Selected AbstractsEthanol-Induced Increase of Agouti-Related Protein (AgRP) Immunoreactivity in the Arcuate Nucleus of the Hypothalamus of C57BL/6J, but not 129/SvJ, Inbred MiceALCOHOLISM, Issue 4 2010Inmaculada Cubero Background:, The melanocortin (MC) system is composed of peptides that are cleaved from the polypeptide precursor, pro-opiomelanocortin (POMC). Previous research has shown that MC receptor (MCR) agonists reduce, and MCR antagonists increase, ethanol consumption in rats and mice. Consistently, genetic deletion of the endogenous MCR antagonist, agouti-related protein (AgRP), causes reductions of ethanol-reinforced lever pressing and binge-like ethanol drinking in C57BL/6J mice. Ethanol also has direct effects on the central MC system, as chronic exposure to an ethanol-containing diet causes significant reductions of ,-melanocyte stimulating hormone (,-MSH) immunoreactivity in specific brain regions of Sprague-Dawley rats. Together, these observations suggest that the central MC system modulates neurobiological responses to ethanol. To further characterize the role of the MC system in responses to ethanol, here we compared AgRP and ,-MSH immunoreactivity in response to an acute injection of saline or ethanol between high ethanol drinking C57BL/6J mice and moderate ethanol drinking 129/SvJ mice. Methods:, Mice received an intraperitoneal (i.p.) injection of ethanol (1.5 g/kg or 3.5 g/kg; mixed in 0.9% saline) or an equivolume of 0.9% saline. Two hours after injection, animals were sacrificed and their brains were processed for AgRP and ,-MSH immunoreactivity. Results:, Results indicated that acute ethanol administration triggered a dose-dependent increase in AgRP immunoreactivity in the arcuate (ARC) of C57BL/6J mice, an effect that was not evident in the 129/SvJ strain. Although acute administration of ethanol did not influence ,-MSH immunoreactivity, C57BL/6J mice had significantly greater overall ,-MSH immunoreactivity in the ARC, dorsomedial, and lateral regions of the hypothalamus relative to the 129/SvJ strain. In contrast, C57BL/6J mice displayed significantly lower ,-MSH immunoreactivity in the medial amygdala. Conclusions:, The results show that acute ethanol exposure has direct effects on endogenous AgRP activity in ethanol preferring C57BL/6J mice. It is suggested that ethanol-induced increases in AgRP may be part of a positive feedback system that stimulates excessive binge-like ethanol drinking in C57BL/6J mice. Inherent differences in ,-MSH immunoreactivity may contribute to differences in neurobiological responses to ethanol that are characteristically observed between the C57BL/6J and 129/SvJ inbred strains of mice. [source] Relative Efficiency of Price Discovery on an Established New Market and the Main Board: Evidence from KoreaASIA-PACIFIC JOURNAL OF FINANCIAL STUDIES, Issue 4 2010Kyong Shik Eom G10; G14; G15 Abstract We examine the relative efficiency of price discovery between the new market (KOSDAQ) and the main board (KOSPI) in the Korean stock markets that have the same trading mechanism (i.e. electronic limit-order book), focusing on the comparisons of each market's efficiency of price discovery in three aspects: speed, degree, and accuracy. We find that, for our entire firm sample, price discovery on KOSDAQ is less efficient than on KOSPI. However, the price discovery of the most liquid group (top 40 stocks) on KOSDAQ turns out to be as efficient as the lowest group (top 160th,200th stocks) among the top 200 liquid stocks on KOSPI. These two quintiles are comparable in terms of their firm characteristics, so it appears that the greater overall efficiency of price discovery on KOSPI is due to the characteristics of its listed firms, rather than any inherent difference between a main board and a new market. We also find evidence that the speed of price discovery is mainly determined by turnover, whereas the accuracy of price discovery is mainly determined by turnover and intraday volatility. All together, our results provide some policy implications for developing or even developed countries eager to establish a viable new market. First, price discovery in a successful or viable new market in an emerging economy behaves as predicted in the market microstructure literature, even though that literature is based primarily on main boards in advanced stock markets. Second, price discovery in the most liquid group in