Selected AbstractsA case of WHIM syndrome associated with diabetes and hypothyroidismPEDIATRIC DIABETES, Issue 7 2009Junji Takaya Abstract: The WHIM syndrome is a rare immunological disorder characterized by warts, hypogammaglobulinemia, infections, and myelokathexis. We hypothesized that immunological or genetic mechanisms may link WHIM syndrome and type 1 diabetes. We report that the young girl with WHIM syndrome developed diabetes and transient hypothyroidism. A nonsense mutation (C,T) truncating the CXC chemokine receptor 4 (CXCR4) C-terminal cytoplasmic tail domain occurred at nucleotide position 1000(R334X) of the CXCR4 gene in one allele of the patient was identified, and the person was diagnosed as having WHIM syndrome. Recent observation suggested that the CXCR4, a G-protein-coupled receptor with a unique ligand, CXCL12, might be involved in the pathogenesis for type 1 diabetes. Taken into consideration the concurrent prevalence of the two disorders and the speculated common pathogenesis associated with the CXCR4, our patient may enable us to understand the genetic damage related to accelerated apoptosis. [source] Climate, climate change and range boundariesDIVERSITY AND DISTRIBUTIONS, Issue 3 2010Chris D. Thomas Abstract Aim, A major issue in ecology, biogeography, conservation biology and invasion biology is the extent to which climate, and hence climate change, contributes to the positions of species' range boundaries. Thirty years of rapid climate warming provides an excellent opportunity to test the hypothesis that climate acts as a major constraint on range boundaries, treating anthropogenic climate change as a large-scale experiment. Location, UK and global data, and literature. Methods, This article analyses the frequencies with which species have responded to climate change by shifting their range boundaries. It does not consider abundance or other changes. Results, For the majority of species, boundaries shifted in a direction that is concordant with being a response to climate change; 84% of all species have expanded in a polewards direction as the climate has warmed (for the best data available), which represents an excess of 68% of species after taking account of the fact that some species may shift in this direction for non-climatic reasons. Other data sets also show an excess of animal range boundaries expanding in the expected direction. Main conclusions, Climate is likely to contribute to the majority of terrestrial and freshwater range boundaries. This generalization excludes species that are endemic to specific islands, lakes, rivers and geological outcrops, although these local endemics are not immune from the effects of climate change. The observed shifts associated with recent climate change are likely to have been brought about through both direct and indirect (changes to species' interactions) effects of climate; indirect effects are discussed in relation to laboratory experiments and invasive species. Recent observations of range boundary shifts are consistent with the hypothesis that climate contributes to, but is not the sole determinant of, the position of the range boundaries of the majority of terrestrial animal species. [source] Distribution of erosion across bedrock channelsEARTH SURFACE PROCESSES AND LANDFORMS, Issue 3 2008Jens M. Turowski Abstract Lateral erosion in bedrock rivers is an important control on the shape of channel cross-sections, and the coupling of channels and hillslopes. Recent observations link lateral erosion to the variability of flow. We propose two mechanisms to explain this. One is based on changing shear stress distributions within the channel with varying flood level, the other on the competition between cover and tool effects in fluvial bedrock erosion. We assess these processes for the Liwu River, Taiwan, and conclude that cover and tool effects dominate the partitioning of lateral and vertical erosion in this case. [source] Comparative study on antibodies to human and bacterial 60 kDa heat shock proteins in a large cohort of patients with coronary heart disease and healthy subjectsEUROPEAN JOURNAL OF CLINICAL INVESTIGATION, Issue 4 2001Z. Prohászka Background Recent observations indicate an association between antibodies against mycobacterial heat shock protein (hsp65) and coronary heart disease (CHD). Previously, we reported on marked differences in antigen specificity and complement activating ability of anti-hsp65 antibodies and auto-antibodies against human heat shock protein, hsp60. Here, we investigated whether there are differences between antih-sp65 and anti-hsp60 antibodies in their association with CHD. Design We measured by ELISA the levels of antibodies to