing conditions. To verify this assumption, we sampled market-sized trout (c. 250 g) from 20 fish farms with different rearing conditions. Growth rate was also studied by sampling trout reared in three different water temperatures from fry to market-size. Transverse sections of vertebrae were microradiographed, then digitized. Total bone area (Tt-B.Ar.) and bone profiles were obtained using BONE PROFILER 3.23 software and a mathematical model was developed to statistically compare bone profiles using 12 parameters in four vertebra regions. Tt-B.Ar. and bone profiles were found to vary with rearing conditions and growing temperatures, indicating obvious influences of these factors on bone remodelling. However, vertebral resorption was found to be a general phenomenon. In trout from 190 to 235 mm in length, vertebrae underwent important remodelling resulting in large resorption of the middle area, while the transition and peripheral areas showed an increase in bone deposition. Changes in vertebra architecture seem to be a good compromise between the need to mobilize stored minerals during growth while maintaining vertebral biomechanical properties. [source] Ultrastructural study of the precursor to fungiform papillae prior to the arrival of sensory nerves in the fetal ratJOURNAL OF MORPHOLOGY, Issue 3 2001Shin-ichi Iwasaki Abstract The structure of precursors to fungiform papillae without taste buds, prior to the arrival of sensory nerve fibers at the papillae, was examined in the fetal rat on embryonic day 13 (E13) and 16 (E16) by light and transmission electron microscopy in an attempt to clarify the mechanism of morphogenesis of these papillae. At E13, a row of rudiments of fungiform papillae was arranged along both sides of the median sulcus of the lingual dorsal surface, and each row consisted of about 10 rudiments. There was no apparent direct contact between papillae rudiments and sensory nerves at this time. Bilaterally towards the lateral side of the tongue, adjacent to these first rudiments of fungiform papillae, a series of cord-like invaginations of the dorsal epithelium of the tongue into the underlying connective tissue, representing additional papillary primordia parallel to the first row, was observed. The basal end of each invagination was enlarged as a round bulge, indented at its tip by a mound of fibroblasts protruding into the bulge. At E16 there was still no apparent direct contact between rudiments of fungiform papillae and sensory nerves. Each rudiment apically contained a spherical core of aggregating cells, which consisted of a dense assembly of large, oval cells unlike those in other areas of the lingual dorsal epithelium. The differentiation of these aggregated cells was unclear. The basal lamina was clearly recognizable between the epithelium of the rudiment of fungiform papillae and the underlying connective tissue. Spherical structures, which appeared to be sections of the cord-like invaginations of the lingual epithelium that appeared on E13, were observed within the connective tissue separated from the dorsal lingual epithelium. Transverse sections of such structures revealed four concentric layers of cells: a central core, an inner shell, an outer shell, and a layer of large cells. Bundles of fibers were arranged in the central core, and the diameters of bundles varied somewhat depending on the depth of the primordia within the connective tissue and their distance from the median sulcus. Ultrastructural features of cells in the outer shell differed significantly in rudiments close to the lingual epithelium as compared to those in deeper areas of connective tissue. Around the outer shell there was a large-cell layer consisting of one to three layers of radially elongated, oval cells that contained many variously sized, electron-dense, round granules. Large numbers of fibroblasts formed dense aggregates around each spherical rudiment, and were separated by the basal lamina from the large-cell epithelial layer. Progressing from deep-lying levels of the rudiments of the papillae to levels close to the lingual surface epithelium, the central core, inner shell, and outer shell gradually disappeared from the invaginated papillary cords. Gilmore MRCP Abstract The objective of this study was to assess neuronal pathology in the spinal cord in multiple sclerosis (MS), both within myelinated and demyelinated tissue. Autopsy material was obtained from 38 MS cases and 21 controls. Transverse sections were taken from three spinal cord levels and stained using Luxol Fast Blue/Cresyl Violet and myelin protein immunohistochemistry. Measurements of neuronal number and size were made for all neurons within the anterior horns of the gray matter. Neurons were classified as motoneurons or interneurons according to size criteria. In comparison with controls, both motoneuron and interneuron number were reduced in MS cases at the upper cervical (interneuron P = 0.0549; motoneuron P = 0.0073) and upper thoracic (interneuron P = 0.0507; motoneuron P = 0.0144), but not the lumbar level. Interneuron cross-sectional area was reduced in MS cases at all levels (upper cervical, P = 0.0000; upper thoracic, P = 0.0002; lumbar, P = 0.0337). Neuronal loss