POTENTIAL TOOLS FOR TRACKING OCEAN CLIMATE: VARIABILITY IN STABLE ISOTOPES IN LIVING CORALLINE ALGAE

Our ability to track long term climate change in coastal regions is limited in temperate and polar regions. Physical oceanographic dynamics in temperature and upwelling events can be recorded as carbon and oxygen stable isotope signals in carbonate producing organisms. Because coralline algae photosynthesize, produce calcium carbonate and are widely distributed, they may provide a new tool for detecting short-term change. However, little is known about how coralline algae incorporate stable isotopes into their calcite thallus structure. The objectives of this study were to determine if growth and isotopic signature differ in articulated coralline algae grown in different oceanographic regimes in Monterey Bay. The articulated alga Calliarthron cheiliospororioides was outplanted at three locations varying in seawater temperature and upwelling strength. New algal growth was measured by staining the algae with Alizarin Red and enumerating the amount of accumulated material at the branch tips. Growth rates varied seasonally and spatially. Low-upwelling daily growth rates averaged 0.044,0.056 mm day,1, while high-upwelling growth rates were 0.083 mm day,1. Isotope ratios were obtained by analyzing microsampled portions of the alga in a mass spectrometer. Changes in the 18O/16O and 13C/12C ratios appear to reflect change in seawater temperature and upwelling strength, respectively. [source]


Capsule Endoscopy in Examination of Esophagus for Lesions After Radiofrequency Catheter Ablation: A Potential Tool to Select Patients With Increased Risk of Complications

Capsule Endoscopy in Examination of Esophagus.,Background: Esophageal injury can result from left atrial radiofrequency ablation (RFA) therapy, with added concern because of its possible relationship to the development of atrial-esophageal (A-E) fistulas. Objective: Evaluate utility of esophageal capsule endoscopy to detect esophageal lesions as a complication of RFA therapy in the treatment of atrial fibrillation (AF). Methods: Consecutive patients with AF who underwent left atrial RFA therapy and received capsule endoscopy within 48 hours postablation. Video was reviewed by a single gastroenterologist. The medical records were also reviewed for symptoms immediately postablation and at the 3-month follow-up. Results: A total of 93 consecutive patients were included and 88 completed the study and were analyzed. The prevalence of esophageal lesions was 17% (15/88 patients). Nine percent (8/88) of these patients had lesions anatomically consistent with the location of the ablation catheter. Six patients with positive capsule findings had symptoms of chest pain (3/6, 50%), throat pain (2/6, 33%), nausea (1/6, 17%), and abdominal pain (1/6, 17%). An additional 24 patients were symptomatic postablation, but with normal capsule findings. All patients with identified lesions by capsule endoscopy received oral proton pump inhibitor therapy, and were instructed to contact the Cleveland Clinic in the event of worsening symptoms. No delayed complications were reported at the 3-month follow-up. Conclusion: This study supports the use of capsule endoscopy as a tool for the detection of esophageal injury post-RFA therapy. PillCam ESO is well tolerated and provides satisfactory images of the areas of interest in the esophagus without potential risk related to insufflation with regular esophagogastroduodenoscopy. Among the genetic variants most commonly assessed by these companies are those found in genes that influence cardiovascular disease risk. However, the interpretation of DNA-based data is complex. The goal of this paper is to carefully examine nutritional genomics as a potential tool for targeted medical nutrition therapy. Electrocardiographic Body Surface Mapping: Potential Tool for the Detection of Transient Myocardial Ischemia in the 21st Century?

Coronary artery disease (CAD) is one of the leading causes of cardiovascular mortality and morbidity worldwide. CAD presents as a wide spectrum of clinical disease from stable angina to ST segment elevation myocardial infarction. The 12-lead electrocardiogram (ECG) has been the main tool for the diagnosis of these events for almost a century but is limited in its diagnostic ability. For patients with suspected angina, the exercise tolerance test is often used to provoke and detect stress-induced ischemia but does not provide a definitive answer in a substantial proportion of patients. Body surface mapping (BSM) is a technique that samples multiple points around the thorax to provide a more comprehensive electrocardiographic data set than the conventional 12-lead ECG. Moreover, recent preliminary data demonstrate that BSM can detect and display transient regional myocardial ischemia in an intuitive fashion, employing subtraction color mapping, making it potentially valuable for diagnosing CAD causing transient regional ischemia. Left Ventricle Afterload Impedance Control by an Axial Flow Ventricular Assist Device: A Potential Tool for Ventricular Recovery

