Peirce Abstract We present a novel multigrid (MG) procedure for the efficient solution of the large non-symmetric system of algebraic equations used to model the evolution of a hydraulically driven fracture in a multi-layered elastic medium. The governing equations involve a highly non-linear coupled system of integro-partial differential equations along with the fracture front free boundary problem. The conditioning of the algebraic equations typically degrades as O(N3). A number of characteristics of this problem present significant new challenges for designing an effective MG strategy. Large changes in the coefficients of the PDE are dealt with by taking the appropriate harmonic averages of the discrete coefficients. Coarse level Green's functions for multiple elastic layers are constructed using a single dual mesh and superposition. Coarse grids that are sub-sets of the finest grid are used to treat mixed variable problems associated with ,pinch points.' Localized approximations to the Jacobian at each MG level are used to devise efficient Gauss,Seidel smoothers and preferential line iterations are used to eliminate grid anisotropy caused by large aspect ratio elements. [source] Accumulation of methylglyoxal in the gingival crevicular fluid of chronic periodontitis patientsJOURNAL OF CLINICAL PERIODONTOLOGY, Issue 4 2003S. Kashket Abstract Background, aims: Methylglyoxal (MG), a toxic product of cellular metabolism, is elevated in tissues and fluids in a number of human diseases. A cross-sectional study was undertaken to determine whether MG accumulates in the gingival crevicular fluid (GCF) of chronic periodontitis patients. Methods: GCF samples were collected for 30 s each from three teeth with pocket depths greater than 3 mm (DD sites), from 14 chronic periodontitis patients. Control samples were taken from three healthy sites (DH sites) in the same patients, as well as from seven subjects who were periodontally healthy (HH sites). Fluid volumes were determined and the strips were placed in 0.5 N perchloric acid. Subsequently, samples were derivatized with o -phenylenediamine and the resulting methylquinoxaline was assayed by high-performance liquid chromatography on Lichrospher® -100 RP-18, with UV detection. Results: Mean pocket depths were 5.7±0.7, 2.7±0.6 and 2.7±0.5 mm (mean±SD) for the DD, DH and HH sites, respectively. Mean MG levels were found to be 208.7±241.7 and 142.9±235.7 pmol/site in the GCF from DD and DH sites, respectively (p=0.0023), but only 11.5±4.4 pmol/site for the HH sites. Bacteroides forsythus has been found to accumulate high levels of MG in culture (unpublished data) and, consistent with this, the sampled diseased sites contained higher levels of B. forsythus than the corresponding healthy sites (2.7±4.2×105 versus 0.7±1.1×105, respectively; p=0.022). Total "red complex" microorganisms were significantly elevated in the DD sites. Conclusions: In view of the known protein- and DNA-modifying effects of MG, the finding of elevated levels of MG in the GCF from chronic periodontitis patients supports the hypothesis that MG may contribute to destructive tissue damage in this disease. Zusammenfassung Hintergrund: Methylglyoxal (MG), ein toxisches Produkt des Zellstoffwechsels, ist bei einer Reihe menschlicher Erkrankungen in Geweben und Körperflüssigkeiten erhöht. Zielsetzung: Querschnittsstudie zur Klärung der Frage, ob sich MG in der Sulkusflüssigkeit (SF) bei Patienten mit chronischer Parodontitis ansammelt. Methoden: Bei 14 Patienten mit chronischer Parodontitis wurde SF für 30 s an 3 Zähnen mit Sondierungstiefen >3 mm (DD-Stellen) gewonnen. Kontrollproben wurden von jeweils 3 gesunden Stellen (DH-Stellen) bei den gleichen Patienten und bei 7 parodontal gesunden Personen gewonnen (HH-Stellen). Die Flüssigkeitsvolumina wurden bestimmt und die Probenstreifen in 0,5 N Perchlorsäure gegeben. Anschließend wurden die Proben mit o-Phenyldiamin versetzt und das resultierende Methylquinoxalin wurde mittels High-Performance-Liquid-Chromatographie in einem Lichrospher®-100 RP-18 bei UV-Detektion nachgewiesen. Ergebnisse: Die mittleren Sondierungstiefen lagen bei 5,7±0,7, an den DD-Stellen, bei 2,7±0,6 (DH) und 2,7±0,5 mm (Mittelwert + Standardabweichung) (HH). Die mittleren MG-Spiegel lagen bei 208,7±241,7 an den DD-Stellen und bei 142,9±235,7 pmol/Stelle an den DH-Stellen (p=0,0023), aber nur bei 11,5±4,4 pmol/Stelle an den HH-Stellen. Es war gezeigt worden, dass Bacteroides forsythus in Kultur hohe Konzentrationen von MG ansammelt (unveröffentlichte Daten) und übereinstimmend damit wurden an den untersuchten DD-Stellen höhere Zahlen von B. forsythus gefunden als an den entsprechenden gesunden Stellen (2,7±4,2×105 versus 0,7±1,1×105; p=0,022). Die Zahl aller Keime des "Roten Komplexes" waren an DD-Stellen signifikant erhöht. Une étude croisée fut réalisée pour déterminer si MG s'accumulait dans le fluide gingival (GCF) des patients atteints de parodontite chronique. Méthodes: Des échantillons de GCF furent prélevés pendant 30 secondes chacun de 3 dents avec des profondeurs de poches de plus de 3 mm (sites DD), chez 14 patients atteints de parodontites chroniques. Des échantillons contrôles furent prélevés sur des sites sains (sites DH) chez les mêmes patients, et aussi chez 7 sujets au parodonte sain (sites HH). Les volumes de fluide furent déterminés et les bandelettes ont été mises dans 0.5 N d'acide perchlorique. Puis, les échantillons furent transformés à l'aide de o-phenylenediamine et la méthylquinoxaline ainsi créée fut analysé par chromatographie liquide à haute performance sur une Lichrospher®-100 RP-18, avec détection aux UV. [source] Aldose Reductase and AGE,RAGE pathways: central roles in the pathogenesis of vascular dysfunction in aging ratsAGING CELL, Issue 5 2010Kellie McCormick Hallam Summary Aging is inevitably accompanied by gradual and irreversible innate endothelial dysfunction. In this study, we tested the hypothesis that accentuation of glucose metabolism via the aldose reductase (AR) pathway contributes to age-related vascular dysfunction. AR protein and activity levels were significantly increased in aged vs. young aortic homogenates from Fischer 344 rats. Immunostaining revealed that the principal site of increased AR protein was the aortic endothelium as well as smooth muscle cells. Studies revealed that endothelial-dependent relaxation (EDR) in response to acetylcholine was impaired in aged rats compared to young rats and that treatment with the AR inhibitor (ARI) zopolrestat significantly improved EDR in aged rats. Methylglyoxal (MG), a key precursor of advanced glycation endproducts (AGEs), was significantly increased in the aortas of aged rats vs. young rats. Consistent with central roles for AR in generation of MG in aging, ARI treatment significantly reduced MG levels in aged rat aorta to those in young rats. Treatment of aged rats with soluble(s) RAGE, a soluble form of the chief signal transduction receptor for AGEs, RAGE, significantly improved EDR in aged rats, thus establishing the contribution of age-related increases in AGEs to endothelial dysfunction. These findings reveal that significant increases in AR expression and activity in aged rat vasculature linked to endothelial dysfunction may be mitigated, at least in part, via ARI and that aging-linked increased flux via AR generates AGEs; species which transduce endothelial injury consequent to their interaction with RAGE. These data demonstrate for the first time that AR mediates aging-related vascular dysfunction, at least in part, via RAGE. The aim of the present study was to compare the value of measuring neonatal urinary ,2 -microglobulin (,2 -MG) levels with fetal blood interleukin (IL)-6 levels in premature infants at risk of developing CLD. Methods:, Premature infants (gestational age <30 weeks) without CLD (n = 19) and with CLD (n = 10) were enrolled. We measured IL-6 levels in umbilical cord blood and ,2 -MG levels in urine obtained within 48 h after birth. Results:, IL-6 and ,2 -MG levels were significantly higher in infants who developed CLD than in those who did not (median IL-6, 54.7 vs 7.6 pg/mL; P < 0.005; ,2 -MG 17.7 vs 9.3 × 104 µg/gCr; P < 0.05). The sensitivity and negative predictive value of ,2 -MG at the cut-off value at 10.0 × 104 µg/gCr (0.90 and 0.92) were comparable to IL-6 at 16 pg/mL (0.90 and 0.94). Conclusion:, We suggest that measuring urinary ,2 -MG in premature