    Changes in capillary luminal diameter in rat soleus muscle after hind-limb suspension

    This study examined the time course change of the capillary luminal diameter and the number of capillaries in the rat soleus muscle during hind-limb suspension. Male Wistar rats were divided into 1 and 3 weeks of hind-limb suspension (HS) groups (HS-1 and HS-3). The HS groups were compared with age-matched control groups. All morphometric parameters with respect to capillary and muscle fibre cross-sectional area were determined in perfusion-fixed soleus muscles. After 1 and 3 weeks of hind-limb suspension, the mean muscle fibre cross-sectional area was significantly decreased in HS-1 (,32.0%) and HS-3 (,59.3%) compared with age-matched control groups. Despite a lower capillary-to-fibre ratio (HS-1, ,19.3%; HS-3, ,21.2%), the capillary density was unchanged in HS-1 and significantly increased in HS-3 compared with age-matched control groups. The mean capillary luminal diameter was significantly smaller in HS-1 (,19.9%) and HS-3 (,21.9%) than in the age-matched control groups. The capillary-to-fibre perimeter ratio which indicates the capillary surface area available for gas exchange between blood and tissue did not significantly differ between control groups and HS groups. In conclusion, the morphometrical adaptations in rat soleus with the suspension involved changes in both the capillary luminal diameter and number of capillaries, and the change in capillary surface area was proportional to the degree of muscle atrophy in HS groups.     Small-scale variability in surface moisture on a fine-grained beach: implications for modeling aeolian transport

    Abstract Small-scale variations in surface moisture content were measured on a fine-grained beach using a Delta-T Theta probe. The resulting data set was used to examine the implications of small-scale variability for estimating aeolian transport potential. Surface moisture measurements were collected on a 40 cm × 40 cm grid at 10 cm intervals, providing a total of 25 measurements for each grid data set. A total of 44 grid data sets were obtained from a representative set of beach sub-environments. Measured moisture contents ranged from about 0% (dry) to 25% (saturated), by weight. The moisture content range within a grid data set was found to vary from less than 1% to almost 15%. The magnitude of within-grid variability varied consistently with the mean moisture content of the grid sets, following an approximately normal distribution. Both very wet and very dry grid data sets exhibited little internal variability in moisture content, while intermediate moisture contents were associated with higher levels of variability. Thus, at intermediate moisture contents it was apparent that some portions of the beach surface could be dry enough to allow aeolian transport (i.e. moisture content is below the critical threshold), while adjacent portions are too wet for transport to occur. To examine the implications of this finding, cumulative distribution functions were calculated to model the relative proportions of beach surface area expected to be above or below specified threshold moisture levels (4%, 7%, and 14%). It was found that the implicit inclusion of small-scale variability in surface moisture levels typically resulted in changes of less than 1% in the beach area available for transport, suggesting that this parameter can be ignored at larger spatial scales.     Integrated selective enrichment target , a microtechnology platform for matrix-assisted laser desorption/ionization-mass spectrometry applied on protein biomarkers in prostate diseases

    Abstract The performance of a miniaturized sample processing platform for matrix-assisted laser desorption/ionization-mass spectrometry (MALDI-MS), manufactured by silicon microfabrication, called integrated selective enrichment target (ISET) technology was evaluated in a biological context. The ISET serves as both sample treatment device and MALDI-MS target, and contains an array of 96 perforated nanovials, which each can be filled with 40 nL of reversed-phase beads. This methodology minimizes the number of sample transfers and the total surface area available for undesired adsorption of the analytes in order to provide high-sensitivity analysis. ISET technology was successfully applied for characterization of proteins coisolated by affinity chromatography of prostate-specific antigen (PSA) from human seminal fluid. The application of ISET sample preparation enabled multiple analyses to be performed on a limited sample volume, which resulted in the discovery that prolactin inducible protein (PIP) was coisolated from the samples.     Benzo[a]pyrene bioavailability from pristine soil and contaminated sediment assessed using two in vitro models

