Discrepancy in measuring CD4 expression on T-lymphocytes using fluorescein conjugates in comparison with unimolar CD4-phycoerythrin conjugates,,

Abstract Background: Numerous methods for quantitative fluorescence calibration (QFC) have been developed to quantify receptor expression on lymphocytes. However, the results from the use of these different QFC methods vary considerably in the literature. To better identify the causes of these discrepancies, we measured CD4 expression using FITC and phycoerythrin (PE) conjugates to stain CYTO-TROLÔ Control Cells and T-lymphocytes in whole blood and isolated cell preparations. We further examined pH of the cellular microenvironment as a cause of discordant results obtained with the FITC conjugate. Methods: Calibration with Quantibrite PE-labeled microspheres and the use of unimolar CD4-PE conjugates provided direct measurement of the antibody bound per cell value (ABC) for CD4 expression on normal T-lymphocytes. Calibration for CD4-FITC monoclonal antibody (Mab) labeled CYTO-TROL Control Cells and normal T-lymphocytes was based on molecules of equivalent soluble fluorochrome (MESF) as determined by FITC-labeled microspheres traceable to NIST RM 8640. The MESF value for CD4-FITC Mab was determined that enabled the conversion of the MESF values obtained for CYTO-TROL cells to ABC. We investigated the likely pH change in the fluorescein microenvironments within FITC-labeled Mab and cells stained with FITC-labeled Mab using a pH sensitive indicator. Results: The mean ABC value for T-lymphocytes prepared from fresh whole blood using CD4-PE conjugate (48,321) was consistent with previous results, and it was much higher than the mean ABC using CD4-FITC Mab (22,156). The mean ABC value for CYTO-TROL cells using CD4-PE conjugate (43,090) was also higher than that using CD4-FITC conjugate (34,734), although the discrepancy was not as great. Further studies suggested the discrepancy in CYTO-TROL results may be accounted for by the low pH of the membrane microenvironment, but the greater discrepancy in T-lymphocytes could not be fully explained. Conclusion: CD4 expression on fresh normal whole blood samples and CYTO-TROL cells can be consistently quantified in ABC units using Quantibrite PE quantification beads and unimolar CD4-PE conjugates. Quantification with CD4-FITC conjugate is not as consistent, but may be improved by the use of CD4 T-cells as biological calibrators. Cross-Modal transfer of the conditioned eyeblink response during interstimulus interval discrimination training in young rats

Abstract Eyeblink classical conditioning (EBC) was observed across a broad developmental period with tasks utilizing two interstimulus intervals (ISIs). In ISI discrimination, two distinct conditioned stimuli (CSs; light and tone) are reinforced with a periocular shock unconditioned stimulus (US) at two different CS,US intervals. Temporal uncertainty is identical in design with the exception that the same CS is presented at both intervals. Developmental changes in conditioning have been reported in each task beyond ages when single-ISI learning is well developed. The present study sought to replicate and extend these previous findings by testing each task at four separate ages. Consistent with previous findings, younger rats (postnatal day,PD23 and 30) trained in ISI discrimination showed evidence of enhanced cross-modal influence of the short CS,US pairing upon long CS conditioning relative to older subjects. ISI discrimination training at PD43,47 yielded outcomes similar to those in adults (PD65,71). Cross-modal transfer effects in this task therefore appear to diminish between PD30 and PD43,47. Comparisons of ISI discrimination with temporal uncertainty indicated that cross-modal transfer in ISI discrimination at the youngest ages did not represent complete generalization across CSs. ISI discrimination undergoes a more protracted developmental emergence than single-cue EBC and may be a more sensitive indicator of developmental disorders involving cerebellar dysfunction. © 2008 Wiley Periodicals, Inc. Post-ingestive effects of nectar alkaloids depend on dominance status of bumblebees

Abstract 1.,Secondary metabolites have acute or chronic post-ingestive effects on animals, ranging from death to growth inhibition to reduced nutrient assimilation. 2.,Although characterised as toxic, the nectar of Gelsemium sempervirens is not lethal to pollinators, even when the concentration of the nectar alkaloid gelsemine is very high. However, little is known about the sublethal costs of nectar alkaloids. 3.,Using a microcolony assay and paired worker bumblebees, the present study measured the effects of artificial nectar containing gelsemine on oocyte development. Oocytes are a sensitive indicator of protein utilisation and general metabolic processes. We also calculated carbohydrate concentrations in the haemolymph to examine energetic costs of gelsemine consumption. 4.,High concentrations of gelsemine significantly reduced mean oocyte width in subordinate bees, while dominant bees showed only a trend towards oocyte inhibition. Gelsemine consumption did not reduce carbohydrate concentrations in haemolymph. 5.,The cost of ingesting gelsemine may be due to direct toxicity of alkaloids or may be an expense associated with detoxifying gelsemine. Detoxification of alkaloids can require reallocation of resources away from essential metabolic functions like reproduction. The risks associated with nectar alkaloid consumption are tied to both the social and nutritional status of the bee. [source]


Dose-related effects following oral exposure of 2,4-dinitrotoluene on the western fence lizard, Sceloporus occidentalis

