    Patterns of isozyme variation as indicators of biogeographic history in Pilgerodendron uviferum (D. Don) Florín

    Abstract. The effects of Pleistocene glaciations on the genetic characteristics of the most austral conifer in the world, Pilgerodendron uviferum, were analysed with specific reference to the hypothesis that the species persisted locally in ice-free areas in temperate South America. It was expected that genetic variation would decrease with latitude, given that ice fields were larger in southern Patagonia and thus refugia were probably located towards the northern distributional limit of the species as suggested by the fossil record. In addition, an increase in among-population genetic divergence was expected with increasing distance to putative glacial refugia. We examined the relationship between location and within-population variability indices of 20 Pilgerodendron populations derived from isozyme analyses. We analysed possible refugia hypotheses by the distribution of allele frequencies using multivariate discriminant analysis. The degree of genetic differentiation with geographical distance between all population pairs was investigated by Mantel tests. Results indicated that Pilgerodendron populations are highly monomorphic, probably reflecting past population bottlenecks and reduced gene flow. Southernmost populations tend to be the least genetically variable and were therefore probably more affected by glacial activity than northern ones. Populations located outside ice limits seem to have been isolated during the glacial period. The presence of centres of genetic diversity, together with the lack of a significant correlation between genetic and geographical distances and the absence of geographical patterns of allelic frequencies at most analysed alleles, may indicate that Pilgerodendron did not advance southward after the last glaciation from a unique northern refugium, but spread from several surviving populations in ice-free areas in Patagonia instead.     Darwin's cross-promotion hypothesis and the evolution of stylar polymorphism

    Abstract There is growing appreciation that the ecological factors which impact on rates of pollen transfer can contribute significantly to reproductive trait evolution in plants. In heterostylous species, several studies support Darwin's claim that the reciprocal positions of stigmas and anthers enhance inter-morph mating in comparison to intra-morph mating and thus the maintenance of the polymorphism. In this study, we evaluate the relative importance of intra-morph and inter-morph pollen transfers in Narcissus assoanus, a species with dimorphic variation in style length but non-reciprocity of anther positions. This stigma-height dimorphism represents a transitional stage in theoretical models of the evolution of distyly. Seed set variation on recipient plants with donor plants of a single morph in experimental arrays in a natural population illustrate that inter-morph cross-pollination is more efficient that intra-morph cross-pollination as a result of high rates of pollen transfer from long-styled to short-styled plants. The observed rates of pollen transfer satisfy the theoretical conditions for the establishment of a stigma-height dimorphism in an ancestral monomorphic long-styled population in pollen-limited situations. These results provide experimental evidence for the Darwinian hypothesis that enhanced inter-morph cross-pollination facilitates not only the maintenance of heterostyly but also the establishment of transitional forms implicated in the evolution of this polymorphism.     Size-Related Advantages for Reproduction in a Slightly Dimorphic Raptor: Opposite Trends between the Sexes

    Despite many comparative analyses and more than 20 proposed hypotheses, there is still little consensus over the factors promoting the evolution of reversed sexual dimorphism (RSD) in raptorial species. Furthermore, intrapopulation studies, which may elucidate how RSD is maintained once evolved, have been surprisingly scarce and only focused on a handful of species with medium to high dimorphism. We examined the reproductive advantages associated with body size and condition, measured in the pre-laying period, in a diurnal raptor with low sexual dimorphism, the black kite (Milvus migrans). The study population was essentially monomorphic in size. For females, there was an evidence of reproductive benefits associated with larger size and/or with better body condition. Larger females had also access to higher quality partners and territories, consistent with the ,intrasexual selection' hypothesis, by which members of the larger sex enjoy size-related advantages in intrasexual competition over a scarce resource, the smaller sex. Opposite trends emerged for males: smaller, leaner males had higher breeding output, consistent with the ,small efficient male' hypothesis. Overall, the fact that we observed in an essentially monomorphic population the same selection pressures previously found in species with marked dimorphism suggests that such reproductive advantages may be counterbalanced in our study model by opposite selection pressures during other stages of the life cycle. This casts some doubts on the evolutionary significance of studies focusing exclusively on reproduction and calls for the need of more comprehensive analyses incorporating trait-mediated differentials in survival and recruitment.     SEASONAL CYCLES OF ALLOZYME-BY-CHROMOSOMAL-INVERSION GAMETIC DISEQUILIBRIUM IN DROSOPHILA SUBOBSCURA

