MINIMAL SELFING, FEW CLONES, AND NO AMONG-HOST GENETIC STRUCTURE IN A HERMAPHRODITIC PARASITE WITH ASEXUAL LARVAL PROPAGATION

Abstract Little is known about actual mating systems in natural populations of parasites or about what constitutes the limits of a parasite deme. These parameters are interesting because they affect levels of genetic diversity, opportunities for local adaptation, and other evolutionary processes. We expect that transmission dynamics and the distribution of parasites among hosts should have a large effect on mating systems and demic structure, but currently we have mostly speculation and very few data. For example, infrapopulations (all the parasites in a single host) should behave as demes if parasite offspring are transmitted as a clump from host to host over several generations. However, if offspring are well mixed, then the parasite component population (all the parasites among a host population) would function as the deme. Similarly, low mean intensities or a high proportion of worms in single infections should increase the selfing rate. For species having an asexual amplification stage, transmission between intermediate and definitive (final) hosts will control the variance in clonal reproductive success, which in turn could have a large influence on effective sizes and rates of inbreeding. We examined demic structure, selfing rates, and the variance in clonal reproductive success in natural populations of Plagioporus shawi, a hermaphroditic trematode that parasitizes salmon. Overall levels of genetic diversity were very high. An a posteriori inference of population structure overwhelmingly supports the component population as the deme, rather than individual infrapopulations. Only a single pair of 597 adult individuals was identified as clones. Thus, the variance in clonal reproductive success was almost zero. Despite being hermaphroditic, P. shawi appears to be almost entirely outcrossing. Genetic estimates of selfing (<5%) were in accordance with the proportion of parasites from single infections. Thus, it appears that individual flukes outcross whenever possible and only resort to selfing when alone. Finally, our data support the hypothesis that aquatic transmission and the use of several intermediate hosts promotes high genetic diversity and well-mixed infrapopulations. [source]


Chromosomal aberrations in peripheral lymphocytes of train engine drivers

Abstract Studies of Swedish railway employees have indicated that railroad engine drivers have an increased cancer morbidity and incidence of chronic lymphatic leukemia. The drivers are exposed to relatively high magnetic fields (MF), ranging from a few to over a hundred ,T. Although the possible genotoxic potential of MF is unclear, some earlier studies have indicated that occupational exposure to MF may increase chromosome aberrations in blood lymphocytes. Since an increased level of chromosomal aberrations has been suggested to predict elevated cancer risk, we performed a cytogenetic analysis on cultured (48 h) peripheral lymphocytes of Swedish train engine drivers. A pilot study of 18 engine drivers indicated a significant difference in the frequency of cells with chromosomal aberrations (gaps included or excluded) in comparison with seven concurrent referents (train dispatchers) and a control group of 16 office workers. The engine drivers had about four times higher frequency of cells with chromosome-type aberrations (excluding gaps) than the office workers (P,<,0.01) and the dispatchers (P,<,0.05). Seventy-eight percent of the engine drivers showed at least one cell per 100 with chromosome-type aberrations compared with 29% among the dispatchers and 31% among the office workers. In a follow-up study, another 30 engine drivers showed an increase (P,<,0.05) in the frequency of cells with chromosome-type aberrations (gaps excluded) as compared with 30 referent policemen. Sixty percent of the engine drivers had one or more cells (per 100 cells) with chromosome-type aberrations compared with 30% among the policemen. In conclusion, the results of the two studies support the hypothesis that exposure to MF at mean intensities of 2,15,,T can induce chromosomal damage. The effects of nitric oxide on magnocellular neurons could involve multiple indirect cyclic GMP-dependent pathways

Abstract Nitric oxide (NO) is known to regulate the release of arginine-vasopressin (AVP) and oxytocin (OT) by the paraventricular nucleus (PVN) and the supraoptic nucleus (SON). The aim of the current study was to identify in these nuclei the NO-producing neurons and the NO-receptive cells in mice. The determination of NO-synthesizing neurons was performed by double immunohistochemistry for the neuronal form of NO synthase (NOS), and AVP or OT. Besides, we visualized the NO-receptive cells by detecting cyclic GMP (cGMP), the major second messenger for NO, by immunohistochemistry on hypothalamus slices. Neuronal NOS was exclusively colocalized with OT in the PVN and the SON, suggesting that NO is mainly synthesized by oxytocinergic neurons in mice. By contrast, cGMP was not observed in magnocellular neurons, but in GABA-, tyrosine hydroxylase- and glutamate-positive fibers, as well as in GFAP-stained cells. The cGMP-immunostaining was abolished by incubating brain slices with a NOS inhibitor (L-NAME). Consequently, we provide the first evidence that NO could regulate the release of AVP and OT indirectly by modulating the activity of the main afferents to magnocellular neurons rather than by acting directly on magnocellular neurons. Moreover, both the NADPH-diaphorase activity and the mean intensity of cGMP-immunofluorescence were increased in monoamine oxidase A knock-out mice (Tg8) compared to control mice (C3H) in both nuclei. This suggests that monoamines could enhance the production of NO, contributing by this way to the fine regulation of AVP and OT release and synthesis. [source]


