Selected AbstractsMyocardial Performance Index (Tei Index) Does Not Reflect Long-Term Changes in Left Ventricular Function after Acute Myocardial InfarctionECHOCARDIOGRAPHY, Issue 1 2003Torstein Hole M.D. Aims: To evaluate whether changes in myocardial performance index (MPI or Tei index) were related to changes in other Doppler echocardiographic parameters after acute myocardial infarction, or had any independent prognostic impact in a 2-year observational study. Methods and Results: Seventy-one patients with acute myocardial infarction without heart failure were examined at baseline, 3 months, and 2 years. MPI was significantly related to end-diastolic and end-systolic volume indexes, ejection fraction, maximal velocity, and time velocity integral of early mitral filling wave at 3 months and 2 years. MPI did not contribute significantly to the prediction of any changes in the measures of diastolic or systolic function at 3 months or 2 years. Baseline MPI was significantly higher in patients who later developed heart failure(0.55 ± 0.16)than in other patients(0.43 ± 0.13, P = 0.006), but had no independent predictive power for the development of heart failure or death relative to end-systolic volume index and deceleration time of early mitral filling wave. Conclusion: MPI did not accurately reflect changes in Doppler and two-dimensional echocardiographic measures of diastolic or systolic function during a 2-year follow-up after acute myocardial infarction, and did not have any independent prognostic impact. (ECHOCARDIOGRAPHY, Volume 20, January 2003) [source] Long-term Changes in Postnatal Susceptibility to Pilocarpine-induced Seizures in Rats Exposed to Gamma Radiation at Different Stages of Prenatal DevelopmentEPILEPSIA, Issue 10 2003Zuzanna Setkowicz Summary:,Purpose: To determine whether brains irradiated at different stages of prenatal development also have different postnatal susceptibility to seizures evoked by pilocarpine. Methods: Pregnant Wistar rats were exposed to a single 1.0-Gy dose of gamma rays on gestation days 13, 15, 17, or 19 (E13, E15, E17, and E19, respectively). On postnatal day 60, their offspring received i.p. pilocarpine injections to evoke status epilepticus. Behavior of the animals was observed continuously for 6 h after the injection, and motor manifestations of seizure activity were rated, and survival times recorded. After 7-day survival, the animals were killed, and their brains were weighed. Results: The average brain weight of animals exposed to irradiation at earlier prenatal stages (E13 or E15) was significantly lower than that after irradiation on E17 or E19. However, effects of the irradiation on the susceptibility to pilocarpine-induced seizures were quite opposite. The intensity of status epilepticus evoked in rats irradiated on E13 or E15 was significantly lower than that in nonirradiated controls or in those irradiated on E17 or E19. Moreover, after irradiation on E13 or E15, survival of the animals was significantly higher in relation not only to other irradiated groups but also to the controls. Conclusions: The results suggest than the extent of neuronal deficit, even if relatively greater, cannot always lead to higher susceptibility of the dysplastic brain to seizures. Functional consequences of the deficit, even if its magnitude is relatively smaller but involving specific brain areas, appear to be critical for the epileptogenesis. [source] Long-term change to fish assemblages and the flow regime in a southeastern U.S. river system after extensive aquatic ecosystem fragmentationECOGRAPHY, Issue 6 2008Christopher M. Taylor The upper Tombigbee River in northeastern Mississippi now exists as a fragment, confluencing with and fed by an extensively modified aquatic landscape now called the Tennessee-Tombigbee Waterway (TTW). We examined the changes to fish assemblages and flow regime after waterway construction based on contemporary comparisons to historical fish collections and discharge data. The river's flow regime has changed markedly since TTW construction. Analysis of discharge data from two stations for 15 years, pre- and post-waterway, indicated significant differences in flow regime including increased minimum and base flows, lower spring and higher late summer-autumn flows, and lower high flow durations, post-TTW. These changes corresponded to significantly reduced regional and local species richness, and strong shifts in fish assemblage structure across a 20 yr time span. Post-waterway fish assemblages were related strongly to measured environmental variables characterizing local habitats. Several lentic-adapted species increased their abundances in lower reaches of the river, including a recent invader to the TTW system, the Mississippi silverside Menidia audens. Fragmentation of river ecosystems via disruption to hydrologic regimes is a major threat to aquatic biodiversity worldwide. Because the flow regime of this fragmented river is in part controlled by waterway operations via five minimum flow control structures, adaptive conservation and management efforts could be implemented in order to maintain and potentially restore the natural flow regime and the ecological integrity of the system. [source] Breeding biology and breeding success of the Lesser Grey Shrike Lanius minor in a stable and dense populationIBIS, Issue 2 2000ANTON KRISTIN The Lesser Grey Shrike Lanius minor is highly endangered throughout Europe, having declined markedly in abundance and range. Long-term changes in climate and agricultural practices have been identified as the main reasons for its decline. To determine which factors influence short-term changes in breeding success, we examined several aspects of its breeding biology. Our investigation revealed that our study area bears a large and stable population of this species. In 1996 and 1997, we recorded 84 and 77 breeding pairs in an area of 20 km2, with an average of 4.20 and 3.85 pairs/km2 respectively. Data on breeding density, clutch size and fledging success from 1989 to 1997 (excluding 1992) indicate a stable breeding population with a constant high breeding success. Reproductive success declined through the season, mainly through seasonal variation in clutch size rather than chick mortality. However, breeding success was generally high (69% and 79% of the nests produced chicks], with low hatching failure and few nest losses. The main cause of breeding failure was nest predation (at least 50% of nest losses), mainly by magpies (at least 66% of depredated nests). Although in this population the Lesser Grey Shrike tends to aggregate in clusters, breeding density had no obvious effect on breeding success and nest predation. [source] Long-term changes in the abundance of flying insectsINSECT CONSERVATION AND DIVERSITY, Issue 4 2009CHRIS R. SHORTALL Abstract., 1.,For the first time, long-term changes in total aerial insect biomass have been estimated for a wide area of Southern Britain. 