Selected AbstractsSources of sulphur in gypsiferous sediments and crusts and pathways of gypsum redistribution in southern TunisiaEARTH SURFACE PROCESSES AND LANDFORMS, Issue 12 2004Nick A. Drake Abstract Southern Tunisia contains one of the most extensive gypsum accumulations in Africa comprising Triassic, Cretaceous, Eocene and Mio-Pliocene marine evaporites, spring deposits, playa sediments, aeolian sands and gypsum crusts. Sulphur isotope analysis (,34S) of bedrock samples, groundwater, playa brines, playa sediments, and gypsiferous crusts provides insight into the sources of gypsum in the region and sheds light on the processes that lead to gypsum crust formation. Results suggest that recycling of marine gypsum is the most likely source of the sulphate in the groundwater, playa sediments and crusts. The low ,34S values found in Eocene and Mio-Pliocene samples suggest that this recycling has been going on for millions of years. Though bedrock appears to be the ultimate source of the gypsum in the crusts, transport of this sulphate to playas, concentration therein, and subsequent dispersal across the landscape by aeolian processes provides the most likely pathway for sur,cial gypsum crust formation. Comparison of these results with those from Australia, Chile and Namibia suggests that, although the source of the sulphur varies from region to region, the processes of sur,cial crust formation appear to be similar. [source] Human sewage identified as likely source of white pox disease of the threatened Caribbean elkhorn coral, Acropora palmataENVIRONMENTAL MICROBIOLOGY, Issue 5 2010Kathryn Patterson Sutherland Summary Caribbean elkhorn coral, Acropora palmata, has been decimated in recent years, resulting in the listing of this species as threatened under the United States Endangered Species Act. A major contributing factor in the decline of this iconic species is white pox disease. In 2002, we identified the faecal enterobacterium, Serratia marcescens, as an etiological agent for white pox. During outbreaks in 2003 a unique strain of S. marcescens was identified in both human sewage and white pox lesions. This strain (PDR60) was also identified from corallivorious snails (Coralliophila abbreviata), reef water, and two non-acroporid coral species, Siderastrea siderea and Solenastrea bournoni. Identification of PDR60 in sewage, diseased Acropora palmata and other reef invertebrates within a discrete time frame suggests a causal link between white pox and sewage contamination on reefs and supports the conclusion that humans are a likely source of this disease. [source] EFFECTIVE POPULATION SIZES AND TEMPORAL STABILITY OF GENETIC STRUCTURE IN RANA PIPIENS, THE NORTHERN LEOPARD FROGEVOLUTION, Issue 11 2004Eric A. Hoffman Abstract Although studies of population genetic structure are very common, whether genetic structure is stable over time has been assessed for very few taxa. The question of stability over time is particularly interesting for frogs because it is not clear to what extent frogs exist in dynamic metapopulations with frequent extinction and recolonization, or in stable patches at equilibrium between drift and gene flow. In this study we collected tissue samples from the same five populations of leopard frogs, Rana pipens, over a 22,30 year time interval (11,15 generations). Genetic structure among the populations was very stable, suggesting that these population were not undergoing frequent extinction and colonization. We also estimated the effective size of each population from the change in allele frequencies over time. There exist few estimates of effective size for frog populations, but the data available suggest that ranid frogs may have much larger ratios of effective size (Ne) to census size (Nc) that toads (bufonidae). Our results indicate that R. pipiens populations have effective sizes on the order of hundreds to at most a few thousand frogs, and Nee/Nc ratios in the range of 0.1,1.0. These estimates of Ne/Nc are consistent with those estimated for other Rana species. Finally, we compared the results of three temporal methods for estimating Ne. Moment and pseudolikelihood methods that assume a closed population gave the most similar point estimates, although the moment estimates were consistently two to four times larger. Wang and Whitlock's new method that jointly estimates Ne and the rate of immigration into a population (m) gave much smaller estimates of Ne and implausibly large estimates of m. This method requires knowing allele frequencies in the source of immigrants, but was thought to be insensitive to inexact estimates. In our case the method may have failed because we did not know the true source of immigrants for each population. The method may be more sensitive to choice of source frequencies than was previously appreciated, and so should be