Accumulation of DDT and mercury in prothonotary warblers (Protonotaria citrea) foraging in a heterogeneously contaminated environment

Abstract Foraging areas of adult prothonotary warblers (Protonotaria citrea) were determined using standard radiotelemetry techniques to determine if soil concentrations of p,p,dichlorodiphenyltrichloroethane (p,p,DDT) and mercury in foraging areas could be used to predict contaminant levels in diets and tissues of nestling warblers. Adult warblers were fitted with transmitters and monitored for approximately 2 d while foraging and feeding 6- to 8-d-old nestlings. Foraging ecology data were integrated with contaminant levels of soil, diets, and tissues into a comprehensive analysis of geographic variation in contaminant exposure and uptake using linear regression. Concentrations of 1,1-dichloro-2,2-bis(p -chlorophenyl)ethylene (DDE) and mercury in nestling tissues varied considerably across the study site. Mean concentration of DDE was greater in eggs than all other tissues, with individual samples ranging from 0.24 to 8.12 ,g/kg. In general, concentrations of DDT in soil were effective in describing the variation of contaminants in adipose samples. Concentrations of mercury in soils accounted for 78% of the variation in kidney samples. This was the best relationship of any of the paired variables. All other relationships showed relatively poor predictive ability. [source]


INTRACELLULAR CYANOBACTERIAL SYMBIONTS IN THE MARINE DIATOM CLIMACODIUM FRAUENFELDIANUM (BACILLARIOPHYCEAE)

The diatom Climacodium frauenfeldianum Grunow was collected in the tropical Atlantic and Pacific Oceans. Observations with epifluorescence microscopy revealed that this diatom contained coccoid symbionts (2.5,3.5 ,m) with a typical cyanobacterial fluorescence in addition to that of their own chloroplasts. Mean concentration of C. frauenfeldianum for 28 stations in the SW tropical Pacific Ocean was 530 x 103 (SE = 1372) cells·m,2, with highest concentration (mean 17.5 cells·L,1) at 40-m depth. The symbiosis was only observed at water temperatures between 26.3 and 28.9° C, with highest concentrations at 27.7° C. Three almost complete 16S rDNA sequences from one sample were determined, and they were identical. The phylogenetic analysis of this 16S rDNA sequence and those from other cyanobacteria and plastids revealed that it was closely related to the 16S rDNA sequence from Cyanothece sp. ATCC 51142. Cyanothece sp. ATCC 51142 is a unicellular nitrogen-fixing cyanobacterium isolated from a coastal marine environment and has ultrastructural features similar to the symbionts of C. frauenfeldianum. The close relationship between Cyanothece sp. and the cyanobacterial symbiont in C. frauenfeldianum suggests the potential for nitrogen fixation in the symbiosis. [source]


Fish consumption and body burden of organochlorines among lower hudson urban anglers

Abstract Background Polychlorinated biphenyls and other organochlorines have contaminated the Hudson River and New York/New Jersey Harbor for many decades. Although body burdens and health effects resulting from exposure to these pollutants have been measured in other populations, little is known about anglers from the lower Hudson area. Methods Anglers were recruited from fishing clubs and along piers on the lower Hudson River. One hundred twenty-five participants provided blood samples and questionnaire information. Serum was analyzed at Mount Sinai School of Medicine for organochlorines. Results Mean concentration of total PCB congeners was 4.78 µg/L. A non-significant adjusted mean difference of ,0.59 µg/L was observed between local fish eaters and non-eaters with higher levels among the non-eaters. Similar negative, but non-significant associations were observed for p,p'-DDT, HCB, and ,-BHC. For pp'-DDE, and several chlordane residues, levels were higher among anglers reporting local fish consumption, but the difference was statistically significant only for TN. Conclusions These findings suggest eating fish from the lower Hudson River and NY/NJ harbor is not associated with an increased body burden of PCBs or most other organochlorines. Am. J. Ind. Biochemical and histopathological effects in pearl dace (Margariscus margarita) chronically exposed to a synthetic estrogen in a whole lake experiment

Abstract Potential effects of exposure to the synthetic estrogen 17,-ethynylestradiol (EE2) were examined in several species of fish from a lake experimentally treated with environmentally relevant concentrations of the contaminant. Ethynylestradiol was added to Lake 260, a small Precambrian shield lake at the Experimental Lakes Area in northwestern Ontario, Canada, from May to October of 2001, 2002, and 2003. Mean concentrations of EE2 in epilimnetic waters ranged between 4.5 and 8.1 ng/L during the three years, with overall means of 6.1 (±2.8), 5.0 (±1.8), and 4.8 (±1.0) ng/L for the three years, respectively. Male and female pearl dace (Margariscus margarita) captured after EE2 additions began contained up to 4,000-fold higher concentrations of the egg yolk precursor vitellogenin than fish captured from the same lake before the EE2 additions or when compared to fish from reference lakes. Edema in the ovaries, inhibited development of testicular tissue, intersex, and histopathological kidney lesions were all evident in fish exposed to EE2. Some indications that EE2 exposure affected in vitro steroidogenic capacity of the ovaries and the testes existed, although results were not always consistent between years. Pearl dace abundance was similar in the lake treated with EE2 and the reference lake. A trend exists toward a reduced overall population of pearl dace from the treated and reference lakes, as do indications that young-of-the-year size classes are less abundant in the EE2-treated lake. Biochemical and histopathological impacts observed in fish exposed to EE2 in this study have not yet been linked to clear population level impacts in pearl dace. Abstract Concentrations of 2,3,7,8-tetrachlorodibenzo- p -dioxin (TCDD) in tree swallows (Tachycineta bicolor) nesting along the Woonasquatucket River northwest of Providence (RI, USA) in 2000 and 2001 were some of the highest ever reported in avian tissues. Mean concentrations in eggs ranged from 300 to > 1,000 pg/g wet weight at the two most contaminated ponds, Allendale and Lyman. Mean egg concentrations at Greystone, the upstream reference pond, were 12 and 29 pg/g. Positive accumulation rates and concentrations in diet samples from 12-day-old nestlings indicated that the contamination was accumulated locally. Concentrations in diet of between 71 and 219 pg/g wet weight were more than 6 and 18 times higher than concentrations considered safe for birds (10,12 pg/g). Hatching success was negatively associated with concentration of TCDD in eggs. Only about half the eggs hatched at Allendale compared with >77% at Greystone. The national average for hatching success in successful nests is 85%. No other contaminants, such as polychlorinated biphenyls and mercury, were present in any sample at concentrations known to affect avian reproduction. Three bioindicators, half-peak coefficient of geometric variation, ethoxyresorufin- O -dealkylase activity, and brain asymmetry were assessed relative to TCDD contamination. [source]


