Hypoxia-sensing properties of the newborn rat ventral medullary surface in vitro

The ventral medullary surface (VMS) is a region known to exert a respiratory stimulant effect during hypercapnia. Several studies have suggested its involvement in the central inhibition of respiratory rhythm caused by hypoxia. We studied brainstem,spinal cord preparations isolated from newborn rats transiently superfused with a very low O2 medium, causing reversible respiratory depression, to characterize the participation of the VMS in hypoxic respiratory adaptation. In the presence of 0.8 mm Ca2+, very low O2 medium induced an increase in c-fos expression throughout the VMS. The reduction of synaptic transmission and blockade of the respiratory drive by 0.2 mm Ca2+,1.6 mm Mg2+ abolished c-fos expression in the medial VMS (at the lateral edge of the pyramidal tract) but not in the perifacial retrotrapezoid nucleus/parafacial respiratory group (RTN/pFRG) VMS, suggesting the existence of perifacial RTN/pFRG hypoxia-sensing neurons. In the presence of Ca2+ (0.8 mm), lesioning experiments suggested a physiological difference in perifacial RTN/pFRG VMS between the lateral VMS (beneath the ventrolateral part of the facial nucleus) and the middle VMS (beneath the ventromedial part of the facial nucleus), at least in newborn rats. The lateral VMS lesion, corresponding principally to the most rostral part of the pFRG, produced hypoxia-induced stimulation, whereas the middle VMS lesion, corresponding to the main part of the RTN, abolished hypoxic excitation. This may involve relay via the medial VMS, which is thought to be the parapyramidal group. [source]


Niche separation of ammonia-oxidizing bacteria across a tidal freshwater marsh

Summary Like many functional groups or guilds of microorganisms, the group of ammonia-oxidizing bacteria (AOB) consists of a number of physiologically different species or lineages. These physiological differences suggest niche differentiation among these bacteria depending on the environmental conditions. Species of AOB might be adapted to different zones in the flooding gradient of a tidal marsh. This issue has been studied by sampling sediments from different sites and depths within a tidal freshwater marsh along the river Scheldt near the village of Appels in Belgium. Samples were taken in February, April, July and October 1998. Communities of AOB in the sediment were analysed on the basis of the 16S rRNA gene by application of polymerase chain reaction in combination with denaturing gradient gel electrophoresis (DGGE). In addition, moisture content and concentrations of ammonium and nitrate were determined as well as the potential ammonia-oxidizing activities. Six different DGGE bands belonging to the ,-subclass of the Proteobacteria were observed across the marsh. The community composition of AOB was determined by the elevation in the flooding gradient as well as by the sampling depth. The presence of plants was less important for the community composition of AOB. DGGE bands affiliated with the Nitrosospira lineage were mostly found in the upper part of the marsh and in the deeper layers of the sediment. Two of the three DGGE bands related to the Nitrosomonas oligotropha lineage were more broadly distributed over the marsh, but were predominantly found in the upper layers of the sediment. Members of the environmental Nitrosomonas lineage 5 were predominantly detected in the deeper layers in the lower parts of the marsh. Potential driving factors for niche differentiation are discussed. [source]


In situ studies of the phylogeny and physiology of filamentous bacteria with attached growth

Summary Among the filamentous bacteria occasionally causing bulking problems in activated sludge treatment plants, three morphotypes with attached microbial growth are common, Eikelboom Type 0041, Type 1851 and Type 1701. A better knowledge of the phylogeny and physiology of these filamentous bacteria is necessary in order to develop control strategies for bulking. In this study we have used a combination of fluorescence in situ hybridization (FISH) and microautoradiography (MAR) to investigate the identity and in situ physiology of the Type 0041-morphotype and its attached bacteria in two wastewater treatment plants. Identification and enumeration of Type 0041 using group-specific 16S rRNA-targeted FISH probes revealed that approximately 15% of the filaments hybridized with a gene probe specific for the TM7 group, a recently recognized major lineage in the bacterial domain. All other filaments morphologically identified as Type 0041 only hybridized to the general bacterial EUB338-probe, indicating that they probably do not belong to commonly isolated bacterial phyla such as the Proteobacteria, Firmicutes, Actinobacteria and Bacteroidetes, for which group-specific probes were used. The phylogenetic heterogeneity of Type 0041 again highlights the inadequacy of a morphology-based classification system. Like the filaments, most of the attached microbial cells were not identified beyond their affiliation to the Bacteria using the group-specific FISH probes. However, several different bacterial phyla were represented in the identified fraction suggesting that the attached microorganisms are phylogenetically diverse. The study of the in situ physiology of Type 0041 using MAR-FISH revealed that both the filaments and the attached bacteria on Type 0041 were versatile in the use of organic substrates and electron acceptors. It was observed that all Type 0041 could consume glucose, but none of the filaments were able to consume acetate under any conditions tested, in contrast to some of the attached bacteria. No significant physiological differences were found between TM7,positive and TM7,negative Type 0041 filaments, and only minor differences were observed between the two treatment plants tested. These are the first data on the physiology of the almost entirely uncharacterized TM7 phylum and show that TM7 filamentous bacteria can uptake carbon substrates under aerobic and anaerobic conditions. V.A.C.® Therapy in the management of paediatric wounds: clinical review and experience

