Fractal analysis of Daphnia motion for acute toxicity bioassay

Abstract To quantify individual behavioral responses to toxic chemicals, the swimming motion of individual Daphniamagna was continuously monitored using a motion analysis system. The fractal dimension was introduced to compare the straightness or complexity of the swimming trajectory before and after exposure to toxic chemicals. Analysis indicated that the swimming trajectory of individual Daphnia has a fractal structure. The basal fractal dimension in the control medium was 1.35±0.01 (n = 50 Daphnia). Exposure to CuSO4 (10 ,g/L), organophosphorus (Dichlorvos; 10 ,g/L), and carbamate (Propoxur; 500 ,g/L) pesticide caused a significant increase in the fractal dimension with a latency of 60 min, reaching a maximal level of 2.26±0.34, 2.43±0.19, and 2.51±0.21, respectively, after a 120-min exposure. The magnitude of the change in the fractal dimension was related to the toxic chemical concentration and the exposure time. Threshold concentrations determined at 60 min of exposure were 10 ,g/L for CuSO4, 5 ,g/L for Dichlorvos, and 500 ,g/L for Propoxur. The toxicity index (EC50) values after 120 min of exposure were 6.31 ,g/L, 7.64 ,g/L, and 466 ,g/L for CuSO4, Dichlorvos, and Propoxur, respectively. Thus, the fractal dimension seems useful for analyzing and comparing complex trails, such as swimming trajectories, which could be used as the endpoint for an acute bioassay. © 2002 Wiley Periodicals, Inc. Increased optical damage resistance of Zr:LiNbO3 crystals

Abstract Zr: LiNbO3 crystals has been grown. The crystal composition and phase are analyzed by X-ray diffration. The optical damage resistance ability of Zr: LiNbO3 crystals is studied by the Sénarmont compensation method and the transmitted beam pattern distortion method. The saturated value of the birefringence change of 6 mol% Zr: LiNbO3 crystal is 1.01×10 -4, which is seven times smaller than that of congruent pure LiNbO3 crystal. The results of UV-Visible and IR absorption spectra of Zr: LiNbO3 crystals powerfully confirm that the optical damage resistance threshold concentration of the Zr4+ ions doped in LiNbO3 crystals is about 6 mol% in the melt. Growth and optical damage resistance of Sc, Er Co-doped LiNbO3 crystals

Abstract A series of Sc:Er:LiNbO3 crystals have been grown by Czochralski method. Their ultraviolet-visible (UV-Vis) absorption spectra was measured and discussed to investigate their defect structure. The optical damage resistance of Sc:Er:LiNbO3 crystals was characterized by the transmitted beam pattern distortion method. It increases remarkably when the concentration of Sc2O3 exceeds a threshold concentration. The optical damage resistance of Sc (3.0mol %):Er:LiNbO3 is much higher than that of the Er:LiNbO3. Electrodeposition of Lead at Boron-Doped Diamond Film Electrodes: Effect of Temperature

Abstract The electrodeposition of lead on boron-doped diamond has been studied with a view to identifying the fundamental parameters controlling the sensitivity and lower detection limit in anodic stripping voltammetry. Chronoamperometric transients are used to explore the deposition, indicating a progressive growth mechanism confirmed by ex situ AFM images. Linear sweep ASV experiments show a threshold concentration of ca 10,6,M below which no lead is detected; this is attributed to the need for nucleation of the solid phase on the electrode. Experiments with variable temperature show that this threshold can be usefully lowered at elevated temperatures. [source]


