    Yeast species composition differs between artisan bakery and spontaneous laboratory sourdoughs

    Abstract Sourdough fermentations are characterized by the combined activity of lactic acid bacteria and yeasts. An investigation of the microbial composition of 21 artisan sourdoughs from 11 different Belgian bakeries yielded 127 yeast isolates. Also, 12 spontaneous 10-day laboratory sourdough fermentations with daily backslopping were performed with rye, wheat, and spelt flour, resulting in the isolation of 217 yeast colonies. The isolates were grouped according to PCR-fingerprints obtained with the primer M13. Representative isolates of each M13 fingerprint group were identified using the D1/D2 region of the large subunit rRNA gene, internal transcribed spacer sequences, and partial actin gene sequences, leading to the detection of six species. The dominant species in the bakery sourdoughs were Saccharomyces cerevisiae and Wickerhamomyces anomalus (formerly Pichia anomala), while the dominant species in the laboratory sourdough fermentations were W. anomalus and Candida glabrata. The presence of S. cerevisiae in the bakery sourdoughs might be due to contamination of the bakery environment with commercial bakers yeast, while the yeasts in the laboratory sourdoughs, which were carried out under aseptic conditions with flour as the only nonsterile component, could only have come from the flour used.     Yeast diversity of Ghanaian cocoa bean heap fermentations

    Abstract The fermentation of the Theobroma cacao beans, involving yeasts, lactic acid bacteria, and acetic acid bacteria, has a major influence on the quality of the resulting cocoa. An assessment of the microbial community of cocoa bean heap fermentations in Ghana resulted in 91 yeast isolates. These were grouped by PCR-fingerprinting with the primer M13. Representative isolates were identified using the D1/D2 region of the large subunit rRNA gene, internal transcribed spacer sequences and partial actin gene sequences leading to the detection of 15 species. Properties of importance for cocoa bean fermentation, namely sucrose, glucose, and citrate assimilation capacity, pH-, ethanol-, and heat-tolerance, were examined for selected isolates. Pichia kudriavzevii (Issatchenkia orientalis), Saccharomyces cerevisiae, and Hanseniaspora opuntiae formed the major components of the yeast community. Hanseniaspora opuntiae was identified conclusively for the first time from cocoa fermentations. Among the less frequently encountered species, Candida carpophila, Candida orthopsilosis, Kodamaea ohmeri, Meyerozyma (Pichia) caribbica, Pichia manshurica, Saccharomycodes ludwigii, and Yamadazyma (Pichia) mexicana were not yet documented from this substrate. Hanseniaspora opuntiae was preferably growing during the earlier phase of fermentation, reflecting its tolerance to low pH and its citrate-negative phenotype, while no specific temporal distribution was recognized for P. kudriavzevii and S. cerevisiae.     IT project managers' construction of successful project management practice: a repertory grid investigation

    Abstract Although effective project management is critical to the success of information technology (IT) projects, little empirical research has investigated skill requirements for IT project managers (PMs). This study addressed this gap by asking 19 practicing IT PMs to describe the skills that successful IT PMs exhibit. A semi-structured interview method known as the repertory grid (RepGrid) technique was used to elicit these skills. Nine skill categories emerged: client management, communication, general management, leadership, personal integrity, planning and control, problem solving, systems development and team development. Our study complements existing research by providing a richer understanding of several skills that were narrowly defined (client management, planning and control, and problem solving) and by introducing two new skill categories that had not been previously discussed (personal integrity and team development). Analysis of the individual RepGrids revealed four distinct ways in which study participants combined skill categories to form archetypes of effective IT PMs. We describe these four IT PM archetypes , General Manager, Problem Solver, Client Representative and Balanced Manager , and discuss how this knowledge can be useful for practitioners, researchers and educators.     A common European foreign policy after Iraq?