    Abstract Allozyme loci are frequently found non randomly associated to the chromosomal inversions in which they are included in Drosophila. Two opposite views compete to explain strong allozyme-by-inversion gametic disequilibria: they result from natural selection or, conversely, merely represent remnants of associations accidentally established at the origin of inversions. Empirical efforts aimed at deciding between adaptive and historical scenarios have focused on the spatial distribution of disequilibria. Yet, the evolutionary significance of these associations remains uncertain. I report here the results of a time-series analysis of the seasonal variation of alleles at six allozyme loci (Acph, Lap, Pept-1, Ao, Mpi, and Xdh) in connection with the O chromosomal polymorphisms of D. subobscura. The findings were: (1) in the segment I of the O chromosome, Lap and Pept-1 allozymes changed seasonally in a cyclical fashion within the ST gene arrangement, but they changed erratically within the 3+4 gene configuration; (2) the frequencies of Lap111 and Pept-10,40 within ST dropped to their lowest values in early and late summer, respectively, when the seasonal level of the ST arrangement is lowest. Furthermore, Lap1,11 and Pept-10,40 covary with ST only within these seasons, yet in a fashion inconsistent with these alleles having a major influence on the dynamics of the inversion; (3) seasonal cycling of alleles within inversions were not detected at Acph, Ao, Mpi, and Xdh, yet these loci are nearly monomorphic at the study population, and/or their sampled series were shorter than those for Lap and Pept-1; and (4) simply monitoring allozyme frequencies separately for each inversion proved to be superior, for evidencing the seasonal cycles of the disequilibria, to the use of the D' coefficient of association. Observed seasonal cycles of allozymes within inversions likely reflect natural selection. [source]


    STABILITY AND EVOLUTION OF OVERLAPPING GENES

    Abstract., When the same sequence of nucleotides codes for regions of more than one functional polypeptide, this sequence contains overlapping genes. Overlap is most common in rapidly evolving genomes with high mutation rates such as viruses, bacteria, and mitochondria. Overlap is thought to be important as: (1) a means of compressing a maximum amount of information into short sequences of structural genes; and (2) as a mechanism for regulating gene expression through translational coupling of functionally related polypeptides. The stability of overlapping codes is examined in relation to the information cost of overlap and the mutation rate of the genome. The degree of overlap in a given population will tend to become monomorphic. Evolution toward partial overlap of genes is shown to depend on a convex cost function of overlap. Overlap does not evolve when expression of overlapping genes is mutually exclusive and produced by rare mutations to the wild-type genome. Assuming overlap increases coupling between functionally related genes, the conditions favoring overlap are explored in relation to the kinetics of gene activation and decay. Coupling is most effective for genes in which the gene overlapping at its 5'end (leading gene) decays rapidly, while the gene overlapping at the 3'end (induced gene) decays slowly. If gene expression can feedback on itself (autocatalysis), then high rates of activation favor overlap. [source]


    Social biology of rodents

    Abstract Herein, I summarize some basic components of rodent social biology. The material in this paper is summarized and condensed from a recent book "Rodent Societies: An Ecological and Evolutionary Perspective" edited by J. O. Wolff and P. W. Sherman (2007). I describe the four basic spacing patterns and illustrate how female territoriality is a function of offspring defense and male mating tactics are a function of female defensibility. The vulnerability of young to infanticide shapes female spacing and mating behavior. Food does not appear to be a defensible resource for rodents, except for those species that larder hoard nonperishable items such as seeds. Philopatry and the formation of kin groups result in genetic sub-structuring of the population, which in turn affects effective population size and genetic diversity. Dispersal is male biased and typically involves emigration from the maternal site to avoid female relatives and to seek unrelated mates. Scent marking is a major form of communication and is used in reproductive competition and to assess prospective mates, but it is also eavesdropped by predators to locate prey. Females do not appear to alter the sex ratio of litters in response to maternal condition but among arvicoline rodents daughters appear to be favored in spring and sons in autumn. Rodents are relatively monomorphic; however, females tend to be larger than males in the smallest species and smaller in the larger species. Predation risk results from an interaction among foraging time and vulnerability and in turn affects behavioral and life history characteristics. [source]


    Association analysis of polymorphisms in the interleukin-4 receptor (alpha) gene with atopic asthma in patients from western Mexico

    Summary We undertook an association analysis between the ile50val, glu375ala, cys406arg, and ser761pro polymorphisms of the IL-4R, gene and atopic asthma, total IgE levels and IL-4 serum levels in a population from western Mexico. We found that the ser761pro polymorphism was monomorphic for ser761, while there was no association between any of the other polymorphisms and the three phenotypes analysed. [source]


