Translocation of viable Aeromonas salmonicida across the intestine of rainbow trout, Oncorhynchus mykiss (Walbaum)

Abstract The pathogenic bacterium Aeromonas salmonicida is the causative agent of the destructive disease furunculosis in salmonids. Horizontal transmission in salmonids has been suggested to occur via the skin, gills and/or intestine. Previous reports are contradictory regarding the role of the intestine as a route of infection. The present study therefore investigates the possibility of bacterial translocation across intestinal epithelia using Ussing chamber technology, in vitro. Intestinal segments were exposed for 90 min to fluorescein isothiocyanate-labelled pathogenic A. salmonicida. Sampling from the serosal side of the Ussing chambers showed that bacteria were able to translocate across the intestinal epithelium in both the proximal and distal regions. Plating and subsequent colony counting showed that the bacteria were viable after translocation. During the 90 min exposure to A. salmonicida, the intestinal segments maintained high viability as measured by electrical parameters. The distal region responded to bacterial exposure by increasing the electrical resistance, indicating an increased mucus secretion. This study thus demonstrates translocation of live A. salmonicida through the intestinal epithelium of rainbow trout, suggesting that the intestine is a possible route of infection in salmonids. Ligation of the caudal mesenteric artery during resection and anastomosis of the colorectal junction for annular adenocarcinoma in two dogs

An 8-year-old terrier cross and a 10-year-old German Shorthaired Pointer presented to the University Veterinary Centre, Sydney, for investigation of long-standing tenesmus and dyschezia. Both patients had an annular adenocarcinoma at the colorectal junction. Exploratory laparotomy was performed and the affected large intestinal segment was removed by resection and anastomosis. In both dogs, the caudal mesenteric artery was intimately associated with the mass, necessitating its ligation and transection. Postoperatively, there was no evidence of anastomosis breakdown in either case and both animals recovered well from surgery. The dogs were euthanased 8 and 10 months, respectively, after surgery because of clinical signs relating to metastatic disease. [source]


Effect of bile salts, lipid, and humic acids on absorption of benzo[a]pyrene by isolated channel catfish (Ictalurus punctatus) intestine segments

Abstract Dietary absorption of lipophilic contaminants may be a significant route of exposure in aquatic organisms. Bile salts, lipids, and humic acids are important factors that may influence the intestinal absorption of a contaminant such as benzo[a]pyrene (BaP). We hypothesized that bile salts, monoglycerides, and free fatty acids would increase BaP intestinal absorption, while triglycerides, humic acids, and sediment would decrease BaP intestinal absorption. We have established and validated an in vitro model to examine modification of 3H-BaP absorption in everted intestinal segments from channel catfish (Ictalurus punctatus). Uptake of BaP into the everted intestinal segments continued to increase over the times examined in this study (60 min) and apparently occurs passively; thus, fugacity-based models of uptake are supported. Absorption of BaP into intestinal cells was significantly decreased by the addition of monoglycerides and free fatty acids to bile salts in the incubation media. Addition of triglycerides decreased BaP absorption even further. Humic acids may have decreased BaP intestinal absorption, while natural sediment may have increased BaP absorption. The results of this study suggest that all lipids may decrease intestinal uptake of lipophilic contaminants if they remain in unabsorbable excess in the intestinal lumen by retaining BaP in lipid/bile micelles. In contrast, if triglycerides are hydrolyzed into monoglycerides/free fatty acids prior to absorption, lipophilic contaminant uptake will likely be facilitated. Thus, it may be the hydrolytic state of lipids that determines its effects on BaP absorption. Humic acids alone may decrease dietary uptake of BaP, but our results suggest that other components in natural sediment may counteract this effect to cause a slight enhancement of BaP uptake. Further studies are needed to determine the dietary conditions necessary for bio-accumulation to contribute significantly to lipophilic contaminant body burdens in benthivorous fish. Finally, the everted intestinal segment technique has the potential to be used in other species and with different contaminants. [source]


Long-term functional outcomes in patients with various types of orthotopic intestinal neobladder