infants soon after birth can monitor FIRS and may provide information on the risk of subsequent CLD development that is as clinically important as information derived from umbilical cord blood IL-6. [source] Influence of Dietary Levels of Magnesium on Growth, Tissue Mineral Content, and Resistance of Channel Catfish Ictalurus punctatus Challenged with Edwardsiella ictaluriJOURNAL OF THE WORLD AQUACULTURE SOCIETY, Issue 1 2003Chhorn Lim Juvenile channel catfish were fed purified diets supplemented with magnesium (Mg) from Mg sulfate at levels of 0, 200, 400, 600, 800, and 1,000 mg/kg and 0, 200, 400, 600, and 800 mg/kg in two separate feeding studies. In study I, the effect of dietary levels of Mg on growth response, vertebral mineral content, and macrophage chemotaxis were evaluated. Study II had similar objectives except that whole body mineral content was measured, and resistance of channel catfish to Edwardsiella ictaluri challenge was also determined. Fish with an average weight of 10.89 g were stocked at a rate of 50 fish/110-L aquarium (study I). In study II, fish with an average weight of 4.14 g were stocked at rates of 40 fish/110-L aquarium. Prior to stocking, each batch of fish was acclimated to laboratory conditions and fed the basal diet for 2 wk. The concentration of Mg in rearing water was 1.8 mg/L. Each diet was fed to fish in quadruplicate and triplicate aquaria to apparent satiation for 10 wk for studies I and II, respectively. Fish fed the basal diet started to die as early as 3 d after the study began (17 d of feeding the diet without Mg supplementation). In both studies, weight gain, survival, and feed efficiency were lowest for fish fed the basal diet but increased with increasing dietary levels of Mg. However, the differences between the values of each of these parameters for fish fed diets containing supplemental Mg were not always significant. Magnesium-deficiency signs observed were anorexia, sluggishness, convulsions, deformed snout, vertebral curvature, muscle flaccidity, and high mortality. Vertebral and whole body ash concentrations were high, but Mg content was low for fish fed the basal and the 200-mg Mg diets. Bone Ca content did not differ among fish fed different diets (study I), but whole body Ca tended to increase for fish fed the basal diet, suggesting the possibility of calcification of soft tissues. Macrophage chemotaxis in the presence of exoantigen was highest for fish fed diets supplemented with Mg at 400 and 200 mgkg for studies I and II, respectively. When expressed in terms of chemotaxis index, however, maximum or near maximum value was observed at a dietary Mg level of 400 mg/kg. Thus, a dietary level of Mg of 400 mg/kg from Mg sulfate was required for optimum growth and survival, maintaining high tissue levels of Mg, prevention of muscle flaccidity and skeletal deformity, and stimulating macrophage chemotaxis. Hypomagnesemia is common in both renal transplant recipients and in diabetics. This study examines the relationship between hypomagnesemia, NODAT and the type of immunosuppression in renal transplant recipients. We conducted a retrospective single-center analysis (2002,2008) in order to assess NODAT the first year posttransplantation as defined by American Diabetes Association criteria. Serum magnesium (Mg) levels were defined as the median of all Mg levels registered during the first month posttransplantation. Patients with NODAT (N = 75; 29.5%) versus non-NODAT had lower Mg levels (p < 0.001). Patients with an Mg level < versus ,1.9 mg/dL showed a faster development of NODAT (log-rank p < 0.001). Mg levels were lower in patients on calcineurin inhibitors (CNI) versus no CNI patients (p < 0.001). Mg levels, albumin, BMI, triglycerides, posttransplantation hyperglycemia, tacrolimus levels and the use of sirolimus were predictors of NODAT in the multivariate analysis. Hypomagnesemia was an independent predictor of NODAT in renal transplant recipients. We confirm that the use of CNI is associated with NODAT, but, to a large extent, this effect seems attributable to the induction of hypomagnesemia. After adjustment for Mg, sirolimus was also associated with NODAT. [source] EFFECTS OF ADMINISTRATION OF ORAL