A neurological examination score for the assessment of spinocerebellar ataxia 3 (SCA3)

Spinocerebellar ataxias (SCAs) are characterized by a heterogeneous set of clinical manifestations. Our aims were to assess the neurological features of SCA3, and to describe and test the feasibility, reliability, and validity of a comprehensive Neurological Examination Score for Spinocerebellar Ataxia (NESSCA). The NESSCA was administered to molecularly diagnosed SCA3 patients at an outpatient neurogenetics clinic. The scale, based on the standardized neurological examination, consisted of 18 items that yielded a total score ranging from 0 to 40. The score's interrater reliability and internal consistency were investigated, and a principal components analysis and a correlation with external measures were performed. Ninety-nine individuals were evaluated. Interrater reliability ranged from 0.8 to 1 across individual items (P < 0.001); internal consistency, indicated by Cronbach's alpha, was 0.77. NESSCA scores were significantly correlated with measures of disease severity: disease stage (rho = 0.76, P < 0.001), duration (rho = 0.56, P < 0.001), and length of CAG repeat (rho = 0.30, P < 0.05). NESSCA was a reliable measure for the assessment of distinct neurological deficits in SCA3 patients. Global scores correlated with all external variables tested, showing NESSCA to be a comprehensive measure of disease severity that is both clinically useful and scientifically valid. [source]


The effect of oxygen tension on the in vitro assay of human osteoblastic connective tissue progenitor cells

Abstract Connective tissue progenitors (CTPs) are defined as the heterogeneous set of stem and progenitor cells that reside in native tissues and are capable of proliferation and differentiation into one or more connective tissue phenotypes. CTPs play important roles in tissue formation, repair, and remodeling. Therefore, in vitro assays of CTP prevalence and biological potential have important scientific and clinical relevance. This study evaluated oxygen tension as an important variable in optimizing in vitro conditions for quantitative assays of human CTPs. Bone marrow aspirates were collected from 20 human subjects and cultured using established medium conditions at ambient oxygen tensions of 1, 5, 10, and 20%. Colony-forming efficiency (CFE), proliferation, and colony density were assessed. CFE and proliferation were greatest at 5% O2. Traditional conditions using atmospheric oxygen tension (20% O2) reduced CFE by as much as 32%. CFE and proliferation at 1% O2 were less than 5% O2 but comparable to that seen at 20% O2, suggesting that CTPs are relatively resilient under hypoxic conditions, a fact that may be relevant to their function in wound repair and their potential use in tissue engineering applications involving transplantation into settings of moderate to severe hypoxia. Changes in Adolescents' Interpersonal Experiences: Are They Being Prepared for Adult Relationships in the Twenty-First Century?

Trends across nations suggest that adulthood in the future will require greater social versatility, including abilities to function in relationships that are less scripted by community norms and that bridge multiple social worlds. This article assesses whether current changes in adolescents' interpersonal experience are likely to give them the social resources and competencies they will need. Changes in families are making them smaller, more diverse in social capital, and more responsive to adolescents. Changes in adolescents' nonfamily experience include more time in institutional settings; more involvement with peers; and more cycles of developing (and ending) relationships with a heterogeneous set of adults, friends, and, for many, romantic partners. The analysis suggests that these changes will provide many youth with greater opportunities to develop the more versatile interpersonal resources required in the future, but that many adolescents will have restricted opportunities to acquire these resources. Visual P3 amplitude and self-reported psychopathic personality traits: Frontal reduction is associated with self-centered impulsivity

Abstract Past studies have examined P3 amplitude as an index of cognitive function related to psychopathy with mixed results. Psychopathy is a heterogeneous set of dissociable traits, and no previous study has examined relationships between P3 and specific traits. A Two Process Theory (TPT) of psychopathy has recently been advanced predicting that P3 reductions are related to only one dimension. We evaluated the relationship between P3 and the two factors of the Psychopathic Personality Inventory (PPI) in 96 undergraduates who performed a visual task. One factor of the PPI, Self-Centered Impulsivity, is related to the dimension of the TPT predicted to underlie P3 reduction. Frontal amplitude reduction was uniquely and inversely related to this trait. The other PPI factor, Fearless Dominance, was associated with faster reaction times. Future work on psychopathic personality and P3 should evaluate whether relationships are unique to one personality dimension. [source]
