A non-synonymous mutation in a conserved site of the MTTP gene is strongly associated with protein activity and fatty acid profile in pigs

Summary Despite the economic interest of the fatty acid profile in pigs, no gene has been convincingly associated with this trait so far. Here, the porcine microsomal triglyceride transfer protein (MTTP) gene, which plays a crucial role in the assembly of nascent lipoproteins, has been analysed as a positional candidate gene for a QTL affecting the fatty acid composition that was previously identified on chromosome 8 in an Iberian by Landrace F2 cross. By resequencing a panel of different breeds, a non-synonymous polymorphism in a conserved residue of the lipid transfer domain of MTTP was identified. Association analyses with this polymorphism showed a strong association with the fatty acid composition of porcine fat, much stronger than the QTL effect, in the F2 cross and in a synthetic Sino-European line. In addition, in vitro activity assays in liver protein extracts have shown that this mutation is also associated with the lipid transfer activity of the MTTP protein (P < 0.1). These results suggest that the detected polymorphism is a potential causal factor of the fatty acid composition QTL. There appears to be an interaction between the porcine MTTP genotype and the type of fat source in the pig diet, which would agree with the previous results on the biology of MTTP biology. [source]


In vitro assessment of antimicrobial activity of carvacrol, thymol and cinnamaldehyde towards Salmonella serotype Typhimurium DT104: effects of pig diets and emulsification in hydrocolloids

Abstract Aims:, To determine the effect of pig diets in vitro on the antimicrobial activity of carvacrol, thymol and cinnamaldehyde, and to identify an emulsifier/stabilizer that can stabilize the essential oil (EO) components in aqueous solution and retain their antimicrobial activity in the presence of the diets. Methods and Results:, Emulsification of essential oil components with hydrocolloid solution was achieved by blending with a Polytron. Antimicrobial activity was measured through in vitro assays to determine the inhibition of bacterial growth by measuring the optical density at 600 nm or plating on nutrition agar after incubation of the mixtures of an EO component with the culture of Salmonella serotype Typhimurium DT104 in the presence or absence of pig diets. The results generated through the in vitro assays indicated that pig diets were able to abolish the antimicrobial activity of EOs. Xanthan, fenugreek and yellow mustard gums were the best in forming stable emulsions of five different EO components among ten different plant polysaccharides and surfactants examined. Emulsification of all the EO components in the fenugreek gum solution did not alter their antimicrobial activity. However, the antimicrobial activity of geraniol was significantly reduced when emulsified with other polysaccharides and surfactants. Both fenugreek and xanthan gum solutions were unable to protect the antimicrobial activity of carvacrol and thymol when mixed with the diets. Although cinnamaldehyde required no emulsification, but a high concentration (equivalent to at least three times of minimum bactericidal concentration for cinnamon oil) to inhibit Salmonella growth significantly in the presence of the diets, emulsification in fenugreek gum appeared to be essential for cinnamaldehyde solution to retain its antimicrobial activity during storage. Conclusions:, The diets for newly weaned pigs were a significant factor limiting the antimicrobial activity of EOs and their components. Cinnamaldehyde required a high concentration to retain its antimicrobial activity in the diets, in addition to its requirement for emulsification to stabilize its activity during the storage. Significance and Impact of the Study:, The assay with the diets used in this study for measuring the antimicrobial activity can be used in vitro for rapid and effective screening of potential antimicrobials for swine production. This study has identified polysaccharides that are able to stabilize EO component solutions. It has also identified cinnamaldehyde for further in vivo studies that may have potential in future application in controlling Salmonella and possibly other enteric pathogens in swine production. Effect of enzymes, flavor and organic acids on nutrient digestibility, performance and carcass traits of growing,finishing pigs fed diets containing dehydrated lucerne meal

Abstract BACKGROUND: We recently determined that 150 g kg,1 of lucerne meal can be fed during the finishing period without detrimental effects on pig performance but lucerne should be limited to less than 75 g kg,1 during the growing period. Lucerne has a high fiber content, is poorly digested and contains antinutritional factors such as saponins which reduce palatability. Therefore, pig performance could be enhanced by supplementing lucerne-containing diets with feed additives such as flavors, enzymes or organic acids. The objective of this study was to determine the effects of these additives on nutrient digestibility, performance and carcass traits when fed to growing-finishing swine fed diets containing lucerne. RESULTS: Flavor and enzyme had no effect on nutrient digestibility, pig performance or carcass traits. During the growing period, inclusion of propionic and acetic acids increased (P < 0.05) the digestibility of dry matter, protein and energy. Weight gain and intake also increased (P < 0.05). None of the additives had any effect on pig performance during the finishing period or on carcass traits. CONCLUSION: During the growing period, including 200 g kg,1 lucerne was detrimental to pig performance. Addition of organic acid improved pig performance and nutrient digestibility. During the finishing period, 200 g kg,1 lucerne can be successfully incorporated into pig diets without negative effects on pig performance. Copyright © 2008 Society of Chemical Industry [source]


Nutrient utilization and manure P excretion in growing pigs fed corn-barley-soybean based diets supplemented with microbial phytase

ABSTRACT The effect of high levels of microbial phytase supplementation in diets for growing pigs was studied in a 2-week performance and nutrient digestibility trial involving 28 growing pigs weighing 16.4 ± 1.06 (mean ± SD) kg. Seven corn-barley-soybean meal-based diets consisting of a positive control (PC) formulated to meet or exceed NRC nutrient requirements; a negative control (NC) with non-phytate P reduced by 0.1% unit from NRC requirement and fed without or with 500 or 1000 U/kg; a doubled negative control (DNC) with no added inorganic P and fed without or with 2000 or 4000 U/kg. Chromic oxide was added as an indigestible marker and all diets were fed as mash. Pigs fed the PC diet had a higher P digestibility compared with those fed the NC (P < 0.02) and the DNC (P < 0.001) diets. Supplementing the NC diet with pyhtase tended to improve P digestibility (P < 0.10). However, addition of phytase to the DNC diet resulted in linear (P < 0.001) and quadratic (P < 0.03) increases in P digestibility with an overall improvement of 8% and 121% at 4000 phytase U/kg of diet, respectively, compared with the PC and DNC diets. Apparent total tract digestibility of N, OM and DM were higher (P < 0.05) in the PC diet compared with the DNC diet, but not the NC diet (P < 0.10). No effect of phytase addition to NC was observed on Ca, N, DM and OM digestibility. Phytase addition to the DNC diet resulted in a linear increase (P < 0.05) in N, DM and OM digestibility but not Ca. Increasing the levels of phytase supplementation in the NC and the DNC diets linearly decreased fecal P (P < 0.05) content by 45 and 42%, respectively. Adding phytase at 1000 or 4000 U/kg increased P retention (P < 0.05) by 14.3 or 15.6% units, respectively, compared with the PC diet. Urinary P excretion was higher in the group fed the PC diet compared with those fed the NC and DNC diets (P < 0.05). The results of this study show that complete removal of inorganic P from growing pig diets coupled with phytase supplementation improves digestibility and retention of P and N, thus reducing manure P excretion without any negative effect on pig performance.