Abstract Ventricular assist devices (VADs) are increasingly used for supporting blood circulation in heart failure patients. To protect or even to restore the myocardial function, a defined loading of the ventricle for training would be important. Therefore, a VAD control strategy was developed that provides an explicitly definable loading condition for the failing ventricle. A mathematical model of the cardiovascular system with an axial flow VAD was used to test the control strategy in the presence of a failing left ventricle, slight physical activity, and a recovering scenario. Furthermore, the proposed control strategy was compared to a conventional constant speed mode during hemodynamic changes (reduced venous return and arterial vasoconstriction). The physiological benefit of the control strategy was manifested by a large increase in the ventricular Frank,Starling reserve and by restoration of normal hemodynamics (5.1 L/min cardiac output at a left atrial pressure of 10 mm Hg vs. 4.2 L/min at 21 mm Hg in the unassisted case). The control strategy automatically reduced the pump speed in response to reduced venous return and kept the pump flow independent of the vasoconstriction condition. Most importantly, the ventricular load was kept stable within 1%, compared to a change of 75% for the constant speed. As a key feature, the proposed control strategy provides a defined and adjustable load to the failing ventricle by an automatic regulation of the VAD speed and allows a controlled training of the myocardium. This, in turn, may represent a potential additional tool to increase the number of patients showing recovery. [source]


Evoked Human Oesophageal Hyperalgesia: A Potential Tool for Analgesic Evaluation?

Therefore, in the development and testing of analgesics for the treatment of visceral pain, it is important to establish an experimental pain model of visceral hypersensitivity. Such a model will mimic the clinical situation to a higher degree than pain models where the receptors and peripheral afferents are briefly activated as with, for example, electrical, thermal, and mechanical stimulations. In this study, a model to evoke experimental hyperalgesia of the oesophagus with a combination of acid and capsaicin was introduced. The study was a randomised, double-blind, cross-over study. Fifteen healthy volunteers were included. Sensory assessments to mechanical, heat, and electrical stimulations were done in the distal oesophagus, before and after perfusion with a 200 ml solution of acid+capsaicin (180 ml HCL 0.1 M and 2 mg capsaicin in 20 ml solvent) or saline. Oesophageal pain assessment and referred pain areas were evaluated. There were reproducible pain assessments between repetitions within the same day and between days (all P > 0.05). Acid+capsaicin perfusion induced 56% reduction of the pain threshold to heat (P = 0.04), 19% reduction of the pain threshold to electrical stimuli (P < 0.001), 78% increase of the referred pain areas to mechanical stimulation (P < 0.001) and 52% increase of the referred pain areas to electrical stimulus (P = 0.045). All volunteers were sensitised to one or more modalities by acid+capsaicin. The model was able to evoke consistent hyperalgesia and may be useful in future pharmacological studies. [source]


Comparison of Impedance Cardiography to Direct Fick and Thermodilution Cardiac Output Determination in Pulmonary Arterial Hypertension