Abstract 2,4-dintitrotoluene (2,4-DNT) is an explosive frequently found in the soil of military installations. Because reptiles can be common on these sites, ecological risk assessments for compounds such as 2,4-DNT could be improved with toxicity data specific to reptiles. Western fence lizards, Sceloporus occidentalis, were used to develop a laboratory toxicity model for reptiles. A hierarchical approach was used; acute to subchronic studies were conducted to provide toxicity data relevant to short- and long-term exposures. First, a modified median lethal dose (LD50) study was conducted on male and female lizards using a stage-wise probit model. The LD50 was 577 mg/kg for female and 380 mg/kg for male lizards. Subsequently, a subacute experiment was conducted to further assess 2,4-DNT toxicity to male lizards and to define exposure levels for a longer term, subchronic study. The subchronic study was conducted for 60 consecutive days; male lizards were exposed to 0, 9, 15, 25, 42, 70 mg/kg/d. Dose-dependent mortality was observed in the three highest dose groups (25, 42, and 70 mg/kg/d); all other animals survived the study duration. Benchmark dose model calculations based on mortality indicated a 5% effect level of 15.8 mg/kg/d. At study termination, a gross necropsy was performed, organ weights were taken, and blood was collected for clinical and hematological analysis. Body weight, kidney weight, food consumption, postdose observations, and blood chemistries all were found to be significantly different from controls at doses above 9 mg/kg/d. Also, preliminary results suggest behavioral observations, and reduced food consumption may be a sensitive indicator of toxicity. The present study indicates Sceloporus occidentalis is suitable for evaluating toxicity of compounds to reptilian species. [source]


Lead accumulation in feathers of nestling black-crowned night herons (Nycticorax nycticorax) experimentally treated in the field

Abstract Although lead can attain high concentrations in feathers, interpretation of the biological significance of this phenomenon is difficult. As part of an effort to develop and validate noninvasive methods to monitor contaminant exposure in free-ranging birds, lead uptake by feathers of nestling black-crowned night herons (Nycticorax nycticorax) was evaluated in a controlled exposure study. Four- to 6-d-old heron nestlings (one/nest) at Chincoteague Bay, Virginia (USA), received a single intraperitoneal injection of dosing vehicle (control, n = 7) or a dose of lead nitrate in water (0.01, 0.05, or 0.25 mg Pb/g body wt of nestling; n = 6 or 7/dose) chosen to yield feather lead concentrations found at low- to moderately polluted sites. Nestlings were euthanized at 15 d of age. Lead accumulation in feathers was associated with concentrations in bone, kidney, and liver (r = 0.32,0.74, p < 0.02) but exhibited only modest dose dependence. Blood delta-aminolevulinic acid dehydratase activity was inhibited by lead, although effects on other biochemical endpoints were marginal. Tarsus growth rate was inversely related to feather lead concentration. Culmen growth rate was depressed in nestlings treated with the highest dose of lead but not correlated with feather lead concentration. These findings provide evidence that feathers of nestling herons are a sensitive indicator of lead exposure and have potential application for the extrapolation of lead concentrations in other tissues and the estimation of environmental lead exposure in birds. Organic carbon additions: effects on soil bio-physical and physico-chemical properties

Summary The effects of organic carbon (OC) additions from farm manures and crop residues on selected soil bio-physical and physico-chemical properties were measured at seven experimental sites, on contrasting soil types, with a history of repeated applications of farm manure or differential rates of inorganic fertilizer nitrogen (N). Repeated (> 7 years annual additions) and relatively large OC inputs (up to 65 t OC ha,1) were needed to produce measurable changes in soil properties, particularly physical properties. However, over all the study sites, there was a positive relationship between OC inputs and changes in total soil OC and ,light' fraction OC (LFOC), with LFOC providing a more sensitive indicator of changes in soil organic matter status. Total soil OC increased by an average of 3% for every 10 t ha,1 manure OC applied, whereas LFOC increased by c. 14%. The measured soil OC increases were equivalent to c. 23% of the manure OC applied (up to 65 t OC ha,1 applied over 9 years) and c. 22% of the crop residue OC applied (up to 32 t OC ha,1 over 23 years). The manure OC inputs (but not crop residue OC inputs) increased topsoil porosity and plant available water capacity, and decreased bulk density by 0.6%, 2.5% and 0.5% with every 10 t ha,1 manure OC applied, respectively. Both OC sources increased the size of the microbial biomass (11% increase in biomass C with 10 t OC ha,1 input), but only manure OC increased its activity (16% increase in the soil respiration rate with 10 t OC ha,1 input). Likewise, the potentially mineralizable N pool only increased with manure N inputs (14% increase with 1 t manure total N ha,1). However, these soil quality benefits need to be balanced with any potential environmental impacts, such as excessive nutrient accumulation, increased nitrate leaching and phosphorus losses and gaseous emissions to the atmosphere. The Selective Heating of Iron Nanoparticles in a Single-Mode Microwave for the Patterned Growths of Carbon Nanofibers and Nanotubes

