League Champions in Mid Table: On the Major Changes in Dutch Prison Policy

The Netherlands has traditionally been known for having perhaps the mildest prison policy of the Western world in terms of prisoner numbers and prisoner treatment. However, over the last 15 to 20 years a dramatic change has taken place. The number of detention years has steadily increased since 1985; subsequently an extensive prison building scheme was launched, which resulted in more than double the number of prison cells. The landmark change in prison policy is probably completed by the coming into force of the Penitentiary Principles Act 1999 [ Penitentiare Benginselenwet] (Ministry of Justice 1997b), that became law on 1 January 1999. The new legislation re-prioritises Dutch prison policy, by emphasising security and sobriety. This article examines causes and effects of these developments. [source]


The role of caveolin-1 in cardiovascular regulation

Abstract Caveolae are omega-shaped membrane invaginations present in essentially all cell types in the cardiovascular system, and numerous functions have been ascribed to these structures. Caveolae formation depends on caveolins, cholesterol and polymerase I and transcript release factor-Cavin (PTRF-Cavin). The current review summarizes and critically discusses the cardiovascular phenotypes reported in caveolin-1-deficient mice. Major changes in the structure and function of heart, lung and blood vessels have been documented, suggesting that caveolae play a critical role at the interface between blood and surrounding tissue. According to an emerging paradigm, many of these changes are secondary to uncoupling of endothelial nitric oxide synthase. Thus, nitric oxide synthase not only synthesizes more nitric oxide in the absence of caveolin-1, but also more superoxide with potential pathogenic consequences. It is further argued that the vasodilating drive from increased nitric oxide production in caveolin-1-deficient mice is balanced by changes in the vascular media that favour increased dynamic resistance regulation. Harnessing the therapeutic opportunities buried in caveolae, while challenging, could expand the arsenal of treatment options in cancer, lung disease and atherosclerosis. [source]


Growth-induced changes in the proteome of Helicobacter pylori

Abstract Helicobacter pylori is a major human pathogen that is responsible for a number of gastrointestinal infections. We have used 2-DE to characterise protein synthesis in bacteria grown either on solid agar-based media or in each of two broth culture media (Brucella and brain heart infusion (BHI) broth). Significant differences were observed in the proteomes of bacteria grown either on agar-based or in broth media. Major changes in protein abundance were identified using principal component analysis (PCA), which delineated the profiles derived for the three key growth conditions (i.e. agar plates, Brucella and BHI broth). Proteins detected across the gel series were identified by peptide mass mapping and Edman sequencing. A number of proteins associated with protein synthesis in general as well as specific amino acid synthesis were depressed in broth-grown bacteria compared to plate-grown bacteria. A similar reduction was also observed in the abundance of proteins involved in detoxification. Two of the most abundant spots, identified as UreB and GroEL, in plate-grown bacteria showed a >140-fold drop in abundance in bacteria grown in Brucella broth compared to bacteria grown on agar plates. Two protein spots induced in bacteria grown in broth culture were both identified as glyceraldehyde 3-phosphate dehydrogenase based on their N -terminal amino acid sequences derived by Edman degradation. The underlying causes of the changes in the proteins abundance were not clear, but it was likely that a significant proportion of the changes were due to the alkaline pH of the broth culture media. The Diverging Geographies of Social and Business Interaction Patterns: a Case Study of Rural South Australia

Major changes in personal mobility and in country town service provision have taken place in rural South Australia in the period 1968,69 to 1992,93. The later part of this period was one of major rural recession across Australia as a whole. The impact of this recession is considered by investigating changes in three different geographies of rural South Australians. These are their affective identification with place, their local social interaction and their commercial service consumption. There has been a shift of both commercial and social activity up through the hierarchy of settlement, but this is much more pronounced in the case of commercial activity. These differential rates of change are weakening the once mutually reinforcing links between community self-identification, social interaction and trading patterns. The full impact of the rural crisis commencing in 1982,83 on social and commercial spatial patterns is unlikely to have been achieved by 1992,93, and adjustment is likely to continue. [source]


Patterns of ant species richness along elevational gradients in an arid ecosystem

ABSTRACT Aim In this study, we examine patterns of local and regional ant species richness along three elevational gradients in an arid ecosystem. In addition, we test the hypothesis that changes in ant species richness with elevation are related to elevation-dependent changes in climate and available area. Location Spring Mountains, Nevada, U.S.A. Methods We used pitfall traps placed at each 100-m elevational band in three canyons in the Spring Mountains. We compiled climate data from 68 nearby weather stations. We used multiple regression analysis to examine the effects of annual precipitation, average July precipitation, and maximum and minimum July temperature on ant species richness at each elevational band. Results We found that patterns of local ant species richness differed among the three gradients we sampled. Ant species richness increased linearly with elevation along two transects and peaked at mid-elevation along a third transect. This suggests that patterns of species richness based on data from single transects may not generalize to larger spatial scales. Cluster analysis of community similarity revealed a high-elevation species assemblage largely distinct from that of lower elevations. Major changes in the identity of ant species present along elevational gradients tended to coincide with changes in the dominant vegetation. Regional species richness, defined here as the total number of unique species within an elevational band in all three gradients combined, tended to increase with increasing elevation. Available area decreased with increasing elevation. Area was therefore correlated negatively with ant species richness and did not explain elevational patterns of ant species richness in the Spring Mountains. Mean July maximum and minimum temperature, July precipitation and annual precipitation combined to explain 80% of the variation in ant species richness. Main conclusions Our results suggest that in arid ecosystems, species richness for some taxa may be highest at high elevations, where lower temperatures and higher precipitation may support higher levels of primary production and cause lower levels of physiological stress. Brain death and its implications for management of the potential organ donor

