Selected AbstractsSpatial analysis of an invasion front of Acer platanoides: dynamic inferences from static dataECOGRAPHY, Issue 3 2005Wei Fang It is an open question whether the invading tree species Acer platanoides is invading and displacing native trees within pre-existing forest stands, or merely preferentially occupying new stands of secondary forest growth at the edges of existing forests. Several threads of spatial pattern analyses were used to assess the invasibility of A. platanoides, and to link the invasion to the structure of a plant community in the deciduous forest of the northeastern United States. The analyses were based on maps of a contiguous 100×50 m area along an A. platanoides infestation gradient. The distribution of A. platanoides was highly aggregated and the population importance value increased from 28.1 to 38.5% according to mortality estimated from standing dead trees, while the distribution of native tree species was close to random and importance value of Quercus spp. decreased from 33.4 to 26.9% over time. The size distributions of each tree species across distance indicated that A. platanoides was progressively invading the interior of the forest while the native species (including A. rubrum) were not spreading back towards the A. platanoides monospecific patch. The null hypothesis of no invasibility was rejected based on quantile regressions. There were negative correlations between A. platanoides density and the densities of native species in different functional groups, and negative correlation of A. platanoides density and the species diversity in forest understory. The null hypothesis that A. platanoides invasion did not suppress native trees or understory was rejected based on Dutilleul's modified t-test for correlation, consistent with experimental results in the same study site. The combination of multiple spatial analyses of static data can be used to infer historical dynamical processes that shape a plant community structure. The concept of "envelop effects" was discussed and further developed. [source] Alteration of subcellular and cellular expression patterns of cyclin B1 in renal cell carcinoma is significantly related to clinical progression and survival of patientsINTERNATIONAL JOURNAL OF CANCER, Issue 4 2006Stephen O. Ikuerowo Abstract Cyclin B1, identified as a regulator of late cell cycle, is involved in the development and progression of a variety of human malignancies. To clarify the role of cyclin B1 in the pathogenesis and prognosis of renal cell carcinoma (RCC), protein expression was compared with clinicopathological characteristics of patients as well as the long-term survival after surgical therapy. Expression analysis was carried out by immunohistochemistry and tissue microarray analysis. The microarrays that represented the primary tumors, their invasion front and normal peritumoral renal parenchyma contained 753 tissue cores obtained from 251 randomly selected nephrectomy specimens. Immunopositivity within the primary tumors was significantly associated with tumor stage (pT) (p < 0.01), lymph node status (pN) (p < 0.01) as well as the presence of systemic metastatic disease (p = 0.01). Subcellular expression in the cytoplasm of tumor cells significantly correlated with pT (p = 0.02) and pN (p = 0.03). When peritumoral tissue samples exhibited a relative amount of <10% of positively reacting epithelial cells, cyclin B positivity was identified to predict long-term survival of patients in univariate analysis (p < 0.01) whereas borderline significance was observed in multivariate statistical analysis (p = 0.05). Increased intratumoral cyclin B1 positivity and aberrant localization of signals within the cytoplasm of tumor cells is positively correlated with the tendency towards tumor progression, indicating the significant role of cyclin B1 in the development and pathogenesis of RCC. The result of uni- and multivariate statistical analysis suggests the prognostic value of cyclin B1 for RCC patients. © 2006 Wiley-Liss, Inc. [source] General guidelines for invasive plant management based on comparative demography of invasive and native plant populationsJOURNAL OF APPLIED ECOLOGY, Issue 4 2008Satu Ramula Summary 1General guidelines for invasive plant management are currently lacking. Population declines may be achieved by focusing control on demographic processes (survival, growth, fecundity) with the greatest impact on population growth rate. However, we often have little demographic information on populations in the early stages of an invasion when control can be most effective. Here we determine whether synthesis of existing demographic data on invasive and native plant populations can address this knowledge problem. 2We compared population dynamics between invasive and native species using published matrix population models for 21 invasive and 179 native plant species. We examined whether the population growth rate responsiveness to survival, growth and fecundity perturbations varied between invasive and native species, and determined which demographic processes of invaders to target for reductions in population growth rate. 3Invaders had higher population growth rates (,) than natives, resulting in differences in demographic processes. Perturbations of growth and fecundity transitions (elasticities) were more important for population growth of invaders, whereas perturbations of survival had greater importance for population growth of natives. 4For both invasive and native species, elasticities of , to survival increased with life span and decreased with ,; while elasticities to growth and fecundity decreased with life span and increased with ,. 