Laboratory simulation of the salt weathering of schist: II.

Abstract Recent developments in long term landform evolution modelling have created a new demand for quantitative salt weathering data, and in particular data describing the size distribution of the weathered rock fragments. To enable future development of rock breakdown models for use in landscape evolution and soil production models, laboratory work was undertaken to extend existing schist/salt weathering fragmentation studies to include an examination of the breakdown of sub-millimetre quartz chlorite schist particles in a seasonally wet tropical climate. Laser particle sizing was used to assess the impact of different experimental procedures on the resulting particle size distribution. The results reveal that salt weathering under a range of realistic simulated tropical wet season conditions produces a significant degree of schist particle breakdown. The fragmentation of the schist is characterized by splitting of the larger fragments into mid-sized product with finer material produced, possibly from the breakdown of mid-sized fragments when weathering is more advanced. Salinity, the salt addition method and temperature were all found to affect weathering rates. Subtle differences in mineralogy also produce variations in weathering patterns and rates. It is also shown that an increase in drying temperature leads to accelerated weathering rates, however, the geometry of the fracture process is not significantly affected. New procedures to decompose geomagnetic field variations and application to volcanic activitiy

SUMMARY We report the development of numerical procedures for extracting long-term geomagnetic field variations caused by volcanic activity from an observed geomagnetic field by using statistical methods. The newly developed procedures are to estimate the trend from the observed data, as well as variations of non-volcanic origin such as periodic components, components related to external geomagnetic variations and observational noise. We also aim at referring to data obtained at a remote standard geomagnetic observatory rather than using a temporarily installed reference site for reasons of data quality. Two different approaches,a Bayesian statistical method and a Kalman filter method,are applied to decompose the geomagnetic field data into four components for comparison. The number of filter coefficients and the degree of condition realizations are optimized on the basis of minimization of the information criteria. The two procedures were evaluated by using a synthetic data set. Generally, the results of both methods are equally sufficient. Subtle differences are seen at the first, several data points due to arbitrarily selected initial values in the case of the Kalman filter method and at the smaller residual for the Bayesian statistical method. The largest differences are in computation time and memory size. The Kalman filter method runs a thousand times faster on a testing workstation and requires less memory than the Bayesian method. The Kalman filter method was applied to the total intensity data at Kuchi-erabu-jima volcano. The result suggests that the procedure works reasonably well. [source]


Mass spectrometric analysis of 7-sulfoxymethyl-12-methylbenz[a]anthracene and related electrophilic polycyclic aromatic hydrocarbon metabolites

Abstract The Meso-region theory of polycyclic aromatic hydrocarbon (PAH) carcinogenesis predicts that the development of pronounced carcinogenicity depends on the introduction of a good leaving group on alkyl side-chains attached to the exceptionally reactive meso-anthracenic or L-region positions of PAHs. Thus, the first step in carcinogenesis by methylated PAHs such as 7,12-dimethylbenz[a]anthracene (DMBA) would be the hydroxylation of the L-region methyl groups, particularly the 7-methyl group. The second would be the formation of a metabolite, e.g. a sulfate ester, which is expected to be a good leaving group capable of generating a highly reactive benzylic carbocation. 7-Hydroxymethyl-12-methylbenz[a]anthracene (7-HMBA) is a metabolite of DMBA, and sulfation of 7-HMBA to a 7-sulfoxymethyl metabolite (7-SMBA) is a known Phase II metabolic process designed to facilitate excretion, but actually enabling more destructive side-reactions. These side-reactions occur with generation of an electrophilic 7-methylene carbonium ion, and/or by in vivo halide exchange to provide neutral side-products more capable of entering cells, especially those of DMBA target tissues. Electrospray ionization mass spectrometry (MS) enabled us to visualize 7-SMBA as an intact m/z 351 conjugate anion by negative mode, and as a released m/z 255 carbonium ion by positive mode. Upon prolonged refrigeration, 7-SMBA accumulated an m/z 383 photooxide, which appeared capable of re-evolving the starting material as visualized by tandem quadrupole MS, or MS/MS. The 7-SMBA carbonium ion provided interpretable fragments when studied by fragment ion MS/MS, including those representing the loss of up to several protons. Subtle differences in this property were encountered upon perturbing 7-SMBA, either by warming it at 37 °C for 2 h or by substituting the initial sulfoxy group with an iodo group. Side-reactions accounting for such proton losses are proposed, and are of interest whether they occur in the mass spectrometer, in solution or both; these proposals include acidity at the 12-methyl position and cyclization between the 12-methyl group and the adjacent C-1 position. It is also suggested that such side-reactions may comprise one route to relieving steric strain arising between the 12-methyl group and the angular benzo ring of 7-SMBA. QUANTITATIVE ANALYSIS OF SHAPE VARIATION IN TYPE AND MODERN POPULATIONS OF MERIDION (BACILLARIOPHYCEAE)

Subtle differences in valve morphology can indicate sexually isolated populations in diatoms (Bacillariophyceae). Shape descriptors, like Legendre coefficients, have been used to quantify differences in valve outline so that morphologically distinct groups can be recognized. Legendre coefficients were used as shape descriptors to quantify differences in valve outline among five populations in the genus Meridion Ag. Two novel populations of Meridion from North America, a North American population of M. constrictum Ralfs, type material for M. circulare (Greve.) Ag., and type material of M. constrictum were included in this analysis. Results of a principle components analysis and discriminate analyses on the shape descriptors of all five populations support the hypotheses that the two novel North American populations are undescribed taxa within Meridion and that the North American M. constrictum can be considered Meridion constrictum sensu stricto. A new taxon, Meridion hohii Rhode, is described. There appears to be greater diversity within the genus Meridion than is currently recognized. [source]