2.,Various indices of biomass were created for standardised samples from four of the Rothamsted Insect Survey 12.2 m tall suction traps for the 30 years from 1973 to 2002. 3.,There was a significant decline in total biomass at Hereford but not at three other sites: Rothamsted, Starcross and Wye. 4.,For the Hereford samples, many insects were identified at least to order level, some to family or species level. These samples were then used to investigate the taxa involved in the decline in biomass at Hereford. 5.,The Hereford samples were dominated by large Diptera, particularly Dilophus febrilis, which showed a significant decline in abundance. 6.,Changes in agricultural practice that could have contributed to the observed declines are discussed, as are potential implications for farmland birds, with suggestions for further work to investigate both cause and effect. Schlünzen Abstract Climate changes and the urban climate of the ,green city' Hamburg and its metropolitan region are analyzed using observational data for temperature and precipitation. Values for Hamburg's synoptic site HH-Fuhlsbüttel start in 1891 and are used to determine climate changes. Additional data from up to 45 climate stations are used to analyze the different aspects of the regional climate and urban effects on the temperature [urban heat island (UHI)] and precipitation [urban precipitation impact (UPI)]. The analysis of the long-term data shows that the climate has already changed. Annual precipitation significantly increases ,0.8 mm/year when focusing on years 1891,2007 and ,1.3 mm/year for 1948,2007. Precipitation increases are largest in November through March and March as well as June for 1978,2007. For April and July of this period, a precipitation decrease is found. The precipitation distribution shows that moderate daily precipitation amounts (,10 mm/day) have increased by about 10% between 1948,1977 and 1978,2007. Precipitation amounts > 10 mm/day have increased by 20% in the same period. Average temperatures significantly increase by 0.07 K/decade (1891,2007), 0.19 K/decade (1948,2007), 0.6 K/decade (1978,2007) with largest significant increases in fall. For the UHI, it is found that the average temperature is higher up to 1.1 K in the densely build-up city area than outside. Values are about halved for more green urban areas but also depend on more local impacts. The minimum temperatures are up to 3 K higher and maximum temperatures slightly lower in the inner city than in the rural during summer. The winter temperatures are higher throughout the urban area. The UHI differences depend on wind speed; this dependence is best described by using the inverse square root of the wind speed. Classification using different wind directions shows that the precipitation is significantly higher (5,20%) for downwind of urban areas compared with the upwind side. Krzy Abstract Decadal changes of ozone mini-hole event appearance over the Northern Hemisphere midlatitudes are examined based on daily total ozone data from seven stations having long records (four decades or more) of ozone observations. The various threshold methods for accepting and rejecting the ozone minima as mini-holes are examined. Mini-hole event activity is seen to be rather stable when averaged over a decadal time scale if the mini-holes are selected as large negative departures (exceeding 20%) relative to the moving long-term total ozone reference. The results are compared with a previous ozone mini-hole climatology derived from satellite data (TOMS measurements on board the Nimbus-7 satellite for the period 1978,93). A nonlinear statistical model (MARS), which takes into account various total ozone dynamical proxies (from NCEP,NCAR reanalysis), is used to study dynamical factors responsible for the ozone extremes over Arosa in the period 1950,99. The model explains as much as 95% of the total variance of the ozone extremes. The model,observation differences averaged over the decadal intervals are rather smooth throughout the whole period analysed. It is suggested that the short-term dynamical processes controlling the appearance of ozone extremes influenced the ozone field in a similar way before and after the onset of abrupt ozone depletion in the early 1980s. The analysis of the ozone profile and the tropopause pressure (from the ozonesondings over Hohenpeissenberg, 1966,99) during mini-hole events shows ,60% ozone reduction in the lower stratosphere and an approximately 50 hPa upward shift of the thermal tropopause there. [source] Long-term changes in distribution and chemistry of middle Miocene to Quaternary volcanism in the Chokai-Kurikoma area across the Northeast Japan ArcISLAND ARC, Issue 1 2004Hirofumi Kondo Abstract To understand the characteristics of long-term spatial and temporal variation in volcanism within a volcanic arc undergoing constant subduction since the cessation of back-arc opening, a detailed investigation of middle Miocene to Quaternary volcanism was carried out within the Chokai-Kurikoma area of the Northeast Japan Arc. This study involved a survey of available literature, with new K,Ar and fission track dating, and chemical analyses. Since 14 Ma, volcanism has occurred within the Chokai-Kurikoma area in specific areas with a ,branch-like' pattern, showing an east,west trend. This is in marked contrast to the widespread distribution of volcanism with a north,south