Scaglione Abstract This paper describes an experimental method used to evaluate the anaerobic ammonium oxidation (Anammox) decay coefficient by means of a batch test. The test was carried out using an experimental procedure based on manometric measurements of the dinitrogen gas that is produced by the Anammox process. The accuracy of the procedure had previously been assessed, and the method was used to determine the specific Anammox activity (SAA mg N2 -N g VSS,1 d,1,) and the maximum nitrogen production rate (MNPR, NmL N2 L,1 d,1) under several different conditions. A specific batch test, which lasted for 148 days, was performed to assess the decay coefficient. The activity decrease was monitored and the estimated value of the decay coefficient was found to be 0.0048 d,1 at 35 °C, for which the corresponding half-life time of the Anammox biomass was 145 days. This value is higher than other values reported in the literature, but in accordance with the slow growth rate of the Anammox bacteria. Copyright © 2009 Society of Chemical Industry [source] Deciphering regulatory mechanisms for secondary metabolite production in the myxobacterium Sorangium cellulosum So ce56MOLECULAR MICROBIOLOGY, Issue 6 2007Shwan Rachid Summary Sorangium cellulosum strains produce approximately 50% of the biologically active secondary metabolites known from myxobacteria. These metabolites include several compounds of biotechnological importance such as the epothilones and chivosazols, which, respectively, stabilize the tubulin and actin skeletons of eukaryotic cells. S. cellulosum is characterized by its slow growth rate, and natural products are typically produced in low yield. In this study, biomagnetic bead separation of promoter-binding proteins and subsequent inactivation experiments were employed to identify the chivosazol regulator, ChiR, as a positive regulator of chivosazol biosynthesis in the genome-sequenced strain So ce56. Overexpression of chiR under the control of T7A1 promoter in a merodiploid mutant resulted in fivefold overproduction of chivosazol in a kinetic shake flask experiment, and 2.5-fold overproduction by fermentation. Using quantitative reverse transcription PCR and gel shift experiments employing heterologously expressed ChiR, we have shown that transcription of the chivosazol biosynthetic genes (chiA,chiF) is directly controlled by this protein. In addition, we have demonstrated that ChiR serves as a pleiotropic regulator in S. cellulosum, because mutant strains lack the ability to develop into regular fruiting bodies. [source] Segregation of the Escherichia coli chromosome terminusMOLECULAR MICROBIOLOGY, Issue 3 2003Yongfang Li Summary We studied the segregation of the replication terminus of the Escherichia coli chromosome by time-lapse and still photomicroscopy. The replicated termini lie together at the cell centre. They rapidly segregate away from each other immediately before cell division. At fast growth rate, the copies move progressively and quickly toward the centres of the new-born cells. At slow growth rate, the termini usually remain near the inner cell pole and migrate to the cell centre in the middle of the cell cycle. A terminus domain of about 160kb, roughly centred on the dif recombination site, segregated as a unit at cell division. Sequences outside this domain segregated before division, giving two separate foci in predivision cells. Resolution of chromosome dimers via the terminus dif site requires the XerC recombinase and an activity of the FtsK protein that is thought to align the dif sequences at the cell centre. We found that anchoring of the termini at the cell centre and proper segregation at cell division occurred normally in the absence of recombination via the XerC recombinase. Anchoring and proper segregation were, however, frequently disrupted when the C-terminal domain of FtsK was truncated. [source] Multiwavelength study of the nuclei of a volume-limited sample of galaxies , II.MONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 4 2007Optical, infrared, radio observations ABSTRACT We present optical and infrared broad-band images, radio maps, and optical spectroscopy for the nuclear region of a sample of nearby galaxies. The galaxies have been drawn from a complete volume-limited sample for which we have already presented X-ray imaging. We modelled the stellar component of the spectroscopic observations to determine the star formation history of our targets. Diagnostic diagrams were used to classify the emission-line spectra and determine the ionizing mechanism driving the nuclear regions. All those sources classified as active galactic nuclei present small Eddington ratios (,10,3,10,6), implying a very slow growth rate of their black holes. We finally investigate the relative numbers of active and normal nuclei as a function of host galaxy luminosity and find that the fraction of active galaxies slowly rises as a function of host absolute magnitude in the MB,,12 to ,22 range. [source] Global lopsided instability in a purely stellar galactic discMONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 1 2007Kanak Saha ABSTRACT It is shown that pure exponential discs in spiral galaxies are capable of supporting slowly varying discrete global lopsided