In a multicenter, prospectively randomized study we compared efficacy and side effects of a dexamethasone-cyclophosphamide (D/C) pulse therapy with a methylprednisolone-azathioprine (M/A) therapy in 22,patients with newly diagnosed pemphigus vulgaris and pemphigus foliaceus. Patients and methods: The 11,patients of the M/A group were treated with daily doses of methylprednisolone (initially 2,mg/kg body weight) and azathioprine (2,,,2,5,mg/kg body weight) which were subsequently tapered. D/C pulse therapy in 11,patients consisted of intravenous administration of 100,mg dexamethasone/d on 3 consecutive days along with cyclophosphamide (500,mg) on day one. Pulses were initially repeated every 2,,,4 weeks and then at increasing intervals. In between the pulses, oral cyclophosphamide (50,mg) was given daily for 6,months. Results: Within 24,months after treatment initiation, 5/11,patients of the D/C group had a remission (complete remissions after discontinuation of therapy in 3,patients) and 6/11,patients had a progression. In the M/A group, there were remissions in 9/11,patients (complete remissions after discontinuation of therapy in 3,patients) and progression in 1/11,patients. There were more relapses in M/A therapy after remission than in D/C therapy. Side effects were more common in the M/A group. These differences were not significant (p > 0,05). Conclusion: Because of the high number of progressions in patients treated with D/C therapy, we can not confirm the encouraging results of earlier reports about pulse D/C therapy. Nevertheless D/C therapy seemed to be better tolerated and, in case of primary efficacy, was associated with fewer recurrences than M/A therapy. Zusammenfassung Hintergrund: Pemphiguserkrankungen sind potentiell lebensbedrohliche blasenbildende Autoimmunerkrankungen, die üblicherweise mit hochdosierten Kortikosteroiden in Kombination mit Immunsuppressiva behandelt werden. In einer multizentrischen, prospektiven, randomisierten Studie verglichen wir die Wirksamkeit und Nebenwirkungen einer Dexamethason-Cyclophosphamid (D/C)-Pulstherapie mit einer Methylprednisolon-Azathioprin (M/A)-Therapie bei 22,Patienten mit neu diagnostiziertem Pemphigus vulgaris und Pemphigus foliaceus. Patienten und Methoden: 11,Patienten der M/A-Gruppe wurden kontinuierlich oral mit Methylprednisolon (initial 2,mg/kg Körpergewicht/Tag) und Azathioprin (2,,,2,5,mg/kg Körpergewicht/Tag) behandelt; die Dosen wurden schrittweise reduziert. Die Therapie bei den 11,Patienten der D/C-Gruppe erfolgte durch intravenöse Gabe von 100,mg Dexamethason/Tag an 3 aufeinander folgenden Tagen und 500,mg Cyclophosphamid am ersten Tag. Die Pulstherapie wurde zunächst alle 2,,,4 Wochen, dann in längeren Abständen wiederholt. Im Intervall wurden 50,mg Cyclophosphamid/Tag oral für 6,Monate verabreicht. Ergebnisse: Innerhalb von 24,Monaten nach Therapiebeginn kam es bei 5 von 11,Patienten der D/C-Gruppe zu einer Remission (komplette Remission nach Absetzen der Therapie bei 3,Patienten); bei 6 der 11,Patienten verlief die Erkrankung progredient. In der M/A-Gruppe kam es bei 9 von 11,Patienten zu einer Remission (komplette Remission nach Absetzen der Therapie bei 3,Patienten) und bei einem Patienten zu einer Progression. In der M/A-Gruppe traten häufiger Rezidive nach Remission auf als in der D/C-Gruppe. Therapienebenwirkungen kamen in der M/A-Gruppe häufiger vor. Diese Unterschiede waren nicht signifikant (p > 0,05). Schlußfolgerungen: Aufgrund der hohen Anzahl von Progressionen bei Patienten der D/C-Gruppe können wir die positiven Ergebnisse früherer Berichte über die D/C-Pulstherapie nicht bestätigen. Dennoch scheint die D/C-Therapie, beim einzelnen Patienten einmal erfolgreich, seltener zu Rezidiven zu führen und möglicherweise auch besser verträglich zu sein als die M/A-Therapie. Ng Abstract Reaction of [Zn(,-TAG){N(SiMe3)2}]2 {TAG = N=C[N(CH2CH3)2N(CH3)2] (DEDMG), N=C{[NCH2CH2CH2CdH2(N,Cd)]N(CH2CH3)2} (DEPYRG) and N=C{[NCH2CH2CH2CH2CeH2(N,Ce)]N(CH2CH3)2} (DEPIPG)} with 2 equiv. of ethanol (EtOH) and 2 equiv. of HOAr {OAr = OC6H3(CMe3)-2-(CH3)-6 (BMP) or OC6H2[C(CH3)3]2 -2,6-(CH3)-4 (4MeDBP)} results in dizinc alkoxides with the general formula [Zn(,-OEt)(OAr)(H-TAG)]2 (1,3). Et2Zn was additionally treated with 2 equiv. of 1,1,3,3-tetramethylguanidine (H-TMG) and H-BMP or HOC6H3(C6H5)2 -2,6 to yield [Zn(BMP)2(H-TMG)2] (4) and [Zn{OC6H3(C6H5)2 -2,6}2(H-TMG)2] (5). Complexes 1, 2, 4, and 5 were characterized by single-crystal X-ray diffraction. Polymerization of rac -lactide with 1,5 and [Zn(,-OMe)(4MeDBP)(H-TMG)]2 (6) were found to generate polylactide (PLA). The bulk powders for all complexes were found to be in agreement with the crystal structures based on elemental analyses, FTIR spectroscopy, and 1H and 13C NMR spectroscopic studies. [source] Postprandial lipemic response to alpha-linolenic acid rich oil, butter, and olive oilEUROPEAN JOURNAL OF LIPID SCIENCE AND TECHNOLOGY, Issue 9 2010Julia Svensson Abstract Postprandial lipemia varies with composition of dietary fat due to partitioning of fatty acids between ,-oxidation, incorporation into TAG, and tissue lipids. Effects of alpha-linolenic acid (ALA) are poorly characterized. Lipase-catalyzed