Experimental Assessment of Coral Reef Rehabilitation Following Blast Fishing

In addition to killing fish and other organisms, the blasts shatter coral skeletons, leaving fields of broken rubble that shift in the current, abrading or burying new coral recruits, and thereby slowing or preventing reef recovery. Successful restoration and rehabilitation efforts can contribute to coral reef conservation. We used field experiments to assess the effectiveness of different low-cost methods for coral reef rehabilitation in Komodo National Park (KNP), Indonesia. Our experiments were conducted at three different spatial scales. At a scale of 1 × 1 m plots, we tested three different rehabilitation methods: rock piles, cement slabs, and netting pinned to the rubble. Significantly more corals per square meter grew on rocks, followed by cement, netting, and untreated rubble, although many plots were scattered by strong water current or buried by rubble after 2.5 years. To test the benefits of the most successful treatment, rocks, at more realistic scales, we established 10 × 10 m plots of rock piles at each of our nine sites in 2000. Three years after installation, coverage by hard corals on the rocks continued to increase, although rehabilitation in high current areas remained the most difficult. In 2002 rehabilitation efforts in KNP were increased over 6000 m2 to test four rock pile designs at each of four rubble field sites. Assuming that there is an adequate larval supply, using rocks for simple, low-budget, large-scale rehabilitation appears to be a viable option for restoring the structural foundation of damaged reefs. Además de matar a peces y otros organismos, las explosiones destruyen esqueletos de corales, dejando campos de escombros rotos que se mueven con la corriente, erosionando o enterrando a reclutas de coral nuevos y por lo tanto disminuyen o previenen la recuperación del coral. Esfuerzos exitosos de restauración y rehabilitación pueden contribuir a la conservación de arrecifes de coral. Usamos experimentos de campo para evaluar la efectividad de diferentes métodos de bajo costo para la rehabilitación de arrecifes de coral en el Parque Nacional Komodo (PNK), Indonesia. Desarrollamos nuestros experimentos en tres escalas espaciales diferentes. A una escala de parcelas de 1 x 1 m, probamos tres métodos de rehabilitación: pilas de rocas, losas de cemento y redes sobre el escombro. Crecieron significativamente más corales por metro cuadrado sobre rocas, seguido por el cemento, redes y escombro sin tratamiento, aunque muchas parcelas fueron dispersadas por la fuerte corriente de agua o enterradas por escombros después de 2.5 años. Para probar los beneficios del tratamiento más exitoso, rocas, a escalas más realistas, en 2000 establecimos parcelas de 10 x10 m con pilas de rocas en cada unos de nuestros nueve sitios. Tres años después, la cobertura de corales duros sobre las rocas continuó incrementando, aunque la rehabilitación en áreas con corrientes fuertes fue la más difícil. En 2002, los esfuerzos de rehabilitación en PNK se incrementaron a 6000 m2 para probar cuatro diseños de pilas de rocas en cada uno de los cuatro sitios con escombros. Asumiendo que hay una adecuada existencia de larvas, la utilización de rocas para rehabilitación simple, de bajo costo y gran escala parece ser una opción viable para la restauración de la base estructural de arrecifes dañados. Abstract Functional organic materials with enhanced two-photon absorption lead to new technologies in the fields of chemistry, biology, and photonics. In this article we review experimental and theoretical methodologies allowing detailed investigation and analysis of two-photon absorption properties of organic chromophores. This includes femtosecond two-photon excited fluorescence experimental setups and quantum-chemical methodologies based on time-dependent density functional theory. We thoroughly analyze physical phenomena and trends leading to large two-photon absorption responses of a few series of model chromophores focusing on the effects of symmetric and asymmetric donor/acceptor substitution and branching. Impact of the fish Garra on the ecology of reservoirs and the occurrence of Microcystis blooms in semi-arid tropical highlands: an experimental assessment using enclosures

