Dysregulated Cytokine Metabolism, Altered Hepatic Methionine Metabolism and Proteasome Dysfunction in Alcoholic Liver Disease

Abstract: Alcoholic liver disease (ALD) remains an important complication and cause of morbidity and mortality from alcohol abuse. Major developments in our understanding of the mechanisms of ALD over the past decade are now being translated into new forms of therapy for this disease process which currently has no FDA approved treatment. Cytokines are low molecular weight mediators of cellular communication, and the pro-inflammatory cytokine tumor necrosis factor (TNF) has been shown to play a pivotal role in the development of experimental ALD. Similarly, TNF levels are elevated in the serum of alcoholic hepatitis patients. Abnormal methionine metabolism is well documented in patients with ALD, with patients having elevated serum methionine levels, but low S-adenosylmethionine levels in the liver. On the other hand, S-adenosylhomocysteine and homocysteine levels are elevated in ALD. Recent studies have documented potential interactions between homocysteine and S-adenosylhomocysteine with TNF in the development of ALD. Altered proteasome function also is now well documented in ALD, and decreased proteasome function can cause hepatocyte apoptosis. Recently it has been shown that decreased proteasome function can also act synergistically to enhance TNF hepatotoxicity. Hepatocytes dying of proteasome dysfunction release pro-inflammatory cytokines such as Interleukin-8 to cause sustained inflammation. This article reviews the interactions of cytokines, altered methionine metabolism, and proteasome dysfunction in the development of ALD. [source]

True-blue Ir(III) phosphors are designed, synthesized, and applied to multilayered organic true-blue-light-emitting diodes with CIEx,y color chromaticity of (0.15,0.11) and maximum external quantum efficiency of ,12%, demonstrating unprecedented performance among all blue-phosphorescent OLEDs ever documented. The molecular-design strategy and subsequent device-fabrication protocol reveal a major development in OLEDs. [source]


Modelling lifetime QALYs and health care costs from different drinking patterns over time: a Markov model

Abstract The negative health consequences of alcohol use and its treatment account for significant health care expenditure worldwide. Long-term modelling techniques are developed in this paper to establish a link between drinking patterns, health consequences and alcohol treatment effectiveness and cost-effectiveness. The overall change in health related quality and quantity of life which results from changes in health-related behaviour is estimated. Specifically, a probabilistic lifetime Markov model is presented where alcohol consumption in grams of alcohol per day and drinking history are used for the categorization of patients into four Markov states. Utility weights are assigned to each drinking state using EQ-5D scores. Mortality and morbidity estimates are state, gender and age specific, and are alcohol-related and non-alcohol-related. The methodology is tested in a case study. This represents a major development in the techniques traditionally used in alcohol economic models, in which short-term costs and outcomes are assessed, omitting potential longer term cost savings and improvements in health related quality of life. Real-time PCR with internal amplification control for detecting tuberculosis: method design and validation

APMIS 2009; 117: 592,7. Real-time PCR has been a major development in the diagnosis of tuberculosis. However, most tests do not include an internal amplification control (IAC), which therefore limits it clinical application. In this study a new, easy to perform real-time PCR test with IAC was designed and validated in clinical samples. The primers amplified a 163-bp fragment of IS6110 of Mycobacterium tuberculosis and the IAC was designed with a fragment of a different microorganism (Chlamydia trachomatis). The interassay and intraassay variation of this test were very low (0.45,1.65% and 0.18,1.80%, respectively). The detection accuracy was validated in 50 samples (25 urine, 25 sputum) with different concentrations of M. tuberculosis, 18 clinical isolates of non-tuberculous mycobacteria and 148 samples with clinical suspicion of pulmonary tuberculosis. The specificity was 100%. The detection limit of this PCR test without IAC was approximately 15 bacteria and with IAC approximately 32 bacteria. This real-time PCR with IAC assay can improve the detection of M. tuberculosis and contribute to standardization of this diagnostic technique. [source]

Prolegomenon to a history of paleoanthropology: The study of human origins as a scientific enterprise.