    Genetic variability of seven dog breeds based on microsatellite markers

    Summary The present study, compared the genetic variability of seven dog breeds and a test sample from Switzerland by means of 26 microsatellite markers. Five loci were excluded from further analyses because one was monomorphic, one not in Hardy,Weinberg equilibrium in all breeds and three in linkage disequilibrium with linked loci. The proportion of shared alleles at the individual level of the remaining 21 microsatellite markers combined with the neighbour-joining method allowed for the clustering of the large majority of the individuals in accordance to their breed. The results were confirmed by analyses using a Bayesian approach for clustering and a Monte Carlo re-sampling method for individual assignment or exclusion to a given population. [source]


    Offspring sex ratio skew in the sexually monomorphic house martin Delichon urbicum

    Sex ratio at conception may be under selection pressure, given that male and female offspring differ in the cost of production or generate different fitness returns under specific conditions. We studied adjustments in the primary, secondary and tertiary sex ratio in house martin Delichon urbicum, which is a sexually monomorphic, socially monogamous, colonial bird. Males of this species engage in extra-pair copulations with heavy males acquiring the highest fertilization success. We analyzed variation in the sex ratio in relation to clutch size and parental characteristics including body condition, wing length, as well as length and pigmentation of the white rump patch during three breeding seasons. The only variable which significantly explained the variation in the sex ratio was wing length of the social father and mother. The proportion of sons among offspring was positively correlated to wing length of the social father and negatively correlated to mother wing length. Social father wing length positively correlated with mean brood body mass at fledging, which may suggest that females that mated with long-winged males produced sons, which acquired the highest total fertilization success. Consequently, our results indicate that house martin females may adaptively adjust offspring sex composition at egg laying in relation to the characteristics of their social mate. [source]


    Sex and age influence intestinal parasite burden in three boreal grouse species

    Parasite infections are often more common in male vertebrates than in females. Sexual selection leading to dimorphism can cause sexual differences in immune defence, behaviour or body size. Possible proximate explanations for male bias in parasitism are the immunosuppressive effect of male sex hormones and the large body size of males which increases the likelihood of being parasitized. To evaluate these hypotheses, we studied the prevalence and abundance of intestinal helminth parasites in three boreal grouse species, the capercaillie Tetrao urogallus, the black grouse Tetrao tetrix, and the hazel grouse Bonasa bonasia. The first two are sexually dimorphic polygynous species while the latter species is sexually monomorphic and monogamous. We found a male-bias in the prevalence and abundance of the nematode Ascaridia compar. The bias was most pronounced in the polygynous black grouse and capercaillie. In the monogamous hazel grouse, there was a slight male-bias in occurrence of ascarids, but no bias in abundance. In juvenile grouse, the male-bias was larger than in adult grouse. No sexual bias was found in regard to the cestodes (Skrjabinia cesticillus, Paroniella urogalli and Hymenolepis spp.). However, age was a factor in cestode prevalence: juvenile grouse were more commonly infected than adults. Differences in growth rates and body size are potential factors that may lead to male-biased parasitism in these grouse species, and their impact requires further studies. [source]


    Intraspecific differences in benefits from feeding in mixed-species flocks

    The Madagascar Paradise Flycatcher Terpsiphone mutata and Common Newtonia Newtonia brunneicauda frequently form two-species flocks in the deciduous dry forest of western Madagascar. In T. mutata, some males have long tails, while other males and females have short tails. When foraging in mixed flocks, each type of bird captured prey more rapidly than otherwise, but the degree of increase in feeding rate was smaller in long-tailed males. When in mixed flocks, all T. mutata caught prey on leaves in the canopy where N.brunneicauda foraged. Long-tailed males changed feeding habits from sallying when not in mixed flocks, whereas short-tailed birds showed no change of feeding habit. The elongated tails of long-tailed males may have made their foraging less efficient owing to decreased agility in the canopy. N. brunneicauda is monomorphic and often formed groups of three to five individuals. In monospecific flocks, subordinates fed at low rates on branches owing to frequent hostile encounters. When foraging in mixed flocks, however, subordinates foraged among leaves, and their feeding rates increased because the frequency of intraspecific interference decreased greatly. Dominants did not show any difference in feeding pattern with social situation. Thus, heterospecific flocking was more advantageous for subordinates. [source]


    Analysis of the Pattern of Initiation of Sustained Ventricular Arrhythmias in Patients with Implantable Defibrillators

