c - fos and estrogen receptor gene expression pattern in the rat uterine epithelium during the estrous cycle

Abstract Different studies in ovariectomized estrogen treated animals support the idea that c - fos plays a role in the proliferation of uterine epithelial cells. However, these studies invite us to reassess the role played by c - fos in epithelial cell types of the endometrium during the estrous cycle. The present study was undertaken to determine the c - fos and estrogen receptor (ER) gene expression pattern in the rat uterine epithelium during the estrous cycle in which natural and cyclic changes of steroid hormones occur, and correlate these changes with the proliferation status of this cellular types. Proliferation was assessed during the estrous cycle using bromodeoxyuridine incorporation to DNA. ER, and , proteins were assessed by immunohistochemistry. The regulation of c - fos gene expression in the uterus of intact animals during the estrous cycle was evaluated using both in situ hybridization and immunohistochemistry. Estradiol (E2) and progesterone (P4) plasma levels were assessed by radioimmunoassay. The results indicated that luminal (LE) and glandular epithelia (GE) presented maximal proliferation during the metestrus (M) and the diestrus (D) days. However, during the proestrus (P) day only LE presented proliferation, and during the estrus (E) day only the stromal cells proliferated. A marked immunostaining for ER, was detected in both LE and GE cells during the early phases of the cycle but diminished on the P and the E day. In contrast, ER, was undetectable in both epithelia during all stages of the cycle. The highest c - fos mRNA level was detected in both epithelia on the M day, followed by a significant reduction during the other days of the cycle. The highest protein content was observed on the M and D days, and the minimal value was detected on the E day. The c-Fos protein level in LE was increased during M and D days, presenting a high correlation with the cellular proliferation pattern of this cell type. In conclusion, the overall results indicate that c-Fos protein presented a good correlation with uterine epithelial cell proliferation of LE. In the case of GE, the same tendency was observed, although no significant correlation was found. Both in LE and GE, c - fos mRNA did not strictly correlate with its protein levels. c - fos seems to have a postranscriptional regulation in uterine epithelial cells during the rat's estrous cycle. Spatial dynamics of supercolonies of the invasive yellow crazy ant, Anoplolepis gracilipes, on Christmas Island, Indian Ocean

ABSTRACT Key to the management of invasive species is an understanding of the scope of an invasion, the rate of proliferation and the rate at which invaded habitats become degraded. This study examines the spatial dynamics of high-density supercolonies of the invasive yellow crazy ant, Anoplolepis gracilipes, on Christmas Island, Indian Ocean, and the associated impacts at their boundaries. Since the early 1990s, A. gracilipes supercolonies have occupied over 30% of the 10,000 ha of rainforest on Christmas Island. Thirty-four discrete high-density supercolonies formed between 1989 and 2003, ranging in size across nearly three orders of magnitude from 0.9 to 787 ha. Supercolonies boundaries are diffuse, and ants were observed in low densities in some cases up to 200 m from the main high-density supercolony. The 13 boundaries examined were all dynamic over a 10,20 observation month period: nine boundaries expanded, and the maximum rate of spread was 0.5 m day,1. Across boundary transition zones, between high-density supercolonies and intact rainforest, yellow crazy ants reduced other ant species richness, occupied red crab burrows and killed resident red crabs, which was the trigger for ,invasional meltdown' on Christmas Island. The highly variable and unpredictable nature of A. gracilipes boundaries poses a challenge for incorporation into a predictive framework, as well as for their management. [source]


