Increased Consumption but Not Operant Self-administration of Ethanol in Mice Lacking the RII, Subunit of Protein Kinase A

Background: Accumulating evidence indicates that adenosine monophosphate (cAMP)-dependent protein kinase A (PKA) is involved in the neurobiological responses to ethanol. Previous reports indicate that mice lacking the RII, subunit of PKA (RII,,/,) voluntarily consume more ethanol than wild-type controls (RII,+/+) using 2-bottle testing procedures. Although such procedures primarily measure consummatory behavior, operant self-administration procedures allow analysis of consummatory as well as appetitive or "ethanol-seeking" behavior (i.e., lever pressing is required to gain access to the ethanol solution). Therefore, we determined whether the high ethanol consumption characteristic of RII,,/, mice would be complemented by increased appetitive ethanol-seeking behavior in an operant paradigm. Methods: RII,,/, (n=8) and RII,+/+ (n=8) mice were initially sucrose-faded until they were lever responding for nonsweetened ethanol (10, 14, and 18%). Following the self-administration testing, RII,+/+ and RII,,/, mice were given access to 2 bottles, one containing water and the other ethanol to replicate the voluntary ethanol drinking data previously from our laboratory. Finally, immediately after voluntary consumption all mice were again tested for self-administration of 10% ethanol. Alterations in the reinforcement schedule were also explored as RII,+/+ and RII,,/, mice were tested for self-administration of 10% ethanol at FR-3 and FR-5 schedules. Results: The RII,,/, mice displayed lower operant responding for ethanol and food reinforcement compared with RII,+/+ controls. However, this effect was driven by a significant increase in lever responses made by female RII,+/+ mice. When the excessive lever responses of the female RII,+/+ mice are accounted for, the RII,,/, mice show ethanol lever responses comparable to controls. Following operant self-administration testing, RII,,/, mice of both sexes consumed more ethanol solution compared with RII,+/+ mice during 2-bottle testing. Conclusions: Increased ingestion of ethanol by RII,,/, mice is likely the result of altered PKA activity within neuronal pathways that control ethanol-consummatory behaviors. Conversely, the RII, subunit of PKA appears not to play a critical role in neuronal pathways that regulate appetitive behaviors directed at obtaining ethanol. Finally, increased operant self-administration of food and ethanol by female wild-type mice was absent in female RII,,/, mice, suggesting that normal PKA signaling may be part of a general, and sex-dependent, mechanism involved with reinforcement-seeking behavior. Physicochemical, Nutritional, and Functional Characterization of Fruits Xoconostle (Opuntia matudae) Pears from Central-México Region

Abstract:, Xoconostle cv. Cuaresmeño (Opuntia matudae) has attracted domestic and international industry attention; however, variations of composition from xoconostle structures have not been evaluated. Industries discard the pulp (endocarp) and peel (pericarp) as wastes and utilize the skin (mesocarp), which is the edible portion. The physicochemical, nutritional, and functional characterization of structures from xoconostle pear from 3 major sites of production in Mexico were assessed. Skin yield ranged from 58% to 64% and was higher to that of peel (22% to 24%) and pulp (12% to 18%) yields. pH, °Brix, and acidity were similar among xoconostle structures. Total fiber showed by peel (18.23% to 20.37%) was 2-fold higher than that of skin. Protein and ether extract were higher in xoconostle pulp compared to that showed by peel and skin. Iron content of xoconostle peel (6 to 9.6 mg/100 g, DWB) was higher to that of skin and pulp and prickly pear pulp. Soluble phenols of peel (840 to 863 mg GAE/100 g, DWB) were almost similar to that of skin (919 to 986 mg GAE/100 g, dry weigh basis); meanwhile, ascorbic acid concentration of skin was 2-fold higher compared to that of peel. The phenolic fraction of xoconostle structures consisted of gallic, vanillic, and 4-hydroxybenzoic acids; catechin, epicatechin, and vanillin were also identified by high-performance liquid chromatography,didoe array detection (HPLC-DAD). Xoconostle peel showed higher antioxidant activity (TEAC) compared to that of skin (2-fold) and pulp (6-fold) of commonly consumed fruits and vegetables. The potential of xoconostle peel and pulp for the production of feed or food is promissory. Practical Application:, Outstanding nutritional and functional properties of xoconostle cv. Cuaresmeño fruits are demonstrated. Increased consumption could contribute positively to improve the diet of rural and urban consumers. The high fiber, mineral, and antioxidant components of xoconostle peel and pulp suggest that these fruit structures, which are currently discarded as waste, have promissory use as feed or food by industry. A LEXICON FOR TEXTURE AND FLAVOR CHARACTERISTICS OF FRESH AND PROCESSED TOMATOES

