    Tail Muscle Free Amino Acid Concentration of Pacific White Shrimp, Litopenaeus vannamei, Fed Diets Containing Protein-bound versus Crystalline Amino Acids

    Tail muscle tissue free amino acids (FAA) concentration is reported for subadult (4.3 g) Litopenaeus vannamei fed diets containing only protein-bound amino acids, crystalline lysine as a supplement to protein-bound lysine, and only crystalline amino acids (CAA). FAA were determined in shrimp at 0.00, 0.25, 0.5, 1, 2, 4, and 8 h postfeeding. Highest total tissue concentrations of essential amino acids and individual amino acids were typically observed 4 h postfeeding for shrimp fed intact and crystalline lysine-supplemented feeds. Those shrimp offered diets containing only CAA showed no tissue maxima. Similar uptake patterns for lysine shown by shrimp fed the intact and crystalline lysine-supplemented diet indicate the potential use of small amounts of crystalline lysine in shrimp feeds.     Plasma Homocysteine, B Vitamins, and Amino Acid Concentrations in Cats with Cardiomyopathy and Arterial Thromboembolism

    Arterial thromboembolism (ATE) is a common complication of cats with cardiomyopathy (CM), but little is known about the pathophysiology of ATE. In people, high plasma concentrations of homocysteine and low B vitamin concentrations are risk factors for peripheral vascular disease. In addition, low plasma arginine concentrations have been linked to endothelial dysfunction. The purpose of this study was to compare concentrations of homocysteine, B vitamins, and amino acids in plasma of normal cats to those of cats with CM and ATE. Plasma concentrations of homocysteine, vitamin B6, vitamin B12, folate, and amino acids were measured in 29 healthy cats, 27 cats with CM alone, and 28 cats with both CM and ATE. No differences were found between groups in homocysteine or folate. Mean vitamin B12 concentration (mean ± standard deviation) was lower in cats with ATE (866 ± 367 pg/mL) and cats with CM (939 ± 389 pg/mL) compared with healthy controls (1,650 ± 700 pg/mL; P < .001). Mean vitamin B6 concentration was lower in cats with ATE (3,247 ± 1,215 pmol/mL) and cats with CM (3,200 ± 906 pmol/mL) compared with healthy control animals (4,380 ± 1,302 pmol/mL; P= .005). Plasma arginine concentrations were lower in cats with ATE (75 ± 33 nmol/mL) compared with cats with CM (106 ± 25 nmol/mL) and healthy control animals (96 ± 25 nmol/ mL; P < .001). Vitamin B12 concentration was significantly correlated with left atrial size. We interpret the results of this study to suggest that vitamin B12 and arginine may play a role in CM and ATE of cats.     Amino acid concentrations in blood serum of horses performing long lasting low-intensity exercise

    Summary The aim of this work was to evaluate the changes in the concentrations, after two rides different for distance covered, of different amino acids in endurance horses. Blood samples have been collected from horses just before the start, at the top of a steep slope (819 m difference in height) and just at the end of a 32-km endurance ride. A second group, competing in a 72 km endurance ride, has also been sampled immediately before and after the race. In serum samples, the concentrations of alanine, arginine, asparagine, glycine, isoleucine, histidine, leucine, lysine, methionine, ornithine, phenylalanine, tryptophan, tyrosine and valine have been measured by high-performance liquid chromatography (HPLC). anova and t -test have been used to study the differences in the concentrations of the amino acids. The pre-ride concentrations of the free amino acids were different between the two races, except for methionine and leucine. Differences between start and end race have been found for both groups for all the considered parameters except asparagine, isoleucine, leucine and lysine for the 72 km ride. Increases have been recorded for the shorter and decreases for the longer ride in the blood serum concentrations. Significant increases have also been found between the starting sampling and the second, at the top of the slope, only for alanine, arginine, asparagines, phenylalanine and lysine. The ride length has a significant impact on blood serum amino acids mobilization and uptake; in the shorter race the increases stand only for mobilization, whereas in the longer the decrease can be considered the effect of the onset of the amino acids catabolism.     Regional and Selective Effects of Oestradiol and Progesterone on NMDA and AMPA Receptors in the Rat Brain

    We investigated the effect of 10 months ovariectomy and a correction therapy, 2 weeks before the rats were killed, of oestradiol, progesterone or their combination on NMDA and AMPA receptor binding in the hippocampus, dentate gyrus, striatum, nucleus accumbens and frontal cortex of the rat brain as well as on amino acid levels in frontal cortex. NMDA and AMPA binding densities were assayed by autoradiography using, respectively, l -[3H]glutamate and [3H]AMPA; amino acid concentrations were measured by high performance liquid chromatograhy (HPLC) coupled with UV detection. Ovariectomy was without effect on NMDA and AMPA binding density in all brain regions assayed except in the hippocampal CA1 region and dentate gyrus where it decreased NMDA binding density compared to intact rats values. Oestradiol restored and increased NMDA binding density in the CA1 subfield and the dentate gyrus of ovariectomized rats but, by contrast, it decreased binding density in the striatum and in the frontal cortex while having no effect in the CA2/3 subfield of the hippocampus and in the nucleus accumbens. Oestradiol was without effect on AMPA binding density in the hippocampus and the dentate gyrus but it reduced AMPA binding density in the striatum, the frontal cortex and the nucleus accumbens. Progesterone, and oestradiol combined with progesterone, decreased NMDA but not AMPA binding density in the frontal cortex of ovariectomized rats, and they were without effect on these receptors in the other brain regions assayed. Amino acid concentrations in the frontal cortex were unchanged after ovariectomy or steroid treatments. The effect of oestradiol in the hippocampus confirmed in the present study and our novel findings in the frontal cortex, striatum and nucleus accumbens may have functional significance for schizophrenia and neurodegenerative diseases.     Amino acid composition of Arctic charr, Salvelinus alpinus (L.) and the prediction of dietary requirements for essential amino acids

