Postendocytic Provitellin Processing in the Growing Oocyte of the Short Horned Grasshopper, Oxyajaponicajaponica (Orthoptera: Acrididae)

ABSTRACT Polyclonal antibodies made against 86 kDa (86 k), 80 kDa (80 k) and 54 kDa (54 k) vitellins of Oxya japonica japonica are used for Western blotting. Anti-80k vitellin antibody is cross-reacted with a 95 kDa (95 k) vitellin. While 95 k vitellin is present both in the female hemolymph and in the oocyte, 80 k vitellin is detected only in the oocyte and the laid egg. In the growing oocytes, as 95 k vitellin is faded out gradually, 80 k vitellin is accumulated increasingly, indicating postendocytic processing of 95 k vitellin brings 80 k vitellin. Further conforming the hypothesis, partial digestion of 95 k vitellin with pepsin and ,-chymotrypsin makes several protein bands of molecular weight around 80 kDa. Thus, the 95k vitellin may have a cleavage site (s) to produce 80 k vitellin which forms fairly stable tertiary structure. In the reduced condition (20 mM glutathion), both 95 k and 80 k vitellins were digested throughly by endogenous proteinase at pH 4. Both 86 k and 54 k vitellins, respectively, show no apparent molecular weight changes in the growing oocyte and in the hemolymph. [source]


Head structures of males of Strepsiptera (Hexapoda) with emphasis on basal splitting events within the order

Abstract Internal and external head structures of males of Strepsiptera were examined and the head of a species of Mengenilla is described in detail. The results suggest a reinterpretation of some structures. The head of basal extant strepsipterans is subprognathous, whereas it is strictly orthognathous in the groundplan of Strepsiptera s.l. The labrum and hypopharynx are not part of the mouthfield sclerite. The labial palps are absent in all strepsipterans. A very slightly modified mandibular articulation is preserved in Eoxenos, whereas it is distinctly reduced in other extant groups. A salivary duct, salivary glands, and a cephalic aorta are absent. The cladistic analysis of 44 characters of the head results in the following branching pattern: (Protoxenos + (Mengea + (Eoxenos + (Mengenilla [Austr.] + Mengenilla) + (Elenchus + Dundoxenos + Xenos + Stylops)))). Most apomorphies of males are associated with the necessity of finding females within a short time span and with a reduced necessity to consume food: large "raspberry" eyes, flabellate antennae with numerous dome-shaped chemoreceptors, Hofeneder's organ, an ovoid sensillum of the maxillary palp, and the simplified condition of the maxilla and the labium. Strepsiptera excl. Protoxenos are supported by the dorsomedian frontal impression, the dorsally shifted antennal insertions, a reduced number of antennal segments, absence of the galea, and probably by the presence of the mouthfield sclerite, which is a unique apomorphic feature. The balloon-gut combined with an unusual air-uptake apparatus is another possible autapomorphy of this clade. It is likely that the last common ancestor of Strepsiptera excl. Protoxenos did not process food. Strepsiptera s.str. are characterized by the strongly reduced condition of the labrum and the absence of the epistomal suture. Eoxenos is the sister group of the remaining Strepsiptera s.str. Synapomorphies of Mengenilla + Stylopidia are the advanced reduction of the mandibular articulation and the secondary absence of the ovoid sensillum. The monophyly of Mengenilla is confirmed, even though a small free labrum is present in Australian species. Derived features of Stylopidia are the absence of the coronal suture and the reduced condition of the frontal suture. Apomorphies that have evolved within Stylopidia are the membranization of parts of the head, the fusion of antennal segments, the increase or decrease of the number of flabellate flagellomeres, reductions and modifications of the mandibles, and modifications of the mouthfield sclerite. The monophyly of Stylopiformia is not unambiguously supported. A position of the mandibles posterior to the mouthfield sclerite (when adducted) is a possible synapomorphy shared by Xenos, Stylops, and other "higher Stylopidia." Effects of proline mutations in the major house dust mite allergen Der f 2 on IgE-binding and histamine-releasing activity

Der f 2 is the major group 2 allergen from house dust mite Dermatophagoides farinae and is composed of 129 amino-acid residues. Wild-type and six proline mutants of Der f 2 (P26A, P34A, P66A, P79A, P95A, and P99A) expressed in Escherichia coli were refolded and purified. Formations of intramolecular disulfide bonds in the purified proteins were confirmed correct. The apparent molecular masses analyzed by gel-filtration were 14,15 kDa. The IgE-binding capacity in the sera of seven mite-allergic patients, inhibitory activity for IgE-binding to immobilized wild-type Der f 2, and activity to stimulate peripheral blood basophils to release histamine in two volunteers were analyzed. P95A and P99A, which slightly differed from the wild-type Der f 2 in their CD spectrum, showed reduced IgE-binding, reduced inhibitory activity, and less histamine-releasing activity than the wild-type. P34A also showed reduced allergenicity. Considering that Pro95, Pro99 and Pro34 are closely located in loops at one end of the tertiary structure of Der f 2, we concluded that these loop regions included an IgE-binding site common to all tested patients. P66A showed reduced IgE-binding in two sera out of seven. P26A and P79A showed no reduced allergenicity. However, in immunoblot analysis after SDS/PAGE under reduced conditions, P79A showed no or markedly reduced IgE-binding while the other mutants showed IgE-binding corresponding to that in the assay using correctly refolded proteins. This suggests that Pro79 is involved in refolding of Der f 2. The findings in this study are important for the understanding of the antigenic structure of mite group 2 allergens and for manipulation of the allergens for specific immunotherapy. [source]


Geochemical characterization of moldavites from a new locality, the Cheb Basin, Czech Republic

Detailed comparison of the Cheb Basin moldavites with moldavites from other substrewn fields in both major and trace element composition shows that the Cheb Basin is a separate substrewn field. The geochemical data obtained are discussed with respect to the source materials and processes leading to formation of moldavites. The data show that three groups of Cheb Basin moldavites exist. Ten samples of group 1 are characterized by the lowest content of Al, Fe, Na, and other elements representing phyllosilicate minerals, and by high Ca + Mg contents related probably to carbonates. They resemble the "poisonous green" moldavites, a subgroup of the Southern Bohemian moldavites. Seven samples of group 2 and 6 samples of group 3 are similar to typical moldavites of the Southern Bohemian substrewn field. These two groups differ from each other mainly in Al contents; with higher contents of Al and the elements associated with phyllosilicate minerals (namely Ba and Sr), group 3 also resembles the Moravian moldavites. Significant positive correlations between K, Ca, Mg, and Mn found in group 2 of the Cheb Basin moldavites and the enrichment in these elements observed generally in all moldavites, as well as other facts, e.g., high K/Na and K/Rb ratios and the reduced conditions during formation of moldavites, have been attributed to possible contribution to the moldavite source materials of the ash produced by burning of vegetation and soil organic matter present at the pre-impact area.