ABSTRACT Usage of negative pressure wound therapy (NPWT) in the management of acute and chronic wounds has grown exponentially in the past decade. Hundreds of studies have been published regarding outcomes and methods of therapy used for adult wounds. This treatment is increasingly being used to manage difficult-to-treat paediatric wounds arising from congenital defects, trauma, infection, tumour, burns, pressure ulceration and postsurgical complications in children, although relatively few studies have been aimed at this population. Given the anatomical and physiological differences between adults and children, a multidisciplinary expert advisory panel was convened to determine appropriate use of NPWT with reticulated open cell foam (NPWT/ROCF) as delivered by Vacuum Assisted Closure® (V.A.C.® Therapy, KCI Licensing, Inc., San Antonio, TX) for the treatment of paediatric wounds. The primary objectives of the expert advisory panel were to exchange state-of-practice information on paediatric wound care, review the published data regarding the use of NPWT/ROCF in paediatric wounds, evaluate the strength of the existing data and establish guidelines on best practices with NPWT/ROCF for the paediatric population. The proposed paediatrics-specific clinical practice guidelines are meant to provide practitioners an evidence base from which decisions could be made regarding the safe and efficacious selection of pressure settings, foam type, dressing change frequency and use of interposing contact layer selections. The guidelines reflect the state of knowledge on effective and appropriate wound care at the time of publication. They are the result of consensus reached by expert advisory panel members based on their individual clinical and published experiences related to the use of NPWT/ROCF in treating paediatric wounds. Best practices are described herein for novice and advanced users of NPWT/ROCF. Recommendations by the expert panel may not be appropriate for use in all circumstances. Decisions to adopt any particular recommendation must be made by the collaborating medical team, including the surgeon and wound care specialist based on available resources, individual patient circumstances and experience with the V.A.C.® Therapy System. Biogeography and microhabitat variation in temperate algal-invertebrate symbioses: zooxanthellae and zoochlorellae in two Pacific intertidal sea anemones, Anthopleura elegantissima and A. xanthogrammica

Abstract. Temperate sea anemones in the genus Anthopleura are unique among cnidarians in harboring two phylogenetically distinct symbiotic algae, zooxanthellae (golden-brown dinophytes, Symbiodinium) and zoochlorellae (green chlorophytes). To determine whether their physiological differences generate patterns in anemone habitat and biogeographic distribution, we sampled symbiotic algae in the small clonal A. elegantissima and the large solitary A. xanthogrammica at 8 field sites (and the other large solitary Anthopleura species at one site) spanning 18° of latitude along 2500 km of the Pacific coast of North America. We found that zoochlorellae predominate in low intertidal habitats and northerly latitudes and in A. xanthogrammica, while zooxanthellae constitute the majority of symbionts in high intertidal habitats and more southerly latitudes and in A. elegantissima. These data are consistent with published predictions based on photosynthetic efficiency of the two algae under varied temperature and light regimes in the laboratory. This anemone-algal system provides a potential biological signal of benthic intertidal communities' responses to El Niño events and long-term climate changes in the Pacific. [source]


Chemical form of dietary l -Carnitine affects plasma but not tissue Carnitine concentrations in male Sprague,Dawley rats