    Taking as read the wide range of other instruments that the EU has for international influence (enlargement, aid, trade, association and other arrangements, etc.), the Common Foreign and Security Policy (CFSP), under pressure from the Kosovo conflict, has been shaped by two important decisions in 1999: the creation of the European Security and Defence Policy (ESDP) to give the EU a military capability when NATO as a whole is not engaged, and the appointment as the new High Representative for the CFSP of a high-profile international statesman rather than a senior civil servant. A major European effort will still be needed if Europe is to be effective militarily, whether in the EU/ESDP or NATO framework. The management of the CFSP has been held back by the doctrine of the equality of all member states regardless of their actual contribution. This in turn leads to a disconnect between theory (policy run by committee in Brussels) and practice (policy run by the High Representative working with particular member states and other actors, notably the US). It has been difficult for Javier Solana to develop the authority to do this, not in competition with the Commission as so widely and mistakenly believed, as with member states themselves, and particularly successive rotating presidencies. It is important that misdiagnosis does not lead to politically correct solutions that end up with the cure worse than the disease. Ways need to be found to assure to the High Representative the authority to work with third countries and with the member states making the real contribution, while retaining the support of all. Then, with its own military capability, the EU can have a CFSP that is the highest common factor rather than the lowest common denominator, with member states ready to attach enough priority to the need for common policies to give Europeans a strong influence in the big foreign policy issues of the day.     A comparison of two pre-enrichment media prior to immunomagnetic separation for the isolation of E. coli O157 from bovine faeces

    Abstract Aims: To compare the sensitivity of two pre-enrichment broth media prior to immunomagnetic separation for the isolation of Escherichia coli O157 from cattle faeces. Methods and Results: One-gram portions of 721 cattle faeces collected from 43 farms were pre-enriched in buffered peptone water containing vancomycin, cefixime and cefsulodin (BPW-VCC) and buffered peptone water without additives (BPW-WOA), respectively. A total of 137 samples were positive for E. coli O157: 127 pre-enriched with BPW-WOA and 89 pre-enriched in BPW-VCC. Representative isolates were tested for phage type, verotoxin and eae (E. coli attaching and effacing) gene sequences, resulting in the recognition of eight different types. All the E. coli O157 types recognized were isolated by both methods except for three different strains, each of which were isolated only on a single occasion: two by BPW-WOA and another by BPW-VCC. Conclusions: The results clearly demonstrate, under the conditions of this study, that BPW without antibiotics was the superior pre-enrichment medium for the isolation of E. coli O157 from cattle faeces. Significance and Impact of the Study: The use of BPW-WOA in preference to BPW-VCC for the isolation of E. coli O157 from cattle faeces in future research and outbreak studies should lead to a higher number of positive isolates.     Continuity and Change in Personality Traits From Adolescence to Midlife: A 25-Year Longitudinal Study Comparing Representative and Adjudicated Men

    The second study examined structural, rank-order, and mean-level continuity. Partial structural continuity was demonstrated through confirmatory factor analysis. Regarding rank-order continuity, the correlations were stronger as age increased, particularly for the adjudicated men. For mean-level continuity, the adjudicated men displayed higher scores from adolescence to midlife for nearly every personality trait related to Disinhibition and Negative Emotionality. Significant decreases were observed in these traits for both samples, supporting the hypothesis of a normative psychological maturation. Although both samples showed this maturation, the adjudicated men displayed a lower rate of change during adolescence and early adulthood. The two samples did not differ in Extraversion and this trait remained more stable, particularly for adjudicated men. [source]


    Clonal groups of high-level gentamicin-resistant Enterococcus faecium isolated from municipal wastewater and clinical samples in Tehran, Iran

    Abstract Aims:, Clonality among high-level gentamicin-resistant Enterococcus faecium (HLGR-EF) isolates obtained from clinical and sewage treatment plants (STP) were investigated using PhePlate system (PhP), ribotyping and pulsed-field gel electrophoresis (PFGE). Methods and Results:, During 1 year study (September 2005,2006), a total of 106 HLGR-EF isolates were collected from clinical (n = 48) and STP (n = 58) samples in Tehran, Iran. Biochemical fingerprinting of these isolates using the PhP showed the presence of 21 PhP types (diversity index, Di = 0·97) among the clinical and 21 PhP types (Di = 0·91) among the STP isolates. Representative isolates of each PhP type (n = 42) were further characterized by the ribotyping method. Sixteen ribotypes were identified among the isolates with five types shared between the clinical and STP isolates. PFGE recognized 24 clonal types among these isolates with three pulsotypes shared between the clinical and STP isolates. Combination of the two techniques (PFGE and ribotyping) resulted in 24 (Di = 0·96) and 16 (Di = 0·93) types among the strains isolated from clinical and STP samples, respectively. Conclusions:, We concluded that the combination of PhP typing, ribotyping and PFGE could be extremely discriminatory when examining HLGR-EF isolates. Significance and Impact of the Study:, The emergence of highly diverse HLGR-EF population in Iran is of serious concern especially because of their multi-resistances. [source]


