Selected AbstractsA VANADIUM BROMOPEROXIDASE CATALYZES THE FORMATION OF HIGH-MOLECULAR-WEIGHT COMPLEXES BETWEEN BROWN ALGAL PHENOLIC SUBSTANCES AND ALGINATES,JOURNAL OF PHYCOLOGY, Issue 1 2009Leonardo Tavares Salgado The interaction between phenolic substances (PS) and alginates (ALG) has been suggested to play a role in the structure of the cell walls of brown seaweeds. However, no clear evidence for this interaction was reported. Vanadium bromoperoxidase (VBPO) has been proposed as a possible catalyst for the binding of PS to ALG. In this work, we studied the interaction between PS and ALG from brown algae using size exclusion chromatography (SEC) and optical tweezers microscopy. The analysis by SEC revealed that ALG forms a high-molecular-weight complex with PS. To study the formation of this molecular complex, we investigated the in vitro interaction of purified ALG from Fucus vesiculosus L. with purified PS from Padina gymnospora (Kütz.) Sond., in the presence or absence of VBPO. The interaction between PS and ALG only occurred when VBPO was added, indicating that the enzyme is essential for the binding process. The interaction of these molecules led to a reduction in ALG viscosity. We propose that VBPO promotes the binding of PS molecules to the ALG uronic acids residues, and we also suggest that PS are components of the brown algal cell walls. [source] Willow genotype, but not drought treatment, affects foliar phenolic concentrations and leaf-beetle resistanceENTOMOLOGIA EXPERIMENTALIS ET APPLICATA, Issue 1 2004Carolyn Glynn Abstract In a greenhouse experiment we examined the effect of willow genotype and irrigation regime (moderate drought and well-watered) on plant growth parameters, foliar nitrogen, and phenolic concentrations, as well as on the preference and performance of the blue leaf beetle, Phratora vulgatissima (L.) (Coleoptera: Chrysomelidae). The 10 vegetatively propagated willow genotypes in the experiments were F2 full-sibling hybrids, originated from a cross between Salix viminalis (L.) (Salicaceae) (high in condensed tannins) and Salix dasyclados (L.) (Salicaceae) (rich in phenolic glycosides). Insect bioassays were conducted on detached leaves in Petri dishes as well as with free-living insects on intact potted plants. The 10-week long irrigation treatments caused statistically significant phenotypic differences in the potted willow saplings. Total biomass was somewhat higher in the well-watered treatment. The root to total biomass ratio was higher in the drought-treatment plants. There was significant genotypic variation in foliar nitrogen concentrations, and they were higher in the drought-treatment plants. There was also a strong genotypic variation in each of the phenolic substances analyzed. Condensed tannins, which accounted for the greatest proportion of total phenolic mass, were higher in the well-watered treatment. There was, however, no difference in levels of the other phenolics (salicylates, cinnamic acid, flavonoids, and chlorogenic acid) between irrigation treatments. The sum of these phenolics was higher in the well-watered treatment. There was a strong variation in P. vulgatissima larval development on different willow genotypes, and larval performance was negatively correlated with levels of salicylates and cinnamic acid. There was, however, no effect of irrigation treatment on larval performance. Phratora vulgatissima preferred to feed on well-watered plants, and we found a preference for oviposition there, but neither feeding nor oviposition site preference was affected by willow genotype. Adult feeding and oviposition preferences were not correlated with larval performance. [source] Sensitivity of Crops to Increased Ultraviolet Radiation in Northern Growing ConditionsJOURNAL OF AGRONOMY AND CROP SCIENCE, Issue 1 2002K. Hakala Crops growing at high latitudes are adapted to low intensities of solar ultraviolet B radiation (UV-B). As a result of destruction of stratospheric ozone layer especially at the Polar Regions of the globe, the intensity of UV-B is projected to increase at high latitudes. In order to find out the consequences of increased UV-B on crops, the sensitivity of different varieties of barley, wheat, oats, clover, timothy, meadow fescue, tall fescue and potato was tested in Jokioinen, Finland. The experiment was conducted in the field, under natural light, with a continuous 30 % increase in UV-B. Both increased UV-B and the control with slightly increased UV-A radiation affected the number of lateral shoots in cereals, and the specific leaf area and the content of phenolic substances in grasses. The effects varied according to plant variety and time of growing season. However, no visible damage, nor changes in biomass and yield production were found in any of the crops. Long-term effects on, e.g. genetic stability and germination of crop seed remain to be studied. Empfindlichkeit von Kulturpflanzenbeständen gegenüber einer Erhöhung der ultravioletten Einstrahlung unter nördlichen Wachstumsbedingungen Kulturpflanzenbestände, die unter hohen Breitengraden wachsen, sind an geringe Intensitäten der solaren ultravioletten B Einstrahlung (UV-B) angepasst. Aufgrund der Destruktion der Stratosphäre, insbesondere der Ozonschicht, im Bereich der polaren Gebiete der Erde, ist eine zunehmende Intensität der UV-B Strahlung in hohen Breitengraden festzustellen. Um die Konsequenzen einer Erhöhung von UV-B auf