Enhancement of the NAD(P)(H) Pool in Saccharomyces cerevisiae

Abstract Asymmetric biosyntheses allow for an efficient production of chiral building blocks. The application of whole cells as biocatalysts for asymmetric syntheses is advantageous because they already contain the essential coenzymes NAD(H) or NADP(H), which additionally can be regenerated in the cells. Unfortunately, reduced catalytic activity compared to the oxidoreductase activity is observed in many cases during whole-cell biotransformation. This may be caused by low intracellular coenzyme pool sizes and/or a decline in intracellular coenzyme concentrations. To enhance the intracellular coenzyme pool sizes, the effects of the precursor metabolites adenine and nicotinic acid on the intracellular accumulation of NAD(H) and NADP(H) were studied in Saccharomyces cerevisiae. Based on the results of simple batch experiments with different precursor additions, fed-batch processes for the production of yeast cells with enhanced NAD(H) or enhanced NADP(H) pool sizes were developed. Supplementation of the feed medium with 95,mM adenine and 9.5,mM nicotinic acid resulted in an increase of the intracellular NAD(H) concentration by a factor of 10 at the end of the fed-batch process compared to the reference process. The final NAD(H) concentration remains unchanged if the feed medium was solely supplemented with 95,mM adenine, but intracellular NADP(H) was increased by a factor of 4. The effects of NADP(H) pool sizes on the asymmetric reduction of ethyl-4-chloro acetoacetate (CAAE) to the corresponding (S)-4-chloro-3-hydroxybutanoate (S-CHBE) was evaluated with S.,cerevisiae,FasB,His6 as an example. An intracellular threshold concentration above 0.07,mM NADP(H) was sufficient to increase the biocatalytic S-CHBE productivity by 25,% compared to lower intracellular NADP(H) concentrations. Biotic ligand model of the acute toxicity of metals.

Abstract The biotic ligand model (BLM) was developed to explain and predict the effects of water chemistry on the acute toxicity of metals to aquatic organisms. The biotic ligand is defined as a specific receptor within an organism where metal complexation leads to acute toxicity. The BLM is designed to predict metal interactions at the biotic ligand within the context of aqueous metal speciation and competitive binding of protective cations such as calcium. Toxicity is defined as accumulation of metal at the biotic ligand at or above a critical threshold concentration. This modeling framework provides mechanistic explanations for the observed effects of aqueous ligands, such as natural organic matter, and water hardness on metal toxicity. In this paper, the development of a copper version of the BLM is described. The calibrated model is then used to calculate LC50 (the lethal concentration for 50% of test organisms) and is evaluated by comparison with published toxicity data sets for freshwater fish (fathead minnow, Pimephales promelas) and Daphnia. Acute toxicity of para -nonylphenol to saltwater animals,

Abstract para -Nonylphenol (PNP), a mixture of alkylphenols used in producing nonionic surfactants, is distributed widely in surface waters and aquatic sediments, where it can affect saltwater species. This article describes a database for acute toxicity of PNP derived for calculating a national saltwater quality criterion. Using a flow-through exposure system with measured concentrations, we tested early life stages of four species of saltwater invertebrates and two species of fish. Static 96-h tests were also conducted on zoeal Homarus americanus, embryo-larval Mulinia lateralis, and larval Pleuronectes americanus. The number of organisms surviving the flow-through test was measured at 2, 4, 8, and 12 h and daily through day 7. Mortality for most species plateaued by 96 h. The ranked sensitivities (96-h 50% lethal concentrations, measured in micrograms per liter) for the species tested were 17 for Pleuronectes americanus, 37.9 (48-h 50% effective concentration) for Mulinia lateralis, 59.4 for Paleomonetes vulgaris, 60.6 for Americamysis bahia (formerly Mysidopsis bahia), 61.6 for Leptocheirus plumulosos, 70 for Menidia beryllina, 71 for Homarus americanus, 142 for Cyprinodon variegatus, and > 195 for Dyspanopius sayii. Values for the seven most sensitive of these species ranged over a factor of only 4.2. The narrow range of responses for PNP implies that exceeding a threshold concentration would endanger a large proportion of the aquatic community. [source]


Evaluation of detergents for the soluble expression of ,-helical and ,-barrel-type integral membrane proteins by a preparative scale individual cell-free expression system