    Revisiting reliability of quantified perineal ultrasound: Bland and Altman analysis of a new protocol for the rectangular coordinate method

    Abstract Aims This study tested the reliability of a new protocol for the rectangular coordinate method of quantifying perineal ultrasound. Methods Representative scans of healthy primiparous females were quantified by positioning a pubic bone template, drawn onto an acetate sheet containing x,y axes, over scans, by aligning the x-axis with the pubic bone central axis. Values for x (Dx) and y (Dy) located the urethrovesical junction (UVJ) at Rest, and at maximal Valsalva and Kegel. Range of motion (V,K) was calculated. Bland and Altman analysis, correlations, and t -tests determined intra- and inter-rater reliability, and variance due to designation of the pubic bone central axis (template control). Results Correlations averaged 0.72, 0.70, and 0.92 for intra-rater, inter-rater, and template control experiments. Dx Rest, Dx Kegel, and V,K were reliable in all experiments. First and second measures for inter-rater Dy Rest and Dy Kegel, and template control Dy Valsalva were significantly different. Bland and Altman analysis showed Dy Rest, Dy Kegel, and Dx and Dy Valsalva for both reliability experiments to have limits of agreement (LOA's) large enough to explain ,50% of the actual value ranges. Template control LOA's explained ,30% of the actual value ranges. Conclusions The reliability of this protocol varied according to the conditions analyzed; accurate reliability assessment of all conditions required Bland and Altman analysis; and the designation of the pubic bone central axis remained a source of variance between investigators.     Characterization of chickpea differentials for pathogenicity assay of ascochyta blight and identification of chickpea accessions resistant to Didymella rabiei

    Forty-eight chickpea germplasm lines, including 22 differentials used in previous studies, were characterized for disease phenotypes following inoculation with six isolates of Didymella (anamorph Ascochyta) rabiei, representing a wide spectrum of pathogenic variation. Representative isolates were also directly compared with six previously identified races on eight chickpea genotypes. Many of the chickpea differentials reacted similarly to inoculation with each isolate of D. rabiei, and several previously identified races caused similar levels of disease on the differentials. This indicates that the number of differentials can be reduced significantly without sacrificing accuracy in describing pathogenic variation of D. rabiei on chickpea. Pathogenic variation among samples of US isolates allowed classification of the isolates into two pathotypes. The distribution of disease phenotypes of the 48 germplasm lines was bimodal after inoculation with pathotype I isolates, whereas the distribution of disease phenotypes was continuous after inoculation with pathotype II isolates. Such distinct distribution patterns suggest that chickpea plants employ different resistance mechanisms to each pathotype and that the two pathotypes may have different genetic mechanisms controlling pathogenicity. The advantages of using the two-pathotype system in assaying pathogenicity of the pathogen and in studying resistance mechanisms of the host are discussed. Three chickpea accessions, PI 559361, PI 559363 and W6 22589, showed a high level of resistance to both pathotypes, and can be employed as resistance sources in chickpea breeding programmes for resistance to ascochyta blight.     Abstract We have chosen the silver arowana (Osteoglossum bicirrhosum), a representative of the most ancient teleost family Osteoglossidae, to address the question of yolk nutrients utilization. Silver arowana have particularly large eggs (1,1.5 cm of diameter) and a unique morphology of the yolk. We present evidence that the yolk cytoplasmic zone (ycz) in the "yolksac juveniles" is a very complex structure involved in sequential processes of yolk hydrolysis, lipoprotein particles synthesis, their transport, and exocytosis. Vacuoles filled with yolk granules in different stages of digestion move from the vitellolysis zone through the ycz to be emptied into the microvillar interspace in the process of exocytosis. The area of the ycz with the abundance of the mitochondria must play an important role in providing energy for both the transport of vacuoles and the release of their contents. Therefore, we postulate that the function of yolk syncytial layer (ysl) as the "early embryonic patterning center" transforms in fish larvae or yolksac juveniles into a predominantly specialized role as the yolk trophoblastic layer (ytl) involved in yolk nutrients utilization. In addition to discovering the mechanism of transformation of the ysl function into ytl function, we suggest that the machinery involved in nutrient mobilization and exocytosis in yolk of arowana yolksac juveniles can be very attractive system for studies of regulatory processes in almost all secretory pathways in animal cells.     TRIPS-Plus Implications for Access to Medicines in Developing Countries: Lessons from Jordan,United States Free Trade Agreement