a new market is more accurate, although slower, than in the lowest group among the liquid stocks on a main board; on balance, the main board and new market are comparable. Finally, the accuracy of price discovery is more (less) impacted by turnover (intraday volatility) on the new market than on the main board. [source] Do Male Veiled Chameleons, Chamaeleo calyptratus, Adjust their Courtship Displays in Response to Female Reproductive Status?ETHOLOGY, Issue 6 2002Erin C. Kelso Variation in male courtship behavior may be due to inherent differences among males or may arise from males adjusting their courtship displays according to female responsiveness. Female veiled chameleons, Chamaeleo calyptratus, exhibit two distinctive suites of body coloration and behavior patterns that vary according to receptive and non-receptive stages of their reproductive cycle. We presented male chameleons with both receptive and non-receptive females, and recorded differences in their mating frequency, courtship intensity and courtship behavior patterns. As expected, males were more likely to court and attempt mating with receptive females. Although fewer males courted non-receptive females, their courtship displays were significantly longer than those directed towards receptive females. Males also adjusted the contents of their displays according to female reproductive condition. Certain behavior patterns were unique to courtship displays directed towards each class of females. Males exhibited the behavior pattern `head roll' only when paired with receptive females, and `chin rub' was displayed only during courtship of non-receptive females. We hypothesize that these differences in male courtship frequency, intensity and content reflect differences in female reproductive value. Although males may benefit from mating with both receptive and non-receptive females, the costs associated with courtship may depend on female responsiveness. Thus, males adjust their courtship tactics accordingly. [source] Corrosion fatigue behaviour of a 15Cr-6Ni precipitation-hardening stainless steel in different tempersFATIGUE & FRACTURE OF ENGINEERING MATERIALS AND STRUCTURES, Issue 6 2000C.-K. Lin Systematic fatigue experiments, including both high-cycle axial fatigue (S,N curves) and fatigue crack growth (FCG, da/dN,,K curves), were performed on a precipitation-hardening martensitic stainless steel in laboratory air and 3.5 wt% NaCl solution. Specimens were prepared in three tempers, i.e. solution-annealed (SA), peak-aged (H900) and overaged (H1150) conditions, to characterize the effects of ageing treatment on the corrosion fatigue (CF) resistance. S,N results indicated that fatigue resistance in all three tempers was dramatically reduced by the aqueous sodium chloride environment. In addition, the smooth-surface specimens in H900 temper exhibited longer CF lives than the H1150 ones, while those in SA condition stood in between. However, for precracked specimens, the H1150 temper provided superior corrosive FCG resistance than the other two tempers. Comparison of the S,N and FCG curves indicated that early growth of crack-like defects and short cracks played the major role in determining the CF life for smooth surface. The differences in the CF strengths for the S,N specimens of the given three tempers were primarily due to their inherent differences in resistance to small crack growth, as they were in the air environment. Mielnick Abstract Increasing atmospheric CO2 concentrations may have a profound effect on the structure and function of plant communities. A previously grazed, central Texas grassland was exposed to a 200-µmol mol,1 to 550 µmol mol,1 CO2 gradient from March to mid-December in 1998 and 1999 using two, 60-m long, polyethylene- covered chambers built directly onto the site. One chamber was operated at subambient CO2 concentrations (200,360 µmol mol,1 daytime) and the other was regulated at superambient concentrations (360,550 µmol mol,1). Continuous CO2 gradients were maintained in each chamber by photosynthesis during the day and respiration at night. Net ecosystem CO2 flux and end-of-year biomass were measured in each of 10, 5-m long sections in each chamber. Net CO2 fluxes were maximal in late May (c. day 150) in 1998 and in late August in 1999 (c. day 240). In both years, fluxes were near zero and similar in both chambers at the beginning and end of the growing