The systemic physiologic changes that occur during and after brain death affect all organs suitable for transplantation. Major changes occur in the cardiovascular, pulmonary, endocrine, and immunological systems, and, if untreated may soon result in cardiovascular collapse and somatic death. Understanding these complex physiologic changes is mandatory for developing effective strategies for donor resuscitation and management in such a way that the functional integrity of potentially transplantable organs is maintained. This review elucidates these physiological changes and their consequences, and based on these consequences the rationale behind current medical management of brain-dead organ donors is discussed. Mid-Holocene and glacial-maximum vegetation geography of the northern continents and Africa

Abstract BIOME 6000 is an international project to map vegetation globally at mid-Holocene (6000 14C yr bp) and last glacial maximum (LGM, 18,000 14C yr bp), with a view to evaluating coupled climate-biosphere model results. Primary palaeoecological data are assigned to biomes using an explicit algorithm based on plant functional types. This paper introduces the second Special Feature on BIOME 6000. Site-based global biome maps are shown with data from North America, Eurasia (except South and Southeast Asia) and Africa at both time periods. A map based on surface samples shows the method's skill in reconstructing present-day biomes. Cold and dry conditions at LGM favoured extensive tundra and steppe. These biomes intergraded in northern Eurasia. Northern hemisphere forest biomes were displaced southward. Boreal evergreen forests (taiga) and temperate deciduous forests were fragmented, while European and East Asian steppes were greatly extended. Tropical moist forests (i.e. tropical rain forest and tropical seasonal forest) in Africa were reduced. In south-western North America, desert and steppe were replaced by open conifer woodland, opposite to the general arid trend but consistent with modelled southward displacement of the jet stream. The Arctic forest limit was shifted slighly north at 6000 14C yr bp in some sectors, but not in all. Northern temperate forest zones were generally shifted greater distances north. Warmer winters as well as summers in several regions are required to explain these shifts. Temperate deciduous forests in Europe were greatly extended, into the Mediterranean region as well as to the north. Steppe encroached on forest biomes in interior North America, but not in central Asia. Enhanced monsoons extended forest biomes in China inland and Sahelian vegetation into the Sahara while the African tropical rain forest was also reduced, consistent with a modelled northward shift of the ITCZ and a more seasonal climate in the equatorial zone. Palaeobiome maps show the outcome of separate, independent migrations of plant taxa in response to climate change. The average composition of biomes at LGM was often markedly different from today. Refugia for the temperate deciduous and tropical rain forest biomes may have existed offshore at LGM, but their characteristic taxa also persisted as components of other biomes. Examples include temperate deciduous trees that survived in cool mixed forest in eastern Europe, and tropical evergreen trees that survived in tropical seasonal forest in Africa. The sequence of biome shifts during a glacial-interglacial cycle may help account for some disjunct distributions of plant taxa. For example, the now-arid Saharan mountains may have linked Mediterranean and African tropical montane floras during enhanced monsoon regimes. Major changes in physical land-surface conditions, shown by the palaeobiome data, have implications for the global climate. The data can be used directly to evaluate the output of coupled atmosphere-biosphere models. The data could also be objectively generalized to yield realistic gridded land-surface maps, for use in sensitivity experiments with atmospheric models. Recent analyses of vegetation-climate feedbacks have focused on the hypothesized positive feedback effects of climate-induced vegetation changes in the Sahara/Sahel region and the Arctic during the mid-Holocene. However, a far wider spectrum of interactions potentially exists and could be investigated, using these data, both for 6000 14C yr bp and for the LGM. [source]


The link between household structure and the level of abstraction in the purchase decision process: an analysis using a functional food

Major changes are affecting consumer trends in the agri-food sector. Suppliers are engaged in launching a succession of product innovations and introducing a range of new marketing strategies to promote food health benefits, while consumers are incorporating the new attributes into their choice structures. This study aims to analyze the consumer cognitive structure for a functional food using a means,end chain approach, through the association pattern technique (APT). A further objective is to verify whether consumer cognitive structures vary with household structure (children vs. no children), as reported in the literature on new product acceptance. The results reveal a higher degree of abstraction in the cognitive structure of households with children, incorporating more values in the means,end chains. In line with other research approaches, our study also confirms the stronger confidence-seeking tendency typically associated with the cognitive structure of this type of household. ,-Oxidation capacity in liver increases during parr-smolt transformation of Atlantic salmon fed vegetable oil and fish oil

