Selected AbstractsNeisseria meningitidis serogroup B: laboratory correlates of protectionFEMS IMMUNOLOGY & MEDICAL MICROBIOLOGY, Issue 2 2002Clementien Vermont Abstract Meningococcal disease in the Western countries is frequently caused by Neisseria meningitidis serogroup B. Major efforts have been made to develop a safe and efficacious vaccine against this serogroup which is suitable for use in infants and young children. To assess the quality of the immune response after vaccination with candidate vaccines, laboratory correlates of protection are needed. For serogroups A and C, serum bactericidal activity (SBA) is a well established predictor for protection, but for serogroup B other mechanisms besides SBA may also be involved in conferring protection from disease. Several laboratory methods for identification and evaluation of the immunogenicity of possible vaccine antigens are described in this review. [source] Development of a historical ice database for the study of climate change in CanadaHYDROLOGICAL PROCESSES, Issue 18 2002Frédéric Lenormand Abstract The Canadian government has been compiling various observations on freshwater and coastal sea ice conditions for many years. However, the records are not easily accessible and are dispersed within different government departments. Given this, a major effort was undertaken in order to gather all available observations into a common database,the Canadian Ice Database (CID). This database will respond to the needs for climate monitoring in Canada, the validation and improvement of numerical ice models and the development of new remote-sensing methods. Indeed, several studies have shown that freshwater ice and sea ice are good proxy indicators of climate variability and change. The first version of CID contains in situ observations from 757 sites distributed across Canada, which were originally kept on digital or paper records at the Meteorological Service of Canada Headquarters and the Canadian Ice Service (CIS). The CID holds 63 546 records covering the period from ice season 1822,23 to 2000,01. An analysis of the database allows one to trace the temporal evolution of the ice networks. The freeze-up/break-up network of 2000,01 only represents 4% of what it was in 1985,86. A drastic decline of the ice thickness and the snow on ice network is also observable. In 1997,98, it represented only 10% of the network that existed in 1984,85. The major budget cuts in Canadian government agencies during the late 1980s and the 1990s offer the most plausible explanation for the drastic decline in the ice observation networks. Weekly ice coverage determination on large lakes from satellite imagery by the CIS and the national volunteer ice monitoring program, IceWatch, may provide a means of reviving, at least, the freeze-up/break-up network. [source] Irrigation and drainage systems research and development in the 21st century,IRRIGATION AND DRAINAGE, Issue 4 2002Bart Schultz irrigation; drainage; développement durable; système de réseau Abstract One critical problem confronting mankind today is how to manage the intensifying competition for water between expanding urban centres, traditional agricultural activities and in-stream water uses dictated by environmental concerns. In the agricultural sector, the dwindling number of economically attractive sites for large-scale irrigation and drainage projects limits the prospects of increasing the gross cultivated area. Therefore, the required increase in agricultural production will necessarily rely largely on a more accurate estimation of crop water requirements on the one hand, and on major improvements in the construction, operation, management and performance of existing irrigation and drainage systems, on the other. The failings of present systems and the inability to sustainably exploit surface and groundwater resources can be attributed essentially to poor planning, design, system management and development. This is partly due to the inability of engineers, planners and managers to adequately quantify the effects of irrigation and drainage projects on water resources and to use these effects as guidelines for improving technology, design and management. To take full advantage of investments in agriculture, a major effort is required to modernize irrigation and drainage systems and to further develop appropriate management strategies compatible with the financial and socio-economic trends, and the environment. This calls for a holistic approach to irrigation and drainage management and monitoring so as to increase food production, conserve water, prevent soil salinization and waterlogging, and to protect the environment. All this requires, among others, enhanced research and a variety of tools such as water control and regulation equipment, remote sensing, geographic information systems, decision support systems and models, as well as field survey and evaluation techniques. To tackle this challenge, we need to focus on the following issues: affordability with respect to the application of new technologies; procedures for integrated planning and management of irrigation and drainage systems; analysis to identify causes and effects constraining irrigation and drainage system performance; evapotranspiration and related