MAGNESIUM ON PLASMA GLUCOSE AND PATHOLOGICAL CHANGES IN THE AORTA AND PANCREAS OF DIABETIC RATSCLINICAL AND EXPERIMENTAL PHARMACOLOGY AND PHYSIOLOGY, Issue 8 2005Nepton Soltani SUMMARY 1.,Magnesium deficiency has recently been proposed as a novel factor implicated in the pathogenesis of the complications of diabetes. The purpose of the present study was to determine the relationship between oral Mg supplementation and changes in plasma glucose, calcium, haemogolobin, Ca/Mg ratio, blood pressure and the histology of the pancreas and vascular system in streptozotocin-induced diabetic rats. 2.,Ten days after the induction of diabetes in male Wistar rats, half the diabetic animals were divided into six groups, receiving 0, 1, 3, 10, 30 or 50 g/L MgSO4 added into the drinking water for 8 weeks. Plasma glucose and Mg were measured at days 1, 2, 3, 5, 7, 14 and 21 to find the optimum dose of Mg and the time-course of its effect. In addition, histological observations were undertaken. Eight weeks later, all animals were decapitated, the pancreas and thoracic aorta were removed carefully and immersed immediately in 10% formaldehyde for histological study. 3.,To evaluate the effects of Mg on plasma glucose, calcium, haemoglobin, Mg and blood pressure, another group of animals was divided into four experimental groups, as follows: (i) non-diabetic controls received tap water for 8 weeks; (ii) acute diabetics received tap water for 10 days; (iii) chronic diabetic controls received tap water for 8 weeks; and (iv) Mg-treated chronic diabetic rats received 10 g/L MgSO4 added into the drinking water 10 days after the induction of diabetes for 8 weeks. 4.,Magnesium dose dependently affects plasma glucose levels. The peak effect was reached during the first 24 h following oral administration. Administration of 10 g/L MgSO4 results in the return of normal structure in the diabetic pancreas and aorta. Moreover, this concentration of MgSO4 causes glucose, haemoglobin, calcium, the Ca/Mg ratio and blood pressure to reach normal levels. Although the Mg level increases slightly following the administration of 10 g/L MgSO4 to diabetic rats, it never reaches control levels. 5.,On the basis of the results of the present study, it may be concluded that chronic Mg administration may have beneficial effects on diabetes. [source] Effects of Fungal Phytase on Utilization of Dietary Protein and Minerals, and Dephosphorylation of Phytic Acid in the Alimentary Tract of Channel Catfish Ictalurus punctatus Fed an All-Plant-Protein DietJOURNAL OF THE WORLD AQUACULTURE SOCIETY, Issue 1 2002Weibing Yan A feeding trial was conducted to quantify the effects of phytase at levels of 0, 500, 1,000, 2,000, 4,000, and 8,000 units (U) per kg diet on utilization of dietary protein and minerals by fingerling (12 g) channel catfish Ictalurus punctatus fed an all-plant-protein diet composed of soybean meal, corn, and wheat middlings. The effects of phytase on dephosphorylation of phytic acid (phytate) in the alimentary tract of catfish also were determined. After 14 wk, mean weight gains (30.2,43.9 g/fish), feed conversion ratios (2.27,2.40 g feed consumed/g weight gain), protein efficiency ratios (1.47,1.61 g weight gaid/g protein consumed), and dietary protein retentions (23.8,26.7%) did not differ significantly (P > 0.05) among treatment groups. A digestibility trial conducted after the feeding trial showed no difference (P > 0.05) in mean digestibility of diet dry matter (49.0,58.3%) or crude protein (85.4-88.5%) among treatment groups. Concentrations of ash (46.7,48.6%), calcium (Ca, 17.9,18.5%), phosphorus (P, 9.1,9.5%), and manganese (Mn, 65.5,74.1 mg/kg) were significantly higher (P , 0.05) in bone of fish fed , 500 U/kg than in bone of fish fed 0 U/kg (ash, 43.5%; Ca, 16.4%; P, 8.4%; and Mn, 49.0 ma/kg), but concentrations of these minerals did not differ (P > 0.05) in bone of fish fed , 500 Uk/g. The magnesium (Mg) content of bone did not differ (P > 0.05) between fish fed 0 U/kg (0.29%) or 500 U/kg (0.34%), but was significantly lower in fish fed 0 U/kg than in fish fed , 1,000 U/kg (0.35,37%). Bone Mg levels did not differ (P > 0.05) among fish fed , 500 U/kg. The amount of zinc (Zn) in bone