Magnetic field intensity study of the 1960 Kilauea lava flow, Hawaii, using the microwave palaeointensity technique

It is extremely valuable to study historic lava flows where the geomagnetic field at their time of extrusion is well known. In this study, two vertical sections, 16 m apart, have been sampled from the approximately 1 m thick 1960 Kilauea lava flow, Hawaii. Variations are seen in the rock-magnetic and palaeomagnetic properties between and within the two sections, indicating that there are small-scale lateral and vertical variations in the lava flow. The two sections showed different responses to microwave palaeointensity analysis. Section H6001 generally gave ideal linear behaviour on plots of natural remanent magnetization (NRM) lost against microwave-induced thermoremanent magnetization (TM,RM) gained, whilst the majority of samples from H6002 showed anomalous two-slope behaviour. When all plots were interpreted by taking the best-fitting line through all points, the flow mean intensity for H6001 was 31.6,±,3.6 ,T and that for H6002 was 37.1,±,6.4 ,T, compared with the expected intensity of 36 ,T. Additional historic flows need to be studied in order to ascertain whether this behaviour is typical of all lava, and whether it is best to always interpret NRM lost/TM,RM gained plots by taking the line of best fit regardless of shape. A computational model for impact failure with shear-induced dilatancy

Abstract It has been observed in plate impact experiments that some brittle solids may undergo elastic deformation at the shock wave front, and fail catastrophically at a later time when they are shocked near but below the apparent Hugoniot elastic limit. Because this phenomenon appears to have features different from those of usual inelastic waves, it has been interpreted as the failure wave. To design an effective numerical procedure for simulating impact failure responses, a three-dimensional computational damage model is developed in this paper. The propagation of the failure wave behind the elastic shock wave is described by a non-linear diffusion equation. Macroscopic shear-induced dilatancy is assumed and treated as a one-to-one measure of the mean intensity of microcracking. The damage evolution in time is determined based on the assumption that the deviatoric strain energy in the elastically compressed material (undamaged) is converted, through the damaging process, into the volumetric potential energy in the comminuted and dilated material. For the ease in large-scale simulations, the coupled damage diffusion equation and the stress wave equation are solved via a staggered manner in a single computational domain. Numerical solutions by using both the finite element method and the material point method, i.e. with and without a rigid mesh connectivity, are presented and compared with the experimental data available. It is shown that the model simulations capture the essential features of the failure wave phenomenon observed in shock glasses, and that the numerical solutions for localized failure are not mesh-dependent. Anguillicoloides crassus infection of European eel, Anguilla anguilla (L.), in inland waters of Estonia: history of introduction, prevalence and intensity

Summary Eel fishery in Estonian inland waters depends entirely on the stocking of glass eels or pre-grown (farmed) eels. Via importation of live eels of 20,30 cm length the non-indigenous swimbladder nematode Anguillicoloides crassus was probably introduced via Germany into Lake Võrtsjärv in 1988, and has since spread to many inland waters of Estonia. In 1992, the parasite was found in eel caught from Lake Võrtsjärv. Between 1992 and 2002 and additionally in 2008, we examined in total 870 eels from Lake Võrtsjärv (270 km2) and in 2008, 63 eels from three small lakes for adult A. crassus. The aim of the study was to obtain information on the variation of A. crassus infection in eels in Estonian lakes, to determine the temporal dynamics of prevalence and intensity of infection, and to establish a relationship between the length of host and intensity of infection in the eels in Lake Võrtsjärv. There appeared to be a pronounced variation in prevalences of infected eels (from 3.7% to 100%) between the four investigated lakes. However, in Lake Võrtsjärv, the prevalence of adult A. crassus infection remained stable (mean about 65%) for many years. The average number of nematode per infected eel (mean intensity) ranged from 12.6 ± 2.5 in 1993 to 4.0 ± 0.6 in 1999 in Lake Võrtsjärv, while it was significantly higher (P < 0.0001) in the period 1992,1998 compared to 1999,2002 and 2008. The mean number of parasites per swimbladder was not related to eel length and no statistical difference was found in the condition factor of infected and non-infected eels. Although under normal environmental conditions A. crassus has not caused serious disease problems to eels in the study area, high intensity of parasite infection may contribute to eel kills due to oxygen deficiency in winter under the ice in Lake Võrtsjärv. First record of Lamproglena pulchella Nordmann 1832 in the Pielach and Melk rivers, Austria