Cardiac output (CO) is an important diagnostic and prognostic tool for patients with ventricular dysfunction. Pulmonary hypertension patients undergo invasive right heart catheterization to determine pulmonary vascular and cardiac hemodynamics. Thermodilution (TD) and direct Fick method are the most common methods of CO determination but are costly and may be associated with complications. The latest generation of impedance cardiography (ICG) provides noninvasive estimation of CO and is now validated. The purpose of this study was to compare ICG measurement of CO to TD and direct Fick in pulmonary hypertension patients. Thirty-nine enrolled patients were analyzed: 44% were male and average age was 50.8±17.4 years. Results for bias and precision of cardiac index were as follows: ICG vs. Fick (,0.13 L/min/m2 and 0.46 L/min/m2), TD vs. Fick (0.10 L/min/m2 and 0.41 L/min/m2), ICG vs. TD (respectively, with a 95% level of agreement between ,0.72 and 0.92 L/min/m2; CO correlation of ICG vs. Fick, TD vs. Fick, and ICG vs. TD was 0.84, 0.89, and 0.80, respectively). ICG provides an accurate, useful, and cost-effective method for determining CO in pulmonary hypertension patients, and is a potential tool for following responses to therapeutic interventions. Abstract Stem cells have been recognized as a potential tool for the development of innovative therapeutic strategies. There are in general two types of stem cells, embryonic and adult stem cells. While embryonic stem cell therapy has been riddled with problems of allogeneic rejection and ethical concerns, adult stem cells have long been used in the treatment of hematological malignancies. With the recognition of additional, potentially therapeutic characteristics, bone marrow,derived stem cells have become a tool in regenerative medicine. The bone marrow is an ideal source of stem cells because it is easily accessible and harbors two types of stem cells. Hematopoietic stem cells give rise to all blood cell types and have been shown to exhibit plasticity, while multipotent marrow stromal cells are the source of osteocytes, chondrocytes, and fat cells and have been shown to support and generate a large number of different cell types. This review describes the general characteristics of these stem cell populations and their current and potential future applications in regenerative medicine. Abstract Quantity prediction of municipal solid waste (MSW) is crucial for design and programming municipal solid waste management system (MSWMS). Because effect of various parameters on MSW quantity and its high fluctuation, prediction of generated MSW is a difficult task that can lead to enormous error. The works presented here involve developing an improved support vector machine (SVM) model, which combines the principal component analysis (PCA) technique with the SVM to forecast the weekly generated waste of Mashhad city. In this study, the PCA technique was first used to reduce and orthogonalize the original input variables (data). Then these treated data were used as new input variables in SVM model. This improved model was evaluated by using weekly time series of waste generation (WG) and the number of trucks that carry waste in week of t. These data have been collected from 2005 to 2008. By comparing the predicted WG with the observed data, the effectiveness of the proposed model was verified. Vegetative growth and development of irrigated forage turnip (Brassica rapa var. rapa)

Abstract Field and greenhouse experiments were conducted to identify visual markers and predictors of changes in the vegetative growth rate of forage turnip (Brassica rapa var. rapa) as a potential tool to improve the timing of inputs of N and irrigation to periods of maximum demand. The onset of root expansion, which was associated with a colour change and the death of cotyledons, was identified as a critical marker for the beginning of the rapid growth of the crop and the accumulation of starch in the storage root but indicators of subsequent changes in vegetative growth rate were not identifiable. The results suggested that management inputs can be more readily targeted to the beginning of the exponential growth phase but targeting of later vegetative growth stages will remain arbitrary. The vegetative growth and development of the crop was also studied to elucidate the process of leaf emergence and senescence (turnover) as they affected both leaf and root yield. The sequential senescence of leaves, which began immediately after cotyledon death, and translocation of carbohydrate to the storage root, coupled with high leaf area index (LAI), probably account for the high growth rates of 220 kg ha,1 day,1 maintained for periods of 10 weeks after the onset of root expansion. High yields can be expected if high LAI is maintained by ensuring that leaf emergence rates are not limited by nutrient or water deficiencies and leaves are protected from insect pests. Forage turnip is particularly robust because new leaf continues to emerge as older and damaged leaves senesce and carbohydrate is stored as starch in the storage root. [source]


Evidence of a founder mutation of BRCA1 in a highly homogeneous population from southern Italy with breast/ovarian cancer

Abstract Several genes have been involved in the pathogenesis of hereditary breast/ovarian cancer (BOC), but mutations in the BRCA1 gene are by far the most recurrent. In this study, we report the identification of a founder mutation in a geographically and historically homogeneous population from Calabria, a south Italian region. A screening performed on 24 patients from unrelated families highlighted the high prevalence of a 5083del19 alteration in the BRCA1 gene, which accounts for 33% of the overall gene mutations. The same mutation was also detected in 4 patients, all of Calabrian origin, referred to us by research centres from the north of Italy. Allelotype analysis, performed on probands and unaffected family members revealed the presence a common allele, therefore suggesting a founder effect due to a common ancestor. Our findings underscore the importance of ethnic background homogeneity in patients' selection and highlight the usefulness of founder mutations as a potential tool for optimisation of preclinical diagnosis in gene carriers and therapeutic approaches in affected individuals. Managing heterogeneity in elephant distribution: interactions between elephant population density and surface-water availability

