COL5A1 signal peptide mutations interfere with protein secretion and cause classic Ehlers-Danlos syndrome,

Abstract Classic Ehlers-Danlos syndrome (EDS) is a heritable connective tissue disease characterized by skin hyperextensibility, atrophic scarring, joint hypermobility and generalized tissue fragility. Mutations in COL5A1 and COL5A2, encoding the type V collagen pro,1- and pro,2-chain, are found in ,50% of patients with classic EDS. The majority of mutations lead to a non-functional COL5A1 allele, as a result of the introduction of a premature stopcodon in one COL5A1 transcript. A minority of mutations affect the structure of the type V collagen central helical domain. We show that mutations in the signal peptide (SP) domain of the preproá1(V)-collagen chain cause classic EDS. The missense mutations (p.L25R and p.L25P) are located in the crucial hydrophobic SP core, which is indispensible for preprotein translocation into the endoplasmic reticulum. As a result, mutant type V procollagen is retained within the cell, leading to a decreased amount of type V collagen in the extracellular matrix and disturbed collagen fibrillogenesis. Our findings further support the observation that decreased availability of type V (pro)collagen is a key factor and a shared mechanism in the pathogenesis of classic EDS. © 2008 Wiley-Liss, Inc. Differential onset of infantile deprivation produces distinctive long-term effects in adult ex-laboratory chimpanzees (Pan troglodytes)

Abstract Maternal or social deprivation during early infancy inevitably produces social deficiencies in juvenile chimpanzees. Hypothesizing such deficiencies to persist into adulthood (a), and, as in humans, a sensitive period in early infancy for attachment formation (b), we predicted and found behavioral differences in resocialized adult ex-laboratory chimpanzees after about 20 years of solitary confinement depending on their age at onset of deprivation: early deprived (ED; mean: 1.2 years) chimpanzees engaged significantly less in social interactions, spent less time associated, and showed more nonsocial idiosyncrasies than did late deprived (LD; mean: 3.6 years) chimpanzees. In addition to these individual attributes relational qualities, specifically the combination of ED and LD chimpanzees within social groups, have an impact on social recovery. LDs can best exploit their social potential in the company of other LDs and EDs tend to stagnate in their recovery when socialized with other EDs. © 2008 Wiley Periodicals, Inc. Abstract Although magnesium alloys were popular in the first half of the 20th century, the bad corrosion properties prevented their breakthrough in industrial mass production. Since the technology for the production of high purity alloys was introduced in the 1970s, magnesium alloys became more and more in the focus of industrial attention. Today magnesium alloys are state-of-the-art in construction parts in automotive industry. Despite its outstanding properties like good castability, low density and nearly unlimited availability the negative aspects like weak corrosion and wear behaviour still limit the application of magnesiums in industry, due to the need of sufficient surface protection (E. Aghion, B. Bronfin, Mater. Sci. Forum2000, 350,351, 19). Today, plasma electrolytic anodisations are state-of-the-art (H. Haferkamp, "Magnesiumkorrosion,Prozesse, Schutz von Anode und Kathode", in: Moderne Beschichtungsverfahren, F.-W. Bach, T. Duda, Eds., Wiley-VCH, Weinheim 2000, ISBN 3-527-30117-8, 242; M. Thoma, Metalloberfläche1984, 38, 393; T. W. Jelinek, Galvanotechnik2003, 94, 46; A. Kuhn, Galvanotechnik2003, 94, 1114). They provide acceptable corrosion resistance and protect the magnesium from mechanical damage due to their high hardness. On the other hand, their high porosity limits their use in combination with electrochemically noble materials, leading to galvanic corrosion (J. In addition, the high surface roughness of the plasma electrolytic anodisations restricts their use in tribological applications, particularly under sliding conditions (H. In order to achieve smooth surfaces with high quality, the PVD technology moves into the centre of interest. Since the 1980s PVD coatings are well established and widely used for different industrial applications, mainly for steel and tool coatings. The authors were the first who carried out serious studies on the development of PVD coatings for magnesium alloys in 1999 (J. Protein intake, growth and lung function of infants with chronic lung disease

Background:, The increased survival rate of extremely preterm infants has not improved the incidence or outcome of infants diagnosed with chronic lung disease (CLD) (Riley, 2008). The relationship between optimal nutrition (particularly protein intake) and chronic lung disease has not been established. The aim of this study was to investigate the association between protein intake, growth and lung function in infants with CLD. Methods:, A CLD database, maintained for the past 10 years, was used to select participants that had reached 1 year of corrected age. Infants who were born during 2001,2006 with a birth weight of <1500 g, and who subsequently had a diagnosis of CLD, were included. Infants with evidence of intra-uterine growth restriction and abnormal cerebral pathology were excluded. Demographic, mean weight gain, protein intake and respiratory support data were collected retrospectively from the medical notes. Growth parameters and need for oxygen and inhalers up to 1 year of corrected age were collected from the CLD follow-up database. SPSS, version 15 (SPSS Inc., Chicago, IL, USA) were used for Pearson's or Spearmans correlation analysis and analysis of variance or the Wilcoxon test, as appropriate. Results:, Sixty infants were studied: 25 females and 35 males. The median (range) post-menstrual age at birth was 26 (22,31) weeks. The most common feed was breast milk; fortified breast milk was used for 37% of the total days studied. The mean (SD) protein intake was 2.28 (0.33) g kg,1 day,1 and the mean (SD) weight gain was 11.67 (1.77) g kg,1 day,1. There was a positive correlation between protein intake and weight gain (r = 0.32, P = 0.013), which was stronger in females (r = 0.51, P = 0.009). Protein intake was significantly associated with head circumference growth in females only (r = 0.47, P = 0.038). Protein intake was inversely related to the number of days spent mechanically ventilated (r = ,0.32, P = 0.015). There was no relationship between protein intake and growth at 1 year corrected age, time spent on continuous positive airway pressure, age weaned off oxygen, or the use of inhalers. There was an inverse correlation between total weeks of oxygen dependence and head circumference at 1 year (r = ,0.35, P = 0.022). Discussion:, The mean protein intake was <3 g kg,1 day,1, which is the minimum requirement for preterm infants (Tsang et al., 2005). This was associated with a sub-optimal weight gain in our participants of <15 g kg,1 day,1 (Steward & Pridham, 2002). The study demonstrates the known association between low protein intake and poor growth with ventilator dependence (Loui et al., 2008). Conclusions:, Low birth weight and low gestational age infants at risk of CLD should receive special attention to optimise their protein intake because sub-optimal protein intake potentially leads to poor growth when on a neonatal intensive care unit.