appears to be predominantly related to local gray matter plaques, whereas interneuron atrophy occurs in both myelinated and demyelinated areas. [source] Guidelines for species descriptions of diplomonad flagellates from fishJOURNAL OF FISH DISEASES, Issue 1 2002S L Poynton Diplomonad flagellates are common commensals of the digestive tract, and less common pathogenic parasites occurring in the digestive tract and systemically in numerous fish species. Many aspects of infections are poorly understood, including host-flagellate specificity, geographic ranges, and pathogenicity of different species. Much confusion is attributable to inadequate determination of genus and species. Although older literature reports Hexamita, Octomitus and Spironucleus from fish, recent studies confirm only Spironucleus. To address this problem, we describe ultrastructural features of trophozoites permitting reliable identification to genus and species, and techniques for their elucidation. Pioneering work by Brugerolle and colleagues established that genera can be distinguished by transmission electron microscopy (TEM). We now demonstrate that at the species level, surface ornamentations (especially at the posterior end of the body), and the pattern of bands of microtubules accompanying the flagellar pocket (in transverse section through the middle of the body), are of particular taxonomic value. Both scanning and TEM are essential for robust species descriptions and type material must be deposited in a recognized reference collection. Taxonomic studies are enhanced by in vitro culture, with tolerance and optimum for different conditions providing important supplementary information. [source] Gliding characteristics of tendon repair in canine flexor digitorum profundus tendonsJOURNAL OF ORTHOPAEDIC RESEARCH, Issue 4 2001Chunfeng Zhao The gliding resistance between the flexor digitorum profundus (FDP) tendon and the proximal pulley system was measured using the method of S. Uchiyama, J.H. Coert, L. Berglund, P.C. Amadio, K.N. An (J. Orthop. Res. 13 (1995) 83) in 108 adult dog digits in vitro. The FDP tendons were then lacerated to 80% of their transverse section. Each tendon was repaired with one of the following six suture techniques: Kessler, modified Kessler, Savage, Lee, Becker and simple running suture alone. Each repaired tendon was then tested again using the same method. The Student,Newman,Keuls test for multiple comparisons was performed for statistical analysis. The average gliding resistances of the Kessler, Savage, and Becker repairs were significantly greater than the resistances of the Lee, modified Kessler, and running suture alone repairs (P < 0.05). The Lee suture technique had a significantly greater resistance than the modified Kessler repair and the running suture (P < 0.05). The results of the peak gliding resistance followed the same trends, except that the modified Kessler repair was significantly higher than the running suture alone (P < 0.05). Suture techniques with a multi-strand core suture, with knots located outside the tendon surface, and with multiple-loops on the tendon surface may result in increased gliding resistance between the tendon and pulley system after tendon repair. © 2001 Orthopaedic Research Society. Roy Abstract The wear behaviour of metallic material is influenced by the friction force, which in turn, is governed by the hardness and oxidation kinetics of the mating surface. In view of this, present investigation is undertaken to find the influence of mating surface on the high temperature wear of 253 MA alloy. This alloy is developed for high temperature application. In this work 253 MA alloy is made to slide against two different types of counter face material, namely 100Cr6 steel and PM 1000 alloy, at five different temperatures. 100Cr6 steel gets soften with increase of temperature whereas PM 1000 alloy retains its strength even at high temperature. The friction coefficient and the thickness loss of 253 MA alloy is measured and compared against both variety of mating surfaces as function of temperatures. The morphology of the worn surfaces and the transverse section of the worn surfaces are examined under scanning electron microscope (SEM) to identify the material removal mechanisms. The results showed that the friction coefficient of test material against PM 1000 alloy is around 40% higher than the friction coefficient against 100Cr6 steel. The transverse section of the worn surface showed presence of a transfer layer, mechanically mixed layer and composite layer, which govern the wear behaviour particularly at elevated temperature. The chemical characteristics of these layers are dependent on the test temperature and the counter face material. [source] Toxic effect of environmental acid-stress on the sperm of a hill-stream fish Devario aequipinnatus: A scanning electron microscopic evaluationMICROSCOPY RESEARCH AND TECHNIQUE, Issue 2 2009Sudip Dey Abstract Environmental stress due to acidic pH of water was found to be one of the major factors leading to toxic effects on the sperm of a hill-stream fish Devario aequipinnatus of Meghalaya, India. The Scanning Electron Microscopy of the transverse section of testes of the