Selected AbstractsIs the Cell Death in Mesial Temporal Sclerosis Apoptotic?EPILEPSIA, Issue 6 2003Hilmi Uysal Summary: Purpose: Mesial temporal sclerosis (MTS) is characterized by neuronal loss in the hippocampus. Studies on experimental models and patients with intractable epilepsy suggest that apoptosis may be involved in neuronal death induced by recurrent seizures. Methods: We searched evidence for apoptotic cell death in temporal lobes resected from drug-resistant epilepsy patients with MTS by using the terminal deoxynucleotidyl transferase (TdT) and digoxigenin-11-dUTP (TUNEL) method and immunohistochemistry for Bcl-2, Bax, and caspase-cleaved actin fragment, fractin. The temporal lobe specimens were obtained from 15 patients (six women and nine men; mean age, 29 ± 8 years). Results: Unlike that in normal adult brain, we observed Bcl-2 immunoreactivity in some of the remaining neurons dispersed throughout the hippocampus proper as well as in most of the reactive astroglia. Bax immunopositivity was increased in almost all neurons. Fractin immunostaining, an indicator of caspase activity, was detected in ,10% of these neurons. Des pite increased Bax expression and activation of caspases, we could not find evidence for DNA fragmentation by TUNEL staining. We also could not detect typical apoptotic changes in nuclear morphology by Hoechst-33258 or hematoxylin counterstaining. Conclusions: These data suggest that either apoptosis is not involved in cell loss in MTS, or a very slow rate of cell demise may have precluded detecting TUNEL-positive neurons dying through apoptosis. Increased Bax expression and activation of caspases support the latter possibility. [source] Sense of coherence and adjustment to divorce among Muslim Arab citizens of IsraelEUROPEAN JOURNAL OF PERSONALITY, Issue 4 2003Orna Cohen This study examined the distinctiveness and causal relationships of Antonovsky's (1979) Sense of Coherence Scale and the Mental Health Index of Veit and Ware (1983) in a sample of 306 divorced Muslim Arab men and women in Israel. The findings, based on confirmatory factor analysis and latent-variable structural modelling, suggest that in the present sample sense of coherence (SOC) and mental health are two independent but correlated constructs. They also suggest that, consistent with Antonovsky's theory, SOC may be better viewed as a mediator between stress and mental health than as a concurrent outcome of stress, although the latter possibility is not ruled out. Finally, the findings indicate that the SOC of the sample was relatively low in comparison with other groups. Copyright © 2003 John Wiley & Sons, Ltd. Nucleotide-sugars, the glycosyltransferase substrates, serve as activated donors of sugar residues during the enzymatic reaction Although very little is known about the catalytic mechanism of these enzymes, it appears that the catalytic activity in most glycosyltransferases is dependent upon the presence of a divalent cation, for example Mn2+ or Mg2+. It is not known whether the ion is bound to the enzyme before its interaction with the substrate, or if it binds the substrate before the enzymatic reaction to modify its conformation to fit better the active site of the enzyme. We have inspected the latter possibility by running four 2-ns molecular dynamics trajectories on fully solvated UDP-glucose in the presence of Mg2+ ions. Our results indicate that the divalent cation interacts strongly with the nucleotide-sugar in solution, and that it can alter its conformational behavior. It is also shown that a conformation of the pyrophosphate moiety that results in an eclipsed or almost eclipsed orientation of two of the oxygen atoms, and which is found in protein interacting with a nucleotide di- or tri-phosphate X-ray data, is energetically favored. The results are also discussed in light of existing NMR data, and are found to be in a good agreement with them. [source] Hypothalamic Vasopressin Gene Expression Increases in Both Males and Females Postpartum in a Biparental RodentJOURNAL OF NEUROENDOCRINOLOGY, Issue 2 2000Z. X. Wang In previous studies, the closely related neuropeptide hormones oxytocin and vasopressin have been implicated in the central mediation of parental behaviour. Several studies in rats and sheep have demonstrated a role for oxytocin in the initiation of maternal behaviour. Recently, a few studies in a biparental species, the prairie vole (Microxytocinus ochrogaster) have suggested that vasopressin is important for paternal care. The present study investigated this latter possibility by measuring changes in vasopressin and oxytocin hypothalamic gene expression 1 day and 6 days following parturition in prairie voles which show paternal care and in montane voles (M. montanus) which lack paternal care. In prairie voles, vasopressin gene expression increased in both males and females postpartum, relative to sexually naive controls. In the non-paternal montane vole, no change in vasopressin gene expression was observed in either sex. In contrast to this species difference in vasopressin gene expression, hypothalamic oxytocin gene expression increased in both prairie and montane vole females, but not in males of either species. To augment measures of gene expression, we assessed vasopressin (V1a) and oxytocin receptor binding in both species. Although forebrain vasopressin V1a receptor binding was not altered following parturition in either species, oxytocin receptor binding increased in the ventromedial nucleus of the hypothalamus in females, but not males, in both prairie and montane voles. In summary, vasopressin gene expression increases in both males and females postpartum in a biparental species and oxytocin gene expression and receptor binding increase selectively in females. These results are consistent with earlier reports of a role for vasopressin in paternal care and for oxytocin in maternal behaviour. [source] Correlated evolution of fruit and leaf size in bird-dispersed plants: species-level variance in fruit traits explained a bit further?OIKOS, Issue 3 2002Carlos M. Herrera The astounding morphological diversity exhibited by the fruits of vertebrate-dispersed plants has been traditionally interpreted as the adaptive outcome of divergent selective pressures exerted on plants by the broad array of frugivorous animals involved in seed dispersal. Although the selective capacity of frugivores provides support to this interpretation, recent studies have challenged it by documenting a strong phylogenetic component associated to interspecific variation in most fruit characteristics. Size-related fruit traits provide a conspicuous exception to this pattern, because they exhibit considerable variation at the between-species level which is largely independent of phylogeny and is correlated with consumption by differently-sized dispersal agents. Substantial species-level variance in size-related traits may reflect genuine disperser-driven diversification, but may also be partly influenced by correlated evolution of fruit size with the size of other plant structures. This latter possibility is tested here for bird-dispersed plants of the Iberian Peninsula using phylogenetically independent contrasts. Results demonstrate the existence of correlated evolution of fruit and leaf size at the species level. As all the plant taxa considered have their fruits eaten, and seeds dispersed, by the same relatively reduced set of frugivorous bird species, results suggest that a significant fraction of the variation in fruit size represented in the species sample may be explained as an indirect consequence of variation in leaf size, rather than being associated with adaptive divergence related to seed dispersal agents.