trend in the 20,14 Ma period. The east,west- trending ,branches' are characterized by regular intervals (50,100 km) of magmatism along the arc. These branches since 14 Ma are remarkably discrepant to the general northwest,southeast or north-northeast,south-southwest direction of the crustal structures that have controlled Neogene to Quaternary tectonic movements in northeast Japan. In addition, evidence indicating clustering and focusing of volcanism into smaller regions since 14 Ma was verified. Comparison of the distribution and chemistry of volcanic rocks for three principal volcanic stages (11,8, 6,3 and 2,0 Ma) revealed that widely but sparsely distributed volcanic rocks had almost the same level of alkali and incompatible element concentrations throughout the area (with the exception of Zr) in the 11,8 Ma stage. However, through the 6,3 Ma stage to the 2,0 Ma stage, the concentration level in the back-arc cluster increased, while that in the volcanic front cluster remained almost constant. Therefore, the degree of partial melting has decreased, most likely with a simultaneous increase in the depth of magma segregation within the back-arc zone, whereas within the volcanic front zone, the conditions of magma generation have changed little over the three stages. In conclusion, the evolution of the thermal structure within the mantle wedge across the arc since 14 Ma has reduced the extent of ascending mantle diapirs into smaller fields. This has resulted in the tendency for the distribution of volcanism to become localized and concentrated into more specific areas in the form of clusters from the late Miocene to Quaternary. Pinnegar Summary 1The intensive exploitation of fish communities often leads to substantial reductions in the abundance of target species, with ramifications for the structure and stability of the ecosystem as a whole. 2We explored changes in the mean trophic level of the Celtic Sea (ICES divisions VII f,j) fish community using commercial landings, survey data and estimates of trophic level derived from the analysis of nitrogen stable isotopes. 3Our analyses showed that there has been a significant decline in the mean trophic level of survey catches from 1982 to 2000 and a decline in the trophic level of landings from 1946 to 1998. 4The decline in mean trophic level through time resulted from a reduction in the abundance of large piscivorous fishes and an increase in smaller pelagic species which feed at a lower trophic level. 5Similar patterns of decline in the trophic level of both catches and landings imply that there have been substantial changes in the underlying structure of the Celtic Sea fish community and not simply a change in fishery preferences. 6We suggest that the reported changes in trophic structure result from reductions in the spawning stock biomass of traditional target species associated with intensive fishing, together with long-term climate variability. 7The relative distribution of fish market prices has changed significantly over the past 22 years, with high trophic level species experiencing greater price rises than lower trophic level species. 8Although decreased abundance of high trophic level species will ultimately have negative economic consequences, the reduction in mean trophic level of the fish community as a whole may allow the system to sustain higher fishery yields. 9Management objectives in this fishery will depend on the relative values that society attaches to economic profit and protein production. [source] Long-term changes in the chromosomal inversion polymorphism of Drosophila subobscura.JOURNAL OF ZOOLOGICAL SYSTEMATICS AND EVOLUTIONARY RESEARCH, Issue 3 2004Abstract The chromosomal polymorphism of 13 European populations of Drosophila subobscura has been compared with that of the same populations collected 15,35 years ago. The chromosomal polymorphism of the old populations differs significantly from that of the new populations, mainly for chromosomes U and O. There is a very good agreement between the geographical space and the genetic space as shown by a graphical representation of the 26 statistical populations (13 old and 13 new) obtained by a principal coordinate analysis. This reflects both the existence of significant latitudinal clines for the frequencies of some chromosomal arrangements in the old and new samples and systematic changes that have taken place in these populations during the period that elapsed between the two surveys. An increase in the frequency of those arrangements typical of southern latitudes and a decrease for those common in northern latitudes is observed in all populations , Mediterranean, Atlantic and Central European. Furthermore, the genetic distances of the new populations to a southern population of reference have decreased in comparison with those of the old populations. These changes could be the result of climatic factors that are correlated with latitude. In particular, the assumption that global warming is responsible for all the changes observed appears rather likely. Whether these systematic changes of the chromosomal polymorphism are a consequence of local adaptations or have been produced by migration from the south remains an open question. Zusamennfassung Der gegenwärtige chromosomale Inversionspolymorphismus in 13 europäischen Populationen von Drosophila subobscura wurde mit dem Zustand in denselben Populationen verglichen, die vor 15 bis 35 Jahren untersucht worden waren. Der Chromosomenpolymorphismus der ,,alten'' Populationen unterscheidet sich signifikant von dem der ,,neuen'' Populationen, besonders für die Chromosomen U und O. Eine sehr gute Übereinstimmung zeigt sich zwischen der geographischen Komponente und der genetischen Komponente bei einer graphischen Darstellung der 26 Populationen (13 alte und 13 neue Populationen) durch eine Koordinatenanalyse. Das spiegelt beides wider, die Existenz von signifikanten breitenabhängigen Klinen in der Häufigkeit einiger chromosomaler Strukturtypen in den alten und den neuen Populationen, aber auch die systematischen Änderungen, die sich bei diesen Populationen im Laufe der Zeit zwischen den beiden Untersuchungen ereignet haben. [source] Sediment