Interest in the history of paleoanthropology and the other disciplines related to human origins studies has grown considerably over the last several decades. Some very informative historical surveys have been written by prominent scientists reflecting on the major developments in their fields. Some well-known early examples include Glyn Daniel's The Idea of Prehistory (1962) and The Origins and Growth of Archaeology (1967), which focus primarily on the history of archaeology, Kenneth Oakley's "The problem of man's antiquity: an historical survey" published in the Bulletin of the British Museum (Natural History) [Geology] (1964), and L. S. B. Leakey's Unveiling Man's Origins; Ten Decades of Thought about Human Evolution (1969), with the latter two focusing on the contributions of geology, paleontology, and biology to the problem of human evolution. The Changing Nexus: Tertiary Education Institutions, the Marketplace and the State

This article examines the evolving relationship between the marketplace, the state and tertiary education institutions. The context of these relations has evolved strikingly in recent years, which have seen three major developments: growing system differentiation, changing governance patterns and diminished direct involvement of governments in the funding and provision of tertiary education. This article first describes the key dimensions of the rise of market forces in tertiary education throughout the world and the main implications of this phenomenon. It then articulates the rationale for continuing public intervention in the sector and, in conclusion, outlines the nature of an appropriate enabling framework for the further development of tertiary education. Large-scale processes in ecology and hydrology

1. Several papers published in the 1980s stressed the importance of scaling issues, the inter-relatedness of patterns and processes at different scales of time and space, to our understanding of ecological systems. Scaling issues are of major theoretical interest and increasingly are of considerable applied importance. 2. In recognition of this, the Natural Environment Research Council, in partnership with the Scottish Executive Rural Affairs Department, funded a Thematic Programme entitled ,Large-scale Processes in Ecology and Hydrology'. The principal aim of this Programme was to integrate recent major developments in information resources and technologies with current theory in order to improve understanding of large-scale patterns and processes and their relationship to patterns and processes at smaller scales. 3. The Thematic Programme, which ran from 1995 until 1999, funded six research projects that have generated a large body of published papers. This volume, dedicated to the findings of the Programme, brings together outputs from all six projects with the aim of ensuring a rapid and widespread dissemination of the Programme's findings. A brief résumé of each of the papers is presented. 4. The papers in this volume cover a wide variety of subjects ranging from ions to the flora and fauna of the United Kingdom. Nonetheless, each study has sought in various ways to quantify observed spatio-temporal patterns at a range of scales, to determine whether those patterns are consistent across scales and to identify the interactions between small-scale patterns and processes and those at larger scales. The importance of the spatial and temporal scales at which studies are conducted, the key role played by dispersal in spatial population dynamics, and the diversity of ways in which large-scale patterns and processes relate to those at smaller scales are highlighted in many of the papers. 4. All of the papers presented here have direct relevance to applied issues. These issues are diverse and include the control of invasive alien species, the conservation of declining, threatened or endangered species, the development of survey techniques, strategies for farmland, woodland and forestry management, and the assessment of pollution sensitivity. Thus, the Thematic Programme has addressed issues of considerable theoretical interest and has at the same time generated results and predictive models that are of considerable practical and policy relevance. Supercritical water for environmental technologies

Abstract OVERVIEW: Supercritical water is a great medium in which to perform chemical reactions and to develop processes. Due to its unique thermo-physico-chemical properties, supercritical water is able to play the role of solvent of organic compounds and/or to react with them. These specific properties have been used since the 1990s to develop new technologies dedicated to the environment and energy. IMPACT: Supercritical water based technologies are innovative and efficient processes having a strong impact on society, the environment and the economy, and is termed a sustainable technology. APPLICATIONS: Three main applications for supercritical water technology are under development: (i) supercritical water oxidation (SCWO); (ii) supercritical water biomass gasification (SCBG); and (iii) hydrolysis of polymers in supercritical water (HPSCW) for composites/plastics recycling. In this paper some fundamentals of supercritical water are first presented to introduce the above three major developments. Then these technologies are reviewed in terms of their present and future industrial development and their impact on the environment and on energy production. The tenth anniversary of Suzuki polycondensation (SPC)