Abstract The fast and cheap synthesis of carbon nanotubes is addressed in a large number of recent publications. At the same time, microwave-assisted synthesis has also gained interest. Besides the fact that reaction kinetics can be positively influenced by the use of microwave irradiation and advanced reaction conditions can be applied, absorption of microwave radiation depends on the material properties, thus resulting in a selective heating mechanism. The selective heating process allows for locally created temperatures high enough to promote the growth of carbon nanofibers and nanotubes on patterned iron catalyst layers. The resulting fibers are micrometers long, and can be synthesized in short time scales of a few minutes, yielding dense films of carbon fibers with uniform height. Here, the selective heating of surface bound iron nanoparticles is investigated in more detail, and experimental evidence for this effect is provided by utilizing a self-assembled monolayer of n -octadecyltrichlorosilane, which acts as a sensitive indicator for locally elevated temperatures. Special emphasis is placed on the development of an improved and controllable experimental setup that permits the safe and fast fabrication of the desired carbon objects. [source]


Predicting population consequences of ocean climate change for an ecosystem sentinel, the seabird Cassin's auklet

Abstract Forecasting the ecological effects of climate change on marine species is critical for informing greenhouse gas mitigation targets and developing marine conservation strategies that remain effective and increase species' resilience under changing climate conditions. Highly productive coastal upwelling systems are predicted to experience substantial effects from climate change, making them priorities for ecological forecasting. We used a population modeling approach to examine the consequences of ocean climate change in the California Current upwelling ecosystem on the population growth rate of the planktivorous seabird Cassin's auklet (Ptychoramphus aleuticus), a demographically sensitive indicator of marine climate change. We use future climate projections for sea surface temperature and upwelling intensity from a regional climate model to forecast changes in the population growth rate of the auklet population at the important Farallon Island colony in central California. Our study projected that the auklet population growth rate will experience an absolute decline of 11,45% by the end of the century, placing this population on a trajectory toward extinction. In addition, future changes in upwelling intensity and timing of peak upwelling are likely to vary across auklet foraging regions in the California Current Ecosystem (CCE), producing a mosaic of climate conditions and ecological impacts across the auklet range. Overall, the Farallon Island Cassin's auklet population has been declining during recent decades, and ocean climate change in this century under a mid-level emissions scenario is projected to accelerate this decline, leading toward population extinction. Because our study species has proven to be a sensitive indicator of oceanographic conditions in the CCE and a powerful predictor of the abundance of other important predators (i.e. salmon), the significant impacts we predicted for the Cassin's auklet provide insights into the consequences that ocean climate change may have for other plankton predators in this system. Understanding the Patient With Migraine: The Evolution From Episodic Headache to Chronic Neurologic Disease.

Traditionally, episodic primary headache disorders are characterized by a return of preheadache (normal) neurologic function between episodes of headache. In contrast, patients with chronic headache often do not return to normal neurologic function between headache attacks. This article proposes that the evolution from episodic migraine to chronic headache may parallel the neurologic disruption observed during the progression of an acute migraine attack and that changes in baseline neurologic function between episodes of headache may be a more sensitive indicator of headache transformation than headache frequency alone. Early recognition of nonheadache changes in nervous system function may offer a more sensitive and specific approach to migraine prevention. [source]


Subtle deficits of attention after surgery: quantifying indicators of sub syndrome delirium

Abstract Objective To determine whether attentional impairments are reliable neuropsychological markers of sub syndrome delirium. Method A prospective cohort study with repeated assessment beginning pre-operatively and continuing through the first post-operative week. Computerized assessments of attention and the Mini-Mental State Examination were administered with one hundred patients admitted for elective orthopedic surgery, 70 years and over and free of dementia. Acute change of cognitive status was used to identify cases of sub syndrome delirium. Results There were significant differences of post-surgical performance between the ,no delirium' and ,sub-syndrome delirium' groups of reaction time, global cognition, accuracy and greater variability of reaction time (p,<,0.041). There were significant within subject main effects on reaction time (p,=,0.001), variability of reaction time (p,=,0.022) and MMSE (p,=,0.000) across the cohort; but no significant interaction effect of ,diagnosis' * ,time' on the computerized measures of attention (p,>,0.195). Conclusion The distinction between people with sub syndrome delirium and no delirium is difficult to quantify but computerized measures of attention might provide a sensitive indicator. Sub syndrome delirium is an observable marker of a clinical abnormality that should be exploited to improve care management for vulnerable patients. Copyright © 2010 John Wiley & Sons, Ltd. Pulse oximeter perfusion index as an early indicator of sympathectomy after epidural anesthesia

Background: The pulse oximeter perfusion index (PI) has been used to indicate sympathectomy-induced vasodilatation. We hypothesized that pulse oximeter PI provides an earlier and clearer indication of sympathectomy following epidural anesthesia than skin temperature and arterial pressure. Methods: Forty patients received lumbar epidural catheters. Patients were randomized to receive either 10 ml 0.5% bupivacaine or 10 ml 0.25% bupivacaine. PI in the toe, mean arterial pressure (MAP) and toe temperature were all assessed at baseline and at 5, 10 and 20 min following epidural anesthesia. The effect of epidural anesthesia over time was assessed by repeated measures analysis of variance. Additionally, we defined clinically evident sympathectomy criteria (a 100% increase in the PI, a 15% decrease in MAP and a 1 °C increase in toe temperature). The numbers of patients demonstrating these changes for each test were compared using the McNemar test for each time point. Results: Twenty-nine subjects had photoplethysmography signals that met a priori signal quality criteria for analysis. By 20 min, PI increased by 326%, compared with a 10% decrease and a 3% increase in MAP and toe temperature, respectively. For PI 15/29, 26/29 and 29/29 of the subjects met the sympathectomy criteria at 5, 10 and 20 min, respectively, compared with 4/29, 6/29 and 18/29 for MAP changes and 3/29, 8/29 and 14/29 for toe temperature changes. Conclusions: PI was an earlier, clearer and more sensitive indicator of the development of epidural-induced sympathectomy than either skin temperature or MAP. [source]