hsp65, hsp60 and E. coli -derived GroEL in three groups: Group I, 357 patients with severe CHD who underwent by-pass surgery; Group II, 67 patients with negative coronary angiography; Group III, 321 healthy blood donors. Antibodies against Helicobacter pylori were also measured by commercial ELISA. Results As calculated by multiple regression analysis, the levels of anti-hsp60 auto-antibodies were significantly higher in Group I compared to Group II (P = 0·007) or Group III (P < 0·0001). By contrast, although concentrations of anti-hsp65 and anti-GroEL antibodies in Group I were higher than in Group III, no significant differences between Group I and Group II were found. Antibodies to the two bacterial hsp strongly correlated to each other, but either did not correlate or weakly correlated to hsp60. In Group I, serum concentrations of anti- H.pylori antibodies significantly correlated with those of anti-hsp65 and anti-GroEL antibodies but they did not correlate with the anti-hsp60 antibodies. Conclusion As to their clinical relevance, a remarkable difference become evident between antibodies to human hsp60 and antibodies against bacterial hsp in the extent of association with CHD. On the basis of these findings and some pertinent literature data, an alternative explanation for the association between high level of anti-hsp antibodies and atherosclerotic vascular diseases is raised. [source] Environmental manipulations early in development alter seizure activity, Ih and HCN1 protein expression later in lifeEUROPEAN JOURNAL OF NEUROSCIENCE, Issue 12 2006Ulrich Schridde Abstract Although absence epilepsy has a genetic origin, evidence from an animal model (Wistar Albino Glaxo/Rijswijk; WAG/Rij) suggests that seizures are sensitive to environmental manipulations. Here, we show that manipulations of the early rearing environment (neonatal handling, maternal deprivation) of WAG/Rij rats leads to a pronounced decrease in seizure activity later in life. Recent observations link seizure activity in WAG/Rij rats to the hyperpolarization-activated cation current (Ih) in the somatosensory cortex, the site of seizure generation. Therefore, we investigated whether the alterations in seizure activity between rats reared differently might be correlated with changes in Ih and its channel subunits hyperpolarization-activated cation channel HCN1, 2 and 4. Whole-cell recordings from layer 5 pyramidal neurons, in situ hybridization and Western blot of the somatosensory cortex revealed an increase in Ih and HCN1 in neonatal handled and maternal deprived, compared to control rats. The increase was specific to HCN1 protein expression and did not involve HCN2/4 protein expression, or mRNA expression of any of the subunits (HCN1, 2, 4). Our findings provide the first evidence that relatively mild changes in the neonatal environment have a long-term impact of absence seizures, Ih and HCN1, and suggest that an increase of Ih and HCN1 is associated with absence seizure reduction. Our findings shed new light on the role of Ih and HCN in brain functioning and development and demonstrate that genetically determined absence seizures are quite sensitive for early interventions. [source] Vulnerability of African mammals to anthropogenic climate change under conservative land transformation assumptionsGLOBAL CHANGE BIOLOGY, Issue 3 2006WILFRIED THUILLER Abstract Recent observations show that human-induced climate change (CC) and land transformation (LT) are threatening wildlife globally. Thus, there is a need to assess the sensitivity of wildlife on large spatial scales and evaluate whether national parks (NPs), a key conservation tools used to protect species, will meet their mandate under future CC and LT conditions. Here, we assess the sensitivity of 277 mammals at African scale to CC at 10, resolution, using static LT assumptions in a ,first-cut' estimate, in the absence of credible future LT trends. We examine the relationship between species' current distribution and macroclimatic variables using generalized additive models, and include LT indirectly as a filter. Future projections are derived using two CC scenarios (for 2050 and 2080) to estimate the spatial patterns of loss and gain in species richness that might ultimately result. We then apply the IUCN Red List criteria A3(c) of potential range loss to evaluate species sensitivity. We finally estimate the sensitivity of 141 NPs in terms of both species richness and turnover. Assuming no spread of species, 10,15% of the species are projected to fall within the critically endangered or extinct categories by 2050 and between 25% and 40% by 2080. Assuming unlimited species spread, less extreme results show proportions