Cell-free expression has become a highly promising tool for the fast and efficient production of integral membrane proteins. The proteins can be produced as precipitates that solubilize in mild detergents usually without any prior denaturation sttif. Alternatively, membrane proteins can be synthesized in a soluble form by adding detergents to the cell-free system. However, the effects of a representative variety of detergents on the production, solubility and activity of a wider range of membrane proteins upon cell-free expression are currently unknown. We therefore analyzed the cell-free expression of three structurally very different membrane proteins, namely the bacterial ,-helical multidrug transporter, EmrE, the ,-barrel nucleoside transporter, Tsx, and the porcine vasopressin receptor of the eukaryotic superfamily of G-protein coupled receptors. All three membrane proteins could be produced in amounts of several mg per one ml of reaction mixture. In general, the detergent 1-myristoyl-2-hydroxy- sn -glycero-3-[phospho- rac -(1-glycerol)] was found to be most effective for the resolubilization of membrane protein precipitates, while long chain polyoxyethylene-alkyl-ethers proved to be most suitable for the soluble expression of all three types of membrane proteins. The yield of soluble expressed membrane protein remained relatively stable above a certain threshold concentration of the detergents. We report, for the first time, the high-level cell-free expression of a ,-barrel type membrane protein in a functional form. Structural and functional variations of the analyzed membrane proteins are evident that correspond with the mode of expression and that depend on the supplied detergent. [source]


Is competition important to arctic zooplankton community structure?

Summary 1. Daphnia pulex and Daphnia middendorffiana are commonly found in the Toolik Lake region of arctic Alaska. These two species are very similar morphologically, although their natural distributions differ markedly: D. pulex is restricted to shallow ponds, while D. middendorffiana is widely distributed and found in a variety of ponds and lakes. We compared the reproductive capabilities of D. pulex and D. middendorffiana grown under similar resource conditions and in the absence of the invertebrate predator Heterocope septentrionalis. In situ life table and mesocosm experiments were conducted in Toolik Lake and Dam Pond, habitats that have historically contained natural populations of D. middendorffiana, but never D. pulex. 2. Daphnia pulex exhibited a significantly higher net growth rate than D. middendorffiana in both life table and mesocosm experiments although D. pulex has never been found in either Toolik Lake or Dam Pond. Daphnia middendorffiana exhibited a negative net growth rate in Dam Pond, which had lower resource levels then Toolik Lake. Therefore, the smaller D. pulex appears to have a lower food threshold concentration than the larger D. middendorffiana. 3. Our results indicate that D. pulex is a superior resource competitor in the Toolik Lake region. These results combined with distributional patterns suggest that the restricted distribution of D. pulex in these arctic lakes and ponds cannot be explained by resource competition alone. We suggest that in the presence of H. septentrionalis, predation is an important factor structuring arctic zooplankton communities in the Toolik Lake region. [source]


Sperm function tests and fertility

Summary Traditionally, the diagnosis of male infertility has depended upon a descriptive evaluation of human semen with emphasis on the number of spermatozoa that are present in the ejaculate, their motility and their morphology. The fundamental tenet underlying this approach is that male fertility can be defined by reference to a threshold concentration of motile, morphologically normal spermatozoa that must be exceeded in order to achieve conception. Many independent studies have demonstrated that this fundamental concept is flawed and, in reality, it is not so much the absolute number of spermatozoa that determines fertility, but their functional competence. In the light of this conclusion, a range of in vitro tests have been developed to monitor various aspects of sperm function including their potential for movement, cervical mucus penetration, capacitation, zona recognition, the acrosome reaction and sperm,oocyte fusion. Such functional assays have been found to predict the fertilizing capacity of human spermatozoa in vitro and in vivo with some accuracy. Recent developments in this field include the introduction of tests to assess the degree to which human spermatozoa have suffered oxidative stress as well as the integrity of their nuclear and mitochondrial DNA. Such assessments not only yield information on the fertilizing capacity of human spermatozoa but also their ability to support normal embryonic development. [source]


Effects of dofetilide on cardiovascular tissues from normo- and hypertensive rats