Summary In Experiment 1, rats (n = 54) were randomly assigned to control or one of the four sources of l -Carnitine supplemented at either 100 or 200 ,mol/kg/day and were allowed to acclimate for 14 days. Following a 12-h fast, plasma samples were obtained at 0, 5, 10, 15, 30, 60, 120, 240, 480 and 720 min after l -Carnitine feeding and assayed for free l -Carnitine concentration. Plasma-free l -Carnitine levels were affected by time after treatment intake (p < 0.0001) and l -Carnitine source (p < 0.0001). The time × source interaction was not statistically significant (p = 0.99). In Experiment 2, rats (n = 54) were randomly assigned to control or one of the four sources of l -Carnitine at either 100 or 200 ,mol/kg/day and were acclimated as in experiment 1. Rats were sacrificed 120 min after feeding. Samples of liver and skeletal muscle were obtained and assayed for free l -Carnitine concentration. Neither skeletal muscle (p = 0.44) or liver (p = 0.59) tissue concentrations of l -Carnitine were affected by any l -Carnitine source as compared with the control. We conclude that some differences exist in plasma concentrations of free l -Carnitine following ingestion of different chemical forms of l -Carnitine. It is unclear if these differences in the circulating concentration of free l -Carnitine translate into any physiological differences for the animal. In this study, chemical form of l -Carnitine had no effect on skeletal muscle or liver tissue concentrations of l -Carnitine in young male Wistar rats. [source]


Seasonal Variation and Correlation with Environmental Factors of Photosynthesis and Water Use Efficiency of Juglans regia and Ziziphus jujuba

Abstract Both the photosynthetic light curves and CO2 curves of Juglans regia L. and Ziziphus jujuba Mill. var. spinosa in three seasons were measured using a LI-6400 portable photosynthesis system. The maximal net photosynthetic rate (Amax), apparent quantum efficiency(,), maximal carboxylation rate (Vcmax) and water use efficiency (WUE) of the two species were calculated based on the curves. The results showed that Amax of J. regia reached its maximum at the late-season, while the highest values of Amax of Z. jujuba occurred at the mid-season. The Amax of J. regia was more affected by relative humidity (RH) of the atmosphere, while that of Z. jujuba was more affected by the air temperature. Light saturation point (LSP) and Light compensation point (LCP) of J. regia had a higher correlation with RH of the atmosphere, those of Z. jujuba, however, had a higher correlation with air temperature. Vcmax of both J. regia and Z. jujuba had negative correlation with RH of the atmosphere. WUE of J. regia would decrease with the rise of the air temperature while that of Z. jujuba increased. Thus it could be seen that RH, temperature and soil moisture had main effect on photosynthesis and WUE of J. regia and Z. jujuba. Incorporating data on the physiological differences among tree species into forest carbon models will greatly improve our ability to predict alterations to the forest carbon budgets under various environmental scenarios such as global climate change, or with differing species composition. Differential osmoregulatory capabilities of common spiny mice (Acomys cahirinus) from adjacent microhabitats

Abstract The osmoregulatory function of common spiny mice Acomys cahirinus living on opposite slopes of the lower Nahal Oren (,Evolution Canyon') on mount Carmel, Israel, was investigated by increasing the salinity of the water source whilst maintaining a high-protein diet. The southern-facing slope (SFS) of this canyon differs from the northern-facing slope (NFS) as it receives considerably more solar radiation and consequently forms a more xeric, sparsely vegetated habitat. During the summer, mice living on the two opposite slopes significantly differed in their urine osmolality, which also increased significantly as dietary salinity increased. Offspring of wild-captured mice, born in captivity, and examined during the winter, continued to show a difference in osmoregulatory function depending on the slope of origin. However, they differed from wild-captured mice, as they did not respond to the increase in dietary salinity by increasing the concentration of their urine, but rather by increasing the volume of urine produced. This study shows that A. cahirinus occupying different microhabitats may exhibit differences in their ability to concentrate urine and thus in their ability to withstand xeric conditions. We suggest that they may also differ genetically, as offspring from the NFS and SFS retain physiological differences, but further studies will be needed to confirm this hypothesis. [source]


Time Development Models for Perfusion Provocations Studied with Laser-Doppler Flowmetry, Applied to Iontophoresis and PORH

ABSTRACT Objective: Clinical acceptance of laser-Doppler perfusion monitoring (LDPM) of microcirculation suffers from lack of quantitatively reliable signal data, due to varying tissue constitution, temperature, hydration, etc. In this article, we show that a novel approach using physiological models for response upon provocations provides quantitatively and clinically relevant time constants. Methods: We investigated this for two provocation protocols: postocclusive reactive hyperemia (PORH) and iontophoresis shots, measured with LDPM on extremities. PORH experiments were performed on patients with peripheral arterial occlusive disease (PAOD) or diabetes mellitus (DM), and on healthy controls. Iontophoresis experiments were performed on pre-eclamptic patients and healthy controls. We developed two dynamical physical models, both based on two characteristic time constants: for PORH, an "arterial" and a "capillary" time constant and, for iontophoresis, a "diffusion" and a "decay" time constant. Results: For the different subject groups, we could extract time constants that could probably be related to physiological differences. For iontophoresis, a shot saturation constant was determined, with very different values for different groups and administered drugs. Conclusions: With these models, the dynamics of the provocations can be investigated and quantitative comparisons between experiments and subject groups become available. The models offer a quantifiable standard that is independent of the type of LDPM instrumentation. [source]


