Role of dietary patterns for dioxin and PCB exposure

Abstract Dietary patterns were related to intake and blood concentrations of polychlorinated dibenzo- p -dioxins and dibenzofurans (PCDD/PCDFs), dioxin-like polychlorinated biphenyls (dl-PCBs) and selected non-dioxin-like-PCBs (ndl-PCBs). Intake calculations were based on an extensive food frequency questionnaire and a congener-specific database on concentrations in Norwegian foods. The study (2003) applied a two-step inclusion strategy recruiting representative (n=73) and high consumers (n=111) of seafood and game. Estimated median intakes of sum PCDD/PCDFs and dl-PCBs of the representative and high consumers were 0.78 and 1.25 pg toxic equivalents (TEQ)/kg bw/day, respectively. Estimated median intakes of ndl-PCBs (sum chlorinated biphenyl (CB)-28, 52, 101, 138, 153, 180) were 4.26 and 6.40 ng/kg bw/day. The median blood concentrations of PCDD/PCDFs/dl-PCBs were 28.7 and 35.1 pg TEQ/g lipid, and ndl-PCBs (sum of CB-101, 138, 153 and 180) 252 and 299 ng/g lipid. The Spearman correlations between dietary intake and serum concentration were r=0.34 (p=0.017) for dl-compounds and r=0.37 (p<0.001) for ndl-PCBs. Oily fish was the major source of dl-compounds and ndl-PCBs in high and representative consumers. Four dietary patterns were identified by principal component analysis. Two were related to high intakes, one dominated by oily fish ((,-3)), the other by fish liver and seagull eggs ("northern coastal"). Only the latter was closely associated with high blood concentrations of dioxins and PCBs. [source]


Uptake and accumulation of sediment-associated 4-nonylphenol in a benthic invertebrate (Lumbriculus variegatus, freshwater oligochaete)

Abstract In the present work, the oligochaete Lumbriculus variegatus was exposed for 56 d to lake sediment spiked with 4-nonylphenol (4-NP), which is a breakdown product of alkylphenol polyethoxylates, an important class of nonionic surfactants. During the exposure period, the content of 4-NP was determined in the oligochaetes, sediment, overlying water, and pore water in order to monitor the distribution of the 4-NP in the compartments of the test system. Concentration of 4-NP in L. variegatus increased linearly over the course of the test, with an uptake rate coefficient of 1.9 × 10,2 (± 0.2 × 10,2; [g carbon/(g lipid-h)]). No steady state was reached at the end of the exposure period, suggesting that the elimination of 4-NP by the organism was negligible. Ingested sediments played an important role in the accumulation of 4-NP in L. variegatus, which may achieve very high 4-NP body concentrations. The 56-d biota sediment accumulation factor (BSAF) was 24 ± 7 g carbon/g lipid. L. variegatus also was exposed to 4-NP-contaminated field sediment, and field oligochaetes and sediments were collected for 4-NP pollution assessment in aquatic ecosystem. The 4-NP uptake with natural sediment was in accordance with that measured with spiked sediments, suggesting that the bioavailability of sediment-associated 4-NP for L. variegatus was not affected by 4-NP sediment concentration and abiotic sediment characteristics. The BSAFs measured in field oligochaetes, ranging from 39 to 55 g carbon/g lipid, was relatively higher than the bioaccumulation factor measured in laboratory tests. The results suggest that 4-NP concentration can reach high levels in benthic oligochaetes; this can be an important way of exposure for their pelagic predators. [source]


Polycyclic aromatic hydrocarbon bioaccumulation by meiobenthic copepods inhabiting a superfund site: Techniques for micromass body burden and total lipid analysis

Abstract Microtechniques for polycyclic aromatic hydrocarbon (PAH) body burden and total lipid analysis were developed and applied to determine the first lipid-normalized bioaccumulation factors for a hydrophobic organic toxicant in a meiobenthic organism (0.063-0.500 mm) living in field-contaminated sediments. The total lipid microtechnique combines the standard Bligh-Dyer extraction method with a colorimetric quantification method for analysis of samples containing 1 to 50 ,g lipid. The microtechnique for body burden analysis quantifies PAHs from tissue samples containing as little as 10 pg PAH. Fluoranthene, benz[a]anthracene, and benzo[a]pyrene biota-sediment accumulation factors (BSAFs) were determined for the meiobenthic copepod Microarthridion littorale living in an estuarine U.S. Environmental Protection Agency Superfund site. Gravid female, nongravid female, and male BSAFs were 0.82, 0.54, and 0.36, respectively, for fluoranthene; 0.50, 0.44, and 0.40, respectively, for benz[a]anthracene; and 0.09, 0.12, and 0.15, respectively, for benzo[a]pyrene. Comparison of nonlipid-normalized bioaccumulation factors (BAFs) to BSAFs indicates that M. littorale bioaccumulated PAHs on a gram lipid basis. The BSAFs declined consistently with increasing PAH log Kow for all copepod sex and reproductive stages. Sex- and stage-specific comparisons of BSAFs suggest that differences in lipid content and quality may lead to differences in BSAF values depending on PAH molecular weight and/or hydrophobicity. MICROBIAL ANALYSIS AND TEXTURAL PROPERTIES OF CANTONESE SAUSAGE

