    Characteristics of Crashes With Farm Equipment That Increase Potential for Injury

    ABSTRACT:,Context:Crash fatality and injury rates are higher on rural roadways than other roadway types. Although slow-moving farm vehicles and equipment are risk factors on rural roads, little is known about the characteristics of crashes with farm vehicles/equipment.Purpose:To describe crashes and injuries for the drivers of farm vehicles/equipment and non-farm vehicles involved in an injury crash. Passengers are not included in this analysis.Methods:Injury crashes were included that involved a farm vehicle/equipment and at least one non-farm vehicle reported in Iowa Department of Transportation crash data from 1995 to 2004. Odds ratios were calculated through logistic regression to identify increased odds for injury among drivers of non-farm vehicles and farm vehicles/equipment. We examined frequently occurring crash characteristics to identify crash scenarios leading to the highest odds for injury.Findings:Non-farm vehicle drivers were 5.23 times more likely to be injured than farm vehicle/equipment drivers (95% CI = 4.12,6.46). The absence of restraint use was a significant predictor of injury for both farm vehicle/equipment drivers (OR = 2.85; 95% CI = 1.14,7.13) and non-farm vehicle drivers (OR = 2.53; 95% CI = 1.54,4.15). Crash characteristics increasing the odds of injury for non-farm vehicle drivers included speeding, passing the farm vehicle/equipment, driving on a county road, having a frontal impact collision, and crashing in darkness. Ejection was the strongest predictor of injury for the farm vehicle/equipment driver.Conclusion:Non-farm vehicle drivers were much more likely to be injured than farm vehicle/equipment drivers, suggesting that farm vehicle/equipment crash prevention should be a priority for all rural road users. Prevention strategies that reduce motor vehicle speed, assist in safe passing, increase seat belt use, and increase conspicuousness of the farm vehicle/equipment are suggested. [source]


    Aortic Valve Closure: Relation to Tissue Velocities by Doppler and Speckle Tracking in Patients with Infarction and at High Heart Rates

    Aim: To resolve the event in tissue Doppler (TDI)- and speckle tracking-based velocity/time curves that most accurately represent aortic valve closure (AVC) in infarcted ventricles and at high heart rates. Methods: We studied the timing of AVC in 13 patients with myocardial infarction and in 8 patients at peak dobutamine stress echo. An acquisition setup for recording alternating B-mode and TDI image frames was used to achieve the same frame rate in both cases (mean 136.7 frames per second [FPS] for infarcted ventricles, mean 136.9 FPS for high heart rates). The reference method was visual assessment of AVC in the high frame rate narrow sector B-mode images of the aortic valve. Results: The initial negative velocities after ejection in the velocity/time curves occurred before AVC, 44.9 ± 21.0 msec before the reference in the high heart rate material, and 25.2 ± 15.2 msec before the reference in the infarction material. Using this time point as a marker for AVC may cause inaccuracies when estimating end-systolic strain. A more accurate but still a practical marker for AVC was the time point of zero crossing after the initial negative velocities after ejection, 5.4 ± 15.3 msec before the reference in high heart rates and 8.2 ± 12.9 msec after the reference in the infarction material. Conclusion: The suggested marker of AVC at high heart rate and in infarcted ventricles was the time point of zero crossing after the initial negative velocities after ejection in velocity/time curves.     Identification and separation of DNA-hybridized nanocolloids by Taylor cone harmonics

    Abstract A rapid (minutes) electrospray bead-based DNA hybridization detection technique is developed by spraying a mixture of hybridized and unhybridized silica nanocolloids. With proper far-field control by external electrodes, the trajectory of the ejected nanobeads from the electrospray is governed by specific harmonics of the Laplace equation, which select discrete polar angles along well-separated field maxima near the conducting Taylor cone. Due to Rayleigh fission and evaporation, beads of different size acquire different total charge after ejection and suffer different normal electrophoretic displacement such that they are ejected along well-separated field maxima and are deposited in distinct rings on an intersecting plane. As the hybridized DNA is of the same dimension as that of the nanocolloid, the nanocolloids are hence easily differentiated from the unhybridized ones. This technique is highly specific as the high shear stress in the microjet shears away any non-specifically bound DNA from the nanocolloid surface. [source]