Diel Changes in Phytoplankton Composition and Abundance in the Surface and Sub-Surface Strata from a Shallow Eutrophic Pond

Abstract Representative phytoplankton assemblages were identified in a eutrophic pond over a 24 hour period. One assemblage characterized species in the surface (neuston) layer and another consisted of algae from 2, 5, and 20 cm sub-surface depths. The surface layer (0 cm) included a similar, but less diverse assemblage of species, and a lower abundance of cells per unit volume, than those at the lower depths. At each of the sub-surface depths (2,20 cm), the major phytoplankton components initially followed similar patterns of abundance in reference to the time and depth of sample collections then later differed in their abundance levels. The dominant algae were chlorophytes, cryptophytes, diatoms, and cyanobacteria. Mean concentrations of total phytoplankton, over the 24 hours for the surface, were 3.3 × 103 cells ml,1, compared to 36.9 × 103 cells ml,1 for depths 2,20 cm. The autotrophic picoplankton abundance was recorded separately from the phytoplankton with mean concentrations of 472.9 × 103 cells ml,1 in the surface layer and 623.0 × 103 cells ml,1 for the three sub-surface depths. Adrenocortical and Pituitary Glucocorticoid Feedback in Abstinent Alcohol-Dependent Women

Background:, The long-term ingestion of alcohol diminishes hypothalamic,pituitary,adrenal (HPA) axis reactivity in alcohol-dependent men, potentially altering future relapse risk. Although sex differences in HPA axis functioning are apparent in healthy controls, disruptions in this system have received little attention in alcohol-dependent women. In this study, we assessed the basal secretory profile of adrenocorticotropic hormone (ACTH) and cortisol, adrenocortical sensitivity in both the presence and absence of endogenous corticotropic pituitary activation, and feedback pituitary glucocorticoid sensitivity to dexamethasone. Methods:, Seven women 4- to 8-week abstinent alcohol-only dependent subjects and 10 age-matched female healthy controls were studied. All subjects were between 30 and 50 years old, not taking oral contraceptives, and were studied during the early follicular phase of their menstrual cycle. Circulating concentrations of ACTH and cortisol were measured in blood samples collected at frequent intervals from 2000 to 0800 hour. A submaximal dose of cosyntropin (0.01 ,g/kg), a synthetic ACTH (1,24), was administered at 0800 hour to assess adrenocortical sensitivity. In a separate session, low-dose cosyntropin was also administered following high-dose dexamethasone (8 mg intravenous) to assess adrenocortical sensitivity in the relative absence of endogenous ACTH. In addition, the ACTH response to dexamethasone was measured to determine the pituitary glucocorticoid negative feedback. Sessions were 5 days apart, and blood draws were obtained every 5 to 10 minutes. Results:, Mean concentrations and pulsatile characteristics of ACTH and cortisol over 12 hours were not statistically different between the 2 groups. Healthy controls had a somewhat higher (p < 0.08) net peak, but not net integrated, cortisol response to cosyntropin relative to the alcohol-dependent women. There were no significant group differences in either the ACTH or cortisol response to dexamethasone nor in the net cortisol response to cosyntropin following dexamethasone. Conclusion:, Significant differences in pituitary,adrenal function were not apparent between alcohol-dependent women and matched controls. Despite the small n, it appears that alcohol-dependent women do not show the same disruptions in HPA activity as alcohol-dependent men. These findings may have relevance for gender-specific treatment effectiveness. [source]


DISTRIBUTION OF SEDIMENT PHOSPHORUS POOLS AND FLUXES IN RELATION TO ALUM TREATMENT,

ABSTRACT: The distribution of sediment physical characteristics, sediment phosphorus (P) pools, and laboratory-based rates of P release from the sediments were used to identify regions and dosage for alum treatment in Wind Lake, Wisconsin. Using variations in sediment moisture content, we identified an erosional zone at depths < 1.4 m and an accumulation zone at depths > 2.6 m. Mean concentrations of porewater P, loosely-bound P, iron- and aluminum-bound P, and mean rates of P release from sediments under anoxic conditions were high in the accumulation zone compared to sediment P characteristics in the erosional zone, indicating focusing of readily mobilized sediment P pools from shallow regions and accumulation to deep regions. We determined that a future alum treatment for control of internal P loading would be most effective at depths > 2.6 in the accumulation zone. The mean rate of anoxic P release from sediments encountered in the accumulation zone (8.3 mg m -2 d -1) was used in conjunction with a summer anoxic period of 122 d, and a treatment area of 1.6 km2 to estimate an internal P load of 1,600 kg to be controlled. Our results suggest that an understanding of the distribution of sediment P pools and P fluxes in lakes provides a strategy for estimating alum dosage and application areas. [source]


Comparison of Selected Endocrine Parameters During Luteal Phase and Pregnancy in German Shepherd Dogs and Beagles