The aim was to test whether dofetilide has some potential for use in the treatment of heart failure. Dofetilide at ,3 times 10,5 M had no effect on the quiescent Wistar Kyoto (WKY) rat aorta, mesenteric and intralobar arteries, or the spontaneous contractions of the WKY rat portal vein. Dofetilide at 10,6 to 3 times 10,5 M relaxed the KCl-contracted aorta. Dofetilide at 10,9 -10,7 M augmented the force of contraction of left ventricle strips from 12- and 18-month-old WKY rats at 2 Hz. Spontaneously hypertensive rats (SHRs) at 12 and 17,21 months of age are models of cardiac hypertrophy and failure, respectively. The augmentation of force at 2 Hz with dofetilide was similar on 12-and 18-month-old WKY rats and 12-month-old SHRs but reduced on the 18-month-old SHR left ventricle. At a higher more physiological frequency, 4 Hz, the threshold concentration of dofetilide required to augment the force responses of 21-month-old SHR left ventricles was markedly increased and the maximum augmenting effect was decreased. Dofetilide at 10,7 -10,5 M reduced the rate of the 17-month-old WKY rat right atrium, and had a similar effect on age-matched SHR right atrium. In summary, dofetilide is a positive inotrope and negative chronotrope in the rat. However, as the positive inotropic effect is not observed with clinically relevant concentrations at a physiological rate in heart failure, dofetilide is unlikely to be useful as a positive inotrope in the treatment of heart failure. Use of ATP bioluminescence to determine the bacterial sensitivity threshold to a bacteriocin

Abstract A new ATP bioluminescence-based method was developed to determine the effectiveness of nisin on a sensitive strain of Lactococcus cremoris. The principle of the method is to quantify the release of adenylic-nucleotides (AN) by a sensitive strain under the action of the bacteriocin, with the complex luciferin,luciferase. Nisin-induced leakage of AN included ATP from a sensitive L. cremoris to the external medium immediately after the contact with the bacteria. The growth of L. cremoris was correlated with the extracellular AN content. The extracellular ATP and AN concentration exhibited a linear correlation to the logarithm of the nisin concentration. For the determination of the effectiveness threshold, the concentration of AN was more sensitive and more reliable than the direct quanti,cation of ATP. The effectiveness threshold, corresponding to a 100% inhibition of L. cremoris growth, was obtained for a null concentration of intracellular nucleotides, i.e. for a ANtot:ANext ratio = 1. For an initial concentration of 1.4 × 107 bacteria/mL, the nisin effectiveness threshold is 3.4 ± 0.01 mg nisin/L. Field studies on the environmental fate of the Cry1Ab Bt-toxin produced by transgenic maize (MON810) and its effect on bacterial communities in the maize rhizosphere

Abstract Field studies were done to assess how much of the transgenic, insecticidal protein, Cry1Ab, encoded by a truncated cry1Ab gene from Bacillus thuringiensis (Bt), was released from Bt-maize MON810 into soil and whether bacterial communities inhabiting the rhizosphere of MON810 maize were different from those of the rhizosphere of nontransgenic maize cultivars. Bacterial community structure was investigated by SSCP (single-strand conformation polymorphism) of PCR-amplified 16S rRNA genes from community DNA. Using an improved extraction and detection protocol based on a commercially available ELISA, it was possible to detect Cry1Ab protein extracted from soils to a threshold concentration of 0.07 ng/g soil. From 100 ng of purified Cry1Ab protein added per gram of soil, only an average of 37% was extractable. At both field sites investigated, the amount of Cry1Ab protein in bulk soil of MON810 field plots was always lower than in the rhizosphere, the latter ranging from 0.1 to 10 ng/g soil. Immunoreactive Cry1Ab protein was also detected at 0.21 ng/g bulk soil 7 months after harvesting, i.e. in April of the following year. At this time, however, higher values were found in residues of leaves (21 ng/g) and of roots (183 ng/g), the latter corresponding to 12% of the Cry1Ab protein present in intact roots. A sampling 2 months later indicated further degradation of the protein. Despite the detection of Cry1Ab protein in the rhizosphere of MON810 maize, the bacterial community structure was less affected by the Cry1Ab protein than by other environmental factors, i.e. the age of the plants or field heterogeneities. The persistence of Cry1Ab protein emphasizes the importance of considering post-harvest effects on nontarget organisms. [source]


Expression of the Pho regulon negatively regulates biofilm formation by Pseudomonas aureofaciens PA147-2