    A Possible Role for Gap Junctions in Generation of Very Fast EEG Oscillations Preceding the Onset of, and Perhaps Initiating, Seizures

    Summary: ,Purpose: We propose an experimentally and clinically testable hypothesis, concerning the origin of very fast (>,70 Hz) EEG oscillations that sometimes precede the onset of focal seizures. These oscillations are important, as they may play a causal role in the initiation of seizures. Methods: Subdural EEG recordings were obtained from children with focal cortical dysplasias and intractable seizures. Intra- and extracellular recordings were performed in rat hippocampal slices, with induction of population activity, as follows: (a) bath-applied tetramethylamine (an intracellular alkalinizing agent, that opens gap junctions); (b) bath-applied carbachol, a cholinergic agonist; and (c) focal pressure ejection of hypertonic K+ solution. Detailed network simulations were performed, the better to understand the cellular mechanisms underlying oscillations. A major feature of the simulations was inclusion of axon,axon gap junctions between principal neurons, as supported by recent experimental data. Results: Very fast oscillations were found in children before seizure onset, but also superimposed on bursts during the seizure, and on interictal bursts. In slice experiments, very fast oscillations had previously been seen on interictal-like bursts; we now show such oscillations before, between, and after epileptiform bursts. Very fast oscillations were also seen superimposed on gamma (30,70 Hz) oscillations induced by carbachol or hypertonic K+, and in the latter case, very fast oscillations became continuous when chemical synapses were blocked. Simulations replicate these data, when axonal gap junctions are included. Conclusions: Electrical coupling between principal neurons, perhaps via axonal gap junctions, could underlie very fast population oscillations, in seizure-prone brain, but possibly also in normal brain. The anticonvulsant potential of gap-junction blockers such as carbenoxolone, now in clinical use for treatment of ulcer disease, should be considered. [source]


    Nursing Synchronization and Milk Ejection Failure as Maternal Strategies to Reduce Allosuckling in Pair-Housed Sows (Sus scrofa domestica)

    Non-offspring nursing (allosuckling) is costly for lactating females. It is reasonable to assume, therefore, that mechanisms have evolved to control the frequency of allosuckling. It is suggested that the synchronization of nursing between females could be such a mechanism. The aim of this study was to investigate whether nursing synchronization is related to the timing of nursing behaviour of paired sows and the behaviour of alien piglets in relation to allosuckling. Specifically, we predicted that: (i) nursing synchronization will increase if there is a real risk of allosuckling compared with a situation where there is none, (ii) nursing synchronization indeed reduces allosuckling and, therefore, those nursings with allosuckling attempts will be less closely synchronized than those without allosuckling attempts and (iii) in non-synchronized nursings, the sows will more often employ another strategy to reduce allosuckling, namely, they will make the nursings non-nutritive by not releasing milk. Ten pairs of sows and their litters were used in this study. For each pair, the two sows were housed individually until their litters were weaned; however, their litters had access to both sows from day 11 post-partum. The behaviour of the 10 pairs of sows with their litters was videotaped for 6 h on days 10, 11, 17 and 24 post-partum. The frequency of allosuckling attempts, non-nutritive nursings and allosuckling during milk ejection was analysed for every pair for every day of observation. The observed frequency of nursing synchronization was high. The median interval between the start of a nursing within one pair of sows was 32 s. The first prediction was supported as synchronization became significantly tighter when piglets were given the possibility to allosuckle on day 11 [Cochran,Mantel,Haenszel (CMH) statistics, p < 0.0001]. The second prediction was also supported; the nursings were less closely synchronized when there were allosuckling attempts (CMH statistics, p < 0.05). Of all nursings, 36% had an allosuckling attempt; however, only 17% resulted in real allosuckling. Evidence in favour of the third prediction was also found as non-nutritive nursings were more frequent in non-synchronized nursings (CMH statistics, p < 0.05). Finally, real allosuckling during milk ejection was unrelated to the degree of nursing synchronization, as in less synchronized nursings, the sows responded to allosuckling attempts by not releasing milk. Our results suggest that sows are not able to completely suppress allosuckling; however, they can reduce its frequency through nursing synchronization and non-nutritive nursings. [source]