Objectives: To evaluate the long-term functional outcomes of various types of orthotopic neobladder (NB). Methods: Eighty-nine patients who underwent orthotopic NB reconstruction after radical cystectomy and were followed for at least 60 months after surgery were included in this retrospective study. The types of NB in this series were: modified Studer type, Hautmann type, Mainz type, Goldwasser type and modified Reddy type in 36, 9, 15, 10 and 19 patients, respectively. Results: Sixty-seven (75.3%) patients could void spontaneously, 15 (16.8%) voided with clean intermittent catheterization (CIC) assistance and seven (7.9%) with CIC alone. After excluding the seven voiding with CIC alone, daytime and night-time continence was achieved in 63 and 50 of the remaining 82 patients, respectively. In addition, the proportion of daytime continence in the modified Studer group was significantly greater than that in the Mainz group, whereas there was no significant difference in the proportion of night-time continence among these groups. Uroflow examination indicated that despite the lack of significant differences in the maximal flow rates and the voided volumes, the post-void residual in the modified Reddy group was significantly smaller than that in the modified Studer group. Blood gas analysis in the 82 patients showed that there were no significant differences in the pH, bicarbonate, chloride and base excess among these groups. Conclusions: Orthotopic NB created with intestinal segments generally has acceptable functional outcomes on long term follow up. These outcomes differ according to the type of NB. To define the optimal procedure for NB reconstruction, it is necessary to understand the long-term functional prospects, on which the subjective success of NB creation is based. [source]


ORIGINAL ARTICLE: Intestinal villus histological alterations in broilers fed dietary dried fermented ginger

Summary To evaluate the effect of dietary dried fermented ginger (DFG) on intestinal villous histological alteration and growth performance, 64 Marshall Chunky male broilers were divided into four groups, each with four replicates of four chickens. Birds were fed the basal commercial mash diet supplemented with DFG at 0 (control), 5, 10 and 20 g/kg for 42 days. With increasing dietary DFG levels, feed intake tended to decrease and significantly decreased in the 20 g/kg DFG group (p < 0.05). Weight gain was higher in all the DFG groups, with the highest in the 10 g/kg DFG group (p < 0.05), resulting in an improved feed efficiency in all the DFG groups. Intestinal villus height, villus area, cell area and cell mitosis in all the intestinal segments were higher in all the DFG groups than in the control group. Protuberated cells and cell clusters were found in all the DFG groups, suggesting that the intestinal villi and cells might be hypertrophied. The present results indicate that dietary DFG can be used as a natural feed additive to induce broiler growth performance as a result of stimulation of morphological maturation and in consequence intestinal function. Comparison of total tract digestibility, development of visceral organs and digestive tract of Mong cai and Yorkshire × Landrace piglets fed diets with different fibre sources

Summary The aim of the study was to evaluate the effects of piglet age and dietary fibre source on the development of visceral organs and the gastrointestinal tract (GIT), and on growth performance and total tract apparent digestibility (TTAD) in local [pure-breed Mong cai (MC)] and exotic [Landrace × Yorkshire (LY)] piglets. The experimental diets contained different fibre sources: C (basal diet), RB (basal diet + rice bran), SPVM (basal diet + sweet potato vine meal) and CReM (basal diet + cassava residue meal). The neutral detergent fibre (NDF) content in diet C and the fibrous diets was 8.8% and 17.1%,17.7% respectively (dry matter basis). Collection of faecal samples to determine TTAD was carried out for five consecutive days before the experiment was finished (63 days). The piglets were killed at the age of 10 days (before being given the same solid feed), 30 days (weaning, 20 days after solid feed introduced) and 63 days (33 days after being given the different fibrous diets) when the length of intestinal segments, weight of organs (liver, heart, kidneys) and empty weight of the GIT (stomach, small intestine, caecum and colon + rectum) were measured. As the age of animals increased, the relative weight of organs and the length of intestines (expressed on a mass-specific basis) decreased (p < 0.05), and the weight of GIT increased (p < 0.001). The piglets fed fibrous diets had heavier GIT than those fed diet C with the highest values in CReM (p < 0.05). The colon + rectum length was not significantly different among C, RB and SPVM, but was shorter than in CReM (p < 0.05). Coefficient of total tract apparent digestibility (CTTAD) of nutrients in the fibrous diets was lower than in C (p < 0.01). Average daily gain (ADG) and feed conversion ratio (FCR) in C, RB and CReM were not different and were better than in SPVM (p < 0.01). There were no significant differences in the weights of organs between the two breeds at day 10, 30 and 63 (p > 0.05). The weight and length of GIT were not significantly different between the two breeds at day 10 and day 30, but were greater for MC at day 63. The caecum and colon + rectum at 10 and 30 days were longer in MC than in LY (p < 0.001). The relative development of GIT post-weaning was higher than pre-weaning, the difference being most apparent in MC. As a result at 63 days, MC had heavier visceral organs and GIT, and longer intestines on fibrous diets than LY (p < 0.05). The MC at 63 days had higher CTTAD of organic matter, gross energy, crude fibre and NDF (p < 0.001) and ether extract and crude protein (p < 0.05), but lower ADG and poorer FCR than LY (p < 0.001). It can be concluded that the GIT of the MC piglets developed more rapidly than LY when they were introduced to solid feed, and that the difference was more marked on the fibrous diets and after weaning, which resulted in higher total tract digestibility of nutrients in MC compared with LY. Cassava residue meal was better digested than RB and SPVM, and supported higher live weight gains. [source]