Contents Concentrations of progesterone, prolactin and relaxin in serum at predetermined intervals after ovulation (day 0) in non-pregnant and pregnant normocyclic Beagles were assayed and results compared with those observed in German Shepherd dogs (GSD) in a previous study. The goal was to determine possible reproductive hormone specificities related to the GSD breed. Furthermore, the effects of medroxyprogesterone acetate (MPA)-treatment in non-pregnant Beagles and of progesterone supplementation in pregnant Beagles on the hormone concentrations were examined. Mean concentrations of progesterone and prolactin were not different in the non-pregnant Beagles compared with those seen in non-pregnant GSD, except at days 50,60, when progesterone concentrations were found to be higher in Beagles (p < 0.05). Mean progesterone concentrations in pregnant Beagles at days 50,60 after ovulation (day 0) were higher (p < 0.05) than in GSD at that time, but not at earlier time periods. Prolactin concentrations were higher (p < 0.05) in Beagles throughout pregnancy compared with those in the GSD. Mean relaxin concentrations were numerically but not significantly lower in GSD than in Beagles throughout pregnancy. A 10-day oral MPA treatment did not affect progesterone or prolactin secretion in normocyclic non-pregnant Beagles. Medroxyprogesterone acetate serum concentrations were approximately 3.9 ng/ml during treatment and decreased to 0.42 and 0.021 ng/ml within 5 and 15 days after end of treatment, respectively. Intramuscular progesterone supplementation from days 30 to 40 in pregnant Beagles resulted in higher concentrations of progesterone in the 36- to 45-day time periods; prolactin and relaxin concentrations were not significantly affected during or after treatment compared with administration of placebo. The results suggest a tendency towards deficient luteal function in the short-cycle GSD bitches previously studied, which in pregnancy may reflect the observed decreased prolactin concentrations; the possibility that GSD relaxin secretion is deficiency required needs further study. As oral treatment with MPA did not affect progesterone and prolactin release, it may be useful for studying luteal function in pregnant bitches with suspected hypoluteoidism. [source]


Determination of uric acid concentrations in human tear fluid, aqueous humour and serum

Abstract. Purpose:, The aim of this investigation was to assess and compare levels of uric acid in human tear fluid, aqueous humour and serum in cataract patients in order to obtain information about uric acid-dependent antioxidative status. Methods:, Reflex tear fluid, aqueous humour and serum were obtained from 103 cataract patients (52 women, 51 men). Uric acid was determined by a reversed phase chromatographic method with electrochemical detection. The Randox test was used to measure the contribution of uric acid to the total antioxidative status (TAS) of these body fluids. Results:, Mean concentrations of uric acid were 20 ± 8 ,g/ml in tear fluid, 18 ± 6 ,g/ml in aqueous humour and 58 ± 15 ,g/ml in serum. Significantly less uric acid was found in tear fluid (p < 0.01) and aqueous humour (p < 0.01) in female cataract patients compared with male cataract patients. There was a significant positive correlation between uric acid content in tear fluid, aqueous humour and serum, respectively (p < 0.01). The contribution of uric acid to TAS (Randox test) amounted to 38% in tear fluid, 10% in aqueous humour and 37% in serum. Conclusions:, Uric acid was found in comparable amounts in tear fluid and aqueous humour, but these were two- to threefold lower than in serum. Female cataract patients had lower uric acid values in tear fluid and aqueous humour compared with male cataract patients. Uric acid is an important antioxidant, as proven by its contribution to TAS as determined by the Randox test. The precise role of uric acid in the antioxidative reactions in tear fluid and aqueous humour is still under debate. [source]


Agrichemicals in surface water and birth defects in the United States

Abstract Objectives: To investigate if live births conceived in months when surface water agrichemicals are highest are at greater risk for birth defects. Methods: Monthly concentrations during 1996,2002 of nitrates, atrazine and other pesticides were calculated using United States Geological Survey's National Water Quality Assessment data. Monthly United States birth defect rates were calculated for live births from 1996 to 2002 using United States Centers for Disease Control and Prevention natality data sets. Birth defect rates by month of last menstrual period (LMP) were then compared to pesticide/nitrate means using logistical regression models. Results: Mean concentrations of agrichemicals were highest in April,July. Total birth defects, and eleven of 22 birth defect subcategories, were more likely to occur in live births with LMPs between April and July. A significant association was found between the season of elevated agrichemicals and birth defects. Conclusion: Elevated concentrations of agrichemicals in surface water in April,July coincided with higher risk of birth defects in live births with LMPs April,July. While a causal link between agrichemicals and birth defects cannot be proven from this study an association might provide clues to common factors shared by both variables. [source]


Effect of perinatal asphyxia on thyroid-stimulating hormone and thyroid hormone levels

Aim: To compare serum concentrations of thyroid hormones,T4, T3, free T4 (FT4) and reverse T3 (rT3),and thyroid-stimulating hormone (TSH) found in the umbilical cord blood of term newborns with and without asphyxia and those found in their arterial blood collected between 18 and 24 h after birth. A further aim of the study was to assess the association between severity of hypoxic-ischemic encephalopathy and altered thyroid hormone and TSH levels, and between mortality and FT4 levels in the arterial blood of newborns between 18 and 24 h of life. Methods: A case-control study was carried out. The case group comprised 17 term newborns (Apgar score ,3 and ,5 at the first and fifth minutes; umbilical cord blood pH ,7.15) who required bag and mask ventilation for at least one minute immediately after birth. The control group consisted of 17 normal, term newborns (Apgar score ,8 and ,9 at the first and fifth minutes; umbilical cord blood pH ,7.2). Cord blood and arterial blood samples were collected immediately after birth and 18 to 24 h after birth, respectively, and were used in the blood gas analysis and to determine serum concentrations of T4, T3, FT4, rT3 and TSH by radioimmunoassay. All newborns were followed-up until hospital discharge or death. Results: Gestational age, birthweight, sex, size for gestational age, mode of delivery and skin color (white and non-white) were similar for both groups. No differences were found in mean levels of cord blood TSH, T4, T3 and FT4 between the groups. In the samples collected 18 to 24 h after birth, mean levels of TSH, T4, T3 and FT4 were significantly lower in the asphyxiated group than in the control group. Mean concentrations of arterial TSH, T4 and T3 between 18 and 24 h of life were lower than concentrations found in the cord blood analysis in asphyxiated newborns, but not in controls. In addition, asphyxiated newborns with moderate/severe hypoxic-ischemic encephalopathy presented significantly lower mean levels of TSH, T4, T3 and FT4 than those of controls. None of the asphyxiated newborns with FT4 ,2.0 ng/dl died; 6 out of the 11 asphyxiated newborns with FT4 < 2.0 ng/dl died. Conclusions: Serum concentrations of TSH, T4, T3 and FT4 are lower in asphyxiated newborns than in normal newborns between 18 and 24 h of life; this suggests central hypothyroidism secondary to asphyxia. Asphyxiated newborns with moderate/severe hypoxic-ischemic encephalopathy present a greater involvement of the thyroid function and consequently a greater risk of death. [source]