    Acute effects of caffeine and tobacco on arterial function and wave travel

    Abstract Background, Caffeine and tobacco consumption are risk factors for heart failure, but their effects remain controversial. It has been hypothesized that they cause alterations in arterial stiffness and arterial wave travel which may increase ventricular loading. In this study the authors examined the influence of these widely used stimulants on wave intensity and arterial stiffness parameters using carotid wave intensity analysis. Materials and methods, A new Doppler-based ultrasound method was used to measure the acute effects of caffeine and tobacco on wave intensity in the right common carotid artery. The measurements enabled changes in arterial stiffness parameters to be recorded. Results, In 17 subjects compared with 10 controls, caffeine increased blood pressure, early systolic wave intensity and wave speed, but late-systolic wave intensity and mid-systolic reflections were unchanged. In 11 smokers studied before and after smoking one cigarette, blood pressure and arterial stiffness increased but wave intensity was unchanged. No changes were observed in the controls. Conclusions, Increased wave intensity during ejection after caffeine suggested sympathomimetic effects on the left ventricular function. Increased wave speed in the common carotid artery implied augmented central loading after caffeine, but the absence of measurable changes in local arterial stiffness in the carotid artery suggested more complex and regional effects. Cigarette smoking acutely increased local arterial stiffness in the common carotid artery. These changes can be detected using wave intensity analysis. [source]


    The nitric oxide/cyclic guanosine monophosphate pathway modulates the inspiratory-related activity of hypoglossal motoneurons in the adult rat

    Abstract Motoneurons integrate interneuronal activity into commands for skeletal muscle contraction and relaxation to perform motor actions. Hypoglossal motoneurons (HMNs) are involved in essential motor functions such as breathing, mastication, swallowing and phonation. We have investigated the role of the gaseous molecule nitric oxide (NO) in the regulation of the inspiratory-related activity of HMNs in order to further understand how neural activity is transformed into motor activity. In adult rats, we observed nitrergic fibers and bouton-like structures in close proximity to motoneurons, which normally lack the molecular machinery to synthesize NO. In addition, immunohistochemistry studies demonstrated that perfusion of animals with a NO donor resulted in an increase in the levels of cyclic guanosine monophosphate (cGMP) in motoneurons, which express the soluble guanylyl cyclase (sGC) in the hypoglossal nucleus. Modulators of the NO/cGMP pathway were micro-iontophoretically applied while performing single-unit extracellular recordings in the adult decerebrated rat. Application of a NO synthase inhibitor or a sGC inhibitor induced a statistically significant reduction in the inspiratory-related activity of HMNs. However, excitatory effects were observed by ejection of a NO donor or a cell-permeable analogue of cGMP. In slice preparations, application to the bath of a NO donor evoked membrane depolarization and a decrease in rheobase, which were prevented by co-addition to the bath of a sGC inhibitor. These effects were not prevented by reduction of the spontaneous synaptic activity. We conclude that NO from afferent fibers anterogradely modulates the inspiratory-related activity of HMNs by a cGMP-dependent mechanism in physiological conditions. [source]


    Substance P excites globus pallidus neurons in vivo

    Abstract Substance P is a member of the neurokinin family. Previous studies have reported the existence of substance P and its high-affinity receptor, neurokinin-1 receptor, in globus pallidus. Employing in vivo extracellular recording combined with behavioural tests, the effects of substance P in globus pallidus of rats were studied. Micropressure ejection of the selective neurokinin-1 receptor agonist [Sar9,Met(O2)11] substance P increased the spontaneous firing rate of pallidal neurons in a concentration-dependent manner, with increases of 27.3% at 0.01, 33.4% at 0.03, 45.5% at 0.1, 38.4% at 0.3 and 36.4% at 1.0 mm. The selective neurokinin-1 receptor antagonist SR140333B prevented the excitatory effects induced by [Sar9,Met(O2)11] substance P. In behaving rats, we observed the postural effects of neurokinin-1 receptor activation in the globus pallidus. Consistent with electrophysiological results, unilateral microinjection of [Sar9,Met(O2)11] substance P (0.1 mm) led to a SR140333B-sensitive contralateral deflection in the presence of systemic haloperidol administration. Combining electrophysiological and behavioural findings, we concluded that substance P produces excitatory effects on globus pallidus neurons via neurokinin-1 receptors. [source]