Like Water for Chocolate: Feasting and Political Ritual among the Late Classic Maya at Xunantunich, Belize

Subtle differences in the context of feasting and manners of food consumption can point to underlying levels of civil and social competition in state-level societies. Haute cuisine and high styles of dining are characteristic of societies with fully developed civil and social hierarchies such as Renaissance Europe and the Postclassic Aztec. Competitive yet socially circumscribed political and social organizations such as the Classic lowland Maya may have prepared elaborate diacritical meals that marked status, but the nature of feasting remained essentially patriarchal. Ancient Maya feasting is recognizable through archaeologically discernible pottery vessel forms that were used to serve festival fare such as tamales and chocolate. Comparison of ceramic assemblages across civic and household contexts at the site of Xunantunich, Belize, demonstrates that drinking chocolate, more so than eating tamales, served as a symbolic cue that established the political significance of events among the Classic Maya. Phorbol esters and adenosine affect the readily releasable neurotransmitter pool by different mechanisms at amphibian motor nerve endings

Phorbol esters and adenosine have been proposed to interact at common sites downstream of calcium entry at amphibian motor nerve endings. We thus studied the actions and interactions of phorbol esters and adenosine using electrophysiological recording techniques in conjunction with both binomial statistical analysis and high-frequency stimulation at the amphibian neuromuscular junction. To begin this study, we confirmed previous observations that synchronous evoked acetylcholine (ACh) release (reflected as endplate potentials, EPPs) is well described by a simple binomial distribution. We then used binomial analysis to study the effects of the phorbol ester phorbol dibutyrate (PDBu, 100 nm) and adenosine (50 µm) on the binomial parameters n (the number of calcium charged ACh quanta available for release) and p (the average probability of release), where the mean level of evoked ACh release (m) =np. We found that PDBu increased m by increasing the parameter n whilst adenosine reduced m by reducing n; neither agent affected the parameter p. PDBu had no effect on either the potency or efficacy of the inhibition produced by adenosine. Subtle differences between these two agents were revealed by the patterns of EPPs evoked by high-frequency trains of stimuli. Phorbol esters increased ACh release during the early phase of stimulation but not during the subsequent plateau phase. The inhibitory effect of adenosine was maximal at the beginning of the train and was still present with reduced efficacy during the plateau phase. When taken together with previous findings, these present results suggest that phorbol esters increase the immediately available store of synaptic vesicles by increasing the number of primed vesicles whilst adenosine acts at a later stage of the secretory process to decrease the number of calcium-charged primed vesicles. On the Nitroxide Quasi-Equilibrium in the Alkoxyamine-Mediated Radical Polymerization of Styrene

Abstract Summary: The range of validity of two popular versions of the nitroxide quasi-equilibrium (NQE) approximation used in the theory of kinetics of alkoxyamine mediated styrene polymerization, are systematically tested by simulation comparing the approximate and exact solutions of the equations describing the system. The validity of the different versions of the NQE approximation is analyzed in terms of the relative magnitude of (dN/dt)/(dP/dt). The approximation with a rigorous NQE, kc[P][N],=,kd[PN], where P, N and PN are living, nitroxide radicals and dormant species respectively, with kinetic constants kc and kd, is found valid only for small values of the equilibrium constant K (10,11,10,12 mol,·,L,1) and its validity is found to depend strongly of the value of K. On the other hand, the relaxed NQE approximation of Fischer and Fukuda, kc[P][N],=,kd[PN]0 was found to be remarkably good up to values of K around 10,8 mol,·,L,1. This upper bound is numerically found to be 2,3 orders of magnitude smaller than the theoretical one given by Fischer. The relaxed NQE is a better one due to the fact that it never completely neglects dN/dt. It is found that the difference between these approximations lies essentially in the number of significant figures taken for the approximation; still this subtle difference results in dramatic changes in the predicted course of the reaction. Some results confirm previous findings, but a deeper understanding of the physico-chemical phenomena and their mathematical representation and another viewpoint of the theory is offered. Additionally, experiments and simulations indicate that polymerization rate data alone are not reliable to estimate the value of K, as recently suggested. Validity of the rigorous nitroxide quasi-equilibrium assumption as a function of the nitroxide equilibrium constant. [source]


Structure-based prediction of modifications in glutarylamidase to allow single-step enzymatic production of 7-aminocephalosporanic acid from cephalosporin C

Abstract Glutarylamidase is an important enzyme employed in the commercial production of 7-aminocephalosporanic acid, a starting compound in the synthesis of cephalosporin antibiotics. 7-aminocephalosporanic acid is obtained from cephalosporin C, a natural antibiotic, either chemically or by a two-step enzymatic process utilizing the enzymes D-amino acid oxidase and glutarylamidase. We have investigated possibilities for redesigning glutarylamidase for the production of 7-aminocephalosporanic acid from cephalosporin C in a single enzymatic step. These studies are based on the structures of glutarylamidase, which we have solved with bound phosphate and ethylene glycol to 2.5 Å resolution and with bound glycerol to 2.4 Å. The phosphate binds near the catalytic serine in a way that mimics the hemiacetal that develops during catalysis, while the glycerol occupies the side-chain binding pocket. Our structures show that the enzyme is not only structurally similar to penicillin G acylase but also employs essentially the same mechanism in which the ,-amino group of the catalytic serine acts as a base. A subtle difference is the presence of two catalytic dyads, His B23/Glu B455 and His B23/Ser B1, that are not seen in penicillin G acylase. In contrast to classical serine proteases, the central histidine of these dyads interacts indirectly with the O, through a hydrogen bond relay network involving the ,-amino group of the serine and a bound water molecule. A plausible model of the enzyme,substrate complex is proposed that leads to the prediction of mutants of glutarylamidase that should enable the enzyme to deacylate cephalosporin C into 7-aminocephalosporanic acid. Brain responses to subject-verb agreement violations in spoken language in developmental dyslexia: an ERP study