fish collected from its natural habitat with acidic pH (5.6,6.0) showed that the sperms were clumped together and their tails were either absent or were of extremely small length. The acrosome and midpiece were also not well differentiated. When the fingerlings from the natural habitat were reared to maturity in aquarium with water from natural habitat after changing the pH to alkaline range (8.0,8.2), the clumping of the sperm was not observed. The sperm tail was found to be well-developed along with well-differentiated acrosome and midpiece. Since the only change in the water quality parameters of the experimental aquarium as compared to those of the natural habitat was the pH, it is evident that the abnormal features of the sperm observed in fish from natural habitat is mainly because of environmental acid stress. Don, a popularly used ethnodrug from Qinghai-Tibetan plateau, was studied to reveal the indispensable morphoanatomic details. The fixed, sectioned, and stained plant materials as well as the epidermis, powder, and maceration materials were studied using light microscope according to the usual microscopic techniques. The results of the microscopic features were systematic described and illustrated. In the root, an endodermal cell was divided into 8-16-22 and 38-50-62 daughter cells in transverse section and in face view, respectively, and 9-11-13 phloem strands were present in primary structure; in the stem, stone cells were observed in the cortex, pericycle, and external phloem while 17-19-21 internal phloem strands were present in an incontinuous ring; in the pedicel, 8-10-12 internal phloem strands were observed to form an incontinuous ring; anisocytic and anomocytic stomata were present in leaf and sepal epidermis; pollen grain was with three germinal apertures and furrows; a few tracheids, a large number of spiral vessels, and various fibers were observed. Also, semiquantitative and quantitative micrographic parameter tables were simultaneously presented. Further, the key authentication parameters were concluded. [source] First record of Palisada maris-rubri (Ceramiales, Rhodophyta) from the Mediterranean Sea along with three proposed transfers to the genus PalisadaPHYCOLOGICAL RESEARCH, Issue 1 2010Donatella Serio SUMMARY The first occurrence of Palisada maris-rubri (K.W. Nam et Saito) K.W. Nam (Ceramiales, Rhodomelaceae) from the Mediterranean Sea, is reported. To date the species was known only from tetrasporic specimens from the type locality (Ras Muhammed, Sinai, Egypt, Red Sea). Mediterranean thalli share nearly all vegetative and reproductive features with Red Sea specimens showing more robust thalli with axes to 3 mm broad and ultimate branchlets to 1000 µm broad, absence of intercellular spaces between medullary cells and epidermal cells in transverse section with a palisade arrangement. Male and cystocarpic thalli are recorded for the first time. Moreover, the analysis of characters of three species of Chondrophycus previously known from the Mediterranean Sea (C. patentirameus (Montagne) K.W. Nam, C. tenerrimus (Cremades) G. Furnari et al. and C. thuyoides (Kützing) G. Furnari) led us to conclude that they belong to the genus Palisada. [source] Characterization of the Swimming Muscle of the Anchovy Engraulis anchoita (Hubbs & Martini 1935)ANATOMIA, HISTOLOGIA, EMBRYOLOGIA, Issue 4 2000C. V. Devincenti The lateral muscle characteristics of individual female anchovies (Engraulis anchoita) were studied by histochemical and ultrastructural techniques. Red, pink and white fibres were discerned on the basis of the activity of the metabolic enzymes succinic dehydrogenase, phosphorylase and myofibrilar ATPase, and the usage of Sudan and periodic-acid Schiff staining techniques. Red fibres were located at the surface, white fibres deep inside and pink fibres in between. The red fibres appeared flattened or ribbon-shaped in transverse section and they were located in rows separated by myosepta of connective tissue. The red and pink fibres were multi-innervated whereas the white fibres were terminally innervated. The relative proportion of the red muscle increased toward the caudal region, the white muscle diminished toward that region, whereas the proportion of pink muscle did not vary according to region, being scarce throughout. The distribution of capillaries was also investigated, and as a result, the vascular supply proved to be scarce in both white and pink muscle. [source] Development of the pons in human fetusesCONGENITAL ANOMALIES, Issue 2 2007Toshihisa Hatta ABSTRACT Morphometric and histological studies of the pons were performed by light microscopy in 28 cases of externally normal human fetuses ranging from 90 to 246 mm in crown-rump length (CRL) and from 13 to 28 weeks of gestation. The brainstems of fetuses were embedded in celloidin or paraffin, and transverse sections were prepared. The pons was divided into two regions at the most ventral margin of the medial lemniscus at the level of the motor trigeminal nucleus. The relationships between the total dorsoventral length, ventral length, and dorsal length of the pons versus CRL and gestational ages were calculated, and empiric formulas were fitted. It was found that