Atlantic salmon Salmo salar were fed diets containing 100% fish oil (FO; capelin oil) or 100% vegetable oil (VO) from start of feeding until the fish reached the size of 2·5 kg. Samples were taken during the period of the parr-smolt transformation (October 2002 to February 2003). The VO diet consisted of a blend of 55% rapeseed oil, 30% palm oil and 15% linseed oil to maintain the sum of saturated, monounsaturated and polyunsaturated fatty acids between the two diets, although with differences in the individual chain length of fatty acids. Na+/K+ -ATPase activity in the gills, total ,-oxidation capacity in muscles and liver and total lipid, glycogen and dry matter content in the muscles were measured during the parr-smolt transformation and after seawater transfer. Na+/K+ -ATPase activity in gills increased prior to seawater transfer, showing an adaptation for seawater survival. Major changes in the lipid and glycogen content in the fillet and in ,-oxidation capacity were found in the tissues measured. ,-oxidation capacity increased significantly in liver and decreased in red muscle, prior to seawater transfer, giving liver an important role in energy production during this period. Results also indicated that feeding Atlantic salmon a diet where 100% of FO was replaced with VO did not have any negative effects on lipid metabolism during parr-smolt transformation. [source]


Modern distribution of saltmarsh testate amoebae: regional variability of zonation and response to environmental variables

Abstract Sea-level reconstruction from biological indicators in saltmarsh sediments requires an understanding of the modern ecology of the organisms concerned. Previous work suggested that testate amoebae are a potential new group of organisms to use for sea-level reconstruction, especially combined with diatoms and foraminifera. This paper analyses data from three saltmarshes on the Taf estuary, South Wales, the River Erme, Devon, and at Brancaster, Norfolk (UK) to (i) test for the presence and zonation of testate amoebae in relation to elevation; (ii) examine the similarity of zonation patterns between marshes; and (iii) explore the relationship between assemblage composition and a wider range of environmental variables. In addition we provide an update on the identification of testate amoebae on saltmarshes. Our results confirm that at all sites the primary environmental gradient is tidal inundation. Major changes in taxa along the tidal gradient are similar except for the lowest elevations, where different taxa become dominant at different sites. Canonical correspondence analysis (CCA) shows that assemblage composition is also strongly related to other variables, independent of the tidal position. Salinity, particle size and organic matter content are particularly important, and there is a statistically significant geographical effect on assemblages. Relationships between sea-level and assemblage composition are often stronger for individual sites, suggesting that local data sets should be used for quantitative sea-level reconstructions. However, the combined data set would provide more robust estimates of past sea-level change from fossil data. Other environmental variables explain as much of the variability in species assemblages as tidal parameters and should be considered more often in sea-level reconstructions based on microfossil indicators. Cell wall phenolics of white and green asparagus

Abstract Cinnamic acids influence physical and textural attributes of plant foods as they play an important role in lignification and cross-linking of cell-wall polymers. Several trans -cinnamic acids have been detected in asparagus cell wall (CW), and, of these, amounts of ferulic acid (FA) and its dehydrodimers have been found to increase significantly during post-harvest storage. The distribution of these compounds along the different sections of white and green spears, as well as their modifications during postharvest storage, has been investigated. It has been observed that the cell walls from the apical, middle and lower sections of the green spears contain equivalent amounts of FA derivatives (about 400 µg g,1 CW), while these compounds are mainly located in the middle (681 µg g,1 CW) and lower section (975 µg g,1 CW) of the white ones. During asparagus post-harvest storage a general increase of FA monomers and dimers took place that affected every section of both green and white spears. Major changes occurred in the middle and lower sections of the white asparagus, where the amounts of ferulic derivatives increased to 1700 and 1678 µg g,1 CW after storage. Weed vegetation of arable land in Central Europe: Gradients of diversity and species composition

Abstract: Question: What are the main broad-scale spatial and temporal gradients in species composition of arable weed communities and what are their underlying environmental variables? Location: Czech Republic and Slovakia. Methods: A selection of 2653 geographically stratified relevés sampled between 1954,2003 was analysed with direct and indirect ordination, regression analysis and analysis of beta diversity. Results: Major changes in weed species composition were associated with a complex gradient of increasing altitude and precipitation and decreasing temperature and base status of the soils. The proportion of hemicryptophytes increased, therophytes and alien species decreased, species richness increased and beta diversity decreased with increasing altitude. The second most important gradient of weed species composition was associated with seasonal changes, resulting in striking differences between weed communities developed in spring and summer. In summer, weed communities tended to have more neophytes, higher species richness and higher beta diversity. The third gradient reflected long-term changes in weed vegetation over past decades. The proportion of hemicryptophytes and neophytes increased, while therophytes and archaeophytes decreased, as did species richness over time. The fourth gradient was due to crop plants. Cultures whose management involves less disturbances, such as cereals, harboured less geophytes and neophytes, and had higher species richness but lower beta diversity than frequently disturbed cultures, such as root crops. Conclusions: Species composition of Central European weed vegetation is mainly influenced by broad-scale climatic and edaphic factors, but its variations due to seasonal dynamics and long-term changes in agricultural management are also striking. Crop plants and crop-specific management affect it to a lesser, but still significant extent. [source]