We report the isolation of insertional mutations to the pstC and pstA genes of the phosphate-specific transport (pst) operon that results in loss of biofilm formation by Pseudomonas aureofaciens PA147-2. Consistent with the known roles of the Pst system in Escherichia coli and Pseudomonas aeruginosa, both P. aureofaciens pst mutants were demonstrated to have defects in inorganic phosphate (Pi) transport and repression of Pho regulon expression. Subsequently, biofilm formation by the wild type was shown to require a threshold concentration of extracellular Pi. The two-component regulatory pair PhoR/PhoB is responsible for upregulation of Pho regulon expression in response to Pi -limiting environments. By generating phoR mutants that were unable to express the Pho regulon, we were able to restore biofilm formation by P. aureofaciens in Pi -limiting conditions. This result suggests that gene(s) within the Pho regulon act to regulate biofilm formation negatively in low-Pi environments, and that phoR mutations uncouple PA147-2 from such regulatory constraints. Furthermore, the inability of pst mutants to repress Pho regulon expression accounts for their inability to form biofilms in non-limiting Pi environments. Preliminary evidence suggests that the Pst system is also required for antifungal activity by PA147-2. During phenotypic analysis of pst mutants, we also uncovered novelties in relation to Pi assimilation and Pho regulon control in P. aureofaciens. [source]


Survivorship, tunneling and feeding behaviors of Coptotermes formosanus (Isoptera: Rhinotermitidae) in response to 2,-acetonaphthone-treated sand

Abstract In laboratory tests, 2,-acetonaphthone was incorporated into sand at different concentrations (4.16,200 mg kg,1) and evaluated for survivorship, feeding and tunneling effects on the Formosan subterranean termite, Coptotermes formosanus Shiraki. 2,-Acetonaphthone at 8.33 mg kg,1 sand significantly reduced survivorship (89,94%), tunnel area (68,91%) and food consumption (84,100%) compared with the control. Most of the dead workers were found at the release point and a few had traveled to the surface of the treated sand. Malformed workers (,27%) with an unexpected symptom of molting failure were observed, particularly at the lowest concentration tested (4.16 mg kg,1). In choice assays, threshold concentration for significant reduction in survivorship, tunnel construction in treated sand and food consumption in the treated-sand side was 8.33 mg kg,1 with termites collected from New Orleans, LA and 35.0 mg kg,1 for those from Lake Charles, LA. Termites actively fed and tunneled in the untreated sand, whereas 2,-acetonaphthone at 140 mg kg,1 completely inhibited consumption of food placed on the treated sand. Photo-stabilization of EPDM,clay nanocomposites: effect of antioxidant on the preparation and durability,

Abstract The present study is to examine the photo-stabilization effect of an antioxidant on the photo-oxidation of ethylene-propylene-diene monomer (EPDM),clay nanocomposites. During the preparation of EPDM,clay nanocomposites via melt processing antioxidants are usually incorporated along with clay, which allows phenolic antioxidant molecules to get adsorbed onto acidic clay platelets and their interaction with metallic impurities reduces the stabilizing efficiency of the antioxidant. The nanocomposites were obtained by solution dispersion followed by melt compounding of EPDM and organophilic montmorillonite (OMMT). The samples were characterized by conventional tools such as X-ray diffraction (XRD), Fourier Transform Infra Red (FT-IR) spectroscopy, and thermo-gravimetric analysis (TGA). It was found, upon photo-irradiation (,,>,290,nm) studies by following the changes in functional groups and surface morphology, that photo-degradation was lowered by the antioxidant and the efficiency of the antioxidant could be improved by initial incorporation of antioxidant in the EPDM matrix. In EPDM,clay nanocomposites, a stabilizing activity of the antioxidant was observed above some threshold concentration of the antioxidant. The relationship between the nanoclay reinforcement and stabilizing efficiency in terms of photo-oxidation and surface morphology for their applicability are discussed. Risk of weathered residual Exxon Valdez oil to pink salmon embryos in Prince William Sound

Abstract It has been hypothesized that pink salmon eggs incubating in intertidal streams transecting Prince William Sound (PWS) beaches oiled by the Exxon Valdez oil spill were exposed to lethal doses of dissolved hydrocarbons. Since polycyclic aromatic hydrocarbon (PAH) levels in the incubation gravel were too low to cause mortality, the allegation is that dissolved high-molecular-weight hydrocarbons (HPAH) leaching from oil deposits on the beach adjacent to the streams were the source of toxicity. To evaluate this hypothesis, we placed pink salmon eggs in PWS beach sediments containing residual oil from the Exxon Valdez oil spill and in control areas without oil. We quantified the hydrocarbon concentrations in the eggs after three weeks of incubation. Tissue PAH concentrations of eggs in oiled sediments were generally <100 ppb and similar to background levels on nonoiled beaches. Even eggs in direct contact with oil in the sediment resulted in tissue PAH loads well below the lethal threshold concentrations established in laboratory bioassays, and very low concentrations of HPAH compounds were present. These results indicate that petroleum hydrocarbons dissolved from oil deposits on intertidal beaches are not at concentrations that pose toxic risk to incubating pink salmon eggs. The evidence does not support the hypothesis that interstitial pore water in previously oiled beaches is highly toxic. [source]