Summary Between April and November 2003, parasitological examinations of the nase Chondrostoma nasus L. and the chub Leuciscus cephalus L. from the neighbouring Melk and Pielach rivers in Lower Austria were conducted. Amongst various gill parasites, Lamproglena pulchella Nordmann 1832 was detected on both fish species, which was the first record of this parasitic crustacean in Austria. Physico-chemical examinations of water samples of the two rivers were carried out during the same period. The results indicate that general water parameters in the Melk were subjected to more vigorous changes than in the Pielach. Critical temperature levels and ammonia concentration as well as drastic changes in the ionic composition occurred more frequently in the Melk River. The observed distribution of L. pulchella indicates its sensitivity to such stress factors: there was no evidence of the parasite in the Melk River until late November and it only then occurred on the gills of L. cephalus with a prevalence of 20% and a mean intensity of 2. In the Pielach River, infestation on chub had already occurred in June with a prevalence of 40% and a mean intensity of 3, rising to 60% and 7 in November; 45% of the nase was also infested in November at a mean intensity of 3. Association among Fas expression in leucocytes, serum Fas and Fas-ligand concentrations and hepatic inflammation and fibrosis in chronic hepatitis C

Abstract Background: Replication of the hepatitis C virus (HCV) in peripheral blood mononuclear cells (PBMC) may impair immune functions and establish persistent infection. The aim of this study was to assess the influence of HCV on PBMC and their susceptibility to apoptosis in relation to liver inflammation and fibrosis. Methods: Eighty-one patients with chronic hepatitis C (CHC) were enrolled in this study. Flow cytometry was used to determine the amount of T cells (CD4+, CD8+), B cells (CD19+), monocytes (CD14+) and natural killer cells (CD16+) in the peripheral blood and the expression of CD95+ (CD95/APO-1) in each subset. Serum concentrations of sFas and sFasL were assessed by the enzyme-linked immunosorbent assay method. Results: An increased expression of Fas was observed in CD4+ and CD8+ cells in CHC. There was a more prominent expression of Fas on CD4+ cells in HCV genotype 1b in contrast to 3a. Increased Fas expression on CD4+ cells was seen in advanced stages of liver disease. Fas expression on monocytes was lower in advanced stages of liver inflammation and fibrosis. Serum sFas concentration was higher in CHC compared with the control group. There was an association between sFasL concentration and inflammatory activity in the liver. Serum sFasL concentration correlated positively with the mean intensity of fluorescence of the Fas receptor in CD4+ and CD8+ cells, granulocytes and monocytes. Conclusion: These findings indicate that there is an increased susceptibility of PBMC to apoptosis, which can be attributed to the constant contact of leucocytes with the inflamed liver tissue, or from direct HCV influence. GEOGRAPHICAL VARIATION OF THE PARASITE, PHYLLOBOTHRIUM DELPHINI (CESTODA), IN BALL'S PORPOISE, PHOCOENOIDES DALLI, IN THE NORTHERN NORTH PACIFIC, BERING SEA, AND SEA OF OKHOTSK

Abstract Prevalence of the larval cestode, Phyllobothrium delphini, was estimated from 2,445 Dall's porpoise, Phocoenoides dalli, from the incidental take of the Japanese high seas salmon drift-net fishery in the northwestern North Pacific Ocean, Bering Sea, and a local hand harpoon fishery in the southern Sea of Okhotsk. Prevalence of P. delphini was 22.7% in the northwestern North Pacific Ocean and 1.4% in the Bering Sea. This parasite was not found in the southern Sea of Okhotsk. Geographical differences in the prevalence of P. delphini may be due, at least in part, to regional differences in abundance of elasmobranchs known to feed on marine mammals and suspected as hosts of the parasite. Estimated intensity of infection of individual porpoises by P. delphini was low (estimated mean intensity of 3.5 plerocercoids per animal). This is a low intensity of infection compared to other species of small cetaceans studied and may be due to both differences in regional abundance of elasmobranchs and the comparatively short life span of P. dalli. Prevalence, self-care behaviors, and self-care activities for peripheral neuropathy symptoms of HIV/AIDS