of fish fed 8,000 U/kg (153.3 mg/kg) was significantly higher than that in fish fed 0 U/kg (115.7 mg/kg) or 500 U/kg (130.3 mg/ kg), but did not differ from Zn levels in bone of fish fed 1,000,4,000 U/kg (134.5,135.8 mg/ kg). Dephosphorylation of phytate occurred primarily in the stomach within 2,8 h after diet ingestion, depending on the level of phytase supplementation. Initial levels of total phytate in the diet decreased 32,94% in stomach contents of fish fed l,000,8,000 U/kg within 2 h after feeding. Eight hours after feeding, stomach contents of fish fed , 1,000 U/kg contained less than 6% of initial total dietary phytate. Stomach contents of fish fed 500 U/kg retained 92% of initial total dietary phytate 2 h after feeding and 15% of total dietary phytate 8 h after feeding. Results of this study indicate that phytase supplementation at levels up to 8,000 U/kg diet did not increase weight gain or improve dietary protein utilization of channel catfish fed an all-plant-protein diet. Addition of phytase at a level of 1,000 U/kg diet was sufficient to significantly increase the Ca, P, Mg, and Mn content of bone, relative to fish fed an unsupplemented diet, and significantly decrease the quantity of total phytate in feces. A phytase level of 8,000 U/kg diet significantly increased the bioavailability of naturally occurring Zn in feed ingredients and increased the rate of phytate dephosphorylation in the stomach, compared with a diet containing no added phytase. Increased utilization of naturally occurring minerals in feed ingredients reduces the need for mineral supplements in diets and results in decreased elimination of minerals in feces. Thus, use of phytase in catfish feeds can be expected to provide both economic and environmental benefits. Halogens were first extracted from powdered samples using a pyrohydrolysis technique, then hydrolysis solutions were analysed by ion chromatography for F and Cl and inductively coupled plasma-mass spectrometry for Br and I. The detection limits in solutions were 100 ,g l,1 for both F and Cl and 10 ng l,1 for Br and I. Considering the extraction procedure, performed on a maximum of 500 mg of sample and producing 100 ml of pyrohydrolysis solution, detection limits in rock samples were 20 mg kg,1 for F and Cl and 2 ,g kg,1 for Br and I. The mean analytical errors on the studied composition ranges were estimated at 10 mg kg,1 for F and Cl, 100 ,g kg,1 for Br and 25 ,g kg,1 for I. The concentration values, based on repeated (generally > 10) sample analysis, were in good agreement generally with published values and narrowed the mean dispersion around mean values. Large dispersions are discussed in terms of samples heterogeneity and contaminations during sample preparation. Basaltic RMs were found to be more suitable for studies of halogen compositions than differentiated rock material, especially granites , the powders of which were heterogeneous in halogens at the 500 mg level. Les limites de détection sont de 100 ,g l,1 pour F et Cl, et de 10 ng l,1 pour Br et I, respectivement. L'extraction des halogènes était réalisée sur 500 mg de poudre de roche, produisant 100 ml de solution d'extraction. Ainsi, pour les échantillons de roche, les limites de détection étaient de 20 mg kg,1 pour F et Cl, et 2 ,g kg,1 pour Br et I. L'erreur analytique moyenne sur la gamme de concentration étudiée est estimée à 10 mg kg,1 pour F et Cl, 100 ,g kg,1 pour Br et 25 ,g kg,1 pour I. Les valeurs de concentrations données, obtenues par l'analyse répétée (> 10) du même échantillon étaient en accord avec les valeurs reportées dans la littérature. Elles présentent en général une plus faible dispersion autour de la valeur moyenne. Dans le cas d'une importante dispersion des résultats, celle-ci est discutée en terme d'hétérogénéité de l'échantillon analysé et de contamination durant la préparation du matériel de référence. Les échantillons de référence de composition basaltique se révèlent être plus appropriés pour étudier les compositions en halogènes que les matériaux correspondant à des roches différenciées, en particulier des granites dont la distribution en halogènes apparaît hétérogène dans les poudres à l'échelle d'un aliquot de 500 mg.