used with caution if the most likely source of immigrants cannot be identified clearly. [source] Gardening at the edge: Documenting the limits of tropical Polynesian kumara horticulture in southern New ZealandGEOARCHAEOLOGY: AN INTERNATIONAL JOURNAL, Issue 3 2004Kari N. Kumara (Ipomoea batatus), a major food source for Maori, was brought to New Zealand from tropical Eastern Polynesia ,700 years ago. Maori successfully adapted their cultivation techniques to grow kumara in New Zealand's cooler, seasonal climate, although most kumara cultivation was limited to the warmer North Island, with cultivation becoming more marginal southward. Banks Peninsula area is considered to be the southernmost limit for kumara gardening. The Okuora Farm archeological site on the southern side of Banks Peninsula has five pits that appear to be of the raised-rim type used for over winter storage of kumara tubers. We conducted a preliminary investigation into the nature of the pits and surrounding 1 km2 area using nondestructive techniques in accordance with Maori designation of food storage sites as tapu. Ground penetrating radar (GPR) investigation of two of the pits revealed subsurface disturbances consistent with postholes and drains, typical of raised rim kumara storage pits. Soil modification typical of kumara gardening was identified on a 1 ha area on a warm north-northwest facing hillside. Several large borrow pits were identified as the likely source of the gravel added to the modified soil, possibly to retain heat and moisture. A plant phytolith study of soil samples identified several that appear to be from kumara. The combination of results strongly suggests this site was one of the southernmost Maori kumara gardening sites yet identified in New Zealand. © 2004 Wiley Periodicals, Inc. [source] Mineral chemical provenance of Neolithic pitchstone artefacts from Ballygalley, County Antrim, Northern IrelandGEOARCHAEOLOGY: AN INTERNATIONAL JOURNAL, Issue 3 2002Jeremy Preston A large number of pitchstone fragments and artefacts have recently been discovered at a Neolithic settlement site in Ballygalley, Co. Antrim, Northern Ireland. They consist predominantly of flakes and un-reworked lumps and cores, with only one complete tool being found. Since no sources of workable pitchstone exist in Ireland, the source must have been the abundant pitchstone volcanic rocks found on the Hebridean Islands of northwest Scotland. The composition of the glass from a number of artefacts is highly siliceous, indicating that they were derived from pitchstones on the Island of Arran; pitchstones from all other Scottish locations are less silica-rich. In addition, analysis of pyroxene and amphibole microcrystallites within the pitchstone suggests that the Corriegills area of Arran is the most likely source of the Ballygalley artefacts, although the precise outcrop has proved elusive. These finds, and others across Ireland, show that raw materials were being transported and probably traded over considerable distances despite there being suitable alternative sources of material for making tools (flint, etc.) available in the local area. [source] Generation and accumulation of oil and condensates in the Wenchang A Sag, western Pearl River Mouth Basin, South China SeaGEOFLUIDS (ELECTRONIC), Issue 4 2009H. J. GAN Abstract The Pearl River Mouth (PRM) Basin is one of four Cenozoic basins in the South China Sea, and the Wenchang A Sag is a secondary depression in the western part of the basin. Both the Wenchang and Enping formations contain good source rocks in the western PRM Basin; however, only the latter has been considered a likely source of the discovered oil and gas. New data from fluid inclusions and the analysis of oil,source rock correlations for the WC10-3 oil and gas pools indicate two stages of petroleum charging, the earlier originating from the Wenchang Formation and the later from the Enping Formation. Kinetics of petroleum generation and structural evolution modeling were employed to further investigate the mechanism of formation of the WC10-3 oil and gas pools. It was shown that the crucial condition for the formation of pools is the time of development of the structural trap. The Wenchang Formation source rocks generated oil from 25 to 14 Ma in the possible source area of the WC10-3 oil and gas pools in the Wenchang A Sag, so that only traps formed earlier than this period could capture oil sourced by the Wenchang Formation. The Enping Formation source rock experienced its oil window from 18 Ma to the present with the main stage of oil generation from 15 to 5 Ma. During this period structural traps in the sag continued to form until movements became weak, so that most pools in the Wenchang A Sag originated from the Enping Formation source rock. The likely dissipation of oil and gas from the earlier