ABSTRACT A lexicon for describing the sensory flavor and texture characteristics of fresh and processed tomatoes was developed. A six-member highly trained, descriptive sensory panel identified, defined and referenced 33 sensory attributes for fresh and processed tomatoes. Forty products including a variety of raw, canned, concentrated and dried tomatoes, as well as tomato-based products including ketchup and simple pasta sauce, were evaluated in the study. These products represented a wide range of sensory characteristics in raw and processed tomatoes. The lexicon established included 5 aroma attributes, 10 texture attributes and 18 flavor attributes including 6 taste and mouthfeel attributes. The lexicon provides attribute descriptors, definitions and references that often are lacking in previous literature. Reducing the number of attributes may be appropriate when testing specific tomato products. PRACTICAL APPLICATIONS Increased consumption of tomato products has expanded interest in improving the sensory characteristics of tomatoes and subsequent products. This research provides a list of sensory flavor and texture terms that can be used to describe the sensory characteristics of both fresh and processed tomatoes. This information will help researchers, breeders and processors better understand the flavor and texture properties of fresh and processed tomatoes, and the tomato characteristics of tomato products. Taste attributes, often used in previous literature, are important but are not enough to describe the characteristics of tomatoes. In addition to flavor attributes, aroma and texture properties are important for describing tomato characteristics but have not been thoroughly established in previous research. [source]


Macular pigment optical density at four retinal loci during 120 days of lutein supplementation

Abstract Background:, Increased consumption of lutein and zeaxanthin has been shown to increase macular pigment optical density (MPOD) in some individuals. Most interventions either obtained infrequent measures of MPOD or measured MPOD at a single retinal locus. Purpose:, The aim of this study was to measure acute changes in MPOD at four retinal loci during lutein intervention. Methods:, For 120 days, three subjects consumed 30 mg of lutein and 2.7 mg of zeaxanthin supplement per day. MPOD was measured with heterochromatic flicker photometry at 20,, 30,, 60, and 120, eccentricity three or four times per week. High-performance liquid chromatography was used to measure serum carotenoid concentrations in blood samples collected at baseline and at 30-day intervals. Results:, At the two most central loci, MPOD significantly increased in all three subjects with a mean change of approximately 0.09 log units at 20, eccentricity and 0.08 log units at 30, eccentricity. MPOD significantly increased in two subjects at 60, eccentricity, and in one subject at 120, eccentricity. The increases in MPOD appeared to be linear and continued after treatment was ended. In all three subjects, log sensitivity at the reference locus decreased linearly. Serum lutein and serum zeaxanthin increased from baseline, reaching peak concentrations after 30 days of supplementation. Conclusion:, The changes in MPOD suggest that carotenoid deposition occurs linearly and may be biased towards the central retina. Further, carotenoid deposition may occur outside the central fovea in interventions with pharmacological doses of carotenoid, resulting in underestimations of psychophysical measures of MPOD. Effects of elevated CO2 associated with maize on multiple generations of the cotton bollworm, Helicoverpa armigera

Abstract Under elevated environmental carbon dioxide (CO2), leaf chewers tend to compensate for decreased leaf nutritional quality with increased consumption; mortality and development times also increase and cause a reduction in the fitness of leaf chewers. However, the effect of elevated CO2 on multiple successive generations of these and other insects is not well understood. Furthermore, information about the direct effects of increased environmental CO2 on developmental time and consumption of herbivores is lacking. In this paper, we tested the hypothesis that cascade effects of elevated CO2 through plants, rather than the direct effects of elevated CO2, are the main factors decreasing the fitness of cotton bollworm, Helicoverpa armigera Hübner (Lepidoptera: Noctuidae). We used two series of experiments to quantify the growth, development, and consumption of H. armigera fed on an artificial diet or C4 plants (maize) grown under two CO2 levels (ambient vs. double ambient). In the first series of experiments, elevated CO2 had no effect on the population abundance or individual consumption for three successive generations of cotton bollworms fed on an artificial diet. In the second series of experiments, elevated CO2 reduced population abundance of cotton bollworm larvae for two successive generations when they were fed maize milky grains. The specific effects were longer larval duration, lower fecundity, and decreased rm of cotton bollworms. Furthermore, elevated CO2 increased individual consumption when cotton bollworm was fed maize milky grains for two successive generations and decreased the population's total consumption in the first generation but increased it in the second generation. The results from this study indicate that: (1) The effects of elevated CO2 on three successive generations of cotton bollworm fed on artificial diet were weak, or even non-existent, and (2) elevated CO2 increased the consumption when cotton bollworm were fed maize. Our study also suggests that the damage inflicted by cotton bollworm on maize (a C4 plant) will be seriously affected by the increases in atmospheric CO2, which is unlike our previous results for spring wheat (a C3 plant). Reach-scale geomorphology affects organic matter and consumer ,13C in a forested Piedmont stream

