Selected AbstractsThe lateral intercellular space as osmotic coupling compartment in isotonic transportACTA PHYSIOLOGICA, Issue 1 2009E. H. Larsen Abstract Solute-coupled water transport and isotonic transport are basic functions of low- and high-resistance epithelia. These functions are studied with the epithelium bathed on the two sides with physiological saline of similar composition. Hence, at transepithelial equilibrium water enters the epithelial cells from both sides, and with the reflection coefficient of tight junction being larger than that of the interspace basement membrane, all of the water leaves the epithelium through the interspace basement membrane. The common design of transporting epithelia leads to the theory that an osmotic coupling of water absorption to ion flow is energized by lateral Na+/K+ pumps. We show that the theory accounts quantitatively for steady- and time dependent states of solute-coupled fluid uptake by toad skin epithelium. Our experimental results exclude definitively three alternative theories of epithelial solute,water coupling: stoichiometric coupling at the molecular level by transport proteins like SGLT1, electro-osmosis and a ,junctional fluid transfer mechanism'. Convection-diffusion out of the lateral space constitutes the fundamental problem of isotonic transport by making the emerging fluid hypertonic relative to the fluid in the lateral intercellular space. In the Na+ recirculation theory the ,surplus of solutes' is returned to the lateral space via the cells energized by the lateral Na+/K+ pumps. We show that this theory accounts quantitatively for isotonic and hypotonic transport at transepithelial osmotic equilibrium as observed in toad skin epithelium in vitro. Our conclusions are further developed for discussing their application to solute,solvent coupling in other vertebrate epithelia such as small intestine, proximal tubule of glomerular kidney and gallbladder. Evidence is discussed that the Na+ recirculation theory is not irreconcilable with the wide range of metabolic cost of Na+ transport observed in fluid-transporting epithelia. [source] Amino acid sources in the adult diet do not affect life span and fecundity in the fruit-feeding butterfly Bicyclus anynanaECOLOGICAL ENTOMOLOGY, Issue 4 2008FREERK MOLLEMAN Abstract 1.,In tropical forests, the adults of many butterfly species feed on fruits rather than nectar from flowers and have long life spans. Rotting fruit and nectar differ from each other in many respects, including sources of amino acids and microbial life. If amino acids in the adult diet can be used for reproduction, this may have facilitated the evolution of extended life spans in this guild. 2.,This issue was addressed by investigating effects of banana, yeast, and amino acids in the adult diet of the fruit-feeding butterfly Bicyclus anynana (Lepidoptera) on longevity and female reproductive output in two experiments. 3.,Results showed that in the fruit-feeding butterfly B. anynana: (i) banana juice, but not sliced banana or added amino acids extend life span compared with a sugar solution of similar composition; (ii) compared with this sugar solution, other cohorts (banana juice-amino acid enriched) did not have significantly higher reproductive outputs; (iii) yeast does not represent a valuable source of nutrients; (iv) caloric restriction may cause decreased life span and rate of reproduction; and (v) increased rates of reproduction have a life span cost. [source] Liveweight gains of lambs from Caucasian clover/ryegrass and white clover/ryegrass swards on soils of high and low fertilityGRASS & FORAGE SCIENCE, Issue 2 2007A. D. Black Summary The high nutritive value and persistence under a wide range of climatic and soil fertility conditions make Caucasian clover a potentially useful forage legume but there is little information about the performance of livestock grazing Caucasian clover/grass swards. This study compared liveweight gains of lambs grazing Caucasian clover/perennial ryegrass and white clover/perennial ryegrass swards on high fertility (Olsen P 20 mg L,1, SO4 -S 12 mg kg,1) and low fertility (Olsen P 11 mg L,1, SO4 -S 7 mg kg,1) soils from 1998 to 2001 in the South Island of New Zealand. Mean annual liveweight gains were 1178 kg ha,1 for Caucasian clover/perennial ryegrass and 1069 kg ha,1 for white clover/perennial ryegrass swards at high fertility compared with 1094 kg ha,1 and 1015 kg ha,1, respectively, at low fertility. There was a higher mean proportion of clover in Caucasian clover/perennial ryegrass (0·19) than white clover/perennial ryegrass (0·11) swards, but there were no differences in total herbage production between the two clover/perennial ryegrass swards. The mean concentration of crude protein in the herbage of Caucasian clover (302 g kg DM,1) was higher