the ventral portion increased in size more rapidly than the dorsal portion. The proportion of the ventral portion in the total dorsoventral length was constitutively higher than that of the dorsal portion in the present range of CRL. In the pontine nuclei, from 235 mm in the CRL, some large cells with rich cytoplasm, pale nuclei, and a distinct nucleolus appeared on the dorsal side of the pyramidal tract. According to Weigert stained preparations, the first myelinated fibers in each motor root of the trigeminal, abducent, and facial nerves were recognized at 130,140 mm in CRL and the medial lemniscus at 230,235 mm. [source] Ultrastructure of spermatozoa of scolecophidian snakes (Lepidosauria, Squamata)ACTA ZOOLOGICA, Issue 3 2007L. Tavares-Bastos Abstract We provide a detailed description of the sperm ultrastructure of three species of scolecophidian snakes, Leptotyphlops koppesi (Leptotyphlopidae), Typhlops reticulatus (Typhlopidae) and Liotyphlops beui (Anomalepididae), and make comparisons with the spermatozoa of Ramphotyphlops waitii (Typhlopidae) (Harding et al. 1995). All the species studied bear synapomorphies of Squamata and Serpentes. Among scolecophidian snakes, we identified eight polymorphic characters. Previous analyses of molecular and somatic morphological data provide equivocal solutions to the relationships among Anomalepididae, Leptotyphlopidae and Typhlopidae. A close relationship between Anomalepididae and Typhlopidae is corroborated by two characters of sperm ultrastructure, presence of an electron-dense structure inside the proximal centriole and rounded mitochondria in transverse sections of T. reticulatus, Li. beui and R. waitii, whereas the absence of the ridge on the acrosome surface of Le. koppesi and T. reticulatus support a closer relationship between Typhlopidae and Leptotyphlopidae. The differences observed in sperm ultrastructure within Typhlopidae suggest the existence of snake intrafamilial polymorphism. The sperm characters of blindsnakes contain significant phylogenetic information and may provide important data for snake phylogenetic reconstructions. [source] Capillary supply and gene expression of angiogenesis-related factors in murine skeletal muscle following denervationEXPERIMENTAL PHYSIOLOGY, Issue 3 2005A. Wagatsuma Capillary supply of skeletal muscle decreases during denervation. To gain insight into the regulation of this process, we investigated capillary supply and gene expression of angiogenesis-related factors in mouse gastrocnemius muscle following denervation for 4 months. Frozen transverse sections were stained for alkaline phosphatase to detect endogenous enzyme in the capillary endothelium. The mRNA for angiogenesis-related factors, including hypoxia inducible factor-1, (HIF-1,), vascular endothelial growth factor (VEGF), kinase insert domain-containing receptor/fetal liver kinase-1 (KDR/Flk-1), fms-like tyrosine kinase (Flt-1), angiopoietin-1 and tyrosine kinase with Ig and epidermal growth factor(EGF) homology domain 2 (Tie-2), was analysed using a semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR). The fibre cross-sectional area after denervation was about 20% of the control value, and the capillary to fibre ratio was significantly lower in denervated than in control muscles. The number of capillaries around each fibre also decreased to about 40% of the control value. These observations suggest that muscle capillarity decreases in response to chronic denervation. RT-PCR analysis showed that the expression of VEGF mRNA was lower in denervated than in control muscles, while the expression of HIF-1, mRNA remained unchanged. The expression levels of the KDR/Flk-1 and Flt-1 genes were decreased in the denervated muscle. The expression levels of angiopoietin-1 but not Tie-2 genes were decreased in the denervated muscle. These findings indicate that reduction in the expression of mRNAs in the VEGF/KDR/Flk-1 and Flt-1 as well as angiopoietin-1/Tie-2 signal pathways might be one of the reasons for the capillary regression during chronic denervation. [source] Fibre type composition of the human psoas major muscle with regard to the level of its originJOURNAL OF ANATOMY, Issue 6 2009Juraj Arbanas Abstract The aim of our study was to explore the fibre type composition of the human psoas major muscle at different levels of its origin, from the first lumbar to the fourth lumbar vertebra, and to compare the muscle fibre size and distribution of different fibre types between levels with respect to its complex postural and dynamic function. Muscle samples were collected from 15 young males (younger than 35 years). Serial transverse sections (5 ,m) of the samples were cut by cryomicrotome. Type I, IIA and IIX muscle fibres were typed using myosin heavy chain identification. The serial sections were analysed using a light microscope with a magnitude of 100×. The differences between measurements were evaluated using a repeated-measures anova and Scheffé test for post-hoc analysis. Our study showed that the human psoas major muscle was composed of type I, IIA and IIX muscle fibres. It had a predominance of type IIA muscle fibres, whereas type I muscle fibres had the largest cross-sectional area. Type IIX