Early Embryonic Development of the Camel Lumbar Spinal Cord Segment

The lumbar spinal cord segment of the camel embryo at CVRL 2.4 to 28 cm was examined. Major changes are occurring in the organization of the lumbar spinal cord segments during this early developmental period. At the CVRL 2.4, 2.7 and 3.6 cm the three primary layers, ependymal cells layer, mantle cells layer, marginal cells layer in the developing lumber spinal cord segment were demonstrated. The mantle layer is the first to show striking differentiation, while the marginal layer is represented by thin outer rim. Proliferation and differentiation of the neuroepithelial cells in the developing spinal cord produce the thick lateral walls, thin roof and floor plates. The spinal ganglion and dorsal root of the spinal nerve are differentiated. At 2.7 cm CVRL differential thickening of the lateral walls produces a shallow longitudinal groove called sulcus limitans, which separates the dorsal part (alar plate) from ventral part (basal plate). The ventral root of the spinal nerve, the spinal cord and ganglion are embedded in loose mesenchyme, which tends to differentiate into spinal meninges. At 3.6 cm CVRL the basal plate, which is the future ventral gray horn, seem to be quite voluminous and the dorsal and ventral roots unite to form the beginning of the spinal nerve. At 5.5 cm CVRL the alar plates enlarge forming the dorsal septum. At 8.4 cm to 10.5 cm CVRL the basal plates enlarge, and bulge ventrally on each side of the midline producing the future ventral medium fissure, and the white and gray matters can be recognized. At 28 cm CVRL the lumen of the spinal cord is differentiated into the central canal bounded dorsally and ventrally by dorsal and ventral gray commissures, and therefore the gray matter takes the appearance of a butterfly. Decadal changes (1996,2006) in coastal ecosystems of the Chagos archipelago determined from rapid assessment

Abstract 1.The atolls and islands comprising Chagos are a biodiversity hotspot of global conservation significance in a remote part of the central Indian Ocean. 2.This study examines the condition of the archipelago's coastal ecosystems by rapid environmental assessment at 21 sites/islands, which were also investigated a decade earlier using the same methodology. Major changes in ecosystem structure and environmental disturbance were determined. 3.Coral fish abundance was significantly lower in 2006 than 1996. Decrease in the physical structural complexity of the reefs, as a result of coral bleaching and mortality induced by the 1998 warming event, may have been a contributing factor. 4.Evidence of collecting/fishing was significantly greater in 2006 than 1996. This is attributed mainly to an illegal fishery for holothurians (sea cucumbers), which has expanded over recent years and now exerts substantial pressure on the resource. The significant decline observed in beach wood, a readily accessible fuel for fishing camps, is consistent with this. 5.Solid waste on islands was high (median 2 to 20 items m,1 beach) in both 1996 and 2006. Potentially harmful biological impacts, determined from other studies, include entanglement, toxic effects and provision of transport for invasives or other ,hitchhiker' species. 6.Significantly higher bird abundances were recorded in protected areas than ,unprotected' areas, attributed mainly to absence of predation by rats. 7.Rapid assessment augments more comprehensive ecosystem investigations. It provides a valuable snapshot of environmental conditions based upon a broad suite of features (ecosystems and disturbances) determined, concurrently, within the same site inspection quadrats and using the same scale of assessment. Radioiodine treatment for benign thyroid diseases

Summary Radioiodine has been in use for over 60 years as a treatment for hyperthyroidism. Major changes in clinical practice have occurred with the realization that accurate dosimetry is incapable of avoiding the risks of hypothyroidism, while more accurate assessment of the risks of other adverse effects of radioiodine such as ophthalmopathy and carcinogenesis have become available. More is also known of the potential for pretreatment with an antithyroid drug to affect the outcome of radioiodine treatment. However, we are still uncertain of the benefits of radioiodine treatment in subclinical hyperthyroidism. During the last two decades there has been wider acceptance of radioiodine as a safe and effective therapy for benign, nontoxic goitre, coupled with waning enthusiasm for the use of levothyroxine, as the risks and benefits of this option have become more apparent. The use of recombinant TSH offers the prospect that radioiodine treatment of nontoxic goitre can be simplified and improved, although more studies of this strategy are urgently required. [source]


Education for All: How Much Will It Cost?

In 1990, a target of universal access to basic education by the year 2000 was set by two global conferences. Ten years later, however, it was clear that the target had not been met. Too many countries had made insufficient progress, and although many of the reasons for this inadequate progress were country-specific, one factor stood out in virtually all countries: inadequate public finance for primary education. In 2000, the Millennium Summit set a new target date for achieving ,education for all' of 2015. This article updates the global and regional cost estimates for reaching that target. The estimates are based on the most recent country-by-country data on budgetary expenditure, population and enrolment trends, and unit cost. The annual additional cost of achieving ,education for all' in developing countries by 2015 is estimated at US$ 9.1 billion. Although this is affordable at the global level, individual countries will need considerably more resources than are currently available. However, official development assistance (ODA) has been declining, and the share of ODA allocated to basic education has changed little over the past decade. Therefore, although affordable, the target of universal basic education by 2015 is likely to be missed, just as it was in 2000, without a major change both in ODA and national budgets. [source]