transesterification was used to produce a novel ALA-oil (35% ALA) from rapeseed and linseed oil. We hypothesized a lower postprandial lipemic response with ALA-oil than with olive oil and butter due to higher ,-oxidation of ALA. A randomized crossover study with 26 healthy men compared the effects on plasma lipids 7,h after a breakfast containing 35,g ALA-rich oil, butter fat, or olive oil. The incremental area under curve for plasma TAG was lower with butter than with olive oil (34%, p<0.05) and ALA-oil (25%, ns). After ALA-oil percentage ALA increased, in TAG to a constant level of 7,mol% and in NEFA to 6% after 7,h. Since total NEFA increased with time the amount of exogenous ALA in NEFA also increased. Butter resulted in lower postprandial lipemia than the oils, the difference exceeding what is expected from the presence of short and medium chain fatty acids in butter. There was a considerable recirculation of ALA into the NEFA pool available for oxidation. Practical application: Enzymatic transesterification was used to produce a dietary oil rich in ALA. By randomizing the partitioning of ALA more evenly between the TAG molecules the risk of oxidation could be reduced. Analyses showed that the ALA-oil was stable during storage for at least 3 months. Enzymatic transesterification could be used as an advantageous method to design an ALA rich dietary oil with new properties regarding fatty acid composition, susceptibility to oxidation, and effects on blood lipids. [source] Cell surface analysis of the lipid-discharging obligate hydrocarbonoclastic species of the genus AlcanivoraxEUROPEAN JOURNAL OF LIPID SCIENCE AND TECHNOLOGY, Issue 6 2010Alvin Brian Lange Abstract This study presents novel information useful for addressing the question how species of the genus Alcanivorax discharge triacylglycerols (TAG) and/or wax esters (WE). The observed structures were referred as "blebs" according to Gauthier et al.1 to avoid confusion with other discharging phenomena. The cells were aerobically cultivated on solid media and not in liquid media to maintain the cells in the native state, and were investigated by transmission electron microscopic (TEM) and scanning electron microscopic (SEM) methods to document the surface structures of the cells. The phenomenon of lipid export could be allocated to three phases: phase I: protrusion formation of the cell membrane occurred; phase II: discharging progressed further with blebs becoming larger; and phase III: the blebs at the cell surface were separated from the cells. Using freeze-fracture micrographs by TEM, vesicle experiments and TLC, we have shown that the blebs contained TAGs and WEs. The results shown in this study will support further research to unravel the unknown discharging mechanism. In addition, the formation of an extensive extracellular matrix was observed by SEM. [source] Comparative analysis of triacylglycerols from Olea europaea L. fruits using HPLC and MALDI-TOFMSEUROPEAN JOURNAL OF LIPID SCIENCE AND TECHNOLOGY, Issue 5 2010Faouzi Sakouhi Abstract MALDI-TOFMS and HPLC are two analytical methods that were used to characterize triacylglycerols (TAG) of the Meski, Sayali, and Picholine Tunisian olive varieties. The HPLC chromatograms of the oils showed the presence of 15 TAG species, among which triolein (OOO) was the most abundant (21,48%). In the Sayali cultivar, OOO was the predominant TAG species followed by POO and LOO. However, the minor TAG molecules were represented by LnLO and LnLP. MALDI mass spectra produced sodiated ([M,+,Na]+) and potassiated ([M,+,K]+) TAG molecules; only the major TAG were potassiated [OOO,+,K] ([OOO,+,K]+, [POO,+,K]+, and [LOO,+,K]+). In contrast to the HPLC chromatograms, the MALDI mass spectra showed 13 peaks of TAG. The major peak was detected at m/z,907, which corresponds to OOO with an Na+ adduct. The results from both HPLC and MALDI techniques predict the fatty acid composition and their percentages for each olive variety. Practical applications: TAG are the main components in vegetable oils. These biomolecules determine the physical, chemical, and nutritional properties of the oils. The nutritional benefits of TAG are related to DAG (moderate plasma lipid level) and esterified FA, which are intermediate biosynthetic molecules of TAG. TAG analysis is necessary to discriminate between oils of different origin, since some oils have similar FA profiles. Olive products, oils, and table olives, are the main diet sources of TAG in the Mediterranean countries. In this work, chromatographic and spectrometric methods were used for TAG analysis and characterization of Tunisian olive varieties. [source] Calorimetric study of milk fat/rapeseed oil blends and their interesterification productsEUROPEAN JOURNAL OF LIPID SCIENCE AND TECHNOLOGY, Issue 4 2009Mario Aguedo Abstract Milk fat (MF) and rapeseed oil (RO) blends were analyzed by differential scanning calorimetry (DSC). It was shown that peak and onset temperatures can be used to determine the percentage of each fat in the blend and that the relative enthalpy of one peak assigned to low-melting triacylglycerols (TAG) can also be used to determine the percentage of RO in the blend. A linear relation was also