Selected AbstractsIn vitro Leishmanicidal activity of naturally occurring chalconesPHYTOTHERAPY RESEARCH, Issue 2 2001Oliver Kayser Abstract A variety of chalcones have been shown to exhibit activity against Leishmania parasites. In contrast to synthetic or semisynthetic chalcones, only a few plant-derived compounds have been investigated. To provide a scientific rational for the antiprotozoal potency of plants used in ethnomedicine and containing chalcones, and in the search for new antiprotozoal drugs, we have carried out a primary screening for in vitro leishmanicidal activity of 20 chalcones isolated from plants. The compounds were tested against extracellular promastigotes of Leishmania donovani, L. infantum, L. enrietii and L. major, and against intracellular amastigote L. donovani residing within murine macrophages. Against the extracellular Leishmania (L. donovani), most compounds were active with EC50 values between 0.07 and 2.01,µg/mL. Some of these chalcones, 2,,4,-dihydroxy-4-methoxychalcone, 2,-hydroxy-3,4-dimethoxychalcone and 2-hydroxy-4,4,-dimethoxychalcone also significantly inhibited the intracellular survival of L. donovani parasites with EC50 values between 0.39 and 0.41,µg/mL. When tested against murine bone marrow-derived macrophages as a mammalian host cell control, all compounds with antileishmanial activities also proved to be cytotoxic to varying extents (EC50 0.19,2.06,µg/mL). Correlations between molecular structures and antileishmanial activity are discussed in detail. [source] Volatile Compounds of Raw Beef from 5 Local Spanish Cattle Breeds Stored Under Modified AtmosphereJOURNAL OF FOOD SCIENCE, Issue 4 2002K. Insausti ABSTRACT Volatile compounds of raw beef from Asturiana de los Valles, Morucha, Parda Alpina, Pirenaica, and Retinta cattle breeds were studied. Steaks were packaged under 60% O2, 30% CO2, and 10% N2, and 53 volatile compounds were tentatively identified by purge-and-trap extraction and gas chromatography with mass spectrometry after 0, 5, 10, and 15 d of chill storage. The degradation of beef quality with increasing storage time was evidenced by the increase in 2,3,3-trimethylpentane, 2,2,5-trimethylhexane, 3-octene, 3-methyl-2-heptene, 2-octene, and 2-propanone and by the decrease in dimethyl sulfide. Consequently, some of the volatile compounds, which are believed to be formed by thermal degradation of meat, might be formed during chill storage, and the rate of formation of some was dependent on the specific compound. [source] Screening of herbal extracts against multi-drug resistant Acinetobacter baumanniiPHYTOTHERAPY RESEARCH, Issue 8 2010Yoko Miyasaki Abstract Antibiotic resistance is increasing resulting in a decreasing number of fully active antimicrobial agents available to treat infections with multi-drug resistant (MDR) bacteria. Herbal medicines may offer alternative treatment options. A direct inoculation method simulating the standard disc diffusion assay was developed to determine in vitro antimicrobial activity of sixty herbal extracts against MDR- Acinetobacter baumannii (A. baumannii). Eighteen herbal extracts inhibited MDR- A. baumannii on agar plates, although the magnitude and quality of bacterial inhibition differed considerably among the antibacterial herbal extracts. Next, minimal inhibitory concentration (MIC) of these antibacterial herbal extracts was calculated using a broth microdilution assay. For most herbal extracts, the larger the zone of inhibition on agar plates, the lower the MIC. In general, hetero-resistance on agar plates correlated with higher MIC. The skip well phenomenon was seen with two herbal extracts. In conclusion, 30% of the screened herbal extracts demonstrated in vitro antibacterial activity against MDR- A. baumannii using similar rigorous testing methods as those commonly employed for assessing antimicrobial activity of synthetic antibacterial agents. Characterization of a specific compound conferring this antibacterial activity of the herbal extracts may help to identify novel antimicrobial agents active against highly resistant bacteria. Alekseeva Summary We investigated the influence of earthworm (Aporrectodea giardi) activity on soil properties and on atrazine (AT) adsorption and biodegradation by comparing a coarse-textured smectite-free wetland soil (Brittany, France) with the earthworm casts derived from the top horizon of this soil. Casts are characterized by lower pH, are enriched in organic carbon (OC) and clay content, have a larger cation exchange capacity, and a greater exchangeable Ca content. The clay mineralogy of the soil studied and casts is characterized by a muscovite,kaolinite,chlorite association. In addition, the clay fraction of the soil contains lepidocrocite (,-FeOOH), which was not found in the casts. Atrazine adsorption isotherms were reasonably well described by the Freundlich equation and were all non-linear. The mean amounts of adsorbed AT for starting concentrations of 3,30 mg litre,1 ranged from 8 to 34%, being largest in earthworm casts. Soil AT adsorption capacity was well correlated with OC content. Non-decomposed organic matter present in the coarse size fractions and specific compounds present in earthworm casts (proteins, mono- and polysaccharides, polyphenols, sugars, lignin) and