stage of charging should be taken into account in assessing the oil potential of the Wenchang A Sag. [source] Chemical and Isotopic Constraints on the Origin of Wadi El-Tarfa Ground Water, Eastern Desert, EgyptGROUND WATER, Issue 5 2000M. Sultan We evaluated the use of the renewable ground water resources of the Eastern Desert to develop sustainable agriculture in Upper Egypt, an alternative that could alleviate some of Egypt's dependence on water from the Nile River. Ground water from shallow aquifers in the Eastern Desert of Egypt, near the intersection of Wadi El-Tarfa and the Nile River, was analyzed for chemical compositions, stable isotope ratios, and tritium activities. The ground water has a range in total dissolved solids of 300 to 5000 mg/L. Values of ,D and ,18O range from -10 to +34 %o and -2 to +5.2 %o, respectively, and defines a line having a slope of 5.7 that intersects the meteoric water line at about ,D = -15 %o on a plot of 8D versus ,18O. These findings indicate that the water might have been derived by a combination of evaporation of and salt addition to regional precipitation. Only one sample could have been derived directly by evaporation and transpiration of modern Nile River water. Salinization of the ground water could have occurred through dissolution of marine aerosol dry fallout, carbonate minerals, gypsum, and other trace evaporitic minerals at and near the ground surface. Tritium activities ranged from 0.04 to 12.9 TU (tritium unite), indicating that all but one of the samples were derived at least partly from precipitation that occurred within the last 45 years. These data indicate that Nubian Aquifer paleowater is not a significant component of the shallow aquifers of this portion of the Eastern Desert. The most likely source of this ground water is sporadic flash flood events yielding locally voluminous recharge that accumulates in coarse sediments and fractured rock beneath alluvial channels. The magnitude of this renewable ground water resource and its potential for supporting sustainable agriculture require further investigation. [source] Analysis and comparison of morphological reconstructions of hippocampal field CA1 pyramidal cellsHIPPOCAMPUS, Issue 3 2005José Ambros-Ingerson Abstract Morphological reconstructions have become a routine and valuable tool for neuroscientists. The accuracy of reconstructions is a matter of considerable interest given that they are widely used in computational studies of neural function. Despite their wide usage, comparisons of reconstructions obtained using various methodologies are lacking. We reviewed reconstructions of hippocampal CA1 pyramidal cells from five published studies and found marked differences in some of the most basic measurements. For four of the five studies means of total cell length clustered in the 11,479,13,417-,m range. The remaining study had a significantly larger value for this index at 16,992 ± 5,788 ,m. Surface area means varied more than 4-fold from 16,074 to 67,102 ,m2. Volume means varied more than 8-fold from 3,828 to 30,384 ,m3. Simulated passive input resistance means varied from 38.0 to 172.1 M,, reflecting the variability in cell dimensions. Estimates of the electrotonic length varied from 1.26 to 1.56. In two reconstructions used in previously published studies, simulated somatic excitatory postsynaptic potentials (EPSPs) varied 2,4-fold in amplitude, time to peak and half-width, for synaptic inputs at similar locations. Substantial jitter on the z -axis was identified as one likely source of the discrepancy in total cell length, while substantial differences in diameter measurements across studies, and sometimes within the same study, accounted for the variability in surface area and volume. While some part of the observed variability is surely due to the diversity of CA1 pyramidal cells, our analysis suggests that a substantial portion stemmed from methodological inconsistencies and from technological limitations. Suggestions are made for improving the quality and usefulness of morphological reconstructions. We conclude that reconstructions across studies have substantial variability in measures that are very relevant to neuronal function. Consequently, modelers are advised to use more than just one reconstructed cell in their simulations of neural function. © 2004 Wiley-Liss, Inc. Komitopoulou Abstract Aims:, To study the behaviour of salmonellae inoculated in the dry, raw materials, related to chocolate confectionery manufacture, as affected by the strain, cell preparation method and storage temperature. Methods and Results:, Outbreak and non outbreak-associated salmonellae were used for the inoculation of cocoa butter oil, crushed cocoa and hazelnut shells, cocoa beans and almond kernels. Dry matrices were inoculated with lawn-collected and broth-grown cells