Selected AbstractsHigh total antioxidant activity and uric acid in tracheobronchial aspirate fluid of preterm infants during oxidative stress: an adaptive response to hyperoxia?ACTA PAEDIATRICA, Issue 3 2000G Vento The effect of O2 exposure, expressed by mean daily fractional inspired oxygen concentration (FiO2), was evaluated during the first 6 d of life in the tracheobronchial aspirate fluid of 16 mechanically ventilated preterm infants in terms of both antioxidant response and oxidative damage, by measuring total antioxidant activity, uric acid concentrations and protein carbonyl content. Each day linear regression analysis was performed and a positive correlation was found between total antioxidant activity and FiO2 during the study period, especially on day 2 of life (r= 0.91, p < 0.0001), but uric acid correlated only in the first 3 d, especially on the 2nd day (r= 0.83, p < 0.0001). No correlation was found between carbonyl content and FiO2. The highest values of total antioxidant activity (416 and 790 ,mol l,1) were found in 2 babies ventilated with highest FiO2: 1 and 0.80, respectively. Total antioxidant activity was not detectable or was very low in the babies not requiring O2 therapy. The highest value of uric acid (270 ,mol l,1) was found in the baby ventilated with 100% oxygen. Uric acid concentrations obtained in these babies were much higher then those reported in the bronchoalveolar lavage fluid of adults. Preterm babies seem to have an antioxidant response in the tracheobronchial aspirate fluid following an oxidative stress and uric acid may be physiologically important as an antioxidant of the respiratory tract, especially during the first days of life. Total polyphenols in the whole fruit and in the peel were determined by the Folin-Ciocalteu method, and the total antioxidant activity was determined by three established methods, diphenyl picrylhydrazyl (DPPH), azinobis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) and ferric reducing/antioxidant power (FRAP). Polyphenolic content for the whole fruit was in the range of 80,196 and for the peel 165,400 (mg Gallic Acid Equivalent (GAE)/100 g fresh weight) for both types of agricultural practices. Antioxidant activities of fruit extracts were also relatively similar and well correlated to their polyphenolic content. High-performance liquid chromatography (HPLC) analysis of the most important polyphenolics (chlorogenic acid, catechin, epicatechin, procyanidin B1 and B2, cyaniding 3-galactoside, phloridzin, quercetin 3-galactoside and quercetin 3-arabinoside) also showed that concentrations do not differentiate significantly between the organic and conventional apples. Statistical significance of differences in antioxidant activities among the same cultivars was relatively small (flesh + peel or peel only) for both types of apples. These results indicate that organic apples do not present higher antioxidant or nutritional value compared with conventionally grown ones, as far as polyphenolic content and total antioxidant activities are concerned. Group I was fed on fresh berseem fodder (Trifolium alexandrinum), whereas group II was fed on berseem fodder and concentrate mixture in 1 : 1 ratio to fulfil their nutritional requirements as per National Research Council (1981). Feed intake and milk yield were recorded daily, and milk samples were collected on 0, 30, 45 and 60 days of experimental feeding for the estimation of fat, vaccenic acid, conjugated linoleic acid (CLA), vitamins A and E, and total antioxidant activity. There was no effect (P > 0.05) of dietary treatments on daily as well as total milk yield and milk fat, solids-not-fat and total solids, although average total CLA (mg/g fat) content in milk during the trial was higher(P < 0.01) in group I (18.92) in comparison to group II (10.94). Vitamin A content averaged 0.46 and 0.27 µg/mL and vitamin E, 1.20 and 1.38 µg/mL in milk of groups I and II, respectively, were higher in the berseem-fed group (P < 0.01) and total antioxidant activity of milk was also higher (P < 0.01) in group I than in group II.