Experimental determination of saltating glass particle dispersion in a turbulent boundary layer

Abstract A horizontal saltation layer of glass particles in air is investigated experimentally over a flat bed and also over a triangular ridge in a wind tunnel. Particle concentrations are measured by light scattering diffusion (LSD) and digital image processing, and velocities using particle image velocimetry (PIV). All the statistical moments of the particle concentration are determined such as mean concentration, root mean square concentration fluctuations, skewness and flatness coefficients. Over the flat bed, it is confirmed that the mean concentration decreases exponentially with height, the mean dispersion height being a significant length scale. It is shown that the concentration distribution follows quite well a lognormal distribution. Over the ridge, measurements were made at the top of the ridge and in the cavity region and are compared with measurements without the ridge. On the hill crest, particles are retarded, the saltation layer decreases in thickness and concentration is increased. Sediment transport in a highly regulated fluvial system during two consecutive floods (lower Ebro River, NE Iberian Peninsula)

Abstract The transfer of sediment through a highly regulated large fluvial system (lower Ebro River) was analysed during two consecutive floods by means of sediment sampling. Suspended sediment and bedload transport were measured upstream and downstream of large reservoirs. The dams substantially altered flood timing, particularly the peaks, which were advanced downstream from the dams for flood control purposes. The suspended sediment yield upstream from the dams was 1 700 000 tonnes, which represented nearly 99 per cent of the total solid yield. The mean concentrations were close to 0·5 g l,1. The sediment yield downstream from the dams was an order of magnitude lower (173 000 tonnes), showing a mean concentration of 0·05 g l,1. The dams captured up to 95 per cent of the fine sediment carried in suspension in the river channel, preventing it from reaching the lowermost reaches of the river and the delta plain. Total bedload transport upstream from the dams was estimated to be about 25 000 tonnes, only 1·5 per cent of the total load. The median bedload rate was 100 gms,1. Below the dams, the river carried 178 000 tonnes, around 51 per cent of the total load, at a mean rate of 250 g ms,1. The results of sediment transport upstream and downstream from the large dams illustrate the magnitude of the sediment deficit in the lower Ebro River. The river mobilized a total of 350 000 tonnes in the downstream reaches, which were not replaced by sediment from upstream. Therefore, sediment was necessarily entrained from the riverbed and channel banks, causing a mean incision of 33 mm over the 27 km long study reach, altogether a significant step towards the long-term degradation of the lower Ebro River. Stimulation of reproductive growth in rainbow trout (Oncorhynchus mykiss) following exposure to treated sewage effluent

Abstract Rainbow trout (Oncorhynchus mykiss) were exposed to 1.5 and 15% v/v secondary treated sewage effluent for 32 weeks in flow-through mesocosms. The exposure encompassed the full period of reproductive development for rainbow trout. Trout did not show any evidence of a dose-dependent change in growth. Fish exposed to 15% effluent were the only group to show mortality (5%) over the duration of the experiment. Trout at the highest effluent concentration had significantly higher liver size than reference water fish. Both male and female trout in the 15% exposure group also exhibited significantly higher gonad weight than the reference group. In female trout, this gonad size increase could be explained by higher egg numbers. Female and male trout both displayed a significant increase in plasma 17,-estradiol levels after exposure to 15% effluent, while neither sex had dose-dependent differences in plasma testosterone. Male trout displayed elevated vitellogenin levels and reduced plasma 11-ketotestosterone concentration after exposure to 15% effluent. Chemical examination of steroidal compounds, including both estrogens and androgens, in the wastewater revealed that only estrone was detectable at a mean concentration of 4.5 ng/L. It is assumed that the effects observed in trout exposed to 15% effluent were consistent with stimulation of reproductive development due to very low levels of estrogens. Overall, long-term exposure to treated sewage effluent containing low levels of estrogen did not have significant negative implications for reproductive development in rainbow trout. [source]


Respiration and annual fungal production associated with decomposing leaf litter in two streams

Summary 1. We compared fungal biomass, production and microbial respiration associated with decomposing leaves in one softwater stream (Payne Creek) and one hardwater stream (Lindsey Spring Branch). 2. Both streams received similar annual leaf litter fall (478,492 g m,2), but Lindsey Spring Branch had higher average monthly standing crop of leaf litter (69 ± 24 g m,2; mean ± SE) than Payne Creek (39 ± 9 g m,2). 3. Leaves sampled from Lindsey Spring Branch contained a higher mean concentration of fungal biomass (71 ± 11 mg g,1) than those from Payne Creek (54 ± 8 mg g,1). Maximum spore concentrations in the water of Lindsay Spring Branch were also higher than those in Payne Creek. These results agreed with litterbag studies of red maple (Acer rubrum) leaves, which decomposed faster (decay rate of 0.014 versus 0.004 day,1), exhibited higher maximum fungal biomass and had higher rates of fungal sporulation in Lindsey Spring Branch than in Payne Creek. 4. Rates of fungal production and respiration per g leaf were similar in the two streams, although rates of fungal production and respiration per square metre were higher in Lindsey Spring Branch than in Payne Creek because of the differences in leaf litter standing crop. 5. Annual fungal production was 16 ± 6 g m,2 (mean ± 95% CI) in Payne Creek and 46 ± 25 g m,2 in Lindsey Spring Branch. Measurements were taken through the autumn of 2 years to obtain an indication of inter-year variability. Fungal production during October to January of the 2 years varied between 3 and 6 g m,2 in Payne Creek and 7,27 g m,2 in Lindsey Spring Branch. 6. Partial organic matter budgets constructed for both streams indicated that 3 ± 1% of leaf litter fall went into fungal production and 7 ± 2% was lost as respiration in Payne Creek. The solute budget of a forest catchment and solute fluxes within a Pinus radiata and a secondary native forest site, southern Chile

