    Multiparametric analysis of normal and postchemotherapy bone marrow: Implication for the detection of leukemia-associated immunophenotypes,

    Abstract Background: The knowledge of normal marrow is mandatory to assess the malignant counterpart of normal cells and define leukemia-associated immunophenotypes (LAIPs). In this study, the expression of a variety of antigens expressed in normal and postchemotherapy bone marrow (BM) was analyzed to provide a frame of reference for the identification of myeloid LAIPs. Methods: Multiparameter four- and six-color flow cytometry was used to define antigen combinations totally absent or present at very minimal levels in marrow cells of normal individuals (n = 20) and patients receiving chemotherapy for acute lymphoblastic leukemia (n = 20). Immature (blast) cells were gated according to CD45/SSC properties. Fifty-three acute myeloid leukemia (AML) samples were studied in six-color combinations. Results: In six-color flow cytometry, 47 phenotypes were totally absent from blast gate in all normal samples. Forty-one other phenotypes were identified in less than 0.05% of blast cells. There was no difference between normal and postchemotherapy BMs. The four-color panel allowed to identify only 30 phenotypes present at a frequency <0.05%. Using the six-color panel, 58% of the absent or infrequent phenotypes in normal BM were found in at least one of 53 AML samples. All AML cases exhibited at least one LAIP. Conclusion: Our results show that the ability to distinguish leukemic from healthy cells is considerably increased by a six-color approach. Furthermore, these absent or infrequent phenotypes in normal BM are identified in AML and can be utilized for minimal residual disease study. © 2007 Clinical Cytometry Society [source]


    Additive Inhibition of Dendritic Cell Allostimulatory Capacity by Alcohol and Hepatitis C Is Not Restored by DC Maturation and Involves Abnormal IL-10 and IL-2 Induction

    Background: Excessive alcohol use results in impaired immunity, and it is associated with increased incidence and progression of chronic hepatitis C virus (HCV) infection. Here we investigated the effects of HCV infection and alcohol on myeloid dendritic cells (DC) that are critical in antiviral immunity. Methods: Immature and mature DCs were generated from monocytes of chronic HCV infected patients (HCV-DC) and controls (N-DC) with IL-4 plus granulocyte-macrophage colony stimulating factor (GM-CSF) in the presence or absence of alcohol (25 mM). DC allostimulatory capacity was tested in mixed lymphocyte reaction (MLR) and cytokine production by ELISA. Results: Allostimulatory capacity of HCV-DCs was reduced compared to N-DCs and it was further inhibited by alcohol treatment (p < 0.01). MLR was also decreased with alcohol-treated N-DCs. DC phenotypic markers and apoptosis were comparable between HCV-DCs and N-DCs irrespective of alcohol treatment. However, HCV-DCs and alcohol-treated N-DCs exhibited elevated IL-10 and reduced IL-12 production. Reduced MLR with HCV-DCs and its further inhibition by alcohol coexisted with decreasing IL-2 levels (p < 0.017). DC maturation partially improved but failed to fully restore the reduced allostimulatory function of either alcohol-treated or alcohol-naïve HCV-DCs (p < 0.018). Conclusions: Alcohol and HCV independently and together inhibit DC allostimulatory capacity, increase IL-10, reduce IL-12 and IL-2 production that cannot be normalized by DC maturation. HCV and alcohol interact to modulate innate and adaptive immune responses via dendritic cells. [source]


    Change over 70 years in a southern California chaparral community related to fire history

    Abstract: Question: What changes in species composition and cover have occurred in chaparral as a function of fire history across an ecoregion? Location: San Diego County, California, USA. Methods: Stands in which 40 mid-elevation chaparral vegetation plots (each 400 m2 in area) were located in the 1930s were resurveyed in 2001. We stratified the stands into Infrequently versus Frequently burned (0,1 versus 2 or more fires recorded in the 91-yr period), and Immature versus Mature (31 yr versus >31 yr since last fire), resulting in four groups. Ten stands were randomly selected from each of these groups for survey. Results: There were no major shifts in life form composition, e.g., live oak trees were not invading chaparral that had experienced little or no fire, nor were subshrubs or herbaceous species replacing shrubs in areas that had experienced more frequent fires. However, there was a notable increase in the frequency of the subshrub Eriogonum fasciculatum across all fire history groups. In the mature stands with infrequent fire, average cover of resprouting shrubs increased (from 72 to 91%) and cover of obligate seeding shrubs (species with fire-cued germination) decreased (from 21 to 6%) significantly. Mature stands with frequent fire showed a significant decrease in resprouter cover (from 87 to 80%) and increase in obligate seeders (from 10 to 16%). Conclusions: While the tremendous changes in land use in southern California have been predicted to cause shifts in chaparral composition, these shifts are difficult to detect because species longevity and fire cycles are on the order of decades to a century. In this study, the expected trends could only be detected in groups that were mature at the time of the second survey. [source]


    Immunoexpression of Aromatase in Immature and Adult Males of the European Bison (Bison bonasus, Linnaeus 1758)