    Glutamatergic input governs periodicity and synchronization of bursting activity in oxytocin neurons in hypothalamic organotypic cultures

    Abstract During suckling, oxytocin (OT) neurons display a bursting electrical activity, consisting of a brief burst of action potentials which is synchronized throughout the OT neuron population and which periodically occurs just before each milk ejection in the lactating rat. To investigate the basis of such synchronization, we performed simultaneous intracellular recordings from pairs of OT neurons identified retrospectively by intracellular fluorescent labelling and immunocytochemistry in organotypic slice cultures derived from postnatal rat hypothalamus. A spontaneous bursting activity was recorded in 65% of OT neurons; the remaining showed only a slow, irregular activity. Application of OT triggered bursts in nonbursting neurons and accelerated bursting activity in spontaneously bursting cells. These cultures included rare vasopressinergic neurons showing no bursting activity and no reaction to OT. Bursts occurred simultaneously in all pairs of bursting OT neurons but, as in vivo, there were differences in burst onset, amplitude and duration. Coordination of firing was not due to electrotonic coupling because depolarizing one neuron in a pair had no effect on the membrane potential of its partner and halothane and proprionate did not desynchronize activity. On the other hand, bursting activity was superimposed on volleys of excitatory postsynaptic potentials (EPSPs) which occurred simultaneously in pairs of neurons. EPSPs, and consequently action potentials, were reversibly blocked by the non-NMDA glutamatergic receptor antagonist CNQX. Taken together, these data, obtained from organotypic cultures, strongly suggest that a local hypothalamic network governs synchronization of bursting firing in OT neurons through synchronous afferent volleys of EPSPs originating from intrahypothalamic glutamatergic inputs.     SHORT COMMUNICATION Inhibition of GABAergic neurotransmission in the ventral tegmental area by cannabinoids

    Abstract It was shown recently that ,9-tetrahydrocannabinol, like several other drugs eliciting euphoria, stimulates dopaminergic neurons projecting from the ventral tegmental area (VTA) to the nucleus accumbens. The aim of the present work was to clarify the mechanism of this stimulatory effect. Our hypothesis was that cannabinoids depress the GABAergic inhibition of dopaminergic neurons in the VTA. Electrophysiological properties of VTA neurons in rat coronal midbrain slices were studied with the patch-clamp technique. GABAA receptor-mediated inhibitory postsynaptic currents (IPSCs) were evoked by electrical stimulation in the vicinity of the recorded neurons. The amplitude of IPSCs was depressed by the synthetic mixed CB1/CB2 cannabinoid receptor agonist WIN55212-2 (10,6 and 10,5 m). The CB1 cannabinoid receptor antagonist SR141716A (10,6 m) prevented the inhibition produced by WIN55212-2 (10,5 m). Two observations showed that IPSCs were depressed with a presynaptic mechanism. WIN55212-2 (10,5 m) did not change the amplitude of miniature IPSCs recorded in the presence of tetrodotoxin. Currents evoked by pressure ejection of muscimol from a pipette were also not changed by WIN55212-2 (10,5 m). The results indicate that activation of CB1 cannabinoid receptors inhibits GABAergic neurotransmission in the VTA with a presynaptic mechanism. Depression of the GABAergic inhibitory input of dopaminergic neurons would increase their firing rate in vivo. Accordingly, dopamine release in the projection region of VTA neurons, the nucleus accumbens, would also increase. [source]