Abstract This article describes the successful transfer of the Suzuki cross-coupling (SCC) reaction to polymer synthesis, one of the major developments within the last decade of polymer synthesis. The polymers prepared by Suzuki polycondensation (SPC) and its Ni-catalyzed reductive counterpart are soluble and processable poly(arylene)s that, because of their rigid and conjugated backbones, are of interest for the materials sciences. Achievable molar masses easily compete with those of traditional polyesters and polyamides. This article also provides insight into some synthetic problems associated with the transfer of SCC from low molar mass organic chemistry to high molar mass polymer chemistry by addressing issues such as monomer purity, stoichiometric balance, achievable molar masses, and defects in the polymer structure. Although the emphasis of this article is synthetic and structural issues, some potential applications of the polyarylenes obtained are briefly mentioned. Together with the enormous developments in the areas of metallocene, ring-opening metathesis, and acyclic diene metathesis polymerization, the success of SPC impressingly underlines the increasing importance of transition-metal-catalyzed CC-bond-forming reactions in polymer synthesis. © 2001 John Wiley & Sons, Inc. Schema Theory and Knowledge-Based Processes in Second Language Reading Comprehension: A Need for Alternative Perspectives

How is knowledge represented and organized in the mind? What role does it play in discourse comprehension and interpretation? What are the exact mechanisms whereby knowledge-based processes are utilised in comprehension? These are questions that have puzzled psycholinguists and cognitive psychologists for years. Despite major developments in the field of second language (L2) reading over the last two decades, many attempts at explaining the role of knowledge in L2 comprehension have been made almost exclusively in the context of schema theory, a perspective that provides an expectation-driven conception of the role of knowledge and considers that preexisting knowledge provides the main guiding context through which information is processed and interpreted. In this article, I first review and critically analyze the major assumptions underlying schema theory and the processes that it postulates underlie knowledge representation and comprehension. Then I consider an alternative perspective, a construction-integration model of discourse comprehension, and discuss how this perspective, when applied to L2 reading comprehension, offers a fundamentally different and more detailed account of the role of knowledge and knowledge-based processes that L2 researchers had previously tried to explain within schema-theoretic principles. ABSTRACT, In the year of Darwin, what were the emerging themes and events that united disparate manifestations of bioanthropological identities? In this review, I draw from conference proceedings, the literature, and electronic social networking to assess six major developments in 2009: the bioanthropological legacy of Darwin on the 200th anniversary of his birth; the efforts of primatologists from a multitude of backgrounds to grapple with the construction of a unified ethics; the remediation of philosophical tensions between field and captive primatology; the coalescence of an explicitly comparative evolutionary anthropology; the role of conference attendance and collaborations in forging disciplinary identity; and the provocative implications of the Ardipithecus ramidus story. The field of biological anthropology continues to evolve and diversify, obscuring a common identity, but as in other organic fractal systems, a common origin as anthropologists leads to descent with modification. Ovarian cancer proteomics: Many technologies one goal

Abstract The last decade has seen major changes in the technologies used to identify markers for diagnosing cancer. This review focuses on recent developments on the evolving field of biomarker discovery, and validation techniques using proteomics platforms for ovarian cancer. It is possible now to diagnose various disease conditions using microliter quantities of body fluids. Currently the major developments were made in three distinct areas: (i) protein profiling, (ii) high-throughput validation techniques, and (iii) solid and liquid phase protein microarray platforms for analyzing candidate markers across subclasses and stages of cancers. The recent addition to the long list of technologies is metabolomics using metabolite profiling and informatics-based filtering of information for biomarker discovery of ovarian cancer. Emerging technologies need to address ways to eliminate the limitations posed by the complex dynamic nature of body fluids as well as ways to enrich low-abundance tumor markers if they were to become a successful biomarker discovery tool. These new technologies hold significant promise in identifying more robust markers for ovarian cancer. Since the prevalence of this disease in the population is low, the test must have a high specificity. [source]

Wildlife Population Assessment: Past Developments and Future Directions

Summary. We review the major developments in wildlife population assessment in the past century. Three major areas are considered: mark-recapture, distance sampling, and harvest models. We speculate on how these fields will develop in the next century. Topics for which we expect to see methodological advances include integration of modeling with Geographic Information Systems, automated survey design algorithms, advances in model-based inference from sample survey data, a common inferential framework for wildlife population assessment methods, improved methods for estimating population trends, the embedding of biological process models into inference, substantially improved models for conservation management, advanced spatiotemporal models of ecosystems, and greater emphasis on incorporating model selection uncertainty into inference. We discuss the kind of developments that might be anticipated in these topics. [source]