    Abstract Embryo somatic tissues, non-somatic yolk-sac materials, and whole, individual fingerlings (age 0+) of Arctic charr, Salvelinus alpinus (L.), as well as a commercial trout diet, were analysed for a wide spectrum of amino acids. Analytical material consisted of prefeeding swim-up fry that were separated into discrete yolk sac and somatic embryo tissue samples. Amino acid concentrations in fry somatic tissue and whole fingerlings were generally very similar to each other, but were lower than those measured in yolk materials. Higher correlations were observed between the majority of specific amino acid concentrations in the trout diet when compared with fingerling data (r2 = 0.91) and fry somatic tissue data (r2 = 0.89), than when correlated with fry yolk sac material (r2 = 0.76). These results indicate that the essential amino acid profiles of fry somatic tissue and whole fingerlings are closer to that of a commercial feed than they are to the endogenous profiles found in the embryonic yolk sac material itself. The dietary ratios of individual essential amino acids were also compared with the total essential amino acid concentrations (A/E ratios) in whole fingerling tissues, and these ratios could be used to accurately estimate the apparent essential amino acid requirements of Arctic charr. The rationale for using carcass amino acid composition data to estimate the dietary essential amino acid requirements of Arctic charr is discussed. [source]


    Flexibility in the composition and concentration of amino acids in honeydew of the drepanosiphid aphid Tuberculatus quercicola

    Abstract 1. Mutualistic interactions between aphids and ants are mediated by honeydew that aphids produce. Previous work showed that when attended by the ant Formica yessensis Forel (Hymenoptera: Formicidae), nymphs of the aphid Tuberculatus quercicola (Matsumura) (Homoptera: Aphididae) developed into significantly smaller adults with lower fecundity than did nymphs that were not ant attended. 2. This study tested the hypothesis that this cost of ant attendance arises through changes in the quality and quantity of honeydew. Ant-attended and ant-excluded aphid colonies were prepared in the field. The composition and concentration of amino acids were compared between the honeydew produced by ant-attended colonies and that produced by ant-excluded colonies. 3. The aphids excreted smaller droplets of honeydew, but also excreted them more frequently, in ant-attended colonies than in ant-excluded colonies. The honeydew of ant-attended aphids contained more types of amino acid, and a significantly higher total concentration of amino acids, than did the honeydew of ant-excluded aphids. 4. These results suggest that the increase in the concentration of amino acids in honeydew leads to a shortage of nitrogen available for aphid growth and reproduction, resulting in lower performance under ant attendance. 5. With the advance of seasons, a significant reduction was found in both the total free amino acid concentration in phloem sap and the frequency of honeydew excretion; however the total concentration of amino acids in the honeydew did not vary significantly during the seasons, suggesting that aphids keep the quality of honeydew constant in order to maintain ant visitation.     THE ROLE OF BACTERIAL SYMBIONTS IN AMINO ACID COMPOSITION OF BLACK BEAN APHIDS

    Abstract To evaluate the role of bacterial symbionts (Buchnera spp.) in the black bean aphids (Aphis craccivora Koch), the aphids were treated with the antibiotic, rifampicin, to eliminate their intracellular symbiotic bacteria. Analysis of protein and amino acid concentration in 7-day-old of aposymbiotic aphids showed that the total protein content per mg fresh weight was significantly reduced by 29%, but free amino acid titers were increased by 17%. The ratio of the essential amino acids was in general only around 20% essential amino acids in phloem sap of broad bean, whereas it was 44% and 37% in symbiotic and aposymbiotic aphids, respectively, suggesting that the composition of the free amino acids was unbalanced. For example, the essential amino acid, threonine represented 21.6% of essential amino acids in symbiotic aphids, but it was only 16.7% in aposymbiotic aphids. Likewise, two nonessential amino acids, tyrosine and serine, represented 8.9% and 5.6% of total amino acids in symbiontic aphids, respectively, but they enhanced to 21.1% and 13.6% in aposymbiotic aphids. It seems likely that the elevated free amino acid concentration in aposymbiotic aphids was caused by the limited protein anabolism as the result of the unbalanced amino acid composition. [source]


    Changes in amino acid composition in the tissues of African catfish (Clarias gariepinus) as a consequence of dietary L-carnitine supplements