Abstract Solute concentrations and fluxes in rainfall, throughfall and stemflow in two forest types, and stream flow in a 90 ha catchment in southern Chile (39°44,S, 73°10,W) were measured. Bulk precipitation pH was 6·1 and conductivity was low. Cation concentrations in rainfall were low (0·58 mg Ca2+ l,1, 0·13 mg K+ l,1, 0·11 mg Mg2+ l,1 and <0·08 mg NH4,N l,1), except for sodium (1·10 mg l,1). Unexpected high levels of nitrate deposition in rainfall (mean concentration 0·38 mg NO3,N l,1, total flux 6·3 kg NO3,N ha,1) were measured. Concentrations of soluble phosphorous in bulk precipitation and stream flow were below detection limits (<0·09 mg l,1) for all events. Stream-flow pH was 6·3 and conductivity was 28·3 ,s. Stream-water chemistry was also dominated by sodium (2·70 mg l,1) followed by Ca, Mg and K (1·31, 0·70 and 0·36 mg l,1). The solute budget indicated a net loss of 3·8 kg Na+ ha,1 year,1, 5·4 kg Mg2+ ha,1 year,1, 1·5 kg Ca2+ ha,1 year,1 and 0·9 kg K+ ha,1 year,1, while 4·9 kg NO3,N ha,1 year,1 was retained by the ecosystem. Stream water is not suitable for domestic use owing to high manganese and, especially, iron concentrations. Throughfall and stemflow chemistry at a pine stand (Pinus radiata D. Don) and a native forest site (Siempreverde type), both located within the catchment, were compared. Nitrate fluxes within both forest sites were similar (1·3 kg NO3,N ha,1 year,1 as throughfall). Cation fluxes in net rainfall (throughfall plus stemflow) at the pine stand generally were higher (34·8 kg Na+ ha,1 year,1, 21·5 kg K+ ha,1 year,1, 5·1 kg Mg2+ ha,1 year,1) compared with the secondary native forest site (24·7 kg Na+ ha,1 year,1, 18·9 kg K+ ha,1 year,1 and 4·4 kg Mg2+ ha,1 year,1). Effect of subtherapeutic concentrations of tylosin on the inhibitory stringency of a mixed anaerobe continuous-flow culture of chicken microflora against Escherichia coli O157:H7

Abstract Aims: The aim of this study was twofold: first to determine the effect of subtherapeutic concentrations of tylosin, a macrolide antibiotic used for growth promotion, on a mixed anaerobic continuous-flow fermentation culture of chicken gastrointestinal microorganisms (CCF) and secondly, to determine if these concentrations would allow persistence of Escherichia coli O157:H7 in CCF. Methods and Results: CCF was treated with tylosin at 10·0, 20·0 and 40·0 ,g ml,1. Tylosin treatment resulted in a significant (P < 0·0001) decrease in total volatile fatty acids (VFAs) from a mean concentration of 101 ± 10·8 ,mol ml,1 in control cultures to 32·0 ± 6·3 and 40·2 ± 9·6 ,mol ml,1 in 10 and 40 ,g ml,1 treated cultures, respectively. Untreated CCF challenged with E. coli O157:H7 cleared the challenge microorganism in 7 days at a rate of 0·96 log10 CFU ml,1 day,1. In contrast, E. coli O157:H7 persisted in all tylosin treated cultures. Conclusions: In the presence of tylosin, E. coli O157:H7 was able to persist in the CCF culture. The significant decrease in the production of VFAs may have been a contributing factor. Significance and Impact of the Study: The use of low-level, growth-promoting antimicrobials may compromise the ability of normal microflora that serve as a natural host defence against infection. ORIGINAL ARTICLE: Investigation of the prediction accuracy of vancomycin concentrations determined by patient-specific parameters as estimated by Bayesian analysis

Summary Background/Objective:, There have been many studies of therapeutic drug monitoring (TDM) of vancomycin (VCM) based on Bayesian analysis, but there have been no reports of the accuracy of prediction based on Bayesian-estimated patient-specific parameters. This study was conducted to compare the accuracy of prediction based on the population pharmacokinetic (PPK) method and Bayesian-estimated parameters. Method:, The subjects were 22 patients who were treated with VCM for MRSA infection and whose blood was sampled twice or more during the administration period. The concentrations between the blood samples were predicted based on the concentrations in the first blood samples based on the PPK method using mean parameters for the Japanese population and Bayesian-estimated patient-specific pharmacokinetic parameters. The mean prediction error (ME), mean absolute error (MAE) and root mean squared error (RMSE) were compared to examine the accuracy of prediction based on Bayesian-estimated patient-specific parameters. Results and discussion:, The mean measured VCM concentration was 10·43 ± 5·19 ,g/mL, whereas the mean concentration predicted based on the PPK method was 8·52 ± 4·34 ,g/mL, with an ME of ,1·91, MAE of 2·93 and RMSE of 3·21. The mean concentration predicted based on patient-specific parameters was 9·62 ± 4·95 ,g/mL with ME of ,0·81, MAE of 1·38 and RMSE of 1·74. The ME and MAE for the concentrations predicted using patient-specific parameters were smaller compared with those predicted using the PPK method (P = 0·0471 and 0·0003, respectively), indicating superior prediction with a significant difference between approaches. Conclusion:, Prediction using Bayesian estimates of patient-specific parameters was better than by the PPK method. However, when using patient-specific parameters it is still necessary to fully understand the clinical status of the patient and frequently determine VCM concentrations. [source]


Consumption of vegetables alters morning urinary 6-sulfatoxymelatonin concentration