muscle fibres were present as a far smaller percentage and had the smallest cross-sectional area. Moreover, the fibre type composition of the psoas major muscle was different between levels of its origin starting from the first lumbar to the fourth lumbar vertebra. We conclude that the fibre type composition of the psoas major muscle indicated its dynamic and postural functions, which supports the fact that it is the main flexor of the hip joint (dynamic function) and stabilizer of the lumbar spine, sacroiliac and hip joints (postural function). The cranial part of the psoas major muscle has a primarily postural role, whereas the caudal part of the muscle has a dynamic role. [source] The histological structure of the malleolar groove of the fibula in man: its direct bearing on the displacement of peroneal tendons and their surgical repairJOURNAL OF ANATOMY, Issue 2 2003T. Kumai Abstract The peroneal (fibularis) tendons are held in place within the malleolar groove by the superior peroneal retinaculum. If this is torn, the tendons can subluxate or dislocate. Understanding the anatomy of the region is important for treating these injuries when it becomes necessary to reconstruct the malleolar groove surgically. Serial transverse sections of the groove were cut from 10 dissecting room cadavers after routine histology processing. The structure of the malleolar groove differed significantly in its proximal and distal parts. Distally, the bone is convex and the shape of the groove is determined by a thick periosteal cushion of fibrocartilage that covers the bone surface. Proximally, the groove shape is determined by the bone itself, and the periosteum is thin and fibrous. The restriction of a periosteal fibrocartilage to the distal end suggests that it serves to adapt the shape of the malleolar groove to that of the tendons within it and thus promotes stress dissipation. Paradoxically, however, it increases the risk of damage to subluxated tendons, because these can be sliced longitudinally by a sharp ridge created from periosteal fibrocartilage when the retinaculum is torn. Our results suggest that if bone-block surgical procedures are used to reconstruct the malleolar groove, they are best restricted to its proximal part. [source] Patterning of the axial musculature in the developing chick embryo (Gallus)JOURNAL OF ANATOMY, Issue 5 2002P. J. Adds While the development and patterning events of the skeletal, myogenic and connective tissues of the developing limb buds of the chick have been relatively well studied, there is little known about the formation of the epaxial muscles and tendons. The epaxial muscles form the postvertebral muscle groups and develop from the myotome of the somite. The myotome develops from myogenic precursors migrating from the dorsomedial lip of the dermomyotome. These myogenic cells differentiate in a cranial to caudal sequence with the muscle fibres also orientated in a cranial to caudal direction. Here we use immunohistological staining both in whole mount and in transverse sections of chick embryos from various stages of development. Antibodies to the myosin heavy chain or tenascin were used to visualise the development of the epaxial muscles and tendons. The first myotomal muscle fibres are differentiated by Hamburger and Hamilton (H & H) stage 10 in the cranial somites, and differentiation proceeds caudally until at least H & H stage 22,23. Further development of the epaxial muscles does not take place until H & H stage 26,27 with the splitting of the myotome into the individual muscles. We demonstrate how the myotome splits into the individual muscles and how some muscle fibres become reorientated into a more oblique orientation. This delayed development and reorientation of muscle fibres is unique to the epaxial muscles. Grandcourt Summary Life history and demographic parameters for Lethrinus borbonicus, Lethrinus microdon, Pomacanthus maculosus and Scolopsis taeniatus in the southern Arabian Gulf were estimated using a combination of size frequency, biological and size-at-age data. Defined structural increments consisting of alternating translucent and opaque bands in transverse sections of sagittal otoliths were validated as annuli. The maximum age estimates ranged from 5 years for Scolopsis taeniatus to 36 years for Pomacanthus maculosus. The size-at-age relationships were highly asymptotic in form with the majority of growth being achieved early in life. There were significant differences in the growth characteristics between sexes for Pomacanthus maculosus, with males approaching a larger asymptotic size at a faster rate than females. With the exception of Scolopsis taeniatus, the mean age at which fish became vulnerable to capture was lower than the mean age at first sexual maturity. The stocks of L. microdon, P. maculosus and S. taeniatus were exploited within sustainable limits, conversely, L. borbonicus was found to be overexploited and recruitment overfishing may have occurred as the relative spawner biomass per recruit was below 30% of the unexploited state. Twotypes of cast Co-Cr plates were prepared, and transverse sections were made at the center ofthe plate. The cut surfaces were butted against one another, and