    A study was undertaken to examine the effect of different amounts of dietary lysine (13 and 21 g kg,1 diet), lipid (80 and 160 g kg,1 diet) and L -carnitine (0.2 and 1.0 g kg,1 diet) on growth performance, proximate composition and amino acid metabolism of the African catfish (Clarias gariepinus). Juvenile African catfish (23 ± 1.5 g/fish) were stocked into 70-L aquaria (16 aquaria, 28 fish/aquarium) connected to a recirculation system during a maximum period of 74 days. All groups were fed at a level of 24 g kg,0.8 day,1 in an experiment run at pair feeding. Animals receiving 1.0 g carnitine accumulated up to six times more carnitine in their tissues than animals receiving 0.2 g (P < 0.05). Acyl-carnitine and free L -carnitine levels increased in the whole body and in tissues. Dietary L -carnitine supplements increased protein-to-fat ratios in the body, but did not affect growth rate. Protein-to-fat ratios were only affected when the biosynthesis capacity of L -carnitine was restricted due to low lysine levels and when there was a shortage of dietary fat. When lysine was offered at 21 g kg,1 feed, dietary L -carnitine supplements did not affect the amino acid concentrations of body tissues. Dietary L -carnitine supplements raised the concentration of glutamic acid,>,aspartic acid,>,glycine > alanine > arginine > serine > threonine in skeletal muscle tissue (P < 0.05). Total amino acid concentration in muscle and liver tissues (dry-matter basis) increased from 506 to 564 and from 138 to 166 mg g,1, respectively, when diets were offered with high L -carnitine, low lysine and low fat levels. These data suggest that dietary L -carnitine supplementation may increase fatty acid oxidation and possibly decrease amino acid combustion for energy. [source]


    NITZSCHIA OVALIS (BACILLARIOPHYCEAE) MONO LAKE STRAIN ACCUMULATES 1,4/2,5 CYCLOHEXANETETROL IN RESPONSE TO INCREASED SALINITY,

    The growth of microalgae in hypersaline conditions requires that cells accumulate osmoprotectants. In many instances, these are polyols. We isolated the diatom Nitzschia ovalis H. J. Arn. from the saline and alkaline water body Mono Lake (CA, USA). This isolate can grow in salinities ranging from 5 to 120 parts per thousand (ppt) of salt but normally at 90 ppt salinity. In this report, we identified the major polyol osmoprotectant as 1,4/2,5 cyclohexanetetrol by electron ionization-mass spectrometry (EI,MS), 1H, 13C nuclear magnetic resonance spectroscopy (NMR), and infrared (IR) and showed an increase in cellular concentration in response to rising salinity. This increase in the cyclitol concentration was evaluated by gas chromatography of the derived tetraacetylated cyclohexanetetrol obtaining an average of 0.7 fmol · cell,1 at 5 ppt and rising to 22.5 fmol · cell,1 at 120 ppt. The 1,4/2,5 cyclohexanetetrol was also detected in the red alga Porphyridium purpureum. Analysis of the free amino acid content in N. ovalis cultures exposed to changes in salinity showed that proline and lysine also accumulate with increased salinity, but the cellular concentration of these amino acids is about 10-fold lower than the concentration of 1,4/2,5 cyclohexanetetrol. The comparison of amino acid concentration per cell with cyclitol suggests that this polyol is important in compensating the cellular osmotic pressure due to increased salinity, but other physiological functions could also be considered.     Nitrate modifies the assimilation pattern of ammonium and urea in wheat seedlings

    Abstract BACKGROUND: In certain plant species, ammonium or urea nutrition can cause negative effects on plant development which can result in toxic symptoms. Some authors suggest that the presence of nitrate can alleviate these symptoms by increasing ammonium and urea assimilation, avoiding its accumulation. In order to study this hypothesis, wheat (Triticum aestivum L.) seedlings were grown with various nitrogen supplies containing the main nitrogen forms (ammonium, nitrate and urea). Amino acids content and the activity of the three main enzymes involved in nitrogen assimilation (nitrate reductase, glutamine synthetase and urease) were studied. RESULTS: The application of nitrate along with urea and/or ammonium was not associated with a time-sustained increase in the activity of glutamine synthetase and urease. Amino acid analysis revealed that nitrate induced changes in amino acid metabolism enhancing its concentration. Likewise the content of protein was also higher in nitrate-treated plants. CONCLUSION: These results suggest that the effect of nitrate is compatible with a rapid and transient increase in the activity of glutamine synthetase and urease during the first hour after the onset of treatments. Nevertheless, a possible effect of nitrate reducing ammonium accumulation through the activation of alternative metabolic pathways different from that involving glutamine synthetase cannot be ruled out. Finally, nitrate effects on amino acid concentration indicate that, whereas ammonium assimilation takes place principally in the root, urea and nitrate assimilation occurred in the shoot, under the conditions of the experiment.     Honeydew amino acids in relation to sugars and their role in the establishment of ant-attendance hierarchy in eight species of aphids feeding on tansy (Tanacetum vulgare)