    Contents Based on recent literature dealing with the role of oestrogens in the male gonad, attempts were undertaken to reveal the site of aromatization within the testis of the European bison (Bison bonasus). Testes were collected from culled animals living in free-ranging populations in Bialowieza Forest, Poland (nine males aged 8 months to 10 years). Moreover, to check for any alterations in the expression of testicular aromatase between American bison (Bison bison) and European bison, testes from one adult 10-year-old individual were also chosen for this study. For immunohistochemistry, 4% formaldehyde fixative was used. Both qualitative and quantitative evaluations of immunohistochemical staining were performed. Leydig cells, Sertoli cells and germ cells exhibited a positive immunoreaction for aromatase in testes of immature and sexually mature bison. A marked increase in aromatase expression was observed in three adult European individuals with impaired spermatogenesis. Consistent with recent data and those of our own, it might be suggested that the strong expression of aromatase negatively affects spermatogenic function in bison testes and may serve as a possible explanation of specific sperm defects observed in European bison bulls. On the contrary, one cannot exclude that differences in the aromatase immunoexpression levels are attributed to the homozygosity, the cause of frequent disease in European bison. [source]

    Factors Associated with the Development of Board Sub,committees

    This study examines the factors associated with the presence of board sub,committees, specifically audit, remuneration and nomination committees. Factors which are hypothesised in this study to affect sub,committee presence are Big 6 auditors, non,executive directors, non,executive chairmen, number of intercorporate relationships of the board and shareholder type. Company size, number of board members and leverage are employed as control variables as suggested by earlier research. An analysis of board sub,committees in the Australian corporate environment is relevant to other jurisdictions as there are no mandatory requirements on either board composition or board sub,committees. There is, however, a mandatory requirement to disclose corporate governance practices which allows for a study of this type to be reliably conducted. A sample of 361 Australian companies drawn from the largest 500 public companies is employed. Audit committee presence is found to be positively associated with Big 6 auditors and the number of intercorporate relationships of the directors of the board. Remuneration committees are also found to be associated with Big 6 auditors and intercorporate relationships and also higher levels of institutional investment. The presence of nomination committees is not associated with auditors, directors or investors, but is associated with board size and leverage. The study concludes that audit committees are a highly developed and mature governance mechanism, and that remuneration committees can be classed as a developing and maturing structure whilst nomination committees are relatively immature.     Nesprins, but not sun proteins, switch isoforms at the nuclear envelope during muscle development

    Abstract Nesprins are a family of nuclear transmembrane proteins anchored via Sun proteins to the nuclear membrane. Analysis of nesprins during human muscle development revealed an increase in nesprin-1-giant during early myogenesis in vitro. During the transition from immature to mature muscle fibres in vivo, nesprin-2 partly replaced nesprin-1 at the nuclear envelope and short nesprin isoforms became dominant. Sun1 and Sun2 proteins remained unchanged during this fibre maturation. In emerin-negative skin fibroblasts, nesprin-2-giant was relocated from the nuclear envelope to the cytoplasm, not to the endoplasmic reticulum, while nesprin-1 remained at the nuclear envelope. In emerin-negative keratinocytes lacking nesprin-1, nesprin-2 remained at the nuclear envelope. HeLa cell nuclear envelopes lacked nesprin-1, which was the dominant form in myoblasts, while a novel 130-kD nesprin-2 isoform dominated Ntera-2 cells.     Posttranslational regulation of BCL2 levels in cerebellar granule cells: A mechanism of neuronal survival

    Abstract Apoptosis can be modulated by K+ and Ca2+ inside the cell and/or in the extracellular milieu. In murine organotypic cultures, membrane potential-regulated Ca2+ signaling through calcineurin phosphatase has a pivotal role in development and maturation of cerebellar granule cells (CGCs). P8 cultures were used to analyze the levels of expression of B cell lymphoma 2 (BCL2) protein, and, after particle-mediated gene transfer in CGCs, to study the posttranslational modifications of BCL2 fused to a fluorescent tag in response to a perturbation of K+/Ca2+ homeostasis. There are no changes in Bcl2 mRNA after real time PCR, whereas the levels of the fusion protein (monitored by calculating the density of transfected CGCs under the fluorescence microscope) and of BCL2 (inWestern blotting) are increased. After using a series of agonists/antagonists for ion channels at the cell membrane or the endoplasmic reticulum (ER), and drugs affecting protein synthesis/degradation, accumulation of BCL2 was related to a reduction in posttranslational cleavage by macroautophagy. The ER functionally links the [K+]e and [Ca2+]i to the BCL2 content in CGCs along two different pathways. The first, triggered by elevated [K+]e under conditions of immaturity, is independent of extracellular Ca2+ and operates via IP3 channels. The second leads to influx of extracellular Ca2+ following activation of ryanodine channels in the presence of physiological [K+]e, when CGCs are maintained in mature status.     The sexually dimorphic expression of L7/SPA, an estrogen receptor coactivator, in zebra finch telencephalon