calculation methods; estimation of crop water requirements; technologies for the design, construction and modernization of irrigation and drainage systems; strategies to improve irrigation and drainage system efficiency; environmental impacts of irrigation and drainage and measures for creating and maintaining sustainability; institutional strengthening, proper financial assessment, capacity building, training and education. Résumé Aujourd'hui le problème critique pour l'humanité est comment manier la compétition intensifiante pour de l'eau entre les centres urbains en expansion, pour des activités traditionnellement agricoles et pour l'usage de l'eau fluviale prescrit par des conditions écologistes. Dans le secteur agricole les perspectives d'agrandir les champs cultivés bruts sont limitées par le nombre diminuant des terrains économiquement attractifs pour des projets d'irrigation et du drainage de grande envergure. Par conséquent l'augmentation nécessaire de la production agricole comptera surtout sur une évaluation plus précise du besoin des plantes d'un côté, et de l'autre sur de grandes améliorations dans la construction, dans l'opération, dans le management et dans la performance des systèmes d'irrigation et du drainage. On peut attribuer les défauts des systèmes actuels et l'incompétence d'exploiter durablement les ressources hydriques de surface et souterraines au planification, au système de la gestion de l'eau et au système du développement. Cela est partiellement dû à l'incapacité des ingénieurs, des planificateurs et des gérants, de quantifier adéquatement les effets des projets d'irrigation et de drainage sur les ressources hydriques et d'utiliser ces résultats pour améliorer la technologie, la planification et la gestion de l'eau. Pour profiter le mieux possible des investissements dans l'agriculture, on exige un effort considérable pour moderniser les systèmes d'irrigation et de drainage et pour développer des stratégies de gestion de l'eau qui doivent être appropriées et compatibles avec les tendances financières et socio-économiques et avec l'environnement. Ceci a besoin d'une procédure holistique pour la gestion et le monitorage de l'eau, pour augmenter la production d'aliments, pour conserver l'eau, pour prévenir la salination du sol et pour protéger l'environnement. Tout cela demande, entre autres choses, une recherche d'avant-garde et une variété d'instruments comme les contrôles du régime hydrique et les appareils de régulation, la télédétection, les systèmes de l'information géographique, les systèmes et les modèles de support de décision et de même les levés sur le terrain et les techniques d'évaluation. Jack Jr. MD Abstract The Alzheimer's Disease Neuroimaging Initiative (ADNI) is a longitudinal multisite observational study of healthy elders, mild cognitive impairment (MCI), and Alzheimer's disease. Magnetic resonance imaging (MRI), (18F)-fluorodeoxyglucose positron emission tomography (FDG PET), urine serum, and cerebrospinal fluid (CSF) biomarkers, as well as clinical/psychometric assessments are acquiredat multiple time points. All data will be cross-linked and made available to the general scientific community. The purpose of this report is to describe the MRI methods employed in ADNI. The ADNI MRI core established specifications thatguided protocol development. A major effort was devoted toevaluating 3D T1 -weighted sequences for morphometric analyses. Several options for this sequence were optimized for the relevant manufacturer platforms and then compared in a reduced-scale clinical trial. The protocol selected for the ADNI study includes: back-to-back 3D magnetization prepared rapid gradient echo (MP-RAGE) scans; B1 -calibration scans when applicable; and an axial proton density-T2 dual contrast (i.e., echo) fast spin echo/turbo spin echo (FSE/TSE) for pathology detection. ADNI MRI methods seek to maximize scientific utility while minimizing the burden placed on participants. The approach taken in ADNI to standardization across sites and platforms of the MRI protocol, postacquisition corrections, and phantom-based monitoring of all scanners could be used as a model for other multisite trials. [source] The impact of food hypersensitivity reported in 9-year-old children by their parents on health-related quality of lifeALLERGY, Issue 2 2008E. Östblom Background:, There are only a few studies on the impact of food hypersensitivity (FHS) in children on health-related quality of life (HRQL). The present study was designed to examine this impact in a population-based birth cohort (BAMSE). Methods:, A nested case,control study was performed within the cohort. The parents of 1378 nine-year-old children filled out a generic questionnaire with 13 subscales (Child Health Questionnaire Parental Form 28 , CHQ-PF28) supplemented with disease-specific questions concerning FHS. There were 212 children with report of FHS. Another 221 children with allergic diseases but not FHS were examined for comparison. Furthermore, the impact of pronounced symptoms of FHS and of increasing levels of food-specific IgE antibodies on HRQL was also analysed. Results:, The children with FHS exhibited significantly lower scores on the subscales physical functioning, role/social limitations , physical