    A spatially advancing turbulent flow and heat transfer in a curved channel

    Abstract Direct numerical simulation was performed for a spatially advancing turbulent flow and heat transfer in a two-dimensional curved channel, where one wall was heated to a constant temperature and the other wall was cooled to a different constant temperature. In the simulation, fully developed flow and temperature from the straight-channel driver was passed through the inlet of the curved-channel domain. The frictional Reynolds number was assigned 150, and the Prandtl number was given 0.71. Since the flow field was examined in the previous paper, the thermal features are mainly targeted in this paper. The turbulent heat flux showed trends consistent with a growing process of large-scale vortices. In the curved part, the wall-normal component of the turbulent heat flux was twice as large as the counterpart in the straight part, suggesting active heat transport of large-scale vortices. In the inner side of the same section, temperature fluctuation was abnormally large compared with the modest fluctuation of the wall-normal velocity. This was caused by the combined effect of the large-scale motion of the vortices and the wide variation of the mean temperature; in such a temperature distribution, large-scale ejection of the hot fluid near the outer wall, which is transported into the near inner-wall region, should have a large impact on the thermal boundary layer near the inner wall. Wave number decomposition was conducted for various statistics, which showed that the contribution of the large-scale vortex to the total turbulent heat flux normal to the wall reached roughly 80% inside the channel 135° downstream from the curved-channel inlet. © 2009 Wiley Periodicals, Inc.     A human phospholamban promoter polymorphism in dilated cardiomyopathy alters transcriptional regulation by glucocorticoids,

    Abstract Depressed calcium handling by the sarcoplasmic reticulum (SR) Ca-ATPase and its regulator phospholamban (PLN) is a key characteristic of human and experimental heart failure. Accumulating evidence indicates that increases in the relative levels of PLN to Ca-ATPase in failing hearts and resulting inhibition of Ca sequestration during diastole, impairs contractility. Here, we identified a genetic variant in the PLN promoter region, which increases its expression and may serve as a genetic modifier in dilated cardiomyopathy (DCM). The variant AF177763.1:g.203A>C (at position ,36,bp relative to the PLN transcriptional start site) was found only in the heterozygous form in 1 out of 296 normal subjects and in 22 out of 381 cardiomyopathy patients (heart failure at age of 18,44 years, ejection fraction=22±9%). In vitro analysis, using luciferase as a reporter gene in rat neonatal cardiomyocytes, indicated that the PLN-variant increased activity by 24% compared to the wild type. Furthermore, the g.203A>C substitution altered the specific sequence of the steroid receptor for the glucocorticoid nuclear receptor (GR)/transcription factor in the PLN promoter, resulting in enhanced binding to the mutated DNA site. These findings suggest that the g.203A>C genetic variant in the human PLN promoter may contribute to depressed contractility and accelerate functional deterioration in heart failure.     Emission of trace toxic metals during pulverized fuel combustion of Czech coals

    Abstract A study of the trace elements emission (As, Se, Cd, Co, Cr, Cu, Zn, Hg, Tl, Pb, Ni, Sn, Sb, V, Mn and Fe) from pulverized coal combustion has been made at six heating and power stations situated in the Czech Republic. The amount of chlorine in coal has considerable influence on volatilization of some elements such as Zn, Cu, Pb, Hg and Tl, which is explained by the formation of thermodynamically stable compounds of these elements with chlorine. Generally, the affinities for Cl follows the order Tl > Cu > Zn > Pb > Co > Mn > Sn > Hg. The experimental data indicates enrichment of some of the trace toxic elements in the emissions (Cu, Zn, As, Se, Cd, Sn, Sb, Hg and Pb) and good agreement was obtained by thermodynamic equilibrium calculations with a few exceptions. In the case of Fe, Mn, Co, Cr and Sn calculated values are overestimated in the bottom ash and there are zero predicted amounts of these elements in the fly ash. In comparison, the results from experiments show up to 80% of these elements retained in fly ash. This implies that there exist additional steps leading to the enrichment by Fe, Mn, Co, Cr and Sn of small particles. Such mechanisms could include the ejection during devolatilization of small inorganic particles from the coal of bottom ash particles, or disintegration of the char containing these metals to small particles of fly ash. On the other hand, there are slightly overestimated or similar values of relative enrichment factors for As, V, Cu, Cd, Sb, Tl and Pb in the fly ashes and zero predicted values for bottom ashes. Our experimental results show about 5% or less of these elements are retained in bottom ashes, so they probably remain in the bottom ash inside unburned parts of coal.     Ejaculatory dysfunction caused by the new ,1 -blocker silodosin: A preliminary study to analyze human ejaculation using color Doppler ultrasonography