    Initiation of Sustained Ventricular Arrhythmias. Introduction: The purpose of this study was to analyze the pattern of initiation of sustained ventricular arrhythmias in patients with varying types of underlying structural heart disease. Methods and Results: The study group consisted of 90 patients with an implantable cardioverter defibrillator. Cardiovascular diagnoses included coronary artery disease in 64 patients (71%). The patients were divided into four groups based on the type and severity of structural heart disease. Two hundred sixty episodes of sustained ventricular arrhythmias were analyzed. The mean coupling interval of the initiating heat of all ventricular arrhythmias was 523 ± 171 msec. The coupling interval of the initiating beat was longer in patients with impaired ventricular function, particularly those with nonischemic dilated cardiomyopathy. The prematurity index was similar regardless of the type of underlying structural heart disease. However, the prematurity index was shorter in patients with polymorphic ventricular tachycardia (VT) compared to those with monomorphic VT. A pause was observed more commonly before the onset of polymorphic VT/ventricular fibrillation than sustained monomorphic VT. Two hundred twenty-two (85%) of the arrhythmia episodes were initiated by a late-coupled premature beat, 33 (13%) were initiated by an early-coupled premature beat, and 5 episodes (2%) were initiated with a short-long-short sequence. The patttern of initiation of the ventricular arrhythmias was similar in all patient groups and for both monomorphic and polymorphic tachycardias. Conclusion: These findings demonstrate that sustained ventricular arrhythmias typically are initiated by late-coupled ventricular premature depolarizations, regardless of the type or severity of underlying structural heart disease or resultant arrhythmia.     Severely reduced sexual reproduction in northern populations of a clonal plant, Decodonverticillatus (Lythraceae)

    Summary 1,In flowering plants the balance between sexual and clonal, asexual reproduction can vary widely. We quantified variation in sexual reproduction in a tristylous, clonal, aquatic plant, Decodon verticillatus, and investigated the role of ecological and genetic factors in causing this variation. 2,We surveyed components of sexual fertility and vegetative growth in 28 populations distributed along a 500-km latitudinal transect in New England, USA. Northerly populations tend to be monomorphic (M) for style length, and probably therefore have reduced sexual reproduction compared with southerly, trimorphic (T) populations. 3,Compared with T populations (n = 10), M populations (n = 18) exhibited large reductions for all components of sexual reproduction, including flower production, pollen deposition, pollen tube growth, fertilization, fruit set and seeds per fruit. Seven M populations produced no seed at all, and the other 11 very little (mean = 24 vs. 1139 seeds per plant in trimorphic populations). Clonal propagation was also greatly reduced in M populations. 4,A survey of three polymorphic allozyme loci detected only single, usually heterozygous, genotypes in 15 M populations, whereas all T populations were genotypically diverse. The other three M populations contained three or fewer genotypes and one always predominated. Sexual recruitment is therefore extremely rare. 5,Comparison of the sexual fertility of M and T populations in a concurrent common glasshouse experiment with our field data revealed that reduced sexual performance in northern M populations is principally due to genetic factors, but is also caused by ecological factors that covary with latitude. 6,This abrupt shift away from sexual reproduction in populations at the northern periphery of the geographical range in D. verticillatus may greatly limit their evolutionary potential and restrict further northward expansion.     A new method for sexing Oriental White Storks

    ABSTRACT For species where males and females are monomorphic, or nearly so, determining the sex of individual birds generally requires either capturing birds or collecting samples, such as feathers, for DNA analysis. We developed a new method, involving the use of photographs, to determine the sex of endangered Oriental White Storks (Ciconia boyciana). Using photographs, we analyzed the lateral features of the heads of 25 captive storks of known sex (12 males and 13 females) and found differences between males and females in the distance from the bill tip to the nape and the distance from the bill tip to the commissural point. These differences were used to generate a discriminant function that was then tested on 22 captive storks at Hyogo Homeland Park (Toyooka, Japan), and we correctly determined the sex of 18 individuals (82%). In addition, the sex of two wild storks was correctly assigned. Our results suggest that good-quality photographs can be useful for determining the sex of both captive and wild Oriental White Storks and, further, that similar methods may prove useful for determining sex in other species of birds. SINOPSIS Para especies monomórficas, o muy parecidas, el determinar el sexo de individuos por lo general requiere capturar las aves o coleccionar muestras, tales como plumas, para hacer análisis de ADN. Desarrollamos un método que envuelve el uso de fotografías, para determinar el sexo en individuos de Ciconia boyciana, la cual es una especie en peligro de extinción. Mediante el uso de fotografías, pudimos analizar las particularidades de la parte lateral de la cabeza de 25 individuos cautivos de sexo conocido (12 machos y 13 hembras). Encontramos diferencias sexuales entre la distancia desde la punta del pico a la nuca, y desde la punta del pico al punto de la comisura mandibular. Estas diferencias fueron utilizadas para generar una función discriminativa que fue puesta a pruebas en 22 individuos cautivos que se encontraban en Hyogo Homeland Park (Toyooka, Japón) y que permitió determinar el sexo correctamente de 18 individuos (82% de estos). Además pudimos determinar correctamente el sexo de dos individuos silvestres. Nuestros resultados sugieren que con buenas fotografías se puede identificar el sexo, tanto de individuos silvestres como cautivos, de Ciconia boyciana y que métodos similares pudieran ser útiles para determinar el sexo de otras especies de aves.     Sexual dimorphism and dichromatism in Steere's Liocichla (Liocichla steerii)