Summary 1. We investigated the spatial (longitudinal position and reach geomorphology) and seasonal (spring and autumn) influences on the variation of ,13C among organic matter sources and consumers in a forested Piedmont river, South Carolina, U.S.A. 2. Six sites were sampled along a continuum and varied in basin area from approximately 30 to 300 km2. Sites fell into two geomorphic categories (i) high-gradient, rock bed (,rock') or (ii) low-gradient, sand bed (,sand') sites. 3. Variation in ,13C was more strongly related to reach geomorphology than longitudinal position. ,13C of biofilm and consumers was consistently enriched at rock sites. Leaf litter (i.e. coarse particulate organic matter, CPOM) ,13C did not vary with bed type. There was significant ,13C enrichment at rock sites for biofilm, seston, fine benthic organic matter (FBOM), and eight of nine consumer trophic guilds (e.g. grazing invertebrates, insectivorous fishes). ,13C of biofilm and four trophic guilds was also positively correlated with drainage area, but the magnitude of enrichment was less than between bed types. 4. ,13C was generally enriched in spring, but this varied among organic matter types, consumers, and by bed type. CPOM and seston were enriched in spring, FBOM was enriched in autumn, and biofilm showed no trend. Five consumer guilds were enriched in spring, and only one fish guild, generalised carnivores, showed enrichment of muscle tissue in autumn. 5. Consumer ,13C enrichment at rock sites suggests greater reliance on algal carbon than for consumers at sand sites, but we also found ,13C enrichment of biofilm at rock sites. Thus, differences in consumer ,13C between bed types could be related to (i) increased consumption of biofilm at rock compared with sand sites, or (ii) consumption of biofilm at rock sites that is enriched relative to biofilm at sand sites or (iii) both mechanisms. 6. ,13C signatures in local food webs appear to respond to processes operating at multiple spatial scales. Overall downstream enrichment of biofilm and consumers was disrupted by strong local effects related to bed morphology. These results suggest that human alteration of channel habitat will have corresponding effects on stream food webs, as assessed by changes in ,13C. [source]


Production and sensory characteristics of flavoured soymilk samples

Abstract The sensory quality characteristics of four flavoured soymilk samples including vanilla, banana, coffee and chocolate were evaluated at varying concentrations of the flavour. This was done to establish the most acceptable flavour and desired concentration for commercial production of soymilk and also to facilitate increased consumption of soymilk for improved nutrition. Hot extraction of the milk from blanched soybeans was achieved by blending in hot water and sieving through muslin cloth. Different concentration levels of the flavours being assessed, namely chocolate, coffee, vanilla and banana, were added. Consumer preference for the various samples was assessed using a nine-point hedonic scale. Addition of flavours was found to improve the sensory characteristics and consumer preference of soymilk. Colour, taste, aroma, mouthfeel and hence overall acceptability of the soymilk samples were improved significantly by the addition of vanilla, banana, coffee and chocolate flavours. The maximum concentrations of the individual flavours for optimal sensory impact were established. Based on the results, the following flavour concentrations are recommended per 100 ml of soymilk: 0.03% vanilla, 0.01% banana, 1.5% coffee and 4% chocolate. [source]


Photoallergic contact dermatitis caused by ultraviolet filters in different sunscreens

Over the last decade, a change in the public awareness regarding the possible danger of excessive sunlight exposure has resulted in an increased consumption of sunscreens. These products contain a broad spectrum of putative sensitizers that can cause contact dermatitis and, upon exposure to ultraviolet (UV) irradiation, photocontact dermatitis. Among these sensitizing compounds, UV filters are the most frequent cause of photoallergic reactions. Although rarely observed, we here describe the occurrence of a photoallergic contact dermatitis in a 55-year-old man after the use of two different sunscreens. Photopatch testing showed hypersensitivity reactions of the delayed type against three different chemical UV filters, 4-tert-butyl-4- methoxy-dibenzoylmethane (Parsol 1789), 2-ethylhexyl-p-methoxycinnamate (Parsol MCX), and isoamyl-p-methoxycinnamate (Neoheliopan). Response of multiple generations of cotton bollworm Helicoverpa armigera Hübner, feeding on spring wheat, to elevated CO2

