Selected AbstractsReceptor architecture of human cingulate cortex: Evaluation of the four-region neurobiological modelHUMAN BRAIN MAPPING, Issue 8 2009Nicola Palomero-Gallagher Abstract The structural and functional organization of the human cingulate cortex is an ongoing focus; however, human imaging studies continue to use the century-old Brodmann concept of a two region cingulate cortex. Recently, a four-region neurobiological model was proposed based on structural, circuitry, and functional imaging observations. It encompasses the anterior cingulate, midcingulate, posterior cingulate, and retrosplenial cortices (ACC, MCC, PCC, and RSC, respectively). For the first time, this study performs multireceptor autoradiography of 15 neurotransmitter receptor ligands and multivariate statistics on human whole brain postmortem samples covering the entire cingulate cortex. We evaluated the validity of Brodmann's duality concept and of the four-region model using a hierarchical clustering analysis of receptor binding according to the degree of similarity of each area's receptor architecture. We could not find support for Brodmann's dual cingulate concept, because the anterior part of his area 24 has significantly higher AMPA, kainate, GABAB, benzodiazepine, and M3 but lower NMDA and GABAA binding site densities than the posterior part. The hierarchical clustering analysis distinguished ACC, MCC, PCC, and RSC as independent regions. The ACC has highest AMPA, kainate, ,2, 5-HT1A, and D1 but lowest GABAA densities. The MCC has lowest AMPA, kainate, ,2, and D1 densities. Area 25 in ACC is similar in receptor-architecture to MCC, particularly the NMDA, GABAA, GABAB, and M2 receptors. The PCC and RSC differ in the higher M1 and ,1 but lower M3 densities of PCC. Thus, multireceptor autoradiography supports the four-region neurobiological model of the cingulate cortex. Thermodynamic structure and AGN-driven shocks in M87MONTHLY NOTICES OF THE ROYAL ASTRONOMICAL SOCIETY, Issue 4 2010E. T. Million ABSTRACT We present the first in a series of papers discussing the thermodynamic properties of M87 and the central regions of the Virgo Cluster in unprecedented detail. Using a deep Chandra exposure (574 ks), we present high-resolution thermodynamic maps created from the spectra of ,16 000 independent regions, each with ,1000 net counts. The excellent spatial resolution of the thermodynamic maps reveals the dramatic and complex temperature, pressure, entropy and metallicity structure of the system. The ,X-ray arms', driven outwards from M87 by the central active galactic nuclei (AGN), are prominent in the brightness, temperature and entropy maps. Excluding the ,X-ray arms', the diffuse cluster gas at a given radius is strikingly isothermal. This suggests either that the ambient cluster gas, beyond the arms, remains relatively undisturbed by AGN uplift or that conduction in the intracluster medium (ICM) is efficient along azimuthal directions, as expected under action of the heat-flux-driven buoyancy instability (HBI). We confirm the presence of a thick (,40 arcsec or ,3 kpc) ring of high-pressure gas at a radius of ,180 arcsec (,14 kpc) from the central AGN. We verify that this feature is associated with a classical shock front, with an average Mach number M= 1.25. Another, younger shock-like feature is observed at a radius of ,40 arcsec (,3 kpc) surrounding the central AGN, with an estimated Mach number M, 1.2. As shown previously, if repeated shocks occur every ,10 Myr, as suggested by these observations, then AGN-driven weak shocks could produce enough energy to offset radiative cooling of the ICM. A high significance enhancement of Fe abundance is observed at radii 350,400 arcsec (27,31 kpc). This ridge is likely formed in the wake of the rising bubbles filled with radio-emitting plasma that drag cool, metal-rich gas out of the central galaxy. We estimate that at least ,1.0 × 106 solar masses of Fe has been lifted and deposited at a radius of 350,400 arcsec; approximately the same mass of Fe is measured in the X-ray bright arms, suggesting that a single generation of buoyant radio bubbles may be responsible for the observed Fe excess at 350,400 arcsec. [source] The evolution of tropical adaptation: comparing taurine and zebu cattleANIMAL GENETICS, Issue 5 2010E. K. F. Chan Summary Beef cattle breeds consist of three major genetic subdivisions. The taurine group is adapted to temperate environments, and the zebu and Sanga groups are both adapted to tropical environments. With the advent of genotyping and sequencing technologies in agriculture, genome-wide exploration of the genetic basis for the differences in tropical adaptation has only just become possible. In this study, approximately 9000 single nucleotide polymorphism markers were genotyped on 317 animals of a selection of taurine, zebu, and composite