    Synchrony of spontaneous calcium activity in mouse neocortex before synaptogenesis

    Abstract Spontaneous calcium activity can be detected in embryonic mouse cortical slices as fluorescence intensity variations, in the presence of a fluorescent calcium indicator. Current methods to detect and quantify these variations depend heavily on experimenters whose judgement may interfere with measurement. In the present work, we developed new software called CalSignal for automatic detection and tracking of cellular bodies and quantification of spontaneous calcium activity on time-series of confocal fluorescence images. Analysis of 28 neocortical slices revealed that 21.0% of detected cells displayed peaks of fluorescence corresponding to spontaneous activity, with a mean frequency of one peak per 4 min. This activity was blocked in the absence of extracellular calcium but was not modified after depletion of calcium stores with thapsigargin or blockade of voltage-gated calcium channels with Ni2+. Further, statistical analysis of calcium activity revealed concomitant activation of distant cells in 24 slices, and the existence of a significant network of synchrony based on such coactivations in 17 slices out of 28. These networks enclosed 84.3% of active cells, scattered throughout the neocortical wall (mean distance between cellular bodies, 111.7 m). Finally, it was possible to identify specific cells which were synchronously active with more neighbouring cells than others. The identity of these nodal cells remains to be investigated to fully comprehend the role of spontaneous calcium activity, before synaptogenesis, in shaping cortical neurogenesis. [source]


    Open source in Swedish companies: where are we?

    Abstract Open Source (OS) is a phenomenon of increasing significance for organizations, offering the prospect of effective alternative business solutions and new business opportunities. A number of surveys have been conducted in various countries with the purpose of understanding the state of practice with respect to OS in companies. In this paper we report on a study of the perceptions of OS and the uptake of OS products and development models in Swedish companies. The study used purposeful sampling of companies that have an expressed interest in OS, and the survey was conducted using a set of pre-prepared questions. Its goal was to investigate the extent to which OS has influenced business thinking, as seen from the standpoint of stakeholders. We found that uptake is much higher than reported in earlier studies, but as with previous studies, activity is still concentrated in small and medium-sized enterprises (SMEs). There is increased evidence of interest beyond the simple use of OS components at the infrastructure level. Further, a significant proportion of the companies studied are supporting the OS community as well as benefiting from it. Support includes participation in existing projects and the release of new software under OS licenses. [source]


    OLEX: new software for visualization and analysis of extended crystal structures

    We have developed new software (OLEX) for the visualization and analysis of extended crystal structures. This software has a Windows-compatible mouse-driven graphical interface which gives full control over all structural elements. OLEX provides the user with tools to construct topological networks, visualize interpenetrating or overlapping fragments, and analyse networks constructed fully or partially by exploiting short interactions. It is also easy to generate conventional ellipsoid, ball-and-stick or packing plots. [source]

    Application of fast-digitizer card Acqiris DP-240 in positron lifetime spectroscopy

    Abstract Based on the available knowledge base in positron annihilation measurements, new software for digital positron lifetime spectrometry has been designed within the frame of joint activity between STU Bratislava and JRC Petten. The tests of the new positron digital-life time setup have been carried out. Detectors with BaF2 scintillators and fast-dynode outputs were chosen for such studies. As the digitiser unit, the card Acqiris DP240 was fully tested. Originally, start and stop timing signals were sampled to digital waveforms separately at a rate of 1 GS/s in 8 bit resolution. However, due to limited quality of collected waveforms which significantly affected post-processing of data, the joined delayed channel mode with 2 GS/s sampling rate was applied for further studies. The full setup was benchmarked by a series of calibration measurements of two high purity materials, specifically Si and Ni. The inter-comparison of conventional analogue- and digital-based processing are discussed in detail. Further, the optimisation of main parameters for waveform-processing are analysed too. Such a new approach in the digital life-time spectroscopy will help to improve the resolution of existing methodology which might enhance the capabilities of this technique for further applications, especially in materials science.     An interactive voice response system.
    Abstract A large full-service clinical research organization in the US needed to replace their legacy interactive voice response system which did not provide 21 CFR Part 11 compliant audit trails. They chose to custom-build the new software using C++ component architecture, eXtensible Markup Language (XML), Hypertext Markup Language (HTML), and Microsoft web telephony engine. The case study described in this article illustrates that when systems developers read, study, understand, and embrace 21 CFR Part 11 before designing and validating their product, the regulations are easily met.