Summary 1Concerns over the ecological impacts of high African elephant Loxodonta africana densities suggest that it may be necessary to control their numbers locally, although the best management approach is still widely debated. Artificial water supply is believed to be a major cause of local overabundance, and could be used as a potential tool to regulate elephant distribution and impact across landscapes, but its effect on elephants at the population scale has never been studied. 2We assessed how dry-season surface-water availability constrained the distribution of an entire elephant population, using aerial and waterhole census data from Hwange National Park, Zimbabwe. The study was initiated in 1986, when the population was released from culling. We studied how artificial waterholes, holding water throughout the dry season, and vegetation production, estimated from a normalized difference vegetation index (NDVI), influenced the long-term distribution of elephant densities. We also investigated how the elephant distribution responded to changes in population density and annual rainfall, a driver of surface-water availability. 3Long-term dry-season elephant densities across the park tended to increase with vegetation production, and increased asymptotically with the density of artificial waterholes. 4Since the culling stopped, dry-season elephant densities have increased in most areas of the park, except in areas of low vegetation production and low water availability. Interannual fluctuations in elephant distribution are linked to rainfall variability through its effect on surface-water availability. During dry years elephants concentrated in areas where artificial pumping maintained surface-water availability during the dry season. 5During dry years elephant numbers at waterholes increased because of reduced surface-water availability, and elephants were distributed more evenly across waterholes, although active waterholes were unevenly distributed across the park. 6Synthesis and applications. Surface-water availability drives the distribution and abundance of elephants within Hwange National Park, and therefore appears to be at the heart of the trade-off between elephant conservation and the extent of their impact on ecosystems. Artificial manipulation of surface water is one of the tools available for the management of elephant populations and should not be overlooked when considering options for controlling elephant numbers in places where they are considered to be overabundant. Can management compensate for atmospheric nutrient deposition in heathland ecosystems?

Summary 1Atmospheric nutrient deposition has contributed to widespread changes in heathlands throughout Europe. As a consequence, management is now being considered as a potential tool with which to compensate for increased nutrient loads. Currently, only limited information is available on the extent to which management measures could compensate for atmospheric nutrient deposition. We hypothesized that low-intensity management measures are not sufficient to counterbalance current nutrient inputs, particularly of nitrogen (N). 2In order to improve heathland management schemes, we evaluated the effectiveness of different management measures in reducing the impact of ongoing atmospheric nutrient loads. We compared the effects of mowing, prescribed burning (low-intensity management) and sod-cutting (high-intensity management) on heathland nutrient budgets [N, calcium (Ca), potassium (K), magnesium (Mg) and phosphorus (P)] in the Lueneburg Heath nature reserve (north-west Germany). Nutrient balances were calculated by analysing the present-day input, the output as a result of the removal of biomass/humus horizons, and changes in leaching rates. 3Nutrient losses by increased leaching following management measures were negligible compared with nutrient losses caused by the removal of above-ground biomass or humus horizons. The total quantities of nutrients removed by sod-cutting were equivalent to between 37 and 176 years of atmospheric input (for N, 89 years). 4In contrast, the quantities of N removed by mowing and prescribed burning were equivalent to only 5 years of atmospheric input. Thus, heathlands subjected to such treatments will accumulate N in the long term. In addition, output,input ratios for phosphorus (P) exceeded those for N in the mowing and sod-cutting experiments. It is therefore likely that heathlands currently (co-) limited by N will shift to being more P-limited in the long term. This will promote species that are well adapted to P-limited sites (e.g. Molinia caerulea). 5Synthesis and applications. This study shows that low-intensity management cannot compensate for atmospheric N loads in the long term. Consequently, high-intensity management measures are an indispensable tool in preserving a long-term balanced N budget in heathlands. In order to maintain a diverse structure, managers need to combine low- and high-intensity management measures. Prescribed burning proved to be the best means of avoiding an increasing P shortage, because this measure causes very low P outputs. [source]