the joints welded with alaser-welding machine at several levels of output energy with the use of two methods. Thefracture force required to break specimens was determined by means of tensile testing. For the0.5-mm-thick specimens, the force required to break the 0.5-mm laser-welded specimens atcurrents of 270 and 300 A was not statistically different (p > 0.05) from the results for thenonwelded control specimens. The force required to break the 1.0-mm specimens double-weldedat a current of 270 A was the highest value among the 1.0-mm laser-welded specimens. Ehrlich Abstract The presence of estrogen receptor , (ER,) in osteocytes was identified immunocytochemically in transverse sections from 560 to 860 ,m distal to the midshaft of normal neonatal and adult male and female rat ulnas (n = 3 of each) and from adult male rat ulnas that had been exposed to 10 days of in vivo daily 10-minute periods of cyclic loading producing peak strains of either ,3000 (n = 3) or ,4000 microstrain (n = 5). Each animal ambulated normally between loading periods, and its contralateral ulna was used as a control. In animals in which limbs were subject to normal locomotor loading alone, 14 ±1.2% SEM of all osteocytes in each bone section were ER, positive. There was no influence of either gender (p = 0.725) or age (p = 0.577) and no interaction between them (p = 0.658). In bones in which normal locomotion was supplemented by short periods of artificial loading, fewer osteocytes expressed ER, (7.5 ± 0.91% SEM) than in contralateral control limbs, which received locomotor loading alone (14 ± 1.68% SEM; p = 0.01; median difference, 6.43; 95% CI, 2.60, 10.25). The distribution of osteocytes expressing ER, was uniform across all sections and thus did not reflect local peak strain magnitude. This suggests that osteocytes respond to strain as a population, rather than as individual strain-responsive cells. These data are consistent with the hypothesis that ER, is involved in bone cells' responses to mechanical strain. High strains appear to decrease ER, expression. In osteoporotic bone, the high strains assumed to accompany postmenopausal bone loss may reduce ER, levels and therefore impair the capacity for appropriate adaptive remodeling. [source] Contribution of Nitric Oxide Synthase to Improved Early Graft Patency in Human Saphenous Vein Graft Harvested by a Novel ,No-Touch' TechniqueJOURNAL OF CARDIAC SURGERY, Issue 6 2002JCS Tsui Aim: Saphenous vein (SV) is the most commonly used conduit in bypass procedures but has a one-year occlusion rate of 15-30%. A new ,no-touch' technique where the SV is harvested with a cushion of surrounding tissue with no distension has led to improved early patency rates of 5% at 18-months. Nitric oxide (NO), synthesised by nitric oxide synthase (NOS) has properties beneficial to graft patency. Our aim was to study the distribution of NOS in SV harvested by this technique and the effect of distension and removal of perivascular tissue on NOS content of SV. Methods: Following ethical committee approval and patients' informed consent, SVs were harvested from ten patients undergoing coronary artery bypass grafting. A segment of vein was harvested by the conventional technique (surrounding tissue stripped and vein distended with saline); another part was stripped but not distended (,control') and the remaining parts harvested by the ,no-touch' technique. Samples of each segment were taken and transverse sections prepared for NOS identification using 3[H]L-NG nitroarginine (NO Arg) autoradiography and NADPH-diaphorase histochemistry. NOS isoforms were studied using standard immunohistochemistry. Endothelial cells and nerves were also identified using immunohistochemistry with CD31 and NF200 respecitvely, to confirm sources of NOS. Morphometric analysis of NADPH-diaphorase staining was carried out to study tissue NOS content. Results: NO Arg binding representing NOS was preserved on the lumen of ,no-touch' vessels whilst that on conventional and control vessels was reduced. NOS was also localised to the medial smooth muscle cells of all vein segments and to the intact adventitia of ,no-touch' segments. This was confirmed by NADPH-diaphorase staining, which revealed a mean reduction of NOS by 19.5% (p < 0.05, ANOVA) in control segments due to stripping of surrounding tissue alone and a reduction of 35.5% (p < 0.01, AVNOVA) in conventional segments due to stripping and distension, compared to ,no-touch' segments. Adventitial NOS sources in ,no-touch' vessels corresponded to vasa vasorum and paravascular nerves. All three NOS isoforms contributed to the preserved NOS in ,no-touch' vessels. Conclusions: Apart from preserved lumenal NOS, NOS sources are also located in the media and adventitia of SV grafts. These are reduced by both adventitial damage and vein distension during conventional vein harvesting. The ,no-touch' technique avoids these procedures, preserving NOS sources. This may result in improved NO availability in SV harvested by this technique, contributing to the improved patency rates reported. Sperling Background: The evaluation of patients with cicatricial alopecia is particularly challenging, and dermatopathologists receive little