Potato spindle tuber viroid: the simplicity paradox resolved?

SUMMARY Taxonomy: , Potato spindle tuber viroid (PSTVd) is the type species of the genus Posipiviroid, family Pospiviroidae. An absence of hammerhead ribozymes and the presence of a ,central conserved region' distinguish PSTVd and related viroids from members of a second viroid family, the Avsunviroidae. Physical properties: , Viroids are small, unencapsidated, circular, single-stranded RNA molecules which replicate autonomously when inoculated into host plants. Because viroids are non-protein-coding RNAs, designation of the more abundant, highly infectious polarity strand as the positive strand is arbitrary. PSTVd assumes a rod-like, highly structured conformation that is resistant to nuclease degradation in vitro. Naturally occurring sequence variants of PSTVd range in size from 356 to 361 nt. Hosts and symptoms: , The natural host range of PSTVd,cultivated potato, certain other Solanum spp., and avocado,appears to be quite limited. Foliar symptoms in potato are often obscure, and the severity of tuber symptoms (elongation with the appearance of prominent bud scales/eyebrows and growth cracks) depends on both temperature and length of infection. PSTVd has a broad experimental host range, especially among solanaceous species, and strains are classified as mild, intermediate or severe based upon the symptoms observed in sensitive tomato cultivars. These symptoms include shortening of internodes, petioles and mid-ribs, severe epinasty and wrinkling of the leaves, and necrosis of mid-ribs, petioles and stems. [source]


Pathogenicity of Phaeoacremonium Species on Grapevines

Abstract Several Phaeoacremonium species have been recently described to include some species involved in disease of decline of woody plants and others associated with human infections. Thirteen species are currently reported on grapevines and they are suspected to be involved in Esca and Petri disease. The pathogenic character of new defined species is still unknown and, therefore, pathogenicity studies were conducted in this work. The pathogenicity of the following species was studied on grapevine seedlings and cuttings: Phaeoacremonium aleophilum, P. angustius, P. inflatipes, P. krajdenii, P. mortoniae, P. parasiticum, P. scolyti, P. venezuelense, P. viticola, and Phaeomoniella chlamydospora included as positive control. Two-month-old grapevine seedlings of Vitis vinifera cv. Malvar and cv. Airen were inoculated by watering 10 individual pots with a spore suspension (107 spores/ml) of each Phaeoacremonium species. All inoculated seedlings showed typical symptoms of a vascular disease 2 months after inoculation. Grapevine cuttings of Vitis vinifera cv. Monastrell were vacuum-inoculated with a spore suspension (108 spores/ml) and individually planted. Plants rated after 5 months showed that all Phaeoacremonium species and P. chlamydospora (used as positive control) caused a significant vascular discoloration, while only Phaeomoniella chlamydospora, Phaoeacremonium mortoniae and P. aleophilum caused a significant root weight reduction compared with a non-inoculated control. Phaoeacremonium parasiticum, P. angustius, P. inflatipes and P. venezuelense caused significant foliar symptoms that included interveinal chlorosis and stunted leaves. [source]


Effects of Brown Streak Virus Disease on Yield and Quality of Cassava in Tanzania

Abstract Brown streak virus disease is the most important biotic constraint to cassava production in the coastal areas of southern Tanzania. Symptoms include foliar chlorosis and sometimes stem lesions. The disease also affects the tuberous roots which develop a yellow/brown, dry, corky necrosis within the starch-bearing tissues, sometimes accompanied by pitting and distortion, that is visible externally. The foliar symptoms of the disease often do not greatly affect plant growth, although the most sensitive cultivars may be stunted and defoliated. The main impact of the disease on the crop is by causing root necrosis. Field experiments were conducted at two sites in Tanzania to determine the effect of the disease on yield and quality of the roots. Cassava brown streak disease (CBSD) decreased root weight and patches of root necrosis made roots unmarketable, although the unaffected parts might still have been suitable for home consumption. The disease therefore has two effects, one on total root yield and one on root quality, which affects marketability. The field trials showed that CBSD can decrease root weight in the most sensitive cultivars by up to 70%. The length of time between the appearance of foliar symptoms and the development of root necrosis is a varietal characteristic. In the most susceptible cultivars, root necrosis may appear within 6 months of planting cuttings derived from symptomatic mother plants. A local cultivar known as cv. Nachinyaya exhibited a form of tolerance to CBSD in which foliar symptoms appeared but the development of root necrosis was delayed allowing the full yield potential to be realized. [source]


Tomato leaf curl geminivirus in Australia: occurrence, detection, sequence diversity and host range

The occurrence of whitefly transmitted geminiviruses in Australia was studied using a mixed DNA probe capable of detecting a range of distinct geminiviruses. The only geminivirus species detected was Tomato leaf curl virus (TLCV), which is spread across a vast geographical region of far-northern coastal Australia, an area inhabited by the Australasian-Oceania biotype of Bemisia tabaci. The newly introduced silverleaf whitefly, B. tabaci biotype B, forms high population densities in the eastern coastal region of Queensland and is currently located approximately 150 km from the nearest known TLCV-infected area. The viral host range appeared to be narrow and of 58 species of crop plants and weeds inoculated using the B biotype, only 11 became infected with the virus, including five that did not show foliar symptoms. A DNA fragment of 694 nt, including the complete C4 open reading frame (ORF), the overlapping N-terminal part of the C1 ORF and the viral iterons involved in replication, was amplified from 11 TLCV field isolates and sequenced. Sequence analysis revealed an overall sequence variation of up to 14% in this region, as well as the presence of distinct viral iterons. [source]