Kulturpflanzenbestände zu bestimmen, wurde die Empfindlichkeit unterschiedlicher Arten wie Gerste, Weizen, Hafer, Klee, Wiesenlieschgras, Wiesenschwingel, Rohrschwingel und Kartoffel in Jokioinen, Finnland, untersucht. Das Experiment wurde im Felde durchgeführt unter natürlichen Lichtbedingungen mit einer kontinuierlichen Zunahme von 30 % für UV-B. Sowohl eine Erhöhung der UV-B als auch die Kontrolle mit leicht erhöhten UV-B Strahlungen beeinflussten die Anzahl der Nebenwurzeln bei den Getreidepflanzen und die spezifische Blattfläche sowie den Gehalt an phenolischen Substanzen bei den Gräsern. Die Wirkungen variierten entsprechend der Pflanzenvarietät und der Anbauzeit. Allerdings wurden keine erkennbare Schädigungen noch Änderungen in der Biomasse und in der Ertragsproduktion bei einer der untersuchten Pflanzenbestände gefunden. Langzeitwirkungen auf die genetische bedingte Stabilität und Keimung der Kulturpflanzen sollte weiterhin untersucht werden. [source] LIPID-LOWERING EFFECTS OF ARONIA MELANOCARPA FRUIT JUICE IN RATS FED CHOLESTEROL-CONTAINING DIETSJOURNAL OF FOOD BIOCHEMISTRY, Issue 5 2007S. VALCHEVA-KUZMANOVA ABSTRACT Aronia melanocarpa fruit juice (AMFJ) is very rich in phenolic antioxidants, mainly flavonoids from the subclass anthocyanins. The aim of this study was to assess the influence of AMFJ on body and liver mass, plasma lipids and lipoprotein profiles, and the histopathology of liver and aorta in rats fed with cholesterol diets. AMFJ was applied orally for 30 days at doses of 5, 10 and 20 mL/kg. In rats fed the cholesterol-containing diets, AMFJ significantly hindered an increase in plasma lipids (total cholesterol, low-density lipoprotein cholesterol and triglycerides) because of cholesterol feeding. Body weight gains, liver weights, and liver and aorta histopathology were not influenced either by high-cholesterol diets or by AMFJ treatment. In conclusion, AMFJ showed lipid-lowering effects in rats with experimentally induced hyperlipidemia, and could be valuable in reducing lipidemia as a factor of cardiovascular risk. PRACTICAL APPLICATIONS Hyperlipidemia characterized by an increase in low-density lipoprotein (LDL) cholesterol and a decrease in high-density lipoprotein cholesterol is one of the major risk factors for atherosclerosis and cardiovascular disease. Plant foods with high contents of phenolic phytochemicals are reported to be inversely correlated with plasma total cholesterol (TC) and LDL cholesterol. Aronia melanocarpa fruits are remarkably rich in phenolic substances. They are used for human consumption as juice, syrup, jam and wine. Our research demonstrated that A. melanocarpa fruit juice hindered the dietary-induced elevation of plasma TC, LDL cholesterol and triglycerides in rats. In view of the results from our experiment, we can suppose that the juice may be further tested for reducing hyperlipidemia in humans and possibly approved a valuable dietary supplement. [source] Phenolic compounds in some apple (Malus domestica Borkh) cultivars of organic and integrated productionJOURNAL OF THE SCIENCE OF FOOD AND AGRICULTURE, Issue 10 2005Robert Veberic Abstract Eleven organically grown apple cultivars and 11 apple cultivars of integrated production from Austria and Slovenia were analyzed by HPLC for the content of phenolic compounds in peel and pulp. We identified chlorogenic acid, p -coumaric acid, procyanidin B3, protocatechuic acid, (,)-epicatechin, phloridzin, rutin and quercetin-3-rhamnoside in apple peel. In apple pulp, (+)-catechin was also identified in all the cultivars. Some other phenols (procyanidin B3, rutin and quercetin-3-rhamnoside) could not be identified or were not properly separated. With regard to the phenolic content in the apple peel, there were no differences between organically grown apple cultivars and apple cultivars of integrated production. Organically grown apples, however, exhibited a higher content of phenolic substances in the apple pulp compared with the apple cultivars of integrated production. This may be due either to the different genotype source or to the growing technology. Higher concentrations of phenolic compounds in organically grown cultivars could be a result of plant response to stress. The apple peel contained higher concentrations of identified phenols than the pulp. The apple peel represents up to 10% of the whole fruit; therefore the phenolsic compounds in the pulp are of greater importance to the consumer than the phenolic compounds in the peel. Here, we report the isolation and structural characterization of two new phenol derivatives, named alpestrin (=3,3,,5,-trimethoxy[1,1,-biphenyl]-4-ol; 10) and alpestriose,A (=6- O -benzoyl-1- O -{6- O -acetyl-3- O -[(4-hydroxy-3,5-dimethoxyphenyl)prop-2-enoyl]- , - D -fructofuranosyl}- , - D -glucopyranoside; 11), and of four known compounds (12,15) from the MeOH extract of Polygala alpestris. The relative in vitro antioxidant activities of these compounds, in comparison with other phenolic substances from Polygala vulgaris, were evaluated by means of the BriggsRauscher (BR) oscillating reaction, a method based on the inhibitory effects of antioxidant free-radical scavengers. The experimental antioxidant-activity values (relative to resorcinol as a standard) were compared with those calculated on the basis of the bond-dissociation enthalpies. The structure/activity relationships for the compounds examined are also discussed. [source] |