dropping to approximately 10,20% by 2080. Spatial patterns of richness loss and gain show contrasting latitudinal patterns with a westward range shift of species around the species-rich equatorial zone in central Africa, and an eastward shift in southern Africa, mainly because of latitudinal aridity gradients across these ecological transition zones. Xeric shrubland NPs may face significant richness losses not compensated by species influxes. Other NPs might expect substantial losses and influxes of species. On balance, the NPs might ultimately realize a substantial shift in the mammalian species composition of a magnitude unprecedented in recent geological time. To conclude, the effects of global CC and LT on wildlife communities may be most noticeable not as a loss of species from their current ranges, but instead as a fundamental change in community composition. [source] Assessing damaged road verges as a suspended sediment source in the Hampshire Avon catchment, southern United KingdomHYDROLOGICAL PROCESSES, Issue 9 2010A. L. Collins Abstract Diffuse sediment pollution impairs water quality, exerts a key control on the transfer and fate of nutrients and contaminants and causes deleterious impacts on freshwater ecology. A variety of catchment sediment sources can contribute to such problems. Sediment control strategies and effective targeting of mitigation options therefore require robust quantitative information on the key sources of the sediment problem at catchment scale. Recent observations by Catchment Sensitive Farming Officers (CSFO's) in England have highlighted road verges damaged and eroded by passing vehicles, particularly large farm machinery, and livestock herd movement as visually important potential sources of local sediment problems. A study was therefore undertaken to assess the relative importance of damaged road verges as a suspended sediment source in three sub-catchments of the Hampshire Avon drainage basin, southern UK. Road verge sediment contributions were apportioned in conjunction with those from agricultural topsoils and channel banks/subsurface sources. Time-integrating isokinetic samplers were deployed to sample suspended sediment fluxes at the outlets of two control sub-catchments drained by the Rivers Chitterne and Till selected to characterize areas with a low road network density and limited visual evidence of verge damage, as well as the River Sem sub-catchment used to represent areas where road verge damage is more prevalent. The findings of a sediment source fingerprinting investigation based on a combination of intermittent sampling campaigns spanning the period 22/5/02,27/4/08 suggested that the respective overall mean relative sediment contributions from damaged road verges were 5 ± 3%, 4 ± 2% and 20 ± 2%. Relative inputs from damaged road verges for any specific sampling period in the River Sem sub-catchment were as high as 33 ± 2%. Reconstruction of historical sources in the same sub-catchment, based on the geochemical record stored in a floodplain depth profile, suggested that the significance of damaged road verges as a sediment source has increased over the past 15,20 years. The findings provide important information on damaged road verges as a primary source of suspended sediment and imply that catchment sediment control strategies and mitigation plans should consider such verges in addition to those agricultural and channel sources traditionally taken into account when attempting to reduce sediment pressures on aquatic resources. Jagiello Summary In antineutrophil cytoplasmatic antibody (ANCA)-associated vasculitides (AAV), bactericidal/permeability increasing protein (BPI) ANCAs are detected. Recent observations suggest that BPI-ANCAs can potentially contribute to a proinflammatory setting in the absence of proteinase 3 (PRTN3) ANCAs during the development of a pulmonary relapse by impeding the elimination of Gram-negative bacteria (GNB). However, it is as yet not clear whether the genetic background contributes to the generation of BPI-ANCAs in Wegener granulomatosis (WG) or if BPI polymorphisms are associated with WG. In this study we genotyped the functionally relevant single nucleotide polymorphism (SNP) A645 (Glu216Lys) of the BPI gene in 201 WG patients and 608 healthy controls. To investigate whether further SNPs might be associated with WG, we also examined an intragenic microsatellite marker. No significant differences were found between patients and controls. Thus BPI polymorphisms do not appear to contribute to genetic predisposition to WG. Moreover, our data do not suggest a genetic background for the generation of BPI-ANCAs in WG. [source] Wnt 3a promotes proliferation and