than that in white clover (287 g kg DM,1) and grass herbage (227 g kg DM,1). Estimated mean metabolizable energy concentrations in the herbage were 12·5 MJ kg DM,1 for the two clovers and 11·6 MJ kg DM,1 for grass herbage. The difference in liveweight gain between swards on soils of high and low fertility was associated with an increase in total herbage production of similar composition and nutritive value, giving a greater number of grazing days for the swards on soils of high than low fertility. [source] Detecting vegetable oil adulteration in hazelnut paste (Corylus avellana L.)INTERNATIONAL JOURNAL OF FOOD SCIENCE & TECHNOLOGY, Issue 3 2009Josep Serra Bonvehi Summary The possibilities of detecting hazelnut paste adulterated with refined and non-refined vegetable oils have been studied. Research was focussed mainly on peanut, high oleic-acid sunflower, corn and soybean oils which have a similar composition to hazelnut oil. The analytical procedures to detect fatty acid (FA), triacylglycerol (TAG) and tocopherol profiles as indicators of adulteration were determined. The better indicators experimentally determined were seven FA (palmitic, stearic, linoleic, linolenic, arachidonic, behenic and lignoceric acids) and different TAG with three unsaturated FA (the code letters used for FA are: P = C16:0; S = C18:0; O = C18:1; L = C18:2;; Ln = C18:3) (LLLn, LLL and OOO), two unsaturated FA (POL, PLL and SOO), and one unsaturated FA (PPL). As expected, when refined vegetable oils were added to hazelnut paste, the increment of stigmasta-3,5-diene allowed detection at levels of 2% oil added. Limits of detection were measured using standard and adulterated hazelnut with different amounts of non-refined vegetable oils added (5%, 10%, 20% and 30%). The distribution of tocopherols and tocotrienols is highly useful, except in the case of added sunflower oil. The differences between the experimental and theoretical values of the TAG with equivalent carbon number (,ECN) of 42 does not improve the detection limit of hazelnut paste adulterated with peanut or sunflower oils. Similarly, tocopherols usually added to refined vegetable oils as an antioxidant were also determined. [source] CHARACTERISTICS OF MUSCLE FROM TWO SPECIES OF BIGEYE SNAPPER, PRIACANTHUS TAYENUS AND PRIACANTHUS MACRACANTHUSJOURNAL OF FOOD BIOCHEMISTRY, Issue 4 2002SOOTTWAT BENJAKUL ABSTRACT Composition and some properties of muscle from two species of bigeye snapper, P. tayenus and P. macracanthus, were investigated. Both species had a similar composition with the same myofibrillar protein content. However, muscle proteins from P. tayenus had higher thermal stability than those from P. macracanthus, as indicated by the higher enthalpy for transitions as well as the lower inactivation rate constant (KD). Upon 15 days of iced storage, natural actomyosin Ca2* -ATP ase and Mg2+ -Ca2+ -ATPase activities decreased, whereas Mg2+ -EGTA-ATPase activity increased, suggesting the denaturation of myosin, actomyosin and troponin/tropomyosin complexes, respectively. Increased surface hydrophobicity and decreased sulfhydryl groups indicated the denaturation possibly occurred via hydrophobic interaction and disulfide formation. Heading and eviscerating offish retarded the denaturation and physicochemical changes of proteins during iced storage. The results indicated that a rapid and proper post harvest handling was of importance to maintain the muscle quality of bigeye snapper. [source] Gas chromatography,mass spectrometry analysis of endogenous cannabinoids in healthy and tumoral human brain and human cells in cultureJOURNAL OF NEUROCHEMISTRY, Issue 2 2001Mauro Maccarrone Endocannabinoids are lipid mediators thought to modulate central and peripheral neural functions. We report here gas chromatography,electron impact mass spectrometry analysis of human brain, showing that lipid extracts contain anandamide and 2-arachidonoylglycerol (2-AG), the most active endocannabinoids known to date. Human brain also contained the endocannabinoid-like compounds N -oleoylethanolamine, N -palmitoylethanolamine and N -stearoylethanolamine. Anandamide and 2-AG (0.16 ± 0.05 and 0.10 ± 0.05 nmol/mg protein, respectively) represented 7.7% and 4.8% of total endocannabinoid-like compounds, respectively. N -Palmitoyethanolamine was the most abundant (50%), followed by N -oleoyl (23.6%) and N -stearoyl (13.9%) ethanolamines. A similar composition in endocannabinoid-like compounds was found in human neuroblastoma CHP100 and lymphoma U937 cells, and also in rat brain. Remarkably, human meningioma specimens showed an approximately six-fold smaller content of all N -acylethanolamines, but not of 2-AG, and a similar decrease was observed in a human glioblastoma. These ex vivo results fully support the purported roles of endocannabinoids in the nervous