Abstract:, The growth, development and consumption of three successive generations of cotton bollworm, Helicoverpa armigera (Hübner), reared on milky grains of spring wheat grown under elevated CO2 (double-ambient vs. ambient) in open-top chambers (OTCs) were examined. Decreases in protein, total amino acid, water and nitrogen content, and increases in total non-structure carbohydrates (TNCs) and ratio of TNC : nitrogen were found in wheat milky grains grown under elevated CO2 conditions. Changes in quality of wheat grains affected the growth, development and food utilization of H. armigera. Significantly longer larval lifespan for the third generation and lower pupal weight for all generations were observed in cotton bollworm fed on milky grains of spring wheat grown under elevated CO2 conditions. Bollworm fecundity was significantly decreased for the second and third generations under elevated CO2 levels. The consumption, frass per larva and relative consumption rate significantly increased in elevated CO2 compared with ambient CO2 conditions. However, the potential population consumption was significant reduced by elevated CO2 in the second and third generations. The results of this study indicate that elevated CO2 levels adversely affect grain quality, resulting in consistently increased consumption per larva for a longer period to produce less fecund bollworm through generations, suggesting that net damage of cotton bollworm on wheat will be less under elevated atmospheric CO2 levels because increased consumption is offset by slower development and reduced fecundity. Size-related shifts in dietary composition of Centropomus parallelus (Perciformes: Centropomidae) in an estuarine ecosystem of the southeastern coast of Brazil

Summary Size-related and seasonal evaluation of the dietary composition of fat snook (Centropomus parallelus Poey 1860) in the upper sector of an estuary of the southeastern coast of Brazil were carried out based on stomach analyses of specimens ranging from 40 to 170 mm standard length. Results reveal that C. parallelus is a carnivorous species feeding mainly on benthic crustaceans. Relatively high stomach replenishment suggests that this environment is an important feeding ground for fat snook juveniles. Multivariate analyses indicated that predator size effect is significantly more important than seasonal variation in determining dietary composition. Predator length was associated with increased consumption of palaemonid shrimps (Macrobrachium spp.) and grapsid crabs, and decreased foraging on tanaids (Kalliapseudes schubarti), thus showing a preference shift from smaller to larger prey. Predator length was also positively associated with an increase in the stomach repletion index. Additionally, allometric growth of both gape and head were consistently correlated with this ontogenetic dietary transition, suggesting that such changes might be related to an individual's ability to capture and consume larger, more elusive prey. The digestive tube is short and grows isometrically, which is in accordance with the carnivorous habit of this estuarine fish and its maintenance through ontogeny. [source]


Regional obesity determinants in the United States: a model of myopic addictive behavior in food consumption

Myopic addiction; Obesity; Sweet foods Abstract Obesity is considered one of the largest public health problems in the United States today. The premise for our study is a body of results from medical research showing that sweetened foods, i.e., an increased consumption of sugars, leads first to sugar addiction and second to carbohydrate addiction and increased consumption of fats. The latter feature is actually responsible for the increase in body mass index (BMI), but the trigger that produces cravings for extra calories is sugar and sweeteners. Based on our results, a myopic model of addictive behavior in food consumption seems to capture the food consuming habits and related outbreak of obesity among the American population. Our results indicate that lower current and past real prices of sugar contribute significantly to higher values of BMI, and increase the likelihood of becoming obese in the United States. [source]


