Immunocytochemistry in liquid-based cervical cytology: Analysis of clinical use following a cross-sectional study

Abstract Cytological screening for cervical cancer is hampered by imperfect sensitivity and low inter-observer reproducibility. Human papillomavirus (HPV) testing lacks specificity as a primary screening method. Studies indicate that immunocytochemical detection of alterations caused by HPV in the host cells can optimise screening. Here, the potential of p16INK4a (cyclin-dependent kinase inhibitor p16) and MIB-1 (Ki-67 proliferation marker) as adjunct molecular markers for cervical lesions was investigated in a prospective, cross-sectional study of 500 samples in the framework of opportunistic screening in Flanders, Belgium. A consecutive series of 200 samples and 100 samples from the cytological categories ASC, LSIL and HSIL were investigated. Surepath samples were interpreted according to the Bethesda 2001 reporting system. HPV testing was done with MY09/MY11 consensus PCR. Immunocytochemistry for p16INK4a and MIB-1 was performed with an automated staining protocol. The number of immunoreactive cells/1,000 cervical cells was assessed. There was a higher mean number of p16INK4A and MIB-1 immunoreactive cells/1,000 cells in HSIL (4.06 ± 1.93 and 11.13 ± 2.83, respectively) compared to other cytological categories. Both markers showed a large spread in counts, for all categories. In cases of HSIL without immunoreactive cells for either marker, low cellularity and long-term storage in water were often the cause of false negativity. This study confirms that positive staining for p16INK4a and MIB-1 is highly correlated with presence of high-grade lesions. These markers could be used as adjuncts to increase the sensitivity of cytological screening as well as the specificity of the HPV test. On the interstellar medium and star formation demographics of galaxies in the local universe

ABSTRACT We present a demographic analysis of integrated star formation and gas properties for a sample of galaxies representative of the overall population at z, 0. This research was undertaken in order to characterize the nature of star formation and interstellar medium (ISM) behaviour in the local Universe, and test the extent to which global star formation rates (SFRs) can be seen as dependent on the interstellar gas content. Archival 21-cm derived H i data are compiled from the literature, and are combined with CO (J =1 , 0) derived H2 masses to calculate and characterize the total gas content for a large sample of local galaxies. The distribution in stellar mass-normalized H i content is found to exhibit the noted characteristic transition at stellar masses of ,3 × 1010 M,, turning off towards low values, but no such transition is observed in the equivalent distribution of molecular gas. H, based SFRs and specific star formation rates (SSFRs) are also compiled for a large (1110) sample of local galaxies. We confirm two transitions as found in previous work: a turnover towards low SFRs at high luminosities, indicative of the quenching of SF characteristic of the red sequence; and a broadening of the SF distribution in low-luminosity dwarf galaxies, again to extremely low SFRs of <10,3 M, yr,1. However, a new finding is that while the upper luminosity transition is mirrored by the turnover in H i content, suggesting that the low SFRs of the red sequence result from a lack of available gas supply, the transition towards a large spread of SFRs in the least luminous dwarf galaxies is not matched by a prominent increase in scatter in gas content. Possible mass-dependent quenching mechanisms are discussed, along with speculations that in low-mass galaxies, the H, luminosity may not faithfully trace the SFR. [source]


Mass segregation in young compact clusters in the Large Magellanic Cloud , III.

Implications for the initial mass function
ABSTRACT The distribution of core radii of rich clusters in the Large Magellanic Cloud (LMC) systematically increases in both upper limit and spread with increasing cluster age. Cluster-to-cluster variations in the stellar initial mass function (IMF) have been suggested as an explanation. We discuss the implications of the observed degree of mass segregation in our sample clusters for the shape of the initial mass function. Our results are based on Hubble Space Telescope/WFPC2 observations of six rich star clusters in the LMC, selected to include three pairs of clusters of similar age, metallicity and distance from the LMC centre, and exhibiting a large spread in core radii between the clusters in each pair. All clusters show clear evidence of mass segregation: (i) their luminosity function slopes steepen with increasing cluster radius, and (ii) the brighter stars are characterized by smaller core radii. For all sample clusters, both the slope of the luminosity function in the cluster centres and the degree of mass segregation are similar to each other, within observational errors of a few tenths of power-law slope fits to the data. This implies that their initial mass functions must have been very similar, down to ,0.8,1.0 M,. We therefore rule out variations in the IMF of the individual sample clusters as the main driver of the increasing spread of cluster core radii with cluster age. [source]