phosphorus characteristics in the clearwater state of Lake Mogan, TurkeyLAKES & RESERVOIRS: RESEARCH AND MANAGEMENT, Issue 3 2008Serap Pulatsü Abstract This study examined the vertical distributions of total phosphorus (TP) and phosphorus fractions, and the iron and organic matter, in the littoral sediment in a macrophyte-dominated, clearwater state in Lake Mogan between September 2005 and August 2006. Benthic macroinvertebrates and total bacteria in the sediment also were determined. No clear seasonal or depth-related (0,20 cm) patterns were found in sediment concentrations for the measured parameters. The phosphorus release was quantitatively very low, and a negative phosphorus release (,0.132 µg m,2 day,1) was measured during the summer months. The TP concentrations of the sediment samples ranged between 675.00 and 1463.80 µg g,1 dry weight (DW), and the trophic level of the lake was eutrophic. On average, inorganic phosphorus fractions comprised the largest fraction (63%), while organic-bound phosphorus (Org , P) constituted 37% of the TP in Lake Mogan. The most important phosphorus-immobilizing factors are high iron content (14 200,47 750 µg g,1 DW), the sediment's clay content (47.80,51.80%), and an abundance of macrophytes at the sampling station. The low abundance of benthic macroinvertebrates (510,850 individuals m,2), which depend on sediments with high iron and low organic matter (5.42,13.30%), played a role in the sediment phosphorus retention. Although bacterial abundance in the surficial sediment appeared to be positively correlated to temperature, the overlying water did not experience anoxic conditions, supporting a state in which bacteria were able to retain phosphorus in their cell structures. Long-term changes in the sediments of Lake Mogan must be monitored lake. In order to optimize the management of the lake, and to determine the longevity of a clearwater state following management measures and continued external phosphorus loading, long-term changes in the sediments of Lake Morgan must be monitored. [source] Long-term changes of aphid vectors of Barley yellow dwarf viruses in north-eastern Italy (Friuli-Venezia Giulia)ANNALS OF APPLIED BIOLOGY, Issue 1 2009P.G. Coceano Abstract Migrations of aphid vectors of Barley yellow dwarf viruses (BYDV) were monitored using a Rothamsted Insect Survey suction trap in Friuli-Venezia Giulia (north-eastern Italy). Catches from 1983 to 2002 were studied for trends, correlations of total catches of each year with those of previous years, correlations between the autumn and the spring + summer catches of the same year and between spring + summer catches of one year with catches of the previous autumn. Infectivity of autumn alates was studied using biological tests, and infectivity indexes were calculated for all vector species and for Rhopalosiphum padi alone. Colonisation of barley and proportion of infected plants were checked in a field close to the suction trap from 1992 to 2002 and related to trap catches. Catches were also correlated to acreage dedicated to cereal and fodder crops in the region. During the 20 years, 15 BYDV vector species were caught in the trap, but only five species were found consistently colonising barley plants during autumn. R. padi was the most numerous species in catches, while Sitobion avenae was the predominant colonising species in the barley field. Relatively to R. padi, S. avenae colonies were about six times more numerous than expected from catches. The yearly abundance of catches of most species did not change significantly during the 20 years, with a few exceptions, significantly correlated to changes in the acreage dedicated to cereal and fodder crops. There was a significant decrease of the autumn catches of both R. padi and the total of BYDV vectors. [source] Inter-decadal variation of the summer precipitation in East China and its association with decreasing Asian summer monsoon.INTERNATIONAL JOURNAL OF CLIMATOLOGY, Issue 9 2008Part I: Observed evidences Abstract In recent two decades, North and Northeast China have suffered from severe and persistent droughts while the Yangtze River basin and South China have undergone much more significant heavy rainfall/floods events. This long-term change in the summer precipitation and associated large-scale monsoon circulation features have been examined by using the new dataset of 740 surface stations for recent 54 years (1951,2004) and about 123-yr (1880,2002) records of precipitation in East China. The following new findings have been highlighted: (1) One dominating mode of the inter-decadal variability of the summer precipitation in China is the near-80-yr oscillation. Other modes of 12-yr and 30,40-yr oscillations also play an important role in affecting regional inter-decadal variability. (2) In recent 54 years, the spatial pattern of the inter-decadal variability of summer precipitation in China is mainly structured with two meridional modes: the dipole pattern and the positive-negative-positive ("+ , + " pattern). In this period, a regime transition of meridional precipitation mode from "+ , + " pattern to dipole pattern has been completed. In the process of southward movement of much precipitation zone, two abrupt climate changing points that occurred in 1978 and 1992, respectively, were identified. (3) Accompanying the afore-described precipitation changes, the East Asian summer monsoon have experienced significant weakening, with northward moisture transport and convergence by the East Asian summer monsoon greatly weakened, thus leading to much deficient moisture supply for precipitation in North China. (4) The significant weakening of the component of the tropical upper-level easterly jet (TEJ) has made a dominating contribution to the weakening of the Asian summer monsoon system. The cooling in the high troposphere at mid- and high latitudes and the possible warming at low latitude in the Asian region is likely to be responsible for the inter-decadal weakening of the TEJ. Hunt Summary 1Population modelling and field measurements of births, growth and deaths were used to investigate the long-term change in abundance of Atriplex vesicaria (Chenopodiaceae), a long-lived, palatable, perennial shrub, under sheep grazing. Of particular interest was whether A. vesicaria is at risk of being eliminated throughout grazed paddocks when the recommended practice of continuous grazing at conservative stocking rates is employed. 