Central fat predicts deterioration of insulin secretion index and fasting glycaemia: 6-year follow-up of subjects at varying risk of Type 2 diabetes mellitus

Abstract Aims To examine the relationships between body composition and changes in fasting glycaemia, and in indices of insulin secretion and insulin action over 6 years in females with a family history of Type 2 diabetes with or without prior gestational diabetes (,at risk' group, AR) and control females (control group, C). Methods At baseline and at follow-up, an oral glucose tolerance test and dual energy X-ray absorptiometry assessment of body composition were performed. Indices of insulin resistance (HOMA R,) and insulin secretion (HOMA ,,) were obtained from fasting insulin and glucose concentrations. Results At baseline, the groups were similar for age, body mass index, fasting levels of plasma glucose and insulin, HOMA R, and HOMA ,,. Despite similar total body fatness, AR had significantly greater waist circumference and central fat (both P < 0.02) compared with C. At follow-up there was a significant increase in central adiposity only in AR, and the fasting plasma glucose (FPG) level was higher in AR compared with C (5.0 ± 0.2 vs. 4.3 ± 0.2 mmol/l, P = 0.02). This rise in plasma glucose in AR was related to a decline in HOMA ,, (r = 0.45, P = 0.0065). Both the baseline and the increments in total and central abdominal fat mass were associated with the time-related decline in HOMA ,,. Conclusions Six years after initial assessment, AR showed deterioration in FPG levels due predominantly to a decline in insulin secretion index without major change in insulin resistance index. Importantly, baseline body fatness (especially central adiposity), as well as increases in fatness with time, were the major predictors of the subsequent decline of insulin secretion index and the consequent rise in FPG. Review of bupropion for smoking cessation

Abstract The advent of bupropion hydrochloride sustained release (Zyban) has heralded a major change in the options available for smoking cessation pharmacotherapy. Bupropion is a selective re-uptake inhibitor of dopamine and noradrenalin which prevents or reduces cravings and other features of nicotine withdrawal. Bupropion is a useful oral and non-nicotine form of pharmacotherapy for smoking cessation. For this review a total of 221 papers were reviewed plus poster presentations. This review examines in detail original clinical trials on efficacy, categorised according to whether they were acute treatment trials in healthy smokers; studies in specific populations such as people with depression, chronic obstructive pulmonary disease (COPD) or cardiovascular disease; or relapse prevention studies. Overall, these studies in varying populations comprising over four thousand subjects, showed bupropion consistently produces a positive effect on smoking cessation outcomes. The evidence highlights the major public health role that bupropion has in smoking cessation. The methodological issues of published clinical trials reporting one year outcomes were examined in detail including: completeness of follow-up; loss to follow-up; intention to treat analysis; blindness of assessment; and validation of smoking status. The review discusses contraindications, adverse effects, dose and overdose, addictive potential, and the role of bupropion in reducing cessation-related weight gain. Bupropion combined with or compared to other pharmacotherapies (nicotine patch; nortriptyline) is considered. Impressive evidence exists for the use of bupropion in smoking cessation among difficult patients who are hard-core smokers such as those with cardiovascular disease, chronic obstructive pulmonary disease (COPD) and depression. Bupropion reduces withdrawal symptoms as well as weight gain and is effective for smoking cessation for people with and without a history of depression or alcoholism. Serious side effects of bupropion use are rare. The major safety issue with bupropion is risk of seizures (estimated at approximately 0.1%) and it should not be prescribed to patients with a current seizure disorder or any history of seizures. In clinical trials of bupropion for smoking cessation no seizures were reported. Allergic reactions occur at a rate of approximately 3% and minor adverse effects are common including dry mouth and insomnia. [source]


Heroin-assisted treatment in Switzerland: a case study in policy change

ABSTRACT Background Switzerland introduced a pragmatic national drug policy when the former conservative abstinence-orientated politics proved unable to cope with an escalating number of users and related negative consequences for public health and public order. The high visibility of ,needle parks' and the size of the acquired immune deficiency disorder (AIDS) epidemic called for a new approach and for national leadership. Aims To describe the intentions, the process and the results of setting up the new treatment approach of prescribing heroin to treatment resistant heroin addicts, as an example of drug policy change. Materials and Methods A systematic collection of relevant documents is analysed and used as evidence for describing the process of policy change. Results Measures to reduce the negative consequences of continued use and to prevent the spread of AIDS were started mainly by private initiatives and soon taken up officially in the ,four-pillar' drug policy (including harm reduction, prevention, treatment and law enforcement). Medical prescription of heroin to chronic, treatment-resistant heroin addicts was one of the innovations, based on extensive scientific and political preparation. Detailed documentation and evaluation, ample communication of results, adaptations made on the basis of results and extensive public debate helped to consolidate the new policy and heroin-assisted treatment, in spite of its limitations as an observational cohort study. All necessary steps were taken to proceed from a scientific experiment to a routine procedure. Discussion Comparable policy changes have been observed in a few other countries, such as The Netherlands and Germany, based on the Swiss experience, with equally positive results of heroin-assisted treatment. These experiments were designed as randomised controlled trials, comparing intravenous heroin against oral methadone, thereby demonstrating the specific value of pharmaceutical diamorphine for maintenance treatment in opiate dependence. The positive impact of policy change and the positive outcomes of heroin-assisted treatment were acknowledged increasingly nationally and internationally, but made it difficult to continue the process of adapting policy to new challenges, due to the low visibility of present drug problems and to changing political priorities. Conclusion A major change in drug policy was effectively realised under typical conditions of a federalist country with a longstanding tradition of democratic consensus building. Facilitating factors were the size and visibility of the heroin problem, the rise of the Aids epidemic, and a pragmatic attitude of tolerating private initiatives opening the way to official policy change. Edible insects: Traditional knowledge or western phobia?