Heightened condition dependence is not a general feature of male eyespan in stalk-eyed flies (Diptera: Diopsidae)

Abstract Stalk-eyed flies are exemplars of sexual selection leading to the evolution of exaggerated male ornaments (eyespan). In Sphyracephala beccarri, there is no evidence for female mate choice for exaggerated male eyespan and only minor sex differences in eyespan. We used S. beccarri to test whether heightened condition dependence only evolves when male eyespan becomes sexually exaggerated. Male eyespan showed heightened condition dependence under food stress compared with a control trait (wing length). However, female eyespan displayed a similar pattern and there was no sex difference in the degree of increased eyespan sensitivity. The finding that eyespan is a sensitive indicator of food stress, even in an unexaggerated state, suggests that this may have acted as a pre-adaptation to the role of eyespan in sexual signalling in other Diopsid species. These results are consistent with handicap theory and Fisher's view of how sexual selection is initiated. [source]


Feeding resumption, morphological changes and mortality during starvation in Japanese flounder larvae

Japanese flounder Paralichthys olivaceus larvae established first feeding 3 days after hatching (DAH) at c. 17° C. Non-fed fish reached irreversible starvation at age 5 DAH. Non-fed fish showed similar feeding rate and feeding intensity as the fed fish when they were provided with prey before 5 DAH, after which the starved larvae did not feed even when prey became available. None of the six morphological measurements examined (total length, body height, eye height, head height, gut height and myotome height) showed significant differences between the non-fed and fed larvae until 5 DAH. Normal development continued only in the fed group, and the non-fed larvae showed reverse growth or body collapse after 5 DAH. Owing to the shrinkage and collapse at the top of head due to starvation, head height could be a sensitive indicator of starvation in Japanese flounder larvae. In the fed treatments, high mortality occurred from first feeding (3 DAH) to irreversible starvation (5 DAH), accounting for about two-thirds to three-quarters of the overall mortality (46,52%) throughout the experiments. This mortality was not prey density or larval density dependent. Mortality during the same period in the non-fed larvae accounted for about a third of the overall mortality (100%). [source]


Influence of protein conformation and adjuvant aggregation on the effectiveness of aluminum hydroxide adjuvant in a model alkaline phosphatase vaccine

Abstract The mechanism(s) of the enhancement of the immune response by addition of aluminum salt adjuvants to parenterally administered protein-based vaccines is still the subject of debate. It has been hypothesized, however, that destabilization of the antigen structure on the surface of the adjuvant may be important for eliciting immune response. Also, it has been suggested that immune response to adjuvanted vaccines is reduced if the adjuvant particles become aggregated before administration because of processing steps such as freeze-drying. In this study, we tested these hypotheses and examined the immune response in a murine model to various liquid, freeze-dried, and spray freeze-dried formulations of a model vaccine, bovine intestinal alkaline phosphatase adsorbed on aluminum hydroxide. Enzymatic activity of the alkaline phosphatase was used as a sensitive indicator of intact native antigen structure. By manipulating the secondary drying temperature during lyophilization, vaccines were produced with varying levels of alkaline phosphatase enzymatic activity and varying degrees of adjuvant aggregation, as assessed by particle size distribution. Phosphatidylethanol and Alcohol Consumption in Reproductive Age Women

Background:, Fetal alcohol disorders are preventable, but self-reported alcohol consumption can be misleading and impede effective treatment. Biomarkers represent an alternative method for assessing alcohol use, and this study evaluated the relationship between blood phosphatidylethanol (PEth) and alcohol use in a sample of reproductive age women. Methods:, Alcohol use was estimated by validated self-report methods in 80 nonpregnant women ages 18 to 35. PEth was measured by a contracted laboratory using a liquid chromatography-tandem mass spectrometry assay. Regression methods appropriate for the distribution of PEth were used to define its relationship to alcohol consumption during the prior 2 weeks and explore the effects of drinking patterns on this association. Receiver operating characteristic analysis was used to estimate the sensitivity of PEth for various drinking levels at 95% specific cutoffs. Results:, PEth had a positive linear association with grams of alcohol consumed (p < 0.001), and was detectable in 93% of subjects consuming an average of 2 or more drinks per day. The relationship between total alcohol consumption and PEth may be stronger in women with recent heavy drinking days. The relationship between drinking and PEth varied considerably between individuals, and sensitivity for a certain amount of drinking was low at a highly specific cutoff concentration. Conclusions:, PEth is a highly sensitive indicator of moderate and heavy alcohol consumption in reproductive age women and may complement the use of self-report alcohol screens when additional objective markers of alcohol use are desirable. However, choosing a highly valid cutoff concentration for PEth to differentiate various levels of alcohol consumption may not be feasible. [source]


Prognostic significance of CEA levels and detection of CEA mRNA in draining venous blood in patients with colorectal cancer