training in the interpretation of scalp biopsy specimens. Accurate interpretation of specimens from patients with hair disease requires both qualitative (morphology of follicles, inflammation, fibrosis, etc.) and quantitative (size, number, follicular phase) information. Much of this data can only be obtained from transverse sections. In most cases, good clinical/pathologic correlation is required, and so clinicians should be expected to provide demographic information as well as a brief description of the pattern of hair loss and a clinical differential diagnosis. Results: The criteria used to classify the various forms of cicatricial alopecia are relatively imprecise, and so classification is controversial and in a state of evolution. There are five fairly distinctive forms of cicatricial alopecia: 1) chronic, cutaneous lupus erythematosus (discoid LE); 2) lichen planopilaris; 3) dissecting cellulitis (perifolliculitis abscedens et suffodiens); 4) acne keloidalis; and 5) central, centrifugal scarring alopecia (follicular degeneration syndrome, folliculitis decalvans, pseudopelade). Not all patients with cicatricial alopecia can be confidently assigned to one of these five entities, and "cicatricial alopecia, unclassified" would be an appropriate label for such cases. Conclusion: The histologic features of five forms of cicatricial alopecia are reviewed. Dermatopathologists can utilize a "checklist" to catalog the diagnostic features of scalp biopsy specimens. In many, but not all, cases the information thus acquired will "match" the clinical and histologic characteristics of a form of cicatricial alopecia. However, because of histologic and clinical overlap between the forms of cicatricial alopecia, a definitive diagnosis cannot always be rendered. [source] Mechanical characteristics of the bone,graft,cement interface after impaction allograftingJOURNAL OF ORTHOPAEDIC RESEARCH, Issue 1 2005Hanspeter Frei Impaction allografting is an attractive procedure for the treatment of failed total hip replacements. The graft,cement,host bone interface after impaction allografting has not been characterized, although it is a potential site of subsidence for this type of revision total hip reconstruction. In six human cadaveric femurs, the cancellous bone was removed proximally and local diaphyseal lytic defects were simulated. After the impaction grafting procedure, the specimens were sectioned in 6 mm transverse sections and pushout tests were performed. From the adjacent sections the percentage cement contact of the PMMA cement with the endosteal bone surface was determined. The host bone interface mechanical properties varied significantly along the femur largely due to different interface morphologies. The apparent host bone interface shear strength was highest around the lesser trochanter and lowest around the tip of the stem. A significant positive correlation was found between the percentage cement contact and the apparent host bone interface shear strength (r2 = 0.52). The sections failed in 69% of the cases through a pure host bone interface failure without cement or allograft failure, 19% failed with local cement failure, and 12% with a local allograft failure. The apparent host bone interface strength was on average 89% lower than values reported for primary total hip replacements and were similar to cemented revisions proximally and lower distally. The digestion and assimilation of nematodes by larval fish were investigated with the aid of a histological and stable isotope approach. Larvae of whitefish Coregonus lavaretus were reared for 8 d with nematodes and compared with an unfed control. Nematodes were readily ingested by 3-d-old larvae. Different stages of nematode digestion could be observed in transverse sections of fish larvae sampled on Day 6 at regular intervals after feeding. Nematodes were produced on corn medium. In this way nematodes with a stable carbon isotope signature clearly different from the isotopic pattern of the fish larvae could be obtained. Stable carbon isotope signatures for lipids and lipid-free matter of fish larvae sampled on Days 2 and 8 after first feeding were clearly influenced by the stable isotopic pattern of the nematodes. The high acceptance of the nematodes by Coregonus lavaretus larvae and the early onset of digestion and nutrient retention positively confirm the potential of PanagreUus redivivus as a live food for first feeding fish larvae. The combination of immunohistochemistry and Western blotting is often used in these cases, but is not always possible if the sample is scarce. We therefore sought to develop a method to quantify relative levels of sarcolemma-associated proteins using digitally captured images of immunolabelled sections of skeletal muscle. Methods: To validate our relative quantification method, we labelled dystrophin and other sarcolemmal proteins in transverse sections of muscle biopsies taken from Duchenne muscular dystrophy and Becker muscular dystrophy patients, a manifesting carrier of Duchenne muscular dystrophy and normal controls. Results: Using this method to quantify relative sarcolemmal protein abundance, we were able to accurately distinguish between the different patients on