established between MF content of the blend and its dropping point (DP), indicating that DP can be linearly related with the above DSC data. A blend of MF/RO 70,:,30 (wt/wt) was then chosen as a model system for enzymatic interesterification (EIE). The applicability of DSC analyses to EIE products was checked and a correct correlation could be established between DSC values and the interesterification degree and DP. Among the data from the DSC profiles, the peak associated with low-melting TAG was the best indicator of the reaction course. In the same way, a high-melting MF stearin fraction was interesterified with RO. In that case, onset temperatures and peak "a" were better reaction indicators than for the interesterified MF/RO blend. We therefore suggest that values from DSC endotherms could be used to monitor EIE of fat blends. [source] Regiospecific distribution of fatty acids in triacylglycerols and phospholipids from broad beans (Vicia faba)EUROPEAN JOURNAL OF LIPID SCIENCE AND TECHNOLOGY, Issue 4 2009Hiromi Yoshida Abstract Regiospecific distributions of fatty acids of triacylglycerols (TAG) and phospholipids (PL) separated from broad beans (Vicia faba) of four cultivars (Minpo, Sanuki, Nintoku and Sanren) were investigated. The major lipid components were PL (47.5,50.5,wt-%) and TAG (47.7,50.1,wt-%), while steryl esters, hydrocarbons, free fatty acids, diacylglycerols and monoacylglycerols were present in minor proportions (1.6,2.4,wt-%). The PL components isolated from the four cultivars were phosphatidylcholine (56.4,58.4,wt-%), phosphatidylethanolamine (20.3,21.7,wt-%) and phosphatidylinositol (16.6,18.6,wt-%). Phosphatidylinositol was unique in that it had the highest saturated fatty acid content among these PL. The principal characteristics of the fatty acid distribution in the TAG and PL were evident in the beans: Unsaturated fatty acids were predominantly concentrated in the sn -2 position while saturated fatty acids primarily occupied the sn -1 or sn -3 position in these lipids. The lipid components and fatty acid distributions were almost the same in the four cultivars and were not influenced by genetic variability and planting location. These results could be useful information to both consumers and producers for the manufacture of traditional broad bean foods in Japan. [source] Triacylglycerol migration and bloom in filled chocolates: Effects of low-temperature storageEUROPEAN JOURNAL OF LIPID SCIENCE AND TECHNOLOGY, Issue 3 2009Frédéric Depypere Abstract This study investigated the effect of storage temperature on triacylglycerol (TAG) migration, visual fat bloom and taste of industrially produced milk chocolates with a hazelnut-based filling. The chocolates were stored for up to 10,months at 18,°C, either directly after production or with the inclusion of a variable time at ,20 or 4,°C immediately after production and prior to further storage at 18,°C. TAG migration from the filling through the chocolate shell was quantified by HPLC analysis of chocolate sampled from the chocolates' surface. Both [OOO/SOS] and [LOO/SOS] were used as markers for oil migration. Compared to storage at 18,°C only, chilling or freezing of the chocolates for part of the storage time was found to reduce the amount of TAG migration. Effects on diffusion, capillary transport and TAG immobilization during the thermal treatment can be raised as possible reasons for this decrease. Furthermore, storage at ,20,°C decreased oil migration during subsequent storage at 18,°C. This suggests a crystallization effect during the storage at ,20,°C, leading to permanent (micro)structural changes. Although a thermal treatment at 4,°C compared to ,20,°C was less effective in retarding TAG migration, storage at low positive temperatures immediately after production appears already beneficial in the prevention of visual fat bloom. Adverse effects of the thermal treatments on the chocolates' taste were not observed. [source] Characterization of volatile compounds and triacylglycerol profiles of nut oils using SPME-GC-MS and MALDI-TOF-MSEUROPEAN JOURNAL OF LIPID SCIENCE AND TECHNOLOGY, Issue 2 2009Stefanie Bail Abstract Several nut oil varieties mainly used as culinary and overall healthy food ingredients were subject of the present study. Headspace solid-phase microextraction combined with gas chromatography-mass spectrometry was employed in order to determine the qualitative composition of volatile compounds. Furthermore, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry was used in order to assess the profiles and relative composition of the prevalent triacylglycerols (TAG) within the oils. The headspace of the majority of oil samples was dominated by high contents of acetic acid (up to 42%) and hexanal (up to 32%). As nut oils are typically gained by cold-pressing from previously roasted nuts, characteristic pyrazine derivatives as well as degradation products of long-chain fatty acids were detected. TAG analysis of these oils revealed a quite homogeneous composition dominated by components of the C52 and C54 group composed mainly of oleic (18:1), linoleic (18:2), stearic (18:0) and palmitic (16:0) acid residues representing together between 65 and 