microbial and fungal biomass contribute to AT adsorption. Weak electrostatic (physical) sorption of AT on organic compounds and on mineral surfaces prevails. For casts, the formation of additional hydrophobic interactions between AT and SOM is proposed. We also studied AT biodegradation by the model bacterium Pseudomonas sp. strain ADP in the presence of soils or earthworm casts. An enhancement of the AT disappearance rate was observed in the presence of all the solid matrices tested compared with that obtained in an aqueous medium. The biodegradation rate was shown to be dependent not only on the OC content of the solid matrix, but mainly on its composition and structure. Four types of receptors for L -glu have been described. This neurotransmitter modulates several neuronal processes, and its dysfunction causes chronic and acute diseases. L -Glu action is terminated by five distinct transporters. Antagonists for these receptors and modulators of these transporters have anticonvulsant and neuroprotective potentials, as observed with the acylpoliamines and peptides isolated from spiders, solitary and social wasp venoms. On the other hand, the major inhibitory neurotransmitter in mammalian nervous tissue is the GABA. Drugs that enhance GABA neurotransmission comprise effective approaches to protecting the brain against neuronal injury. Is this study, we demonstrate for the first time the inhibition of the [3H]L -glu binding to its specific sites in synaptosomal membranes from rat cerebral cortex, produced by 0.027 U of Paratemnus elongatus venom (EC50). The venom of P. elongatus changes Km and Vmax into the high affinity uptake of the L -glu and decreases Km and Vmax into the parameters of the GABA uptake from rat synaptosomes. This leads us to speculate on the possible presence of selective and specific compounds in this venom that act in L -glu and GABA dynamics, and therefore, that can serve as tools and new drug models for understanding these neurotransmissions. © 2006 Wiley Periodicals, Inc. BDNF exerts its action through interactions with two transmembrane receptors, either separately or in concert. BDNF has been implicated in several neurological disorders, and irregularities in BDNF function may have severe consequences. Administration of BDNF as a drug has thus far yielded few practicable results, and the potential side effects when using a multifunctional protein are substantial. In an effort to produce more specific compounds without side effects, small peptides mimicking protein function have been developed. The present study characterized two mimetic peptides, Betrofin 3 and Betrofin 4, derived from the BDNF sequence. Both Betrofins bound the cognate BDNF receptors, TrkB and p75NTR, and induced neurite outgrowth and enhanced neuronal survival, probably by inducing signaling through tha Akt and MAPK pathways. Distinct, charged residues within the Betrofin sequences were identified as important for generating the neuritogenic response, which was also inhibited when BDNF was added together with either Betrofin, indicating partial agonistic effects of the peptides. Thus, two peptides derived from BDNF induced neurite outgrowth and enhanced neuronal survival, probably through binding to BDNF receptors. © 2009 Wiley-Liss, Inc. [source] OILS FROM CENOZOIC RIFT-BASINS IN CENTRAL AND NORTHERN THAILAND: SOURCE AND THERMAL MATURITYJOURNAL OF PETROLEUM GEOLOGY, Issue 1 2007H.I. Petersen Oil is produced from the Suphan Buri, Phitsanulok and Fang Basins onshore central and northern Thailand. Most of the Cenozoic rift-basins onshore Thailand are 2,4 km deep, but the Phitsanulok Basin is the deepest with a basin-fill up to 8 km thick. In this basin, the Sirikit field produces ,18,000,24,000 bbl/day of crude oil. In the Suphan Buri Basin, about 400 bbl/day of crude oil is produced from the U Thong and Sang Kajai fields. Approximately 800 bbl/day of crude oil is produced from the Fang field (Fang Basin), which in reality consists of a number of minor structures including Ban Thi, Pong Nok, San Sai, Nong Yao and Mae Soon. A total of eight oil samples were collected from these structures and from the Sirikit, U Thong and Sang Kajai fields. The oils were subjected to MPLC and HPLC separation and were analysed by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS and GC-MS-MS). The U Thong oil was investigated in more detail by separating the oil into a number of fractions suited for the analysis of various specific compounds. The Sirikit oil appears to be the most mature, whereas the Suphan Buri oils and the oil from the San Sai structure (Fang Basin) are the least mature. Apart from the San Sai oil, the other oils in the Fang Basin are of similar maturity. The oils contain small amounts of asphaltenes and the asphaltene-free fractions are completely dominated by saturated hydrocarbons (generally >60%). Long-chain n-alkanes extend to at least C40 and the oils are thus highly waxy. In general the oils were generated from freshwater lacustrine source rocks containing a large proportion of algal material, as indicated by the presence of long-chain n-alkanes, low C3122R/C30 hopane ratios, the presence of 28-Nor-spergulane, T26/T25 (tricyclic triterpanes) ratios of 1.07,1.57 and tetracyclic polyprenoid (TTP) ratios close to 1. Occasional saline conditions may have occurred during deposition of the Sirikit, Ban Thi and Pong Nok source rocks. The Fang Basin oils were sourced from two different kitchens, one feeding the Ban Thi and Pong Nok structures and one feeding the Mae Soon, Nong Yao and San Sai structures. The presence ofcadalene, tetracyclic C24 compounds, oleanane, lupane, bicadinane and trace amounts ofnorpimarane or norisopimarane indicate a contribution from higher land plant organic matter to the oils. The terrestrial organic matter may occur disseminated in the lacustrine facies or concentrated in coal seams associated with the lacustrine mudstones. Thermally immature oil shales (lacustrine mudstones) and coals exposed in numerous basins in central and northern Thailand could upon maturation generate oils with a composition comparable to the investigated oils. [source] Semi-quantitative and structural metabolic phenotyping by direct infusion ion trap mass spectrometry and its application in genetical metabolomicsRAPID COMMUNICATIONS IN MASS SPECTROMETRY, Issue 15 2009Albert Koulman The identification of quantitative trait loci (QTL) for plant metabolites requires the quantitation of these metabolites across a large range of progeny. We developed a rapid metabolic profiling method using both untargeted and targeted direct infusion tandem mass spectrometry (DIMSMS) with a linear ion trap mass spectrometer yielding sufficient precision and accuracy for the quantification of a large number of metabolites in a high-throughput environment. The untargeted DIMSMS method uses top-down data-dependent fragmentation yielding MS2 and MS3 spectra. We have developed software tools to assess the structural homogeneity of the MS2 and MS3 spectra hence their utility for phenotyping and genetical metabolomics. In addition we used a targeted DIMS(MS) method for rapid quantitation of specific compounds. This method was compared with targeted LC/MS/MS methods for these compounds. The DIMSMS methods showed sufficient precision and accuracy for QTL discovery. We phenotyped 200 individual Loliumperenne genotypes from a mapping population harvested in two consecutive years. Computational and statistical analyses identified 246 nominal m/z bins with sufficient precision and homogeneity for QTL discovery. Comparison of the data for specific metabolites obtained by DIMSMS with the results from targeted LC/MS/MS analysis showed that quantitation by this metabolic profiling method is reasonably accurate. [source] High-resolution extracted ion chromatography, a new tool for metabolomics and lipidomics using a second-generation orbitrap mass spectrometerRAPID COMMUNICATIONS IN MASS SPECTROMETRY, Issue 10 2009Albert Koulman Most analytical methods in metabolomics are based on one of two strategies. The first strategy is aimed at specifically analysing a limited number of known metabolites or compound classes. Alternatively, an unbiased approach can be used for profiling as many features as possible in a given metabolome without prior knowledge of the identity of these features. Using high-resolution mass spectrometry with instruments capable of measuring m/z ratios with sufficiently low mass measurement uncertainties and simultaneous high scan speeds, it is possible to combine these two strategies, allowing unbiased profiling of biological samples and targeted analysis of specific compounds at the same time without compromises. Such high mass accuracy and mass resolving power reduces the number of candidate metabolites occupying the same retention time and m/z ratio space to a minimum. In this study, we demonstrate how targeted analysis of phospholipids as well as unbiased profiling is achievable using a benchtop orbitrap instrument after high-speed reversed-phase chromatography. [source] Floral scent of bat-pollinated species: West Africa vs. the New WorldBIOLOGICAL JOURNAL OF THE LINNEAN SOCIETY, Issue 2 2004STEFAN PETTERSSON Floral scent of seven West African bat-pollinated tree species, belonging to six families, was collected in situ from flowering individuals using headspace adsorption. The seven species shared neither any specific compounds nor any other discernible pattern in their floral scent composition. Most of the identified compounds are common in the floral scent of species pollinated by a variety of animals. Adansonia digitata (Bombacaceae) was the only African species found to have a substantial proportion of sulphur compounds in its floral scent. This feature contrasts with the sampled New World bat-pollinated plants, which frequently contain these compounds. The floral scent of Ceiba pentandra (Bombacaceae), native to both South America and Africa, contained no sulphur substances, contradicting a previous study in the New World that identified the major floral compounds as dimethyl disulphide and dimethyl trisulphide. We suggest that the differences in the floral scent of C. pentandra, including the absence of sulphur compounds in the African variety, result from the different selective regimes exerted by the Pteropotidae bats, in Africa, and Phyllostomidae bats, in the New World, that visit their flowers. © 2004 The Linnean Society of London, Biological Journal of the Linnean Society, 2004, 82, 161,168.