Influence of deoxynivalenol on the D -glucose transport across the isolated epithelium of different intestinal segments of laying hens

Summary Deoxynivalenol (DON) decreases glucose absorption in the proximal jejunum of laying hens in vitro and this effect is apparently mediated by the inhibition of the sodium d -glucose co-transporter. DON could modulate the sugar transport of other intestinal regions of chickens. For this purpose, we have measured the effects of DON on the Na+d -glucose co-transporter, by addition of DON after and before a glucose addition in the isolated epithelium from chicken duodenum, jejunum, ileum, caecum and colon by using the Ussing chamber technique in the voltage clamp technique. The data showed in all segments of the gut that the addition of d -glucose on the mucosal side produced an increase in the current (Isc) compared with the basal values, the Isc after glucose addition to the small intestine was greater than the Isc of the large intestine compared with the basal values, specially of the jejunum (p < 0.002), indicating that the jejunum is the segment that is the best prepared for Na+ - d -glucose co-transport. Further addition of 10 ,g DON/ml to the mucosal solution decreased the Isc in all segments and the Isc returned to the basal value, especially in the duodenum and mid jejunum (p < 0.05). In contrast, the addition of 5 mmol d -glucose/l on the mucosal side after incubation of the tissues with DON in all segments had no effect on the Isc (p > 0.05), suggesting that DON previously inhibited the Na+d -glucose co-transport. The blocking effects of DON in duodenum and jejunum were greater than the other regions of the gut. It can be concluded that the small intestine of laying hens has the most relevant role in the carrier mediated glucose transport and the large intestine, having non-significant capacity to transport sugars, appears to offer a minor contribution to glucose transport because the surface area is small. The effect of d -glucose on the Isc was reversed by DON in all segments, especially in the duodenum and jejunum, suggesting that DON entirely inhibited Na+ - d -glucose co-transport. This finding indicates that the inhibition of Na+ co-transport system in all segments could be an important mode of action for DON toxicity of hens. Zusammenfassung Deoxynivalenol (DON) erniedrigt in vitro die Glukoseabsorption im proximalen Jejunum von Legehennen. Dieser Effekt ist vermutlich durch eine Hemmung des Natrium- d -Glukose-Cotransportsystems bedingt. DON könnte außerdem den Glukosetransport in anderen Segmenten des Darms beeinflußen. Zu diesem Zweck haben wir Wirkungen von DON auf das Natrium- d -Glukose-Cotransportsystem gemessen, indem wir DON nach und vor einer Glukosezugabe auf isolierte Darmepithelien des Duodenums, Jejunums, Ileums, Caecums und des Kolons mittels der Ussing-Kammer-Technik in der Volt-Klemmtechnik einwirken ließen. Die erzielten Daten wiesen in allen Segmenten des Darms verglichen mit den Basalwerten einen Anstieg im Strom (Isc) auf, wobei die Isc des Dünndarms bei Glukosegabe signifikant größer als die des Dickdarms waren, was darauf hinweist, dass das Jejunum am besten für den Glukosetransport geeignet war. Eine DON-Zugabe von 10 ,g/ml zur mukosalen Lösung schwächte den Isc in allen Segmenten, wobei die Isc speziell im Duodenum und mittleren Jejunum zum Ausgangswert zurück kehrten. Im Gegensatz dazu brachte die mukosale Glukosezugabe nach der DON-Inkubation keinen signifikanten Anstieg der Isc (p > 0,05), was auf eine durch DON hervorgerufene Blockade des Natrium- d -Glukose-Cotransportsystems schließen ließ. Es kann daraus geschlossen werden, dass der Dünndarm von Legehennen den bedeutendsten Einfluß im Glukosetransportmechanismus nimmt und der Dickdarm aufgrund einer kleineren Oberfläche einen geringeren Beitrag zum Glukosetransport leistet. Dem Isc steigernden Effekt der Glukose konnte signifikant durch DON in den Darmsegmenten besonders im Duodenum und im Jejunum entgegen gewirkt werden, was auf eine umfassende Hemmung des Natrium- d -Glukose-Cotransportsystems hinweist. Die Resultate weisen darauf hin, dass eine Hemmung des Natrium- d -Glukose-Cotransportsystems in allen Darmsegmenten eine wichtige Rolle in der DON-Toxizität für die Henne darstellen könnte. Profile of P-glycoprotein distribution in the rat and its possible influence on the salbutamol intestinal absorption process

