Tolerance of barley seed germination to cold- and drought-stress expressed as seed vigour

Abstract In barley, high seed vigour is a precondition for rapid and homogenous field emergence and good malting quality. Seed vigour was defined as germination percentage under stress conditions (10°C, drought stress - 2 Bars) in 7-8 barley varieties grown in 7-8 locations in the Czech Republic over 7 years. Three of the 7 years were not suitable for high seed quality, probably because of unsuitable weather, as average seed vigour reached only 61, 77 and 86%, respectively. In the remaining 4 years, the average vigour exceeded 94%. The impact of variety on seed vigour was higher in the ,bad years' and the impact of location was higher in the other years. Varieties with higher vigour from all locations in the ,bad years' were identified. Lower vigour was related to the high occurrence of fungi (indicated by ergosterol assays) and to lower field emergence rates of seed samples. The results support the possibility of selecting for improvement of barley seed vigour, which is related to tolerance to various conditions during emergence and to homogenous malting. [source]


Preparation and application of a novel environmentally friendly organic seed coating for rice

Abstract BACKGROUND: Traditional rice seed coating techniques involve the use of chemical pesticides, which can damage the seed in the process and cause possible physical and environmental damage. Increasing knowledge and concern about the traditional applications have brought new attention to the industry and the search for a novel coating agent that is effective, safe and environmentally friendly. A new type of organic rice seed coating agent was developed using liquid-based polymeric adhesives. By using chitosan as the main raw material, modified with sodium hydroxide and polymerised with plant growth regulators and other additives, the novel seed coating agent is a safer, cheaper and more environmentally friendly alternative. RESULTS: The novel seed coating agent significantly enhanced sprout growth over traditional agents. We found it has obvious biological advantages: it stimulates the seedling growth of rice, advances the growth of root, improves root activity and increases the crop yield in the germination test and field trial. Compared with the traditional rice seed coating agent, the crop yield of seeds coated by the novel seed coating agent was increased by 5%, and at 25% less cost. The fungal inhibition test of the novel seed coating agent and acute toxicity test on fish showed that it has an obvious fungal inhibitory effect and a higher safety index during usage and disposal. CONCLUSION: This result suggests that treating seeds with the novel seed coating agent has significant agricultural implications through the enhanced seed vigour as reflected in growth. It is efficient and effective, resulting in better seed and crop protection. The novel seed coating agent demonstrates unique characteristics with great economic and environmental benefits. Rapid determination of rice seed vigour by spontaneous chemiluminescence and singlet oxygen generation during early imbibition

Abstract Using a highly sensitive single photon counter, a spontaneous chemiluminescence (CL) study on rice (Oryza sativa L.) seeds stored in different years was carried out. We first observed that the degree of ageing in rice seeds was related to the intensity of spontaneous CL during early imbibition (0,30 min). Rice seeds stored for a shorter time had a stronger intensity of CL in early imbibition. The germination rate of rice seeds showed an obvious positive correlation with the intensity of spontaneous CL. Singlet oxygen (1O2) in rice seeds during early imbibition was investigated by a CL method using a cypridina luciferin analogue, 2-methyl-6-(p -methoxyphenyl)-3,7-dihydroimidazo [1,2,] pyrazin-3-one (MCLA), as a selective CL probe. Additional experimental evidence for the formation of 1O2 came from the quenching effect of sodium azide (NaN3) on MCLA-mediated rice seeds' CL. Analysis based on the experimental results demonstrated that spontaneous CL in rice seeds during early imbibition was mainly contributed by singlet oxygen (1O2).