Abstract This study investigates the presence and latency of the P600 component in response to subject,verb agreement violations in spoken language in people with and without developmental dyslexia. The two groups performed at-ceiling level on judging the sentences on their grammaticality, but the ERP data revealed subtle differences between them. The P600 tended to peak later in the left posterior region in the dyslexic group compared with the control group. In addition, the group of dyslexic subjects did not show a P600 in response to sentences with a plural NP subject. These results suggest that brain activation involved in syntactic repair is more affected by linguistic complexity in developmental dyslexia compared with non-dyslexic individuals. A catchment scale evaluation of the SIBERIA and CAESAR landscape evolution models

Abstract Landscape evolution models provide a way to determine erosion rates and landscape stability over times scales from tens to thousands of years. The SIBERIA and CAESAR landscape evolution models both have the capability to simulate catchment,wide erosion and deposition over these time scales. They are both cellular, operate over a digital elevation model of the landscape, and represent fluvial and slope processes. However, they were initially developed to solve research questions at different time and space scales and subsequently the perspective, detail and process representation vary considerably between the models. Notably, CAESAR simulates individual events with a greater emphasis on fluvial processes whereas SIBERIA averages erosion rates across annual time scales. This paper describes how both models are applied to Tin Camp Creek, Northern Territory, Australia, where soil erosion rates have been closely monitored over the last 10 years. Results simulating 10,000 years of erosion are similar, yet also pick up subtle differences that indicate the relative strengths and weaknesses of the two models. The results from both the SIBERIA and CAESAR models compare well with independent field data determined for the site over different time scales. Representative hillslope cross-sections are very similar between the models. Geomorphologically there was little difference between the modelled catchments after 1000 years but significant differences were revealed at longer simulation times. Importantly, both models show that they are sensitive to input parameters and that hydrology and erosion parameter derivation has long-term implications for sediment transport prediction. Therefore selection of input parameters is critical. This study also provides a good example of how different models may be better suited to different applications or research questions. The role of moisture cycling in the weathering of a quartz chlorite schist in a tropical environment: findings of a laboratory simulation

Abstract Long-term weathering of a quartz chlorite schist via wetting and drying was studied under a simulated tropical climate. Cubic rock samples (15 mm × 15 mm × 15 mm) were cut from larger rocks and subjected to time-compressed climatic conditions simulating the tropical wet season climate at the Ranger Uranium Mine in the Northern Territory, Australia. Fragmentation, moisture content and moisture uptake rate were monitored over 5000 cycles of wetting and drying. To determine the impact of climatic variables, five climatic regimes were simulated, varying water application, temperature and drying. One of the climatic regimes reproduced observed temperature and moisture variability at the Ranger Uranium Mine, but over a compressed time scale. It is shown that wetting and drying is capable of weathering quartz chlorite schist with changes expected over a real time period of decades. While wetting and drying alone does produce changes to rock morphology, the incorporation of temperature variation further enhances weathering rates. Although little fragmentation occurred in experiments, significant changes to internal pore structure were observed, which could potentially enhance other weathering mechanisms. Moisture variability is shown to lead to higher weathering rates than are observed when samples are subjected only to leaching. Finally, experiments were conducted on two rock samples from the same source having only subtle differences in mineralogy. The samples exhibited quite different weathering rates leading to the conclusion that our knowledge of the role of rock type and composition in weathering is insufficient for the accurate determination of weathering rates. Intraoperative transesophageal echocardiography has become an integral component of the repair of congenital heart defects. It currently has a direct impact on reducing morbidity and mortality in the pediatric cardiac population. To establish a successful program, it is important to follow guidelines for training as well as having a systematic approach to the evaluation of this patient population. This article addresses the specific indications in a patient population as practiced at the Hospital For Sick Children, Toronto. While there may be subtle differences between programs, the objectives are to provide excellent service to the pediatric cardiac patient in the operating room. [source]


Chromosomal Polymorphism in Korean Natural Populations of Drosophila immigrans

ABSTRACT To analyze chromosome inversions of Drosophila immigrans, wild flies were captured from large vineyards located in the suburbs of Yecheon and Gyeongsan from October 1999 to 2001. With the egg samples obtained singly at each of the 799 females of D. immigrans, cytological examinations were carried out for the type and frequency of inversions. Two types of different inversions were found only in the second chromosome. The inversions detected were known to be the cosmopolitan inversion "A" and "B". The mean frequency of inversion A was estimated to be 0.074 in Yecheon and 0.066 in Gyeongsan and that of B was to be 0.026 in Yecheon and 0.021 in Gyeongsan, respectively. In the frequency ratio, inversion A was significantly higher than that of B. The present populations of D. immigrans showed subtle differences from other Korean populations in inversion frequencies. To account for the local variations observed in inversion frequencies, several hypotheses are discussed such as founding event or selective force. [source]