Abstract Background and Objectives: The aims of this study were to determine carcinoembryonic antigen (CEA) levels and incidence of tumor cells using the RT-PCR technique in draining venous blood of patients with colorectal cancer, correlate the results with various histopathologic factors and determine their significance as prognostic factors. Methods: From 1995 to 2000, 108 patients with adenocarcinoma of the colon or rectum, underwent curative surgery and enrolled in this prospective study. Results: The 5-year survival group had significantly lower portal CEA levels compared to the hepatic metastasis outcome group. CEA mRNA was positive in the draining venous blood from 12 (11.1%) out of 108 patients included in the study. The rate of positive tumor cell detection in portal blood was significantly higher in the hepatic metastasis outcome group than in the 5-year survival and recurrence group. The proportion of patients with portal CEA ,5 ng/ml was greater in patients with higher stage than in patients with lower stage. Conclusions: Positive CEA mRNA in draining venous blood predicted hepatic metastases and local recurrence with accuracy over 80% but with low sensitivity of 30% and 9%, respectively. Moreover, CEA level was a sensitive indicator in hepatic metastases as sensitivity was 95% and a specific indicator in predicting 5-year survival with specificity 84%. J. Surg. Complementary (secondary) metabolites in a soft coral: sex-specific variability, inter-clonal variability, and competition

Abstract Sex-specific interactions involving competition for space between the dioecious alcyonacean soft coral Sarcophyton glaucum and the scleractinian coral Acropora robusta were assessed experimentally on Bald Rock, central region of the Great Barrier Reef. To examine this, plus inter-clonal responses, one male colony of S. glaucum, known to produce sarcophytoxide as its predominant complementary (secondary) metabolite, was sectioned, producing 10 clones. The same was done for a female colony. These two sets of clones were then relocated to grids and placed in contact with Acropora clones. Relocated and non-relocated controls were also monitored. High levels of tissue necrosis were observed in the hard coral under contact conditions with both the male and female clones after 20 days. The development of a protective polysaccharide layer in the alcyonacean was also observed. Differences observed in the concentrations of complementary metabolites within the two different S. glaucum colonies were related to sex. Both under competition and non-competition conditions, females exhibited significantly higher concentrations of sarcophytoxide than males, and this increased with time. Fatty ester concentration was also higher in females than males, varying significantly through time, and falling dramatically just after spawning. Fatty ester concentrations decreased linearly through time in the male clones. When involved in competition for space, females possessed higher concentrations of fatty esters than males, both at the site of contact and in non-contact sites, again, decreasing after spawning. No significant changes in sarcophytoxide levels were noted in the parental colonies, but such changes were observed in fatty esters, with the female producing higher concentrations until after spawning. The use of these two variates in the form of a ratio (sarcophytoxide concentration:fatty ester concentration) yielded a variable Rho (,) which was a more sensitive indicator of biochemical change than either of its components alone. These two sets of compounds appeared to have a negative association through time and varied highly significantly between sexes. The diterpene sarcophytoxide may be considered an allelopathic or stress metabolite, while the lipids act as energy storage metabolites. [source]


Molecular targets of botulinum toxin at the mammalian neuromuscular junction

Abstract The molecular targets of botulinum neurotoxins (BoNTs) are SNARE (soluble N -ethylmaleimide-sensitive factor- attachment protein- receptor) proteins necessary for neurotransmitter release. BoNT are powerful therapeutic agents in the treatment of numerous neurological disorders. The goals of this study were to (1) assess toxin diffusion by measuring substrate cleavage in adjacent and distant muscles, and (2) characterize the clinical course using SNARE protein chemistry. A small volume of BoNT/A was injected unilaterally into the mouse gastrocnemius muscle. Motor impairment was limited to the toxin-treated limb. No systemic illness or deaths occurred. At five time points, a subset of mice were killed, and muscles from both hindlimbs, and the diaphragm, were collected. Protein samples were examined for changes in SNAP-25 (synaptosomal-associated protein of Mr = 25 kDa) using immunochemistry. SNAP-25 cleavage product was noted in the toxin-treated limb as early as 1 day postinjection and continued through day 28. Onset and peak levels of substrate cleavage corresponded to the onset and peak clinical response. Cleavage was observed in adjacent and distant muscles, demonstrating that substrate cleavage is a sensitive indicator of toxin diffusion. Significant increases in full-length SNAP-25 and vesicle-associated membrane protein II were evident early in the impaired limb and continued through day 28. Reaction norms of life history traits in response to zinc in Thlaspi caerulescens from metalliferous and nonmetalliferous sites

Summary ,,We examined phenotypic plasticity of fitness components in response to zinc (Zn) in the Zn hyperaccumulator, Thlaspi caerulescens. ,,Two populations from Zn-enriched soils (M) and two populations from normal soils (NM) were grown in pots at three Zn concentrations (0, 1000 and 8000 mg kg,1 Zn), for an entire life cycle. Growth, Zn accumulation and fitness components were assessed. ,,Based on vegetative growth, M and NM populations had similar Zn tolerance at 1000 mg kg,1 Zn. However, reproductive output was markedly decreased in NM at 1000 and 8000 mg kg,1 Zn. In M populations, Zn did not affect fitness. However, low Zn status enhanced reproductive output in year 1 compared with year 2 and decreased survival after the first flowering season. ,,M populations are able to achieve equal fitness across a broad range of Zn concentrations in soil by different combinations of fecundity and longevity. No cost of higher tolerance was demonstrated in M populations. Reproductive traits appeared to be a more sensitive indicator of tolerance than vegetative growth. [source]