A cautionary note on the use of species presence and absence data in deriving sediment criteria

Abstract In recent years, a variety of approaches to deriving sediment quality guidelines have been developed. One approach relies on establishing an empirical relationship between the concentration of a contaminant in sediment and the condition of some biological indicator, for example, combining measured sediment concentrations of contaminants combined with data on colocated benthic species to measure in situ community effects of contamination. Biological threshold concentrations derived in this manner are being considered or have already been adopted by some regulatory agencies as a means for deriving sediment guidelines (e.g., Canada's Provincial Sediment Quality Guidelines). In order to test the validity of this method, we constructed several Monte Carlo simulations to illustrate that the methodology used to develop these guidelines is flawed by the effects of sampling and statistical artifacts that emerge from undersampling a lognormal density function. As a case study, this paper will present the screening level concentration method used by the Ontario Ministry of the Environment (Toronto, ON, Canada) and provide the results of several probabilistic exercises highlighting these issues. We present a word of caution on the applicability of methods that rely exclusively on statistical and mathematical relationships between invertebrate data and sediment concentrations to derive sediment quality guidelines. Body size and food thresholds for zero growth in Dreissena polymorpha: a mechanism underlying intraspecific competition

Summary 1. Dreissena polymorpha is an extraordinarily successful invasive species that shows high recruitment of small juvenile mussels on established mussel banks. Such juvenile settlement on, and overgrowth of, large adult mussels; however, leads to competition with adults, and often at high densities and low-food concentrations. 2. The concept of food thresholds for zero growth has been a powerful approach to explaining size-related exploitative competition in different zooplankton species. We applied it to investigate whether food threshold concentrations for zero growth (C0) differ between juvenile and adult zebra mussels. 3. By determining body mass growth at various concentrations of a diet mixture (Nannochloropsis limnetica and Isochrysis aff. galbana) we demonstrate that the threshold food concentration for growth of juvenile mussels (C0 = 0.08 mg C L,1) is substantially lower than that for adults (C0 = 0.36 mg C L,1). 4. This indicates that, at low food availability, juvenile zebra mussels are competitively superior to their larger conspecifics. Within zebra mussel banks plankton food is substantially depleted and so the observed mechanism might ensure juvenile success and therefore the regeneration of mussel banks in nature. [source]


Redox Processes and Water Quality of Selected Principal Aquifer Systems

Reduction/oxidation (redox) conditions in 15 principal aquifer (PA) systems of the United States, and their impact on several water quality issues, were assessed from a large data base collected by the National Water-Quality Assessment Program of the USGS. The logic of these assessments was based on the observed ecological succession of electron acceptors such as dissolved oxygen, nitrate, and sulfate and threshold concentrations of these substrates needed to support active microbial metabolism. Similarly, the utilization of solid-phase electron acceptors such as Mn(IV) and Fe(III) is indicated by the production of dissolved manganese and iron. An internally consistent set of threshold concentration criteria was developed and applied to a large data set of 1692 water samples from the PAs to assess ambient redox conditions. The indicated redox conditions then were related to the occurrence of selected natural (arsenic) and anthropogenic (nitrate and volatile organic compounds) contaminants in ground water. For the natural and anthropogenic contaminants assessed in this study, considering redox conditions as defined by this framework of redox indicator species and threshold concentrations explained many water quality trends observed at a regional scale. An important finding of this study was that samples indicating mixed redox processes provide information on redox heterogeneity that is useful for assessing common water quality issues. Given the interpretive power of the redox framework and given that it is relatively inexpensive and easy to measure the chemical parameters included in the framework, those parameters should be included in routine water quality monitoring programs whenever possible. Estimating Background and Threshold Nitrate Concentrations Using Probability Graphs