Abstract With an increasing human population and environmental degradation, the world faces a major problem in providing adequate animal based proteins. Many traditional societies have used or still use insects as a protein source, while westernized societies are reluctant to use insects, despite being the major consumers of animal proteins. We now need to consider insects as a source of food for humans in a manner that acknowledges both the role of entomophagy in indigenous societies and the need for westernized societies to reduce the size of their environmental footprint with regard to food production. The situation on continents such as Africa, Asia, and Central and South America has some parallels to Australia in that there are two forces in operation: the sustainable traditional use of edible insects and the "westernization" of these societies leading to a movement away from entomophagy. However, the potential to reach a compromise is greater in these continents because entomophagy is already accepted. The major challenges will be establishing sustainable production systems that include food safety and security as well as environmental protection. Whether this will happen or not will depend upon: (i) a major change in attitude in westernized societies towards entomophagy; (ii) pressure to conserve remaining habitats in a sustainable manner; (iii) economic impetus to develop food production systems that include insects; and (iv) an acknowledgement that achieving adequate nutrition on a global basis will involve different diets in much of the developed world. Subcellular distribution of zinc in Daphnia magna and implication for toxicity

Abstract We examined the subcellular partitioning of zinc (Zn) in Daphnia magna both under acute and chronic exposures. In the acute Zn toxicity tests, the daphnids were exposed to different Zn concentrations for 48,h or to one lethal concentration (1,000,µg/L) for different durations (time to death for up to 47,h). Significant mortality of daphnids was observed when the newly accumulated Zn concentration reached a threshold level of approximately 40,µg/g wet weight (or 320,µg/g dry wt), approximately 3.5 times higher than the background tissue concentration (92,µg/g dry wt). Chronic exposure (14 d) to Zn resulted in nonobservable effect on survivorship and growth at newly accumulated tissue concentration of over 40,µg/g wet weight. With increasing Zn acute exposure, more Zn was partitioned into the cellular debris fraction, indicating that this fraction was presumably the first targeted site of binding for Zn upon entering the animals. The importance of other subcellular fractions either decreased accordingly or remained comparable. We found that the metal-sensitive fraction (Zn distribution in the organelles and heat-denatured proteins) did not predict the acute Zn toxicity in Daphnia. During chronic exposure, however, no major change of the subcellular partitioning of Zn with increasing Zn exposure was documented. Zinc was mainly found in the organelles and heat-stable protein fractions during chronic exposure, suggesting that any subcellular repartitioning occurred primarily during acute exposure. Metallothioneins were induced upon chronic Zn exposure, but its induction evidently lagged behind the Zn accumulation. Our present study showed that the subcellular fractionation approach could not be readily used to predict the acute and chronic toxicities of Zn in Daphnia. Influence of isolation on the recovery of pond mesocosms from the application of an insecticide.

Abstract The immediate response and recovery of the macrobenthic communities of nonisolated and isolated freshwater outdoor 9 m3 mesocosms following an acute stress caused by the addition of deltamethrin were studied over a 14-month period. To discriminate between internal and external recovery mechanisms, half of the treated ponds were covered by 1-mm mesh lids that restricted aerial recolonization. Both structural (abundance of the different taxonomic groups) and functional (litter breakdown) parameters were monitored. Insects were broadly reduced in numbers by deltamethrin addition. In general, noninsect groups were not affected or increased in abundance in deltamethrin-treated ponds, probably because of relative insensitivity to deltamethrin, reduced predation, and lower competition for food. No major change in litter breakdown rates were seen, probably because of functional redundancy among the macrobenthic community. Chironominae larvae recovered in open, treated mesocosms 62 d after deltamethrin addition and most insect groups recovered 84 d after the treatment date. However, the presence of lids significantly reduced insect recovery rate, suggesting that it largely depends on the immigration of winged forms (i.e., external recovery) from surrounding non- or less affected systems. These results indicate that the recovery time of macrobenthic communities in an affected natural pond would depend on spatial characteristics of the landscape and also the season that exposure occurs. Isolated ecosystems would display posttreatment insect recovery dynamics very different from highly connected ones, evolving toward alternate pseudoequilibrium states, possibly with lower biodiversity but with preserved functionality. Consequences for higher tier risk assessment of pesticides are discussed. [source]


Let eating disorder patients decide: Providing choice may reduce early drop-out from inpatient treatment