    Abstract A major route of exposure to hydrophobic organic contaminants (HOCs), such as benzo[a]pyrene (BaP), is ingestion. Matrix-bound HOCs may become bioavailable after mobilization by the gastrointestinal fluids followed by sorption to the intestinal epithelium. The purpose of this research was to measure the bioavailability of [14C]-BaP bound to pristine soils or field-contaminated sediment using an in vitro model of gastrointestinal digestion followed by sorption to human enterocytes (Caco-2 cells) or to a surrogate membrane, ethylene vinyl acetate (EVA) thin film. Although Caco-2 cells had a twofold higher lipid-normalized fugacity capacity than EVA, [14C]-BaP uptake by Caco-2 lipids and EVA thin film demonstrated a linear relationship within the range of BaP concentrations tested. These results suggest that EVA thin film is a good membrane surrogate for passive uptake of BaP. The in vitro system provided enough sensitivity to detect matrix effects on bioavailability; after 5 h, significantly lower concentrations of [14C]-BaP were sorbed into Caco-2 cells from soil containing a higher percentage of organic matter compared to soil with a lower percentage of organic matter. The [14C]-BaP desorption rate from Caco-2 lipids consistently was twofold higher than from EVA thin film for all matrices tested. The more rapid kinetics observed with Caco-2 cells probably were due to the greater surface area available for absorption/desorption in the cells. After 5 h, the uptake of BaP into Caco-2 lipid was similar in live and metabolically inert Caco-2 cells, suggesting that the primary route of BaP uptake is by passive diffusion. Moreover, the driving force for uptake is the fugacity gradient that exists between the gastrointestinal fluid and the membrane. [source]


    Is food availability limiting African Penguins Spheniscus demersus at Boulders?

    A comparison of foraging effort at mainland, island colonies
    The African Penguin Spheniscus demersus (Vulnerable) formed three new colonies during the 1980s, two on the South African mainland (Stony Point and Boulders) and one on Robben Island. One of the mainland colonies, at Boulders, Simon's Town, is in a suburban area, resulting in conflict with humans. Growth of the Boulders colony was initially rapid, largely through immigration, but has since slowed, possibly as a result of density-dependent effects either on land (where there has been active management to limit the spread of the colony) or at sea. We test the latter hypothesis by comparing the foraging effort of Penguins feeding small chicks at island and mainland sites, and relate this to the foraging area available to birds. Three-dimensional foraging paths of African Penguins were reconstructed using GPS and time,depth loggers. There were no intercolony differences in the rate at which birds dived during the day (33 dives/h), in diving depths (mean 17 m, max. 69 m) or in travelling speeds. The maximum speed recorded was 2.85 m/s, with birds travelling faster when commuting (average 1.18 m/s) than when foraging (0.93 m/s) or resting at sea (0.66 m/s during the day, 0.41 m/s at night). There were strong correlations between foraging trip duration, foraging range and total distance travelled. Foraging effort was correlated with chick age at Robben Island, but not at Boulders. Contrary to Ashmole's hypothesis, birds from Boulders (c. 1000 pairs) travelled further (46,53 km) and foraged for longer (13.2 h) than did birds from Robben Island (c. 7000 pairs) and Dassen Island (c. 21 000 pairs) (33 km, 10.3 h). The mean foraging range also differed significantly between mainland (18,20 km) and island colonies (9 km). The area available to central-place-foraging seabirds breeding on the mainland is typically less than that for seabirds breeding on islands, but the greater foraging range of Boulders birds results in an absolute foraging area roughly twice that of island colonies, and the area per pair is an order of magnitude greater for the relatively small Boulders colony. Ashmole's hypothesis assumes relatively uniform prey availability among colonies, but our results suggest this does not apply in this case. The greater foraging effort of Boulders birds probably reflects reduced prey availability in False Bay, and thus the recent slowing in growth at the colony may be the result of differential immigration rather than management actions to limit the spatial growth of the colony.     Predicting the effects of marine climate change on the invertebrate prey of the birds of rocky shores