ABSTRACT Cantonese sausage is one of the traditional meat products in China. In the present study, eight Cantonese sausage samples from different manufacturers were collected. Their microbial and textural properties were characterized. The chemical compositions were determined as follows: moisture, 15.18 ± 3.40%; protein, 33.75 ± 7.26% (expressed in dry matter); fat, 42.12 ± 6.64% (expressed in dry matter); total carbohydrates, 9.11 ± 3.73%; sodium chloride, 6.13 ± 0.41%; and sodium nitrite, 35.55 ± 6.69 mg/kg. The pH was determined to be 6.22 ± 0.14, while the acid value was 2.02 ± 0.71 mg potassium hydroxide/g lipid, and the peroxide value was 0.03 ± 0.04 g potassium iodide/100 g lipid. The total viable counts were 5.55 ± 1.48 log10 cfu/g, lactic acid bacteria were 3.59 ± 0.68 log10 cfu/g, staphylococci/micrococci were 3.96 ± 0.54 log10 cfu/g and yeasts/molds were 3.55 ± 0.47 log10 cfu/g. Staphylococci/micrococci were the dominant microbial group in Cantonese sausage. This was different from other sausages. The textural profiles of Cantonese sausage were determined as follows: average values of hardness, springiness, cohesiveness, gumminess and chewiness were 3,066.56 g, 0.96 mm, 0.57, 1,736.76 g and 1,658.26 g × mm, respectively. Analysis of between-variable relationships indicated that the pH value of Cantonese sausage was significantly correlated to its microbial and textural properties. PRACTICAL APPLICATIONS Cantonese sausage has gained much popularity and acceptance in China and many other countries. Commercial products from different manufacturers were collected in this work. The microbiological and textural characteristics of Cantonese sausage as well as the chemical compositions were evaluated. It is helpful to understand this type of sausage in order to improve its quality. [source]


Importance of maternal transfer of the photoreactive polycyclic aromatic hydrocarbon fluoranthene from benthic adult bivalves to their pelagic larvae

Abstract Laboratory experiments were conducted to determine if maternal transfer of polycyclicaromatichydrocarbons (P AH) from benthic adult bivalves could result in phototoxicity to their pelagic larvae when exposed to ultraviolet light (UV). In these experiments, adult bivalves were exposed to water or sediments amended with the model photoreactive PAH, fluoranthene. Elevated adult fluoranthene tissue concentrations were correlated with embryo concentrations. Embryonic mortality was enhanced by exposure to ultraviolet light when adult tissue fluoranthene levels were greater than 5,000 ,g/g lipid. While the exposure concentrations used in this experimental system were high, ultraviolet light exposure levels were low. Also, some of the PAHs are more potent than the model compound used in this study. These results indicate that maternal transfer of photoactive PAHs from organisms living in contaminated sediments may represent a significant adverse effect to populations of benthic bivalves. Metabolites of polychlorinated biphenyls in human liver and adipose tissue

Abstract A method for analysis of hydroxylated metabolites of polychlorinated biphenyls (OH-PCBs) was developed and adopted to the previously described method for determination of PCBs and methylsulfonyl-PCBs (MeSO2 -PCBs) in human liver and adipose tissue. The concentrations of OH-PCBs (14 congeners), MeSO2 -PCBs (24 congeners), and PCBs (17 congeners) in five paired samples of human liver and adipose tissue are reported. The sum of OH-PCB congeners was higher in liver (7,175 ng/g lipids) than in adipose tissue (0.3,9 ng/g lipids). In both liver and adipose tissue, 2,2,,3,4,4,,5,-hexachloro-3,-biphenylol (3,-OH-CB138) and 2,2,,3,3,,4,5,-hexachloro-4,-biphenylol (4,-OH-CB130) were the predominant hydroxylated PCB metabolites. The sum of OH-PCB congeners were of the same order of magnitude as those of methylsulfonyl metabolites of PCB in the same samples, 12 to 358 ng/g lipids and 2 to 9 ng/g lipids in liver and adipose tissue, respectively. The levels of PCBs were similar in liver and adipose tissue, 459 to 2,085 ng/g lipids and 561 to 2,343 ng/g lipids, respectively. The sum of OH-PCBs and the sum of MeSO2 -PCBs correlated to the sum of PCBs. The determined PCB metabolites constituted 3 to 26% of total PCB concentration in the liver and 0.3 to 0.8% of total PCBs in the adipose tissue samples. [source]