and general health in the generic instrument. Furthermore, children with food-related symptoms from the lower airways were scored lower on Self Esteem, Parental Impact , time and Family Cohesion. Sensitization per se did not alter these patterns, but high levels of food-specific IgE-antibodies affected mental health and general health negatively. A physician's diagnosis of food allergy did not affect any of the subscales negatively. Conclusions:, Parents reported that FHS exerts a negative impact on the HRQL of 9-year-old children, in particular in children with symptoms from the lower airways or if the FHS is associated with high levels of food-specific IgE-antibodies. Healthcare-givers must put major effort into improving and maintaining the HRQL of these children. [source] Interspecies comparison of prostate cancer gene-expression profiles reveals genes associated with aggressive tumorsTHE PROSTATE, Issue 10 2009Itai Kela Abstract Prostate cancer (PC) is a heterogeneous disease whose aggressive phenotype is the second leading cause of cancer-related death in men. The identification of key molecules and pathways that play a pivotal role in PC progression towards an aggressive form is crucial. A major effort towards this end has been taken by global analyses of gene expression profiles. However, the large body of data did not provide a definitive idea about the genes which are associated with the aggressive growth of PC. In order to identify such genes, we performed an interspecies comparison between several human data sets and high quality microarray data that we generated from the transgenic adenocarcinoma of mouse prostate (TRAMP) strain. The TRAMP PC mimics the histological and pathological appearance as well as the aggressive phenotype of human PC (huPC). Analysis of the microarray data, derived from microdissected TRAMP specimens removed at different stages of the disease yielded genetic signatures delineating the TRAMP PC development and progression. Comparison of the TRAMP data with a set of genes representing the core expression signature of huPC yielded a limited set genes. Some of these genes are known predictors of poor prognosis in huPC. Interestingly, the modulation of genes responsible for the invasive phenotype of huPC occurs in TRAMP already during the transition to prostate intraepithelial neoplasia (PIN) and onwards to localized tumors. [source] Combining experimental data for structure determination of flexible multimeric macromolecules by molecular replacementACTA CRYSTALLOGRAPHICA SECTION D, Issue 5 2006Stefano Trapani A major effort has been made by the structural biology community to develop user-friendly software for the use of biologists. However, structural projects become more and more challenging and their solution often relies on a combination of information from various sources. Here, it is described how X-ray data, normal-mode analysis (NMA) and electron-microscopy (EM) data can be successfully combined in order to obtain a molecular-replacement (MR) solution for crystal structures containing multimeric molecules. NMA is used to simulate computationally the inherent internal flexibility of the monomer and thus enhance, together with the crystal noncrystallographic symmetry (NCS), the MR capabilities. NCS is also used to obtain a reliable EM reconstruction, which is then employed as a filter to construct oligomers starting from monomers. The feasibility of the direct use of EM reconstructions as a template for MR when the X-ray and EM data resolutions overlap is also discussed. On 25 April 1998, the mineral waste retaining wall at the Swedish-owned pyrite mine at Aznalcóllar (Seville, Spain) burst,2 causing the most harmful environmental and socio-economic disaster in the history of the River Guadiamar basin. The damage was so great that the regional government decided in May 1998 to finance a comprehensive, multidisciplinary research initiative with the objective of eradicating or at least minimising all of the negative social, economic and environmental impacts. This paper utilises a Strengths, Weaknesses, Opportunities and Threats (SWOT) analysis to identify eight strategic measures aimed at providing policymakers with key guidelines on implementing a sustainable development model, in a broad sense. Empirical evidence, though, reveals that, to date, major efforts to tackle the negative impacts have centred on environmental concerns and that the socio-economic consequences have not been completely mitigated. [source] Resurgent Metropolis: Economy, Society and Urbanization in an Interconnected WorldINTERNATIONAL JOURNAL OF URBAN AND REGIONAL RESEARCH, Issue 3 2008ALLEN J. SCOTT Abstract An urban problematic is identified by reference to the essential characteristics of cities as spatially polarized ensembles of human activity marked by high levels of internal symbiosis. The roots of the crisis of the classical industrial metropolis of the twentieth century are pinpointed, and the emergence of a new kind of urban economic dynamic over the 1980s and 1990s is discussed. I argue that this new dynamic is based in high degree upon the growth and spread of cognitive-cultural production systems. Along with these developments have come radical transformations of urban space and