The biosynthesis of Juvenile Hormone, its degradation and titres in females of the true armyworm: a comparison of migratory and non-migratory populations

Summary In a previous study [McNeil et al. (1996) Archives of Insect Biochemistry and Physiology, 32, 575,584], patterns of sexual maturation and Juvenile Hormone (JH) biosynthesis were compared in virgin females from migratory (North American) and non-migratory (Azorean) populations of the true armyworm moth, Pseudaletia unipuncta Haworth (Lepidoptera: Noctuidae). Sexual maturation occurred at a significantly earlier age after emergence in the non-migrant population, and the rates of biosynthesis of JH in vitro suggested that lower titres of JH may be required to initiate the onset of calling behaviour (pheromone emission) and ovarian development in Azorean females. To examine the physiological differences in the reproductive biology of migratory and non-migratory populations in greater detail, the haemolymph titres of JH and JH esterase activity were compared in virgin females as a function of age. In addition, the effects of mating on JH biosynthesis in vitro, JH titres, JH esterase activity and egg production were measured in the two populations. As expected, JH titres rose more rapidly after emergence in Azorean females than in their North American counterparts but, contrary to our prediction, the maximum levels were also higher in the non-migrant population. Activity of JH esterase was much higher in Azorean females on the day of emergence. However, by the second day both populations had similar activity levels (about 17 nmol JH/min/ml) and exhibited a similar age-related decline in subsequent days. Mating did not affect the rate of JH biosynthesis in vitro but resulted in a significant increase in the titres of JH in the haemolymph of both populations. The maximum titre (a five-fold increase) occurred within 24 h of mating in Azorean females. In North American individuals the increase was greater (seven-fold) but did not occur until 48 h after mating. No difference in the activity of JH esterase was observed between mated and virgin North American females. By contrast, while there was an age-related decline in the activity of JH esterase in mated Azorean females, as seen in both North American groups, activity levels in virgin females remained constant with age. In all females, mating resulted in a significant increase in egg production within 24 h. The Azores is a volcanic archipelago, so these non-migratory populations were probably founded by immigrants originating from migratory continental populations. It is clear from our results that the change from a life history that includes migration to a non-migratory one involved more than just a temporal shift in the timing of the production of JH. Furthermore, the interpopulation differences in titres of JH and mating-induced changes reported here cannot be fully explained by the observed differences in the patterns of activity of JH esterase and JH biosynthesis in vitro. Blood-feeding and immunogenic Aedes aegypti saliva proteins

Abstract Mosquito-transmitted pathogens pass through the insect's midgut (MG) and salivary gland (SG). What occurs in these organs in response to a blood meal is poorly understood, but identifying the physiological differences between sugar-fed and blood-fed (BF) mosquitoes could shed light on factors important in pathogens transmission. We compared differential protein expression in the MGs and SGs of female Aedes aegypti mosquitoes after a sugar- or blood-based diet. No difference was observed in the MG protein expression levels but certain SG proteins were highly expressed only in BF mosquitoes. In sugar-fed mosquitoes, housekeeping proteins were highly expressed (especially those related to energy metabolism) and actin was up-regulated. The immunofluorescence assay shows that there is no disruption of the SG cytoskeletal after the blood meal. We have generated for the first time the 2-DE profiles of immunogenic Ae. aegypti SG BF-related proteins. These new data could contribute to the understanding of the physiological processes that appear during the blood meal. [source]


Sex, age, and family differences in the chemical composition of owl monkey (Aotus nancymaae) subcaudal scent secretions