95% of the investigated nut oils. The TAG profiles showed characteristic patterns which can be used as ,fingerprints' of the genuine oils. Nut oils exhibiting quite similar fatty acid composition (e.g. hazelnut, pistachio and beech oil) could be clearly discriminated based on TAG showing significant differences between the oils. [source] Positional distribution of fatty acids in triacylglycerols and phospholipids from adzuki beans (Vigna angularis)EUROPEAN JOURNAL OF LIPID SCIENCE AND TECHNOLOGY, Issue 2 2008Hiromi Yoshida Abstract The fatty acid distributions of triacylglycerols (TAG) and major phospholipids (PL) obtained from adzuki beans (Vigna angularis) were investigated. The total lipids extracted from the beans were separated by thin-layer chromatography (TLC) into eight fractions. The major lipid components were PL (63.5,wt-%), TAG (21.2,wt-%), steryl esters (7.5,wt-%) and hydrocarbons (5.1,wt-%), while free fatty acids, diacylglycerols (1,3-DAG and 1,2-DAG) and monoacylglycerols were also present in minor proportions (0.2,1.1,wt-%). The major PL components isolated from the beans were phosphatidylcholine (45.3,wt-%), phosphatidylethanolamine (25.8,wt-%) and phosphatidylinositol (21.5,wt-%). Phosphatidylinositol was unique in that it had the highest saturated fatty acid content among the three PL. With a few exceptions, however, the principal characteristics of the fatty acid distribution in the TAG and three PL were evident in the beans: Unsaturated fatty acids were predominantly concentrated in the sn -2 position while saturated fatty acids primary occupied the sn -1 or sn -3 position in the oils of the adzuki beans. In general, these results could be useful to both consumers and producers for the manufacture of traditional adzuki foods in Japan. [source] Using lipid profiles and genotypes for the characterization of Corsican olive oilsEUROPEAN JOURNAL OF LIPID SCIENCE AND TECHNOLOGY, Issue 1 2008Virginie Bronzini de Caraffa Abstract In 2004, the Corsican producers of olive oils obtained a French protected designation of origin (PDO) "huile d'olive de Corse", but up to now specifications of Corsican oil production do not clearly indicate the oil attributes related to the territory of production. That is why the fatty acid and triacylglycerol (TAG) compositions of olive oils from the nine main cultivars used to produce oils under PDO were determined and related to the olive variety. The results showed (i),that the nine cultivars covered only four olive varieties, as revealed by random-amplified polymorphic DNA markers, (ii),that the lipid composition of oils is strongly dependent on the variety, and (iii),that the lipid composition of the four varieties is completely discriminated on the basis of the proportions of four TAG (OOO, OOL, PoOO, OOL) and one fatty acid (18:0). These results clearly establish the relationships between some characteristics of oils and the area of production (Corsica) for at least three varieties that are originated from Corsica. For the fourth variety, other investigations on minor compounds and on sensory attributes of oils must be undertaken to link some oil traits to the territory of production. [source] Evaluation of the stability of blends of sunflower and rice bran oilEUROPEAN JOURNAL OF LIPID SCIENCE AND TECHNOLOGY, Issue 5 2007Samia Mezouari Abstract Blends of sunflower oil (SFO) and rice bran oil (RBO) were evaluated for their stability. Additionally, known amounts of natural antioxidants extracted from RBO were added to SFO, and their protective effect was compared to that of the blends. The results found indicate that by raising the amount of RBO, from 10 to 50%, an increase of OLO, OLP, PPL, OOO, PPO, OPO, 18:1 and 16:0 occurred, followed by a decrease of LLL, LLO, and 18:2. These changes in fatty acid and triacylglycerol (TAG) composition led to an increase of the oil stability index at 120,°C and a reduction of polymer TAG formation in the heated blends at 180,°C during 8,h. A comparable protective effect of natural antioxidants to that of blending was observed in a 50,:,50 blend, by remarkably increasing the induction period. [source] Comparative analysis of triacylglycerol composition, melting properties and polymorphic behavior of palm oil and fractionsEUROPEAN JOURNAL OF LIPID SCIENCE AND TECHNOLOGY, Issue 4 2007Sabine Braipson-Danthine Abstract Palm oil is without doubt the most widely fractionated oil. Dry fractionation is based on differences in the melting points of triacylglycerols (TAG) which will crystallize selectively during the cooling process. Unfortunately, limitations due to intersolubility, closely linked to polymorphism, induce formation of co-crystals at each crystallization step. For this reason, only restricted TAG enrichments are observed. In this work, a series of samples (24) of palm oil, solid and liquid fractions (stearins, mid fractions, oleins and superoleins) have been selected and examined in terms of TAG composition (by HPLC), differential scanning calorimetry (DSC) melting profile and variable temperature powder X-ray diffraction pattern. Three major endotherms [low-melting, high-melting and very high-melting peaks (LMP, HMP and VHMP)] are detected in