    Abstract., The ratio of the concentration of honeydew total amino acids to total sugars in the honeydew of eight species of aphids, all feeding on tansy, Tanacetum vulgare (L.), was determined and correlated with honeydew production and ant-attendance. The honeydew of the five ant-attended aphid species [Metopeurum fuscoviride (Stroyan), Trama troglodytes (v. Hayd), Aphis vandergooti (Börner), Brachycardus cardui (L.), Aphis fabae (Scopoli)] was rich in total amino acids, ranging from 12.9 to 20.8 nmol µL,1 compared with the unattended aphid Macrosiphoniella tanacetaria (Kalt.) with only 3 nmol µL,1. Asparagine, glutamine, glutamic acid and serine (all nonessential amino acids) were the predominant amino acids in the honeydew of all species. The total concentration of amino acids in the phloem sap of tansy was much higher (78.7 nmol µL,1) then in the honeydew samples, and the predominant amino acids were glutamate (34.3%) and threonine (17.7%). A somewhat unexpected result was the finding that those aphid species with the highest total amino acid concentration in the honeydew always had the highest concentration of sugars. The lowest amino acid,sugar combined value was 104,28.8 nmol µL,1 in the non ant-attended species M. tanacetaria, and the highest value was an average of 270,89.9 nmol µL,1 for the three most intensely attended aphid species M. fuscoviride, A. vandergooti and T. troglodytes. There is no evidence that any single amino acid or group of amino acids in the honeydew acted as an attractant for ant-attendance in these eight aphid species. The richness of the honeydew (rate of secretion × total concentration of sugars), along with the presence of the attractant sugar melezitose, comprised the critical factors determining the extent of ant-attendance of the aphids feeding on T. vulgare. The high total amino acid concentration in sugar-rich honeydews can be explained by the high flow-through of nutrients in aphids that are particularly well attended by ants. [source]


    Defined Protein-Free NS0 Myeloma Cell Cultures: Stimulation of Proliferation by Conditioned Medium Factors

    A chemically defined, protein-free, and animal-component-free medium, designated RITM01, has been developed for NS0 myeloma cells. The basal medium used was a commercial serum-free and protein-free hybridoma medium, which was supplemented with phosphatidylcholine, cholesterol, ,-cyclodextrin, and ferric citrate. Increasing the amino acid concentration significantly improved cell growth. An NS0 cell line, constitutively producing a human IgG1 antibody, reached a peak cell density of 3 × 106 cells mL,1 in this medium. The antibody yield was 195 mg L,1 in batch culture, which is a 3-fold increase compared to that of a standard serum-supplemented medium, even though the cell yield was the same. The increase in antibody yield was a consequence of a longer growth phase and a slight increase in specific antibody production rate at low specific proliferation rates. Adaptation of the NS0 myeloma cell line to the protein-free conditions required about 3 weeks before viability and cell densities were stabilized. Most probably, changes in gene expression and phenotypic behavior necessary for cell survival and proliferation occurred. We hypothesize that mitogenic factors produced by the cells themselves are involved in autocrine control of proliferation. To investigate the presence of such factors, the effect of conditioned (spent) medium (CM) on cell growth and proliferation was studied. Ten-fold concentrated CM, harvested at a cell density of 2 × 106 cells mL,1, had a clear positive effect on proliferation even if supplied at only 2.5% (v/v). CM was found to contain significant amounts of extracellular proteins other than the antibody. Fractionation of CM on a gel filtration column and subsequent supplementation of new NS0 cultures with the individual fractions showed that factors eluting at 20,25 kDa decreased the lag phase and increased the peak cell density as compared to control cultures. Identification of autocrine factors involved in regulation of proliferation may lead to completely new strategies for control of growth and product formation in animal cell processes. [source]


    Forearm and leg amino acid metabolism in the basal state and during combined insulin and amino acid stimulation after a 3-day fast

    Abstract Aim:, Fasting is characterized by a progressive loss of protein, but data on protein kinetics are unclear and few have studied the effects of re-feeding. The present study was designed to test the hypothesis that a combined infusion of insulin and amino acids after fasting would induce compensatory increases in protein synthesis and reductions in protein breakdown at the whole body level and in muscle. Methods:, We included 10 healthy male volunteers and studied them twice: (1) in the post-absorptive state and (2) after 72 h of fasting. Amino acid kinetics was measured using labelled phenylalanine and tyrosine, whole body energy expenditure was assessed and urea nitrogen synthesis rates were calculated. Results:, After fasting we observed an increase in arterial blood concentration of branched chain amino acids and a decrease in gluconeogenic amino acids (P < 0.05). Isotopically determined whole body, forearm and leg phenylalanine fluxes were unaltered apart from a 30% decrease in phenylalanine-to-tyrosine conversion (2.0 vs. 1.4 ,mol kg,1 h,1, P < 0.01). During infusion of insulin and amino acids, amino acid concentrations increased. Conclusion:, Our data indicate that after a 72-h fast basal and insulin/amino acid-stimulated regional phenylalanine fluxes in leg and forearm muscle are unaltered. During fasting concentrations of gluconeogenic amino acids decrease and hepatic and/or renal phenylalanine-to-tyrosine conversion decreases. Thus, as opposed to glucose and lipid metabolism, fasting does not induce insulin resistance as regards amino acid metabolism. [source]


    Volatile composition and proteolysis in traditionally produced mature Kashar cheese