Abstract:, Melatonin, which is contained in certain vegetables, may have an influence on circulatory melatonin concentrations. This study examined the effects of the consumption of vegetables on 6-sulfatoxymelatonin concentrations in morning urine. Ninety-four healthy women aged 24,55 were recruited through a city public health center in Japan. The women randomly allocated to the intervention group were requested to consume high amounts of six selected vegetables, with a target of 350 g/day for 65 days, while those in the control group were asked to avoid the same six vegetables during the same period. First-void morning urine was collected before and at the end of the intervention period, and creatinine-adjusted 6-sulfatoxymelatonin concentrations were measured. At the end of the intervention period, daily mean intake of melatonin from the six vegetables was 1288.0 ng in the intervention group and 5.3 ng in the control group. In the intervention group, the mean concentration of 6-sulfatoxymelatonin changed from 48.1 [95% confidence interval (CI): 40.4,57.2] ng/mg creatinine to 49.6 (95% CI: 42.8,57.3) ng/mg creatinine across the intervention period. In the control group, the mean concentration of 6-sulfatoxymelatonin changed from 55.5 (95% CI: 48.7,63.2) ng/mg creatinine to 50.8 (95% CI: 44.0,58.7) ng/mg creatinine across the intervention period. A comparison of the two groups with regard to the changes in the 6-sulfatoxymelatonin concentrations across the intervention period showed a significant difference (P = 0.03). The results indicate that increased consumption of vegetables raises circulatory melatonin concentrations. [source]

ABSTRACT: Several chlorinated solvent plumes threaten the sole-source aquifer underlying the Massachusetts Military Reservation at the western end of Cape Cod. Sensitive surface water features including ponds, cranberry bogs, and coastal wetlands are hydraulically connected to the aquifer. For one of the plumes (CS-10 the original remedy of 120 extraction and reinjection wells has the potential for significant disruption of surface water hydrology, through the localized drawdown and mounding of the water table. Recirculating wells with in-well air stripping offer a cost-effective alternative to conventional pump-and-treat technology that does not adversely affect the configuration of the water table. Pilot testing of a two well system, pumping 300 gpm, showed a capture radius of > 200 feet per well, in-well trichloroethylene (TCE) removal efficiencies of 92 to 98 percent per recirculation cycle, an average of three recirculation cycles within the capture zone, and no measurable effect on water table elevations at any point within the recirculation/treatment zone. During 120 days of operation, the mean concentration of TCE in the treatment zone was reduced by 83 percent, from 1,111 ,g/l to 184 ,g/l. Full-scale design projections indicate that 60 wells at an average spacing of 160 feet, having an aggregate recirculation 11 MGD, can contain the CS-b plume without ground water extraction or adverse hydraulic effects on surface water resources. The estimated capital costs for such a system are about $7 million, and annual operations-and-maintenance costs should be about $1.4 million, 40 percent of those associated with a pump and treat system over a 20-year period. Sensitive enzyme-linked immunosorbent assay and rapid one-step immunochromatographic strip for fumonisin B1 in grain-based food and feed samples

Abstract BACKGROUND: Maize contaminated with mycotoxin fumonisin B1 poses a global threat to agricultural production. In this study, polyclonal antibodies (pAb) specific to fumonisin B1 were generated from rabbits immunised with fumonisin B1,keyhole limpet haemocyanin (KLH). These antibodies were used to establish a sensitive competitive direct enzyme-linked immunosorbent assay (cdELISA) and gold nanoparticle immunochromatographic strip for detecting fumonisin B1 levels in maize-based foods and feeds. RESULTS: In cdELISA, fumonisins B1, B2 and B3 at concentrations of 0.42, 0.58 and 81.5 ng mL,1 respectively caused 50% inhibition (IC50) of binding of fumonisin B1,horseradish peroxidase (HRP) to the antibodies. Effective on-site detection of fumonisin B1 was achieved by developing a rapid and sensitive pAb-based gold nanoparticle immunochromatographic strip. This strip had a detection limit of 5 ng mL,1 for fumonisin B1 in maize-based samples. Additionally, the whole analytical process could be completed within 10 min. Close examination of 15 maize-based samples by cdELISA revealed that 11 were fumonisin-positive, with a mean concentration of 435 ± 20.1 ng g,1. These results correlated well with those obtained by immunochromatographic strip. CONCLUSION: Both cdELISA and immunochromatographic strip methods established in this study are sensitive for rapid detection of fumonisins in agricultural commodities. Pharmacokinetics of ketamine in plasma and milk of mature Holstein cows

Plasma and milk concentrations were determined using a high-performance liquid chromatography assay. Pharmacokinetic parameters were estimated using a noncompartmental method. Following i.v. administration, plasma Tmax was 0.083 h and plasma Cmax was 18 135 ± 22 720 ng/mL. Plasma AUC was 4484 ± 1,398 ng·h/mL. Plasma t˝, was 1.80 ± 0.50 h and mean residence time was 0.794 ± 0.318 h with total body clearance of 1.29 ± 0.70 L/h/kg. The mean plasma steady-state volume of distribution was calculated as 0.990 ± 0.530 L/kg and volume of distribution based on area was calculated as 3.23 ± 1.51 L/kg. The last measurable time for ketamine detection in plasma was 8.0 h with a mean concentration of 24.9 ± 11.8 ng/mL. Milk Tmax was detected at 0.67 ± 0.26 h with Cmax of 2495 ± 904 ng/mL. Milk AUC till the last time was 6593 ± 2617 ng·h/mL with mean AUC milk to AUC plasma ratio of 1.99 ± 2.15. The last measurable time that ketamine was detected in milk was 44 ± 10.0 h with a mean concentration of 16.0 ± 9.0 ng/mL. [source]


Characterization of the pharmacokinetic disposition of levofloxacin in stallions after intravenous and intramuscular administration

