Myosin diversity in the diatom Phaeodactylum tricornutum,

Abstract This report describes the domain architecture of ten myosins cloned from the pennate diatom Phaeodactylum tricornutum. Several of the P. tricornutum myosins show similarity to myosins from the centric diatom Thalassiosira pseudonana as well as to one myosin from the oomycete Phytophthora ramorum. The P. tricornutum myosins, ranging in size from 126 kDa to over 250 kDa, all possess the canonical head, neck and tail domains common to most myosins, though variations in each of these domains is evident. Among the features distinguishing several of the diatom myosin head domains are N-terminal SH3-like domains, variations in or near the P-loop and Loop 1 regions close to the nucleotide binding pocket, and extended converter domains. Variations in the length of the neck domain or lever arm, defined by the light chain-binding IQ motifs, are apparent with the different diatom myosins predicted to contain from one to nine IQ motifs. Protein domains found within the P. tricornutum myosin tails include regions of coiled-coil structure, ankyrin repeats, CBS domain pairs, a PB1 domain, a kinase domain and a FYVE-finger motif. As many of these features have never before been characterized in myosins of any type, it is likely that these new diatom myosins will expand the repertoire of known myosin behaviors. © 2010 Wiley-Liss, Inc. Centrioles to basal bodies in the spermiogenesis of Mastotermes darwiniensis (Insecta, Isoptera)

Abstract In addition to their role in centrosome organization, the centrioles have another distinct function as basal bodies for the formation of cilia and flagella. Centriole duplication has been reported to require two alternate assembly pathways: template or de novo. Since spermiogenesis in the termite Mastotermes darwiniensis lead to the formation of multiflagellate sperm, this process represents a useful model system in which to follow basal body formation and flagella assembly. We present evidence of a possible de novo pathway for basal body formation in the differentiating germ cell. This cell also contains typical centrosomal proteins, such as centrosomin, pericentrin-like protein, ,-tubulin, that undergo redistribution as spermatid differentiation proceeds. The spermatid centrioles are long structures formed by nine doublet rather than triplet microtubules provided with short projections extending towards the surrounding cytoplasm and with links between doublets. The sperm basal bodies are aligned in parallel beneath the nucleus. They consist of long regions close to the nucleus showing nine doublets in a cartwheel array devoid of any projections; on the contrary, the short region close to the plasma membrane, where the sperm flagella emerge, is characterized by projections similar to those observed in the centrioles linking the basal body to the plasma membrane. It is hypothesized that this appearance is in connection with the centriole elongation and further with the flagellar axonemal organization. Involvement of the thalamocortical network in TLE with and without mesiotemporal sclerosis

Summary Purpose:, The thalamus plays an important role in seizure propagation in temporal lobe epilepsy (TLE). This study investigated how structural abnormalities in the focus, ipsilateral thalamus and extrafocal cortical structures relate to each other in TLE with mesiotemporal sclerosis (TLE-MTS) and without hippocampal sclerosis (TLE-no). Methods:, T1 and high-resolution T2 images were acquired on a 4T magnet in 29 controls, 15 TLE-MTS cases, and 14 TLE-no. Thalamus volumes were obtained by warping a labeled atlas onto each subject's brain. Deformation-based morphometry was used to identify regions of thalamic volume loss and FreeSurfer for cortical thickness measurements. CA1 volumes were obtained from high-resolution T2 images. Multiple regression analysis and correlation analyses for voxel- and vertex-based analyses were performed in SPM2 and FreeSurfer. Results:, TLE-MTS had bilateral volume loss in the anterior thalamus, which was correlated with CA1 volume and cortical thinning in the mesiotemporal lobe. TLE-no had less severe volume loss in the dorsal lateral nucleus, which was correlated with thinning in the mesiotemporal region but not with extratemporal thinning. Discussion:, The findings suggest that seizure propagation from the presumed epileptogenic focus or regions close to it into the thalamus occurs in TLE-MTS and TLE-no and results in circumscribed neuronal loss in the thalamus. However, seizure spread beyond the thalamus seems not to be responsible for the extensive extratemporal cortical abnormalities in TLE. [source]