Abstract Ion channels play essential roles in human physiology and toxicology. Cardiac contraction, neural transmission, temperature sensing, insulin release, regulation of apoptosis, cellular pH and oxidative stress, as well as detection of active compounds from chilli, are some of the processes in which ion channels have an important role. Regulation of ion channels by several chemicals including those found in air, water and soil represents an interesting potential link between environmental pollution and human diseases; for instance, de novo expression of ion channels in response to exposure to carcinogens is being considered as a potential tool for cancer diagnosis and therapy. Non-specific binding of several drugs to ion channels is responsible for a huge number of undesirable side-effects, and testing guidelines for several drugs now require ion channel screening for pharmaceutical safety. Animal toxins targeting human ion channels have serious effects on the population and have also provided a remarkable tool to study the molecular structure and function of ion channels. In this review, we will summarize the participation of ion channels in biological processes extensively used in toxicological studies, including cardiac function, apoptosis and cell proliferation. Major findings on the adverse effects of drugs on ion channels as well as the regulation of these proteins by different chemicals, including some pesticides, are also reviewed. Association of ion channels and toxicology in several biological processes strongly suggests these proteins to be excellent candidates to follow the toxic effects of xenobiotics, and as potential early indicators of life-threatening situations including chronic degenerative diseases. Fibrous Dysplasia as a Stem Cell Disease,

Abstract At a time when significant attention is devoted worldwide to stem cells as a potential tool for curing incurable diseases, fibrous dysplasia of bone (FD) provides a paradigm for stem cell diseases. Consideration of the time and mechanism of the causative mutations and of nature of the pluripotent cells that mutate in early embryonic development indicates that, as a disease of the entire organism, FD can be seen as a disease of pluripotent embryonic cells. As a disease of bone as an organ, in turn, FD can be seen as a disease of postnatal skeletal stem cells, which give rise to dysfunctional osteoblasts. Recognizing FD as a stem cell disease provides a novel conceptual angle and a way to generate appropriate models of the disease, which will continue to provide further insight into its natural history and pathogenesis. In addition, skeletal stem cells may represent a tool for innovative treatments. These can be conceived as directed to alter the in vivo behavior of mutated stem cells, to replace mutated cells through local transplantation, or to correct the genetic defect in the stem cells themselves. In vitro and in vivo models are currently being generated that will permit exploration of these avenues in depth. [source]

Rietveld analysis of X-ray powder diffraction patterns as a potential tool for the identification of impact-deformed carbonate rocks

Entire XRD patterns, single peak profiles and Rietveld refined parameters of carbonate samples from the Sierra Madera impact crater, west Texas, unshocked equivalent samples from 95 miles north of the crater and the Mission Canyon Formation of southwest Montana and western Wyoming were used to evaluate the use of X-ray powder diffraction as a potential tool for distinguishing impact deformed rocks from unshocked and tectonically deformed rocks. At Sierra Madera dolostone and limestone samples were collected from the crater rim (lower shock intensity) and the central uplift (higher shock intensity). Unshocked equivalent dolostone samples were collected from well cores drilled outside of the impact crater. Carbonate rocks of the Mission Canyon Formation were sampled along a transect across the tectonic front of the Sevier and Laramide orogenic belts. Whereas calcite subjected to significant shock intensities at the Sierra Madera impact crater can be differentiated from tectonically deformed calcite from the Mission Canyon Formation using Rietveld refined peak profiles, weakly shocked calcite from the crater rim appears to be indistinguishable from the tectonically deformed calcite. In contrast, Rietveld analysis readily distinguishes shocked Sierra Madera dolomite from unshocked equivalent dolostone samples from outside the crater and tectonically deformed Mission Canyon Formation dolomite. Gravitational flexion by elliptical dark matter haloes

ABSTRACT We present equations for the gravitational lensing flexion expected for an elliptical lens mass distribution. These can be reduced to one-dimensional finite integrals, thus saving significant computing time over a full two-dimensional calculation. We estimate constraints on galaxy halo ellipticities for a range of potential future surveys, finding that the constraints from the two different types of flexion are comparable and are up to two orders of magnitude tighter than those from shear. Flexion therefore appears to be a very promising potential tool for constraining the shapes of galaxy haloes from future surveys. [source]

The present and future of public sector extension in Africa: contracting out or contracting in?