5For long-lived invaders, simulated reductions in either survival, growth or fecundity transitions were generally insufficient to produce population declines, whereas multiple reductions in either survival + growth or survival + fecundity were more effective. For short-lived invaders, simulated reductions in growth or fecundity and all pairwise multiple reductions produced population declines. 6Synthesis and applications. Life history and population growth rate of invasive species are important in the selection of control targets. For rapidly growing populations of short-lived invaders, growth and fecundity transitions should be prioritized as control targets over survival transitions. For long-lived invaders, simultaneous reductions in more than one demographic process, preferably survival and growth, are usually required to ensure population decline. These general guidelines can be applied to rapidly growing new plant invasions and at the invasion front where detailed demographic data on invasive species are lacking. Increased GLUT-1 expression has been reported in many human cancers. We hypothesized that a oral squamous cell carcinoma, characterized by high frequency of lymph node metastasis, distant metastasis or local recurrences, was associated with GLUT-1 overexpression in invasion front. Methods:, GLUT-1 immunostaining in invasion front was studied on 24 oral squamous cell carcinomas, and revealed the correlation with the clinical characteristics. Result:, The analysis showed that all oral squamous cell carcinoma patients and GLUT-1 expression correlated the depth of the tumors (P = 0.023 < 0.05). Furthermore the survival of patients who had overexpression of invasion front was significant shorter than that of patients with GLUT-1 weakly positive (P = 0.046 < 0.05). No significant association was noted between GLUT-1 immunostaining and either age, gender, subsites, tumor size, or lymph node status. Conclusion:, The present study shows that GLUT-1 served as a marker indicating that tumors with deep invasion tended to result in a worse prognosis in patients due to either lymph node metastasis, a recurrence of the primary lesion or distant metastasis. [source] Jaw bone remodeling at the invasion front of gingival squamous cell carcinomasJOURNAL OF ORAL PATHOLOGY & MEDICINE, Issue 1 2003Masahiro Ito Abstract Background:, It is still unknown how jaw bone remodeling occurs at actual invasion sites of oral squamous cell carcinomas. Since there is no other human carcinomas which make a direct invasion of the bone, gingival carcinomas are valuable examples. Methods:, Twelve surgical specimens of gingival squamous cell carcinoma were examined histopathologically and immunohistochemically for remodeling of bone and its surrounding tissue. Results:, Three types of bone interfaces with carcinomatous invasion were distinguished. These included areas with bone resorption, smooth bone surface and new bone formation. In the bone-resorption area, numerous osteoclasts were located along the bone surface, which was surrounded by myxoid stroma. The myxoid stroma was characterized by immunopositivity for heparan sulfate proteoglycan (HSPG), abundant vascularity and macrophagic infiltration. In the bone-formation area, rows of osteoblasts were aligned on the bone surface. The stroma around osteoblasts was also HSPG-immunopositive, poor in vascularity but rich in activated fibroblasts. In the smooth-bone area, the stroma showed an organizing phase of granulation tissue with slender fibroblasts and mature collagen fibers but with less vascularity and inflammatory infiltrates. Conclusion:, The results indicate that the stromal architecture, especially in terms of its inflammatory cellular, vascular and matrix compositions, is strictly regulated in the timing and site of jaw bone remodeling which is causes by carcinomatous invasion. [source] Transcriptional activation of the ,- catenin gene at the invasion front of colorectal liver metastases,THE JOURNAL OF PATHOLOGY, Issue 3 2009Obul R Bandapalli Abstract ,-Catenin is a pivotal molecule of the Wnt-signalling pathway, involved in regulation of developmental and oncogenic processes as well as in intercellular adhesion. So far, ,-catenin has been thought to be regulated mainly at the protein level. Here, we provide evidence for a transcriptional mechanism of ,-catenin regulation at the invasion front of colorectal liver metastases. In a nude mouse/LS174T cell xenograft model of colorectal liver metastases, we observed ,-catenin up-regulation at the mRNA and protein levels and a 13.7-fold increase of ,-catenin promoter activity in the cancer cells of the invasion front. In addition, the promoter activity was five-fold higher in the interior of the tumour than in cells growing in cell culture. In vitro studies revealed binding of TCF-4 to the ,-catenin promoter and reduced promoter activity by over-expression of dominant negative TCF-4, or ,-catenin knock-down and increased activity by ,-catenin over-expression, indicating that ,-catenin acts as co-transcription factor of its own promoter. In 55% (7/13) of clinical specimens, ,-catenin mRNA was markedly elevated in the cancer cells of the invasion front. Elevation of mRNA was paralleled by increased nuclear and cytoplasmic ,-catenin protein concentrations. These data indicate that transcriptional regulation contributes to the dynamic changes of ,-catenin levels upon the confrontation of tumour cells with the host microenvironment. [source] Native Australian frogs avoid the scent of invasive cane toadsAUSTRAL ECOLOGY, Issue 1 2009LÍGIA PIZZATTO Abstract Invasive species can affect the ecosystems they colonize by modifying the behaviour of native taxa; for example, avoidance