    Counterfeiting in performance- and image-enhancing drugs

    Abstract The current drastic escalation in obesity may be contributing to the exponential rise in drugs used for image enhancement. Drugs such as anabolic-androgenic steroids (AAS) are perceived as a viable method of achieving a perfect physique. They are also the most widely abused drugs in sport. The Internet has encouraged the abuse of expensive drugs, particularly human growth hormone (hGH), resulting in increased importation for personal use. The substantial increase in this market has opened up avenues for counterfeiting, estimated as a multi-million pound business. The acute adverse effects from contaminated vials may result in a variety of pathologies including communicable diseases. In 2007, in the UK, a series of intramuscular abscesses, requiring surgical treatment, led us to study samples obtained from the underground market. The analysis of 38 parenteral samples and 19 oral samples of tablets was performed by a World Anti-Doping Agency (WADA) accredited laboratory, in an attempt to establish the extent of available counterfeit products. Fifty-three per cent (20) of the injectable AAS esters and 21% (4) of the oral tablets were counterfeit. Culture and sensitivity revealed the presence of skin commensal organisms, which may have contributed to the development of the abscesses. Users of AAS and hGH for sport, including bodybuilding, are currently risking their health because of counterfeit and poorly controlled products. Copyright © 2009 John Wiley & Sons, Ltd. [source]


    Gas6 and protein S

    Vitamin K-dependent ligands for the Axl receptor tyrosine kinase subfamily
    Gas6 and protein S are two homologous secreted proteins that depend on vitamin K for their execution of a range of biological functions. A discrete subset of these functions is mediated through their binding to and activation of the receptor tyrosine kinases Axl, Sky and Mer. Furthermore, a hallmark of the Gas6,Axl system is the unique ability of Gas6 and protein S to tether their non receptor-binding regions to the negatively charged membranes of apoptotic cells. Numerous studies have shown the Gas6,Axl system to regulate cell survival, proliferation, migration, adhesion and phagocytosis. Consequently, altered activity/expression of its components has been detected in a variety of pathologies such as cancer and vascular, autoimmune and kidney disorders. Moreover, Axl overactivation can equally occur without ligand binding, which has implications for tumorigenesis. Further knowledge of this exquisite ligand,receptor system and the circumstances of its activation should provide the basis for development of novel therapies for the above diseases. [source]


    Angiogenesis in the female reproductive organs: pathological implications

    Summary. The female reproductive organs (ovary, uterus, and placenta) are some of the few adult tissues that exhibit regular intervals of rapid growth. They also are highly vascular and have high rates of blood flow. Angiogenesis, or vascular growth, is therefore an important component of the growth and function of these tissues. As with many other tissues, vascular endothelial growth factors (VEGFs) and fibroblast growth factors (FGFs) appear to be major angiogenic factors in the female reproductive organs. A variety of pathologies of the female reproductive organs are associated with disturbances of the angiogenic process, including dysfunctional uterine bleeding, endometrial hyperplasia and carcinoma, endometriosis, failed implantation and subnormal foetal growth, myometrial fibroids (uterine leiomyomas) and adenomyosis, ovarian hyperstimulation syndrome, ovarian carcinoma, and polycystic ovary syndrome. These pathologies are also associated with altered expression of VEGFs and/or FGFs. In the near future, angiogenic or antiangiogenic compounds may prove to be effective therapeutic agents for treating these pathologies. In addition, monitoring of angiogenesis or angiogenic factor expression may provide a means of assessing the efficacy of these therapies. [source]


    Nucleolar organizer regions (NORs) evaluation of lingual salivary glands of chronic alcoholics

    Abstract Background: ,Chronic alcoholism has been associated with structural and physiological changes in salivary glands. Studies on a variety of pathologies have suggested that variation in number of nucleolar organizer regions (NORs) reveals conditions of cellular activity. The aim of this work was to examine, through the AgNOR technique, changes in number and size of NORs in lingual salivary glands of chronic alcoholics. Methods:, Samples of mucous and serous lingual salivary glands were obtained from tongues from autopsies of individuals whose cause of death was hepatic alcoholic cirrhosis. Lingual organs from individuals whose cause of death was accidental were used as controls. Number and size of the AgNORs and nuclear area, in ductal and acinar cells, were evaluated through a digital image analyzer. Results:, Statistical analysis revealed differences (P , 0.05) in number of AgNORs in mucous acini and ductal cells. Also, we observed changes in the area of the NORs. Conclusion:, These results suggest that in alcoholics the activity of glandular cells, mainly in ductal epithelium, could be affected, modifying synthesis, transport and salivary secretions.     Fructus Ligustrum lucidi inhibits inflammatory mediator release through inhibition of nuclear factor-,B in mouse peritoneal macrophages

    ABSTRACT Fructus Ligustrum lucidi (FLL) is a widely used herbal medicine for the treatment of a variety of pathologies. We have investigated the anti-inflammatory mechanism of FLL in mouse peritoneal macrophages. FLL exerted an anti-inflammatory action through inhibition of lipopolysaccharide (LPS)-induced tumour necrosis factor (TNF)-, production in mouse peritoneal macrophages. The maximal inhibition rate of TNF-, production by FLL (0.5 mg mL,1) was 60.88 + 0.30%. In the inflammatory process, nitric oxide (NO) and prostaglandin E2 (PGE2) increased in peritoneal macrophages. FLL decreased the protein level of NO and PGE2 in LPS-stimulated mouse peritoneal macrophages. In addition, FLL inhibited nuclear factor-,B activation and I,B-, degradation by the decrease in I,B-, phosphorylation. Our study suggested that FLL reduced inflammation via an important molecular mechanism, which might explain its beneficial effect in the regulation of inflammatory reactions. [source]