and were stored at 5 and 21°C for 21 days. Results demonstrated that all strains survived in all dry matrices. Lawn-collected cells survived considerably better than broth-collected cells, at either temperature, and outbreak-associated strains of Salmonella serotype Enteritidis PT30 and Salmonella serotype Oranienburg appeared to be among the best surviving strains. Conclusions:, The work demonstrated that salmonellae can survive storage for 3,4 weeks in dry raw materials and that survival is dependent on the source of strains, cell preparation and inoculation methodology, and storage temperature. Significance and Impact of the Study:, The data contributes to understanding the parameters to consider when assessing the risk of dry raw materials as the most likely source of salmonellae in chocolate processing plants. It also demonstrates the importance of implementing effective lethal processes and segregation procedures to prevent cross-contamination to ensure the safety of confectionery products regarding Salmonella. [source] Evolutionary divergence and possible incipient speciation in post-glacial populations of a cosmopolitan aquatic plantJOURNAL OF EVOLUTIONARY BIOLOGY, Issue 1 2005G. Nies Abstract Habitat configuration is expected to have a major influence on genetic exchange and evolutionary divergence among populations. Aquatic organisms occur in two fundamentally different habitat types, the sea and freshwater lakes, making them excellent models to study the contrasting effects of continuity vs. isolation on genetic divergence. We compared the divergence in post-glacial populations of a cosmopolitan aquatic plant, the pondweed Potamogeton pectinatus that simultaneously occurs in freshwater lakes and coastal marine sites. Relative levels of gene flow were inferred in 12 lake and 14 Baltic Sea populations in northern Germany using nine highly polymorphic microsatellite markers developed for P. pectinatus. We found highly significant isolation-by-distance in both habitat types (P < 0.001). Genetic differentiation increased approximately 2.5-times faster among freshwater populations compared with those from the Baltic Sea. As different levels of genetic drift or population history cannot explain these differences, higher population connectivity in the sea relative to freshwater populations is the most likely source of contrasting evolutionary divergence. These findings are consistent with the notion that freshwater angiosperms are more conducive to allopatric speciation than their life-history counterparts in the sea, the relative species poor seagrasses. Surprisingly, population pairs from different habitat types revealed almost maximal genetic divergence expected for complete reproductive isolation, regardless of their respective geographical distance. Hence, the barrier to gene flow between lake and sea habitat types cannot be due to dispersal limitation. We may thus have identified a case of rapid incipient speciation in post-glacial populations of a widespread aquatic plant. [source] Characterization of a human fetal spinal cord stem cell line, NSI-566RSC, and its induction to functional motoneuronsJOURNAL OF TISSUE ENGINEERING AND REGENERATIVE MEDICINE, Issue 3 2010Xiufang Guo Abstract Specific neuronal subtypes, especially motoneurons (MNs), derived from human stem cells provide a significant therapeutic potential for spinal cord diseases, such as amyotrophic lateral sclerosis (ALS) and spinal cord injury. So far, in vitro, MNs have only been successfully induced from embryonic stem cells (hESC) and human fetal cortical progenitors. Although neural progenitors from spinal cord would be a likely source for generating MNs, there has been no study reporting successful in vitro differentiation of MNs from spinal cord progenitors. This study first characterized a polyclonal spinal cord stem cell line isolated from an 8 week-old fetus. Then a paradigm was introduced to successfully induce MNs from this cell line, which was demonstrated by immunostaining using the MN markers HB9, Islet1 and choline acetyl transferase (ChAT). The combination of HB9 and ChAT immunostainings indicated that ,20% of the cells were MNs after this induction protocol. The presence of other cell types in the differentiated culture was also analysed. Finally, the electrophysiological properties of these differentiated MNs were characterized to confirm their functional integrity. The majority of these MNs fired repetitive action potentials (APs), which is an indicator of functional maturation. The recordings of spontaneous excitatory postsynaptic currents (EPSCs) confirmed the formation of synapses onto these MNs. This study reports the first successful differentiation of MNs from human spinal cord stem cells in vitro, providing a novel approach for obtaining functional MNs when designing the