Making the Leap from Researcher to Planner: Lessons from Avian Conservation Planning in the Dominican Republic

Published accounts of national, multidisciplinary planning efforts and priority setting for avian conservation are not common. I describe the process and results of a broad-based, grassroots-oriented avian conservation planning workshop held in the Dominican Republic in which we designed a coordinated strategy for avian conservation in the country. The planning process sought to (1) increase communication and cooperation among conservationists; (2) familiarize participants with resources pertinent to avian conservation; (3) encourage the transfer of information between researchers and managers; (4) promote the concepts of long-term avian monitoring, avian conservation plans, and species management plans; and (5) develop a common, multidisciplinary strategy to promote the conservation of birds in the Dominican Republic. The workshop highlighted group discussions among research biologists, managers, educators, and public policy specialists to assess avian conservation needs and priorities with respect to each discipline and has since galvanized a significant portion of the conservation community around several cooperative projects involving diverse segments of the community. Avian biologists can play a significant role in conservation efforts through a willingness to work with key players in diverse fields and to envision holistic, multidisciplinary approaches to conservation issues. Resumen: Cuando los biologícos investigadores incursionan en la biología de la conservación enfrentan nuevos desafíos, especialmente en países extranjeros, al intentar prestar apoyo para esfuerzos de planificación de la conservación. Los informes publicados de esfuerzos de planificación nacional, multidisciplinaria y de establecimiento de prioridades para la conservación de aves no son comunes. Describo el proceso y los resultados de un taller nacional de planificación para conservación de aves en la República Dominicana que utilizaba un proceso fundamental de base amplia donde creamos una estrategia coordinada para la conservación de aves del país. El proceso de planeación buscaba (1) aumentar comunicación y cooperación entre conservacionistas, (2) familiarizar a los participantes con los recursos disponibles para la conservación de aves, (3) estimular la transferencia de información entre investigadores y manejadores, (4) promover los conceptos del monitoreo de aves a largo plazo, planes de conservación de especies y planes de manejo de especies y (5) desarrollar una estrategia multidisciplinaria común para promover la conservación de aves en la República Dominicana. El taller puso a relieve discusiones de grupo entre investigadores, manejadores, educadores y especialistas en política pública para evaluar las necesidaes y prioridades para la conservación de aves con respecto a cada disciplina, desde entonces se ha estimulado a una porción significativa de la comunidad conservacionista alrededor de proyectos de cooperación que involucran a diversos segmentos de la comunidad. Los ornitólogos pueden jugar un papel significativo en los esfuerzos de conservación mediante una buena disposición para trabajar con personas clave en diversas disciplinas y visualizar de una manera integral y multidisciplinaria las estrategias para abordar asuntos de conservación. Functional tissue engineering for tendon repair: A multidisciplinary strategy using mesenchymal stem cells, bioscaffolds, and mechanical stimulation,

Abstract Over the past 8 years, our group has been continuously improving tendon repair using a functional tissue engineering (FTE) paradigm. This paradigm was motivated by inconsistent clinical results after tendon repair and reconstruction, and the modest biomechanical improvements we observed after repair of rabbit central patellar tendon defects using mesenchymal stem cell-gel-suture constructs. Although possessing a significantly higher stiffness and failure force than for natural healing, these first generation constructs were quite weak compared to normal tendon. Fundamental to the new FTE paradigm was the need to determine in vivo forces to which the repair tissue might be exposed. We first recorded these force patterns in two normal tendon models and then compared these peak forces to those for repairs of central defects in the rabbit patellar tendon model (PT). Replacing the suture with end-posts in culture and lowering the mesenchymal stem cell (MSC) concentration of these constructs resulted in failure forces greater than peak in vivo forces that were measured for all the studied activities. Augmenting the gel with a type I collagen sponge further increased repair stiffness and maximum force, and resulted in the repair tangent stiffness matching normal stiffness up to peak in vivo forces. Mechanically stimulating these constructs in bioreactors further enhanced repair biomechanics compared to normal. We are now optimizing components of the mechanical signal that is delivered in culture to further improve construct and repair outcome. Our contributions in the area of tendon functional tissue engineering have the potential to create functional load-bearing repairs that will revolutionize surgical reconstruction after tendon and ligament injury. © 2007 Orthopaedic Research Society. Characterization of conantokin Rl -A: molecular phylogeny as structure/function study

Abstract A multidisciplinary strategy for discovery of new Conus venom peptides combines molecular genetics and phylogenetics with peptide chemistry and neuropharmacology. Here we describe application of this approach to the conantokin family of conopeptides targeting NMDA receptors. A new conantokin from Conus rolani, ConRl -A, was identified using molecular phylogeny and subsequently synthesized and functionally characterized. ConRl -A is a 24-residue peptide containing three ,-carboxyglutamic acid residues with a number of unique sequence features compared to conantokins previously characterized. The HPLC elution of ConRl -A suggested that this peptide exists as two distinct, slowly exchanging conformers. ConRl -A is predominantly helical (estimated helicity of 50%), both in the presence and absence of Ca++. The order of potency for blocking the four NMDA receptor subtypes by ConRl -A was NR2B > NR2D > NR2A > NR2C. This peptide has a greater discrimination between NR2B and NR2C than any other ligand reported so far. In summary, ConRl -A is a new member of the conantokin family that expands our understanding of structure/function of this group of peptidic ligands targeted to NMDA receptors. Thus, incorporating phylogeny in the discovery of novel ligands for the given family of ion channels or receptors is an efficient means of exploring the megadiverse group of peptides from the genus Conus. A mitochondrial ATPase 6 mutation is associated with Leigh syndrome in a family and affects proton flow and adenosine triphosphate output when modeled in Escherichia coli

A multidisciplinary strategy was used to identify the molecular defect in a family with Leigh syndrome (LS). The propositus presented severe developmental delay, an ataxic-spastic gait and seizures. She died at 3.5 y of age from cardiorespiratory arrest. Postmortem examination disclosed pathological features typical of LS. A 12-y-old sister is affected with the same disease. Respiratory chain enzyme complex activities in skeletal muscle biopsy were normal. Adenosine triphosphate (ATP) synthesis during oxidative phosphorylation in skin fibroblasts mitochondria showed a severely hampered ATP production. Mitochondrial DNA sequencing revealed a new mutation in the ATPase 6 gene (T9176G). Site-directed mutagenesis in Escherichia coli strains was used to measure H+ pumping and ATP synthesis. Results were comparable to findings obtained in human cells. These data corroborate the use of E. coli strains as a feasible "animal" model for functional studies in pathogenic mutations of the ATPase 6 gene. [source]