the DSC melting profiles (5,°C/min). The VHMP is only recorded for palm stearin which contains more SSS components. The HMP contribution is weak for palm olein and even not observed for palm super oleins. The LMP is usually made up of UUU, SUU and SUS components; SUS components are observed in both LMP and HMP; the HMP is also made up of some SSS, except for palm oleins and super oleins. Sub-,2, sub-,1, ,, ,'2, ,'1 and , polymorphic forms are recorded; the LMP components preferentially crystallize in sub-,2, sub-,1 and ,,forms; the HMP components generally crystallize in ,'2 and ,'1, with a tendency to exhibit ,,crystals, depending on the SSS content. Components of the VHMP have an increased tendency to stabilize in the ,,form; in view of the results, we can assume that there is a clear relationship between TAG composition, melting properties and polymorphic behavior and of palm oil and fractions. [source] Production and behavioural responses of high- and low-yielding dairy cows to different periods of access to grazing or to a maize silage and soyabean meal diet fed indoorsGRASS & FORAGE SCIENCE, Issue 4 2006O. Hernandez-Mendo Abstract The study examined whether high-yielding cows grazing pasture respond differently from low-yielding cows in milk production and feeding behaviour, to increasing the time made available for eating a maize silage and soyabean meal (TAMS) diet indoors and reducing the time available for grazing (TAG). Two experiments, each lasting 42 d, were carried out in spring (Experiment 1) and autumn (Experiment 2) using Holstein-Friesian cows at two different levels of milk yield (MYL). Milk production and feeding behaviour were examined for TAG + TAMS systems of 19 h, TAG plus 1 h TAMS (19 + 1), and 5 h TAG plus 15 h TAMS (5 + 15). There were two levels of concentrate (0 and 6 kg d,1), and in the spring experiment two sward heights (4,6 and 8,10 cm) were also studied. Milk yield, persistency of milk yield, liveweight change and estimated total DM intake were significantly higher on the 5 + 15 than on the 19 + 1 grazing system in Experiment 1 but not in Experiment 2. There were no significant interactions of TAG + TAMS treatment with MYL for any production or behavioural measurements except for maize silage feeding time, where high MYL cows spent a significantly greater time eating maize silage than low MYL cows on the 5 + 15 treatment but not on the 19 + 1 treatment. It can be concluded that high- and low-yielding cows respond similarly in milk production and feeding behaviour to different combinations of TAG and TAMS. In autumn, estimated daily intakes of herbage were lower on both grazing treatments relative to spring, resulting from lower rates of herbage intake with no compensatory increase in grazing time. In contrast, rates of intake of maize silage were higher in autumn especially on the 19 + 1 system. These results may imply a change of preference from herbage to maize silage between spring and autumn. [source] Detecting vegetable oil adulteration in hazelnut paste (Corylus avellana L.)INTERNATIONAL JOURNAL OF FOOD SCIENCE & TECHNOLOGY, Issue 3 2009Josep Serra Bonvehi Summary The possibilities of detecting hazelnut paste adulterated with refined and non-refined vegetable oils have been studied. Research was focussed mainly on peanut, high oleic-acid sunflower, corn and soybean oils which have a similar composition to hazelnut oil. The analytical procedures to detect fatty acid (FA), triacylglycerol (TAG) and tocopherol profiles as indicators of adulteration were determined. The better indicators experimentally determined were seven FA (palmitic, stearic, linoleic, linolenic, arachidonic, behenic and lignoceric acids) and different TAG with three unsaturated FA (the code letters used for FA are: P = C16:0; S = C18:0; O = C18:1; L = C18:2;; Ln = C18:3) (LLLn, LLL and OOO), two unsaturated FA (POL, PLL and SOO), and one unsaturated FA (PPL). As expected, when refined vegetable oils were added to hazelnut paste, the increment of stigmasta-3,5-diene allowed detection at levels of 2% oil added. Limits of detection were measured using standard and adulterated hazelnut with different amounts of non-refined vegetable oils added (5%, 10%, 20% and 30%). The distribution of tocopherols and tocotrienols is highly useful, except in the case of added sunflower oil. The differences between the experimental and theoretical values of the TAG with equivalent carbon number (,ECN) of 42 does not improve the detection limit of hazelnut paste adulterated with peanut or sunflower oils. Similarly, tocopherols usually added to refined vegetable oils as an antioxidant were also determined. The changes in slip melting point (SMP), solid fat content (SFC), carbon number (CN) triacylglycerol (TAG) composition, induction period (IP) of oxidation at 120 °C and IP of crystallisation at 20 °C of blends before and after treatments were investigated. Both BT and TB methods resulted in an increase in the CN48 TAG molecules, SMP and SFC, and a decrease in the IP of oxidation and crystallisation of initial blends. Samples made by TB method had higher CN48 TAG content, SMP, SFC and IP of oxidation, and lower IP of crystallisation than those made