    Summary Twelve samples of raw milk mature Kashar cheese at different stages of ripening were collected from retail outlets. The average pH, moisture, fat-in-dry matter, protein, salt-in-dry matter and titratable acidity contents of the samples were 5.33, 39.39%, 45.20%, 27.33%, 6.62% and 0.65% (as lactic acid), respectively. Indices of proteolysis varied from 10.72% to 23.75% and 7.09% to 12.26% for pH 4.6-soluble and 12% trichloroacetic acid-soluble nitrogen fractions, respectively, and total free amino acid concentrations ranged from 6.36 to 36.03 mg Leu g,1 of cheese. The cheeses were analysed for volatile compounds by Solid Phase Microextraction and Gas Chromatography-Mass Spectrometry (GC-MS). A total of 113 compounds were detected and identified belonging to the following chemical groups: acids (eleven), esters (sixteen), ketones (sixteen), aldehydes (six), alcohols (twenty-seven), sulphur compounds (seven), terpenes (seven) and miscellaneous compounds (twenty-three). The potential effect of each compound on the flavour profile of Kashar cheese is discussed. Acids, esters, ketones and alcohols were found at considerable levels in the samples. Kashar cheeses obtained from different retail outlets displayed some differences in terms of chemical composition, proteolysis and patterns of aroma compounds; and may be attributed to their production technologies and age-related variations.     Regional and Selective Effects of Oestradiol and Progesterone on NMDA and AMPA Receptors in the Rat Brain

    We investigated the effect of 10 months ovariectomy and a correction therapy, 2 weeks before the rats were killed, of oestradiol, progesterone or their combination on NMDA and AMPA receptor binding in the hippocampus, dentate gyrus, striatum, nucleus accumbens and frontal cortex of the rat brain as well as on amino acid levels in frontal cortex. NMDA and AMPA binding densities were assayed by autoradiography using, respectively, l -[3H]glutamate and [3H]AMPA; amino acid concentrations were measured by high performance liquid chromatograhy (HPLC) coupled with UV detection. Ovariectomy was without effect on NMDA and AMPA binding density in all brain regions assayed except in the hippocampal CA1 region and dentate gyrus where it decreased NMDA binding density compared to intact rats values. Oestradiol restored and increased NMDA binding density in the CA1 subfield and the dentate gyrus of ovariectomized rats but, by contrast, it decreased binding density in the striatum and in the frontal cortex while having no effect in the CA2/3 subfield of the hippocampus and in the nucleus accumbens. Oestradiol was without effect on AMPA binding density in the hippocampus and the dentate gyrus but it reduced AMPA binding density in the striatum, the frontal cortex and the nucleus accumbens. Progesterone, and oestradiol combined with progesterone, decreased NMDA but not AMPA binding density in the frontal cortex of ovariectomized rats, and they were without effect on these receptors in the other brain regions assayed. Amino acid concentrations in the frontal cortex were unchanged after ovariectomy or steroid treatments. The effect of oestradiol in the hippocampus confirmed in the present study and our novel findings in the frontal cortex, striatum and nucleus accumbens may have functional significance for schizophrenia and neurodegenerative diseases.     Mulberry nutrient management for silk production in Hubei Province of China

    Abstract The silk industry is important for south China's rural economy. Leaves of mulberry (Morus spp.) are used for silkworm production. Hubei province is one of the main silk-producing provinces in China. The objectives of this research were to survey the fertilization practices in the mulberry-producing regions in the province and to determine the best nutrition-management practice for mulberry plantations. A survey and a series of field experiments with N, P, K, and micronutrients were conducted from 2001 to 2002. In addition, a silkworm-growth experiment was also conducted by feeding leaves harvested from various fertilization treatments. The results indicate that poor soil fertility and unbalanced fertilization were the main factors limiting mulberry-leaf yield and quality in Hubei province. Nitrogen fertilization of mulberry has reached a high level (454,kg ha,1 y,1) in Hubei province, but P- and K-fertilization rates have not been matched with N-fertilization rates as farmers are not aware of the significance of P and K. Balanced fertilization showed positive nutrient interactions with respect to mulberry yield and quality. Potassium application increased yield and quality (protein and sugar concentration) of mulberry leaves. Silkworm growth and cocoon quality were improved when silkworms were fed with the leaves derived from K-fertilized plants in comparison with those taken from control plots. Application of Mg, S, and B also significantly improved leaf sugar, essential and total amino acid concentrations, but did not increase leaf yield significantly. It is concluded that a fertilizer dose of 375,kg N ha,1, 66,kg P ha,1, and 125,kg K ha,1 is suitable for the cultivation of mulberry in the Hubei province along with Mg, S, and B, wherever necessary, for the improvement of yield and quality of mulberry leaves.     Effects of L-Asparaginase on Plasma Amino Acid Profiles and Tumor Burden in Cats with Lymphoma