season. Average daily CO2 flux in 1998 was 13 g CO2 m,2 day,1 in the subambient chamber and 20 g CO2 m,2 day,1 in the superambient chamber; comparable averages were 15 and 26 g CO2 m,2 day,1 in 1999. Flux was positively and linearly correlated with end-of-year above-ground biomass but flux was not linearly correlated with CO2 concentration; a finding likely to be explained by inherent differences in vegetation. Because C3 plants were the dominant functional group, we adjusted average daily flux in each section by dividing the flux by the average percentage C3 cover. Adjusted fluxes were better correlated with CO2 concentration, although scatter remained. Our results indicate that after accounting for vegetation differences, CO2 flux increased linearly with CO2 concentration. Ananthapadmanabhan Synopsis Skin-cleansing compositions based on alkyl carboxylates (soaps) have a higher irritation potential than those based on syndet surfactants such as alkyl isethionates or alkyl ether sulphates. Contributing factors include inherent differences in the irritation potential of soaps and syndet surfactants, pH-induced changes in surfactant solution chemistry, and the direct effects of pH on the physical properties of the stratum corneum (SC). Past work has not directly addressed the effect of solution pH on the SC itself and its potential role in cleanser-induced skin irritation. In the current work, alterations to SC properties induced by buffered pH solutions and two strongly ionizable surfactants, sodium dodecyl sulphate and sodium lauryl ether sulphate, at different pH values are measured. By utilizing optical coherence tomography (OCT) and infrared (IR) spectroscopy we have directly measured physical changes in SC proteins and lipids. Our results indicate that SC swelling, which reflects alterations to SC structural proteins, is increased significantly at pH 10, compared to pH 4 and 6.5. The transition temperature (Tm) of SC lipids is found to increase at pH 10, compared to pH 4 and 6.5, suggesting a more rigid SC lipid matrix. Surfactants cause a further increase in swelling and lipid rigidity. Some aspects of what these results mean for SC physical properties as well as their implications to potential mechanisms of surfactant-induced skin irritation are discussed. Résumé Les compositions nettoyantes pour la peau à base d'alkyl carboxylates (savons) ont un potentiel irritant supérieur à celles à base de syndet tensioactifs tels que les alkyl isothionates ou les alkyl ether sulfates. Les facteurs en cause comprennent les différences de potentiel irritant inhérentes aux savons et aux syndet tensioactifs, les modifications de la chimie de la solution de tensioactif dues aux pH, et les effets directs du pH sur les propriétés physiques de la couche cornée (CC). Les travaux antérieurs n'ont pas traité directement l'effet du pH de la solution sur la couche cornée elle-même et son rôle potentiel dans l'irritation de la peau due à la solution nettoyante. Dans la présente étude on a mesuré les altérations des propriétés de la CC causées par des solutions à pH tamponné et deux tensioactifs fortement ionisables, le dodecyl sulfate de sodium et le lauryl ether sulfate de sodium, à différentes valeurs de pH. En utilisant la tomographie optique (OCT) et la spectroscopie à infrarouge (IR) on a mesuré directement les modifications physiques des protéines et des lipides de la CC. Nos résultats montrent que le gonflement de la CC, qui traduit des altérations des protéines structurales de la CC, augmente significativement à pH 10, par comparaison au pH 4 et 6.5. On observe que la température de transition (Tm) des lipides de la CC augmente à pH 10, par comparaison au pH 4 et 6.5, suggérant une matrice lipidique de la CC plus rigide. Les tensioactifs provoquent une augmentation plus importante du gonflement et de la rigidité lipidique. On aborde certains aspects de la signification de ces résultats vis-à-vis des propriétés physiques de la couche cornée ainsi que leurs conséquences sur les mécanismes potentiels de l'irritation de la peau causée par les tensioactifs. It compares the incidence and scope of collaborations in the voluntary sector with those of the commercial sector; while being mindful of some important inherent differences between the two milieus. The findings are illustrated with examples of charity collaborations, both past and present. The paper concludes with