suppresses osteogenic differentiation of adult human mesenchymal stem cells,JOURNAL OF CELLULAR BIOCHEMISTRY, Issue 6 2004Genevieve M. Boland Abstract Multipotential adult mesenchymal stem cells (MSCs) are able to differentiate along several known lineages, and lineage commitment is tightly regulated through specific cellular mediators and interactions. Recent observations of a low/high bone-mass phenotype in patients expressing a loss-/gain-of-function mutation in LRP5, a coreceptor of the Wnt family of signaling molecules, suggest the importance of Wnt signaling in bone formation, possibly involving MSCs. To analyze the role of Wnt signaling in mesenchymal osteogenesis, we have profiled the expression of WNTs and their receptors, FRIZZLEDs (FZDs), and several secreted Wnt inhibitors, such as SFRPs, and examined the effect of Wnt 3a, as a representative canonical Wnt member, during MSC osteogenesis in vitro. WNT11, FZD6, SFRP2, and SFRP3 are upregulated during MSC osteogenesis, while WNT9A and FZD7 are downregulated. MSCs also respond to exogenous Wnt 3a, based on increased ,-catenin nuclearization and activation of a Wnt-responsive promoter, and the magnitude of this response depends on the MSC differentiation state. Wnt 3a exposure inhibits MSC osteogenic differentiation, with decreased matrix mineralization and reduced alkaline phosphatase mRNA and activity. Wnt 3a treatment of fully osteogenically differentiated MSCs also suppresses osteoblastic marker gene expression. The Wnt 3a effect is accompanied by increased cell number, resulting from both increased proliferation and decreased apoptosis, particularly during expansion of undifferentiated MSCs. The osteo-suppressive effects of Wnt 3a are fully reversible, i.e., treatment prior to osteogenic induction does not compromise subsequent MSC osteogenesis. The results also showed that sFRP3 treatment attenuates some of the observed Wnt 3a effects on MSCs, and that inhibition of canonical Wnt signaling using a dominant negative TCF1 enhances MSC osteogenesis. Interestingly, expression of Wnt 5a, a non-canonical Wnt member, appeared to promote osteogenesis. Taken together, these findings suggest that canonical Wnt signaling functions in maintaining an undifferentiated, proliferating progenitor MSC population, whereas non-canonical Wnts facilitate osteogenic differentiation. Release from canonical Wnt regulation is a prerequisite for MSC differentiation. Thus, loss-/gain-of-function mutations of LRP5 would perturb Wnt signaling and depress/promote bone formation by affecting the progenitor cell pool. Macaluso Ras genes are evolutionary conserved and codify for a monomeric G protein binding GTP (active form) or GDP (inactive form). The ras genes are ubiquitously expressed although mRNA analysis suggests different level expression in tissue. Mutations in each ras gene frequently were found in different tumors, suggesting their involvement in the development of specific neoplasia. These mutations lead to a constitutive active and potentially oncogenic protein that could cause a deregulation of cell cycle. Ras protein moderates cellular responses at several mitogens and/or differentiation factors and at external stimuli. These stimuli activate a series of signal transduction pathways that either can be independent or interconnected at different points. Recent observations begin to clarify the complex relationship between Ras activation, apoptosis, and cellular proliferation. A greater understanding of these processes would help to identify the factors directly responsible for cell cycle deregulation in several tumors, moreover it would help the design of specific therapeutic strategies, for the control on the proliferation of neoplastic cells. [source] Current Experimental Perspectives on the Clinical Progression of Alcoholic Liver DiseaseALCOHOLISM, Issue 10 2009Katja Breitkopf Chronic alcohol abuse is an important cause of morbidity and mortality throughout the world. Liver damage due to chronic alcohol intoxication initially leads to accumulation of lipids within the liver and with ongoing exposure this condition of steatosis may first progress to an inflammatory stage which leads the way for fibrogenesis and finally cirrhosis of the liver. While the earlier stages of the disease are considered reversible, cirrhotic destruction of the liver architecture beyond certain limits causes irreversible damage of the organ and often represents the basis for cancer development. This review will summarize current knowledge about the molecular mechanisms underlying the different stages of alcoholic liver disease (ALD). Recent observations have led to the identification of new molecular mechanisms and