Because of the ubiquitous nature of anthropogenic nitrate () in many parts of the world, determining background concentrations of in shallow ground water from natural sources is probably impossible in most environments. Present-day background must now include diffuse sources of such as disruption of soils and oxidation of organic matter, and atmospheric inputs from products of combustion and evaporation of ammonia from fertilizer and livestock waste. Anomalies can be defined as derived from nitrogen (N) inputs to the environment from anthropogenic activities, including synthetic fertilizers, livestock waste, and septic effluent. Cumulative probability graphs were used to identify threshold concentrations separating background and anomalous NO3 -N concentrations and to assist in the determination of sources of N contamination for 232 spring water samples and 200 well water samples from karst aquifers. Thresholds were 0.4, 2.5, and 6.7 mg/L for spring water samples, and 0.1, 2.1, and 17 mg/L for well water samples. The 0.4 and 0.1 mg/L values are assumed to represent thresholds for present-day precipitation. Thresholds at 2.5 and 2.1 mg/L are interpreted to represent present-day background concentrations of NO3 -N. The population of spring water samples with concentrations between 2.5 and 6.7 mg/L represents an amalgam of all sources of in the ground water basins that feed each spring; concentrations >6.7 mg/L were typically samples collected soon after springtime application of synthetic fertilizer. The 17 mg/L threshold (adjusted to 15 mg/L) for well water samples is interpreted as the level above which livestock wastes dominate the N sources. [source]


Approaches for derivation of environmental quality criteria for substances applied in risk assessment of discharges from offshore drilling operations

Abstract In order to achieve the offshore petroleum industries "zero harm" goal to the environment, the environmental impact factor for drilling discharges was developed as a tool to identify and quantify the environmental risks associated with disposal of drilling discharges to the marine environment. As an initial step in this work the main categories of substances associated with drilling discharges and assumed to contribute to toxic or nontoxic stress were identified and evaluated for inclusion in the risk assessment. The selection were based on the known toxicological properties of the substances, or the total amount discharged together with their potential for accumulation in the water column or sediments to levels that could be expected to cause toxic or nontoxic stress to the biota. Based on these criteria 3 categories of chemicals were identified for risk assessment the water column and sediments: Natural organic substances, metals, and drilling fluid chemicals. Several approaches for deriving the environmentally safe threshold concentrations as predicted no effect concentrations were evaluated in the process. For the water column consensus were reached for using the species sensitivity distribution approach for metals and the assessment factor approach for natural organic substances and added drilling chemicals. For the sediments the equilibrium partitioning approach was selected for all three categories of chemicals. The theoretically derived sediment quality criteria were compared to field-derived threshold effect values based on statistical approaches applied on sediment monitoring data from the Norwegian Continental Shelf. The basis for derivation of predicted no effect concentration values for drilling discharges should be consistent with the principles of environmental risk assessment as described in the Technical Guidance Document on Risk Assessment issued by the European Union. COMPARISON OF HEADSPACE SOLID PHASE MICROEXTRACTION AND XAD-2 METHODS TO EXTRACT VOLATILE COMPOUNDS PRODUCED BY SACCHAROMYCES DURING WINE FERMENTATIONS

ABSTRACT A modified headspace solid phase microextraction (HS-SPME) method was compared with Amberlite® XAD-2 resin for the extraction of volatile compounds. In the HS-SPME method, volatiles were extracted using an 85 ,m polyacrylate fiber from wines that contained a standardized amount of ethanol (10% v/v), NaCl (0.325 g/mL) and internal standards (dodecanol and nonanoic acid). Both extraction procedures yielded high relative recoveries (>92%) and reproducibilities (coefficient of variations , 11%) for the different higher alcohols, esters and medium-chain fatty acids. Overall, limits of detection for the HS-SPME and XAD-2 methods were below sensory threshold concentrations. HS-SPME and XAD-2 performed similarly in the analysis of a Riesling wine; however, the HS-SPME method did not require organic solvents and was generally quicker to perform. In applying the HS-SPME method, differences in concentrations of volatile compounds produced in Riesling and Chenin blanc wines by 11 different yeast strains were noted. [source]