Survival, movement, and resource use of the butterfly Parnassius clodius

Abstract., 1. A mark,recapture study was conducted on the American Apollo butterfly Parnassius clodius Menetries during three field seasons (1998,2000) to examine its movement patterns over the course of a season within a sagebrush meadow in Grand Teton National Park, Wyoming, U.S.A. The study examined how resources affected butterfly distribution patterns and used mark,recapture data to gain insight into movement differences between sexes and over time. 2. The average straight-line movement of P. clodius was 202 m day,1, adjusted for sampling effort at different distances. Movement estimates in all 3 years were highly correlated with the average distance between plots sampled. 3. Butterfly abundance was correlated positively with per cent cover of its host plant Dicentra uniflora, but this relationship decreased in importance during the peak of the flight period when individuals may be more interested in finding mates. There was a weak, positive correlation between butterfly abundance and the abundance of its primary nectar source, Eriogonum umbellatum in 1999, but no relationship in 2000. 4. Survival, recapture, and transition probabilities were estimated using open population, capture,recapture models. Survival and recapture probability decreased over the course of each season, while the probability of moving between plots increased. Recapture probability was significantly lower for females than for males among all 3 years, but there was no difference between the sexes in survival rate. [source]


Natural-gradient tracer experiments in epikarst: a test study in the Acqua dei Faggi experimental site, southern Italy

Abstract Two natural-gradient tracer experiments were carried out using borehole fluorometers in order to characterize the internal structure of epikarstic horizons and analyze subsurface flow within these high-conductivity layers. The experiments were carried out in a test site in southern Italy where the epikarst is made up of an upper part with pervasive karstification and a lower part without pervasive karstification. Injection and observation boreholes were 6.9 m apart. An initial experiment demonstrated that wider (conduits) and narrower (fractures and bedding planes) openings coexist in a well-connected network within the lower epikarst. The adjusted aperture of the opening network (105 ,m) suggests that conduits are subordinately developed. The lower epikarstic horizon is hydraulically similar to granular porous media and Darcy's law can be applied to describe groundwater flow. A small value of longitudinal dispersivity (0.13 m) shows that variations in the velocity field in the direction of flow are less significant than those typical of carbonate systems at the same experiment scale. A second experiment demonstrated that longitudinal dispersivity (2.42 m) in the upper epikarst is in agreement with findings in other carbonates at the same experiment scale. However, despite the higher dispersivity and more pervasive karstification, the mean tracer velocity (3.7 m day,1) in the upper epikarst is slightly lower than the velocity in the lower epikarst (13.6 m day,1). Solute movement through intact columns of cryoturbated Upper Chalk

Abstract Cryoturbated Upper Chalk is a dichotomous porous medium wherein the intra-fragment porosity provides water storage and the inter-fragment porosity provides potential pathways for relatively rapid flow near saturation. Chloride tracer movement through 43 cm long and 45 cm diameter undisturbed chalk columns was studied at water application rates of 0·3, 1·0, and 1·5 cm h,1. Microscale heterogeneity in effluent was recorded using a grid collection system consisting of 98 funnel-shaped cells each 3·5 cm in diameter. The total porosity of the columns was 0·47 ± 0·02 m3 m,3, approximately 13% of pores were , 15 µm diameter, and the saturated hydraulic conductivity was 12·66 ± 1·31 m day,1. Although the column remained unsaturated during the leaching even at all application rates, proportionate flow through macropores increased as the application rate decreased. The number of dry cells (with 0 ml of effluent) increased as application rate decreased. Half of the leachate was collected from 15, 19 and 22 cells at 0·3, 1·0, 1·5 cm h,1 application rates respectively. Similar breakthrough curves (BTCs) were obtained at all three application rates when plotted as a function of cumulative drainage, but they were distinctly different when plotted as a function of time. The BTCs indicate that the columns have similar drainage requirement irrespective of application rates, as the rise to the maxima (C/Co) is almost similar. However, the time required to achieve that leaching requirement varies with application rates, and residence time was less in the case of a higher application rate. A two-region convection,dispersion model was used to describe the BTCs and fitted well (r2 = 0·97,0·99). There was a linear relationship between dispersion coefficient and pore water velocity (correlation coefficient r = 0·95). Characteristics of preferential flow and groundwater discharge to Shingobee Lake, Minnesota, USA