    Abstract Sex differences in the zebra finch (Taeniopygia guttata) brain are robust and include differences in morphology (song control nuclei in males are significantly larger) and behavior (only males sing courtship songs). In zebra finches, hormonal manipulations during development fail to reverse sex differences in song nuclei size and suggest that the classical model of sexual differentiation is incomplete for birds. Coactivators act to initiate transcriptional activity of steroid receptors, and may help explain why hormonal manipulations alone are not sufficient to demasculinize the male zebra finch brain. The present study investigated the expression and localization of L7/SPA (an estrogen receptor coactivator) mRNA and protein expression across the development of zebra finch song nuclei from males and females collected on P1 (song nuclei not yet formed), P10 (posthatch day 10, song nuclei formed), P30 (30 days posthatch, sexually immature but song nuclei formed and birds learning to sing), and adult birds (older than 65 days and sexually mature). Northern blot analysis showed a significant sex difference in P1 and adult L7/SPA mRNA expression while Western blot analysis also showed enhanced expression in the male brain at all age points. Both in situ hybridization and immunohistochemistry demonstrated that L7/SPA mRNA and protein were located in the song nuclei as well as expressed globally. Elevated coactivator expression may be a possible mechanism controlling the development of male song control nuclei, and coactivators such as L7/SPA may be important regulators of the masculinizing effects of estradiol on brain sexual differentiation. © 2007 Wiley Periodicals, Inc.     Ultrastructural and histochemical study of the salivary glands of Aplysia depilans (Mollusca, Opisthobranchia)

    Abstract The digestive system of the sea hare, Aplysia depilans, includes a pair of ribbon-shaped salivary glands. A central duct and a large blood vessel run close to each other along the length of these glands and both are surrounded by a layer of muscle cells. Three cell types form the glandular epithelium: granular cells, vacuolated cells and mucocytes. The granular cells possess cilia and spherical secretion granules, located primarily in the apical region. The granules of immature cells have a low electron density and are mainly formed by neutral polysaccharides with small amounts of proteins. The granules of mature cells are larger, have a high electron density and are mainly formed by proteins with lower amounts of neutral polysaccharides. Transition stages between immature and mature granular cells are observed. The vacuolated cells are large and frequently pyramidal in shape, but after the application of histochemical techniques almost all vacuoles remain uncoloured. The numerous vacuoles contain flocculent material in a clear background and the mitochondria possess large crystalline structures in the matrix. A pyramidal shape is also typical of the mucocytes, which are filled with vesicles containing granular masses surrounded by a network of secretion material. These large cells are strongly stained by Alcian blue, revealing the presence of acidic mucopolysaccharides. This is the first ultrastructural study of the salivary glands in opisthobranch gastropods. [source]


    Coastal conglomerates around the Hadjer el Khamis inselbergs (western Chad, central Africa): new evidence for Lake Mega-Chad episodes

    Abstract This paper reports on a study dealing with the rhyolitic inselbergs of Hadjer el Khamis that formed palaeoislands during Lake Mega-Chad events. Field observations have shown that: (1) conglomeratic patches of immature to mature clasts are preserved at the feet of the Hadjer el Khamis inselbergs; (2) in cross-section, their pro,le reveals a well de,ned cliff,platform junction at a constant elevation (325 m). The monolithological clasts show all degrees of roundness, from angular cobbles to well rounded pebbles. This wide range of maturity suggests a coastal origin for these cobbles. The system was permanently fed with angular clasts, which were progressively worn by waves. Cobbles that were wave-worked for the longest time are the best rounded. The cliff,platform junction is the result of erosion by waves, which attacked and undercut the inselberg cliffs during Lake Mega-Chad events. Asymmetrical erosion pro,les moreover suggest a wind regime dominated by SW to NE oriented winds. These interpretations have two implications. The elevation of the cliff,platform junction is an indication of the highest water level of Lake Mega-Chad at 320,325 m, which is in agreement with other observations elsewhere in the basin. The SW to NE oriented winds show that monsoon-related winds were prevalent during Lake Mega-Chad events, suggesting the Inter-Tropical Convergence Zone was located higher in latitude than today.     First-line treatment with bortezomib rapidly stimulates both osteoblast activity and bone matrix deposition in patients with multiple myeloma, and stimulates osteoblast proliferation and differentiation in vitro

    Abstract Objectives:, The aim of the study was to investigate the effect of bortezomib on osteoblast proliferation and differentiation, as well as on bone matrix deposition for the first time in bisphosphonate-naïve, previously untreated patients with myeloma. Methods:, Twenty newly diagnosed patients received four cycles of bortezomib treatment, initially as monotherapy and then combined with a glucocorticoid from cycle two to four. Bone remodeling markers were monitored closely during treatment. Furthermore, the effects of bortezomib and a glucocorticoid on immature and mature osteoblasts were also studied in vitro. Results:, Treatment with bortezomib caused a significant increase in bone-specific alkaline phosphatase and pro-collagen type I N-terminal propeptide, a novel bone formation marker. The addition of a glucocorticoid resulted in a transient decrease in collagen deposition. In vitro bortezomib induced osteoblast proliferation and differentiation. Differentiation but not proliferation was inhibited by glucocorticoid treatment. Conclusions:, Bortezomib used as first-line treatment significantly increased collagen deposition in patients with multiple myeloma and osteolytic lesions, but the addition of a glucocorticoid to the treatment transiently inhibited the positive effect of bortezomib, suggesting that bortezomib may result in better healing of osteolytic lesions when used without glucocorticoids in patients that have obtained remission with a previous therapy. The potential bone-healing properties of single-agent bortezomib are currently being explored in a clinical study in patients who have undergone high-dose therapy and autologous stem cell transplantation.     Long-term maternal imprinting of the specific B cell repertoire by maternal antibodies