    By the end of the 21st century models of climate change predict that the air temperature over most of the British Isles will increase by between 2 and 3 °C and sea-level will rise by 40,50 cm. Over that period it will become windier and mean wave height will increase, as will the frequency of storms. These changes in climate and weather will impact the intertidal zone of the UK and will cause distribution changes in many of the common invertebrate species that live there. Where these changes are severe they may well impact on patterns of distribution of ducks and wading birds. In the British Isles a number of organisms live close to their geographical limits of distribution. Some of these species might be expected to extend their range as climatic restraints are relaxed. Species currently limited by cool summers or winter cold will move northwards. In most cases the effects on the distribution of waterbirds will be small. For example, the replacement of the Northern Limpet Patella vulgata by the Southern Limpet P. depressa is unlikely to adversely affect Eurasian Oystercatchers Haematopus ostralegus. Of wider concern is the possibility that as climate warms the abundance and productivity of brown algae will decrease. This is likely to have two significant effects for waders. First, it would represent a loss of potentially rich feeding grounds for species such as Ruddy Turnstone Arenaria interpres that feed on small easily desiccated invertebrates living on or below the seaweed. Secondly, as algae die or are broken away the resulting debris is exported to sediment habitats where it considerably boosts the in situ production of bacteria at the base of the food web. An increase in sea-level will only have a major impact on the extent of rocky shore invertebrate communities where shore topography prevents the upward migration of the biota. Where a seawall limits shores, for example, biological production will be curtailed as the area available for colonization decreases. Increases in the size of waves and the frequency of storms will mimic increasing exposure and there will be a significant reduction in algal production in areas that are affected. [source]


    Distributive mixing profiles for co-rotating twin-screw extruders

    Distributive mixing was experimentally measured during polymer melt blending along the length of a co-rotating twin-screw extruder. A mixing limited interfacial reaction between two reactive polymer tracers was employed to gain direct evidence of the generation of interfacial area. Model reactions were performed to validate this novel experimental method. In particular, the conversion was a direct indicator of the interfacial area available for the reaction. Specially designed sampling ports were used to obtain polymer samples along the length of the extruder during its continuous operation. The mixing capabilities of conveying sections and kneading blocks were compared over a wide range of operating conditions. In conveying sections, distributive mixing was controlled by the fully filled fraction.     A hollow fiber membrane photo-bioreactor for CO2 sequestration from combustion gas coupled with wastewater treatment: a process engineering approach

    Abstract BACKGROUND: In the presence of light, micro-algae convert CO2 and nutrients to biomass that can be used as a biofuel. In closed photo-bioreactors, however, light and CO2 availability often limit algae production and can be difficult to control using traditional diffuser systems. In this research, a hollow fiber membrane photo-bioreactor (HFMPB) was investigated to: (1) increase the interfacial contact area available for gas transfer, (2) treat high nutrient strength (412 mg NO3, -N L,1) wastewater, and (3) produce algal biomass that can be used as a biofuel. RESULTS: A bench scale HFMPB was inoculated with Spirulina platensis and operated with a 2-15% CO2 supply. A mass transfer model was developed and found to be a good tool to estimate CO2 mass transfer coefficients at varying liquid velocities. Overall mass transfer coefficients were 1.8 × 10,6, 2.8 × 10,6, 5.6 × 10,6m s,1 at Reynolds numbers of 38, 63, and 138, respectively. A maximum CO2 removal efficiency of 85% was observed at an inlet CO2 concentration of 2% and a gas residence time (membrane-lumen) of 8.6 s. The corresponding algal biomass concentrations and NO3 removal efficiencies were 2131 mg L,1 and 68%, respectively. CONCLUSION: The results show that the combination of CO2 sequestration, wastewater treatment and biofuel production in an HFMPB is a promising alternative for greenhouse gas mitigation.     Feeding on roots in the humus layer by adult pine weevil, Hylobius abietis