Abstract Small-scale heterogeneities and large changes in hydraulic gradient over short distances can create preferential groundwater flow paths that discharge to lakes. A 170 m2 grid within an area of springs and seeps along the shore of Shingobee Lake, Minnesota, was intensively instrumented to characterize groundwater-lake interaction within underlying organic-rich soil and sandy glacial sediments. Seepage meters in the lake and piezometer nests, installed at depths of 0·5 and 1·0 m below the ground surface and lakebed, were used to estimate groundwater flow. Statistical analysis of hydraulic conductivity estimated from slug tests indicated a range from 21 to 4·8 × 10,3 m day,1 and small spatial correlation. Although hydraulic gradients are overall upward and toward the lake, surface water that flows onto an area about 2 m onshore results in downward flow and localized recharge. Most flow occurred within 3 m of the shore through more permeable pathways. Seepage meter and Darcy law estimates of groundwater discharge agreed well within error limits. In the small area examined, discharge decreases irregularly with distance into the lake, indicating that sediment heterogeneity plays an important role in the distribution of groundwater discharge. Temperature gradients showed some relationship to discharge, but neither temperature profiles nor specific electrical conductance could provide a more convenient method to map groundwater,lake interaction. These results suggest that site-specific data may be needed to evaluate local water budget and to protect the water quality and quantity of discharge-dominated lakes. A simple model for the determination of the relative utilization efficiency of protein by blacklip abalone (Haliotis rubra Leach)

Abstract A 6-week growth rate trial of 6.7 mm ± 1.35 SD long blacklip abalone (Haliotis rubra) fed one of four graded protein feeds (26%, 32%, 37%, 45% crude protein, CP) resulted in a maximum average growth rate of 81 ,m day,1 (percentage daily weight gain of 5.2% day,1) with the 45% CP diet. Based on the results, a new, simple, non-destructive method to calculate the relative utilization efficiencies (RUE) of nutrients is presented to estimate the relative contributions of a nutrient to growth, body maintenance and the excess used for energy. Calculations of RUE of protein estimated that at the maximum growth rate, 59% of the CP was responsible for growth, while 31% was used for body maintenance and an excess of 10% was lost and presumably catabolized for energy and carbohydrate anabolism. At the %CP where the highest proportion of CP was used for growth, i.e. the maximum RUE of protein for growth (slightly higher at 62%), 35% was used for body maintenance and only 3% of the excess was lost to energy. Patterns of growth of juvenile pink abalone Haliotis corrugata fed re-hydrated natural feeds at a laboratory and a hatchery

Abstract The feasibility of obtaining a similar growth response from juvenile pink abalone Haliotis corrugata at a research laboratory and a hatchery, when using natural feeds was evaluated. Four macroalgae, Egregia menziesii, Eisenia arborea, Macrocystis pyrifera, Gracilaria sp., and the surfgrass Phyllospadix torreyi were used as feeds. Response patterns of abalone were very similar at both facilities in terms of final length, weight, and survival, varying from 13.0 to 15.50 mm, 0.31 to 0.52 g, and 60.0% to 78.6%, respectively. Better growth was obtained when E. menziesii, M. pyrifera, and Gracilaria sp. were offered. Poor results were obtained with surfgrass. The feed conversion ratio was determined at the laboratory and did not vary significantly, ranging from 42.3 to 199.0; although a significant inverse correlation was observed with growth rate. Growth in length and weight and survival rates varied within 1.6,20.2 ,m day,1, 0.123,1.664 mgday,1, 0.4,0.64%day,1, respectively. Mean growth rate in length (14.7,m day,1) and weight (1.18 mg day,1) at the hatchery were significantly higher than that obtained at the laboratory (9.4 ,m day,1 and 0.77 mg day,1), which is most likely a consequence of more suitable water temperature at the hatchery. Mean survival rate was significantly higher at the laboratory (53.1%day,1) than at the hatchery (46.1%day,1). [source]


Comparative performance of juvenile red abalone, Haliotis rufescens, reared in laboratory with fresh kelp and balanced diets