system. [source] Evaluation of Brewer's Waste as Partial Replacement of Fish Meal Protein in Nile Tilapia, Oreochromis niloticus, DietsJOURNAL OF THE WORLD AQUACULTURE SOCIETY, Issue 4 2008Desale B. Zerai A 10-wk feeding trial experiment involving five different diets with increasing levels of brewer's waste (32% crude protein) was carried out to evaluate the use of brewer's waste in tilapia diets in place of fish meal. Growth performance was compared against a control diet formulated to have similar composition to a typical commercial diet. Four experimental diets replaced successively 25, 50, 75, and 100% of the fish meal protein with brewer's waste. The diets were isonitrogenous and isocaloric. Results indicated that weight gain did not differ significantly (P > 0.05) with up to 50% replacement. Feed intake and utilization were depressed at high levels of brewer's waste. In addition, methionine of high replacement level diets was low. The results of the digestibility trial demonstrated that the brewer's waste used in this study has an apparent digestibility coefficient for protein of 70%. It was concluded that 50% of the fish meal protein in a typical commercial diet could be replaced with brewer's waste with no adverse effect on growth and feed utilization for tilapia. [source] The clay mineralogy of sediments related to the marine Mjølnir impact craterMETEORITICS & PLANETARY SCIENCE, Issue 10 2003Henning DYPVIK It was formed about 142 ± 2.6 Myr ago by the impact of a 1,2 km asteroid into the shallow shelf clays of the Hekkingen Formation and the underlying Triassic to Jurassic sedimentary strata. A core recovered from the central high within the crater contains slump and avalanche deposits from the collapse of the transient crater and central high. These beds are overlain by gravity flow conglomerates, with laminated shales and marls on top. Here, impact and post-impact deposits in this core are studied with focus on clay mineralogy obtained from XRD decomposition and simulation analysis methods. The clay-sized fractions are dominated by kaolinite, illite, mixed-layered clay minerals and quartz. Detailed analyses showed rather similar composition throughout the core, but some noticeable differences were detected, including varying crystal size of kaolinite and different types of illites and illite/smectite. These minerals may have been formed by diagenetic changes in the more porous/fractured beds in the crater compared to time-equivalent beds outside the crater rim. Long-term post-impact changes in clay mineralogy are assumed to have been minor, due to the shallow burial depth and minor thermal influence from impact-heated target rocks. Instead, the clay mineral assemblages, especially the abundance of chlorite, reflect the impact and post-impact reworking of older material. Previously, an ejecta layer (the Sindre Bed) was recognized in a nearby well outside the crater, represented by an increase in smectite-rich clay minerals, genetically equivalent to the smectite occurring in proximal ejecta deposits of the Chicxulub crater. Such alteration products from impact glasses were not detected in this study, indicating that little, if any, impact glass was deposited within the upper part of the crater fill. Crater-fill deposits inherited their mineral composition from Triassic and Jurassic sediments underlying the impact site. Nutrient provisioning in aphids depends both on the composition of ingested phloem sap and on the biosynthetic capabilities of the aphid and its intracellular symbionts. Amino acid budgets for three aphid species, Rhopalosiphum padi (L.), Schizaphis graminum (Rondani) and Diuraphis noxia (Mordvilko), were compared on a single host plant species, wheat Triticum aestivum L. Ingestion of amino acids from phloem, elimination of amino acids in honeydew, and the content of amino acids in aphids tissue were measured. From these values, ingestion rates were estimated and compared to honeydew and to estimated composition of aphid proteins. Ingestion rate was lowest in D. noxia due to low growth rate and low honeydew production; intermediate in S. graminum due to higher growth rate and intermediate honeydew production; and highest in R. padi, which had the highest rates for both variables. Both D. noxia and S. graminum induced increases in the amino acid content of ingested phloem. These changes in phloem content, combined with differences in ingestion rates, resulted in large differences among aphids in estimated rates of ingestion of individual amino acids. In honeydew, most essential amino acids were found in low amounts compared with the amounts ingested, especially for methionine and lysine. A few amino acids (arginine, cystine, histidine and tryptophan) were more abundant in honeydew of some aphids, suggesting oversupply. Aphid species