    Abstract 1,The consumption by adult pine weevil, Hylobius abietis, of the bark of roots present in the humus layer was assessed in a field study conducted in southern Sweden during two years (1998 and 2002). The study sites were divided into two areas: (i) a shelterwood where 80,100 mature Scots pine trees per hectare remained after cutting and (ii) a clearcut where no trees were left. 2,On average, 3741 m2 per hectare of root bark was present in the humus layer, of which 135 m2 was not coniferous but comprised species such as bilberry and broadleaved trees. 3,The mean area debarked by pine weevils was 2.9 m2 per hectare; 2.6 m2 of conifer roots and 0.3 m2 of bilberry roots. Roots of broadleaved trees were almost never consumed. No clear preferences for roots of a specific level of vitality were observed. 4,No consistent difference between the shelterwood and clearcut was found, either in the amount of root bark area available or in the extent of root feeding by pine weevil. 5,A weak negative correlation between debarked areas on roots and seedlings was found, indicating that root feeding may have reduced damage to seedlings. 6,It is concluded that conifer roots in the humus layer constitute a major food source for the pine weevil and can be utilized for a considerable period in both clearcuts and shelterwoods.     Mining and sustainable development: considerations for minerals supply

    Abstract Sustainable development involves meeting the needs of human societies while maintaining viable biological and physical Earth systems. The needs include minerals: metals, fuels, industrial and construction materials. There will continue to be considerable demand for virgin mineral resources, even if levels of recycling and efficiency of use are optimal, and rates of population growth and globalisation decrease significantly. This article aims to stimulate debate on strategic issues for minerals supply. While the world has considerable stocks of mineral resources overall, international considerations of the environmental and social aspects of sustainable development are beginning to result in limitations on where mining will be conducted and what types of deposits will be mined. Current and emerging trends favour large mines in parts of the world where mining can be conducted within acceptable limits of environmental and social impact. Finding new deposits that meet such criteria will be all the more challenging given a disturbing global decline in the rate of discovery of major economic resources over the last decade, and the decreasing land area available for exploration and mining. To attract responsible exploration and mining, governments of mining nations will need to provide: regional-scale geo-scientific datasets as required to attract and guide future generations of exploration; resource access through multiple and sequential land use regimes, and frameworks for dealing with indigenous peoples' issues; and arrangements for consideration of mining proposals and regulation of mines that ensure responsible management of environmental and social issues. The minerals industry will need to continue to pursue advances in technologies for exploration, mining, processing, waste management and rehabilitation, and in public reporting of environmental and social performance.     Disproportional effects of Igf2 knockout on placental morphology and diffusional exchange characteristics in the mouse

    Both complete knockout of the Igf2 gene (Igf2null+/,) and knockout of its placental specific transcript alone (Igf2P0+/,) lead to fetal growth restriction in mice. However, in the Igf2null+/, this growth restriction occurs concurrently in gestation with placental growth restriction, whereas, placental growth restriction precedes fetal growth restriction in the Igf2P0+/, mouse. Previous studies have shown that the Igf2P0+/, placenta has proportionate reductions in its cellular compartments and its diffusional exchange characteristics. Yet, nothing is known about the structural development or diffusional exchange characteristics of the Igf2null+/, mouse. Hence, this study compares the structural properties (using stereology) and diffusional exchange characteristics (using measurement of permeability,surface area product, P.S, of three inert hydrophilic tracers) of the Igf2null+/, and the Igf2P0+/, placenta to identify the role of Igf2 in the development of the labyrinthine exchange membrane and its functional consequences. Our data show disproportionate effects of complete Igf2 ablation on the compartments of the placenta, not seen when the placental-specific transcript alone is deleted. Furthermore, although the theoretical diffusing capacity (calculated from the stereological data) of the Igf2null+/, placenta was reduced relative to control, there was no effect of the complete knockout on permeability surface area available for small hydrophilic tracers. This is in contrast to the Igf2P0+/, placenta, where theoretical diffusion capacity and P.S values were reduced similarly. Total ablation of the Igf2 gene from the fetoplacental unit in the mouse therefore results in a disproportionate growth of placental compartments whereas, deleting the placental specific transcript of Igf2 alone results in proportional placental growth restriction. Thus, placental phenotype depends on the degree of Igf2 gene ablation and the interplay between placental and fetal Igf2 in the mouse. [source]