    Abstract Maternal antibodies protect newborns whilst they are immunologically immature. This study shows that maternal antibodies can also shape the B cell repertoire of the offspring long after the maternal antibodies themselves become undetectable. VHDJH gene-targeted (VI10) mice expressing a heavy chain specific for vesicular stomatitis virus (VSV) produce a 20-fold increased spontaneous titer of VSV-neutralizing antibodies. When transferred from mother to offspring, these antibodies prevented accumulation of Ag-specific transitional type,2 and marginal zone B cells with an activated phenotype and favored selection to the B cell follicles. This effect was B cell-intrinsic and lasted up to adulthood. The pups nursed by mothers producing specific antibodies developed higher endogenous antibody titers of this specificity which perpetuated the effects of specific B cell selection into the mature follicular compartment, presumably by blocking auto-Ag-dependent development of transitional type,2 B cells in the spleen. This repertoire change was functional, as following infection of adult mice with VSV, those pups that had received specific maternal antibodies as neonates had increased pre-immune titers and mounted strong early IgG neutralizing antibodies.     Immunisation with BCG and recombinant MVA85A induces long-lasting, polyfunctional Mycobacterium tuberculosis -specific CD4+ memory T lymphocyte populations

    Abstract In the search for effective vaccines against intracellular pathogens such as HIV, tuberculosis and malaria, recombinant viral vectors are increasingly being used to boost previously primed T cell responses. Published data have shown prime-boost vaccination with BCG-MVA85A (modified vaccinia virus Ankara expressing antigen 85A) to be highly immunogenic in humans as measured by ex vivo IFN-, ELISPOT. Here, we used polychromatic flow cytometry to investigate the phenotypic and functional profile of these vaccine-induced Mycobacterium tuberculosis (M.tb) antigen 85A-specific responses in greater detail. Promisingly, antigen 85A-specific CD4+ T cells were found to be highly polyfunctional, producing IFN-,, TNF-,, IL-2 and MIP-1,. Surface staining showed the responding CD4+ T cells to be relatively immature (CD45RO+ CD27intCD57,); this observation was supported by the robust proliferative responses observed following antigenic stimulation. Furthermore, these phenotypic and functional properties were independent of clonotypic composition and epitope specificity, which was maintained through the different phases of the vaccine-induced immune response. Overall, these data strongly support the use of MVA85A in humans as a boosting agent to expand polyfunctional M.tb -specific CD4+ T cells capable of significant secondary responses. [source]


    Deletion of the LIME adaptor protein minimally affects T and B cell development and function

    Abstract LIME (Lck-interacting membrane protein) is a transmembrane adaptor that associates with the Lck and Fyn protein tyrosine kinases and with the C-terminal Src kinase (Csk). To delineate the role of LIME in vivo, LIME-deficient mice were generated. Although Lime transcripts were expressed in immature and mature B and T cells, the absence of LIME impeded neither the development nor the function of B and T cells. TCR transgenic mice deprived of LIME showed, however, a 1.8-fold enhancement in positive selection. Since B cells and activated T cells express LIME and the related adaptor NTAL, mice lacking both adaptors were generated. Double-deficient mice showed no defect in the development and function of B and T cells, and the lack of LIME had no effect on the autoimmune syndrome that develops in aged NTAL-deficient mice. In contrast to a previous report, we further showed that this autoimmune syndrome develops in the absence of T cells. Therefore, our in vivo results refute all the previous roles postulated for LIME on the basis of studies of transformed B and T cells and demonstrate that LIME has no seminal role in the signaling cassette operated by antigen receptors and coreceptors.     Interaction between the CCR5 chemokine receptors and microbial HSP70

    Abstract Evidence is presented that the microbial 70-kD heat shock protein (HSP70) binds to CCR5 chemokine receptors in CCR5-transfected cell lines and in primary human cells. Significant CCR5-mediated calcium mobilization was stimulated by HSP70 and inhibited with TAK,779, which is a specific CCR5 antagonist. HSP70-mediated activation of the p38 MAPK phosphorylation signaling pathway was also demonstrated in CCR5-transfected HEK 293 cells. Direct binding of three extracellular peptides of CCR5 to HSP70 was demonstrated by surface plasmon resonance. Functional evidence of an interaction between HSP70, CCR5 and CD40 was shown by enhanced production of CCL5 by HEK 293 cells transfected with both CD40 and CCR5. Primary monocyte-derived immature DC stimulated with HSP70 produced IL-12 p40, which showed dose-dependent inhibition of >90% on treatment with both TAK 779 and anti-CD40 mAb. Stimulation of IL-12 p40 or TNF-, by HSP70 was related to the differential cell surface expression of CCR5 in primary human immature and mature DC, and those with the homozygous ,,32 CCR5 mutation. These findings may be of significance in the interaction between HSP70 and immune responses of CCR5+ T cells in HIV-1 infection, as well as in inflammatory bowel disease.     Multifocal structure of the T cell , dendritic cell synapse