Summary 1.,Many man-made reservoirs in the semi-arid highlands of Northern Ethiopia (Tigray) are characterised by the occurrence of intensive blooms of cyanobacteria and a dominance of small riverine fishes belonging to the genus Garra. 2.,We carried out enclosure experiments to test for the effect of these small fish on abiotic characteristics, phytoplankton biomass and zooplankton community structure in the pelagic of two reservoirs (Gereb Awso and Tsinkanet). Two experiments were carried out in each of the reservoirs, one at the end of the rainy season (highest water level) and one at the end of the dry season (lowest water level). 3.,The presence of Garra in general increased the amount of suspended matter, nutrient concentrations (total nitrogen and total phosphorus), phytoplankton and Microcystis biomass (including the proportion of Microcystis in the phytoplankton community), and reduced water transparency. The positive effect of the presence of Garra on nutrient concentrations and phytoplankton productivity indicate that Garra has the potential to affect food web functioning indirectly through bottom-up effects, by enhancing nutrient concentrations through sediment resuspension and excretion of nutrients. Indeed, population densities of the cladoceran zooplankton taxa Ceriodaphnia and Diaphanosoma also showed an overall increase in enclosures with Garra. 4.,However, our data also provide some evidence for a potential of Garra to exert top-down control on large bodied daphnids (Daphnia carinata, D. barbata), although such effect varied among experiments. The limited capability of Garra to control zooplankton communities mainly reflects the low efficiency of these small, riverine and benthos-oriented fish in foraging on zooplankton and suggests the existence of an unoccupied niche for zooplanktivorous fish in the majority of the reservoirs. 5.,Although the main effects of Garra on the pelagic food web seemed to be mediated by bottom-up mechanisms, our results also indicate that one of the key variables, the relative abundance of Microcystis, was impacted by Daphnia -mediated trophic cascade effects. Abstract Numerical and experimental assessment of modern composite structures provided with knowledge-based joints for their behaviour control is treated in the present paper. Special connection strip, joining composite materials, is adopted. The wave approach of the back propagation neural network in micro- and macromechanical modelling is used for the numerical analysis of the problem. Enhancing diversity of species-poor grasslands: an experimental assessment of multiple constraints

Summary 1Many grasslands in north-west Europe are productive but species-poor communities resulting from intensive agriculture. Reducing the intensity of management under agri-environment schemes has often failed to increase botanical diversity. We investigated biotic and abiotic constraints on diversification by manipulating seed and microsite availability, soil fertility, resource competition, herbivory and deficiencies in the soil microbial community. 2The effectiveness of 13 restoration treatments was investigated over 4 years in a randomized block experiment established in two productive grasslands in central-east and south-west England. 3Severe disturbance involving turf removal followed by seed addition was the most effective and reliable means of increasing grassland diversity. Disturbance by multiple harrowing was moderately effective but was enhanced by molluscicide application to reduce seedling herbivory and by sowing the hemiparasite Rhinanthus to reduce competition from grasses. 4Low-level disturbance by grazing or slot-seeding was ineffective in increasing diversity. Inoculation with soil microbial communities from species-rich grasslands had no effect on botanical diversity. Nitrogen and potassium fertilizer addition accelerated off-take of phosphorus in cut herbage but did not cause a reduction in soil phosphorus or increase botanical diversity. 5Different grazing management regimes had little impact on diversity. This may reflect the constraining effect of the July hay cut on species dispersal and colonization. 6Synthesis and applications. Three alternative approaches to grassland diversification, with different outcomes, are recommended. (i) High intervention deturfing, which would create patches with low competitive conditions for rapid and reliable establishment of the target community. For reasons of cost and practicality this can only be done over small areas but will form source populations for subsequent spread. (ii) Moderate intervention (harrowing or slot-seeding) over large areas, which would establish a limited number of desirable, generalist species that perform well in restoration. This method is low cost and rapid but the increases in biodiversity are less predictable. (iii) Phased restoration, which would complement the above approaches. Productivity and competition are reduced over 3,5 years using Rhinanthus or fertilizers to accelerate phosphorus off-take. After this time harrowing and seeding should allow a wide range of more specialist species to establish. However, further research is required to determine the long-term effectiveness of these approaches. [source]