some possible ways forward for both practitioners and policy makers. Patil Aims: In recent years, interest has been growing in the search for novel bioemulsifiers. Many bacterial genera including Acinetobacter have been reported to produce bioemulsifiers. The present study aims to screen Acinetobacter isolates from healthy human skin for bioemulsifier production. Methods and Results: Acinetobacter junii SC14 produced maximum bioemulsifier in the presence of almond oil during stationary growth phase at 37°C and pH 7·2. Partially purified, nondialysable bioemulsifier from SC14 was a proteoglycan. The protein and polysaccharide fractions resulted in 95·2% reconstitution of the emulsification activity. The role of esterase in the release of cell-bound emulsifier and the contribution of capsular polysaccharide to the emulsification activity were observed. Conclusion:Acinetobacter strains from human skin exhibited better emulsification activity than that by burn wound or soil isolates, owing to the inherent differences in chemical microenvironment of their habitats. Significance and Impact of the Study: Investigation of skin commensals, especially acinetobacters, would lead to the discovery of novel bioemulsifiers with interesting properties. Attempts of screening and strain improvement directed towards skin commensals will open up new avenues for strains producing bioemulsifier on a commercial scale. This article documents the growing importance of the services sector and of services purchasing. Next, it develops a supply chain framework appropriate for a services supply chain by comparing and contrasting the applicability of three product-based manufacturing models: Global Supply Chain Forum Framework, SCOR and Hewlett-Packard's Supply Chain Management Model. Finally, this research describes the challenges for procurement professionals managing purchases for a services supply chain and provides suggestions for use of supply chain management theory, and practices for improvement. However, glycoproteins require particular attention as inherent differences in the N-glycosylation pathways of plants and mammals result in the production of glycoproteins bearing core-xylose and core-,(1,3)-fucose glyco-epitopes. For treatments requiring large quantities of repeatedly administered glycoproteins, the immunological properties of these non-mammalian glycans are a concern. Recombinant glycoproteins could be retained within the endoplasmic reticulum (ER) to prevent such glycan modifications occurring in the late Golgi compartment. Therefore, we analysed cPIPP, a mouse/human chimeric IgG1 antibody binding to the ,-subunit of human chorionic gonadotropin (hCG), fused to a C-terminal KDEL sequence, to investigate the efficiency of ER retrieval and the consequences in terms of N-glycosylation. The KDEL-tagged cPIPP antibody was expressed in transgenic tobacco plants or Agrobacterium -infiltrated tobacco and winter cherry leaves. N-Glycan analysis showed that the resulting plantibodies contained only high-mannose (Man)-type Man-6 to Man-9 oligosaccharides. In contrast, the cPIPP antibody lacking the KDEL sequence was found to carry complex N-glycans containing core-xylose and core-,(1,3)-fucose, thereby demonstrating the secretion competence of the antibody. Furthermore, fusion of KDEL to the diabody derivative of PIPP, which contains an N-glycosylation site within the heavy chain variable domain, also resulted in a molecule lacking complex glycans. The complete absence of xylose and fucose residues clearly shows that the KDEL-mediated ER retrieval of cPIPP or its diabody derivative is efficient in preventing the formation of non-mammalian complex oligosaccharides. [source] Volumetric and lateralized differences in selected brain regions of chimpanzees (Pan troglodytes) and bonobos (Pan paniscus)AMERICAN JOURNAL OF PRIMATOLOGY, Issue 12 2009William D. Hopkins Abstract The two species of Pan, bonobos and common chimpanzees, have been reported to have different social organization, cognitive and linguistic abilities and motor skill, despite their close biological relationship. Here, we examined whether bonobos and chimpanzee differ in selected brain regions that may map to these different social and cognitive abilities. Eight chimpanzees and eight bonobos matched on age, sex and rearing experiences were magnetic resonance images scanned and volumetric measures were