Structure elucidation for curvularides,A,E (1,5) was accomplished by analysis of spectroscopic data, as well as by single-crystal X-ray crystallography. Curvularide,B (2) exhibited antifungal activity against Candida albicans, and it also showed synergistic activity with a fluconazole drug. [source] HP(2,9)-magainin 2(1,12), a synthetic hybrid peptide, exerts its antifungal effect on Candida albicans by damaging the plasma membraneJOURNAL OF PEPTIDE SCIENCE, Issue 4 2004Yoonkyung Park Abstract In our previous study, HP(2,9)-MA(1,12), HP-MA for short, a hybrid peptide incorporating residues 2,9 of Helicobacter pylori ribosomal protein L1 (HP) and residues 1,12 of magainin 2 (MA) was shown to have strong antibacterial activity. In this study the antifungal activity of HP-MA was evaluated using various fungi, and it was shown that the activity was increased when compared with the parent peptides. In order to investigate the fungicidal mechanism(s) of HP-MA its action against fungal cell membranes was examined by the potassium-release test, which showed that HP-MA caused an increase in the amount of K+ released from the cells. Furthermore, HP-MA induced significant morphological changes. These facts suggested that the fungicidal effect of HP-MA involves damaging the fungal cell membranes. CD investigators suggested that the ,-helical structure of these peptides plays an important role in their antibiotic effect, but that ,-helicity is less directly correlated with the enhanced antibiotic activity of the hybrid. Nagi Reddy Dissociation pathways of a series of alkali-cationized hybrid peptides, viz., Boc- ,,, - and - ,,, -carbopeptides, synthesized from C-linked carbo- ,3 -amino acids [Caa (S)] and , -alanine (L-Ala), have been investigated by electrospray ionization tandem mass spectrometry. The positional isomers (six pairs) of the cationized ,,, - and ,,, -peptides can be differentiated by the collision-induced dissociation (CID) spectra of their [M,+,Cat-Boc,+,H]+ ions which give characteristic series of alkali-cationized C- (x, y, z) and N-terminal (a, b, c) ions. Another noteworthy difference is cationized ,,, -peptides eliminate a molecule of ammonia whereas this pathway is absent for ,,, -peptides. This is useful for identifying the presence of a , -amino acid at the N-terminus. The CID spectra of [M,+,Cat-Boc,+,H]+ ions of these peptide acids show abundant rearrangement [bn,+,17,+,Cat]+ (n,=,1 to n,1) ions which is diagnostic for distinguishing between , - and , -amino acid at the C-terminus. MSn experiments of [bn,+,Li,H]+ ions from these hybrid peptides showed the loss of CO and 72 u giving rise to [an,+,Li,H]+ and cationized nitrile product ions which render support to earlier proposals that b or [bn,+,Cat,H]+ ions have protonated or cationized oxazolinone structures, respectively. Hongbiao Abstract:, Ten kinds of hybrid peptides containing the N-terminal residues of cecropin B (CB) and C-terminal of thanatin (TH) were constructed and expressed as gluthathion S-transferase (GST)-fusion proteins. Variants were screened for the better biological activity, which was paralleled with the degree of growth inhibition of the transformant cells. The hybrid CB-TH g was selected as the best one among those hybrids by in vivo monitoring method and was chemical synthesized for in vitro antimicrobial activity analysis. The hybrid peptide showed rescued activity against several test strains when compared with the truncated isoforms of TH, suggesting that the peptides with different structure and mechanism could be used as templates for hybrid peptides design.