    Abstract The structure of immunological synapses formed between murine naive T cells and mature dendritic cells has been subjected to a quantitative analysis. Immunofluorescence images of synapses formed in the absence of antigen show a diffuse synaptic accumulation of CD3 and LFA-1. In electron microscopy, these antigen-free synapses present a number of tight appositions (cleft size ,15,nm), all along the synapse. These tight appositions cover a significantly larger surface fraction of antigen-dependent synapses. In immunofluorescence, antigen-dependent synapses show multiple patches of CD3 and LFA-1 with a variable overlap. A similar distribution is observed for PKC, and talin. A concentric organization characteristic of prototypical synapses is rarely observed, even when dendritic cells are paralyzed by cytoskeletal poisons. In T,DC synapses, the interaction surface is composed of several tens of submicronic contact spots, with no large-scale segregation of CD3 and LFA-1. As a comparison, in T,B synapses, a central cluster of CD3 is frequently observed by immunofluorescence, and electron microscopy reveals a central tight apposition. Our data show that it is inappropriate to consider the concentric structure as a "mature synapse" and multifocal structures as immature. [source]


    Impaired lymphocyte development and function in Clast5/Stra13/DEC1-transgenic mice

    Abstract Clast5/Stra13/DEC1 is a member of the helix-loop-helix family of transcriptional repressors. We have previously shown that Clast5 is rapidly down-regulated upon B,cell activation and its overexpression inhibits cell cycle progression in B,lymphoma cells. In the present study, we show that Clast5 expression is developmentally regulated during B,cell differentiation, being expressed at theprogenitor B,cells, down-regulated at the precursor B,cells, elevated in immature and mature resting B,lymphocytes, and down-regulated again in germinal center B,ells. To investigate the function of Clast5 in regulating lymphocyte development, we have generated transgenic mice expressing Clast5 in B- and T-lineage cells (Clast5-Tg). Clast5-Tg mice grew and bred normally but their spleen and thymus cellularity was reduced compared with control littermates. The development of B,cells in the bone marrow and T,cells in the thymus was impaired, with the expansion of progenitor B and T,cells most strongly affected. The frequency of IL-7-responsive cells in the bone marrow of Clast5-Tg mice was reduced by >80% and their proliferative response to IL-7 was also compromised. Mature B,cells from Clast5-Tg mice were hyporesponsive to antigen receptor cross-linking and exhibited mild reduction in the proliferative response to CD40 ligation or lipopolysaccharide stimulation. Moreover, thedevelopment of germinal center B,cells and antibody production against a T-dependent antigen were reduced in Clast5-Tg mice. These results reveal a critical role for Clast5/Stra13/DEC1 in negatively regulating lymphocyte development and function in vivo. [source]


    Trypanosoma cruzi infection modulates intrathymic contents of extracellular matrix ligands and receptors and alters thymocyte migration

    Abstract Several T cell abnormalities have been described in the course of acute Trypanosoma cruzi infection in mice, including severe effects on the thymus. In the present study, looking at the expression of extracellular matrix ligands in the thymus, we observed that deposits of fibronectin and laminin increased progressively during the course of infection, reaching a maximum at the peak of parasitemia and thymic atrophy. Concomitantly, membrane expression of fibronectin and laminin receptors (VLA-4, VLA-5 and VLA-6) was also enhanced on thymocyte subsets of infected mice. These results correlated with changes in intrathymic thymocyte migration ability during the acute phase of infection, when a higher fibronectin-dependent transmigratory activity of CD4+CD8+ thymocytes was observed. Strikingly, we detected higher frequency of immature and high VLA-expressing CD4+CD8+ T cells in the peripheral lymphoid organs of infected mice at thepeak of parasitemia. These cells seemed to be thymus dependent, since significantly lower amounts of them were found in thymectomized mice, and some of them carry "prohibited" V, segments of the TCR. Our data suggest an imbalance in the intrathymic cell trafficking following acute T. cruzi infection, likely due to dysregulated extracellular matrix-dependent interactions. [source]


    Adult neurogenesis in the central olfactory pathway in the absence of receptor neuron turnover in Libinia emarginata