mediators of ALD. For example, plasminogen activator inhibitor 1 was shown to play a central role for steatosis, the anti-inflammatory adipokine, adiponectin profoundly regulates liver macrophage function and excessive hepatic deposition of iron is caused by chronic ethanol intoxication and increases the risk of hepatocellular carcinoma development. Until recently, peroxisomes were viewed as a class of static organelles that developed by growth and fission from pre-existing organelles. Recent observations have challenged this view by providing evidence that peroxisomes may be part of the endomembrane system and constitute a highly dynamic population of organelles that arises and is removed upon environmental demands. Additionally, evidence is now accumulating that peroxisomes may arise by alternative methods. This review summarizes relevant recent data on this subject. [source] Evolution of balanced genetic polymorphismMOLECULAR ECOLOGY, Issue 12 2000Adam Richman Abstract Extreme genetic polymorphism maintained by balancing selection (so called because many alleles are maintained in a balance by a mechanism of rare allele advantage) is intimately associated with the important task of self/non-self-discrimination. Widely disparate self-recognition systems of plants, animals and fungi share several general features, including the maintenance of large numbers of alleles at relatively even frequency, and persistence of this variation over very long time periods. Because the evolutionary dynamics of balanced polymorphism are very different from those of neutral genetic variation, data on balanced polymorphism have been used as a novel source for inference of the history of populations. This review highlights the unique evolutionary properties of balanced genetic polymorphism, and the use of theoretical understanding in analysis and application of empirical data for inference of population history. However, a second goal of this review is to point out where current theory is incomplete. Recent observations suggest that entirely novel selective forces may act in concert with balancing selection, and these novel forces may be extremely potent in shaping genetic variation at self-recognition loci. [source] Modification of the gamma-ray spectra by internal absorption in optically violently variable blazars: the example cases of 3C 273 and 3C 279MONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 2 2008J. Sitarek ABSTRACT Recent observations with the low-threshold Cherenkov telescopes proved that sub-TeV ,-rays are able to arrive from active galaxies at relatively large distances in spite of the expected severe absorption in the extragalactic background light (EBL). We calculate the ,-ray spectra at TeV energies from two example optically violently variable quasars, 3C 273 and 3C 279, assuming that ,-rays are injected in the inner parts of the jets launched by the accretion discs. It is assumed that ,-rays in the broad energy range (from MeV up to TeV) are produced in these blazars with a power-law spectrum with the spectral index as observed from these objects by the EGRET telescope at GeV energies. We take into account the internal absorption of these ,-rays by considering a number of models for the radiation field surrounding the jet. The classical picture of a relativistic blob in a jet for the injection of primary ,-rays is considered, with the injection rate of ,-rays as observed by the EGRET telescope in the GeV energy range. The results of calculations are compared with positive detection and the upper limits on the sub-TeV ,-ray fluxes from these two sources. It is concluded that, even with the Stecker EBL model, the level of ,-ray emission from 3C 279 is close to the recent measurements in the sub-TeV ,-ray energies, provided that the injected ,-ray spectrum extends from the GeV energies over the next two decades with this same spectral index. We also suggest that a flare with a time-scale of a few days from 3C 273 could be detected by the MAGIC II stereo telescopes. We present Hartree,Fock calculations of the polarization-dependent photoionization cross-sections of the He atom in strong magnetic fields ranging from 1012 to 1014 G. Convenient fitting formulae for the cross-sections are given along with the related oscillator strengths for various bound,bound transitions. The effects of finite nucleus mass on the radiative absorption cross-sections are examined using perturbation theory. [source] Fate of clumps in damped Ly, systemsMONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 4 2006Ilian T. Iliev ABSTRACT Recent observations have revealed that damped Ly, clouds (DLAs) host star formation activity. In order to examine if such