Computer-aided NMR assay for detecting natively folded structural domains,

Abstract Structural genomics projects require strategies for rapidly recognizing protein sequences appropriate for routine structure determination. For large proteins, this strategy includes the dissection of proteins into structural domains that form stable native structures. However, protein dissection essentially remains an empirical and often a tedious process. Here, we describe a simple strategy for rapidly identifying structural domains and assessing their structures. This approach combines the computational prediction of sequence regions corresponding to putative domains with an experimental assessment of their structures and stabilities by NMR and biochemical methods. We tested this approach with nine putative domains predicted from a set of 108 Thermus thermophilus HB8 sequences using PASS, a domain prediction program we previously reported. To facilitate the experimental assessment of the domain structures, we developed a generic 6-hour His-tag-based purification protocol, which enables the sample quality evaluation of a putative structural domain in a single day. As a result, we observed that half of the predicted structural domains were indeed natively folded, as judged by their HSQC spectra. Furthermore, two of the natively folded domains were novel, without related sequences classified in the Pfam and SMART databases, which is a significant result with regard to the ability of structural genomics projects to uniformly cover the protein fold space. An experimental assessment on the effects of photoperiod treatments on the somatic and gonadal growth of the juvenile European purple sea urchin Paracentrotus lividus

Abstract Determining the optimum light conditions for sea urchins reared in land-based systems is vital for the future use and assessment of possible commercial systems of sea urchin farming. The effects of two different light regimes, complete darkness and a long day photoperiod of 16 h light:8 h darkness, on the somatic and gonadal growth of the European sea urchin Paracentrotus lividus (19.5,23.0 mm) was investigated using the commercial UrchinPlatterÔ System over a 6-month period (5 March to 5 September). Hatchery-produced P. lividus were transported to the Aquaculture Fisheries Development Centre (AFDC, University College, Cork UCC). Before arrival at the AFDC, sea urchins were reared on a diet of Laminaria digitata. Females were the predominant species of the animal group, displaying a reproductive Stage III (growing stage) where gametogenesis was commencing. Results show that darkness supports higher somatic growth than the photoperiod treatment. Feeding rates were higher for sea urchins reared under darkness with gonadal growth increasing for both experimental treatments. Individuals reared under darkness had a higher per cent change in gonad index from the initial sample taken at the beginning of the experiment. [source]


Catalysts for water,gas shift processing of coal-derived syngases,

Abstract Although the gasification of coal is an efficient means of producing syngas, the carbon content of coal is such that gasification produces significantly higher ratios of carbon oxides to hydrogen than those obtained by the steam reforming of natural gas. The CO:H2 ratio can be adjusted, and more hydrogen produced, by the subsequent application of the water,gas shift (WGS) reaction. This article presents a review of technologies associated with the catalytic WGS reaction in a fixed-bed reactor that might be incorporated into a coal gasification-based system for H2 production with CO2 capture. The main output from this review is the identification of key project areas requiring further research. The performance of existing, commercially available catalysts,designed for use in natural gas reforming processes,with coal-derived syngases is an important aspect of developing technologies for coal-based H2 production. This article presents an experimental assessment of the performance of selected commercially available WGS catalysts, two high-temperature catalysts (HT01 and HT02) and a sour shift catalyst (SS01), with such syngases. For the three commercial catalysts investigated in this study, CO reaction order is found to be in a range of 0.75,1. The effect of changes in H2O concentration over HT01 is insignificant, whereas H2O reaction orders determined using HT02 and SS01 are found to be significantly positive even at high H2O:C ratios. The CO conversion rate is significantly reduced by increasing CO2 concentration, whereas increasing H2 concentration also causes a slight reduction in CO conversion rate for the three commercial catalysts investigated. Parental care in response to natural variation in nest predation pressure in six sunfish (Centrarchidae: Teleostei) species