Cognitive and emotional outcome after pediatric liver transplantation

Abstract: The aim of the study was to evaluate the cognitive and emotional development after pediatric liver transplantation. A total of 21 patients, aged 4,16.9 yr (median 9.6 yr) were tested 1,9 yr (median 4.2 yr) after the transplantation. The pretransplant diagnoses included biliary atresia (eight patients), various metabolic diseases (n = 6), acute liver failure (n = 3), and miscellaneous (n = 4). The cognitive functions were tested with Wechsler preschool and primary scale of intelligence (WPPSI)-R or Wechsler intelligence scale for children (WISC)-III according to age. The Piers,Harris self-concept scale and the evaluation of human figure drawings according to Koppitz were used to detect emotional problems. All tests in all patients were performed by the same psychologist. A significantly lower result on cognitive tests was seen when compared with the expected normal values (p < 0.01). The number of patients with results within or under the lower normal range was higher than expected. Although the mean value of the Piers,Harris self-concept scale was normal, there was a large spread within the group. Indicators of emotional problems were found in the human figure drawings of 50% of the patients. To some extent, low cognitive scores coincided with low scores on self-concept scale and indicators of emotional difficulties. We conclude that the high degree of cognitive and emotional problems after liver transplantation is an important argument for routine psychologic follow-up and support in these patients. [source]


Intercomparison of model simulations of mixed-phase clouds observed during the ARM Mixed-Phase Arctic Cloud Experiment.

Abstract Results are presented from an intercomparison of single-column and cloud-resolving model simulations of a deep, multilayered, mixed-phase cloud system observed during the Atmospheric Radiation Measurement (ARM) Mixed-Phase Arctic Cloud Experiment. This cloud system was associated with strong surface turbulent sensible and latent heat fluxes as cold air flowed over the open Arctic Ocean, combined with a low pressure system that supplied moisture at mid-levels. The simulations, performed by 13 single-column and 4 cloud-resolving models, generally overestimate liquid water path and strongly underestimate ice water path, although there is a large spread among models. This finding is in contrast with results for the single-layer, low-level mixed-phase stratocumulus case in Part I, as well as previous studies of shallow mixed-phase Arctic clouds, that showed an underprediction of liquid water path. These results suggest important differences in the ability of models to simulate deeper Arctic mixed-phase clouds versus the shallow, single-layered mixed-phase clouds in Part I. The observed liquid-ice mass ratios were much smaller than in Part I, despite the similarity of cloud temperatures. Thus, models employing microphysics schemes with temperature-based partitioning of cloud liquid and ice masses are not able to produce results consistent with observations for both cases. Models with more sophisticated, two-moment treatment of cloud microphysics produce a somewhat smaller liquid water path closer to observations. Cloud-resolving models tend to produce a larger cloud fraction than single-column models. The liquid water path and cloud fraction have a large impact on the cloud radiative forcing at the surface, which is dominated by long-wave flux. Subseasonal extremes of precipitation and active-break cycles of the Indian summer monsoon in a climate-change scenario

Abstract Changes to the behaviour of subseasonal precipitation extremes and active-break cycles of the Indian summer monsoon are assessed in this study using pre-industrial and 2 × CO2 integrations of the Hadley Centre coupled model HadCM3, which is able to simulate the monsoon seasonal cycle reasonably. At 2 × CO2, mean summer rainfall increases slightly, especially over central and northern India. The mean intensity of daily precipitation during the monsoon is found to increase, consistent with fewer wet days, and there are increases to heavy rain events beyond changes in the mean alone. The chance of reaching particular thresholds of heavy rainfall is found to approximately double over northern India, increasing the likelihood of damaging floods on a seasonal basis. The local distribution of such projections is uncertain, however, given the large spread in mean monsoon rainfall change and associated extremes amongst even the most recent coupled climate models. The measured increase of the heaviest precipitation events over India is found to be broadly in line with the degree of atmospheric warming and associated increases in specific humidity, lending a degree of predictability to changes in rainfall extremes. Active-break cycles of the Indian summer monsoon, important particularly due to their effect on agricultural output, are shown to be reasonably represented in HadCM3, in particular with some degree of northward propagation. We note an intensification of both active and break events, particularly when measured against the annual cycle, although there is no suggestion of any change to the duration or likelihood of monsoon breaks. PROVENANCE OF MARBLES FROM NAXOS BASED ON MICROSTRUCTURAL AND GEOCHEMICAL CHARACTERIZATION

A detailed quantitative microstructural study coupled with cathodoluminescence and geochemical analyses on marbles from Naxos demonstrates that the analysis of microstructures is the most sensitive method to define the origin of marbles within, and between, different regions. Microstructure examination can only be used as an accurate provenance tool if a correction for the second-phase content is considered. If second phases are not considered, a large spread of different microstructures occurs within sample sites, making a separation between neighbouring outcrops difficult or impossible. Moreover, this study shows that the origin of a marble is defined more precisely if the microstructural observations are coupled with cathodoluminescence data. [source]