the basis of the relative amount of dystrophin present. Conclusions: This comparative method adds value to techniques that are already part of the diagnostic process and can be used with minimal variation of the standardized protocols, without using extra amounts of valuable biopsy samples. Comparative quantification of sarcolemmal proteins on immunostained muscle sections will be of use to establish both the abundance and localization of the protein. Moreover, it can be applied to assess the efficacy of experimental therapies where only partial restoration or upregulation of the protein may occur. Craven The 5-hydroxytryptamine (5-HT, serotonin) system has been implicated in the pathophysiology and treatment of schizophrenia. In this study, we addressed the hypothesis that a deficit of 5-HT neurones, either inherited or acquired, is central to the developmental pathology of the disorder. We examined putative 5-HT neurones of the dorsal raphe nucleus (DRN) in post mortem, formalin-fixed tissue from 15 schizophrenic patients and 20 control subjects matched for age and gender. No significant difference was detected between these groups in the number or size (cross-sectional area or diameter) of tryptophan-hydroxylase-immunoreactive cell profiles viewed in transverse sections collected from the level of the trochlear decussation to the emergence of the trigeminal nerve. Profile number was not affected by age, gender, side of the brainstem (left or right) or post mortem interval; however, time in formalin correlated negatively with the number of neurones counted. Moreover, a significant negative correlation was detected between time in formalin and the levels of immunoreaction product (optical density), which in turn correlated positively with our profile counts. A positive correlation was found between the age of subjects and our estimates of cell size. Our results do not support the proposal that an abnormality in the number and/or size of DRN 5-HT neurones is central to the aetiopathology of schizophrenia. [source] Assessment of fetal lung development by quantitative ultrasonic tissue characterization: a methodical studyPRENATAL DIAGNOSIS, Issue 9 2004Ismail Tekesin Abstract Objectives This study was performed to evaluate the quantitative ultrasonic tissue characterization of the normal fetal lung development by using acoustic raw data captured after preprocessing. Methods One hundred and sixty-two patients with completed gestational ages between 22 and 37 weeks were enrolled in this study. Longitudinal and transverse sections of the fetal thorax and upper abdomen were imaged. A region of interest of constant size was defined and the tissue-specific gray scale was determined by using an interactive software. Results A total of 162 patients met the inclusion criteria. The echogenicity of the fetal lung showed a particular changing pattern during pregnancy: the mean gray value of the fetal lung (MGV) is almost the same as the MGV of the fetal liver at 22 and 23 weeks, decreases between 22 and 31 weeks and increases between 31 and 37 weeks. The MGV of the fetal liver decreases significantly from 24 weeks to 31 weeks and increases significantly again toward 37 weeks. We stated that the MGV of the lung is smaller than the MGV of the liver during 31 weeks of gestation and the relation reverses in late gestation. At term, the MGV of the liver is greater than the MGV of the lung. The lung-to-liver ratio is <1 between 24 and 29 weeks and >1 between 30 and 35 weeks. Conclusion The echogenicity of the fetal lung showed a particular changing pattern during pregnancy, which corresponds to morphologic changes of the fetal lung development. [source] A role for HKT1 in sodium uptake by wheat rootsTHE PLANT JOURNAL, Issue 2 2002Sophie Laurie Summary The high affinity potassium transporter, HKT1 from wheat was introduced into Florida wheat in sense and antisense orientation under control of a ubiquitin promoter. Ten transgenic lines expressing the transgene were identified and two of these showed strong down-regulation of the native HKT1 transcript. One line (271) was expressing the antisense construct and the other (223) was expressing a truncated sense construct. The two lines were examined further for phenotype relating to cation transport. Membrane depolarisations were measured in low (0.1 mm) K+ and high (100 mm) NaCl. Under these conditions there was no difference between line 271 and the control at low K+, but at high Na+ there was a rapid depolarisation that was significantly larger in control plants. 22Na uptake was measured in this line and there was a significant decrease in uptake at 100 mm NaCl in the transgenic line when compared with the control. The two transgenic lines were grown at high NaCl (200 mm) and analysed for growth and root sodium content. Lines 271 and 223 showed enhanced growth under salinity when compared with the control and had lower sodium in the root. Secondary ion mass spectrometry (SIMS) analysis of transverse sections of the root showed that Na+ and K+ were strongly localised to stelar regions when compared with other ions, and that the Na+ : K+ ratios were reduced in salt-stressed transgenic tissue when compared with the control.