Abstract The intrinsic absorption of salbutamol in different intestinal segments of the rat was measured and related with the corresponding intestinal P-glycoprotein (P-gp) expression levels. The apparent absorption rate constants (ka, h,1) observed in each fraction by means of the "in situ" rat gut absorption method after perfusion of a 0.29-mM isotonic solution of salbutamol were used as absorption indexes. In a separate series of studies, a semiquantitative analysis of the mRNA expression of P-gp by means of polymerase chain reaction and Western blot with an antibody raised against the P-gp were also performed. The "in situ" ka values determined in the different segments (h,1) showed that the absorption is not homogeneous along the intestinal tract, that is, 0.499,±,0.054 for colon, 0.474,±,0.052 for the proximal segment, 0.345,±,0.014 for the mean, and 0.330,±,0.023 for the distal fraction. Addition of verapamil to the perfusion fluid did provide a better absorption of salbutamol in the distal segment. Heterogeneous expression of melatonin receptor MT1 mRNA in the rat intestine under control and fasting conditions

Abstract:, Melatonin is found in mammalian central nervous system and various peripheral tissues including gastrointestinal tract (GIT) where it participates in the regulation of intestinal motility, blood flow, immunomodulation, ion transport, cell proliferation and scavenging of free radicals. Some of these effects are achieved via melatonin binding to specific receptors, MT1 and MT2. As no thorough study on the expression of these receptors in the GIT has yet been done, the aim of this study was to determine the MT1 mRNA expression in the rat intestine under both control and fasting conditions. Our results suggest that MT1 mRNA is present in epithelial as well as subepithelial layer, with higher expression in the latter in all intestinal segments studied. The highest signal of the MT1 transcript along the rostro,caudal intestinal axis was found both in epithelial and subepithelial layers of the duodenum. Nevertheless, duodenal MT1 mRNA expression did not reach the level found in pituitary gland. In a 12:12-hr light:dark cycle a MT1 receptor expression in the subepithelial layer of rat distal colon did not manifest a significant diurnal rhythm. Short-term fasting increased the expression of MT1 transcript in the subepithelial layer of both the small and large intestine. During long-term fasting the increase persisted only in distal colon while a return to control levels was observed in small intestinal segments. In conclusion we demonstrated heterogeneous expression of MT1 receptor in the rat intestine and showed that its expression is up-regulated by nutritional deprivation. [source]


Motor patterns and propulsion in the rat intestine in vivo recorded by spatio-temporal maps

Abstract, We have used spatio-temporal maps derived from video images to investigate propagated contractions of the rat small intestine in vivo. The abdomen, including an exteriorized segment of jejunum, was housed in a humid chamber with a viewing window. Video records were converted to spatio-temporal maps of jejunal diameter changes. Intraluminal pressure and fluid outflow were measured. Contractions occupied 3.8 ± 0.2 cm of intestine and propagated anally at 3.1 ± 0.2 mm s,1 when baseline pressure was 4 mmHg. Contractions at any one point lasted 8.7 ± 0.6 s. Contractions often occurred in clusters; within cluster frequencies were 2.28 ± 0.04 min,1. Pressure waves, with amplitudes greater than about 9 mmHg, expelled fluid when the baseline pressure was 4 mmHg. In the presence ofl -NAME, circular muscle contractions occurred at a high frequency, but they were not propagated. We conclude that video recording methods give good spatio-temporal resolution of intestinal movement when applied in vivo. They reveal neurally-mediated propulsive contractions, similar to those previously recorded from intestinal segments in vitro. The propagated contractions had speeds of propagation that were slower and frequencies of occurrence that were less than speeds and frequencies of slow waves in the rat small intestine. [source]