obtained for the whole brain, cerebellum, striatum, motor-hand area, hippocampus, inferior frontal gyrus and planum temporale. Chimpanzees had significantly larger cerebellum and borderline significantly larger hippocampus and putamen, after adjusting for brain size, compared with bonobos. Bonobos showed greater leftward asymmetries in the striatum and motor-hand area compared with chimpanzees. No significant differences in either the volume or lateralization for the so-called language homologs were found between species. The results suggest that the two species of Pan are quite similar neurologically, though some volumetric and lateralized differences may reflect inherent differences in social organization, cognition and motor skills. Storer Abstract Rubrospinal motoneurons (RSMN) represent a population of androgen receptor-expressing central motoneurons with limited regenerative potential relative to their peripheral counterparts. A key determinant of regenerative capability lies in the nucleolar reaction of injured neurons. To date, characterization of the nucleolar reaction in injured central motoneurons has not been accomplished. Furthermore, it has been documented that testosterone propionate (TP) augments peripheral motoneuron regeneration through regulation of the nucleolar reaction to injury. In this study, the effects of injury alone, or in conjunction with TP, on the nucleolar response of injured RSMN were examined using in situ hybridization (ISH) techniques. Castrated adult male hamsters were subjected to right spinal cord hemisection at the C7/T1 vertebral level. Half the animals were subcutaneously implanted with one Silastic TP capsule, with the other half sham implanted. ISH for precursor 45S and mature 28S rRNA was accomplished with a 3H-labeled ribosomal DNA probe specific to the external transcribed spacer region or to the 28S region of the ribosomal gene, respectively. Postoperative times of 2, 6, and 24 hours were selected for examination of precursor 45S rRNA (i.e., rRNA transcriptional activation) levels and 0.25, 2, 4, and 14 days for examination of mature rRNA (i.e., ribosome) levels. Transcriptional activation of the rRNA gene was rapidly and transiently increased in injured RSMN, analogously to previously documented effects of injury on rRNA transcription in peripheral motoneurons, but, in contrast, this did not translate into an increase in mature ribosomes. TP administration failed to affect positively the nucleolar response of injured RSMN at all. From this study, a key component underlying inherent differences in the regenerative capacity of peripheral vs. central motoneurons has been identified, which can be targeted in future experiments designed to enhance the regenerative potential of selective neuronal populations. [source] Unique Early Gene Expression Patterns in Human Adult-to-Adult Living Donor Liver Grafts Compared to Deceased Donor GraftsAMERICAN JOURNAL OF TRANSPLANTATION, Issue 4 2009J. De Jonge Because of inherent differences between deceased donor (DD) and living donor (LD) liver grafts, we hypothesize that the molecular signatures will be unique, correlating with specific biologic pathways and clinical patterns. Microarray profiles of 63 biopsies in 13 DD and 8 LD liver grafts done at serial time points (procurement, backbench and postreperfusion) were compared between groups using class comparisons, network and biological function analyses. Specific genes were validated by quantitative PCR and immunopathology. Clinical findings were also compared. Following reperfusion, 579 genes in DD grafts and 1324 genes in LDs were differentially expressed (p < 0.005). Many upregulated LD genes were related to regeneration, biosynthesis and cell cycle, and a large number of downregulated genes were linked to hepatic metabolism and energy pathways correlating with posttransplant clinical laboratory findings. There was significant upregulation of inflammatory/immune genes in both DD and LD, each with a distinct pattern. Gene expression patterns of select genes associated with inflammation and regeneration in LD and DD grafts correlated with protein expression. Unique patterns of early gene expression are seen in LD and DD liver grafts, correlating with protein expression and clinical results, demonstrating distinct inflammatory profiles and significant downregulation of metabolic pathways in LD grafts. Two independent