    Abstract Life-long neurogenesis is a characteristic feature of the olfactory pathways of a phylogenetically diverse array of animals. In both vertebrates and invertebrates, the life-long addition of olfactory interneurons in the brain occurs in parallel with the continuous proliferation of olfactory receptor neurons in the olfactory organ. It has been proposed that these two processes are related functionally, with new olfactory interneurons being added to accommodate the new olfactory receptor neurons added in the periphery. While this has not been tested directly because the two processes are not readily separable, this question can be addressed in the olfactory pathway of the crab, Libinia emarginata. Unlike most decapod crustaceans, which moult and grow throughout life, L. emarginata has a terminal, maturational moult after which animals become anecdysic (stop moulting). Because the addition of new receptor neurons in crustaceans is associated with moulting, a comparison of neurogenesis in immature and mature L. emarginata provides an opportunity to examine the interdependence of central and peripheral neurogenesis in the olfactory pathway. This study demonstrates that the continuous addition of olfactory receptor neurons in L. emarginata ceases at the terminal moult but that proliferation and differentiation of olfactory interneurons in the brain continues in mature animals. Contrary to the general assumption, therefore, continuous neurogenesis in the central olfactory pathway of this species does not occur as part of a process involving the coregulation of central and peripheral neurogenesis. These findings suggest that peripheral neurogenesis is not a requirement for continuous neurogenesis in the central olfactory pathway. [source]


    GABA and glycine are protective to mature but toxic to immature rat cortical neurons under hypoxia

    Abstract Although recent studies suggest that ,-aminobutyric acid (GABA) and glycine may be ,inhibitory' to mature neurons, but ,excitatory' to immature neurons under normoxia, it is unknown whether inhibitory neurotransmitters are differentially involved in neuronal response to hypoxia in immature and mature neurons. In the present study, we exposed rat cortical neurons to hypoxia (1% O2) and examined the effects of three major inhibitory neurotransmitters (GABA, glycine and taurine) on the hypoxic neurons at different neuronal ages [days in vitro (DIV)4,20]. Our data showed that the cortical neurons expressed both GABAA and glycine receptors with differential developmental profiles. GABA (10,2000 µm) was neuroprotective to hypoxic neurons of DIV20, but enhanced hypoxic injury in neurons of <,DIV20. Glycine at low concentrations (10,100 µm) exhibited a similar pattern to GABA. However, higher concentrations of glycine (1000,2000 µm) for long-term exposure (48,72 h) displayed neuroprotection at all ages (DIV4,20). Taurine (10,2000 µm), unlike GABA and glycine, displayed protection only in DIV4 neurons, and was slightly toxic to neurons >,DIV4. In comparison with delta-opioid receptor (DOR)-induced protection in DIV20 neurons exposed to 72 h of hypoxia, glycine-induced protection was weaker than that of DOR but stronger than that of GABA and taurine. These data suggest that the effects of the inhibitory neurotransmitters on hypoxic cortical neurons are age-dependent, with GABA and glycine being neurotoxic to immature neurons and neuroprotective to mature neurons.     Differential maturation of motoneurons innervating ankle flexor and extensor muscles in the neonatal rat

    Abstract The first postnatal week is a critical period for the development of posture in the rat. The use of ankle extensor muscles in postural reactions increases during this period. Changes in excitability of motoneurons are probably an important factor underlying this maturation. The aim of this study was to identify whether variations in the maturation exist between motor pools innervating antagonistic muscles. Intracellular recordings in the in vitro brain stem,spinal cord preparation of neonatal rats (from postnatal day 0,5) were used to examine the developmental changes in excitability of motoneurons innervating the ankle flexors (F-MNs) and the antigravity ankle extensors (E-MNs). No significant difference in resting potential, action potential threshold, input resistance or rheobase was observed at birth. The age-related increase in rheobase was more pronounced for F-MNs than for E-MNs. The development of discharge properties of E-MNs lagged behind that of F-MNs. More F-MNs than E-MNs were able to fire repetitively in response to current injection at birth. F-MNs discharged at a higher frequency than E-MNs at all ages. Differences in the duration of action potential afterhyperpolarization accounted, at least partly, for the differences in discharge frequency between E-MNs and F-MNs at birth, and for the age-related increase in firing rate. These results suggest that E-MNs are more immature at birth than F-MNs and that there is a differential development of motoneurons innervating antagonistic muscles. This may be a critical factor in the development of posture and locomotion. [source]


    ORIGINAL ARTICLE: Propensity of marine reserves to reduce the evolutionary effects of fishing in a migratory species

    Abstract Evolutionary effects of fishing can have unwanted consequences diminishing a fishery's value and sustainability. Reserves, or no-take areas, have been proposed as a management tool for reducing fisheries-induced selection, but their effectiveness for migratory species has remained unexplored. Here we develop an eco-genetic model to predict the effects of marine reserves on fisheries-induced evolution under migration. To represent a stock that undergoes an annual migration between feeding and spawning grounds, we draw model parameters from Atlantic cod (Gadus morhua) in the northern part of its range. Our analysis leads to the following conclusions: (i) a reserve in a stock's feeding grounds, protecting immature and mature fish alike, reduces fisheries-induced evolution, even though protected and unprotected population components mix on the spawning grounds; (ii) in contrast, a reserve in a stock's spawning grounds, protecting only mature fish, has little mitigating effects on fisheries-induced evolution and can sometimes even exacerbate its magnitude; (iii) evolutionary changes that are already underway may be difficult to reverse with a reserve; (iv) directly after a reserve is created or enlarged, most reserve scenarios result in yield losses; and (v) timescale is very important: short-term yield losses immediately after a reserve's creation can give way to long-term gains.     SYNTHESIS: Evolutionary consequences of fishing and their implications for salmon