Abstract Premature drop-out from treatment is a highly prevalent phenomenon among eating disorder (ED) patients. In a specialized inpatient treatment unit a major change was made in the admission strategy in 2001, giving a maximum of personal choice to the patients. A quasi-experimental research was carried out comparing 87 patients treated till 2000 (,old' strategy) with 87 patients treated from 2001 on (,new' strategy). The results indicate that the provision of choice at the beginning of treatment significantly reduced drop-out during the first weeks of inpatient treatment. No differences between both strategies on later drop-out and weight change (in anorexia nervosa patients) during inpatient treatment were found. The results are discussed in the light of the importance placed on dynamics of personal choice, autonomy and volition within the framework of the self-determination theory (SDT). Altered kynurenine metabolism correlates with infarct volume in stroke

Abstract Inflammation and oxidative stress are involved in brain damage following stroke, and tryptophan oxidation along the kynurenine pathway contributes to the modulation of oxidative stress partly via the glutamate receptor agonist quinolinic acid and antagonist kynurenic acid, and via redox-active compounds such as 3-hydroxyanthranilic acid. We have confirmed that following a stroke, patients show early elevations of plasma neopterin, S100B and peroxidation markers, the latter two correlating with infarct volume assessed from computed tomography (CT) scans, and being consistent with a rapid inflammatory response. We now report that the kynurenine pathway of tryptophan metabolism was also activated, with an increased kynurenine : tryptophan ratio, but with a highly significant decrease in the ratio of 3-hydroxyanthranilic acid : anthranilic acid, which was strongly correlated with infarct volume. Levels of kynurenic acid were significantly raised in patients who died within 21 days compared with those who survived. The results suggest that increased tryptophan catabolism is initiated before or immediately after a stroke, and is related to the inflammatory response and oxidative stress, with a major change in 3-hydroxyanthranilic acid levels. Together with previous evidence that inhibiting the kynurenine pathway reduces brain damage in animal models of stroke and cerebral inflammation, and that increased kynurenine metabolism directly promotes oxidative stress, it is proposed that oxidative tryptophan metabolism may contribute to the oxidative stress and brain damage following stroke. Some form of anti-inflammatory intervention between the rise of S100B and the activation of microglia, including inhibition of the kynurenine pathway, may be valuable in modifying patient morbidity and mortality. Calcium modulates endopeptidase 24.15 (EC 3.4.24.15) membrane association, secondary structure and substrate specificity

The metalloendopeptidase 24.15 (EP24.15) is ubiquitously present in the extracellular environment as a secreted protein. Outside the cell, this enzyme degrades several neuropeptides containing from 5 to 17 amino acids (e.g. gonadotropin releasing hormone, bradykinin, opioids and neurotensin). The constitutive secretion of EP24.15 from glioma C6 cells was demonstrated to be stimulated linearly by reduced concentrations of extracellular calcium. In the present report we demonstrate that extracellular calcium concentration has no effect on the total amount of the extracellular (cell associated + medium) enzyme. Indeed, immuno-cytochemical analyses by confocal and electron microscopy suggested that the absence of calcium favors the enzyme shedding from the plasma membrane into the medium. Two putative calcium-binding sites on EP24.15 (D93 and D159) were altered by site-directed mutagenesis to investigate their possible contribution to binding of the enzyme at the cell surface. These mutated recombinant proteins behave similarly to the wild-type enzyme regarding enzymatic activity, secondary structure, calcium sensitivity and immunoreactivity. However, immunocytochemical analyses by confocal microscopy consistently show a reduced ability of the D93A mutant to associate with the plasma membrane of glioma C6 cells when compared with the wild-type enzyme. These data and the model of the enzyme's structure as determined by X-ray diffraction suggest that D93 is located at the enzyme surface and is consistent with membrane association of EP24.15. Moreover, calcium was also observed to induce a major change in the EP24.15 cleavage site on distinctive fluorogenic substrates. These data suggest that calcium may be an important modulator of ep24.15 cell function. [source]


Phosphorus decrease and climate variability: mediators of synchrony in phytoplankton changes among European peri-alpine lakes

Summary 1. In an attempt to discern long-term regional patterns in phytoplankton community composition we analysed data from five deep peri-alpine lake basins that have been included in long-term monitoring programmes since the beginning of the 1970s. Local management measures have led to synchronous declines in phosphorus concentrations by more than 50% in all four lakes. Their trophic state now ranges from mesotrophic to oligotrophic. 2. No coherence in phytoplankton biomass was observed among lakes, or any significant decrease in response to phosphorus (P)-reduction (oligotrophication), except in Lakes Constance and Walen. 3. Multivariate analyses identified long-term changes in phytoplankton composition, which occurred coherently in all lakes despite the differing absolute phosphorus concentrations. 4. In all lakes, the phytoplankton species benefiting from oligotrophication included mixotrophic species and/or species indicative of oligo-mesotrophic conditions. 5. A major change in community composition occurred in all lakes at the end of the 1980s. During this period there was also a major shift in climatic conditions during winter and early spring, suggesting an impact of climatic factors. 6. Our results provide evidence that synchronous long-term changes in geographically separated phytoplankton communities may occur even when overall biomass changes are not synchronous. [source]