by BT method. Correlation between SFC at 20 °C and saturated fatty acid (SFA) content of the treated blends indicated that the SFA must be higher than 33.1% and 26.8% for BT and TB methods, respectively, to obtain fats suitable for use as vanaspati. In 1996 a number of specimens became available to the European Monotreme and Marsupial Taxon Advisory Group (TAG), which recommended that the animals should be distributed to three zoos (London ZSL, Poznan Zoo and Berlin Zoologischer Garten) so that a management and breeding protocol could be developed in case this fairly common species becomes threatened in the future. London ZSL acquired 3.3 Striped possums to establish a founder captive population and two joeys were born, one in April 1999 and the other in March 2000. Purebred Yorkshire (n = 131), Duroc (n = 136), Hampshire (n = 49), Spotted (n = 35), Chester White (n = 74), Poland China (n = 51), Berkshire (n = 169) and Landrace (n = 82) pigs (n = 727; 427 barrows and 300 gilts) from the 1994 and 2001 National Barrow Show Sire Progeny Tests were used. For statistical analyses, a mixed model was used that included fixed effects of breed, sex, HAL-1843TM genotype, year, slaughter date within each year, interaction of breed × sex and random effects of sire and dam within breed. Breeds and sex were significantly associated with the percentages of the majority fatty acids in TAG. Duroc pigs had greater total saturated fatty acids (SFA) and lower total monounsaturated fatty acids (MUFA) (p < 0.05) contents than did pigs of all other breeds except Berkshire (p > 0.05). The concentration of total polyunsaturated fatty acids (PUFA) was the greatest in Hampshire pigs (p < 0.05). The content of total SFA was greater (p < 0.01), whereas the concentrations of total MUFA and PUFA were lower (p < 0.01) in barrows than those in gilts. The contents of major SFA in PL did not differ significantly among pigs from different breeds and sex groups. However, breed and sex significantly affected the concentrations of major MUFA and PUFA in PL and strong negative correlation between the total contents of MUFA and PUFA in PL was observed in the current study. Chester White pigs had greater total MUFA and lower total PUFA contents (p < 0.05) in PL than did pigs of all other breeds except Spotted (p > 0.05). In contrast to breed and sex effects, the concentrations of fatty acids in PL were more affected by HAL-1843TM genotype than those in TAG. The content of C16:0, a major SFA in PL, differed significantly in pigs with different HAL-1843TM genotypes. In conclusion, these results suggest that breed and sex are important sources of the variations for fatty acid composition of TAG and PL in LM. The aim of this study was to study the final fatty acid (FA) contribution and oxidation stability of an infant formula prepared using the structured lipid DCAO (DHA-containing customized amaranth oil). DCAO was included as complementary fat in a "prototype" infant formula, and prepared in parallel with a "control" infant formula under the same processing conditions. The same ingredients but different complementary fat sources were used. A blend of the most commonly used vegetable oils (palm olein, soybean, coconut, and high-oleic sunflower oils) for infant formula was used instead of DCAO in the "control" formula. Additionally, "prototype" and "control" infant formulas were compared to a "commercial" product in terms of FA composition. The oxidative stability index (OSI) of the extracted fats from "prototype,""control," and "commercial" infant formulas were evaluated and compared to the OSI of the substrate fat replacers used. DCAO was the least stable compared to other fat analogs. The use of commercial antioxidants in DCAO containing products should prevent oxidation and therefore increase their stability. [source] Synthesis of Diacylglycerols Containing CLA by Lipase-Catalyzed EsterificationJOURNAL OF FOOD SCIENCE, Issue 7 2006In-Hwan Kim ABSTRACT:, Diacylglycerols (DAG) were prepared by esterification of glycerol with conjugated linoleic acid (CLA) in the presence of an immobilized 1,3-regiospecific lipase from Rhizomucor miehei and vacuum conditions. The effects of several parameters, namely, temperature, enzyme loading, stirring speed, and vacuum, on the concentration and the purity of the DAG were studied. The reaction temperature influenced both the reaction rate and the concentration of the DAG. The rate of DAG synthesis increased as the enzyme loading increased. However, for high enzyme loadings, the concentration of triacylglycerols (TAG) increased significantly at long reaction times and, as a result, the purity of the DAG decreased. When the stirring speed increased from 150 to 450 rpm, the DAG concentration increased significantly. However, at stirring speeds above 450 rpm, no significant increases in DAG concentration were observed. When the pressure was decreased from 20 to 3 mmHg, the maximum concentration of DAG increased from 76.0% to 80.5%. No increase in the DAG concentration was observed when the pressure was decreased from 3 to 1 mm Hg, even though a slightly higher DAG purity was achieved at 1 mm Hg. For the range of absolute pressures tested, the