differed in the composition of free amino acids in tissue but showed very similar composition in protein, implying similar requirements among the aphids. In R. padi and D. noxia, most essential amino acids were ingested in amounts insufficient for growth, implying dependence on symbiont provisioning. In S. graminum, most amino acids were ingested in amounts apparently sufficient for growth. [source] Structure of reactively extruded rigid PVC/PMMA blendsPOLYMERS FOR ADVANCED TECHNOLOGIES, Issue 6 2005Y. Haba Abstract A novel route for producing polymer blends by reactive extrusion is described, starting from poly (vinyl chloride)/methyl methacrylate (PVC/MMA) dry blend and successive polymerization of MMA in an extruder. Small angle X-ray scattering (SAXS) measurements were applied to study the monomer's mode of penetration into the PVC particles and to characterize the supermolecular structure of the reactive poly(vinyl chloride)/poly(methyl methacrylate) (PVC/PMMA) blends obtained, as compared to the corresponding physical blends of similar composition. These measurements indicate that the monomer molecules can easily penetrate into the PVC sub-primary particles, separating the PVC chains. Moreover, the increased mobility of the PVC chains enables formation of an ordered lamellar structure, with an average d -spacing of 4.1,nm. The same characteristic lamellar structure is further detected upon compression molding or extrusion of PVC and PVC/PMMA blends. In this case the mobility of the PVC chains is enabled through thermal energy. Dynamic mechanical thermal analysis (DMTA) and SAXS measurements of reactive and physical PVC/PMMA blends indicate that miscibility occurs between the PVC and PMMA chains. The studied reactive PVC/PMMA blends are found to be miscible, while the physical PVC/PMMA blends are only partially miscible. It can be suggested that the miscible PMMA chains weaken dipole,dipole interactions between the PVC chains, leading to high mobility and resulting in an increased PVC crystallinity degree and decreased PVC glass transition temperature (Tg). [source] PVC modification through polymerization of a monomer absorbed in porous suspension-type PVC particlesJOURNAL OF VINYL & ADDITIVE TECHNOLOGY, Issue 3 2004M. Narkis In-situ polymerization is the polymerization of one monomer in the presence of another polymer. It can be performed by sequential emulsion polymerization, or by reactions in the melt, in the solid phase, or in solution. The current report describes two methods to obtain poly(vinyl chloride) (PVC) modification through polymerization of a monomer absorbed in commercial porous suspension-type PVC particles. The generated modified PVC products differ significantly in their structure and properties. The first approach includes absorption of a monomer/peroxide solution within porous suspension-type PVC particles, followed by polymerization/crosslinking in the solid state at 80°C in an aqueous stabilizer-free dispersion. The monomer/crosslinker pairs selected are styrene/DVB (divinyl benzene), methylmethacrylate/EGDMA (ethylene glycol dimethacrylate), butyl acrylate/EGDMA, and ethylhexyl acrylate/EGDMA. The influence of composition and nature of the polymerizing/crosslinking constituents on the modified PVC particle structure was studied by microscopy methods, porosity measurements, and dynamic mechanical behavior (DMTA). The level of molecular grafting between PVC and the modifying polymer was determined by solvent extraction experiments. This work shows that the different monomers used represent distinct courses of monomer transport through the PVC particles. The characteristics of the modified PVC particle indicate that the polymerization/crosslinking process occurs in both the PVC bulk, i.e., within the walls constituting a particle, and in the PVC pores. No indication of chemical intermolecular interaction within the modified PVC particles was found. In the second approach, a solution of monomer, initiator, and a crosslinking agent is absorbed in commercial suspension-type porous PVC particles, thus forming a dry blend. This dry blend is subsequently reactively polymerized in a twin-screw extruder at an elevated temperature, 180°C, in the molten state. The properties of the reactively extruded PVC/PMMA blends are compared with those of physical blends at similar compositions. Owing to the high polymerization temperature, short-chain polymers are formed in the reactive polymerization process. Reactively extruded PVC/PMMA blends are transparent, form single-phase morphology, have a single Tg, and show mechanical properties comparable with those of the neat PVC. [source] The Cornubian Batholith: an Example of Magmatic Fractionation on a Crustal ScaleRESOURCE GEOLOGY, Issue 3 2006Bruce W. Chappell Abstract. The Cornubian Batholith