breeds to characterize any systematic differences between these groups. We identified 91 intra-breed-class markers; 78 were polymorphic only within the zebu animals, while 13 were polymorphic only in the taurine animals. There were no fixed differences (fixed for alternate alleles between the two breed types) between zebu and taurine animals. We found 14 regions with significantly different allele frequencies between zebu and taurine animals indicative of variable selection pressure or genetic drift. We also found 12 independent regions of differential extended haplotype homozygosity (EHH), indicative of recent selection or rapid fixation of the alternate allele within a short period of time in one of the two breed classes. A preliminary functional genomics analysis of these regions pointed towards signatures of tropical attributes including keratins, heat-shock proteins and heat resistance genes. We anticipate this investigation to be a stepping-stone for future studies to identify genomic regions specific to the two cattle groups, and to subsequently assist in the discrimination between temperate and tropically adapted cattle. [source] Association between the interleukin-1 family gene cluster and psoriatic arthritisARTHRITIS & RHEUMATISM, Issue 7 2006Proton Rahman Objective The interleukin-1 (IL-1) cytokine elicits a wide variety of biologic activities that initiate and promote an inflammatory response. The loci in the IL1 gene cluster have recently been associated with ankylosing spondylitis (AS). Since there is clinical and immunologic overlap between psoriatic arthritis (PsA) and AS, we wanted to examine the association between a panel of single-nucleotide polymorphisms (SNPs) in the IL1 gene family cluster and chromosome 2q12,13 in a PsA cohort. Methods Two hundred twelve PsA patients and 150 ethnically matched controls were genotyped with 11 SNPs in IL1A, 9 SNPs in IL1B, and 9 SNPs in IL1F5,10. Univariate analyses of the 29 single markers and short intragenic haplotypes identified several associated regions. Seventeen markers of interest were noted and further investigated to determine which markers or short haplotypes independently predict case,control status, using a stepwise logistic model. Results Two regions contributing independently to risk of disease in PsA were noted: a region spanned by markers rs3783547, rs3783543, and rs17561 in IL1A, and a region near the end of IL1B, through IL1F7, IL1F8, and into IL1F10. The best model contained markers rs3811047, rs1562304, and rs3811058, and 1 haplotype constructed from the 3 markers in region 1, with a likelihood ratio of 25.34 (4 degrees of freedom). Conclusion The IL1 locus appears to be a high-priority susceptibility locus in PsA, with at least 2 independent regions that confer increased risk. [source] Implementation in Australia of molecular diagnostic techniques for the rapid detection of foot and mouth disease virusAUSTRALIAN VETERINARY JOURNAL, Issue 7 2004DB BOYLE Objective To evaluate and implement rapid molecular diagnostic techniques for the detection of foot and mouth disease virus (FMDV) suitable for use in Australia. Design Two PCR TaqMan assays targeted to the FMDV internal ribosome entry site or the 3D polymerase coding region for the rapid detection of FMDV were evaluated using non-infectious materials to determine the test most appropriate for implementation as part of Australia's national preparedness for the rapid detection and diagnosis of FMD outbreaks. Results Two published tests (PCR TaqMan assays targeted to the FMDV IRES region or the FMDV 3D polymerase coding region) were evaluated for their ability to detect FMDV genetic material in non-infectious FMDV ELISA antigen stocks held at Australian Animal Health Laboratory. Both tests were able to detect FMDV genetic material from strains O1 Manisa, O-3039, A22, A24, A Malaysia, C, Asia 1 and SAT 1, 2 and 3. With the exception of Asia 1, the TaqMan assay targeted to the FMD 3D polymerase coding region had Ct values equal to or lower than for the TaqMan assay targeted to the IRES region suggesting that this test may provide broader serotype detection and sensitivity. However, the TaqMan assay directed to the FMDV IRES is the only one to date to have undergone substantial evaluation using clinical samples collected during an outbreak. The greatest differences observed were for O-3039, SAT 1, and 3. Conclusion Given the ease of setting up both tests, AAHL currently runs both tests on highly suspect FMD investigations to provide independent confirmation of the absence of FMDV because the tests are focused on two independent regions of the FMDV genome. These tests add substantially to Australia's preparedness for FMD diagnosis complementing the already well-established virus isolation and antigen capture ELISA tests for index case diagnosis of FMD in Australia.