Elective Surgical Patients as Living Organ Donors: A Clinical and Ethical Innovation

We propose a new model for living organ donation that would invite elective laparoscopic cholecystectomy patients to become volunteer, unrelated living kidney donors. Such donors would be surgical patients first and living donors second, in contrast to the current system, which ,creates' a surgical patient by operating on a healthy individual. Elective surgery patients have accepted the risks of anesthesia and surgery for their own surgical needs but would face additional surgical risks when a donor nephrectomy is combined with their cholecystectomy procedure. Because these two procedures have never been performed together, the precise level of additional risk entailed in such a combined approach is unknown and will require further study. However, considering the large number of elective cholecystectomies performed each year in the United States, if as few as 5% of elective cholecystectomy patients agreed to also serve as living kidney donors, the number of living kidney donors would increase substantially. If this proposal is accepted by a minority of patients and surgeons, and proves safe and effective in a protocol study, it could be applied to other elective abdominal surgery procedures and used to obtain other abdominal donor organs (e.g. liver and intestinal segments) for transplantation. In situ kinetic modelling of intestinal efflux in rats: functional characterization of segmental differences and correlation with in vitro results

Abstract The objective was to devise and apply a novel modelling approach to combine segmental in situ rat perfusion data and in vitro cell culture data, in order to elucidate the contribution of efflux in drug absorption kinetics. The fluoroquinolone CNV97100 was used as a model P-gp substrate. In situ intestinal perfusion was performed in rat duodenum, jejunum, ileum and colon to measure the influence of P-gp expression on efflux. Inhibition studies of CNV97100 were performed in the presence of verapamil, quinidine, cyclosporin A and p -aminohippuric acid. Absorption/efflux parameters were modelled simultaneously, using data from both in situ studies as well as in vitro studies. The maximal efflux velocity was modelled as a baseline value, corrected for each segment based on the expression level. CNV97100 passive diffusional permeability (Pdiff) and its affinity for the efflux carrier (Km) were assumed to be the same in all segments. The results indicate the new approach to combine in situ data and in vitro data succeed in yielding a unified, quantitative model for absorption/efflux. The model incorporated a quantitative relationship between P-gp expression level and the efflux functionality, both across in situ and in vitro systems, as well across different intestinal segments in the in situ studies. Permeability values decreased from duodenum to ileum in accordance with the increasing P-gp expression levels in rat intestine. The developed model reflects a strong correlation between in vitro and in situ results, including intrinsic differences in surface area. The successful application of a model approach to combine absorption data from two different experimental systems holds promise for future efforts to predict absorption results from one system to a second system. Permeability of intestinal mucosa from urinary reservoirs in man and rat

Objective To evaluate the barrier properties of intestinal mucosa chronically exposed to urine and to evaluate possible differences between ileal and colonic segments used in the reconstruction of the urinary tract. Materials and methods Mucosal specimens from patients with continent reservoirs with an abdominal stoma, or orthotopic neobladders constructed from colonic segments, were obtained at revisional surgery. Control segments were obtained during right-sided hemicolectomy. In addition, ileal and colonic segments from enterocystoplasties in rats were assessed. The mucosa-to-serosa passage of marker molecules, i.e. 14C-mannitol, 3H-glucose, fluorescein isothiocyanate-dextran 4400 and ovalbumin, was measured using modified Ussing diffusion chambers. Results In man, there were no permeability differences between segments exposed to urine and control segments for any of the marker molecules. In rats, there was less passage of markers in ileal and colonic transplanted segments than in intestinal segments from sham-operated animals. Conclusions Intestinal mucosa that has been in chronic contact with urine maintains its barrier function; in the rat model the permeability was even decreased. In addition, there were no detectable differences between ileal and colonic segments in this model. [source]