star formation activity can be triggered by ionization fronts, we perform high-resolution hydrodynamics and radiative transfer simulations of the effect of radiative feedback from propagating ionization fronts on high-density clumps. We examine two sources of ultraviolet (UV) radiation field to which high-redshift (z, 3) galaxies could be exposed: one corresponding to the UV radiation originating from stars within the DLA, itself, and the other corresponding to the UV background radiation. We find that, for larger clouds, the propagating I-fronts created by local stellar sources can trigger cooling instability and collapse of significant part, up to 85 per cent, of the cloud, creating conditions for star formation in a time-scale of a few Myr. The passage of the I-front also triggers collapse of smaller clumps (with radii below ,4 pc), but in these cases the resulting cold and dense gas does not reach conditions conducive to star formation. Assuming that 85 per cent of the gas initially in the clump is converted into stars, we obtain a star formation rate of ,0.25 M, yr,1 kpc,2. This is somewhat higher than the value derived from recent observations. On the other hand, the background UV radiation which has harder spectrum fails to trigger cooling and collapse. Instead, the hard photons which have long mean free-path heat the dense clumps, which as a result expand and essentially dissolve in the ambient medium. Therefore, the star formation activity in DLAs is strongly regulated by the radiative feedback, both from the external UV background and internal stellar sources and we predict quiescent evolution of DLAs (not starburst-like evolution). [source] Metallicity, planetary formation and migrationMONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 3 2003Mario Livio ABSTRACT Recent observations show a clear correlation between the probability of hosting a planet and the metallicity of the parent star. As radial velocity surveys are biased, however, towards detecting planets with short orbital periods, the probability,metallicity correlation could merely reflect a dependence of migration rates on metallicity. We investigated the possibility, but find no basis to suggest that the migration process is sensitive to the metallicity. The indication is, therefore, that a higher metallicity results in a higher probability for planet formation. [source] Kilohertz quasi-periodic oscillations difference frequency exceeds inferred spin frequency in 4U 1636,53MONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 1 2002Peter G. Jonker Abstract Recent observations of the low-mass X-ray binary 4U 1636,53 with the Rossi X-ray Timing Explorer show, for the first time, a kilohertz quasi-periodic oscillation (kHz QPO) peak separation that exceeds the neutron star spin frequency as inferred from burst oscillations. This strongly challenges the sonic-point beat-frequency model for the kHz QPOs found in low-mass X-ray binaries. We detect two simultaneous kHz QPOs with a frequency separation of 323.3 ± 4.3 Hz in an average Fourier power spectrum of observations obtained in 2001 September and 2002 January. The lower kHz QPO frequency varied between 644 and 769 Hz. In previous observations of this source the peak separation frequency was ,250 Hz when the lower kHz QPO frequency was ,900 Hz. Burst oscillations occur in 4U 1636,53 at ,581 Hz and also possibly at half that frequency (290.5 Hz). This is the first source where the peak separation frequency is observed to change from less than (half) the burst oscillation frequency to more than that. This observation contradicts all previously formulated implementations of the sonic-point beat-frequency model except those where the disc in 4U 1636,53 switches from prograde to retrograde. [source] Winds from massive stars: implications for the afterglows of , -ray burstsMONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 3 2001Enrico Ramirez-Ruiz Recent observations suggest that long-duration , -ray bursts (GRBs) and their afterglows are produced by highly relativistic jets emitted in core-collapse explosions. The pre-explosive ambient medium provides a natural test for the most likely progenitors of GRBs. Those stars that shed their envelopes most readily have short jet crossing times and are more likely to produce a GRB. We construct a simple computational scheme to explore the expected contribution of the presupernova ejecta of single Wolf,Rayet (WR) stars to the circumstellar environment. Using detailed stellar tracks for the evolution of massive stars, we discuss the effects that the initial main-sequence mass, metallicity, rotation and membership in a binary system have on the ambient medium. We extend