A molecular assessment of the iron stress response in the two phylogenetic clades of Trichodesmium

Summary Trichodesmium spp. play key roles in global carbon and nitrogen budgets and thus defining what controls their productivity is important for understanding climate change. While iron availability has been shown to be an important chemical factor for controlling both growth and nitrogen fixation rates in Trichodesmium, all culture experiments to date have focused solely on representatives from one clade of Trichodesmium. Genomic sequence analysis determined that the Trichodesmium erythraeum (IMS101) genome contains many of the archetypical genes involved in the prokaryotic iron stress response. Focusing on three of these genes, isiB, idiA and feoB, we found that all three showed an iron stress response in axenic T. erythraeum (IMS101), and their sequences were well conserved across four species in our Trichodesmium culture collection [consisting of two T. erythraeum strains (IMS101 and GBRTRLI101), two Trichodesmium tenue strains (Z-1 and H9-4), Trichodesmium thiebautii and Trichodesmium spiralis]. With clade-specific quantitative PCR (qPCR) primers for one of these genes, isiB, we found that high isiB expression at low Fe levels corresponded to specific reductions in N2 fixation rates in both major phylogenetic clades of Trichodesmium (the T. erythraeum clade and T. tenue clade). With regard to the two clades, the most significant difference determined was temperature optima, while more subtle differences in growth, N2 fixation rate and gene expression responses to Fe stress were also observed. However the apparent conservation of the Fe stress response in the Trichodesmium genus suggests that it is an important adaptation for their niche in the oligotrophic ocean. [source]


Display Plasticity in Response to a Robotic Lizard: Signal Matching or Song Sharing in Lizards?

Many territorial songbirds alter the structure of their songs after listening to and interacting repeatedly with the same neighbors. Here, we use a robotic lizard to test for similar learned changes in signal structure in male Sagebrush lizards, Sceloporus graciosus. Subjects were exposed to two types of headbob displays (species-typical and unusual) both in short-term tests and in repeated exposures for 10 d. We found no evidence for immediate changes in signal structure to match a particular opponent (signal matching) or long-term changes after repeated exposure (,song' sharing). If anything, the lizards' displays became less like that of the robotic stimulus over time. Further tests of other taxa are needed to identify the evolutionary forces that lead to these forms of behavioral plasticity and to determine whether song sharing and signal matching are unique characteristic of songbirds. Lizards also became more agitated and produced more highly aggressive displays of their own when confronted with headbob displays that violated the basic syntactic structure of their display system, confirming that they were paying attention to subtle differences in display structure despite the artificial nature of the treatments. Thus, our study also adds to the growing evidence supporting the use of robotic playbacks to study animal communication. [source]


Studies on Nickel(II) Complexes with Amide-Based Ligands: Syntheses, Structures, Electrochemistry and Oxidation Chemistry

Abstract The present work discusses the nickel chemistry in a set of amide-based open-chain ligands with subtle differences in the backbone or terminal amine substituents. The ligands coordinate to the Ni2+ ion through the Namide and Namine atoms maintaining a square-planar geometry. Absorption spectra and NMR studies reveal that the solid-state square-planar geometry is retained in solution. The electrochemical results suggest that the NiIII/NiII redox couple primarily depends on the N4 donors, which is composed of two Namide and twoNamine atoms and not on the peripheral substituents. All four ligands with variable backbone and substituents are equally competent in stabilizing the NiIII state. On the basis of electrochemical findings, chemical oxidations were carried out, and they reveal generation of the NiIII state in two cases, whereas decomposition was observed in others. Preliminary alkene epoxidation reactions suggest that the present nickel complexes transiently stabilize the higher oxidation state of the nickel ion that possibly participates in the oxidation of the substrates.(© Wiley-VCH Verlag GmbH & Co. Methylone and mCPP, two new drugs of abuse?

Recently, two new ecstasy-like substances, methylone and mCPP, were found in street drugs in the Netherlands by the Drugs Information and Monitoring System (DIMS). Methylone (3,4-methylenedioxymethcathinone) is the main ingredient of a new liquid designer drug that appeared on the Dutch drug market, called ,Explosion'. mCPP (meta-chlorophenylpiperazine) is a substance often used as a probe for the serotonin function in psychiatric research, and has now been found in street drugs, both in tablets and powders. Methylone as well as mCPP act on monoaminergic systems, resembling MDMA (3,4-methylenedioxymethamphetamine), with mCPP mainly affecting the serotonin system. The subjective effects of both new substances exhibit subtle differences with those of MDMA. Only little is known about the harmfulness of both methylone and mCPP. However, because of similarities between these substances and MDMA, risks common to MDMA cannot be excluded. [source]