    Background: L-Asparaginase (Elspara), is an Escherichia coli -derived enzyme that depletes lymphoma cells of asparagine, inhibiting protein synthesis and resulting in cell death. The single agent response rate in cats with lymphoma and impact of L-asparaginase on plasma amino acid concentrations is unknown. Hypotheses: L-Asparaginase significantly reduces plasma asparagine concentrations and has demonstrable efficacy against untreated lymphoma in cats. Animals: Thirteen cats with confirmed lymphoma (LSA) of any anatomic site were given 1 dose 400 IU/kg IM) of L-asparaginase for initial LSA treatment. Methods: Plasma collected at 0, 2, and 7 days after L-asparaginase therapy was assayed for ammonia, asparagine, aspartic acid, glutamine, and glutamic acid concentrations. Cats were restaged 7 days later to assess tumor response. Results: Eight cats had T-cell LSA, 4 cats had B-cell LSA, and 1 cat's immunophenotype was unknown. Two complete and 2 partial responses to L-asparaginase were seen. Four cats had stable disease, and 5 cats had progressive disease. Ammonia and aspartic acid concentrations were increased from baseline at 2 and 7 days posttreatment. Asparagine concentrations were decreased from baseline at 2 days but not 7 days posttreatment. Glutamic acid concentrations were increased at day 2 compared to day 7 posttreatment but not compared to baseline. Glutamine concentrations were unchanged. Conclusions and Clinical Importance: L-Asparaginase significantly reduced asparagine concentrations within 2 days of treatment, but this effect was lost within 7 days. The apparent overall response rate of feline LSA to L-asparaginase in this study was 30%. [source]


    Effects of etomidate on free intracellular amino acid concentrations in polymorphonuclear leucocytes in vitro

    Background: Previous studies have shown the inhibitory effects of etomidate on polymorphonuclear leucocyte (PMN) function. No reports exist, however, regarding free intracellular amino acid metabolism, although physiological cell metabolism and basic cell functions rely upon a balanced intracellular amino acid content and the cell membrane-mediated separation of cellular amino acids from the extracellular plasma amino acid pool. Thus, in the current study, we evaluated the effects of etomidate on free intracellular amino acid metabolism in PMN. Methods: With ethics committee approval, blood was withdrawn from 35 healthy volunteers and incubated (1 h) either with 0 ,g/ml, 0.0156 ,g/ml, 0.0625 ,g/ml or 0.5 ,g/ml of etomidate as well as with its additives (propylene glycol and Lipofundin MCT® 10%). The PMN were separated using standardized Percoll® -gradient and centrifugation procedure before deep-freezing and lyophilization techniques were employed. All PMN samples were dissolved in methanol/H2O, and the concentrations of free intracellular amino acids were monitored using both novel advanced PMN-separation and high-performance liquid chromatography techniques. Results: Etomidate influenced important free amino acid profiles in PMN in a dose-dependent manner, indicating complex changes of cellular amino acid turnover. Neither propylene glycol nor Lipofundin MCT® 10% changed free amino acid concentrations in PMN. Conclusions: For the first time, the effects of etomidate on free intracellular amino acid metabolism in PMN have been investigated. Our results draw attention to the biochemical pathways which may be involved in etomidate-induced alterations in PMN function and cellular immunocompetence. [source]


    Local and systemic effects of two herbivores with different feeding mechanisms on primary metabolism of cotton leaves

    ABSTRACT Caterpillars and spider mites are herbivores with different feeding mechanisms. Spider mites feed on the cell content via stylets, while caterpillars, as chewing herbivores, remove larger amounts of photosynthetically active tissue. We investigated local and systemic effects of short-term caterpillar and spider mite herbivory on cotton in terms of primary metabolism and growth processes. After short-term caterpillar feeding, leaf growth and water content were decreased in damaged leaves. The glutamate/glutamine ratio increased and other free amino acids were also affected. In contrast, mild spider mite infestation did not affect leaf growth or amino acid composition, but led to an increase in total nitrogen and sucrose concentrations. Both herbivores induced locally increased dark respiration, suggesting an increased mobilization of storage compounds potentially available for synthesis of defensive substances, but did not affect assimilation and transpiration. Systemically induced leaves were not significantly affected by the treatments performed in this study. The results show that cotton plants do not compensate the loss of photosynthetic tissue with higher photosynthetic efficiency of the remaining tissue. However, early plant responses to different herbivores leave their signature in primary metabolism, affecting leaf growth. Changes in amino acid concentrations, total nitrogen and sucrose content may affect subsequent herbivore performance. [source]


    Measurement of xylem sap amino acid concentrations in conjunction with whole tree transpiration estimates spring N remobilization by cherry (Prunus avium L.) trees

    Abstract Prunus avium trees were grown in sand culture for one vegetative season with contrasting N supplies, in order to precondition their N storage capacities. During the spring of the second year a constant amount of 15N was supplied to all the trees, and the recovery of unlabelled N in the new biomass production was used as a direct measure of N remobilization. Destructive harvests were taken during spring to determine the pattern of N remobilization and uptake. Measurements of both xylem sap amino acid profiles and whole tree transpiration rates were taken, to determine whether specific amino acids are translocated as a consequence of N remobilization and if remobilization can be quantified by calculating the flux of these amino acids in the xylem. Whereas remobilization started immediately after bud burst, N derived from uptake by root appeared in the leaves only 3 weeks later. The tree internal N status affected both the amount of N remobilization and its dynamics. The concentration of xylem sap amino acids peaked shortly after bud burst, concurrently with the period of fastest remobilization. Few amino acids and amides (Gln, Asn and Asp) were responsible for most of N translocated through the xylem; however, their relative concentration varied over spring, demonstrating that the transport of remobilized N occurred mainly with Gln whereas transport of N taken up from roots occurred mainly with Asn. Coupling measurements of amino acid N in the xylem sap with transpiration values was well correlated with the recovery of unlabelled N in the new biomass production. These results are discussed in relation to the possibility of measuring the spring remobilization of N in field-grown trees by calculating the flux of N translocation in the xylem. [source]