the theory of GRB afterglows in winds to consider the effect of the relativistic fireball propagating through the WR ejecta. Specific predictions are made for the interaction of the relativistic blast wave with the density bumps that arise when the progenitor star rapidly loses a large fraction of its initial mass or when the ejected wind interacts with the external medium and decelerates. A re-brightening of the afterglow with a spectrum redder than the typical synchrotron spectrum (as seen in GRB 970508, GRB 980326 and GRB 000911) is predicted. We also calculate the luminosity of the reflected echo that arises when circumstellar material Compton-scatters the prompt radiation, and examine the spectral signatures expected from the interaction of the GRB afterglow with the ejected medium. Wirth Abstract This paper systematically investigates differences between the thermal and the dynamical tropopause for upper-tropospheric balanced flow anomalies. Idealized cyclonic and anticyclonic anomalies are considered, which are either axisymmetric or plane symmetric. Given a distribution of potential vorticity (PV), the inversion problem is solved numerically to obtain the corresponding balanced flow (i.e. wind and temperature). The control parameter is the aspect ratio of the PV anomaly, which governs the partitioning into a thermal and a dynamical anomaly. For PV anomalies of intermediate and tall aspect ratios, the location of the thermal tropopause differs significantly from the location of the dynamical tropopause. The thermal tropopause is rather indistinct for intermediate aspect ratios, while it is sharp and well defined for both tall and shallow anomalies. A barotropic deformation flow field superimposed on a plane symmetric anomaly induces an ageostrophic wind which modifies the static stability throughout the PV anomaly such that the thermal and dynamical tropopauses evolve differently. Recent observations concerning the correlation between the thermal and ozone tropopauses can be interpreted consistently in terms of the present results. Budding Abstract We use minimal empirical modelling techniques to interpret recent (2006,2007) photometry and spectroscopy of AB Dor. We compare, in particular, broadband (B and V) maculation effects with emission features in high-resolution Ca II K-line spectroscopy. We also compare emission effects in the Ca II Kand H, lines observed at different rotational phases. We refer to a broader multiwavelength campaign, of which these optical data were a part, involving X-ray and microwave observations to be published later. The broadband light curves are characterized by one outstanding macula, whereas the emission lines suggest 4 possible main chromospheric activity sites (,faculae'). These appear at a similar latitude and with comparable size to the main umbra, but there are significant displacements in longitude. However, one strong facular concentration near phase zero may have a physical relationship to the main macula. The derived longitudes of these features would have been affected by differential rotation operating over the several months between the spectroscopic and photometric observations, but the difference of at least ,30° between facula and umbra appears too great to allow their coincidence. The possibility of a large bipolar surface structure is considered, keeping in mind the bipolar character of solar activity centres: the activity of rapidly rotating cool stars being generally compared with that of the Sun, scaled up by a few orders of magnitude. Observed microwave activity may link to this same main photospheric and chromospheric centre picked up by the optical analysis. Recent observations suggest that "the H3K27me3 modification" acts to maintain Polycomb repressive complex (PRC) 2, the enzyme that creates this modification, on replicating chromatin. This could in turn promote propagation of H3K27me3 on newly replicated daughter chromatin, and promote recruitment of PRC1. Other work suggests that PRC1-class complexes can be maintained on replicating chromatin, at least in vitro, independently of H3K27me3. Metcalf Abstract In Drosophila, testis-specific TBP-associated factors (tTAFs) predominantly localize to spermatocyte nucleoli and regulate the transcription of genes necessary for spermatocyte entry into meiosis. tTAFs are paralogs of generally expressed TAF subunits of transcription factor IID (TFIID). Our recent observation that the generally expressed TAF1 isoform TAF1-2 is greatly enriched in testes prompted us to explore the functional relationship between general TAFs and tTAFs during spermatogenesis. Analysis by immunofluorescence microscopy revealed that among the general TFIID