    Effects of the Cardiff Bay tidal barrage on the abundance, ecology and behaviour of shelducks Tadorna tadorna

    Abstract 1.Closure of the Taff/Ely Estuary by the Cardiff Bay barrage in 1999 resulted in the replacement of intertidal mudflats by a permanent freshwater lake. This led to an 89% reduction in the population of shelducks Tadorna tadorna. 2.The birds switched from foraging mainly for Nereis diversicolor and Hydrobia ulvae by scything with the bill as they walked across the mudflats at low tide, to feeding on benthic chironomid midge larvae while swimming in shallow water around the margins of the lake. 3.The population decline occurred as a consequence of a decrease in the area available for foraging,,,from about 1,km2 of mudflats to about 0.1,km2 of water shallow enough for shelducks to reach the bottom when dabbling, head dipping and upending. 4.Contrary to expectation, the amount of time shelducks spent feeding was similar pre- and post-barrage, and their body and plumage condition improved. 5.A tidal rhythm in activity persisted, with a reduced amount of feeding at high tide, probably because of the slight rise in water levels that sometimes occurred at this time.     Anatomical delineation of a safety zone for drilling the internal acoustic meatus during surgery for vestibular schwanomma by retrosigmoid suboccipital approach

    Abstract Drilling the internal acoustic meatus (IAM) is an important step during surgical removal of vestibular schwanommas. During this maneuver, the proximity of the jugular bulb and the saccus endolymphaticus to the IAM makes these structures prone to injury. There have been studies in the past on the relationship between a high jugular bulb (HJB) and the IAM, but there is no description of a safety area available for drilling the meatus. In the present study, measurements were made between the IAM and the nearest points on the top of the jugular bulb, the saccus endolymphaticus and the petrous ridge. The incidence of a HJB was also calculated. It was found that about 5 mm of bone could be safely drilled away from the posterior lip of the IAM without damaging either the jugular bulb or the saccus endolymphaticus in most of the cases. The incidence of HJB was found to be 38.6% with 18.1% having a very high bulb. Thus, a significant proportion of patients are likely to have a HJB. Since there are significant individual variations, a preoperative radiological study for delineation of important bony landmarks is necessary in all patients undergoing surgery for vestibular schwanommas.     Geometric effects in high-temperature vapour-phase lubrication using hydrocarbon feed gases

    Abstract The effectiveness of ,far-field' vapour-phase lubrication, in which areas of a bearing surface that are cycled through the contact are exposed to vapour while outside the contact, has been demonstrated in both sliding and combined roll slide tests using acetylene vapours to deposit pyrolytic graphite. Friction coefficients as low as , = 0.008 have been measured for steel at 540°C with far-field acetylene concentrations as low as 5%. Effective vapour-phase lubrication depends on solid lubricant deposition that exceeds the contact's capacity to remove solid lubricant through wear. While the rate of removal is increased by increasing the sliding velocity, in far-field vapour-phase lubrication the rate of lubricant deposition, and therefore the lubrication effectiveness, is augmented by increased areas available for far-field deposition, such as those provided by performing wear tests with increased wear-track diameters. These geometric concepts may be considered in rolling-element bearing and gear set applications where vapour-phase lubrication is to be employed. [source]