    ABSTRACT Although sexual differences in birds can be extreme, differences between males and females in body size and plumage color are more subtle in many species. We used a genetic-based approach to determine the sex of male and female Steere's Liocichla (Liocichla steerii) and examine the degree of size dimorphism and plumage dichromatism in this apparently monomorphic species. We found that males were significantly larger than females. In addition, Steere's Liocichla have a prominent yellow plumage patch on the lores that was significantly larger in males than females for both live birds and museum specimens. We also used reflectance spectrometry to quantify the color of the yellow-green breast feathers of Steere's Liocichla and found no significant differences between males and females in brightness, intensity, saturation, or hue. However, females tended to have brighter breast plumage, particularly at long wavelengths. Collectively, these color variables were useful in discriminating birds according to sex when used in a discriminant function analysis. Our study suggests that sexual selection may be more widespread than once assumed, even among birds considered monomorphic, and emphasizes the need for additional data from tropical and subtropical species. SINOPSIS Aunque las diferencias sexuales entre algunas especies puede ser estrechas, un muchas de estas las diferencias corporales entre los sexos pudiera ser sutiles. Abordamos el asunto basados en genética para determinar el sexo en Liocichla steerii y examinamos el grado de dimorfismo sexual y dicromatismo en el plumaje en esta especie la cual aparentemente es monomórfica. Encontramos que los machos eran significativamente de mayor tamaño que las hembras. Además encontramos en los machos un parcho amarillo en los lores significativamente más grande que en las hembras, tanto en aves del estado silvestre como en pieles de museos. Usamos espectrometría para cuantificar el color de las plumas verde-amarillentas del pecho de las aves y no encontramos diferencias significativas entre los sexos, con respecto a la brillantez, insensidad, saturación o matiz color. Sin embargo, las hembras tuvieron la tendencia a tener el plumaje más brillante en el pecho particularmente en el espectro de onda larga. Colectivamente, estas variables de color fueron útiles para discriminar entre el sexo de las aves, particularmente cuando fueron examinadas utilizando un análisis discriminante de funciones. Nuestro estudio sugiere que la selección sexual pudiera estar más ampliamente distribuida que lo previamente asumido, aún en aves consideradas monomórficas.     Sex differences in behaviour as an indirect consequence of mating system

    A considerable literature has been devoted to documenting differences between the sexes. However, relatively little attention has hitherto been directed towards those differences that arise as an indirect consequence of mating system even though they can have profound implications for the daily lives of the animals involved. In this review we focus on differences in the non-reproductive behaviour of fish and relate these to sexual dimorphism in size and morphology, and to variance in fitness between the sexes. In line with our expectation, differences in distributional ecology, schooling, aggression, predator avoidance and foraging are exaggerated in sexually dimorphic species and polygamous mating systems. Nonetheless, the behaviour of males and females may also differ in sexually monomorphic and monogamous species. We conclude by highlighting promising directions for further research. [source]


    Microsatellite DNA variation in Bornean orangutans (Pongo pygmaeus)

    Orangutans (Pongo pygmaeus) on the islands of Borneo and Sumatra are considered two separate subspecies. However, the genetic relationships between isolated populations on Borneo are not clear. This study determined the extent of variation within the Bornean subspecies of orangutan, using microsatellite DNA analysis. Blood samples were collected from 96 individuals of known origin from East, West and Central Kalimantan. Human microsatellite primer pairs located at human map position D2S141, D4S431, D11S925, D16S420 and D17S791 were suitable for use in primates. D4S431 appeared monomorphic for all orangutans. In three cases (D2S141 East and West and D16S420 West), a highly significant excess of homozygous allele frequencies was detected, but with other primer pairs no significant difference in allele frequencies occurred. We conclude that the divergence between the different populations on Borneo is less than the variation within the populations. There was also evidence that inbreeding occurred within the populations.     Differentiation and Genetic Structure of Sclerophoma pythiophila Strains on Pinus sylvestris in Poland