concentrations of 1,2-DAG were less than 1%. Ortega ABSTRACT: Interesterification (acidolysis) of fully hydrogenated soybean oil (melting point = 69.9 °C) with conjugated linoleic acid (CLA) was carried out in a batch reactor at 75 °C. Lipases from Candida antarctica, Rhizomucor miehei, Pseudomonas sp., and Thermomyces lanuginosus were used at 5% (wt/wt) of the total substrate load. The lipase from Rhizomucor miehei produced the fastest reaction rates, and the greatest extent of incorporation of CLA residues in acylglycerols was achieved in 12 h. Lipases from C. antarctica and T. lanuginosus produced slower initial rates, and maximum extents of incorporation of CLA residues were achieved in 24 h. The lipase from Pseudomonas sp. produced the slowest initial rate. The corresponding maximum extent of incorporation was reached in 48 h. Differential scanning calorimetry analysis of the triacylglycerol (TAG) fractions produced by C. antarctica, R. miehei, and T. lanuginosus lipases after purification by solid phase extraction showed little variation in melting point (60.4 °C, 62.8 °C, and 60.1 °C, respectively). By contrast, the corresponding TAG fraction produced by the Pseudomonas sp. lipase melted at 48.4 °C. The solute adsorption isotherms (bi-Langmuir model) were determined for both the two CSPs using the perturbation method. It was demonstrated that the sugar units were involved in the reduction of the apparent enantioselectivity through two phenomena: (i) the inhibition of some enantioselective contacts with low-affinity binding regions of the aglycone and (ii) a decrease in the stereoselective properties of the aglycone high-affinity binding pocket. The phenomenon (ii) was governed by both a decrease in the ratio of the enantiomer adsorption constant and a strong reduction of the site accessibility for D- and L-tryptophan. In addition, a temperature effect study was performed to investigate the chiral recognition mechanism at the aglycone high-affinity pocket. An enthalpy-entropy compensation analysis derived from the Grunwald model as well as the comparison with the literature data demonstrated that the enantioselective binding mode was dependent on an interface dehydration process. The change in the enantioselective process observed between the TAG and teicoplanin CSP was characterized by a difference of ca. 2,3 ordered water molecules released from the species interface. Rice bran oil, palm stearin and coconut oil were used as substrates for lipase-catalyzed reaction. RESULTS: After interesterification, 137,150 g kg,1 medium-chain fatty acid was incorporated into the triacylglycerol (TAG) of the interesterified fats. Solid fat contents at 25 °C were 15.5,34.2%, and slip melting point ranged from 27.5 to 34.3 °C. POP and PPP (,-tending TAG) in palm stearin decreased after interesterification. X-ray diffraction analysis demonstrated that the interesterified fats contained mostly ,, polymorphic forms, which is a desirable property for margarines. CONCLUSIONS: The interesterified fats showed desirable physical properties and suitable crystal form (,, polymorph) for possible use as a spreadable margarine stock. Therefore, our result suggested that the interesterified fat without trans fatty acid could be used as an alternative to partially hydrogenated fat. Hearts from obese diabetic and lean littermate control rats were perfused with VLDL and CM from diabetic and control rats. Metabolic fate of the lipoprotein TAG and contractile function were examined. Myocardial utilisation of both VLDL- and CM-TAG was increased in the diabetic state. Diabetic hearts oxidised diabetic lipoprotein-TAG to a greater extent than control lipoproteins; glucose oxidation was decreased. There was no difference in lipoprotein-TAG assimilation into diabetic heart lipids; diabetic lipoproteins were, however, a poor substrate for control heart tissue lipid accumulation. Although the proportion of exogenous lipid incorporated into tissue TAG was increased in diabetic hearts perfused with control lipoproteins, this effect was not seen in diabetic hearts perfused with diabetic lipoproteins. Myocardial heparin-releasable lipoprotein lipase (LPL) activity was moderately increased in the diabetic state, and diabetic lipoproteins increased tissue-residual LPL activity. Cardiac hydraulic work was decreased only in diabetic hearts perfused with diabetic CM. Compositional analysis of diabetic variant lipoproteins indicated changes in size and apoprotein content. Alterations in cardiac TAG-rich lipoprotein metabolism in type 2 diabetes are due to changes in both the diabetic myocardium and the diabetic lipoprotein particle; decreased contractile function is not related to cardiac lipid accumulation from TAG-rich lipoproteins but may be associated with changes in TAG-fatty acid oxidation. Smith Muscle contraction increases glucose uptake and fatty acid (FA) metabolism in isolated rat skeletal muscle, due at least in part to an increase in AMP-activated kinase activity (AMPK). However, the extent to which AMPK plays a role in the regulation of substrate utilization during contraction is not fully understood. We examined the acute effects of 5-aminoimidazole-4-carboxamide