Abstract As part of a larger randomized controlled trial examining the efficacy of an HIV/AIDS symptom management manual (n = 775), this study examined the prevalence of peripheral neuropathy in HIV-infected individuals at 12 sites in the USA, Puerto Rico, and Africa. Neuropathy was reported by 44% of the sample; however, only 29.4% reported initiating self-care behaviors to address the neuropathy symptoms. Antiretroviral therapy was found to increase the frequency of neuropathy symptoms, with an increased mean intensity of 28%. A principal axis factor analysis with Promax rotation was used to assess the relationships in the frequency of use of the 18 self-care activities for neuropathy, revealing three distinct factors: (i) an interactive self-care factor; (ii) a complementary medicine factor; and (iii) a third factor consisting of the negative health items of smoking, alcohol, and street drugs. The study's results suggest that peripheral neuropathy is a common symptom and the presence of neuropathy is associated with self-care behaviors to ameliorate HIV symptoms. The implications for nursing practice include the assessment and evaluation of nursing interventions related to management strategies for neuropathy. [source]


Fluorescent Detection of Rat Parathyroid Glands via 5-Aminolevulinic Acid,

Abstract Objective: Anatomic identification of parathyroid glands during surgery is challenging and time consuming. We sought to determine whether 5-aminolevulinic acid (5-ALA) could produce parathyroid gland fluorescence to improve their detection in a preclinical model. Methods: Thirty-two rats were administered 0 to 700 mg/kg of 5-ALA by intraperitoneal injection prior to neck exploration under the illumination of a blue light (380,440 nm). Tissue fluorescence was assessed at 1, 2, or 4 hours postinjection and then removed for histologic confirmation of parathyroid tissue. Results: Rat parathyroid glands could not be visualized under ambient light. At dosages of 300 mg/kg or greater, bilateral parathyroid glands were visualized in 18 of 19 rats using blue light illumination. At dosages less than 300 mg/kg, parathyroid gland fluorescence was detected in only 1 of 13 rats. At 2 hours after 5-ALA administration, the net mean intensity of parathyroid gland fluorescence was optimal with a dose of 500 mg/kg. At both 1 and 4 hours after 5-ALA injection, the net mean intensity of parathyroid gland fluorescence was optimal at the highest dose (700 mg/kg) and positively correlated with dosage increases. Conclusion: 5-ALA can be used to selectively detect parathyroid tissue from surrounding tissue in a preclinical model. Our data support the use of this technique in the clinical setting. [source]


Comparison of Combination Plasmapheresis/IVIg/Anti-CD20 Versus High-Dose IVIg in the Treatment of Antibody-Mediated Rejection

Different strategies appear to improve the success in treatment of antibody-mediated rejection (AMR), although no one best method has yet emerged. The objective of this study was to compare the efficacy of the combination of Plasmapheresis/intravenous immunoglobulin (IVIg)/anti-CD20-based regimes versus high-dose IVIg alone in the treatment of AMR. Group A (12 patients) was treated with high-dose IVIg between January 2000 and December 2003; group B (12 patients) was treated by Plasmapheresis/IVIg/anti-CD20 between January 2004 and December 2005. Graft survival at 36 months was 91.7% in group B versus 50% in group A (p = 0.02). Donor-specific human leukocyte antigens (DSA) levels detected by Luminex single antigen (Luminex SA) and ELISA, 3 months postrejection are significantly lower in group B than in group A: DSA ELISA class 2 score 6,8 (p = 0.02), DSA mean intensity of fluorescence (MFI) max (p = 0.009) and DSA mean MFI (p = 0.0004). The persistence of elevated DSA levels posttreatment is more frequent in patients with graft loss as compared to those with preserved renal function: score 6,8 on ELISA (p = 0.04); mean MFI (p = 0.00009) and MFImax (p = 0.018). We conclude that: (1) high dose IVIg alone is inferior to Plasmapheresis/IVIg/anti-CD20 as therapy for AMR and (2)DSA postrejection can be quantified using solid phase assays, showing that 3 months after AMR, DSA levels are higher in patients with graft loss. Salmon lice infection of wild sea trout and Arctic char in marine and freshwaters: the effects of salmon farms