Height and relative leg length as indicators of the quality of the environment among Mozambican juveniles and adolescents

The growth status of Mozambique adolescents was assessed to test the hypothesis that relative leg length is a more sensitive indicator of the quality of the environment than the total height. The sample comprised 690 boys and 727 girls, aged between 9 and 17 years, from Maputo. It is divided between those living in the Centre of Maputo and those living in the slums on the periphery of the city. Height, weight, and sitting height were measured and the sitting height ratio was calculated. The hypothesis that relative leg length is more sensitive than total stature as an indicator of environmental quality is not uniformly confirmed. Overall, mean stature is greater for the centre group than the slum group, but relative leg length as measured by the sitting height ratio does not differ. Compared with African-American references (NHANES II), all centre girls, 9- to 14-year-old slum girls, all slum boys, and the oldest centre boys show relatively shorter legs. These findings show that within the Mozambique sample, relative leg length is not sensitive enough to distinguish the quality of the living environment. Mozambique was a colony of Portugal until 1975. Civil unrest and warfare characterized the late Colonial period and the postindependence period until a peace settlement was concluded in 1992. It is possible that all socioeconomic status groups within the country suffered sufficiently to reduce relative leg length compared with the better-off African-American reference sample. The effects of socioeconomic status on endochondral and appositional bone growth, and acquisition of cortical bone in children from 19th century Birmingham, England

Abstract Endochondral growth, appositional growth, and acquisition of cortical bone thickness in the femur are investigated in subadult skeletons (N = 43, dental age range birth to 12 years) from the 19th -century AD burial site of St. Martin's churchyard, Birmingham, England. Endochondral growth is monitored using diaphyseal femoral length. Appositional growth is monitored using radiographic midshaft mediolateral width and acquisition of cortical bone using combined mediolateral cortical thickness measured at the midshaft from radiographs. The methodology involves plotting these variables against dental age. Growth is compared in children of differing socioeconomic status. Higher and lower status individuals are identified in the assemblage by their burial in brick vaults in the case of the former and in earth-cut graves in the case of the latter. The relationships between bone dimensions and dental age are described using a polynomial regression procedure, and analysis of regression residuals is used to evaluate differences in bone dimension-for-dental age between the two status groups. Results show that lower socioeconomic status individuals had lower cortical thickness-for-dental age than those of higher status. This was interpreted as likely reflecting poorer nutrition in the children of lower socioeconomic backgrounds. There was no patterning with respect to socioeconomic status in femur diaphyseal length or midshaft width. The results support the idea that, for skeletal populations, growth in cortical thickness may be a more sensitive indicator of adverse conditions in childhood than growth in bone length or width. Endothelial Gene Expression in Kidney Transplants with Alloantibody Indicates Antibody-Mediated Damage Despite Lack of C4d Staining

Anti-HLA alloantibody is a risk factor for graft loss, but does not indicate which kidneys are experiencing antibody-mediated rejection (ABMR). C4d staining in biopsies is specific for ABMR but insensitive. We hypothesized that altered expression of endothelial genes due to alloantibody acting on the microcirculation would be sensitive indicator of ABMR. We identified 119 endothelial-associated transcripts (ENDATs) from literature, and studied their expression by microarrays in 173 renal allograft biopsies for cause. Mean ENDAT expression was increased in all rejection but was higher in ABMR than in T-cell-mediated rejection and correlated with histopathologic lesions of ABMR, and alloantibody. Many individual ENDATs were increased in ABMR and predicted graft loss. Kidneys with high ENDATs and antibody showed increased lesions of ABMR and worse prognosis in comparison to controls. Only 40% of kidneys with high ENDAT expression and chronic ABMR or graft loss were diagnosed by C4d positivity. High ENDAT expression with antibody predicts graft loss with higher sensitivity (77% vs. 31%) and slightly lower specificity (71% vs. 94%) than C4d. The results were validated in independent set of 82 kidneys. High renal endothelial transcript expression in patients with alloantibody is indicator of active antibody-mediated allograft damage and poor graft outcome. [source]


Gas sensing in microplates with optodes: Influence of oxygen exchange between sample, air, and plate material

Abstract Microplates with integrated optical oxygen sensors are a new tool to study metabolic rates and enzyme activities. Precise measurements are possible only if oxygen exchange between the sample and the environment is known. In this study we quantify gas exchange in plastic microplates. Dissolved oxygen was detected using either an oxygen-sensitive film fixed at the bottom of each well or a needle-type sensor. The diffusion of oxygen into wells sealed with different foils, paraffin oil, and paraffin wax, respectively, was quantified. Although foil covers showed the lowest oxygen permeability, they include an inevitable gas phase between sample and sealing and are difficult to manage. The use of oil was found to be critical due to the extensive shaking caused by movement of the plates during measurements in microplate readers. Thus, paraffin wax was the choice material because it avoids convection of the sample and is easy to handle. Furthermore, without shaking, significant gradients in pO2 levels within a single well of a polystyrene microplate covered with paraffin oil were detected with the needle-type sensor. Higher pO2 levels were obtained near the surface of the sample as well as near the wall of the well. A significant diffusion of oxygen through the plastic plate material was found using plates based on polystyrene. Thus, the location of a sensor element within the well has an effect on the measured pO2 level. Using a sensor film fixed on the bottom of a well or using a dissolved pO2 -sensitive indicator results in pO2 offset and in apparently lower respiration rates or enzyme activities. Oxygen diffusion through a polystyrene microplate was simulated for measurements without convection,that is, for samples without oxygen diffusion through the cover and for unshaken measurements using permeable sealings. Evaluation of the atrophogenic potential of different glucocorticoids using optical coherence tomography, 20-MHz ultrasound and profilometry; a double-blind, placebo-controlled trial