2Time-invariant matrix population models indicated that the A. vesicaria population was in decline over much of the study paddock, but the rate of decline was greatest nearer to the water point (population growth rate , , 0·8). Time-varying stochastic matrix models projected that the A. vesicaria population would become locally extinct at most sites up to approximately 2200 m from water, occurring first closer to water (within 12,29 years). The population was stable (i.e. , , 1) at sites greater than 2200 m from water over the projection period of 100 years. 3Decreases in adult survival and recruitment made the largest contributions to reductions in the population growth rate. However, there were spatial patterns centred on the water point in the degree to which particular demographic processes contributed to these reductions, because of a grazing gradient and the differential sensitivity of demographic processes to grazing. Thus decreases in recruitment contributed to reductions in the population growth rate at greater distances. Such responses, together with the sensitivity of the population growth rate to these processes, determined the spatial pattern in population growth. 4The results suggest that piospheres (i.e. the zone of impact) continue to expand over many years under set-stocking so that the area around the water point that is devoid of A. vesicaria becomes larger. The process of expansion appears to first involve the inhibition of recruitment, followed by eventual mortality of established shrubs. 5The large contribution of adult survival to the population growth rate in A. vesicaria suggests that minimizing the mortality of established adults should be a priority for management. This is likely to involve resting from grazing at critical times such as during extended dry periods. This may also permit increased levels of recruitment during subsequent moister periods. [source] The Logarithmic-To-Linear Shift: One Learning Sequence, Many Tasks, Many Time ScalesMIND, BRAIN, AND EDUCATION, Issue 3 2009Robert S. Siegler ABSTRACT The relation between short-term and long-term change (also known as learning and development) has been of great interest throughout the history of developmental psychology. Werner and Vygotsky believed that the two involved basically similar progressions of qualitatively distinct knowledge states; behaviorists such as Kendler and Kendler believed that the two involved similar patterns of continuous growth; Piaget believed that the two were basically dissimilar, with only development involving qualitative reorganization of existing knowledge and acquisition of new cognitive structures. This article examines the viability of these three accounts in accounting for the development of numerical representations. A review of this literature indicated that Werner's and Vygotsky's position (and that of modern dynamic systems and information processing theorists) provided the most accurate account of the data. In particular, both changes over periods of years and changes within a single experimental session indicated that children progress from logarithmic to linear representations of numerical magnitudes, at times showing abrupt changes across a large range of numbers. The pattern occurs with representations of whole number magnitudes at different ages for different numerical ranges; thus, children progress from logarithmic to linear representations of the 0,100 range between kindergarten and second grade, whereas they make the same transition in the 0,1,000 range between second and fourth grade. Similar changes are seen on tasks involving fractions; these changes yield the paradoxical finding that young children at times estimate fractional magnitudes more accurately than adults do. Several different educational interventions based on this analysis of changes in numerical representations have yielded promising results. Smith Abstract 1.Historical data are often one of the only resources available for documenting and assessing causes of environmental change, particularly in developing regions where funding for ecological studies is limited. In this paper, previously unpublished data from a year-long study (1977) of the fish community of the Espiritu Santo estuary are presented. This dataset is among the oldest and most extensive surveys of a Caribbean island estuarine fish community. 2.A comparison of these historical data with data collected in June and July 2004 using identical sampling methods allowed description of potential long-term changes in the fish community, identification of vulnerable species, and assessment of potential drivers of change. 3.Results strongly suggest a decline in species richness and abundance in the Espiritu Santo estuarine fish community, with greater declines in freshwater-tolerant than marine or euryhaline species. Declines in freshwater inflow to the estuary, due to large-scale upstream water abstractions for municipal use, have increased since the initial 1977 survey. 