comprises six major and several smaller bodies of S-type granite in southwestern England. These late-Variscan granites comprise two-mica granites, and much less abundant Li-mica granites that are restricted to one of the major bodies (St Austell) and smaller bodies. Some of these intrusive rocks are associated with major Sn mineralization. This paper is concerned with the geochemistry of the two-mica granites, which are felsic, strongly peraluminous, and have a high total alkali content and low Na:K. Rocks with very similar compositions to these granites occur elsewhere, including the Variscan granites of continental Europe, and in southeastern Australia. In detail all of the major plutons of this batholith have distinctive compositions, except for Bodmin Moor and Carnmenellis which cannot be discriminated from each other compositionally. A comparison with experimental data shows that the granites attained their major element composition under conditions of crystal-liquid equilibrium, with the final melt being saturated in H2O, at temperatures close to 770d,C and pressures about 50 MPa. That temperature estimate is in good agreement with values obtained from zircon saturation thermometry. The specific minimum-temperature composition excludes the possibility of widespread transfer of elements during hydrothermal alteration. Minor elements that are relatively very abundant are Li, B, Cs and U, while F, Ga, Ge, Rb, Sn, Ta, W and Tl are quite abundant and P is high for felsic rocks. Sr, Ba, and the trace transition metals Sc to Zn, are low, but not as low as they commonly are in very felsic granites. These trace element abundances, and the EL2O-saturation, resulted from the fractional crystallization of a melt derived by the partial melting of feldspathic greywackes in the crust. The Cornubian granites have compositions very similar to the more felsic rocks of the Koetong Suite of southeastern Australia, where a full range of granites formed at the various stages of magmatic fractionation postulated for the Cornubian granites, can be observed. The operation of fractional crystallization in the Cornubian granites is confirmed by the high P abundances in the feldspars, with P contents of the plagioclase crystals correlating with Ab-con-tent Most of the granites represent solidified melt compositions but within the Dartmoor pluton there is a significant component of granites that are cumulative, shown by their higher Ca contents. The Cornubian plutons define areas of high heat flow, of a magnitude which requires that fractionated magmas were transported laterally from their sources and concentrated in the exposed plutons. The generation of these granite plutons therefore involved magmatic fractionation during the stages of partial melting, removal of unmelted material from that melt, and fractional crystallization. During the later stages of those processes, movement of those magmas occurred on a crustal scale. [source] Utilization of waste material resulting from trout processing in gilthead bream (Sparus aurata L.) dietsAQUACULTURE RESEARCH, Issue 2001Y P Kotzamanis Abstract Fish processing creates a large amount of waste of high nutrient content which, if not properly processed for use in human or animal nutrition, is likely to be deposited in the environment creating pollution problems. Waste parts from rainbow trout processing for smoking, consisting of heads, bones, tails and intestines, were used as feed ingredients for gilthead bream diets. Heads, bones and tails had similar compositions, their weighed mean indicating about 700 g kg,1 moisture, 150 g kg,1 protein and 110 g kg,1 fat. Intestines contained higher lipid (350 g kg,1) and lower moisture (560 g kg,1) and protein content (80 g kg,1). Seasonal changes in composition indicated significant differences. Three experimental diets were formulated having the same proximate composition on a dry weight basis. The control diet (A) contained fish meal as the main protein source and fish oil as the oil supplement. In diet B part of the protein and most of lipid was provided by trout waste and in diet C most of the lipid was provided by trout intestines. Gilthead bream fingerlings of 4 g initial weight were fed to apparent satiation for 72 days, at a temperature of 20 °C, to an average final weight of 19 g. All diets were fed in a dry form. The experiment was performed in duplicate. Growth and feed utilization data were high and similar among groups. The body composition of the resulting fish did not show any difference among dietary treatments. Differences in liver lipid and fatty acid content were found between all dietary treatments. The growth and body composition data from this preliminary experiment indicated that trout waste could be used successfully as a dietary ingredient of sea bream diets. [source] |
