Electrokinetic supercharging for highly efficient peptide preconcentration in capillary zone electrophoresis

Abstract Electrokinetic supercharging has been integrated in CZE for the development of a highly sensitive methodology for protein tryptic digest analysis. A careful choice of the experimental conditions led to sensitivity enhancement factors between 1000 and 10,000 whilst maintaining a satisfactory resolution. Peptides in the low nanomolar concentration range have been detected despite the use of the poorly sensitive UV absorbance detection mode. The buffer system used in this study is fully suitable for coupling CE to MS. [source]


Performance of very-high-order upwind schemes for DNS of compressible wall-turbulence

Abstract The purpose of the present paper is to evaluate very-high-order upwind schemes for the direct numerical simulation (DNS) of compressible wall-turbulence. We study upwind-biased (UW) and weighted essentially nonoscillatory (WENO) schemes of increasingly higher order-of-accuracy (J. Comp. Phys. 2000; 160:405,452), extended up to WENO17 (AIAA Paper 2009-1612, 2009). Analysis of the advection,diffusion equation, both as ,x,0 (consistency), and for fixed finite cell-Reynolds-number Re,x (grid-resolution), indicates that the very-high-order upwind schemes have satisfactory resolution in terms of points-per-wavelength (PPW). Computational results for compressible channel flow (Re,[180, 230]; M,CL,[0.35, 1.5]) are examined to assess the influence of the spatial order of accuracy and the computational grid-resolution on predicted turbulence statistics, by comparison with existing compressible and incompressible DNS databases. Despite the use of baseline O(,t2) time-integration and O(,x2) discretization of the viscous terms, comparative studies of various orders-of-accuracy for the convective terms demonstrate that very-high-order upwind schemes can reproduce all the DNS details obtained by pseudospectral schemes, on computational grids of only slightly higher density. Internal Thoracic Arterial Grafts Evaluation by Multislice CT Scan:

A Preliminary Study
The aim of this study is to evaluate the multidetector multislice CT Scan (MCTS) as a means of postoperative evaluation of ITA coronary artery bypass grafts. Methods: Twenty-eight patients having been operated on for coronary artery bypass with ITA during a 6-months period, benefited, 7 days after surgery, from a patency and anastomotic site control of ITA with a MCTS associated with cardiac gating (Light Speed, General Electric, USA). Results: Internal thoracic artery bypasses are visualized perfectly on all their courses, with possibility of 3D reconstructions, showing the relationship between cardiac cavities and the arterial bypasses. The anastomotic site on the LAD was, in selected cases, perfectly visualized. Sequential bypasses with left ITA are well visualized as well as T or Y right-to-left ITA grafts. However, surgical clips create some image artefacts. Conclusions: The postoperative control of ITAs are possible by MCTS with a satisfactory resolution. This makes it possible to check the patency of ITAs, their course on the heart surface, and the location and quality of anastomosis with a noninvasive reproductive method. [source]


Clinical trial: the treatment of gastro-oesophageal reflux disease in primary care , prospective randomized comparison of rabeprazole 20 mg with esomeprazole 20 and 40 mg

Summary Background, A trial of empirical PPI therapy is usual practice for most patients with symptoms of gastro-oesophageal reflux disease (GERD) in primary care. Aim, To determine if the 4-week efficacy of rabeprazole 20 mg for resolving heartburn and regurgitation symptoms is non-inferior to esomeprazole 40 mg or 20 mg. Methods, In all, 1392 patients were randomized to rabeprazole 20 mg, esomeprazole 20 mg or 40 mg once daily. Patients, doctors and assessors were blinded. Symptom resolution data were collected on days 0,7 and day-28 using the Patient Assessment of Upper Gastrointestinal Disorders Symptom Severity Index with a shortened version used on days 8,27. Results, Rabeprazole 20 mg was non-inferior to esomeprazole 40 mg for complete resolution of regurgitation and satisfactory resolution of heartburn and regurgitation. For complete heartburn resolution, the efficacy of rabeprazole 20 mg and esomeprazole 40 mg was statistically indistinguishable, although the non-inferiority test was inconclusive. Rabeprazole 20 mg was non-inferior to esomeprazole 20 mg for all outcomes. Conclusions, In uninvestigated GERD patients, rabeprazole 20 mg was non-inferior to esomeprazole 40 mg for complete and satisfactory relief of regurgitation and satisfactory relief of heartburn, and not different for complete resolution of heartburn. The U.S. Financial Sanctions against North Korea

In September 2005, the U.S. imposed financial restrictions on North Korea after blaming the North for illicit financial activities, including counterfeiting and money-laundering. The U.S. financial sanctions against the North had direct and immediate impact on the ongoing six-party negotiations on North Korea's nuclear issue. North Korea insisted on the lifting of U.S. financial sanctions as the precondition for returning to the negotiating table and consequently the six-party talks on North Korea's nuclear issue stalled. The third session of the fifth round of six-party talks in February 2007 produced an important accord on North Korea's nuclear question. This agreement was made possible after the U.S. and the DPRK reached a compromise on the financial sanctions issue. This article discusses the U.S. financial sanctions against North Korea and their implications for North Korea's nuclear question. It begins with an overview of the U.S. financial restrictions. This study then examines the nexus between the financial sanctions and the impasse at the six-party talks on North Korea's nuclear issue. This is followed by a discussion of the breakthrough on the financial restrictions issue and the landmark agreement on North Korea's nuclear issue in February 2007. In this study, the authors argue that a mutually satisfactory resolution of the BDA dispute holds the key to a peaceful settlement of the second North Korean nuclear crisis. With the BDA dispute behind, the six-party talks should gain momentum and prepare a road map for implementing the denuclearization of the Korean peninsula. Both the U.S. and North Korea should not miss this golden opportunity and make earnest efforts to build a firm foundation for peace and stability on the Korean peninsula and in Northeast Asia. I defend contextualism against an objection that it cannot explain how ,knows' and its cognates function inside propositional attitude reports. A validated enantiospecific method for determination and purity assay of clopridogrel

Abstract A new and accurate HPLC method using ,-cyclodextrin chemically bonded to spherical silica particles as chiral stationary phase (CSP) was developed and validated for determination of S -clopidogrel and its impurities R -enantiomer and S -acid as a hydrolytic product. The effects of acetonitrile and methanol content in the mobile phase and temperature on the resolution and retention of enantiomers were investigated. A satisfactory resolution of S -clopidogrel active form and its impurities was achieved on ChiraDex® column (5 ,m, 4 × 250 mm) at a flow rate of 1.0 ml/min and 17°C using acetonitrile, methanol and 0.01 M potassium dihydrogen phosphate solution (15:5:80 v/v/v) as mobile phase. The detection wavelength was set at 220 nm. The method was validated in terms of accuracy, precision, linearity, and robustness. The limit of detection for R- enantiomer and S -acid were 0.75 and 0.09 ,g/ml, respectively, injection volume being 20 ,l. Finally, the molecular modeling of the inclusion complexes between the analytes and ,-cyclodextrin was performed to investigate the mechanism of the enantiorecognition and to study the quantitative structure,retention relationships.
