Frequent occurrence of high affinity T cells against MELOE-1 makes this antigen an attractive target for melanoma immunotherapy

Abstract We recently showed that the infusion of tumor infiltrating lymphocytes specific for the MELOE-1 antigen was associated with a prolonged relapse-free survival for HLA-A2+ melanoma patients who received tumor infiltrating lymphocytes therapy. Here, we characterized the MELOE-1/A2-specific T-cell repertoire in healthy donors and melanoma patients to further support an immunotherapy targeting this epitope. Using tetramer enrichment followed by multicolor staining, we found that MELOE-1-specific T cells were present in the blood of healthy donors and patients at similar frequencies (around 1 in 1×105 CD8+ cells). These cells mainly displayed a naïve phenotype in 4/6 healthy donors and 3/6 patients, whereas high proportions of memory cells were observed in the remaining individuals of both groups. There was a recurrent usage of the V,12.1 chain for 17/18 MELOE-1-specific T-cell clones derived from healthy donors or patients, associated with diverse V, chains and V(D)J junctional sequences. All clones derived from melanoma patients (9/9) were reactive against the MELOE-136,44 peptide and against HLA-A2+ melanoma cell lines. This study documents the existence of a large TCR repertoire specific for the MELOE-1/A2 epitope and its capacity to give rise to antitumor CTL that supports the development of immunotherapies targeting this epitope. Alcohol Drinking Patterns and Health Care Utilization in a Managed Care Organization

Objective. To estimate the relationship between current drinking patterns and health care utilization over the previous two years in a managed care organization (MCO) among individuals who were screened for their alcohol use. Study Design. Three primary care clinics at a large western MCO administered a short health and lifestyle questionnaire to all adult patients on their first visit to the clinic from March 1998 through December 1998. Patients who exceeded the National Institute on Alcohol Abuse and Alcoholism (NIAAA) guidelines for moderate drinking were given a more comprehensive alcohol screening using a modified version of the Alcohol Use Disorders Identification Test (AUDIT). Health care encounter data for two years preceding the screening visit were linked to the remaining individuals who responded to one or both instruments. Using both quantity,frequency and AUDIT-based drinking pattern variables, we estimated negative binomial models of the relationship between drinking patterns and days of health care use, controlling for demographic characteristics and other variables. Principal Findings. For both the quantity,frequency and AUDIT-based drinking pattern variables, current alcohol use is generally associated with less health care utilization relative to abstainers. This relationship holds even for heavier drinkers, although the differences are not always statistically significant. With some exceptions, the overall trend is that more extensive drinking patterns are associated with lower health care use. Conclusions. Based on our sample, we find little evidence that alcohol use is associated with increased health care utilization. On the contrary, we find that alcohol use is generally associated with decreased health care utilization regardless of drinking pattern. [source]


Feeding habits and trophic morphology of inshore lizardfish (Synodus foetens) on the central continental shelf off Veracruz, Gulf of Mexico

Summary The inshore lizardfish, Synodus foetens, is one of the most common coastal demersal predators on the continental shelf of the Gulf of Mexico (GOM), but the biology and feeding ecology of this species is virtually unknown. Between November 2001 and January 2003 (10 collections), 603 individuals of S. foetens ranging from 112 to 420 mm standard length (SL) and 13 to 630 g (wet weight) were collected from the continental shelf of Alvarado, Veracruz, Mexico. About 60% of the individuals had empty stomachs with the stomach fullness of the remaining individuals being distributed as follows: 5% full (24.8%), 50,75% full (13.5%), and completely full (1.7%). The mean (± SD) wet weight of stomach contents was 12.1 ± 10.8 g during the rainy season, and 19.0 ± 13.0 g during the nortes season. Seventeen prey items were identified, with the majority thereof being fish. The most important prey items were Upeneus parvus, Loligo pealei, Engyophrys senta, Trachurus lathami, and Anchoa hepsetus. Seasonal changes in the diet were observed, with U. parvus and L. pealei being the most important prey during the nortes season, whereas E. senta and L. pealei were the main items during the rainy season. Prey size selection was evident among size classes of S. foetens, although no trophic overlap was observed among size classes (Cik , 0.004). High trophic level values were determined for all size classes by season (rainy season = 4.67, nortes season = 4.84), and indicated this species belongs to the piscivorous trophic guild that preys upon both demersal and pelagic species. Synchrony in the downstream migration of smolts and upstream migration of adult Atlantic salmon in the subarctic River Utsjoki

Seasonal and diel migration timing of wild Atlantic salmon Salmo salar smolts and adults were investigated annually (2001,2004) in the subarctic River Utsjoki, a tributary of the large River Teno (70° N), using underwater video monitoring. Submerged video cameras provided information on the exact timing and intensity of both migrations in a natural river channel, without disturbing the fish. In contrast to the mainly nocturnal migration pattern described from temperate rivers, the River Utsjoki smolts and adults migrated throughout the day. Smolts migrated most intensively during hours of rising (0300,0900 hours) and high sun (0900,1500 hours), while adults favoured the period of low sun (2100,0300 hours). Smolt migrations started in June and lasted on average 42 days. Adults usually ascended the site 2,3 weeks before the first descending smolts were observed and the adult migrations extended to the end of August. Seasonal synchrony was observed between smolt and adult migrations in years of slowly warming water, whereas in a year of exceptionally warm early summer (2002), smolts migrated earlier than adults. Thus, water temperature seemed to be an important environmental factor triggering the smolt migration, while the migration of adults was probably more fixed to a certain season. Weak positive correlations between fish counts and water temperature were observed, indicating that increasing water temperature may have promoted both smolt and adult migrations. The influence of discharge was negligible, although increasing discharge late in the season may have activated the remaining individuals in both groups. [source]