4.This is the first study to examine long-term change in the fish community of a tropical island estuary. The developing organisms including rodents, fish and amphibians are particularly sensitive to exposure to estrogenic chemicals during a critical window. Exposure to estrogens during the critical period induces long-term changes in reproductive as well as non-reproductive organs, including persistent molecular alterations. The antenatal mouse model can be utilized as an indicator of possible long-term consequences of exposure to exogenous estrogenic compounds including possible environmental endocrine disrupters. Many chemicals released into the environment potentially disrupt the endocrine system in wildlife and humans, some of which exhibit estrogenic activity by binding to the estrogen receptors. Estrogen responsive genes, therefore, need to be identified to understand the molecular basis of estrogenic actions. In order to understand molecular mechanisms of estrogenic chemicals on developing organisms, we are identifying estrogen responsive genes using cDNA microarray, quantitative RT-PCR, and differential display methods, and genes related to the estrogen-independent vaginal changes in mice induced by estrogens during the critical window. In this review, discussion of our own findings related to endocrine distuptor issue will be provided. Symons Abstract People with mental retardation, autism, and related developmental disabilities who self-injure are treated with a wide array of behavioral techniques and psychotropic medications. Despite numerous reports documenting short-term and some long-term changes in self-injury associated with the opiate antagonist naltrexone hydrochloride, no quantitative review of its efficacy has been reported. We conducted a quantitative synthesis of the peer-reviewed published literature from 1983 to 2003 documenting the use of naltrexone for the treatment of self-injurious behavior (SIB). Individual-level results were analyzed given subject and study characteristics. A sample of 27 research articles involving 86 subjects with self-injury was reviewed. Eighty percent of subjects were reported to improve relative to baseline (i.e., SIB reduced) during naltrexone administration and 47% of subjects SIB was reduced by 50% or greater. In studies reporting dose levels in milligrams, males were more likely than females to respond. No significant relations were found between treatment outcomes and autism status or form of self-injury. Rosenfalck SUMMARY Aim To evaluate the long-term effect of changes in body composition induced by weight loss on insulin sensitivity (SI), non-insulin mediated glucose disposal, glucose effectiveness (SG) and ,-cell function. Design Glucose metabolism was evaluated before and after participation in a two-year weight loss trial of Orlistat vs. placebo, combined with an energy and fat restricted diet. Subjects Twelve obese patients (11 women, 1 man), age 45.8 ± 10.5 years, body weight (BW) 99.7 ± 13.3 kg, BMI 35.3 ± 2.8 kg/m2. Measurements At inclusion and 2 years later an oral glucose tolerance test (OGTT) and a frequently sampled intravenous glucose tolerance test (FSIGT) were performed. Body composition was estimated by a dual-energy X-ray absorptiometry (DXA) whole body scanning. Results The patients obtained varying changes in BW ranging from a weight loss of 17.8 kg to a weight gain of 6.0 kg. Corresponding changes in fat mass (FM) varied from a 40% reduction to a 19% increase. A significant decrease in both fasting (p =,0.038) and 2 h (p =,0.047) blood glucose at OGTT was found. The improvement in insulin sensitivity (SI) estimated by means of Bergmans Minimal Model, was significantly and linearly correlated to change in total FM (r = , 0.83, p =,0.0026). A multiple regression analysis showed that changes in truncal FM was the strongest predictor of change in SI explaining 67% of the variation. First phase insulin response (AIRg) remained unchanged whereas insulin disposition index increased significantly (p =,0.044). At inclusion five patients had impaired glucose tolerance of which four, who lost weight, were normalized at the retest 2 years later. Conclusion In obese subjects long-term minimal or moderate changes in weight were found to be linearly associated with changes in insulin sensitivity. In obese subjects with impaired glucose tolerance even a minor weight loss was able to normalize glucose tolerance. CLEMENS Abstract The substituted amphetamines 3,4-methylenedioxymethamphetamine (MDMA, ,Ecstasy') and methamphetamine (METH, ,ice', ,speed') are increasingly popular drugs amongst party-drug users. Studies with humans have investigated the acute and possible long-term adverse effects of these drugs, yet outcomes of such studies are often ambiguous due to a variety of confounding factors. Studies employing animal models have value in determining the acute and long-term effects of MDMA and METH on brain and behaviour. Self-administration studies show that intravenous METH is a particularly potent reinforcer in rats and other species. In contrast, MDMA appears to have powerful effects in enhancing social behaviour in laboratory animals. Brief exposure to MDMA or METH may produce long-term reductions in dopamine, serotonin and noradrenaline in the brain and alterations in the density of various receptor and transporter proteins. However it is still unclear, particularly in the case of MDMA, whether this reflects a ,neurotoxic' effect of the drug. Lasting alterations in social behaviour, anxiety, depressive symptoms and memory have been demonstrated in laboratory rats given MDMA or METH and this matches long-term changes reported in some human studies. Recent laboratory studies suggest that MDMA/METH combinations may produce greater adverse neurochemical and behavioural effects than either drug alone. This is of some concern given recent evidence that party drug users may be frequently exposed to this combination of drugs. [source] Response of collembolan communities to land-use change and grassland successionECOGRAPHY, Issue 2 2007Matthieu Chauvat This study focuses on the long-term changes of collembolan communities occurring after the conversion of arable land to managed grassland. We analysed collembolan communities at grassland sites of different age that had been gradually converted over a period of 50 yr. Abundance and biomass responded rapidly and very positively to the conversion of arable land to grassland, while species richness was not affected. Collembolan assemblages changed only little during grassland maturation. The impact of land-use change on community structure was more obvious at the functional level because the colonization processes observed in our study mostly relied on hemiedaphic species. Vegetation and soil parameters were good predictors of collembolan community structure during development of managed grassland. The present study demonstrated that past landscape patterns and processes like land-use conversion and subsequent succession had a considerable impact on the present day pattern of species richness and