Abstract The abundance of salmon lice and the physiological effects of infection were examined in two stocks of sympatric sea trout and anadromous Arctic char in northern Norway. One stock feed in a coastal area with extensive salmon farming (exposed locality), while the other feed in a region with little farming activity (unexposed locality). The results showed that the lice infection was significantly higher at the exposed locality, at which the mean intensity of infection peaked in June and July at over 100 and 200 lice larvae per fish respectively. At the exposed locality we also observed a premature return to freshwater of the most heavily infected fish. Such behaviour has previously been interpreted as a response by the fish to reduce the stress caused by the infection and/or to enhance survival. Blood samples taken from sea trout at sea at the exposed locality showed a positive correlation between intensity of parasite infection and an increase in the plasma cortisol, chloride and blood glucose concentrations, while the correlations from sea trout in freshwater were more casual. Several indices pointed towards an excessive mortality of the heaviest infected fish, and 47% of the fish caught in freshwater and 32% of those captured at sea carried lice at intensities above the level that has been shown to induce mortality in laboratory experiments. Furthermore, almost half of all fish from the exposed locality had lice intensities that would probably cause osmoregulatory imbalance. High salmon lice infections may therefore have profound negative effects upon wild populations of sea trout. At the unexposed location, the infection intensities were low, and few fish carried more than 10 lice. These are probably within the normal range of natural infection and such intensities are not expected to affect the stock negatively. [source]


Microarray-based gene expression analysis as a process characterization tool to establish comparability of complex biological products: Scale-up of a whole-cell immunotherapy product

Abstract Whole-cell immunotherapies and other cellular therapies have shown promising results in clinical trials. Due to the complex nature of the whole cell product and of the sometimes limited correlation of clinical potency with the proposed mechanism of action, these cellular immunotherapy products are generally not considered well characterized. Therefore, one major challenge in the product development of whole cell therapies is the ability to demonstrate comparability of product after changes in the manufacturing process. Such changes are nearly inevitable with increase in manufacturing experience leading to improved and robust processes that may have higher commercial feasibility. In order to comprehensively assess the impact of the process changes on the final product, and thus establish comparability, a matrix of characterization assays (in addition to lot release assays) assessing the various aspects of the cellular product are required. In this study, we assessed the capability of DNA-microarray-based, gene-expression analysis as a characterization tool using GVAX cancer immunotherapy cells manufactured by Cell Genesys, Inc. The GVAX immunotherapy product consists two prostate cancer cell lines (CG1940 and CG8711) engineered to secrete human GM-CSF. To demonstrate the capability of the assay, we assessed the transcriptional changes in the product when produced in the presence or absence of fetal bovine serum, and under normal and hypoxic conditions, both changes intended to stress the cell lines. We then assessed the impact of an approximately 10-fold process scale-up on the final product at the transcriptional level. These data were used to develop comparisons and statistical analyses suitable for characterizing culture reproducibility and cellular product similarity. Use of gene-expression data for process characterization proved to be a reproducible and sensitive method for detecting differences due to small or large changes in culture conditions as might be encountered in process scale-up or unanticipated bioprocess failures. Gene expression analysis demonstrated that cell products of representative lots under the same production process and at the same production scale were statistically identical. Large process changes that resulted from the artificial stress conditions used (absence of FBS and induction of hypoxia) displayed profoundly different gene expression patterns. We propose the use of simple t -test analysis in combination with the herein introduced expression ratio with mean intensity (ERMI) analysis as useful tools for process characterization by global gene expression analysis. Influence of disease features on adherence to prophylactic migraine medication

Objectives,,, Randomized controlled trials of prophylactic treatments for migraine focus on the effects in an ideal situation and underestimate the impact of non-adherence, which in this study was examined in a natural setting. Materials and methods,,, A sample of 174 adult migraineurs with a current prescription of pharmacological prophylaxis were consecutively recruited at a specialist clinic. Logistic regression analysis was performed to analyse the association between adherence (self-reported with the Medication Adherence Report Scale) and number of years with migraine, frequency of attacks, number of days with migraine per month, attack duration, presence of cardinal features, mean intensity of pain, and recovery between attacks. Results,,, One third (35%) were non-adherent. Neither demographic characteristics nor any of the disease specific variables were significantly associated with adherence. Conclusion,,, Characteristics of the disease per se did not predict non-adherence which was also observed among patients with severe migraine. The full benefit of drugs cannot be realized at currently achieved levels of adherence. [source]