therapeutic strategy for spinal cord diseases or injuries. Leong Abstract Aims:, To determine the incidence and severity of infection by ochratoxin A (OA)-producing fungi in Vietnamese green coffee beans. Methods and Results:,Aspergillus carbonarius, A. niger and yellow Aspergilli (A. ochraceus and related species in section Circumdati) were isolated by direct plating of surface-disinfected Robusta (65 samples) and Arabica (11 samples) coffee beans from southern and central Vietnam. Significantly, more Robusta than Arabica beans were infected by fungi. Aspergillus niger infected 89% of Robusta beans, whereas A. carbonarius and yellow Aspergilli each infected 12,14% of beans. OA was not produced by A. niger (98 isolates) or A. ochraceus (77 isolates), but was detected in 110 of 113 isolates of A. carbonarius, 10 isolates of A. westerdijkiae and one isolate of A. steynii. The maximum OA observed in samples severely infected with toxigenic species was 1·8 ,g kg,1; however, no relationship between extent of infection and OA contamination was observed. Conclusions:,Aspergillus niger is the dominant species infecting Vietnamese coffee beans, yet A. carbonarius is the likely source of OA contamination. Significance and Impact of Study:, Vietnamese green coffee beans were more severely infected with fungi than the levels reported for beans from other parts of the world, yet OA contamination appears to be infrequent. Ibenyassine Abstract Aims:, Fresh fruits and vegetables are increasingly recognized as vectors for food-borne illness. On farm contamination through contaminated irrigation water is considered likely source of the pathogen for several outbreaks. The purpose of this study is to investigate the possible similarity of strains of Escherichia coli isolated from the soil and vegetables irrigated by treated wastewater. Methods and Results:, Seventy-five strains of enteropathogenic Escherichia coli isolated from vegetables, soil and irrigation water were tested for sensitivity to antibiotics and shown to be sensitive. The result of enterobacterial repetitive intergenic consensus (ERIC)-PCR shows similarities between analysed strains isolated from the three different samples. Moreover strains of E. coli isolated from vegetables over different periods of time have the same ERIC-PCR profile. Conclusions:, The isolated strains of enteropathogenic E. coli can persist in soil and in vegetables growing in fields treated with contaminated irrigation water for an extended period of time. Significance and Impact of the Study:, Contaminated irrigation water can transport pathogenic bacteria, which persists in the soil for a long period of time and contaminates the vegetables growing in the field irrigated by this contaminated water. [source] Keck infrared observations of GRO J0422+32 in quiescenceMONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 2 2007Mark T. Reynolds ABSTRACT We present Keck K -band photometry and low-resolution H - and K -band spectroscopy of the X-ray nova GRO J0422+32 obtained while the system was in the quiescent state. No clear ellipsoidal modulation is present in the light curve, which is instead dominated by a strong flickering component. In the K band, we observe strong Br, emission, with an equivalent width of 38 ± 5 Å. From this, we conclude that the accretion disc is the most likely source of the observed photometric contamination, and that previous infrared-based attempts to constrain the mass of the putative black hole in this system are prone to considerable uncertainty. We finally proceed to show how it is possible to place meaningful constraints on some of the binary parameters of this system, even in the presence of a relatively high level of contamination from the disc. Daniels BS Abstract Background Previous analyses suggest that worker radiation dose may be significantly increased by routine occupational X-ray examinations. Medical exposures are investigated for 570 civilian workers employed at the Portsmouth Naval Shipyard (PNS) at Kittery, Maine. The research objective was to determine the radiation exposure contribution of work-related chest X-rays (WRX) relative to conventional workplace radiation sources. Methods Methods were developed to estimate absorbed doses to the active (hematopoietic) bone marrow from X-ray examinations and workplace exposures using data extracted from worker dosimetry records (8,468) and health records (2,453). Dose distributions were examined for radiation and non-radiation workers. Results Photofluorographic chest examinations resulted in 82% of the dose from medical sources. Radiation workers received 26% of their collective dose from WRX and received 66% more WRX exposure than non-radiation workers. Conclusions WRX can result in a significant fraction of the total dose, especially for radiation workers who were