Public sector extension has come under increasing pressure to downsize and reform. Contracting out,the use of public sector funds to contract non-governmental and private service providers,is often held up as a potential tool in reform efforts. Much has been written about the possible advantages of contracting out of agricultural extension and it is being encouraged and promoted by numerous international organizations. However, a look at field experience in Africa shows that contracting out is relatively infrequent, especially compared with the reverse,contracting in,where private sector and non-governmental organizations finance public sector extension delivery. Case studies from Uganda and Mozambique indicate that on the ground attempts to come up with solutions to providing services to farmers are resulting in innovative contracting approaches and combined public and private institutional arrangements. Contracting in and public,private coalition approaches, in contrast to purely public sector extension (characterized by ineffectiveness and inefficiencies) and purely private for profit extension (which may ignore public goods and concerns), may help achieve extension services which are both demand led and which internalize public concerns such as environmental protection, food security and socio-economic equity. These coalition approaches can be improved and facilitated. They deserve greater analysis and may contribute to a better understanding of extension contracting and the roles of private and public organizations. Copyright © 2000 John Wiley & Sons, Ltd. Real-time reaction monitoring by probe electrospray ionization mass spectrometry

Probe electrospray ionization (PESI) is a modified version of the electrospray ionization (ESI), where the capillary for sampling and spraying is replaced by a solid needle. High tolerance to salts and direct ambient sampling are major advantages of PESI compared with conventional ESI. In this study, PESI-MS was used to monitor some biological and chemical reactions in real-time, such as acid-induced protein denaturation, hydrogen/deuterium exchange (HDX) of peptides, and Schiff base formation. By using PESI-MS, time-resolved mass spectra and ion chromatograms can be obtained reproducibly. Real-time PESI-MS monitoring can give direct and detailed information on each chemical species taking part in reactions, and this is valuable for a better understanding of the whole reaction process and for the optimization of reaction parameters. PESI-MS can be considered as a potential tool for real-time reaction monitoring due to its simplicity in instrumental setup, direct sampling with minimum sample preparation and low sample consumption. Sawdust Addition Reduces the Productivity of Nitrogen-Enriched Mountain Grasslands

Abstract Anthropogenic nutrient enrichment of mountain grasslands has boosted grasses and fast-growing unpalatable plants at the expense of slow-growing species, resulting in a significant loss in biodiversity. A potential tool to reduce nutrient availability and aboveground productivity without destroying the perennial vegetation is carbon (C) addition. However, little is known about its suitability under severe climatic conditions. Here, we report the results of a 3-year field study assessing the effects of sawdust addition on soil nutrients, aboveground productivity, and vegetational composition of 10 grazed and ungrazed mountain grasslands. Of particular interest was the effect of C addition on grasses and on the tall unpalatable weed Veratrum album. After 3 years, soil pH, ammonium, and plant-available phosphorus were not altered by sawdust application, and nitrate concentrations were marginally higher in treatment plots. However, the biomass of grasses and forbs (without V. album) was 20,25% lower in sawdust-amended plots, whereas the biomass of V. album was marginally higher. Sawdust addition reduced the cover of grasses but did not affect evenness, vegetation diversity, or plant species richness, although species richness generally increased with decreasing biomass at our sites. Our results suggest that sawdust addition is a potent tool to reduce within a relatively short time the aboveground productivity and grass cover in both grazed and ungrazed mountain grasslands as long as they are not dominated by tall unpalatable weeds. The technique has the advantage that it preserves the topsoil and the perennial soil seed bank. [source]


3D Computerized Model for Measuring Strain and Displacement of the Brachial Plexus Following Placement of Reverse Shoulder Prosthesis