    Abstract We review the evidence for fisheries-induced evolution in anadromous salmonids. Salmon are exposed to a variety of fishing gears and intensities as immature or maturing individuals. We evaluate the evidence that fishing is causing evolutionary changes to traits including body size, migration timing and age of maturation, and we discuss the implications for fisheries and conservation. Few studies have fully evaluated the ingredients of fisheries-induced evolution: selection intensity, genetic variability, correlation among traits under selection, and response to selection. Most studies are limited in their ability to separate genetic responses from phenotypic plasticity, and environmental change complicates interpretation. However, strong evidence for selection intensity and for genetic variability in salmon fitness traits indicates that fishing can cause detectable evolution within ten or fewer generations. Evolutionary issues are therefore meaningful considerations in salmon fishery management. Evolutionary biologists have rarely been involved in the development of salmon fishing policy, yet evolutionary biology is relevant to the long-term success of fisheries. Future management might consider fishing policy to (i) allow experimental testing of evolutionary responses to exploitation and (ii) improve the long-term sustainability of the fishery by mitigating unfavorable evolutionary responses to fishing. We provide suggestions for how this might be done. [source]

Abstract:, To circumvent the costly isolation procedure associated with tissue mast cells (MC), two human MC lines, i.e. HMC-1 and LAD2, are frequently employed, but their relation to mature MC is unknown. Here, we quantitatively assessed their expression of MC markers in direct comparison to skin MC (sMC). sMC expressed all lineage markers at highest and HMC-1 cells at lowest levels. LAD2 cells expressed comparable high-affinity IgE receptor , (Fc,RI,) and Fc,RI, but less Fc,RI, than sMC and displayed slightly reduced, but robust Fc,RI-mediated histamine release. Only minor differences were found for total histamine content and c-Kit expression. Huge, and to this level unexpected, differences were found for MC tryptase and chymase, with sMC >>> LAD2 > HMC-1. Taken together, HMC-1 cells represent very immature malignantly transformed MC, whereas LAD2 cells can be considered intermediately differentiated. Because of the minute levels of MC proteases, MC lines can serve as surrogates of tissue MC to a limited degree only. [source]


    Modeling the influence of oceanic-climatic changes on the dynamics of Pacific saury in the northwestern Pacific using a life cycle model

    Abstract A life cycle model for Pacific saury (Cololabis saira) was developed to clarify the possible causes of interannual and decadal variability in its abundance. In the model, the population of saury is composed of two spawning cohorts: one spawned in the Kuroshio region during autumn,winter and the other spawned in the Kuroshio-Oyashio Transition Zone during winter,spring. The life cycle of saury was divided into six stages: namely egg, larval, juvenile, young, immature and adult stages. The life cycle model combines growth, survival, fishing and reproductive processes, in which the effects of sea surface temperature (SST) in the Kuroshio region and El Niño-Southern Oscillation (ENSO) events on the winter-spawning cohorts, the effects of SST in the Oyashio region on the spring-spawning cohorts, and the effects of fishing on the two spawning cohorts are taken into account. Results of basic modeling, in which environments are assumed stable and the stock is affected by fishing only, shows that the interannual fluctuations in the abundance are small and could hardly explain the observed large annual changes in abundance. On the contrary, results of modeling incorporating the effects of oceanic-climatic changes corresponded well with actual interannual-decadal variations in abundance. These results suggest the following environmental effects: (1) SST in the Kuroshio region affects decadal changes in abundance; (2) ENSO events influence the survival of the winter spawning cohort and result in large interannual variations in the abundance. It is concluded that large-scale climatic and oceanic changes strongly affect the abundance of saury. [source]


    Diving behavior of immature, feeding Pacific bluefin tuna (Thunnus thynnus orientalis) in relation to season and area: the East China Sea and the Kuroshio,Oyashio transition region

    Abstract Twenty-four archival tags were recovered from Pacific bluefin tuna previously released off Tsushima Island in the East China Sea. By analysis of the time-series data of the pressure and the ambient and internal temperature from the 24 tags, we examined the relationship between the tuna's pattern of diving and the thermocline depth. In the East China Sea, diving and feeding events occurred throughout almost the entire day in both winter and summer, suggesting that the purpose of diving is for feeding. In summer, the feeding frequency was greater than that in winter, which corresponds to the fact that growth is more rapid in summer than in winter. During summer in the Kuroshio,Oyashio transition region, on the other hand, feeding events were much more frequent than those in the East China Sea, in spite of a lower diving frequency. The mean horizontal distance traveled was also significantly higher and it seems that in this area they may move horizontally to feed on prey accumulated at the surface. We conclude that, in addition to the ambient temperature structure, the vertical and horizontal distribution of prey species plays an important role in the feeding behavior of Pacific bluefin tuna. One bluefin tuna migrated to the Oyashio frontal area, where both the horizontal and the vertical thermal gradients are much steeper. The fish spent most of the time on the warmer side of the front and often traveled horizontally to the colder side during the day, perhaps to feed. This implies that there is a thermal barrier effect, in this case from the Oyashio front, on their behavior. The frequency of feeding events was low, although all the monitored fish dived every dawn and dusk, irrespective of the seasons or location. It is possible that these twice-daily diving patterns occurred in response to the change in ambient light at sunrise and sunset. [source]