The target of the present study was to investigate the plasma disposition kinetics of levofloxacin in stallions (n = 6) following a single intravenous (i.v.) bolus or intramuscular (i.m.) injection at a dose rate of 4 mg/kg bwt, using a two-phase crossover design with 15 days as an interval period. Plasma samples were collected at appropriate times during a 48-h administration interval, and were analyzed using a microbiological assay method. The plasma levofloxacin disposition was best fitted to a two-compartment open model after i.v. dosing. The half-lives of distribution and elimination were 0.21 ± 0.13 and 2.58 ± 0.51 h, respectively. The volume of distribution at steady-state was 0.81 ± 0.26 L/kg, the total body clearance (Cltot) was 0.21 ± 0.18 L/h/kg, and the areas under the concentration,time curves (AUCs) were 18.79 ± 4.57 ,g.h/mL. Following i.m. administration, the mean t1/2el and AUC values were 2.94 ± 0.78 h and 17.21 ± 4.36 ,g.h/mL. The bioavailability was high (91.76% ± 12.68%), with a peak plasma mean concentration (Cmax) of 2.85 ± 0.89 ,g/mL attained at 1.56 ± 0.71 h (Tmax). The in vitro protein binding percentage was 27.84%. Calculation of efficacy predictors showed that levofloxacin might have a good therapeutic profile against Gram-negative and Gram-positive bacteria, with an MIC , 0.1 ,g/mL. [source]


Clinical efficacy of intravenous administration of marbofloxacin in a Staphylococcus aureus infection in tissue cages in ponies

Tissue cages (TC), implanted subcutaneously in the neck in eight ponies, were inoculated with Staphylococcus aureus (S. aureus) to determine the clinical efficacy of marbofloxacin in the treatment of this infection. From 21 h after inoculation, marbofloxacin (6 mg/kg) was administered intravenously (i.v.) once daily for 7 days. Samples of the tissue cage fluid (TCF) were taken to determine marbofloxacin concentrations (days 1, 3 and 7), using high-pressure liquid chromatography, and numbers of viable bacteria [colony forming units (CFU)] (days 1, 3, 7, 14 and 21). Statistical analysis was used to compare CFU before and after treatment. Clinical signs and CFU were used to evaluate the efficacy of treatment. Although, there was a slight decrease in CFU in all TC initially, the infection was not eliminated by marbofloxacin treatment in any of the ponies and abscesses formed. As the MIC (0.25 ,g/mL) did not change during treatment and the concentration of marbofloxacin during treatment (mean concentration in TCF was 0.89 ,g/mL on day 1, 0.80 ,g/mL on day 3 and 2.77 ,g/mL on day 7) was above MIC, we consider that the treatment failure might be attributable to the formation of a biofilm by S. aureus. Based on the present results, i.v. administration of marbofloxacin alone is not suitable for the elimination of S. aureus infections from secluded sites. [source]


Clenbuterol in the horse: urinary concentrations determined by ELISA and GC/MS after clinical doses

Clenbuterol is a ,2 agonist/antagonist bronchodilator marketed as Ventipulmin® and is the only member of this group of drugs approved by the US Food and Drug Administration (FDA) for use in horses. Clenbuterol is a class 3 drug in the Association of Racing Commissioners International (ARCI) classification system; therefore, its identification in postrace samples may lead to sanctions. Recently, the sensitivity of postrace testing for clenbuterol has been substantially increased. The objective of this study was to determine the ,detection times' for clenbuterol after administration of an oral clinical dose (0.8 g/kg, b.i.d.) of Ventipulmin syrup. Five horses received oral clenbuterol (0.8 g/kg, b.i.d.) for 10 days, and urine concentrations of clenbuterol were determined by an enhanced enzyme-linked immunoabsorbent assay (ELISA) test and gas chromatography/mass spectrometric (GC/MS) analysis by two different methods for 30 days after administration. Twenty-four hours after the last administration, urine concentrations of apparent clenbuterol, as measured by ELISA, averaged about 500 ng/mL, dropping to about 1 ng/mL by day 5 posttreatment. However, there was a later transient increase in the mean concentrations of apparent clenbuterol in urine, peaking at 7 ng/mL on day 10 postadministration. The urine samples were also analysed using mass spectral quantification of both the trimethylsilyl (TMS) and methane boronic acid (MBA) derivatives of clenbuterol. Analysis using the TMS method showed that, at 24 h after the last administration, the mean concentration of recovered clenbuterol was about 22 ng/mL. Thereafter, clenbuterol concentrations fell below the limit of detection of the TMS-method by day 5 after administration but became transiently detectable again at day 10, with a mean concentration of about 1 ng/mL. Derivatization with MBA offers significant advantages over TMS for the mass spectral detection of clenbuterol, primarily because MBA derivatization yields a high molecular weight base peak of 243 m/z, which is ideal for quantitative purposes. Therefore, mass spectral analyses of selected urine samples, including the transient peak on day 10, were repeated using MBA derivatization, and comparable results were obtained. The results show that clenbuterol was undetectable in horse urine by day 5 after administration. However, an unexpected secondary peak of clenbuterol was observed at day 10 after administration that averaged ,1 ng/mL. Because of this secondary peak, the detection time for clenbuterol (0.8 g/kg, b.i.d. × 10 days) is at least 11 days if the threshold for detection is set at 1 ng/mL. [source]


Association of polymorphisms within the transforming growth factor-,1 gene with diabetic nephropathy and serum cholesterol and triglyceride concentrations