modes, which can explain the observed features of lopsidedness in the stellar discs. Using linearized fluid dynamical equations with the softened self-gravity and pressure of the perturbation as the collective effect, we derive self-consistently a quadratic eigenvalue equation for the lopsided perturbation in the galactic disc. On solving this, we find that the ground-state mode shows the observed characteristics of the lopsidedness in a galactic disc, namely the fractional Fourier amplitude A1, increases smoothly with the radius. These lopsided patterns precess in the disc with a very slow pattern speed with no preferred sense of precession. We show that the lopsided modes in the stellar disc are long-lived because of a substantial reduction (approximately a factor of 10 compared to the local free precession rate) in the differential precession. The numerical solution of the equations shows that the ground-state lopsided modes are either very slowly precessing stationary normal mode oscillations of the disc or growing modes with a slow growth rate depending on the relative importance of the collective effect of the self-gravity. N -body simulations are performed to test the spontaneous growth of lopsidedness in a pure stellar disc. Both approaches are then compared and interpreted in terms of long-lived global m= 1 instabilities, with almost zero pattern speed. [source] Epithelioid srcoma of the penis: A case report and review of the literaturePATHOLOGY INTERNATIONAL, Issue 7 2000Giulio Rossi A rare case of epithelioid sarcoma (ES) of the penis is presented. The patient, a 35-year-old man, was initially treated as having Peyronie's disease, but the subsequent appearance of a subcutaneous nodule displayed a ,distal-type' ES. At immunohistochemical phenotypification, the tumor was positive for vimentin, cytokeratins and epithelial membrane antigen (EMA), as well as for some other multidirectional antibodies, including a membranous reaction for CD99. The review of 11 similar cases so far reported in the literature led to the conclusion that the clinicopathological characteristics of penile ES are basically the same as those of tumors in more classical locations: the age at diagnosis ranged from 23 to 43 years, the interval between first manifestations and diagnosis averaged 37 months (delayed diagnosis being common because of the slow growth rate and the harmless appearance of the lesion), the typical sign was a superficial nodule or mass, usually accompanied (better preceded) by urethral stenosis, dysuria and erectile disturbances. Total or partial penectomy was the treatment of choice in most patients, but lack of adequate follow up did not permit any definitive conclusion to be reached regarding its efficacy. [source] A Newborn with Nodular Ulcerated Lesion on a Giant Congenital NevusPEDIATRIC DERMATOLOGY, Issue 4 2000J. Borbujo M.D. It is usually small (less than 5 mm), sometimes multiple, with a slow growth rate, and has a black or dark brown, smooth, shiny surface. It usually involutes spontaneously. We report a newborn infant who, at birth, had a giant congenital nevus with a nodular, ulcerative, hemorrhagic lesion within it. Physical and neurologic examinations were normal. Radiologic studies at birth and subsequently were normal. A fragment of the lesion was biopsied and histologic findings were compatible with a diagnosis of proliferative nodule in a giant congenital nevus. The rest of the nodule regressed spontaneously after 4 months. Barker Abstract 1.Over the past two decades the number of fisheries targeting shark resources has increased dramatically. A combination of factors, including relatively slow growth rate, low fecundity and late age of maturity, result in low recovery rates from exploitation for most shark species. This, in turn, is reflected in the poor record of sustainability of shark fisheries. 2.One of the greatest challenges is to find a way to deal with the substantial levels of shark bycatch taken in many non-target fisheries. Poor general recording of shark landings and paucity of shark landing data at the species level also undermine the development of effective shark management strategies.3. This paper reviews the problems that must be faced worldwide if shark resources are to be managed sustainably and lays out a comprehensive set of prioritized management strategies to facilitate the sustainable management of global shark fisheries. It is acknowledged that the majority of sharks are harvested in developing countries and that the management of shark resources in developing and developed countries will need to incorporate different management strategies relevant to local socio-economic agendas. The management recommendations deal with methods to improve the global regulation of fisheries, ways to improve global conservation ethics and encourage active participation in management, as well as means by which specific management strategies may be implemented. MYERS Summary 1To survive in forest understoreys, seedlings must depend on carbohydrate reserves when they experience negative carbon balance imposed by occasional light reduction and tissue loss to herbivores and diseases. We present the first experimental evidence in support of this hypothesis, using seven woody neotropical species. 