more likely to be subjected to routine medical monitoring. Omission of WRX from the total dose is a likely source of bias that can lead to dose category misclassification and may skew the epidemiologic dose,response assessment for cancers induced by the workplace. [source] Genetic diversity within and among Atlantic cod (Gadus morhua) farmed in marine cages: a proof-of-concept study for the identification of escapeesANIMAL GENETICS, Issue 5 2010K. A. Glover Summary This study presents a molecular genetic characterization of Atlantic cod reared in commercial marine farms. Samples consisted of approximately 47 fish collected from nine cages located on four farms throughout Norway. In addition, 28 farmed escapees were recaptured in the sea (443 fish in total). Nine microsatellite loci and the Pan I gene were analysed, revealing a total of 181 alleles. Each sample contained 43,63% of total allelic variation. Comparing variation with published data for wild cod indicates that lower genetic variation exists within single cages than in wild populations. Significant linkage disequilibrium was observed amongst pairs of loci in all samples, suggesting a low number of contributing parental fish. Global FST was 0.049, and the highest pairwise FST value (pooled loci) was 0.085. For single loci, the Pan I gene was the most diagnostic, displaying a global FST of 0.203. Simulations amongst the samples collected on farms revealed an overall correct self-assignment percentage of 75%, demonstrating a high probability of identifying individuals to their farm of origin. Identification of the 28 escapees revealed a single cage as the most likely source of origin for half of the escapees, whilst the remaining fish were assigned to a mixture of samples, suggesting more than one source of escapees. HOLMES ABSTRACT The human AIDS viruses , HIV-1 and HIV-2 , impose major burdens on the health and economic status of many developing countries. Surveys of other animal species have revealed that related viruses , the SIVs , are widespread in a large number of African simian primates where they do not appear to cause disease. Phylogenetic analyses indicate that these SIVs are the reservoirs for the human viruses, with SIVsm from the sooty mangabey monkey the most likely source of HIV-2, and SIVcpz from the common chimpanzee the progenitor population for HIV-1. Although it is clear that AIDS has a zoonotic origin, it is less certain when HIV-1 and HIV-2 first entered human populations and whether cross-species viral transmission is common among primates. Within infected individuals the process of HIV evolution takes the form of an arms race, with the virus continually fixing mutations by natural selection which allow it to escape from host immune responses. The arms race is less intense in SIV-infected monkeys, where a weaker immune response generates less selective pressure on the virus. Such a difference in virus-host interaction, along with a broadening of co-receptor usage such that HIV strains are able to infect cells with both CCR5 and CXCR4 chemokine receptors, may explain the increased virulence of HIV in humans compared to SIV in other primates. [source] Control of misincorporation of serine for asparagine during antibody production using CHO cellsBIOTECHNOLOGY & BIOENGINEERING, Issue 1 2010Anurag Khetan Abstract A recombinant monoclonal antibody produced by Chinese hamster ovary (CHO) cell fed-batch culture was found to have amino acid sequence misincorporation upon analysis by intact mass and peptide mapping mass spectrometry. A detailed analysis revealed multiple sites for asparagine were being randomly substituted by serine, pointing to mistranslation as the likely source. Results from time-course analysis of cell culture suggest that misincorporation was occurring midway through the fed-batch process and was correlated to asparagine reduction to below detectable levels in the culture. Separate shake flask experiments were carried out that confirmed starvation of asparagine and not excess of serine in the medium as the root cause of the phenomenon. Reduction in serine concentration under asparagine starvation conditions helped reduce extent of misincorporation. Supplementation with glutamine also helped reduce extent of misincorporation. Maintenance of asparagine at low levels in 2,L bench-scale culture via controlled supplementation of asparagine-containing feed eliminated the occurrence of misincorporation. [source] Apportionment of polycyclic aromatic hydrocarbon sources in lower Fox River, USA, sediments by a chemical mass balance modelENVIRONMENTAL TOXICOLOGY & CHEMISTRY, Issue 6 2000Ming-Chien Su Abstract Four sediment cores were collected from the lower Fox River, Wisconsin, USA, to identify possible sources of polycyclic aromatic hydrocarbons (PAHs) using a chemical mass balance