    Abstract Between 1996 and 2006, 97 strains of Sclerophoma pythiophila were isolated from 3 to 10-year-old trees of Pinus sylvestris from three regions of Poland differing by climatic conditions and geographic location. On the basis of 56 random amplified microsatellites (RAMS) markers obtained with the use of five primers, very high level of genetic variability of strains (mean Jaccard's coefficient 0.56) was ascertained. Among the analysed markers only one was completely monomorphic, whereas six were monomorphic in 90%. Greater genetic similarity was ascertained for strains in southern Poland (0.58) in comparison with strains deriving from northern Poland (0.52). Correlation between the percentage share of polymorphic loci and the average annual temperature of places of strain isolation (r = 0.62) were shown, as well as the number of days with snow cover (r = 0.79). On the basis of the amova analysis, it was ascertained that 11.6% of genetic variability was located between regions of the strains origin and 88.4% within the regions. [source]


    Genetic variability and differentiation in red deer (Cervus elaphus) from Scotland and England

    Abstract Samples from 69 British red deer Cervus elaphus scoticus from seven populations in Scotland and England were analysed with respect to variability within and differentiation among stocks using 11 polymorphic microsatellite loci and 439 bp of the mitochondrial control region. The results clearly showed the effects of anthropogenic factors on British red deer. On the whole, variability values were within the species' reported range. The island population of Islay, Scotland, however, while showing average microsatellite variability, exhibited no mitochondrial variation at all. One microsatellite locus was monomorphic in three Scottish populations (Islay, Dunachton and Achnacarry). Overall and pairwise FST values indicate considerable differentiation among the populations studied, but Dunachton and Achnacarry, two adjacent populations free from recorded introductions, showed only a little differentiation and were paired in trees based on genetic distances. In terms of variability, no statistically significant differences were observed between island and mainland populations and the overall test of isolation by distance was negative. Possible reasons for the genetic patterns observed, such as differences in human impact on the populations, are discussed. [source]


    Histologic classification of ductal carcinoma in situ

    Abstract Prior to the current mammographic era, ductal carcinoma in situ (DCIS) usually presented as a large mass, was classified morphologically by architecture, and treated by mastectomy. The introduction of screening mammography led to an increase in the incidence of DCIS, a decrease in the average size of DCIS, and an increased emphasis on its heterogeneous nature. Thus, a reproducible and prognostically relevant classification system for DCIS is necessary. The ultimate goal of this classification is proper selection of patients for whom lumpectomy would suffice rather than mastectomy. Features to evaluate include: extent and size of disease, adequacy of resection margins, and histology. While none of the proposed histological classification systems were endorsed at the recent Consensus Conference on the Classification of DCIS, nuclear grade was the most important feature common to most of them. Architecture was given secondary importance. By definition, DCIS is a non-invasive clonal proliferation of epithelial cells originating in the terminal duct lobular unit, which would be expected to be monomorphic; however, it is the degree of nuclear pleomorphism that is primarily used to separate DCIS into low, intermediate, and high grades. Architecturally, DCIS has been divided into the following types: comedo, solid, cribriform, micropapillary, and papillary. Different architectural patterns and grades may be present in a given particular case; however, some combinations of patterns occur more frequently than others. Interobserver studies have shown nuclear grading to be interpreted with greater consistency than architecture, and nuclear grading methods have correlated with biological and molecular marker studies.     Genetic drift outweighs balancing selection in shaping post-bottleneck major histocompatibility complex variation in New Zealand robins (Petroicidae)