2We transplanted seedlings that had recently expanded their first photosynthetic cotyledon or leaf to the forest understorey (1% of full sun) and quantified initial biomass and total non-structural carbohydrate (TNC) in stems, roots and storage cotyledons. We then randomly assigned seedlings to control and two stress treatments: light reduction (0.08% of full sun for 8 weeks) and complete defoliation. 3First-year survival of control seedlings, a comparative measure of shade tolerance, differed widely among species. The two stress treatments reduced survival and relative growth rates (RGR) of all species. Shade-tolerant species were little impacted by the stress treatments, whereas the two least shade-tolerant species experienced 100% mortality. 4In all treatments, 8-week and first-year survival was positively correlated with initial TNC pool size in stems and roots. By contrast, survival was generally not correlated with initial TNC concentration in any organ, TNC pools in cotyledons, seed mass or seedling biomass. 5TNC in stems and roots, but not in cotyledons, decreased in response to light reduction and defoliation over 8 weeks. Leaf area recovery of defoliated seedlings was positively correlated with initial TNC pools in stems and roots. 6First-year survival in each treatment was negatively correlated with 0,8 week RGR of control seedlings, suggesting higher stress tolerance of species with inherently slow growth rates in shade. RGR of control seedlings from 0 to 8 weeks was negatively correlated with initial TNC pools, but not concentrations, in stems and roots. After 8 weeks, RGR was positive for all species, without clear relationships with survival or TNC. 7We conclude that carbohydrate storage in stems and roots enhances long-term survival in shade by enabling seedlings to cope with periods of biotic and abiotic stress. Carbohydrate storage is a key functional trait that can explain species differences in growth and survival that lead to species coexistence through niche assembly processes and life-history trade-offs. [source] Snow metamorphism as revealed by scanning electron microscopyMICROSCOPY RESEARCH AND TECHNIQUE, Issue 1 2003Florent Dominé Abstract Current theories of snow metamorphism indicate that sublimating snow crystals have rounded shapes, while growing crystals have shapes that depend on growth rates. At slow growth rates, crystals are rounded. At moderate rates, they have flat faces with rounded edges. At fast growth rates, crystals have flat faces with sharp edges, and they have hollow faces at very fast growth rates. The main growth/sublimation mechanism is thought to be by the homogeneous nucleation of new layers at or near crystal edges. It was also suggested that the equilibrium shape of snow crystals would be temperature dependent: rounded above ,10.5°C, and faceted below. To test these paradigms, we have performed SEM investigations of snow samples having undergone metamorphism under natural conditions, and of snow samples subjected to isothermal metamorphism at ,4° and ,15°C in the laboratory. In general, current theories predicting crystal shapes as a function of growth rates, and of whether crystals are growing or sublimating, are verified. However, the transition in equilibrium shapes from rounded to faceted at ,10.5°C is not observed in our isothermal experiments that reveal a predominance of rounded shapes after more than a month of metamorphism at ,4 and ,15°C. Some small crystals with flat faces that also have sharp angles at ,15°C, are observed in our isothermal experiments. These faces are newly formed, and contradict current theory. [source] The effect of soil on the growth performance of tropical species with contrasting distributionsOIKOS, Issue 10 2008Tania Brenes-Arguedas Within the tropics, a marked gradient in rainfall between dry and wet forests correlates with a well documented turnover of plant species. While water availability along these gradients is an important determinant of species distributions, other abiotic and biotic factors correlate with rainfall and may also contribute to limit species distribution. One of these is soil fertility, which is often lower in the wetter forests. To test its possible role in species distribution along a rainfall gradient, we performed a screen-house experiment where we measured the growth performance of seedlings of 23 species with contrasting distributions across the Isthmus of Panama. We grew seedlings in soils collected from the drier Pacific side and the wetter Atlantic side. Differences in soil fertility across the Isthmus were large enough to significantly influence the growth performance of the seedlings. However, we found no evidence of home-soil advantage among species with contrasting distributions. Dry-distribution species grew on average slower than wet-distribution species suggesting a cost to drought adaptations. The response to soil differences correlated with the growth rate of the species, such that fast-growing species responded more to changes in soil quality. We hypothesize that inherently slow growth rates of some dry-distribution tropical species may be a more important factor limiting their colonization of wetter sites along the rainfall gradient.