ABSTRACT Aim: The TGF-, gene participates in the development of chronic kidney disease. We investigated whether the 869 T > C, 915 G > C and ,800 G > A polymorphisms of TGF-,1 are associated with diabetic nephropathy (DN). Methods: Polymorphisms were genotyped in 439 type 2 diabetes mellitus patients, 233 with diabetic nephropathy (DN+) and 206 without (DN,). The sample was characterized for relevant clinical and biochemical parameters. Results: The 869 T > C (P = 0.016; odds ratio (OR) = 1.818, 95% confidence interval (CI) = 1.128,2.930) and the 915 G > C polymorphisms (P = 0.008, OR = 4.073, 95% CI = 1.355,12.249) were associated with diabetic nephropathy. The 869 T > C variant was associated with total cholesterol levels: CC + CT genotypes had a mean cholesterol concentration of 5.62 ± 1.40 mmol/L vs a mean concentration of 5.15 ± 1.40 mmol/L for the TT genotype (P = 0.011). Triglycerides were also higher in CC + CT genotypes (2.49 ± 1.56 mmol/L) in comparison with TT homozygotes (2.1 ± 1.22 mmol/L, P = 0.042). Multivariate logistic regression showed that the polymorphisms 869 T > C and 915 G > C were independent predictors for DN (P = 0.049 and 0.046, respectively). Conclusion: The 869 T > C and 915 G > C polymorphisms within the TGF-,1 gene were associated with DN+. Lower cholesterol and triglycerides levels were observed in TT homozygotes for the 869 T > C polymorphism. The TGF-,1 869 T allele seems to confer protection against DN+. [source]


Residues of zeta-cypermethrin in bovine tissues and milk following pour-on and spray application

Abstract The depletion of zeta-cypermethrin residues in bovine tissues and milk was studied. Beef cattle were treated three times at 3-week intervals with 1,ml 10,kg,1 body weight of a 25,g,litre,1 or 50,g,litre,1 pour-on formulation (2.5 and 5.0,mg zeta-cypermethrin kg,1 body weight) or 100,mg,kg,1 spray to simulate a likely worst-case treatment regime. Friesian and Jersey dairy cows were treated once with 2.5,mg zeta-cypermethrin,kg,1 in a pour-on formulation. Muscle, liver and kidney residue concentrations were generally less than the limit of detection (LOD,=, 0.01,mg,kg,1). Residues in renal-fat and back-fat samples from animals treated with 2.5,mg,kg,1 all exceeded the limit of quantitation (LOQ,=, 0.05,mg,kg,1), peaking at 10 days after treatment. Only two of five kidney fat samples were above the LOQ after 34 days, but none of the back-fat samples exceeded the LOQ at 28 days after treatment. Following spray treatments, fat residues were detectable in some animals but were below the LOQ at all sampling intervals. Zeta-cypermethrin was quantifiable (LOQ,=, 0.01,mg,kg,1) in only one whole-milk sample from the Friesian cows (0.015,mg,kg,1, 2 days after treatment). In whole milk from Jersey cows, the mean concentration of zeta-cypermethrin peaked 1 day after treatment, at 0.015,mg,kg,1, and the highest individual sample concentration was 0.025,mg,kg,1 at 3 days after treatment. Residues in milk were not quantifiable beginning 4 days after treatment. The mean concentrations of zeta-cypermethrin in milk fat from Friesian and Jersey cows peaked two days after treatment at 0.197,mg,kg,1 and 0.377,mg,kg,1, respectively, and the highest individual sample concentrations were 2 days after treatment at 0.47,mg,kg,1 and 0.98,mg,kg,1, respectively. © 2001 Society of Chemical Industry [source]


Low-dose broad-band UVA in morphea using a new method for evaluation

Until recently, various therapies for localized scleroderma have been used with limited success. Recently, phototherapy, with or without psoralen, was proposed as a successful treatment modality. The aim of this study was to evaluate the effect of broad-band low-dose ultraviolet A (UVA) phototherapy in patients with localized scleroderma, using a new method for evaluation. Twelve patients complaining of morphea were exposed to UVA irradiation at a dose of 20 J/cm2 3 times per week for 20 sessions. Selected covered plaques served as internal controls. The efficacy of therapy was judged clinically by sequential inspection and palpation. In biopsy specimens from exposed and covered plaques stained with hematoxylin and eosin (H & E) and Masson trichrome stains, the concentration of collagen per dermal surface area was measured with the use of a computerized image analyzer. All patients reported remarkable softening of skin lesions, confirmed by sequential palpatory assessment. A significant reduction in the mean concentration of collagen per surface area was detected in the plaques exposed to UVA (the P value being 0.007, P<0.01), whereas in the covered plaques the difference was not statistically significant (the P value being 0.10, P>0.05). The conclusion is that low-dose broad-band UVA phototherapy is a very effective and safe treatment modality for localized scleroderma. [source]


Differential Antibody Recognition of Four Allelic Variants of the Merozoite Surface Protein-2 (MSP-2) of Plasmodium falciparum

ABSTRACT. The merozoite surface protein-2 (MSP-2) is a major vaccine candidate for the asexual blood stage of Plasmodium falciparum. MSP-2 is essentially dimorphic, and allelic families are named after the representative isolates FC27 and IC1. The polymorphic central region contains immunodominant repeats, which vary in number, length, and sequence within and between allelic families. We have examined the antibody recognition of repeat regions from both MSP-2 allelic families expressed as recombinant fusion peptides. The results are summarized as follows. (1) Immunization of mice with the fusion peptides elicited IgG antibodies that cross-reacted with the native MSP-2 molecule in an allelic family-specific manner. (2) These mouse antibodies recognized the recombinant proteins in both a variant-specific and a family-specific manner, as shown in inhibition immunoassays. Antibodies raised against the peptide FC27 seemed to be essentially variant-specific, since the soluble form of the S20 antigen (a member of FC27 family) had relatively little inhibitory effect on them. (3) The overall pattern of human IgG antibody responses to MSP-2 in Karitiana Indians, a population continuously exposed to hypoendemic malaria in the Brazilian Amazon Region, differs from that described in hyperendemic areas in Africa and Papua New Guinea in two important features: there was no clear age-dependent increase in the prevalence and mean concentration of specific IgG antibodies, and there was no skewing towards the IgG, subclass in antibody responses. (4) The relatively poor correlation between concentrations of IgG antibodies that are specific for members of the same allelic family suggests that recognition of MSP-2 peptides by naturally acquired antibodies was largely variant-specific in this population. The potential role of naturally acquired variant-specific antibodies in immune evasion, by selecting mutant parasites carrying insertions or deletions of repeat sequences, is briefly discussed.