    Since the establishment of the World Trade Organization (WTO) in 1995 and implementation of the Agreement on Trade-Related Aspects of Intellectual Property Rights (TRIPS) as a result, the United States (US) sought to impose still higher levels of intellectual property rights on developing countries, a phenomenon that is commonly known today as TRIPS-Plus. The Jordan,US FTA, signed in 2001, contains several TRIPS-Plus rules that restrict the poor's access to medicines, and is today touted by US officials and the US Trade Representative (USTR) as a success, and providing a wide range of benefits. These benefits not only include a higher growth rate, but also more specific benefits to the pharmaceutical sector in particular, such as an improved ability to develop generic medicine and engage in new innovative research, as well as increasing the presence of and collaboration with multinational drug makers. This article analyzes in detail the TRIPS-Plus provisions of the Jordan,US FTA. It challenges the claims that the FTA brings general and specific benefits to developing countries, and provides fresh evidence which strongly suggests that benefits from the Jordan,US FTA have been largely exaggerated while the costs underestimated.     Responsible, Representative and Accountable Government

    As an evaluation of the health of Australia's political system, this article offers a perspective different from the lament over the loss of responsible government. It finds that responsible government is not compatible with representative democracy. Peculiar to Australia is conflict between ,responsible party government' and ,responsible parliamentary government'. Nevertheless, the system is healthy. A parliament-as-a-whole approach identifies key holistic functions of manifest and latent legitimation and accountability that bolster legitimacy. Political accountability is enhanced by the watchdog role of the media. Public accountability is enriched by the links between citizens and administrative review. Critical changes include the guarantee of senate independence and the removal of senate power over supply. These changes would confine the theory and practice of responsible government to the House of Representatives, promote accountability, and thus increase the legitimacy of Australian parliamentary democracy.     Hyperinvasive genotypes of Neisseria meningitidis in France

    Abstract Clinical isolates of Neisseria meningitidis from cases of meningococcal disease, collected between January 2000 and December 2004, were identified and typed at the French National Reference Centre. A representative subset of 546 isolates from among 2882 isolates was further genotyped by multilocus sequence typing to determine their genetic lineages (clonal complexes) and the degree of diversification among different clonal complexes. Representative isolates of the main clonal complexes were tested for their virulence in mice and for proapoptotic effects on human epithelial cells. High genetic diversity in some genetic lineages (ST-32 and ST-41/44) was correlated with heterogeneity in virulence in mice and proapoptotic effects on human epithelial cells. In contrast, the homogeneous genetic structure of isolates of the ST-11 clonal complex, regardless of their serogroup, correlated positively with a fatal outcome of the infection, increased virulence in mice and increased proapoptotic effects on human epithelial cells.     Representative marketing-oriented study on implants in the Austrian population.