of chemical cues from the invader may modify habitat use (shelter site selection) by native species. In laboratory trials, we show that metamorphs of most (but not all) native frog species on a tropical Australian floodplain avoid the scent of invasive cane toads (Bufo marinus Linnaeus 1758). Cane toads also avoid conspecific scent. This response might reduce vulnerability of metamorph frogs and toads to larger predatory toads. However, similar avoidance of one type of pungency control (garlic), and the presence of this avoidance behaviour in frogs at the toad invasion front (and hence, with no prior exposure to toads), suggest that this may not be an evolved toad-specific response. Instead, our data support the simpler hypothesis that the metamorph anurans tend to avoid shelter sites that contain strong and unfamiliar scents. Temporal and spatial differences in activity of frogs versus toads, plus the abundance of suitable retreat sites during the wet season (the primary time of frog activity), suggest that avoiding toad scent will have only a minor impact on the behaviour of native frogs. However, this behavioural impact may be important when environmental conditions bring toads and frogs into closer contact. [source] Spawning site selection by feral cane toads (Bufo marinus) at an invasion front in tropical AustraliaAUSTRAL ECOLOGY, Issue 5 2006MATTIAS HAGMAN Abstract Spawning sites are a critical and often scarce resource for aquatic-breeding amphibians, including invasive species such as the cane toad (Bufo marinus). If toads select spawning sites based on habitat characteristics, we can potentially manipulate those characteristics to either enhance or reduce their suitability as breeding sites. We surveyed 25 spawning sites used by cane toads, and 25 adjacent unused sites, in an area of tropical Australia recently invaded by these feral anurans. Water chemistry (pH, conductivity, salinity, turbidity) was virtually identical between the two sets of waterbodies, but habitat characteristics were very different. Toads selectively oviposited in shallow pools with gradual rather than steep slopes, and with open (unvegetated) gradually sloping muddy banks. They avoided flowing water, and pools with steep surrounds. In these respects, cane toads broadly resemble previously studied toad species in other parts of the world, as well as conspecifics within their natural range in South America. [source] Escape from natural enemies during climate-driven range expansion: a case studyECOLOGICAL ENTOMOLOGY, Issue 3 2008ROSA MENÉNDEZ Abstract 1.,A major, and largely unexplored, uncertainty in projecting the impact of climate change on biodiversity is the consequence of altered interspecific interactions, for example between parasitoids and their hosts. The present study investigated parasitism in the Brown Argus butterfly, Aricia agestis; a species that has expanded northward in Britain during the last 30 years in association with climate warming. 2.,Aricia agestis larvae suffered lower mortality from parasitoids in newly colonised areas compared with long-established populations. This result was consistent over four consecutive generations (2 years) when comparing one population of each type, and also when several populations within the historical and recently colonised range of the species were compared within a single year. Thus, A. agestis appears to be partially escaping from parasitism as it expands northwards. 3.,Reduced parasitism occurred despite the fact that several of the parasitoid species associated with A. agestis were already present in the newly colonised areas, supported predominantly by an alternative host species, the Common Blue butterfly, Polyommatus icarus. 4.,As the species expand their distributions into areas of increased climatic suitability, invasion fronts may escape from natural enemies, enhancing rates of range expansion. The results suggest that the decoupling of interspecific interactions may allow some species to exploit a wider range of environments and to do so more rapidly than previously thought possible. [source] Tenascin-C in primary malignant melanoma of the skinHISTOPATHOLOGY, Issue 4 2004S Ilmonen Aims :,To investigate the expression and the prognostic role of glycoprotein Tenascin-C (Tn-C) in primary melanoma of the skin. Methods and results :,The immunohistochemical expression of Tn-C was studied in 98 primary melanomas and related to inflammation, invasion, and patient outcome. Patients were followed up for disease recurrence for 0.04,7.4 years (median 3.9) and for survival for 0.5 to 12.1 years (median 9.3). The expression of Tn-C was evaluated for each tumour invasion border; the stromal and intracytoplasmic Tn-C of the melanoma islets were also recorded. Tn-C is widely expressed in primary melanoma samples, the staining pattern varying from focal to diffuse in different parts of the tumour. No correlation existed between intensity of Tn-C staining and inflammation. No stromal Tn-C was detected at the upper dermal lateral border in 12 patients, nor at the deep, dermal or subcutaneous border in 14 patients. These patients showed better disease-free survival (DFS) than did those cases with focal or diffuse staining (P = 0.06, P = 0.05). Also, absence of intracytoplasmic Tn-C was a beneficial prognostic factor for DFS (P = 0.04). In multivariate analysis, tumour ulceration and intracytoplasmic Tn-C expression of melanoma cells were independent adverse prognostic factors for DFS. Conclusions :,In primary melanoma of the skin, absence of Tn-C in the stroma of invasion fronts and within tumour cells seems to be related to a more benign disease behaviour with a lower risk of developing metastases.