model. The cores, which were obtained in 1995 from areas close to Green Bay, Wisconsin, USA, had total PAH concentrations between 19.3 and 0.34 ppm. To determine historical trends of PAH inputs, 210Pb and 137Cs dating was used, and elemental carbon particle analysis was done to characterize particles from the combustion of coal, wood, and petroleum. Source fingerprints were taken from the literature. Our results indicate that coke oven emissions, highway dust, coal gasification, and wood burning are likely sources of PAHs in the lower Fox River. Coke oven emissions are in the range of 40 to 90% of total PAHs, and this fraction decreases from 1930 to 1990, except in core Fox River,A (FR-A). The overall highway dust (HWY) contribution is between 10 and 75%, and this fraction increases from 1930 to present, except in core FR-A. The wood burning (WB) contribution (i.e., wood burning or coal gasification) is less than 7% in cores FR-B, FR-C, and FR-D. In core FR-A, a maximum (,23%) is found around 1960. The contribution of wood burning has changed from less than 6% in 1950 to between 3 and 10% in 1995. Evidence of aerobic biodegradation or photolysis in the sediment of phenanthrene, with a half-life of approximately 0.5 years has been found at the site of core FR-D, which is the shallowest (1.1 m) of the four core sites. [source] Trends and methodological impacts in soil CO2 efflux partitioning: A metaanalytical reviewGLOBAL CHANGE BIOLOGY, Issue 6 2006JENS-ARNE SUBKE Abstract Partitioning soil carbon dioxide (CO2) efflux (RS) into autotrophic (RA; including plant roots and closely associated organisms) and heterotrophic (RH) components has received considerable attention, as differential responses of these components to environmental change have profound implications for the soil and ecosystem C balance. The increasing number of partitioning studies allows a more detailed analysis of experimental constraints than was previously possible. We present results of an exhaustive literature search of partitioning studies and analyse global trends in flux partitioning between biomes and ecosystem types by means of a metaanalysis. Across all data, an overall decline in the RH/RS ratio for increasing annual RS fluxes emerged. For forest ecosystems, boreal coniferous sites showed significantly higher (P<0.05) RH/RS ratios than temperate sites, while both temperate or tropical deciduous forests did not differ in ratios from any of the other forest types. While chronosequence studies report consistent declines in the RH/RS ratio with age, no difference could be detected for different age groups in the global data set. Different methodologies showed generally good agreement if the range of RS under which they had been measured was considered, with the exception of studies estimating RH by means of root mass regressions against RS, which resulted in consistently lower RH/RS estimates out of all methods included. Additionally, the time step over which fluxes were partitioned did not affect RH/RS ratios consistently. To put results into context, we review the most common techniques and point out the likely sources of errors associated with them. In order to improve soil CO2 efflux partitioning in future experiments, we include methodological recommendations, and also highlight the potential interactions between soil components that may be overlooked as a consequence of the partitioning process itself. Kcnj10 brain expression differences have been established between ethanol-preferring C57Bl/6J and ethanol-avoiding BALB/cJ mice, but its differential expression in other tissues and strains have largely been overlooked. A nonsynonymous single nucleotide polymorphism exists between C57Bl/6J and ethanol-avoiding DBA/2J mice which changes amino acid 262 from threonine (C57Bl/6J) to serine (DBA/2J). This Kcnj10 SNP and its expression may serve as valuable markers in predicting the ethanol preference phenotype in mice. Methods:, The evolutionary divergence of the Kir gene family was characterized using phylogenetic analysis involving the 16 mouse Kir channels. Kcnj10 expression differences in the brain, liver, lung, heart, spleen, kidney, testes, and muscle of male C57Bl/6J and DBA/2J mice at different developmental stages were examined using semiquantitative RT-PCR analysis. A SNP analysis was conducted to assess the association of Kcnj10 Thr262Ser SNP and the ethanol preference phenotype in F2 mice derived from the reciprocal crosses of the C57Bl/6J and DBA/2J strains. Results:, Evolutionary analysis supports gene duplication and genetic recombination as likely sources of diversity within the Kir gene family. Semiquantitative RT-PCR analysis revealed significantly higher Kcnj10 expression in the brain, spleen, and kidney of both strains when compared to other tissues from the same strain. There were