riboside (AICAR; 2 mm), a pharmacological activator of AMPK, on FA metabolism and glucose oxidation during high intensity tetanic contraction in isolated rat soleus muscle strips. Muscle strips were exposed to two different FA concentrations (low fatty acid, LFA, 0.2 mm; high fatty acid, HFA, 1 mm) to examine the role that FA availability may play in both exogenous and endogenous FA metabolism with contraction and AICAR. Synergistic increases in AMPK ,2 activity (+45%; P < 0.05) were observed after 30 min of contraction with AICAR, which further increased exogenous FA oxidation (LFA: +71%, P < 0.05; HFA: +46%, P < 0.05) regardless of FA availability. While there were no changes in triacylglycerol (TAG) esterification, AICAR did increase the ratio of FA partitioned to oxidation relative to TAG esterification (LFA: +65%, P < 0.05). AICAR significantly blunted endogenous TAG hydrolysis (LFA: ,294%, P < 0.001; HFA: ,117%, P < 0.05), but had no effect on endogenous oxidation rates, suggesting a better matching between TAG hydrolysis and subsequent oxidative needs of the muscle. There was no effect of AICAR on the already elevated rates of glucose oxidation during contraction. These results suggest that FA metabolism is very sensitive to AMPK ,2 stimulation during contraction. Eight novel SNPs were identified, which were AJ276223:g.2064T>A (SNP1), g.2155C>T (SNP2), g.2203G>T (SNP3), g.2268T>C (SNP4), g.2274G>A (SNP5), g.2340A>G (SNP6), g.2350T>C (SNP7) and g.2370A>G (SNP8). Complete linkage disequilibrium was observed among SNP1, 2, 4, 5, 6 and 8. Phenotypic data were collected from 573 cross-bred steers with six sire breeds, including Hereford, Angus, Brangus, Beefmaster, Bonsmara and Romosinuano. The genotypes of SNP1/2/4/5/6/8 were significantly associated with adjusted backfat thickness. The genotypes of SNP3 were significantly associated with triacylglycerol (TAG) content and fatty acid composition of longissimus dorsi muscle (LM) in Brangus-, Romosinuano- and Bonsmara-sired cattle. Cattle with g.2203GG genotype had greater concentrations of TAG, total lipid, total saturated fatty acid and total monounsaturated fatty acid than did cattle with g.2203GT genotype. The genotypes of SNP7 were significantly associated with fatty acid composition of LM. Cattle with genotype g.2350TC had greater amounts of several fatty acids in LM than did cattle with genotype g.2350CC. OXLEY Abstract Calanoid copepods are a rich source of marine lipid for potential use in aquafeeds. Copepod oil is primarily composed of wax esters (WE) and there are concerns over the efficiency of wax ester, versus triacylglycerol (TAG), digestion and utilization in fish. As smoltification represents a period of major physiological adaptation, the present study examined the digestibility of a high WE diet (Calanus oil; 48% WE, 26% TAG), compared with a TAG diet (fish oil; 58% TAG), in Atlantic salmon freshwater presmolts and seawater postsmolts, of similar age (9 months) and weight (112 g and 141 g initial, respectively), over a 98-day period at constant temperature. Fish grew significantly better, and possessed lower feed conversion ratios (FCR), in seawater than freshwater. However, total lipid apparent digestibility coefficient (ADC) values were significantly lower in seawater fish, as were total fasted bile volumes. Dietary Calanus oil also had a significant effect, reducing growth and lipid ADC values in both freshwater and seawater groups. Postsmolts fed dietary Calanus oil had the poorest lipid ADC values and analysis of faecal lipid class composition revealed that 33% of the remaining lipid was WE and 32% fatty alcohols. Dietary prevalent 22:1n-11 and 20:1n-9 fatty alcohols were particularly poorly utilized. A decrease in primary bile acid, taurocholate, concentration was observed in the bile of dietary Calanus oil groups which could be related to the lower cholesterol content of the diet. MARTÍN Abstract Total lipid (TL), lipid classes and fatty acid composition of neutral (NL) and polar (PL) lipids were studied in the gonads, liver and muscle of gilthead seabream males (Sparus aurata) fed a control diet (diet C) or an n-3 and n-6 highly unsaturated fatty acids (HUFA)-deficient diet (diet D), at different stages of the reproductive cycle. Between pre-spermatogenesis (November) and spermatogenesis (March), the lipid content was high and particularly rich in cholesterol, phosphatidylcholine and phosphatidylethanolamine in gonads from both dietary groups. At post-spermatogenesis (June), TL and especially PL dramatically decreased in the gonads from both groups. However, at this period diet C fish gonads were richer in triacylglycerides (TAG) than those from diet D fish. The liver lipid contents and particularly TAG were over 200% lower in June than in March for both groups. Nevertheless, the most noteworthy depletion of lipids during this period was achieved by the n-3 HUFA in diet D fish. Conversely, arachidonic acid (20:4n-6) did not decrease in NL or PL from gonads and liver in groups C and D. Muscle lipids from diet C fish were relatively insensitive to seasonal influences. However, in June, the muscle TAG content was significantly reduced in diet D fish. [source] |