A comparison of different models of stroke on behaviour and brain morphology

Abstract We compared the effects of three models of permanent ischemia, as well as cortical aspiration, on behaviour and brain morphology. Rats received a stroke either by devascularization or by two different procedures of medial cerebral artery occlusion (MCAO; small vs. large). Animals were trained in a reaching task, forepaw asymmetry, forepaw inhibition, sunflower seed task and tongue extension. Behaviour was assessed 1 week after the lesion and at 2-week intervals for a total of 9 weeks. One week after the surgery all animals were severely impaired on all tasks and although they improved over time they only reached preoperative base lines on tongue extension. Animals with small MCAOs performed better in reaching and sunflower tasks; no other behavioural differences were detected among the groups. Pyramidal cells in forelimb and cingulate areas as well as spiny neurons of the striatum were examined for dendritic branching and spine density using a Golgi,Cox procedure. Each lesion type had a different impact on cell morphology. Overall, different changes (atrophy or hypertrophy) were observed with each kind of lesion and these changes were specific for the region (forelimb, cingulate, striatum) and the condition (intact vs. damaged hemisphere). These results suggest that: (i) different lesions to the motor cortex produce subtle differences in behaviour, and (ii) the method used to induce the lesion produces striking differences in cortical and subcortical plasticity. Abstract The preparation of several difluoroborondipyrromethene (Bodipy) dyads is described incorporating covalently attached hydroquinone/quinone groups at the 2-position (BD-SHQ, BD-SQ, BD-SPHQ, BD-SPQ). The compounds, currently under investigation as chemical sensors for reactive oxygen species, show various levels of fluorescence depending on the oxidation state of the appended group. The 19F NMR spectrum for BD-SHQ in CDCl3 at room temperature reveals the two fluorines are inequivalent on the NMR timescale. In contrast, the 19F NMR spectrum for the counterpart quinone compound, BD-SQ, is consistent with two equivalent fluorine atoms. The two results are interpreted as the quinone is free to rotate around the connector bond, whereas this motion is restricted for the hydroquinone group and makes the fluorines chemically inequivalent. Cyclic voltammograms recorded for all derivatives in CH2Cl2 electrolyte solution are consistent with typical Bodipy-based redox chemistry; the potentials of which depend on factors such as presence of the phenylene spacer and oxidation state of the appended group. A comparison of the electrochemical behaviour with the counterpart meso derivatives reveals some interesting trends which are associated with the location of the HOMO/LUMOs. The absorption profiles for the compounds in CH3CN are again consistent with Bodipy-based derivatives, though there are some subtle differences in the band-shapes of the closely-coupled systems. In particular, the absorption spectra for the dyad, BD-SQ, in a wide range of solvents are appreciably broader than for BD-SHQ. Femtosecond transient absorption spectroscopy performed on the hydroquinone derivatives, BD-SHQ and its meso analogue is interpreted as electron transfer occurs from the hydroquinone unit to the first-excited singlet (S1) state of the Bodipy center, followed by ultrafast charge recombination to reinstate the ground state. The coupling of OH vibrations to the return electron transfer process is invoked to explain the lack of clear identification of the charge-separated state in the transient records. [source]


The effect of linguistic abstraction on interpersonal distance

It is well known that people describe positive behaviors of others close to them (e.g., in-group member, friend) in abstract terms, but with concrete terms in the case of people who they are not close to (e.g., out-group member, enemy). In contrast, negative behaviors of people who we are close to are described in concrete terms, but in abstract terms for people who are distant. However, the communicative impact of such subtle differences in language use on a receiver who is also the actor of the behavior being described has never been addressed. We hypothesized and found that a positive abstract message compared to a positive concrete message leads to perceived proximity to the sender, while a negative abstract message compared to a negative concrete message leads to perceived distance. Communicating expectancies about others

The linguistic expectancy bias hypothesis predicts that, in general, person impressions are shared with others via subtle differences in the level of linguistic abstraction that is used to communicate expected and unexpected information about an individual. In a two-part communication experiment, we examined this hypothesis. In the first part of the experiment communicators were asked to provide a description of an event where a good friend had behaved in an expected or unexpected way. In the second part, recipients of these stories who were blind to the conditions under which the description was generated judged whether the story target's behavior was due to dispositional or situational factors. Behaviors in expected events were judged to be more dispositional relative to behaviors in unexpected events. As predicted, the level of linguistic abstraction mediated this effect. Characterization of Zebrafish Cx43.4 Connexin and its Channels

Connexins (Cx) form intercellular junctional channels which are responsible for metabolic and electrical coupling. We report here on the biochemical and immunohistochemical characterization of zebrafish connexin zfCx43.4, an orthologue of mammalian and avian Cx45, and the electrophysiological properties of junctional channels formed by this protein. The investigations were performed on transfected COS-7 cells or HeLa cells. Using site-directed antibodies, zfCx43.4 cDNA (GenBank accession no. X96712) was demonstrated to code for a protein with a Mr of 45 000. In transfected cells, zfCx43.4 was localized in cell-cell contact areas as expected for a gap junction protein. zfCx43.4 channels were shown to transfer Lucifer Yellow. The multichannel currents were sensitive to the transjunctional voltage (Vj). Their properties were consistent with a two-state model and yielded the following Boltzmann parameters for negative/positive Vj: Vj,0= -38.4/41.9 mV; gj,min= 0.19/0.18; z = 2.6/2.3. These parameters deviate somewhat from those of zfCx43.4 channels expressed in Xenopus oocytes and from those of Cx45, an orthologue of zfCx43.4, expressed in mammalian cells or Xenopus oocytes. Conceivably, the subtle differences may reflect differences in experimental methods and/or in the expression system. The single channel currents yielded two prominent levels attributable to a main conductance state (,j,main= 33.2 ± 1.5 pS) and a residual conductance state (,j,residual= 11.9 ± 0.6 pS). [source]