Summary Background, Skin atrophy is one of the main side-effects of topical corticosteroid therapy. Although the use of high-frequency ultrasound is an established method that has been studied previously, it allows measurements of the slow-reacting dermal thickness only. Objectives, To investigate the decreasing epidermal thickness, which occurs earlier, we used optical coherence tomography (OCT), a high-resolution noninvasive imaging technique, and compared it with 20-MHz ultrasound and profilometry. Patients/methods, In this double-blind placebo-controlled trial 20 healthy volunteers applied four different corticosteroids and the cream base formulation as placebo to the volar part of both arms once a day over a 4-week period. The epidermal thickness, the dermal thickness and the skin surface roughness were assessed using OCT, high-frequency ultrasound and profilometry. Results, Each of the three methods allowed the detection and monitoring of significant corticosteroid-induced skin atrophy and its reversibility. The changes correlated with the potency of the steroids. The epidermal thickness decreased significantly in all test areas, even in the placebo and the untreated fields. As expected, the reduction in epidermal thickness was more pronounced and could be detected earlier by OCT than the reduction of dermal thickness using ultrasound. The epidermal surface roughness investigated using profilometry showed a slight smoothing. Conclusions, OCT allows a simple, fast and noninvasive in vivo measurement of the epidermal thickness. To evaluate the atrophogenic potential of corticosteroids it is more suitable than high-frequency ultrasound as epidermal thickness decreases earlier. In addition, epidermal thickness is a more sensitive indicator of steroid atrophy as the degree of thinning is much higher compared with the dermal atrophy. Profilometry might give further information; however, it would not be suitable for clinical use as the results were generally less pronounced. In the future, OCT might be useful to detect corticosteroid-induced side-effects at the beginning for monitoring the therapy. [source]


Post-Impact Assessment of Oil Pollution in Agbada West Plain of Niger Delta, Nigeria: Field Reconnaissance and Total Extractable Hydrocarbon Content

Typical post-impact assessment of an oil-spill-impacted site in Agbada west plain of the Niger Delta basin of Nigeria was carried out 13 months after recorded incidence of spillage, using empirical indices of reconnaissance and extractable hydrocarbon content. Field-reconnaissance surveys revealed lower species' numbers and diversity of surface and subterranean flora and fauna. The presence and absence of such plant species as Elaeis guineensis, Musanga cecropiodes, and Andropogon gayanus, as well as animal species like earthworms and nematodes in sampled plots, corroborated the evidence provided by the total extractable hydrocarbon content (of (2.53±0.43)×102,mg/kg and (2.00±0.76)×102,mg/kg at surface and subsurface depths, resp.) on the level of degradation and/or regeneration at the polluted site. The most important evidence that oiling must have been responsible, at least in part, for the reduction in species' number and diversity was provided by the plant cover, Andropogan gayanus, which occurred at a lower density of 0.9,plants/m2 in the oil-impacted plots and 14.00 plants/m2 in the unimpacted reference plots of an adjacent, geographically similar area. The presence of this species on site thus presented a significant difference (P<0.05) of over 85%, and indicated exhaustion or impoverishment of impacted soils. Elaeis guineensis, with a population density of 0.1,stands/m2, provided evidence of past cultivation on site, while species like Musanga cecropioides, with a density of 0.5,stands/m2, at the unimpacted site, confirmed the abandonment of the farmstead over a long period of time. For the fauna, the most sensitive indicator of hydrocarbon toxicity was provided by nematode Xiphinema sp. that were completely absent in the oil-impacted site. [source]


Early prediction of anthracycline induced cardiotoxicity

Abstract Aim: The purpose of this study is to evaluate echocardiographically determined cardiac functions with serum levels of brain natriuretic peptide (BNP), cardiac troponin I (cTnI) and total antioxidant status (TAOS) in childhood leukemia treated with chemotherapeutics containing anthracyclines. Methods: A study group of 29 patients who have been followed for acute lymphoblastic leukemia (ALL) and administered a treatment protocol containing chemotherapy of anthracyclines were included in the analysis. Levels of BNP, cTnI and TAOS were studied in serum samples of the patients. Results: We demonstrated that as the drug dosage increased, systolic ejection fraction (EF) and shortening fraction (FS) values decreased (EF r2= 0.2327, FS r2= 0.251). On the other hand, increased dosage of anthracycline therapy was associated with significant raise in plasma BNP levels (r2= 0.246) and significant decrease in serum TAOS levels (r2= 0.317) without any change in serum cTnI levels. Conclusion: Our study suggest that serum TAOS and BNP levels may be useful as an early and sensitive indicator of anthracycline induced cardiotoxicity. [source]