social life, as well as major efforts on the part of many cities to assert a role for themselves as national and international cultural centers. This argument is the basis of what we might call the resurgent metropolis hypothesis. The effects of globalization are shown to play a critical role in the genesis and geography of urban resurgence. Three major policy dilemmas of resurgent cities are highlighted, namely, their internal institutional fragmentation, their increasing character as economic agents on the world stage and the concomitant importance of collective approaches to the construction of localized competitive advantage, and their deepening social disintegration and segmentation. Résumé Une problématique urbaine est dégagée à propos des caractéristiques essentielles des villes définies comme des ensembles d'activité humaine polarisés dans l'espace et marqués par une symbiose interne poussée. Les racines de la crise qu'a subie la métropole industrielle classique au xxe siècle sont mises en évidence. Est aussi étudié un nouveau type de dynamique économique urbaine apparu au cours des années 1980-1990, cette dynamique étant largement fondée sur la croissance et la diffusion des systèmes de production cognitifs culturels. Lederman Helping students develop informed views of nature of science (NOS) has been and continues to be a central goal for kindergarten through Grade 12 (K,12) science education. Since the early 1960s, major efforts have been undertaken to enhance K,12 students and science teachers' NOS views. However, the crucial component of assessing learners' NOS views remains an issue in research on NOS. This article aims to (a) trace the development of a new open-ended instrument, the Views of Nature of Science Questionnaire (VNOS), which in conjunction with individual interviews aims to provide meaningful assessments of learners' NOS views; (b) outline the NOS framework that underlies the development of the VNOS; (c) present evidence regarding the validity of the VNOS; (d) elucidate the use of the VNOS and associated interviews, and the range of NOS aspects that it aims to assess; and (e) discuss the usefulness of rich descriptive NOS profiles that the VNOS provides in research related to teaching and learning about NOS. The VNOS comes in response to some calls within the science education community to go back to developing standardized forced-choice paper and pencil NOS assessment instruments designed for mass administrations to large samples. We believe that these calls ignore much of what was learned from research on teaching and learning about NOS over the past 30 years. Muller Background:, Despite major efforts, factors that predict or correspond to the level of allergic symptoms remain elusive. Given our previous observations of mucosal interleukin-10 (IL-10) expression by local tissue cells and its described role as immune modulator, we hypothesized that, in allergic rhinitis, nasal mucosal IL-10 expression could influence the severity of symptoms. Methods:, In this study, we investigated endothelial IL-10 expression in nasal mucosa of healthy- and house dust mite allergic patients, both before and after provocation, and under nasal steroid therapy. Nasal turbinate biopsies were taken from healthy individuals as well as from house dust mite allergic patients, both before and after provocation. Allergic patients received fluticasone proprionate aqueous nasal spray or control treatment. In the allergic patients, endothelial IL-10 scores based on immunohistochemical stainings were correlated with allergic symptoms, measured by visual analog scores. Results:, At baseline, variable levels of endothelial IL-10 were detected in nasal biopsies. After nasal provocation, but not at baseline, endothelial IL-10 expression corresponded very closely to the allergic symptoms after allergen provocation. Low symptom scores were correlated with high endothelial IL-10 scores. This correlation disappeared after fluticason propionate treatment. Conclusions:, There is a large variation in the level of endothelial IL-10 expression both in healthy individuals and in house dust mite allergic patients. Endothelial IL-10 expression may affect local immune reactions resulting in reduced levels of allergic symptoms. Durant Neoinstitutionalists applying the logic of rational choice institutionalism have leavened our understanding of public agency design and evolution in the domestic and national security policy domains. This paper seeks to advance theory building in empirically grounded ways by assessing the explanatory power of an important theoretical perspective (rational choice institutionalism), in an understudied "hybrid" policy domain where domestic and national security aims interact (domestic environmental policy and national security policy), and in an organizational type (the U.S. military) that has drawn scant attention from students of bureaucracy in political science, public administration, or public management. Analysis of three major efforts to green the U.S. military suggests that the patterns of politics accompanying agency evolution involving hybrid policy domains differ from domestic and national security domains in ways that limit the generalizability of rational choice institutionalism.