states of a cis -MoO2F4 octahedron are characteristic of static and dynamic disorder in structure (I). The dynamically disordered Mo atom is displaced from the symmetry axis producing four possible orientations of an anion that allow O and F atoms to be identified in separate orientations owing to the inherent differences between the Mo,O and Mo,F bonding. After the phase transition at lower temperature, (I) transforms into the statically disordered structure (II) with three possible orientations of the cis -MoO2F4 octahedron. In this case, it also seemed possible to distinguish between O and F atoms on a local scale. H atoms of two independent NH4 groups in (II) which form bifurcated N,H...F(O) hydrogen bonds were localized. [source] Investigating the Feasibility of Stem Cell Enrichment Mediated by Immobilized SelectinsBIOTECHNOLOGY PROGRESS, Issue 6 2007Nichola Charles Hematopoietic stem cell therapy is used to treat both malignant and non-malignant diseases, and enrichment of the hematopoietic stem and progenitor cells (HSPCs) has the potential to reduce the likelihood of graft vs host disease or relapse, potentially fatal complications associated with the therapy. Current commercial HSPC isolation technologies rely solely on the CD34 surface marker, and while they have proven to be invaluable, they can be time-consuming with variable recoveries reported. We propose that selectin-mediated enrichment could prove to be a quick and effective method for recovering HSPCs from adult bone marrow (ABM) on the basis of differences in rolling velocities and independently of CD34 expression. Purified CD34+ ABM cells and the unselected CD34, ABM cells were perfused over immobilized P-, E-, and L-selectin-IgG at physiologic wall shear stresses, and rolling velocities and cell retention data were collected. CD34+ ABM cells generally exhibited lower rolling velocities and higher retention than the unselected CD34, ABM cells on all three selectins. For initial CD34+ ABM cell concentrations ranging from 1% to 5%, we predict an increase in purity ranging from 5.2% to 36.1%, depending on the selectin used. Additionally, selectin-mediated cell enrichment is not limited to subsets of cells with inherent differences in rolling velocities. CD34+ KG1a cells and CD34, HL60 cells exhibited nearly identical rolling velocities on immobilized P-selectin-IgG over the entire range of shear stresses studied. However, when anti-CD34 antibody was co-immobilized with the P-selectin-IgG, the rolling velocity of the CD34+ KG1a cells was significantly reduced, making selectin-mediated cell enrichment a feasible option. Optimal cell enrichment in immobilized selectin surfaces can be achieved within 10 min, much faster than most current commercially available systems. [source] Non-invasive assessment of endothelial function , relation between vasodilatory responses in skin microcirculation and brachial arteryCLINICAL PHYSIOLOGY AND FUNCTIONAL IMAGING, Issue 6 2004Jimmy Hansell Summary Objective:, To compare different non-invasive methods for determination of human endothelial function in peripheral circulation. Design:, Observational, cross-sectional study in 39 healthy subjects (21 females, age 17,56 years). Setting:, Vascular research laboratory at university hospital. Methods:, Laser Doppler (LD) flowmetry was used to compare skin microvascular perfusion changes during postocclusive reactive hyperaemia with those induced by iontophoretic administration of acetylcholine (ACh), an endothelial-dependent vasodilator. LD measurements were compared with ultrasonographic measurements of postocclusive flow-mediated dilatation (FMD) in the brachial artery (n = 21). Results:, Local ACh induced a larger and more sustained skin perfusion increase than reactive hyperaemia after 4 min of regional arterial occlusion (P<0·001). A significant correlation was found between the magnitude of ACh-induced vasodilatation and peak reactive hyperaemia, both in absolute (r = 0·62, P<0·001) and relative terms (r = 0·58, P<0·001). A correlation was also found between brachial artery FMD and the magnitude of ACh-induced skin perfusion increase (r = 0·43, P<0·05) but not between FMD and reactive hyperaemia. Conclusion:, Endothelial function, an early marker of cardiovascular risk, can be non-invasively assessed and graded by LD and FMD-measurements and despite inherent differences, both methods do correlate.