Human impacts on the species,area relationship in reef fish assemblages

Abstract The relationship between species richness and area is one of the oldest, most recognized patterns in ecology. Here we provide empirical evidence for strong impacts of fisheries exploitation on the slope of the species,area relationship (SAR). Using comparative field surveys of fish on protected and exploited reefs in three oceans and the Mediterranean Sea, we show that exploitation consistently depresses the slope of the SAR for both power-law and exponential models. The magnitude of change appears to be proportional to fishing intensity. Results are independent of taxonomic resolution and robust across coral and rocky reefs, sampling protocols and statistical methods. Changes in species richness, relative abundance and patch occupancy all appear to contribute to this pattern. We conclude that exploitation pressure impacts the fundamental scaling of biodiversity as well as the species richness and spatial distribution patterns of reef fish. We propose that species,area curves can be sensitive indicators of community-level changes in biodiversity, and may be useful in quantifying the human imprint on reef biodiversity, and potentially elsewhere. Ammonium perchlorate effects on thyroid function and growth in bobwhite quail chicks

Abstract Bobwhite quail chicks were used to investigate ammonium perchlorate (AP; NH4ClO4) effects on thyroid function and growth. Beginning at 3 to 4 d posthatch, we evaluated organismal thyroid status (circulating hormones), activation of the hypo-thalamic-pituitary-thyroid axis (thyroid wt) and thyroidal hormone content over a wide range of AP concentrations (50 ,g/L , 4,000 mg/L) in drinking water, for relatively short (2-week) and longer (8-week) exposures. Thyroidal thyroxine (T4) content, the most sensitive index of decreased thyroid function, decreased markedly in response to increasing perchlorate exposure. Thyroid weight and plasma T4 were less sensitive indicators and similar in their ability to detect thyroid changes. Growth measurements (body wt and skeletal growth) were very insensitive indices. Because thyroids contain large hormone stores, with low exposures or short time periods, these stores can be used to maintain circulating hormones, at least temporarily. Most depletion of thyroidal T4 occurred during the first two weeks of AP exposure. Subsequent decreases were at a slower rate presumably because thyrotropin stimulation of the thyroids at least partially compensated for some of the perchlorate effect. Additional studies of the interactions between AP concentration and exposure time are needed for understanding the complex nature of thyroid responses to perchlorate. [source]


HISTORIC CYCLES OF FRAGMENTATION AND EXPANSION IN PARNASSIUS SMINTHEUS (PAPILIONIDAE) INFERRED USING MITOCHONDRIAL DNA

Abstract Climate oscillations of the Quaternary drove the repeated expansion and contraction of ecosystems. Alpine organisms were probably isolated in sky island refugia during warm interglacials, such as now, and expanded their range by migrating down-slope during glacial periods. We used population genetic and phylogenetic approaches to infer how paleoclimatic events influenced the distribution of genetic variation in the predominantly alpine butterfly Parnassius smintheus. We sequenced a 789 bp region of cytochrome oxidase I for 385 individuals from 20 locations throughout the Rocky Mountains, ranging from southern Colorado to northern Montana. Analyses revealed at lease two centers of diversity in the northern and southern Rocky Mountains and strong population structure. Nested clade analysis suggested that the species experienced repeated cycles of population expansion and fragmentation. The estimated ages of these events, assuming a molecular clock, corresponded with paleoclimatic data on habitat expansion and contraction over the past 400,000 years. We propose that alpine butterflies persisted in an archipelago of isolated sky islands during interglacials and that populations expanded and became more connected during cold glacial periods. An archipelago model implies that the effects of genetic drift and selection varied among populations, depending on their latitude, area, and local environment. Alpine organisms are sensitive indicators of climate change and their history can be used to predict how high-elevation ecosystems might respond to further climate warming. [source]


Habitat-specific normal and reverse diel vertical migration in the plankton-feeding basking shark

Summary 1Megaplanktivores such as filter-feeding sharks and baleen whales are at the apex of a short food chain (phytoplankton,zooplankton,vertebrate) and are sensitive indicators of sea-surface plankton availability. Even though they spend the majority of their time below the surface it is still not known how most of these species utilize vertical habitat and adapt to short-term changes in food availability. 2A key factor likely to control vertical habitat selection by planktivorous sharks is the diel vertical migration (DVM) of zooplankton; however, no study has determined whether specific ocean-habitat type influences their behavioural strategy. Based on the first high-resolution dive data collected for a plankton-feeding fish species we show that DVM patterns of the basking shark Cetorhinus maximus reflect habitat type and zooplankton behaviour. 3In deep, well-stratified waters sharks exhibited normal DVM (dusk ascent,dawn descent) by tracking migrating sound-scattering layers characterized by Calanus and euphausiids. Sharks occupying shallow, inner-shelf areas near thermal fronts conducted reverse DVM (dusk descent,dawn ascent) possibly due to zooplankton predator,prey interactions that resulted in reverse DVM of Calanus. 4These opposite DVM patterns resulted in the probability of daytime-surface sighting differing between these habitats by as much as two orders of magnitude. Ship-borne surveys undertaken at the same time as trackings reflected these behavioural differences. 5The tendency of basking sharks to feed or rest for long periods at the surface has made them vulnerable to harpoon fisheries. Ship-borne and aerial surveys also use surface occurrence to assess distribution and abundance for conservation purposes. Our study indicates that without bias reduction for habitat-specific DVM patterns, current surveys could under- or overestimate shark abundance by at least 10-fold. [source]