    Effect of amino acids on cryopreservation of cynomolgus monkey (macaca fascicularis) sperm

    Abstract The effects of three amino acids (proline, glutamine, and glycine) added to the freezing medium Tes-Tris-egg yolk (TTE) for cryopreservation of cynomolgus monkey (Macaca fascicularis) spermatozoa were studied. This is the first report on the effects of amino acids on nonhuman primate sperm cryopreservation. The addition of 5mM proline, 10mM glutamine, and 10 or 20mM glycine each significantly improved post-thaw sperm motility and membrane and acrosome integrity compared with the control (TTE alone). However, a significant decrease in motility and membrane/acrosome integrity was observed when amino acid concentrations increased to 60mM for proline and glutamine, and 80mM for glycine.     Cerebrospinal fluid of brain trauma patients inhibits in vitro neuronal network function via NMDA receptors,

    Neurological diseases frequently induce pathological changes of cerebrospinal fluid (CSF) that might secondarily influence brain activity, as the CSF,brain barrier is partially permeable. However, functional effects of CSF on neuronal network activity have not been specified to date. Here, we report that CSF specimens from patients with reduced Glasgow Coma Scale values caused by severe traumatic brain injury suppress synchronous activity of in vitro-generated neuronal networks in comparison with controls. We present evidence that underlying mechanisms include increased N -methyl- D- aspartate receptor activity mediated by a CSF fraction containing elevated amino acid concentrations.     Predicting the optimal dietary essential amino acid profile for growth of juvenile yellow perch with whole body amino acid concentrations

    Abstract Rapid methods of estimating dietary essential amino acid (EAA) requirements might facilitate increases in aquaculture production, particularly for new or emerging industries. We conducted a 12-week feeding study to test the hypothesis that whole body EAA concentrations and the quantified methionine requirement could be used to predict the remaining dietary EAA requirements for juvenile all-female yellow perch. Six purified diets were developed and fed to triplicate groups of fish for 12 weeks. The diets contained the EAA profile of fishmeal (FM), the profile as predicted by whole body analysis, the quantified methionine requirement and resulting A/E ratios (PRED), PRED + 20% of all EAA (PRED20), PRED + 40% of all EAA (PRED40), PRED + 20% threonine, isoleucine and tryptophan (PRED320), and PRED + 40% threonine, isoleucine and typtophan (PRED340). Mean weight gain and feed consumption were significantly higher in fish fed PRED20 (35.7 ± 3.2 and 55.0 ± 5.3 g, respectively) than in fish fed FSM (25.1 ± 0.4 g wt gain, 41.0 ± 1.9 g cons), PRED (23.4 ± 2.3 g wt gain, 40.1 ± 4.2 g cons) and PRED340 (22.9 ± 3.3 g wt gain, 35.0 ± 3.8 g cons). There was no significant difference in feed efficiency among treatments. We recommend an EAA profile similar to PRED20 for feeding all-female juvenile yellow perch. [source]


    Amino acid composition of Arctic charr, Salvelinus alpinus (L.) and the prediction of dietary requirements for essential amino acids

    Abstract Embryo somatic tissues, non-somatic yolk-sac materials, and whole, individual fingerlings (age 0+) of Arctic charr, Salvelinus alpinus (L.), as well as a commercial trout diet, were analysed for a wide spectrum of amino acids. Analytical material consisted of prefeeding swim-up fry that were separated into discrete yolk sac and somatic embryo tissue samples. Amino acid concentrations in fry somatic tissue and whole fingerlings were generally very similar to each other, but were lower than those measured in yolk materials. Higher correlations were observed between the majority of specific amino acid concentrations in the trout diet when compared with fingerling data (r2 = 0.91) and fry somatic tissue data (r2 = 0.89), than when correlated with fry yolk sac material (r2 = 0.76). These results indicate that the essential amino acid profiles of fry somatic tissue and whole fingerlings are closer to that of a commercial feed than they are to the endogenous profiles found in the embryonic yolk sac material itself. The dietary ratios of individual essential amino acids were also compared with the total essential amino acid concentrations (A/E ratios) in whole fingerling tissues, and these ratios could be used to accurately estimate the apparent essential amino acid requirements of Arctic charr. The rationale for using carcass amino acid composition data to estimate the dietary essential amino acid requirements of Arctic charr is discussed. [source]


    The whole amino acid profile as indicator of the nutritional condition in cultured marine fish larvae