Abstract The aim of the present study was to develop a method for three-dimensional (3D) reconstruction of the brachial plexus to study its morphology and to calculate strain and displacement in relation to changed nerve position. The brachial plexus was finely dissected and injected with contrast medium and leaden markers were implanted into the nerves at predefined places. A reverse shoulder prosthesis was inserted in a cadaveric specimen what induced positional change in the upper limb nerves. Computed tomography (CT) was performed before and after this surgical intervention. The computer assisted image processing package Mimics® was used to reconstruct the pre- and postoperative brachial plexus in 3D. The results show that the current interactive model is a realistic and detailed representation of the specimen used, which allows 3D study of the brachial plexus in different configurations. The model estimated strains up to 15.3% and 19.3% for the lateral and the medial root of the median nerve as a consequence of placing a reverse shoulder prosthesis. Furthermore, the model succeeded in calculating the displacement of the brachial plexus by tracking each implanted lead marker. The presented brachial plexus 3D model currently can be used in vitro for cadaver biomechanical analyses of nerve movement to improve diagnosis and treatment of peripheral neuropathies. The model can also be applied to study the exact location of the plexus in unusual upper limb positions like during axillary radiation therapy and it is a potential tool to optimize the approaches of brachial plexus anesthetic blocks. Prostatic stromal cells derived from benign prostatic hyperplasia specimens possess stem cell like property

Abstract INTRODUCTION The hyper-proliferative activity of stromal smooth muscle (SM) cells is believed to be responsible for the pathogenesis of benign prostatic hyperplasia (BPH). We have observed that those stromal cells can differentiate into unrelated specialized cells. We thus hypothesize that stromal cells derived from adults prostate specimens may contain adult stem cells. To test this hypothesis, human prostate stromal primary cultures were established and used for characterization of their stem cell properties. METHODS Immunoblotting, immunohistochemistry, RT-PCR, and tissue culture techniques were used to characterize the primary cultured human prostate-derived stromal cells for their stem cell and differentiation properties. The plasticity of these stromal cells was analyzed using cell culture and histology techniques. RESULTS Primary cultured prostate stromal cells from BPH patient possess polygonal and elongated fibroblast/myofibroblast cellular morphology. They are positive in CD30, CD34, CD44, NSE, CD133, Flt-1, stem cell factor (SCF), and neuron-specific enolase (NSE), but negative in C-Kit, stem cell antigen (SCA), SH2, CD11b. Expression of SM myogenic markers in these cells may be induced by sodium butyrate (NaBu) treatment. Induction to osteogenic and adipogenic differentiation in these cells is also evident. CONCLUSIONS Our study on primary stromal cells from BPH patients have yielded many interesting findings that these prostate stroma cells possess: (1) mesenchymal stem cell (MSC) markers; (2) strong proliferative potential; and (3) ability to differentiate or transdifferentiate to myogenic, adipogenic, and osteogenic lineages. These cell preparations may serve as a potential tool for studies in prostate adult stem cell research and the regulation of benign prostatic hyperplasia. Monitoring of sympathetic tone to assess postoperative pain: skin conductance vs surgical stress index

Summary The number of fluctuations in skin conductance per second has been described as a potential tool for monitoring postoperative pain. More recently, the surgical stress index has shown promising correlations with intra-operative painful stimuli. We compared both methods for their ability to assess postoperative pain, in 100 postoperative patients who were also asked to quantify their level of pain at different time points in the recovery room. The number of fluctuations per second and surgical stress index were significantly different between pain scoring , 5/10 and > 5/10 on a numeric rating scale (mean (SE) number of fluctuations per second 0.12 (0.02) vs 0.21 (0.03), respectively; p = 0.017, and surgical stress index 57 (1.4) vs 64 (1.9) points, respectively; p = 0.001). Both number of fluctuations in skin conductance per second and surgical stress index identified timepoints with moderate to severe pain with only moderate sensitivity and specificity. [source]