NO synthase isoforms specifically modify peroxynitrite reactivity

Nitric oxide synthases (NOSs) are multi-domain hemothiolate proteins that are the sole source of nitric oxide (NO) in mammals. NOSs can also be a source or a sink for peroxynitrite (PN), an oxidant that is suspected to be involved in numerous physiopathological processes. In a previous study, we showed that the oxygenase domain of the inducible NOS (iNOSoxy) reacts with PN and changes its oxidative reactivity [Maréchal A, Mattioli TA, Stuehr DJ & Santolini J (2007) J Biol Chem282, 14101,14112]. Here we report a similar analysis on two other NOS isoforms, neuronal NOS (nNOS) and a bacterial NOS-like protein (bsNOS). All NOSs accelerated PN decomposition, with accumulation of a similar heme intermediate. The kinetics of PN decomposition and heme transitions were comparable among NOSs. However, their effects on PN reactivity differ greatly. All isoforms suppressed PN two-electron oxidative activity, but iNOSoxy enhanced PN one-electron oxidation and nitration potencies, the oxygenase domain of nNOS (nNOSoxy) affected them minimally, and bsNOS abolished all PN reactivities. This led to the loss of both NOS and PN decomposition activities for nNOSoxy and iNOSoxy, which may be linked to the reported alterations in their electronic absorption spectra. Bacterial bsNOS was affected to a lesser extent by reaction with PN. We propose that these differences in PN reactivity among NOSs might arise from subtle differences in their heme pockets, and could reflect the physiological specificity of each NOS isoform, ranging from oxidative stress amplification (iNOS) to detoxification (bsNOS). The influence of predatory fish on mayfly drift: extrapolating from experiments to nature

1.,A knowledge of how individual behaviour affects populations in nature is needed to understand many ecologically important processes, such as the dispersal of larval insects in streams. The influence of chemical cues from drift-feeding fish on the drift dispersal of mayflies has been documented in small experimental channels (i.e. < 3 m), but their influence on dispersal in natural systems (e.g. 30 m stream reaches) is unclear. 2.,Using surveys in 10 Rocky Mountain streams in Western Colorado we examined whether the effects of predatory brook trout (Salvelinus fontinalis) on mayfly drift, that were apparent in stream-side channels, could also be detected in natural streams. 3.,In channel experiments, the drift of Baetis bicaudatus (Baetidae) was more responsive to variation in the concentration of chemical cues from brook trout than that of another mayfly, Epeorus deceptivus (Heptageniidae). The rate of brook trout predation on drifting mayflies of both species in a 2-m long observation tank was higher during the day (60,75%) but still measurable at night (5,10%). Epeorus individuals released into the water column were more vulnerable to trout predation by both day and night than were Baetis larvae treated similarly. 4.,Drift of all mayfly taxa in five fishless streams was aperiodic, whereas their drift was nocturnal in five trout streams. The propensity of mayflies to drift was decreased during the day and increased during the night in trout streams compared with fishless streams. In contrast to the channel experiments, fish biomass and density did not alter the nocturnal nature nor magnitude of mayfly drift in natural streams. 5.,In combination, these results indicate that mayflies respond to subtle differences in concentration of fish cues in experimental channels. However, temporal and spatial variation in fish cues available to mayflies in natural streams may have obscured our ability to detect responses at larger scales. [source]


Explaining isotope trophic-step fractionation: why herbivorous fish are different

Summary 1An assumed constant trophic fractionation of 15N/14N between consumer and diet (usually 3·4 for diet,muscle tissue differences) allows inferences to be made about feeding interactions and trophic level in food web studies. However, considerable variability surrounds this constant, which may conceal subtle differences about the trophodynamics of consumers. 2The feeding ecologies of herbivores and carnivores differ in terms of diet quality (in C : N terms) and food processing mechanisms, which may affect fractionation. 3We present a new model that explores how consumer feeding rates, excretion rates and diet quality determine the 15N/14N ratios in the consumer's tissues and hence influence the magnitude of trophic fractionation. 4Three herbivorous reef fish Acanthurus sohal, Zebrasoma xanthurum and Pomacentrus arabicus were chosen as study organisms. Empirical estimates of diet,tissue stable isotope fractionation were made in the field, and model parameters were derived from feeding observations and literature data. 5The trophic fractionation values of A. sohal, Z. xanthurum and P. arabicus were 4·69, 4·47 and 5·25, respectively, by empirical measurement, and 4·41, 4·30 and 5·68, respectively, by model, indicating that herbivores have a higher trophic fractionation than the currently accepted value of 3·4. 6The model was most sensitive to the excretion rate, which may differ between herbivores and carnivorous animals. This model is the first to determine stable isotope signatures of a consumer's diet mixture without applying a constant fractionation value. [source]


A Wet/Wet Differential Pressure Sensor for Measuring Vertical Hydraulic Gradient

Vertical hydraulic gradient is commonly measured in rivers, lakes, and streams for studies of groundwater,surface water interaction. While a number of methods with subtle differences have been applied, these methods can generally be separated into two categories; measuring surface water elevation and pressure in the subsurface separately or making direct measurements of the head difference with a manometer. Making separate head measurements allows for the use of electronic pressure sensors, providing large datasets that are particularly useful when the vertical hydraulic gradient fluctuates over time. On the other hand, using a manometer-based method provides an easier and more rapid measurement with a simpler computation to calculate the vertical hydraulic gradient. In this study, we evaluated a wet/wet differential pressure sensor for use in measuring vertical hydraulic gradient. This approach combines the advantage of high-temporal frequency measurements obtained with instrumented piezometers with the simplicity and reduced potential for human-induced error obtained with a manometer board method. Our results showed that the wet/wet differential pressure sensor provided results comparable to more traditional methods, making it an acceptable method for future use. [source]


,, T-cell lymphomas: a homogeneous entity?