Abstract Juvenile Haliotis rufescens were reared in the laboratory in order to investigate the extent to which fresh kelp and formulated feeds with 250 g kg,1 (25P) and 380 g kg,1 protein content (38P) affected their growth rate, gut residence time (GRT), food consumption (C), food conversion ratio (FCR) and digestibility. Abalone from 38P attained the highest growth rate (70.5 ± 4.2 ,m day,1; 98.3 ± 6.95 ,g day,1), followed by 25P (47.9 ± 2.79 ,m day,1; 67.4 ± 2.82 ,g day,1) and kelp (23.6 ± 3.36 ,m day,1; 28.2 ± 4.11 ,g day,1). No significant differences were observed in consumption rate among treatments (0.61,0.68% body weight per day), yet kelp-fed abalone exhibited higher FCR (2.44), protein efficiency ratio (4.42), and apparent digestibility of dry matter (69.5%), protein (69.8%) and gross energy (79.2%) than 38P organisms (59.8, 62.4 and 62.2%, respectively). They also showed longer GRT (23.1 ± 0.93 h). This study demonstrated that formulated diets with 250 g kg,1 and 380 g kg,1 protein inclusion can sustain higher growth rates of juvenile H. rufescens than fresh algae. These differences seem to be due to the amount of dietary protein. Kelp meal appears to improve the consumption and digestibility of balanced diets, and its inclusion in formulated diets is recommended. [source]


Effect of antibiotic treatment on the growth and survival of juvenile northern Chilean scallop, Argopecten purpuratus Lamarck (1819), and associated microflora in experimental cultures

Abstract Juvenile northern Chilean scallops of 937±55 ,m shell height were exposed to five different concentrations of chloramphenicol (CHL) (5, 10, 25, 50 and 100 ,g mL,1), plus a control without antibiotics. To determine the effect of CHL on the accompanying microflora, the number of colony-forming units (CFU) that grew on TGE culture medium was counted in the seawater of containers with juveniles, and in containers with microalgae used as food. Both were exposed to the same concentrations of CHL. The growth rates of juveniles treated with CHL and the control without antibiotic showed highly significant differences (P=0.0001). The growth rate was inversely proportional to the CHL concentration. The control sample presented the highest growth rate (84.4±14.3 ,m day,1), followed by the sample treated with 5 ,g mL,1 (64.2±14.3 ,m day,1). The survival in the control and in the treated samples with 5,50 ,g mL,1 was rather high, with a mean value of 95%. Only the sample treated with 100 ,g mL,1 had a low survival (36.7%). The CFU count was larger in the containers with juveniles, when compared with the ones with food. The CFU count tended to decrease with increasing CHL concentration in the juveniles. [source]


Effects of the initial size, stocking density and sorting on the growth of juvenile Pacific abalone, Haliotis discus hannai Ino

Abstract During winter months, a novel overwintering mode of transferring juvenile abalones to open seawaters in southern China rather than keeping them in closed land-based nursery systems in northern China is a popular practice. The initial size, stocking density and sorting are among the first considerations when establishing an abalone culture system. This study aimed to investigate the effects of these factors on the growth of juvenile Pacific abalone, Haliotis discus hannai Ino, during overwintering. Juvenile abalones were reared in multi-tier basket form for overwintering in open seawaters in southern China for 106 days. The daily growth rates (DGRs) in the shell length of all experimental groups ranged from 67.08 to 135.75 ,m day,1, while the specific growth rates (SGRs) were 0.2447,0.3259% day,1. Variance analysis indicated that both DGRs and SGRs in shell length were significantly affected by the initial body size and stocking density. Furthermore, the effects of stocking density on DGRs and SGRs varied with the initial size. However, sorting abalones according to their initial sizes may not be necessary in practice as sorting did not alter growth significantly at all densities in this study. Factors potentially affecting abalone growth such as genetic control and intraspecific competition were discussed. [source]