community composition of Collembola within a landscape. Our results strongly differ from those obtained for other invertebrate groups, highlighting on the one hand the very diverse reactions of invertebrates to a common factor, and on the other hand the need to survey more than one taxa in order to draw conclusions on effects of land-use change on faunistic communities. Design Prospective cohort: British Regional Heart Study. Setting General practices in 24 towns in England, Wales and Scotland. Participants Non-smoking men: 2125 studied at baseline [questionnaire (Q1): 1978,80, aged 40,59 years], 3046 studied 20 years later (Q20: 1998,2000, aged 60,79 years) and 1208 studied at both times. Non-smokers were men reporting no current smoking with cotinine < 15 ng/ml at Q1 and/or Q20. Measurements Serum cotinine to assess ETS exposure. Findings In cross-sectional analysis, geometric mean cotinine level declined from 1.36 ng/ml [95% confidence interval (CI): 1.31, 1.42] at Q1 to 0.19 ng/ml (95% CI: 0.18, 0.19) at Q20. The prevalence of cotinine levels , 0.7 ng/ml [associated with low coronary heart disease (CHD) risk] rose from 27.1% at Q1 to 83.3% at Q20. Manual social class and northern region of residence were associated with higher mean cotinine levels both at Q1 and Q20; older age was associated with lower cotinine level at Q20 only. Among 1208 persistent non-smokers, cotinine fell by 1.47 ng/ml (95% CI: 1.37, 1.57), 86% decline. Absolute falls in cotinine were greater in manual occupational groups, in the Midlands and Scotland compared to southern England, although percentage decline was very similar across groups. Conclusions A marked decline in ETS exposure occurred in Britain between 1978 and 2000, which is likely to have reduced ETS-related disease risks appreciably before the introduction of legislation banning smoking in public places. [source] Governing long-term social,ecological change: what can the adaptive management and transition management approaches learn from each other?ENVIRONMENTAL POLICY AND GOVERNANCE, Issue 1 2009Timothy J. Foxon Abstract Maintaining social welfare and opportunity in the face of severe ecological pressures requires frameworks for managing and governing long-term social,ecological change. In this paper we analyse two recent frameworks, adaptive management and transition management, outlining what they could learn from each other. Though usually applied in different domains, the two conceptual frameworks aim to integrate bottom-up and top-down approaches, and share a focus on the ability of systems to learn and develop adaptive capacity whilst facing external shocks and long-term pressures. Both also emphasize learning from experimentation in complex systems, but transition management focuses more on the ability to steer long-term changes in system functions, whilst adaptive management emphasizes the maintenance of system functions in the face of external change. The combination of iterative learning and stakeholder participation from adaptive management has the potential to incorporate vital feedbacks into transition management, which in turn offers a longer-term perspective from which to learn about and manage socio-technical and social,ecological change. It is argued that by combining insights from both frameworks it may be possible to foster more robust and resilient governance of social,ecological systems than could be achieved by either approach alone. The paper concludes by critically reflecting upon the challenges and benefits of combining elements of each approach, as has been attempted in the methodology of a research project investigating social,ecological change in UK uplands. Methods: We performed Western blot analysis of signaling proteins in PSDs isolated from cortical tissue. Results: Seizures induce a strong upregulation of TrkB, the receptor for brain-derived neurotrophic factor (BDNF), whereas components of the N -methyl- d -aspartate (NMDA)-receptor complex are downregulated in both human and rat PSDs. Conclusions: These data show that long-term changes in PSD composition occur as a consequence of epileptic seizure activity. [source] Display Plasticity in Response to a Robotic Lizard: Signal Matching or Song Sharing in Lizards?ETHOLOGY, Issue 10 2006C. Brian Smith Many territorial songbirds alter the structure of their songs after listening to and interacting repeatedly with the same neighbors. Here, we use a robotic lizard to test for similar learned changes in signal structure in male Sagebrush lizards, Sceloporus graciosus. Subjects were exposed to two types of headbob displays (species-typical and unusual) both in short-term tests and in repeated exposures for 10 d. We found no evidence for immediate changes in signal structure to match a particular opponent (signal matching) or long-term changes after repeated exposure (,song' sharing). If anything, the lizards' displays became less like that of the robotic stimulus over time. Further tests of other taxa are needed to identify the evolutionary forces that lead to these forms of behavioral plasticity and to determine whether song sharing and signal matching are unique characteristic of songbirds. Lizards also became more agitated and produced more highly aggressive displays of their own when confronted with headbob displays that violated the basic syntactic structure of their display system, confirming that they were paying attention to subtle differences in display structure despite the artificial nature of the treatments. Thus, our study also adds to the growing evidence supporting the use of robotic playbacks to study animal communication. [source] PRECLINICAL STUDY: Atypical development of behavioural sensitization to 3,4-methylenedioxymethamphetamine (MDMA, ,Ecstasy') in adolescent rats and its expression in adulthood: role of the MDMA chiralityADDICTION BIOLOGY, Issue 1 2010Nora Von Ameln ABSTRACT Despite the great popularity of 3,4-methylenedioxymethamphetamine (MDMA, Ecstasy) as a drug of abuse, not much is known about the detailed mechanisms of the acute and subchronic effects of the drug. There is especially a lack of information about the distinct behavioural effects of its optical