    Abstract: The number of dental implants inserted annually worldwide has been estimated to come close to a million. But the level of information available to patients about realistic, evidence-based treatment options by implants is often enough more than fragmentary, and what is disseminated by the media and the industry does not always reflect evidence-based empirical data. This survey of 1000 adults presented with 18 questions was designed to shed light on several points. These were (1) level of subjective patient information, (2) sources of information and prejudices, (3) future demand for implant treatment and target groups for patient information campaigns, and (4) potential misinformation, information deficits, discrepancies of information and how these come about. Of those questioned, 20% said unprompted that implants were a possibility to replace missing teeth. When prompted, 72% said that they knew about dental implants. Most of those questioned felt poorly informed about the options for replacing missing teeth and many knew less about implants than about other alternatives. The dentist was said to be the desired source of information, but 77% of those questioned reported that their dentists did not practice implant dentistry. More than 79% of those questioned did not know whether their dentist worked with implants. Forty-four percent thought that implants should only be placed by specially trained doctors. Sixty-one percent were of the opinion that dentists who provide implant dentistry were better qualified than their nonimplanting colleagues. Half of those questioned attributed implant failures to allergies and incompatibilities, the other half to poor medical care. Only 29% incriminated poor oral hygiene as a cause of implant failure. Future strategies should be geared to more professional public relations and patient information. Internationally operating qualified implant institutions could contribute much to balance discrepant information. Mais le niveau d'information disponible du patient en ce qui concerne les options de traitement basées sur l'évidence est souvent fragmentaire, et l'information dispensée par les média et l'industrie ne reflète pas toujours ces options. Cette enquête réalisée sur mille adultes ayant reçu un questionnaire de 18 questions a été effectuée afin de mettre en évidence certains points. Ces derniers étaient 1) le niveau de l'information subjective du patient, 2) les sources d'information et préjudices, 3) la demande future pour le traitement implantaire et les groupes cibles pour les campagnes d'information des patients, 4) les mauvaises informations potentielles, les déficits d'information, les désaccords de l'information et les causes de ces derniers. De ces personnes questionnées, 20% ont répondu spontanément que les implants étaient un moyen de remplacer les dents manquantes. Lorsqu'ils étaient interrogés, 72% ont répondu connaître les implants dentaires. La plupart de ces personnes se sentaient cependant peu informées sur les options de remplacement des dents manquantes et beaucoup d'entre-elles possédaient très peu d'information sur les implants par rapport aux autres possibilités existantes. Le dentiste semblait être la source préférentielle d'information mais 77 % des personnes questionnées ont rapporté que leur dentiste ne plaçait pas d'implant. Plus de 79% ne savaient pas si leur dentiste pratiquait l'implantologie. Quarante-quatre pour cent étaient persuadés que les implants ne pouvaient être placés que par des spécialistes. Aber die den Patienten zur Verfügung stehende Information über realistische, auf Evidenz basierende Behandlungsmöglichkeiten mit Implantaten ist mehr als lückenhaft. Was durch die Medien und durch die Industrie verbreitet wird, wiederspiegelt nicht immer auf Evidenz basierende empirische Daten. Diese Untersuchung an 1000 Erwachsenen, welchen 18 Fragen gestellt wurden, wurde entwickelt, um Licht auf verschiedene Punkte zu werfen. Die Punkte betrafen (1) Stand der subjektiven Patienteninformation; (2) Informationsquellen und Vorurteile; (3) zukünftiger Bedarf an Implantatbehandlungen und Zielgruppen für Propaganda zur Patienteninformation; (4) potentielle Fehlinformation, Informationsdefizite, Diskrepanzen in der Information und wie diese zustande kommen.     Representative marketing-oriented study on implants in the Austrian population.

    Abstract: Oral implantology is an established subspecialty of restorative dental and oral surgery. While an extensive body of evidence on the fundamentals of osseointegration and associated factors has been published, marketing-oriented analyses based on representative public opinion polls of implant acceptance, patient-perceived cost and patient satisfaction are scarce. In this study, an attempt was made to address these points by questioning a representative sample of 1000 adults in the household setting. The interviewees were presented with 14 questions. Of those familiar with implants as one of the treatment alternatives, 61% reported they would accept implants if the need arose. Implant acceptance was highest among males and interviewees below the age of 30 years. The interest in implants increased with increasing family incomes. Four percent of those questioned already had implants. Twenty-five percent knew someone who had undergone implant treatment. All those questioned found implant-supported rehabilitation to be very expensive. Many of them blamed the dentists for the high cost. One detail was particularly evident: satisfaction among implanted patients was clearly higher than satisfaction rates perceived by them from what they were told about implants by others. First-hand experiences with implants proved to be less biased than reported second-hand information. Tandis que la proportion d'évidences s'accroît en ce qui concerne la recherche sur l'ostéoïntégration et les facteurs associés, les analyses orientées sur le marketing et l'opinion du public sur l'acceptation de l'implant, le prix et la satisfaction se font rares. In dieser Studie wird der Versuch unternommen, diese Punkte mit Hilfe einer Befragung einer repräsentativen Gruppe von 1000 Erwachsenen anzusprechen. Den Befragten wurden 14 Fragen gestellt. Von denen, welche über Implantate als Behandlungsalternative Bescheid wussten, gaben 61% an, sie würden Implantate bei sich akzeptieren, falls die Notwendigkeit dafür besteht.     We examine the major interest groups in the debate over allowing the re-importation of prescription drugs by utilizing a logit model and instrumental variables. Consistent with political support approach, the evidence suggests that Representatives are maximizing their electoral prospects: contributions from pharmaceutical manufacturers shrink the probability of voting for the bill; and Representatives are sensitive to their constituencies , employees of pharmaceutical manufacturing and senior citizens. Representatives' gender and ideology regarding free trade and subsidies are also determining factors. However, the decision was, by and large, a partisan one: party affiliation was the most important factor in passing the bill. [source]