    Abstract The Chatham Island black robin, Petroica traversi, is a highly inbred, endangered passerine with extremely low levels of variation at hypervariable neutral DNA markers. In this study we investigated variation in major histocompatibility complex (MHC) class II genes in both the black robin and its nonendangered relative, the South Island robin Petroica australis australis. Previous studies have shown that Petroica have at least four expressed class II B MHC genes. In this study, the sequences of introns flanking exon 2 of these loci were characterized to design primers for peptide-binding region (PBR) sequence analysis. Intron sequences were comprised of varying numbers of repeated units, with highly conserved regions immediately flanking exon 2. Polymerase chain reaction primers designed to this region amplified three or four sequences per black robin individual, and eight to 14 sequences per South Island robin individual. MHC genes are fitness-related genes thought to be under balancing selection, so they may be more likely to retain variation in bottlenecked populations. To test this, we compared MHC variation in the black robin with artificially bottlenecked populations of South Island robin, and with their respective source populations, using restriction fragment length polymorphism analyses and DNA sequencing of the PBR. Our results indicate that the black robin is monomorphic at class II B MHC loci, while both source and bottlenecked populations of South Island robin have retained moderate levels of variation. Comparison of MHC variation with minisatellite DNA variation indicates that genetic drift outweighs balancing selection in determining MHC diversity in the bottlenecked populations. However, balancing selection appears to influence MHC diversity over evolutionary timescales, and the effects of gene conversion are evident. [source]


    Genetic variation in eastern North American and putatively introduced populations of Ceratocystis fimbriata f. platani

    Abstract The plant pathogenic fungus Ceratocystis fimbriata f. platani attacks Platanus species (London plane, oriental plane and American sycamore) and has killed tens of thousands of plantation trees and street trees in the eastern United States, southern Europe and Modesto, California. Nuclear and mitochondrial DNA fingerprints and alleles of eight polymorphic microsatellite markers of isolates of C. fimbriata from these regions delineated major differences in gene diversities. The 33 isolates from the eastern United States had a moderate degree of gene diversity, and unique genotypes were found at each of seven collection sites. Fingerprints of 27 isolates from 21 collection sites in southern Europe were identical with each other; microsatellite markers were monomorphic within the European population, except that three isolates differed at one locus each, due perhaps to recent mutations. The genetic variability of C. fimbriata f. platani in the eastern United States suggests that the fungus is indigenous to this region. The genetic homogeneity of the fungus in Europe suggests that this population has gone through a recent genetic bottleneck, perhaps from the introduction of a single genotype. This supports the hypothesis that the pathogen was introduced to Europe through Naples, Italy during World War II on infected crating material from the eastern United States. The Californian population may also have resulted from introduction of one or a few related genotypes because it, too, had a single nuclear and mitochondrial genotype and limited variation in microsatellite alleles.     Isolation, characterization and PCR multiplexing of polymorphic microsatellite markers in the edible dormouse, Glis glis

    Abstract We isolated and characterized 10 dinucleotide microsatellite loci in the edible dormouse, Glis glis (Linnaeus). Four multiplex panels were developed. Loci were amplified in samples from two geographically distant populations (Torgny in Belgium and Montseny in Spain). All loci were polymorphic in Spain but four were monomorphic in Belgium. Individuals from Belgium and Spain exhibited an average allelic diversity of 1.9 and 3.3 and an observed heterozygosity ranging from 0.08 to 0.47 and from 0.04 to 0.72, respectively. [source]


    Tetranucleotide microsatellite loci from the striped dolphin (Stenella coeruleoalba Meyen, 1833)

    Abstract Five tetranucleotide microsatellite loci were isolated from the striped dolphin (Stenella coeruleoalba). Characterization of these loci revealed between three and 11 alleles at each locus and observed heterozygosity ranged from 0.192 to 0.775. All sets of primers amplified homologous loci when tested on four related species and showed polymorphisms in most locus,species combinations. A sixth microsatellite, which appeared to be monomorphic in the striped dolphin, was also shown to be polymorphic in three related species. [source]


    Development and characterization of novel microsatellite markers from the olive ridley sea turtle (Lepidochelys olivacea)

    Abstract Olive ridley turtles, although widely distributed globally and in Indian coastal waters, have undergone declines in recent years due to anthropogenic factors, particularly fishery-related mortality. Assessment of genetic variability in existing populations is critical to the development of effective conservation strategies. Here we describe the development of six highly polymorphic microsatellite loci from a simple sequence repeat-enriched genomic DNA library of olive ridley turtle. Characterization of five of these loci using 83 individual olive ridley turtles revealed eight to 24 alleles per locus, high observed and expected heterozygosity values and broad cross-species amplifications. The sixth microsatellite was found to be monomorphic in the olive ridley samples but was polymorphic in two related marine turtle species. These microsatellites thus provide efficient genetic markers to understand the population structure, phylogeography and species relationships of olive ridley and other marine turtle species.     New polymorphic microsatellite loci for different camel species

    Abstract New microsatellite loci were screened and sequenced from the genomic DNA of male Camelus bactrianus. Among 32 loci, 23 were amplified in bactrian and dromedary species, 19 in llama and 20 in alpaca. The different species had similar fragment lengths per locus, with more striking similarities between bactrian and dromedary and between llama and alpaca, respectively. Seven loci had more than 10 alleles each, nine were monomorphic in all species, and one was monomorphic in Old World and polymorphic in New World camels. The results show that the informative microsatellite loci can be widely applied to several species. [source]