,, T-cells comprise an immunologically distinct lymphoid population, characterized by specific morphological, phenotypical and functional properties. Therefore it seems reasonable to speculate that neoplasms derived from this particular T-cell subset display distinct features. Indeed, the prototype ,, T-cell lymphoma, hepatosplenic T-cell lymphoma constitutes a unique clinicopathological entitity which is intimately associated with a ,, T-cell phenotype. However, ,, T-cell lymphomas have also been described in other extranodal sites where, unlike reactive ,, T-cells and hepatosplenic ,, T-cell lymphomas, they display an important morphological heterogeneity. Moreover, these nonhepatosplenic ,, T-cell lymphomas are essentially not that different from their ,, T-cell receptor for antigen (TCR)-expressing counterparts and thus may be incorporated in the established T-cell lymphoma subclasses. However, subtle differences regarding their histopathological appearance as well as their biological behaviour indicate that further studies to determine the exact significance of TCR expression are required. Such inquiries may contribute to the general understanding of T-cell lymphomagenesis in general, which is still obscure. [source]


Pyogenic liver abscesses: a comparison of older and younger patients

Background Pyogenic liver abscess is a life-threatening disease. Few studies have specifically explored the way in which the clinical features and management of elderly patients with pyogenic liver abscess differ from those of younger individuals. Methods A retrospective study was undertaken to evaluate whether older patients with pyogenic liver abscess have distinctive presenting features or whether their management and outcome differ from that of younger patients. A total of 133 patients with liver abscess treated in five hospitals during 13 years comprised two groups: 78 patients aged 60 years or above (older group) and 55 patients below age 60 years (younger group). Clinical features, laboratory data, imaging and microbiological findings, management and outcome were determined in each group. Results The older group contained more patients with associated diseases (p = 0.03), nausea and vomiting at presentation (p = 0.02), higher APACHE II (Acute Physiological and Chronic Health Evaluation II scale) score (p < 0.001) and blood urea nitrogen (p < 0.001) and serum creatinine levels (p = 0.002). Multiple abscess (p = 0.05) and bilobar (p = 0.03) abscess were also commoner in this group. By contrast, in the younger group men predominated (p = 0.01), and there was a higher overall complication rate (p = 0.05). Time to diagnosis, hospital mortality rate and other variables analysed were similar in both groups. Discussion Elderly patients with pyogenic liver abscess have some subtle differences in clinical and laboratory presentation, but these do not appear to delay diagnosis. Active management is tolerated well, with a lower morbidity rate than in younger patients and no difference in the mortality rate. [source]