    Comparison of dental licensure, specialization and continuing education in five countries

    Dental practice and education are becoming more globalized. Greater practitioner and patient mobility, the free flow of information, increasingly global standards of care and new legal and economic frameworks (such as European Union [EU] legislation) are forcing a review of dental licensure, specialization and continuing education systems. The objective of this study was to compare these systems in Canada, France, Germany, the UK and the US. Representatives from the five countries completed a 29-item questionnaire, and the information was collated and summarized qualitatively. Statutory bodies are responsible for licensing and re-licensing in all countries. In the two North American countries, this responsibility rests with individual states, and in Europe, with the countries themselves, mainly governed by the legal framework of the EU. In some countries, re-licensure requires completion of continuing education credits. Approaches to dental specialization tend to differ widely with regard to definition of specialities, course and duration of training, training facilities, and accreditation of training programmes. In most countries, continuing education is provided by a number of different entities, such as universities, dental associations, companies, institutes and private individuals. Accreditation and recognition of continuing education is primarily process-driven, not outcome-orientated. Working towards a global infrastructure for dental licensing, specialization and continuing education depends on a thorough understanding of the international commonalities and differences identified in this article.     Horizontal Coherence and the External Competences of the European Union

    First considered as a political requirement, its best expression is located in the Article 3 of the Treaty on the European Union. The practice of European foreign policy has demonstrated the importance of this requirement: the control of exports of dual-use goods as well as the adoption of sanctions, whether on the basis of Article 301 EC, or in application of international agreements with third states, have given the example of overlapping competences. Besides, this overlap has been reflected in the internal organisation of the institutions (allocation of portfolios in the European Commission, conflict between the Political Committee and the Committee of the Permanent Representatives in the Council of the EU). However, the answers to these problems have been far too timid: the adoption of an integrated approach (conflict prevention) as well as the institutional adaptations of the Treaty of Amsterdam do not compensate for the absence of a vision of the European foreign policy which would overcome the old cleavage between federalism and intergovernmentalism. Indeed, it seems to us that much more innovative solutions are needed, such as an evolution towards the binding character of the coherence requirement, which would pave the way to a coherent European foreign policy, comprising external relations and CFSP (including the defence dimension).     EVOLUTION OF TEMPORAL ISOLATION IN THE WILD: GENETIC DIVERGENCE IN TIMING OF MIGRATION AND BREEDING BY INTRODUCED CHINOOK SALMON POPULATIONS

    Abstract. The timing of migration and breeding are key life-history traits; they are not only adaptations of populations to their environments, but can serve to increase reproductive isolation, facilitating further divergence among populations. As part of a study of divergence of chinook salmon, Oncorhynchus tshawytscha, populations, established in New Zealand from a common source in the early 1900s, we tested the hypotheses that the timing of migration and breeding are under genetic control and that the populations genetically differ in these traits despite phenotypic overlap in timing in the wild. Representatives of families from two populations were collected within a day or two of each other, reared in a common environment, and then released to sea from each of two different rivers, while other family representatives were retained in fresh water to maturity. The date of maturation of fish held in fresh water and the dates of return from the ocean and maturation of fish released to sea all showed significant differences between the two populations and among families within populations. The very high heritabilities and genetic correlations estimated for migration and maturation date indicated that these traits would respond rapidly to selection. Combined with the results of related studies on these chinook salmon populations, it appears that spawning time may not only evolve during the initial phases of divergence, but it may play an important role in accelerating divergence in other traits.     Patient and clinician collaboration in the design of a national randomized breast cancer trial