no significant differences in tissue-specific mRNA levels between strains except in the testes. Genotype distributions of the Kcnj10 Thr262Ser SNP were different between low- and high-drinkers. A significant difference in the average ethanol preference level of each genotype was also observed. Conclusion:, Our results suggest a role for Kcnj10 in ethanol preference determination in mice. PATTERSON Abstract Although recruitment of pelagic larvae is a fundamental and well-documented process in the dynamics of benthic marine populations, identifying the sources of recruitment, or the degree to which populations are connected via dispersal of larvae, has remained elusive for most marine taxa. In this study we used natural environmental markers (trace elements) recorded in fish otoliths (ear stones) as tags of natal origin. Specifically, we used the otolith core and edge chemistries of a locally endemic wrasse (Coris bulbifrons) from Lord Howe Island (LHI), Australia, and a widely distributed species (Coris picta) from three potential mainland source regions, to determine the likely sources of recruitment to C. picta populations on LHI. The use of a local endemic species, which is by definition self-recruiting, is a novel approach for ground-truthing the dispersal history of non-endemic coral reef fish. Discriminant function analyses were able to separate LHI from mainland fish, using both edge and core signatures, with a high degree of accuracy, suggesting at least some of the C. picta collected on LHI were of local origin. This result was corroborated when half of the C. bulbifrons and LHI C. picta were introduced as unknowns into a discriminant function analysis using the remaining C. bulbifrons, LHI C. picta, and the mainland C. picta as a training data set. Overall, our findings suggest that both long distance dispersal and local retention are important sources of recruitment to populations of C. picta on LHI and that otolith chemistry of endemic species could be a useful benchmark for determining the prevalence of self-recruitment in insular populations of other widespread species. The major and trace element compositions are comparable to Post-Archean upper continental crust (PA-UCC), but have slight enrichment in the LILE, with the exception of Cs and Sr, and a slight depletion in ferromagnesian elements, HFS elements, such as Nb and Ta, and some major elements, such as CaO and Na2O. The geochemical data reveal that the collected metasedimentary rocks have experienced intermediate source weathering with chemical index of alteration values ranging from 72 to 78, varying degrees of K-metasomatism, and post-depositional loss of Na, as well as negligible sorting, and are derived from the weathering of mostly felsic and non-mafic rocks. The selected Lu,Hf isotopic analysis on detrital zircon points to both the Trans-North China Orogen and Eastern Block of the north China craton as the most likely sources for the metasedimentary rocks of the Jiangxian Group. However, a contribution of detritus from the Western Block of the north China craton can be ruled out. The sediments were probably deposited in a back-arc basin within an active continental margin setting. The purpose of the study was to assay multiple cytokines and chemokines in tears, to compare subgroups of allergic conjunctivitis (AC) with controls, and in culture supernatants to determine whether conjunctival fibroblasts produce some of these cytokines. Methods Fifty to one hundred microlitre tears were obtained from patients with active seasonal allergic conjunctivitis (SAC; n=12), vernal keratoconjunctivitis (VKC; n=18), atopic keratoconjunctivitis (AKC; n=6) and non-atopic controls (n=14). Primary conjunctival fibroblasts grown in vitro were stimulated with IL-4, IL-13 or TNF-, for 24 h. Cell-free tear and culture supernatants were assayed for IL-1,, IL-2, IL-4, IL-5, IL-6, IL-8, IL-10, IL-12, IL-13, IFN-,, TNF-,, eotaxin, MCP-1 and RANTES using multiplex bead analysis. Induction of chemokine gene expression was determined by PCR. Results IL-1,, IL-2, IL-5, IL-6, IL-12, IL-13, MCP-1 were increased in all tears groups compared with controls, with highly significant correlations between many of these molecules. In addition IL-4, IFN-,, and IL-10 were elevated in SAC and VKC, while eotaxin and TNF-, were only increased in VKC. IL-6, IL-8, MCP-1, RANTES and eotaxin were detected from fibroblasts cultures, and were all up-regulated by TNF-,. By PCR, fibroblasts expressed MCP-1 transcripts constitutively, whereas IP-10 and Mig were up-regulated by TNF-,. Conclusions Differential cytokine levels support tears as a useful indicator of immune mechanisms occurring during AC. The striking similarities in chemokine profiles between tears and fibroblasts suggest these cells as likely sources of chemokines in tears. [source] |