Ethnic skin types: are there differences in skin structure and function?,

Synopsis People of skin of colour comprise the majority of the world's population and Asian subjects comprise more than half of the total population of the earth. Even so, the literature on the characteristics of the subjects with skin of colour is limited. Several groups over the past decades have attempted to decipher the underlying differences in skin structure and function in different ethnic skin types. However, most of these studies have been of small scale and in some studies interindividual differences in skin quality overwhelm any racial differences. There has been a recent call for more studies to address genetic together with phenotypic differences among different racial groups and in this respect several large-scale studies have been conducted recently. The most obvious ethnic skin difference relates to skin colour which is dominated by the presence of melanin. The photoprotection derived from this polymer influences the rate of the skin aging changes between the different racial groups. However, all racial groups are eventually subjected to the photoaging process. Generally Caucasians have an earlier onset and greater skin wrinkling and sagging signs than other skin types and in general increased pigmentary problems are seen in skin of colour although one large study reported that East Asians living in the U.S.A. had the least pigment spots. Induction of a hyperpigmentary response is thought to be through signaling by the protease-activated receptor-2 which together with its activating protease is increased in the epidermis of subjects with skin of colour. Changes in skin biophysical properties with age demonstrate that the more darkly pigmented subjects retaining younger skin properties compared with the more lightly pigmented groups. However, despite having a more compact stratum corneum (SC) there are conflicting reports on barrier function in these subjects. Nevertheless, upon a chemical or mechanical challenge the SC barrier function is reported to be stronger in subjects with darker skin despite having the reported lowest ceramide levels. One has to remember that barrier function relates to the total architecture of the SC and not just its lipid levels. Asian skin is reported to possess a similar basal transepidermal water loss (TEWL) to Caucasian skin and similar ceramide levels but upon mechanical challenge it has the weakest barrier function. Differences in intercellular cohesion are obviously apparent. In contrast reduced SC natural moisturizing factor levels have been reported compared with Caucasian and African American skin. These differences will contribute to differences in desquamation but few data are available. One recent study has shown reduced epidermal Cathepsin L2 levels in darker skin types which if also occurs in the SC could contribute to the known skin ashing problems these subjects experience. In very general terms as the desquamatory enzymes are extruded with the lamellar granules subjects with lowered SC lipid levels are expected to have lowered desquamatory enzyme levels. Increased pores size, sebum secretion and skin surface microflora occur in Negroid subjects. Equally increased mast cell granule size occurs in these subjects. The frequency of skin sensitivity is quite similar across different racial groups but the stimuli for its induction shows subtle differences. Nevertheless, several studies indicate that Asian skin maybe more sensitive to exogenous chemicals probably due to a thinner SC and higher eccrine gland density. In conclusion, we know more of the biophysical and somatosensory characteristics of ethnic skin types but clearly, there is still more to learn and especially about the inherent underlying biological differences in ethnic skin types. Résumé, Les gens qui ont une peau de couleur représentent la majorité de la population mondiale et les sujets asiatiques en représentent plus de la moitié. Pourtant la littérature consacrée aux caractéristiques de ces sujets est limitée. Plusieurs groupes de travail ont essayé au cours des dernières années de comprendre les différences sous-jacentes de la structure et de la fonction de la peau de différentes ethnies. Maisla plupart de ces études ont été réalisées à petite échelle et dans certains cas les différences observées entre les individus au niveau de la qualité de la peau ne font pas ressortir de différence entre races. Récemment, un besoin d'études reliant les diffèrences génétiques et phénotypiques entre différents groupes raciaux s'est fait sentir et de ce fait beaucoup d'études à grande èchelle ont été entreprises. La différence la plus évidente, entre les peaux ethniques, est leur couleur liée à la présence de la mélanine. La photoprotection induite par ce polymère influence le taux de vieillissement de la peau entre les différents groupes raciaux qui finalement sont tous sujets au processus de photovieillissement. Généralement, les caucasiens ont des signes plus précoces et plus importants de formation de rides et de relâchement de la peau; en général, les problèmes d'augmentation de la pigmentation sont observés sur les peaux de couleur, bien qu'une grande étude ait rapporté que des sujets originaires de l'Asie de l'Est vivant aux U.S.A. avaient le moins de taches pigmentaires. On pense que la réponse d'une induction hyperpigmentaire est due à un signal envoyé par le récepteur 2 activé par une protéase. Le récepteur 2 augmente en même temps que la protéase activatrice dans l'épiderme des sujets ayant une peau de couleur. Les changements dans les propriètés biophysiques de la peau en fonction de l'âge montrent que les sujets qui ont la pigmentation la plus sombre gardent une peau plus jeune par comparaison aux groupes qui possèdent une pigmentation moins forte. Toutefois, bien qu'ayant un stratum corneum plus compact, il existe des rapports divergents sur la fonction barrière de ces sujets. Dans le cas d'agression chimique ou mécanique, la fonction barrière du stratum corneum est considérée plus forte chez les sujets à peau plus foncée, malgré leurs taux plus faibles encéramide. On doit garder à l'esprit que la fonction barrière du stratum corneum dépend de toute son architecture et pas seulement de sa teneur en lipides. On considère que la peau asiatique à unePIE (TEWL) basale similaire à la peau caucasienne, ainsi que des taux en céramides comparables, mais on constate que dans le cas d'agression mécanique, elle possède un effet barrière le plus faible. Des différences dans la cohésion intercellulaire sont évidentes. A contrario, on a mis en évidence des taux d'hydratation (NMF) plus faibles dans son stratum corneum, comparativement à la peau caucasienne et afro-américaine. Ces différences expliquent les variations au niveau de la desquamation, mais on a très peu de données sur ce sujet. Une étude récente a mis en évidence des taux réduits de Cathepsin L2 dans l'épiderme des types de peau plus sombre, ce qui, si cela se produisait dans le stratum corneum, expliquerait les problèmes biens connus de cendrage de la peau que ces sujets connaissent. En terme très gènéral, étant donné que les enzymes liées à la desquamation sont libérées avec les granules lamellaires, on s'attend à ce que les sujets ayant des taux de lipides faibles dans le stratum corneum aient des taux d'enzymes liés à la desquamation faibles. On constate chez les sujets noirs une augmentation de la taille des pores, de la sécrétion du sébum et de la microflore cutanée. On observe également chez ces sujets une augmentation de la taille des granules mastocellulaires. Le phénomène de peau sensible se retrouve à une fréquence similaire dans les différents groupes raciaux, mais il existe des différences subtiles dans lesstimuli nécessaires pour l'induire. En tout cas, plusieurs études montrent que la peau asiatique est peut-être plus sensible aux produits chimiques exogènes, ce qui probablement est dûà un stratum corneum plus mince et à une densité de glandes eccrines plus élevées. En conclusion, c'est sur les caractéristiques biophysiques et somato-sensorielles des différents types de peaux ethniques que nous en savons plus, mais il est clair qu'il nous reste à comprendre encore beaucoup de choses principalement sur leurs différences biologiques. Aroma, touch and well-being: following the mind to wellness

Fragrances have been used since ancient times to deliver a variety of benefits. The term aromatherapy was first coined in the 1920s by a French chemist named Rene-Maurice Gattefossé, referring specifically to the use of natural fragrance essential oils to treat injury and disease. More recently, the discovery that touch is an important part of healthy human development has led to a greater understanding of its role as an essential part of maintaining wellness. With the growing mainstream acceptance of holistic health care, herbal remedies and nutraceuticals, aromatherapy has become much more broadly defined to include benefits which are strictly cosmetic in nature. Today, a variety of products ranging from candles to dish-washing liquids are marketed with ,aromatherapy' benefits. Although there is no legal definition of aromatherapy in the U.S.A., specific claims can affect whether a product is considered a cosmetic or a pharmaceutical. Outside of the U.S.A., the term aromatherapy itself has very specific connotations which can affect the regulatory status of potential product introductions. In recent years, a number of new tools such as psychophysical measures and brain imaging techniques have greatly enhanced our knowledge of how touch and aromas are interpreted by the mind and body. This symposium will review the current state of our understanding in this area with the objective of providing a clearer understanding of the sometimes subtle differences of the role of fragrance and touch in maintaining well-being and enhancing personal attractiveness vs. those which are pharmacological in nature.