Mechanisms of particle dispersion in a turbulent, square duct flow

Abstract Particle dispersion in a square duct flow is studied using large eddy simulation combined with Lagrangian particle tracking under conditions of one-way coupling. The flow has a bulk Re = 250 k, with six particle sizes ranging from 5 to 1000 ,m. Results obtained for the fluid phase show good agreement with experimental data. For particles, predictions demonstrate that secondary flows within the duct dominate small particle dispersion and result in a uniform distribution, whereas gravity promotes the deposition of large particles on the duct floor. For the largest particles, the secondary flows contribute to particle concentration in corners on the duct floor, with these particles also clustering in low-velocity regions close to the floor. A detailed analysis of the influence of the flow on particle distribution is provided through consideration of the particle dispersion function, with the mechanisms of particle dispersion elucidated using a dynamical analysis. © 2009 American Institute of Chemical Engineers AIChE J, 2009 [source]


Bipolar Fatigue Caused by Field Screening in Pb(Zr,Ti)O3 Ceramics

Bipolar cycling of lead zirconate titanate ceramics can lead to massive material damage in regions close to the electrode. The damaged region can be identified by color changes, and the microstructure in this region shows signs of interface melting. This damaged region can screen the sample volume from the applied voltages and reduced fields are applied to the undamaged part of the sample. This has two effects. The first one is that the bulk is effectively subjected to smaller fields, but the measured parameters are assigned to the applied field, yielding apparent fatigue curves. The second effect is that with further cycling, field screening protects the bulk of the sample from fatigue due to the reduced effective fields. If the damaged region is mechanically removed and the ferroelectric hystereses are measured again, nearly unfatigued parameters are obtained. [source]


Inferring X-ray coronal structures from Zeeman,Doppler images

Abstract We have modelled the X-ray emission from the young rapid rotator AB Doradus (Prot= 0.514 d) using as a basis Zeeman,Doppler maps of the surface magnetic field. This allows us to reconcile the apparently conflicting observations of a high X-ray emission measure and coronal density with a low rotational modulation in the X-ray band. The technique is to extrapolate the coronal field from the surface maps by assuming the field to be potential. We then determine the coronal density for an isothermal corona by solving hydrostatic equilibrium along each field line and scaling the surface plasma pressure with the surface magnetic pressure. We set the density to zero along those field lines that are open and those where at any point along their length the plasma pressure exceeds the magnetic pressure. We then calculate the optically thin X-ray emission measure and rotational modulation for models with a range of coronal densities. Although the corona can be very extended, much of the emission comes from high-latitude regions close to the stellar surface. Since these are always in view as the star rotates, there is little rotational modulation. We find that emission measures in the observed range 1052.8,1053.3 cm,3 can be reproduced with densities in the range 109,1010.7 cm,3 for coronae at temperatures of 106,107 K. [source]


Generation of a flanking sequence-tag database for activation-tagging lines in japonica rice

Summary We have generated 47 932 T-DNA tag lines in japonica rice using activation-tagging vectors that contain tetramerized 35S enhancer sequences. To facilitate use of those lines, we isolated the genomic sequences flanking the inserted T-DNA via inverse polymerase chain reaction. For most of the lines, we performed four sets of amplifications using two different restriction enzymes toward both directions. In analyzing 41 234 lines, we obtained 27 621 flanking sequence tags (FSTs), among which 12 505 were integrated into genic regions and 15 116 into intergenic regions. Mapping of the FSTs on chromosomes revealed that T-DNA integration frequency was generally proportional to chromosome size. However, T-DNA insertions were non-uniformly distributed on each chromosome: higher at the distal ends and lower in regions close to the centromeres. In addition, several regions showed extreme peaks and valleys of insertion frequency, suggesting hot and cold spots for T-DNA integration. The density of insertion events was somewhat correlated with expressed, rather than predicted, gene density along each chromosome. Analyses of expression patterns near the inserted enhancer showed that at least half the test lines displayed greater expression of the tagged genes. Whereas in most of the increased lines expression patterns after activation were similar to those in the wild type, thereby maintaining the endogenous patterns, the remaining lines showed changes in expression in the activation tagged lines. In this case, ectopic expression was most frequently observed in mature leaves.