Abstract,,, Parental care is an important, energetically costly component of the life history of many fishes. Despite this importance, little is known about how different species of fish vary parental care in response to natural nest predator burdens. In this study, underwater videography was used to quantify parental care activity of six species of syntopic nesting male centrarchid fishes in Lake Opinicon, Ontario, in response to natural predators. This approach was used to test the hypothesis that as offspring develop from eggs to wrigglers, parental care activity should decrease or remain static for fish guarding nests with low predator burden and increase for those with high predator burden, reflecting different external risks. Principal components analysis (PCA) was used to derive common aeration and nest defence variables. Aeration and predator defence activity of the fish varied extensively among species. Parental care behaviours indicative of defence and vigilance (e.g., turning, departures, time away from nest, displays) tended to be highest for species that had the most predation attempts, although this was not entirely consistent. There was also a positive relationship between the defence PCA metric and attempted predation. Defence did not vary with stage of offspring development, although interactions between defence and developmental stage were noted for several species. A trade-off between aeration and defence was not observed. In fact, species that provide high levels of aeration also simultaneously provide high levels of defence. Stage-specific patterns of defence in this study were less apparent than those documented by studies using responses to staged predator intrusions making it unclear as to the extent that fish were responding to the level of the risk to offspring than to the value of the brood. Therefore, combined use of observational and experimental assessments of parental care investment may be most appropriate for refining current theoretical paradigms. [source]


Responses of periphyton and insects to experimental manipulation of riparian buffer width along forest streams

Summary 1Riparian trees regulate aquatic ecosystem processes, such as inputs of light, organic matter and nutrients, that can be altered dramatically when these trees are harvested. Riparian buffers (uncut strips of vegetation) are widely used to mitigate the impact of clear-cut logging on aquatic ecosystems but there have been few experimental assessments of their effectiveness. 2Forests along 13 headwater stream reaches in south-western British Columbia, Canada, were clear-cut in 1998, creating three riparian buffer treatments (30-m buffer, 10-m buffer and clear-cut to the stream edge), or left as uncut controls, each treatment having three or four replicates. 3We predicted that periphyton biomass and insect consumers would increase as buffer width decreased, because of increased solar flux. We used two complementary studies to test this prediction. 4In one study, we compared benthic communities before and after logging in all 13 streams; a second study focused on periphyton and insect colonization dynamics over 6-week periods in each of four seasons in four streams, one in each treatment. 5Photosynthetically active radiation, and mean and maximum water temperature, increased as buffer width narrowed. 6Periphyton biomass, periphyton inorganic mass and Chironomidae abundance also increased as buffer width narrowed, with the largest differences occurring in the clear-cut and 10-m buffer treatments. 7Photosynthetically active radiation, water temperature, periphyton biomass and periphyton inorganic mass were significantly greater in the 30-m buffer treatment than in controls during some seasons. 8.,Synthesis and applications. We have shown that a gradient of riparian buffer widths created a gradient in light and temperature that led to non-linear increases in periphyton biomass and insect abundance. For example, Chironomidae abundance was generally greater in the 10-m and 30-m buffer treatments than in controls, whereas this was not always the case in the clear-cut treatment. This pattern may be due to the high sediment content of the periphyton mat in the clear-cut treatment, which potentially limited the response of some insects to increased food resources. Overall, our results indicate that uncut riparian buffers of 30-m or more on both sides of the stream were needed to limit biotic and abiotic changes associated with clear-cut logging in headwater, forested watersheds. Experimental superficial candidiasis on tissue models

Summary Candida species are common pathogens causing superficial mycoses primarily affecting the mucosa and the skin in humans. Crucial steps during pathogenesis of superficial candidiasis comprise fungal adhesion, colonisation and subsequent penetration of the respective tissues. Exploring these pathological events and perhaps fungal and tissue responses towards drug treatment is imperative in the management of this infection. Unfortunately, pathological biopsies of superficial candidiasis do not exhibit the early changes in the host,pathogen interaction as the tissues are already invaded by the fungi. In vivo experimental assessments of pathological processes of superficial candidiasis are also limited because of the difficulties in providing reproducible and comparable conditions in the host environment. Conversely, in vitro models have helped studying fungal,host interactions under more defined and controlled conditions. Some common in vitro models used to simulate superficial candidiasis are chick chorioallantoic membrane, mucosal explants and single layer or multiple layer cell cultures. Interestingly, these experimental approaches share advantages as well as disadvantages when compared with in vivo conditions. Hence, this review intends to discuss about the experimental superficial candidiasis produced in various tissue models and their advantages as well as disadvantages with a particular reference to further improvement of validity and reliability of such experiments.