Deconstructing helicoplacoids: reinterpreting the most enigmatic Cambrian echinoderms

Abstract Helicoplacoids, the most enigmatic group of Early Cambrian echinoderms, have a spirally plated, inverted pear- to spindle-shaped theca with numerous interambulacral pleats and a long triradiate ambulacrum. They occur in western Laurentia along with the earliest edrioasteroids and epispire-bearing eocrinoids(?), but unlike these other groups, helicoplacoids do not show pentameral symmetry around a centrally located, upward-facing mouth. It is uncertain where the major body openings were located in helicoplacoids, where new plates were added during thecal growth, and whether helicoplacoids were primitive, pre-pentameral, stem-group echinoderms or derived, relatively specialized forms that had lost several ambulacra. We made highly modified, side-layout, plating diagrams of different helicoplacoid species by cutting the theca apart half-way between the spiraling ambulacra, leaving four to six interambulacral pleats on each side. This deconstructed thecal strip resulted in an elongate sigmoid-shaped plating diagram with a long central single or paired ambulacrum that is interpreted as three ambulacral branches (A down, C and D up) around a central mouth on the side of the theca. The unrolled plating diagram resembles an elongate French curve drawing template with nearly parallel sides, a large or small sigmoidal top (where the theca is rounded and the interambulacral pleats converge), and an open deviated bottom, where the interambulacral pleats turn down to a truncated conical base. The two upward-extending ambulacra are only one to two interambulacral pleats apart, which only slightly increases the plating strip width. New interambulacral plates are added at the summit and base, and interambulacral pleats rarely branch elsewhere. New floor and cover plates are added at the top and bottom distal ambulacral tips. If this plating model is correct, the anus (which has still not been identified) should be located near the thecal summit where the C and D ambulacra end, and all imperforate extraxial plating has apparently been lost from the theca. If upright, spirally pleated helicoplacoids with three ambulacra were derived from attached, domal or pyrgate, pentameral edrioasteroids, numerous changes must have occurred in the thecal shape, number of ambulacra, and interambulacral areas. Reform in a Cold Climate: Change in US Campaign Finance Law

The Bipartisan Campaign Reform Act (BCRA) of 2002 was the first major change in US federal campaign finance law in a quarter of a century. Many attempts at reform had failed in that period. Few members of Congress were enthusiasts for reform, the two parties and two chambers had conflicting interests to protect, successive presidents did not promote the issue and public pressure for reform was weak. When reform was achieved in 2002, many of these formidable obstacles remained in place. This paper draws on the literature of public interest reform and policy innovation to attribute the change to a policy entrepreneur whose resources had undergone a sharp increase, the neutralization of opposition, the impact of an event (the bankruptcy of the Enron Corporation) and membership turnover in Congress. The substantial support for the bill in Congress from Democrats, the party with most to lose from reform, is attributed to the inescapability of past commitments. [source]

Speciation chronology of rockhopper penguins inferred from molecular, geological and palaeoceanographic data

Abstract Aim, The Southern Ocean is split into several biogeographical provinces between convergence zones that separate watermasses of different temperatures. Recent molecular phylogenies have uncovered a strong phylogeographic structure among rockhopper penguin populations, Eudyptes chrysocome sensu lato, from different biogeographical provinces. These studies suggested a reclassification as three species in two major clades, corresponding, respectively, to warm, subtropical and cold sub-Antarctic watermasses rather than to geographic proximity. Such a phylogeographic pattern, also observed in plants, invertebrates and fishes of the Southern Ocean, suggests that past changes in the positions of watermasses may have affected the evolutionary history of penguins. We calculated divergence times among various rockhopper penguin clades and calibrated these data with palaeomagmatic and palaeoceanographic events to generate a speciation chronology in rockhopper penguins. Location, Southern Ocean. Methods, Divergence times between populations were calculated using five distinct mitochondrial DNA loci, and assuming a molecular clock model as implemented in mdiv. The molecular evolution rate of rockhopper penguins was calibrated using the radiochronological age of St Paul Island and Amsterdam Island in the southern Indian Ocean. Separations within other clades were correlated with palaeoceanographic data using this calibrated rate. Results, The split between the Atlantic and Indian populations of rockhopper penguins was dated as 0.25 Ma, using the date of emergence of St Paul and Amsterdam islands, and the divergence between sub-Antarctic and subtropical rockhopper penguins was dated as c. 0.9 Ma (i.e. during the mid-Pleistocene transition, a major change in the Earth's climate cycles). Main conclusions, The mid-Pleistocene transition is known to have caused a major southward shift in watermasses in the Southern Ocean, thus changing the environment around the northernmost rockhopper penguin breeding sites. This ecological isolation of northernmost populations may have caused vicariant speciation, splitting the species into two major clades. After the emergence of St Paul and Amsterdam islands in the subtropical Indian Ocean 0.25 Ma, these islands were colonized by penguins from the subtropical Atlantic, 6000 km away, rather than by penguins from the sub-Antarctic Indian Ocean, 5000 km closer.