Abstract Numerous behavioral studies have shown that animals use olfactory cues as inbreeding avoidance or kin avoidance mechanisms, implying that scent is unique to families. However, few studies have analyzed the chemical profile of a scent and ascertained the messages that are conveyed in scent secretions. Owl monkeys (Aotus nancymaae) are socially monogamous primates that utilize scent when interacting with foreign conspecifics. This suggests there is a difference in the chemical composition of scent marks. We chemically analyzed sub-caudal gland samples from three families of captive owl monkeys (Aotus nancymaae). Samples were analyzed by capillary GC-MS and relative retention time and fragment pattern was compared with known standards. Gland samples were high in large plant-based shikikate metabolites and fatty ketones; alcohols, acids, and acetates were virtually absent. Gender, age, and family could be reliably classified using discriminant analysis (92.9, 100, and 100%, respectively). Female scent profiles were greater in concentration of aromatic plant metabolites, possibly the result of a different diet or physiological differences in female metabolism as compared to male. Offspring of adult age still living in their natal group showed a less complex chemical profile than their parents. Finally, each family had its own unique and complex chemical profile. Effect of dietary protein and energy levels on growth, oxygen consumption, haemolymph and digestive gland carbohydrates, nitrogen excretion and osmotic pressure of Litopenaeus vannamei (Boone) and L. setiferus (Linne) juveniles (Crustacea, Decapoda; Penaeidae)

Abstract The influence of protein and energy levels on growth rate, survival, pre- and post-prandial oxygen consumption, ammonia excretion, haemolymph glucose (HG), glycogen in digestive gland and osmotic pressure (OP) in white shrimp Litopenaeus vannamei (Boone) and L. setiferus (Linne) juveniles was studied. Diets containing a high-quality protein at a protein/energy (P/E) ratio of 16, 26, 31 and 36 mg kJ,1 were fed at 20% of shrimp body weight of two sizes: < 1 g and >,1 g. Both species showed a optimum P/E ratio of 36 mg kJ,1 (33,44% protein and 6,23% carbohydrate) in juveniles <,1 g. For shrimp >,1 g, L. setiferus showed a higher growth rate in the diet with 16 mg kJ,1 (27% protein; 32% carbohydrate) and L. vannamei between 26 and 36 mg kJ,1 (33,44% protein and 6,23% carbohydrate). In both experiments, the growth rate of L. vannamei was 2,3 times that observed in L. setiferus. Routine oxygen consumption and apparent heat increment (AHI) of L. setiferus juveniles was two times higher than that observed in L. vannamei juveniles, which could indicate that L. setiferus has a higher metabolic rate. The O/N ratio varied according to protein level, with higher values (O/N = 180) with a 16-mg kJ,1 diet and lower values (O/N = 73) with a 36-mg kJ,1 diet in L. setiferus juveniles. A similar variation in O/N ratio was obtained in L. vannamei fed with all diets with an interval between 22 and 50. An inverse relation between ammonia excretion and HG, and digestive gland glycogen (DGG) in relation to an increase in the P/E ratio indicate that both shrimp species are well adapted to use carbohydrates and/or proteins from their diet. The higher values of hyper-osmotic capacity (hyper-OC) were observed in L. setiferus <,1 g fed with 36 mg kJ,1 and the lowest in L. vannamei <,1 g fed with 31 mg kJ,1. Intermediate values of hyper-OC were observed in both species fed all diets indicating that osmotic factors of juveniles <,1 g of both species are more affected by the P/E ratio than juveniles >,1 g. All results showed that juveniles >,1 g of both species are less dependent of P/E ratio than juveniles <,1 g. Litopenaeus vannamei is a most tolerant shrimp species with a high capacity to use a wide range of dietary P/E ratios for growth, which may be due to its lower energy requirements. Litopenaeus setiferus showed a lower capacity to accept different P/E ratios but the optimum P/E ratio obtained with this species shows that L. setiferus accept diets with a high carbohydrate level as well. These results demonstrate that there are nutritional and physiological differences that explain the differences that have been observed when both species were cultured in commercial ponds. [source]


Do physical activity levels influence the cardiovascular-related physiological characteristics of postmenopausal women?

Aim:, This study aimed to determine whether cardiovascular-related physiological differences existed among postmenopausal women in relation to their physical activity levels. Methods:, Participants were postmenopausal women (n= 101) resident in North Queensland. A self-report questionnaire determined recent exercise history. Anthropometric and physiological measures were obtained. Participants also performed a six-minute graded exercise test to determine cardiorespiratory fitness. Results:, Compared with the women who exercised, those women who did not exercise had a lower level of cardiorespiratory fitness (P= 0.00) and higher resting diastolic blood pressure (P= 0.01), BMI (P= 0.00) and WHR (P= 0.02). Discriminant function analysis found that a combination of BMI and cardiorespiratory fitness discriminated between the two groups. Conclusions:, Postmenopausal women who performed moderate-intensity physical activity had more favourable cardiovascular-related physiological characteristics. Health professionals should encourage more postmenopausal women to participate in moderate-intensity activity to reduce the risk of cardiovascular disease. [source]