WATER QUALITY IMPACTS AND INDICATORS OF METABOLIC ACTIVITY OF THE ZEBRA MUSSEL INVASION OF THE SENECA RIVER,

ABSTRACT: The conspicuous shifts in summertime values of common measures of water qualify that have persisted for 10 years (1993 to 2002) in the Seneca River, New York, as a result of the zebra mussel invasion are documented. Resolution of patterns in time and space is supported by water quality monitoring that extends back to the late 1970s. Patterns are evaluated to describe the stability of impacts and quantify metabolic activity of the invader. The water quality impacts that have persisted unabated for 10 years since the invasion are the most severe documented for a river in North America. Changes in summer median conditions since the invasion include: (1) a 16-fold decrease in chlorophyll concentration (Chi), (2) a 2.5-fold increase in Secchi disc transparency, (3) a 17-fold increase in soluble reactive phosphorus concentration, (4) a 3.7-fold increase in total ammonia concentration, (5) a greater than 25 percent decrease in dissolved oxygen (DO) concentration, and (6) a decrease in pH of 0.55 units. The strength of these signatures has been driven by anthropogenic influences that include upstream nutrient loading and morphometric modifications of the river, and the functioning of Cross Lake, through which the river flows. This hypereutrophic lake sustains dense zebra mussel populations and related water quality impacts in the river downstream of the lake outflow by acting as a source of veligers and suitable food for this bivalve. Evidence is presented that levels of metabolic activity of the zebra mussel in this river have been resource limited, manifested through increased consumption of Chl and DO with increased delivery of these constituents in the lake's outflow. High-protein Weight-loss Diets: Are They Safe and Do They Work?

A Review of the Experimental, Epidemiologic Data
Recommendations for increased consumption of protein are among the most common approaches of popular or fad diets. This review summarizes the effects of dietary protein on satiety, energy intake, thermogenesis, and weight loss, as well as its effect on a variety of health outcomes in adults. In short-term studies, dietary protein modulates energy intake via the sensation of satiety and increases total energy expenditure by increasing the thermic effect of feeding. Whereas these effects did not contribute to weight and fat loss in those studies in which energy intake was fixed, one ad libitum study does suggest that a high-protein diet results in a greater decrease in energy intake, and therefore greater weight and fat loss. In terms of safety, there is little long-term information on the health effects of high-protein diets. From the available data, however, it is evident that the consumption of protein greater than two to three times the U.S. Recommended Daily Allowance contributes to urinary calcium loss and may, in the long term, predispose to bone loss. Caution with these diets is recommended in those individuals who may be predisposed to nephrolithiasis or kidney disease, and particularly in those with diabetes mellitus. [source]


Preventive effects of drinking green tea on cancer and cardiovascular disease: Epidemiological evidence for multiple targeting prevention

Abstract The significance of drinking green tea in prevention of two of the main lifestyle-related diseases, cancer and cardiovascular disease, was demonstrated in terms of a prospective cohort study on a total of 8,552 general residents in Saitama Prefecture, Japan. On the basis of the follow-up study, we revealed decreased relative risk of cancer incidence for those consuming over 10 cups a day, compared with those consuming below 3 cups: 0.54 (95% men, 0.57 (0.34,0.98) for women, and 0.59 (0.35,0.98) for both sexes. Furthermore, a significant delay in cancer onset was associated with increased consumption of green tea. Next, decreased relative risk of death from cardiovascular disease was 0.58 (0.34,0.99) for men, 0.82 (0.49,1.38) for women, and 0.72 (0.60,1.04) for members of both sexes consuming over 10 cups a day. Finally, we evaluated the life-prolonging effects of drinking green tea on cumulative survival, using the life table. [source]

Abstract BACKGROUND Neural tube defect (NTD) rates can be lowered by increased consumption of folic acid (FA) by women before and during early pregnancy. The crude proportion of reproductive-aged women taking FA supplements has been used to predict a decline of the NTD rate in the general population. In this study we examine the potential error in using the crude proportion to predict NTD risk reduction, and offer an alternative method. METHODS The crude proportion measures the number of women taking FA. It ignores the substantial variability by maternal age in the probability of giving birth. Age-specific fertility rates (ASFRs) reflect the probability that a woman in a specific age group will give birth in a given year. In this study, we show how to calculate a proportion weighted by ASFRs to predict a decline in the NTD rate, and to assess the effectiveness of FA consumption in preventing NTDs. RESULTS Our results show that a crude proportion of 50% of women (15,49 years old) taking FA is associated with a range of 24,77% in weighted proportions. Assuming a 40% risk reduction from taking 400 ,g of FA daily, the expected NTD rate decline could vary from 9.6% to 30.6%, depending on the age distribution of women taking FA. CONCLUSIONS The ASFR-weighted proportion estimates the proportion of babies born to women taking FA, as opposed to the crude proportion of women taking FA. We recommend using the ASFR-weighted proportion to predict an NTD rate decline and measure the success of FA education campaigns. We found that when women in high-fertility age groups increased their FA consumption, the decline in the NTD rate was greater than when women in low-fertility age groups did so. Our findings suggest that the more efficient approach to NTD prevention is to focus on women with a higher probability of giving birth. For example, by focusing on <50% of women of childbearing age (20,34 years), as much as 76% of the maximum NTD rate reduction can be achieved. Birth Defects Research (Part A), 2003.