    Microsatellite markers for diverse Salix species

    Abstract Forty-six microsatellites were isolated from an enriched library of Salix burjatica and tested on 20 individuals (of nine species/hybrids) from the National Willows Collection (IACR-Long Ashton Research Station, UK). Twenty-nine were monomorphic, gave multilocus or unscorable patterns, or were duplicates. The remaining 17 microsatellites gave 2,22 alleles/locus. Three microsatellites successfully cross-amplified in 31 additional Salix species. A further six were tested on panels comprising 6,25 individuals from the 31 species. Cross-amplification was successful in all cases. These results suggest that the microsatellites isolated here should prove useful for population studies in a wide range of Salix species. [source]


    Eleven new microsatellites for hop (Humulus lupulus L.)

    Abstract We present a new set of 11 polymorphic microsatellite primer sequences for use with Humulus lupulus. Microsatellite-enriched libraries for GAn and GTn types of repeats were produced. Sequencing of 72 clones revealed 42 unique inserts containing microsatellites, out of which 19 primer pairs were designed and microsatellite amplification was tested on 39 wild hops and cultivars. Eleven primer pairs showed single locus amplification with 2,13 alleles, average 7.2, of which 17 unique alleles were discovered. One primer pair amplified too strong stutter bands, one locus was monomorphic and multilocus amplification was obtained with the remaining six primer pairs. [source]


    Trichome production and spatiotemporal variation in herbivory in the perennial herb Arabidopsis lyrata

    Allocation theory suggests that the optimal level of resistance against herbivores should vary with the risk of herbivory if allocation to resistance is costly. The perennial herb Arabidopsis lyrata has a genetically based polymorphism for trichome production and occurs in a glabrous and a trichome-producing form. Leaf trichomes (hairs) can protect plants against insect herbivores, and may increase tolerance to drought and UV-radiation. To examine the functional significance of trichome production, we documented the frequency of glabrous plants and damage by insect herbivores in 30 A. lyrata populations in Sweden and Norway. The proportion of glabrous plants ranged from 0.10 to 0.71 (median=0.44) in polymorphic populations; 7 of 12 populations in Norway and 14 of 18 populations in Sweden were monomorphic glabrous, i.e. with fewer than 5% trichome-producing plants. The mean proportion of the leaf area removed by herbivores varied substantially among populations and years. With few exceptions, glabrous plants were more damaged than trichome-producing plants in polymorphic populations. The intensity of herbivory quantified as the mean damage to glabrous plants tended to be higher in polymorphic populations than in populations monomorphic for the glabrous morph and was higher in Sweden than in Norway. In Norway, both the magnitude of herbivore damage and the frequency of trichome-producing plants tended to decrease with increasing altitude. The results indicate that leaf trichomes contribute to resistance against herbivorous insects in A. lyrata, and suggest that herbivore-mediated selection contributes to the maintenance of the polymorphism in trichome production.     Nontransmural Scar Detected by Magnetic Resonance Imaging and Origin of Ventricular Tachycardia in Structural Heart Disease

    Background: Contrast-enhanced magnetic resonance imaging (CMR) identifies scar tissue as an area of delayed enhancement (DE). The scar region might be the substrate for ventricular tachycardia (VT). However, the relationship between the occurrence of VT and the characteristics of scar tissue has not been fully studied. Methods: CMR was performed in 34 patients with monomorphic, sustained VT and dilated cardiomyopathy (DCM, n = 18), ischemic cardiomyopathy (ICM, n = 10), or idiopathic VT (IVT, n = 6). The VT exit site was assessed by a detailed analysis of the QRS morphology, including bundle branch block type, limb lead polarity, and precordial R-wave transition. On CMR imaging, the transmural score of each of the 17 segments was assigned, using a computer-assisted, semiautomatic technique, to measure the DE areas. Segmental scars were classified as nontransmural when DE was 1,75% and transmural when DE was 76,100% of the left ventricular mass in each segment. Results: A scar was detected in all patients with DCM or ICM. Nontransmural scar tissue was often found at the VT exit site, in patients with DCM or ICM. In contrast, no scar was found in patients with IVT. Conclusions: CMR clarified the characteristics and distribution of scar tissue in patients with structural heart disease, and the presence and location of scar tissue might predict the VT exit site in these patients.