    Abstract The effect of variations in the amount or quality of food provided on the amino acid profile of larvae, was tested in two marine fish species, the Senegal sole (Solea senegalensis) and the sea bass (Dicentrarchus labrax). The objective was to assess whether such nutritionally suboptimum dietary patterns, which simulate situations that may occur in hatcheries, result in variations in the concentration of one or more amino acids that could be used as indicators of nutritional condition of larvae. Restriction in the normal food ration by 60% had no clear effect on the amino acid profile of sea bass, while the concentration of some amino acids (Arg, Ala and Phe) showed significant variations in sole. Feeding on artificial diets, which have shown their inadequacy a priori, resulted also in no significant effect on the profile of sea bass, but in a dramatic modification of the amino acid profile in sole. In addition, changes in the amino acid profiles considered as a whole were evaluated using the cluster analysis instead of paired comparisons between amino acid concentrations in each treatment. The analysis clearly separated profiles of larvae fed restrictedly or inadequately from their controls, irrespective of the species. This demonstrated the potential value of evaluating whole changes in amino acid profile as nutritional indicator. Besides, it was demonstrated that sensitivity of larvae to nutritional changes that may be reflected in the amino acid profile is greatly conditioned by the species and developmental stage.     Biochemical responses of matrinxăBrycon cephalus (Günther, 1869) after sustained swimming

    Abstract Juvenile matrinxă, Brycon cephalus, were submitted to sustained swimming for 72 days at 1.0 body length s,1. Exercised fish (EF) grew more than non-EF and their feed conversion ratio (FCR) improved; haematological responses demonstrated a decrease in haemoglobin and mean cell haemoglobin contents and increase in the mean cell volume. In the plasma, sodium, ammonia and amino acid concentrations increased; plasma triglycerides decreased while free fatty acids increased. Liver glucose, free amino acids, ammonia, the rate protein per fish weight and total lipid content increased, while the glycogen per fish ratio declined. Glutamate dehydrogenase (GDH) activity increased while pyruvate kinase (PK) and lactate dehydrogenase (LDH) decreased. White muscle glucose, lactate, the glycogen per fish-weight ratio and total lipid content exhibited a decrease in their values; ammonia, free amino acids and the protein per fish-weight ratio increased. GDH and PK decreased their activities. In the red muscle glycogen store, the glycogen per fish-weight ratio and glucose were reduced. Juvenile matrinxăs, under sustained swimming, were physiologically and biochemically adapted to exercise as indicated by improved blood flow, transport and oxygen uptake, FCR, amino acid and protein incorporation and growth. Continuous exercise is a good practice for B. cephalus cultivation. [source]


    The patterns of plasma free amino acids after force-feeding in rainbow trout Oncorhynchus mykiss (Walbaum) with and without dorsal aorta cannulation

    Abstract Two experiments were conducted to compare the patterns of plasma free amino acid concentrations after force-feeding in rainbow trout Oncorhynchus mykiss (Walbaum) with and without dorsal aorta cannulation. In the first experiment, 35 rainbow trout averaging 504 ± 7.8 g (mean ± SD) were divided into seven groups of five fish each. After 48 h starvation, a group of fish was anaesthetized and blood samples were taken at one of the following time periods: 0, 4, 8, 12, 24, 36 and 48 h after feeding. In the second experiment, five dorsal aorta cannulated rainbow trout averaging 511 ± 6.2 g (mean ± SD) were kept in a cage. After 48 h starvation, the fish were anaesthetized and blood samples were taken from the same fish at 0, 4, 8, 12, 24, 36 and 48 h after feeding. In the first experiment, the concentration of all plasma free amino acids except histidine and glycine peaked at 4 h and returned to the basal level 24 h after feeding. In the second experiment, the concentration of all plasma free amino acids except isoleucine, leucine, phenylalanine and tryptophan also peaked at 4 h and returned to the basal level 24 h after feeding. These results showed that the pattern of plasma free amino acid concentrations from fish with and without dorsal aorta cannulation were similar. [source]


    Effects of timing and rate of N supply on leaf nitrogen status, grape yield and juice composition from Shiraz grapevines grafted to one of three different rootstocks

    Abstract Yeast cells have a minimum N requirement to ferment a must through to dryness, so that grape N content (hence must N) becomes critical in meeting that prerequisite. Viticultural practices aimed at meeting that N requirement are of special relevance because interactions between rootstock and vineyard nitrogen supply strongly influence scion mineral nutrient status as well as shoot vigour, and via those processes, fruit composition. Such outcomes were investigated in a field trial involving Shiraz on three rootstocks viz. Teleki 5C, Schwarzmann and Ramsey. Five N supply regimes, varying from 0 to 80 kg/(haseason), were imposed through a drip-irrigation system during two periods (either flowering to veraison, or post-harvest to leaf-fall, or both) over three successive growing seasons. Post-harvest N supply increased scion leaf N and nitrate N concentrations at flowering for vines on Teleki 5C and Schwarzmann. By veraison, N recently applied in the flowering to veraison period elevated these indicators of N status in all vines on all rootstocks. Grape yields from vines on Teleki 5C and Schwarzmann were elevated by N supply after harvest, whereas juice soluble solids levels were lowered. Free amino acids in Shiraz juice were dominated by non-assimilable N, amounting to about 50% or more of the total free amino-N in the juice. Increasing N supply increased free amino acid concentrations in the juice of berries from vines on all rootstocks, but only vines on Schwarzmann derived any benefit from N supplied after harvest. The highest concentrations of free amino acids were measured in the berries from vines on Schwarzmann receiving 80 kg N/(ha.season). Of immediate practical relevance for N management of Shiraz grapevines on either Teleki 5C or Ramsey rootstocks, the minimum value for assimilable free amino-N concentration required to ferment a must through to dryness was not achieved if vineyard N application was limited to the post-harvest period.