isomers (enantiomers) R- and S-MDMA compared with the racemic RS-MDMA. For this purpose, adolescent rats were repetitively treated during two treatment stages (stage 1: days 1,10; stage 2: days 15, 17, 19) with RS-MDMA (5 or 10 mg/kg) or each of the respective enantiomers (5 mg/kg). The repeated treatment started on postnatal day (PND) 32 and locomotor activity was measured on each day by means of a photobeam-equipped activity box system. RS-MDMA or S-MDMA administration led acutely to massive hyperlocomotion and subchronically, to the development of behavioural sensitization after a short habituation period. R-MDMA was free of hyperactivating effects and even decreased locomotor behaviour upon repeated treatment. Nevertheless, co-administration of R-MDMA increased the hyperactivity of S-MDMA and made the S-MDMA induced behavioural sensitization state-dependent. The animals pre-treated with R-MDMA showed a sensitized response in adulthood when tested with RS-MDMA. Our results indicated that even in the absence of substantial neurotoxicity, both MDMA enantiomers can lead to long-term changes in brain circuitry and concomitant behavioural changes when repeatedly administered in adolescence. The sensitization development was most pronounced in the animals treated with S- and RS-MDMA; the animals with R-MDMA did not develop sensitization under repeated treatment but expressed a sensitized response when challenged with RS-MDMA. Decoding of the photoperiod involves circadian clock mechanisms, although the molecular basis remains unclear. Recent studies have shown that the ependymal cell layer lining the infundibular recess of the third ventricle (EC) is a key structure for the photoperiodic gonadal response. The EC exhibits daylength-dependent changes in the expression of photoperiodic output genes, including the type 2 deiodinase gene (Dio2,). Here we investigated whether clock genes (Per1 and Bmal1) and the albumin D-binding protein gene (Dbp) are expressed in the EC of Syrian hamsters, and whether their expression differs under long-day and short-day conditions. Expression of all three genes followed a diurnal rhythm; expression of Per1 and Dbp in the EC peaked around lights-off, and expression of Bmal1 peaked in the early light phase. The amplitude of Per1 and Dbp expression was higher in hamsters kept under long-day conditions than in those kept under short-day conditions. Notably, the expression of these genes was not modified by exogenous melatonin within 25 h after injection, whereas Dio2 expression was inhibited 19 h after injection. Targeted melatonin receptor (MT1, MT2, and both MT1 and MT2) disruption in melatonin-proficient C3H mice did not affect the rhythmic expression of Per1 in the EC. These data show the existence of a molecular clock in the rodent EC. In the hamster, this clock responds to long-term changes in the photoperiod, but is independent of acute melatonin signals. In mice, the EC clock is not affected by deletion of melatonin receptors. Borgland Abstract The orexins (hypocretins) play a crucial role in arousal, feeding and reward. Highly relevant to these functions, orexin-containing neurons from the lateral hypothalamus project densely to the ventral tegmental area (VTA), which is the origin of dopamine projections implicated in motivation and reward. Orexin A/hypocretin 1 (oxA/hcrt-1) can enable long-term changes associated with drugs of abuse; however, the effects of orexin B/hypocretin 2 (oxB/hcrt-2) on excitatory synaptic transmission in the VTA are unknown. We used whole-cell patch-clamp electrophysiology in rat horizontal midbrain slices to examine the effects of oxB/hcrt-2 on excitatory synaptic transmission. We observed that oxB/hcrt-2 has distinct effects from oxA/hcrt-1 in the VTA. oxB/Hcrt-2 (100 nm) increased presynaptic glutamate release in addition to a postsynaptic potentiation of NMDA receptors (NMDARs). The oxB/hcrt-2-mediated postsynaptic potentiation of NMDARs was mediated via activation of orexin/hypocretin 2 (OX2/Hcrt-2) receptors and protein kinase C (PKC). Furthermore, the increase in transmitter release probability was also PKC-dependent, but not through activation of orexin/hypocretin 1 (OX1/Hcrt-1) or OX2/Hcrt-2 receptors. Finally, oxB/hcrt-2 or the selective OX2/Hcrt-2 receptor agonist ala11 - d -leu15 -orexin B, significantly reduced spike-timing-induced long-term potentiation. Taken together, these results support a dual role for oxB/hcrt-2 in mediating enhanced glutamatergic transmission in the VTA, and suggest that oxA/hcrt-1 and oxB/hcrt-2 exert different functional roles in modulating the enhancement of the motivational components of arousal and feeding. Velho Abstract Song behavior in songbirds induces the expression of activity-dependent genes in brain areas involved in perceptual processing, production and learning of song. This genomic response is thought to represent a link between neuronal activation and long-term changes in song-processing circuits of the songbird brain. Here we demonstrate that Arc, an activity-regulated gene whose product has dendritic localization and is associated with synaptic plasticity, is rapidly induced by song in the brain of zebra finches. We show that, in the context of song auditory stimulation, Arc expression is induced in several telencephalic auditory areas, most prominently the caudomedial nidopallium and mesopallium, whereas in the context of singing, Arc is also induced in song control areas, namely nucleus HVC, used as a proper name, the robust nucleus of the arcopallium and the interface nucleus of the nidopallium. We also show that song-induced Arc expression co-localizes at the cellular level with those of the transcriptional regulators zenk and c-fos, and that the song induction of these three genes is dependent on activation of the mitogen-activated protein kinase signaling pathway. These findings provide evidence for an involvement of Arc in the brain's response to birdsong. They also demonstrate that genes representing distinct genomic and cellular regulatory programs, namely early effectors and transcription factors, are co-activated in the same neuronal cells by a naturally learned stimulus.