subunits examined (TATA-box binding protein [TBP], TAF1, TAF4, TAF5, and TAF9), only TAF1 colocalized with the tTAF Mia in spermatocyte nucleoli. Nucleolar localization of TAF1, but not Mia, was disrupted in tTAF mutant flies, and TAF1 dissociated from DNA prior to Mia as spermatocytes entered meiosis. Taken together, our results suggest stepwise assembly of a testis-specific TFIID complex (tTFIID) whereby a TAF1 isoform, presumably TAF1-2, is recruited to a core subassembly of tTAFs in spermatocyte nucleoli. Hofheinz Abstract The 2d gauge theory on the lattice is equivalent to the twisted Eguchi,Kawai model, which we simulated at N ranging from 25 to 515. We observe a clear large N scaling for the 1- and 2-point function of Wilson loops, as well as the 2-point function of Polyakov lines. The 2-point functions agree with a universal wave function renormalization. The large N double scaling limit corresponds to the continuum limit of non-commutative gauge theory, so the observed large N scaling demonstrates the non-perturbative renormalizability of this non-commutative field theory. The area law for the Wilson loops holds at small physical area as in commutative 2d planar gauge theory, but at large areas we find an oscillating behavior instead. In that regime the phase of the Wilson loop grows linearly with the area. This agrees with the Aharonov-Bohm effect in the presence of a constant magnetic field, identified with the inverse non-commutativity parameter. Next we investigate the 3d ,,4 model with two non-commutative coordinates and explore its phase diagram. Our results agree with a conjecture by Gubser and Sondhi in d = 4, who predicted that the ordered regime splits into a uniform phase and a phase dominated by stripe patterns. We further present results for the correlators and the dispersion relation. In non-commutative field theory the Lorentz invariance is explicitly broken, which leads to a deformation of the dispersion relation. In one loop perturbation theory this deformation involves an additional infrared divergent term. Our data agree with this perturbative result. We also confirm the recent observation by Ambjř rn and Catterall that stripes occur even in d = 2, although they imply the spontaneous breaking of the translation symmetry. This activity of 5-ASA is probably the consequence of a number of effects determined on colo-rectal cancer cells, consisting of reduced proliferation, increased apoptosis and activation of cell cycle checkpoints and DNA repair processes. A recent observation has suggested that inhibition of ,-catenin signalling could induce these cellular effects. Aim, To characterize better the capacity of 5-ASA to inhibit the ,-catenin signalling pathway. Methods, Genes belonging to the ,-catenin signalling pathway were analysed in colo-rectal cancer cell lines treated with 5-ASA using a combination of laboratory assays that are able to detect their phenotypic expression and functional activity. Results, The results obtained indicated that 5-ASA induces the expression of a protein called ,-protocadherin that belongs to the cadherin superfamily and is able to sequester ,-catenin on the plasmatic membrane of treated cells hampering its function. Conclusion, These findings suggest that ,-protocadherin might be employed as a biological marker to monitor the chemopreventive efficacy of 5-ASA. Recent studies have shown that exosomes are also found in vivo in body fluids such as blood, urine, amniotic fluid, malignant ascites, bronchoalveolar lavage fluid, synovial fluid, and breast milk. While the biological function of exosomes is still unclear, they can mediate communication between cells, facilitating processes such as antigen presentation and in trans signaling to neighboring cells. Exosome-like vesicles identified in Drosophila (referred to as argosomes) may be potential vehicles for the spread of morphogens in epithelia. The advent of current MS-based proteomic technologies has contributed significantly to our understanding of the molecular composition of exosomes. In addition to a common set of membrane and cytosolic proteins, it is becoming increasingly apparent that exosomes harbor distinct subsets of proteins that may be linked to cell-type associated functions. The secretion of exosomes by tumor cells and their implication in the transport and propagation of infectious cargo such as prions and retroviruses such as HIV suggest their participation in pathological situations. Interestingly, the recent observation that exosomes contain both mRNA and microRNA, which can be transferred to another cell, and be functional in that new environment, is an exciting new development in the unraveling exosome saga.