Anti-JC virus antibodies: Implications for PML Risk Stratification

Objective A study was undertaken to establish an enzyme-linked immunosorbent assay (ELISA) to detect JC virus (JCV)-specific antibodies in multiple sclerosis (MS) patients, and to evaluate its potential utility for identifying patients at higher or lower risk (ie, risk stratification) of developing progressive multifocal leukoencephalopathy (PML). Methods A 2-step assay for detecting and confirming the presence of anti-JCV antibodies in human serum and plasma was developed and demonstrated to be both sensitive and specific. ELISA cutpoints were statistically established using sera from >800 MS patients from natalizumab clinical studies. Subsequently, this assay was used to determine the presence of anti-JCV antibodies in natalizumab-treated PML patients where serum samples were collected 16-180 months prior to the diagnosis of PML. Results In our evaluation of natalizumab-treated MS patients, 53.6% tested positive for anti-JCV antibodies, with a 95% confidence interval of 49.9 to 57.3%. The false-negative rate of the ELISA was calculated to be approximately 2.5%, with an upper 1-sided confidence limit of 4.4%. Notably, we observed anti-JCV antibodies in all 17 available pre-PML sera samples, which was significantly different from the 53.6% seropositivity observed in the overall MS study population (p < 0.0001). Interpretation This 2-step assay provides a means to classify MS patients as having detectable or not detectable levels of anti-JCV antibodies. The finding that all 17 of the pre-PML samples that were available tested seropositive, and none tested seronegative, warrants further research on the clinical utility of the anti-JCV antibody assay as a potential tool for stratifying MS patients for higher or lower risk of developing PML. Functional analysis of the osteoarthritis susceptibility,associated GDF5 regulatory polymorphism

Objective Single-nucleotide polymorphism (SNP) rs143383 (T to C) in the 5,-untranslated region (5,-UTR) of GDF5 has recently been reported to be associated with osteoarthritis (OA) susceptibility, with lower expression of the risk-associated T allele observed in vitro and in vivo. The in vivo studies were performed on cartilage tissue from OA patients. The present study was undertaken to expand the analysis of the effect of this SNP on GDF5 allelic expression to more joint tissue types, to investigate for cis and trans factors that interact with the SNP, and to examine novel cis -acting GDF5 regulatory polymorphisms. Methods Tissue samples were collected from OA patients undergoing joint replacement of the hip or knee. Nucleic acid was extracted, and, using rs143383 and an assay that discriminates and quantifies allelic expression, the relative amount of GDF5 expression from the T and C alleles was measured. Additional common variants in the GDF5 transcript sequence were interrogated as potential regulatory elements using allelic expression and luciferase reporter assays, and electrophoretic mobility shift assays were used to search for trans factors binding to rs143383. Results We observed a consistent allelic expression imbalance of GDF5 in all tissues tested, implying that the functional effect mediated by rs143383 on GDF5 expression is joint-wide. We identified a second polymorphism, located in the 3,-UTR of GDF5, that influenced allelic expression of the gene independent of rs143383. Finally, we observed differential binding of deformed epidermal autoregulatory factor 1 (DEAF-1) to the 2 alleles of rs143383. Conclusion These findings show that the OA susceptibility mediated by polymorphism in GDF5 is not restricted to cartilage, emphasizing the need to consider the disease as involving the whole joint. The existence of an additional cis -acting regulatory polymorphism highlights the complexity of the regulation of expression of this important OA susceptibility locus. DEAF-1 is a trans -acting factor that merits further investigation as a potential tool for modulating GDF5 expression. [source]


An exergy calculator tool for process simulation

Abstract The constant tightening of environmental regulations and the ongoing need to reduce operating costs have posed a challenge for the design of any chemical process. Process engineers use process simulators to help them perform calculations that will, ultimately, result in design parameters or operating conditions for a plant or process. Exergy is a potential indicator that can aid in the design of energy efficient chemical processes and plants. The exergy concept has been increasingly used as a tool to locate the critical energy use in many industrial processes, both chemical and non-chemical. However, currently most process simulators in the market do not offer the capability of calculating the exergy of a process. An open-source exergy calculator has been created by embedding the calculation procedure in an open-source chemical process simulator. This improves process simulation by including a potential tool for design teams to quickly evaluate several process options in detail in order to understand their energy utilisation. A simple exergy analysis for a gas processing facility is used to demonstrate the capabilities of the tool. The analysis shows where the largest quantities of exergy are being consumed within the plant, thus pointing to areas where improvement in energy usage can be made.