    An evaluation of the potential influence of SST and currents on the oceanic migration of juvenile and immature chum salmon (Oncorhynchus keta) by a simulation model

    Abstract Using a salmon migration model based on the assumption that swimming orientation is temperature dependent, we investigated the determining factors of the migration of juvenile and immature chum salmon (Oncorhynchus keta) in the North Pacific. We compared the predictions of the model with catch data of immature and juvenile chum salmon collected by Japanese research vessels from 1972 to 1999. The salmon migration model reproduced the observed distributions of immature chum salmon and indicates that passive transport by wind-driven and geostrophic currents plays an important role in the eastward migration of Asian salmon. These factors result in a non-symmetric distribution of Asian and North American chum salmon in the open ocean. The directional swimming component contributes to the northward migration in summer. The model results indicate that during the first winter Asian chum salmon swim northward against the southward wind-driven currents to stay in the western North Pacific. This suggests that Asian chum salmon require more energy to migrate than other stocks during the first winter of their ocean life. [source]


    Fish community characteristics of the lower Gambia River floodplains: a study in the last major undisturbed West African river

    Summary 1.,The Gambia River is the last major West African river that has not been impounded. However, a hydroelectric dam is being constructed and substantial changes to the hydrology and ecology of the system are expected. 2.,Little information is available on the impact of water impoundments in semi-arid regions on downstream floodplain fish communities, due to the scarcity of pre-intervention data. Because profound impacts on physical habitat, salinity and nutrient transport can occur downstream of such impoundments, a knowledge of the species-habitat associations of biota such as fishes is necessary for understanding likely changes and how to limit them. 3.,Fish were sampled using cast and hand nets along two transects on the floodplain, and with fyke nets in two ,bolongs' (creeks) from May to November 2005 and 2006 in the lower reaches of the Gambia River, close to the salt water front where ecological changes due to the construction of the dam are likely to be pronounced. 4.,Greatest fish species richness was associated with low conductivity, low pH and deep water. Bolongs held greater species richness compared with other floodplain habitats, probably because they acted as conduits for fish moving on and off the floodplain. Species richness and catch biomass increased rapidly following the first rains and then declined. 5.,Using a multivariate analysis, three main species groups were identified on the floodplain; one associated with deeper water, one with less brackish water and one with shallow, open water. Tilapia guineensis was the commonest species on the floodplains. 6.,The floodplains provide nursery habitats as many fish captured were immature, particularly for species where adults are mainly encountered in the main channel. Several small-sized floodplain specialists were also represented by a high proportion of mature individuals. 7.,Impoundment is expected to reduce seasonal flooding of the floodplain in the lower Gambia River, downstream of the impoundment, resulting in reduced occurrence of aquatic habitats, especially bolongs, together with lower dissolved oxygen and increased salinity, leading to alteration of the floodplain fish communities, benefiting salt-tolerant species, reducing overall species richness and probably reducing floodplain fish production.     Population size, weight distribution and food in a persistent population of the rare medicinal leech, Hirudo medicinalis

    Summary 1. It is important for species recovery and conservation management projects to know the minimum viable population size for rare and endangered species, such as the medicinal leech, Hirudo medicinalis. Therefore, using a catch-removal method, this study estimated every two years (1986, 1988, 1990, 1992) the total number of medicinal leeches in a tarn in the English Lake District, and the number of mature adults in the population. 2. Four samples were taken each year in June and July, when water temperatures exceeded 20 °C. Population size was estimated both by maximum likelihood and regression methods. All leeches were weighed alive and size groups were separated by polymodal frequency analysis. A small sample of the blood meal in each leech gut was taken before the leeches were returned to the tarn, and was used to estimate the proportion of mammalian and non-mammalian blood in the meals. 3. Both methods of estimation produced similar values, increasing confidence in the population estimates. Values for the total population in June and July varied among years from 248 to 288, the maximum value being only 16% higher than the minimum. Values for the number of mature leeches varied from 48 to 58 (19,20% of the total population), and this was an estimate of the effective population size. 4. There were four size groups. The largest mature leeches (live weight >5 g) in group IV formed only 1% of the population, and the smallest (0.02,0.5 g) in group I 14,17%. Most leeches were in two overlapping groups of immature (64,67% of population) and mature (18%) leeches with size ranges of 0.4,3.4 g and 2.5,5 g respectively. The percentage of leeches in each size group was very consistent among years. Blood meals were found in 38,44% of the leeches in group I, 45,50% in group II, 70,75% in group III, and 100% in group IV, but mammalian blood was present only in larger mature leeches (>3.5 g). 5. Medicinal leeches were first detected in the tarn in 1980 and are still present in 2007, so the population has persisted for at least 27 years. Compared with minimum viable population sizes for other species, including many endangered species, values for this medicinal leech population are extremely low, but may be typical of some rare freshwater invertebrates in isolated habitats.