    Abstract Objective, To show breast cancer patient involvement in the design of a national randomized trial of hormone replacement therapy (HRT) in symptomatic patients will increase accrual. Setting and participants, Three stakeholder groups [(1) researchers from the Lynda Jackson Macmillan Centre, (2) the Consumers' Advisory Group for Clinical Trials (CAG-CT), (3) clinicians responsible for a pilot randomized HRT study in breast cancer patients] developed this collaborative study. Methods, (1) Nine focus group discussions were conducted to identify issues relevant to breast cancer patients about HRT and a national trial: six involved women from breast cancer support groups nationwide and three patients who had previously participated in the pilot randomized HRT study. (2) Recommendations from the focus groups (analysed by Grounded Theory) were debated by the research stakeholders and focus group representatives at a 1-day meeting and consensus reached (using a voting system) on mutual priorities for incorporation into the design of a national HRT trial. (3) Representatives from the CAG-CT and focus groups participated in subsequent national HRT steering committee meetings to ensure that these priorities were accounted for and the resulting trial design summary was circulated to the CAG-CT and all focus group representatives for comment. Results, Focus groups demonstrated that the complexity of factors relating to trial participation was not just restricted to the research topic in question. Patient,clinician interaction provided a platform for negotiating potential conflicts over trial design and outcomes. Patient feedback suggested that mutually agreed priorities were accounted for in the trial design. Interpretation, Clinical research planning should involve all research stakeholders at the outset. Quantifying the impact of patient involvement in terms of trial accrual may be too simple given the complexity of their motivations for participating in trials. [source]

    Molecular phylogenetic evidence for an extracellular Cu Zn superoxide dismutase gene in insects

    Abstract Representatives of three ancient gene families of the antioxidant enzyme superoxide dismutase (SOD) can be found in most metazoans. In mammals and Caenorhabditis elegans, there is at least one gene each of the cytoplasmic, mitochondrial and extracellular lineages of SOD genes. The cytoplasmic SOD was one of the first enzymes to be implicated in ageing due to its protection against damaging oxygen free radicals. In contrast to other metazoans, insects were thought to lack a gene for the extracellular SOD. We have cloned and sequenced an SOD mRNA in the ant Lasius niger that appears to belong to this extracellular family. Subsequent searches and analyses of SOD gene sequences in insect databases revealed that insects do indeed express all three SOD genes including the extracellular form. We conclude that insects as well as other metazoans appear to have the full repertoire of the three families of SOD. [source]


    Cotylea (Polycladida): a cladistic analysis of morphology

    Abstract. Polyclad flatworms are acoelomate bilaterians found in benthic communities worldwide, predominantly in marine environments. Current polyclad systematics is unstable, with two non-concordant classification schemes resulting in a poor understanding of within-group relationships. Here we present the first phylogenetic framework for the suborder Cotylea using a morphological matrix. Representatives of 34 genera distributed among all cotylean families (except four, excluded due to their dubious taxonomic status) were investigated. The number of families included ranges from a conservative eight to a revisionary 11. Outgroup analysis indicated that the suborder is monophyletic and defined by the presence of a ventral adhesive structure, a short posteriorly positioned vagina, and cement glands. Of the eight to 11 families included, we confirmed that three were monophyletic: Boniniidae, Prosthiostomidae, and Pseudocerotidae. Boniniidae was consistently recovered as the sister group to other Cotylea, based on the retention of the plesiomorphic Lang's vesicle. The clade consisting of Anonymus, Marcusia, and Pericelis is sister to the Boniniidae and the rest of the Cotylea. Above this clade there is little resolution at the base of the sister group. The Euryleptidae are found to be paraphyletic and give rise to the Pseudocerotidae. Neither classification scheme received unequivocal support. The intrafamilial relationships of the diverse Pseudocerotidae and Euryleptidae were examined. Color pattern characters (used for species identification) were highly homoplasious but increased cladogram resolution within genera. The monophyly of seven genera within the Pseudocerotidae and Euryleptidae was not supported and many genera showed no autapomorphies, highlighting the need for taxonomic revision of these families.