    Objectives: In order to clinically investigate the mechanism of ejaculatory dysfunction attributable to the ,1 -blocker silodosin, a real-time observation of ejaculation by healthy males was performed. Methods: Following intake of silodosin, a newly developed selective ,1 -blocker for benign prostatic hypertrophy, ejaculation was dynamically observed using color Doppler ultrasound in three healthy males. Normal ejaculation was also investigated in the same manner. Results: With silodosin intake, no antegrade ejaculation was observed in cases 1 or 2. In case 1, seminal fluid slowly but continuously flowed out from the seminal vesicles into the bladder. In case 2, only a small amount of seminal fluid flowed into the bladder during the ejaculatory sensation. In case 3, ejection of a small amount of semen from the external urethral orifice was observed and inflow of a small amount of seminal fluid into the bladder was also captured. Without silodosin intake, all three subjects exhibited antegrade ejaculation. Conclusions: The mechanism of ejaculatory dysfunction is intricately related to retrograde ejaculation (retrograde inflow of seminal fluid), insufficient contraction of the seminal vesicles, and insufficient rhythmic contraction of the muscles of the pelvic floor.     Behavioral responses of nesting female dark-eyed juncos Junco hyemalis to hetero- and conspecific passerine preen oils

    Several studies have suggested a greater role for olfactory cues in avian social interactions than previously recognized, but few have explicitly investigated the effect of odor on parental behavior. We present results from a preliminary study in which we applied hetero- and conspecific preen gland secretions, which are known to contain volatile compounds, to the nests and eggs of incubating female dark-eyed juncos Junco hyemalis. The responses to these two conditions were compared to the responses of females whose nests were treated with their own preen oil as a control condition, and to females whose nests were treated with the vehicle only. We found that females significantly reduced incubation bout length, a form of parental care, in response to alien secretions, more so if they came from a heterospecific than a conspecific. Females did not reduce incubation bout length in response to their own preen oil or to a vehicle-only control. These results suggest that odors in the nest may influence avian parental care. However, the behavioral change was only temporary and had no effect on later hatching success. In our study population, brood parasitism by brown-headed cowbirds is common, but resulting nest abandonment is rare; juncos are frequently able to successfully breed even with cowbird nestlings in their nests. Thus, we suggest that more extreme behavioral responses to alien odor, such as nest abandonment or egg ejection, may not be adaptive and should not be expected. [source]


    Maternal clutch reduction in the pied flycatcher Ficedula hypoleuca: an undescribed clutch size adjustment mechanism

    During eight years of study of a population of pied flycatchers Ficedula hypoleuca breeding in central Spain, we observed throughout incubation 22 cases of intact eggs being placed on the nest rim and clearly outside of the nest bowl and remaining there, with consequent embryo mortality. We assume that the removal of eggs had been performed by the incubating female. There is no evidence that pied flycatcher parents can detect embryo mortality and eject unhatchable eggs. Maternal egg ejection constitutes a direct mechanism of clutch size adjustment during incubation and before hatching of any young of the brood. This adjustment of clutch size happened when the mean and minimum temperature of the day before ejection were especially low. Also, clutch-reducing females were significantly more prone to show moult-breeding overlap than other females, suggesting a lower disposition to invest in reproduction. Clutch-reducing females were also frequently older than four years and had indications of a lower immunocompetence than females not ejecting eggs. Our results support the hypothesis that adverse conditions can elicit parental family size adjustment at every stage of reproduction without the need to depend on mechanisms of sibling competition.     Responses of great reed warblers Acrocephalus arundinaceus to experimental brood parasitism: the effects of a cuckoo Cuculus canorus dummy and egg mimicry

    Egg rejection behaviour towards parasitic eggs was studied in a great reed warbler Acrocephalus arundinaceus population in central Hungary, which was heavily (about 65%) parasitised by the common cuckoo Cuculus canorus. Clutches were experimentally parasitised during the egg-laying period with artificial, moderately mimetic cuckoo eggs or with conspecific eggs that were good mimics of the hosts' eggs. Great reed warblers rejected 76.2% of the artificial cuckoo eggs, mainly by ejection, but accepted most of the conspecific eggs (87.5%). Cuckoo eggs in naturally parasitised clutches were rejected at a lower rate (32%). When, in addition to the egg mimicry experiments, a stuffed cuckoo was placed near the nest, accompanied by the recording of a female cuckoo call, hosts' rejection rate of the artificial cuckoo egg increased from 76% to 96%. The sight of the cuckoo, on the other hand, did not influence host's rejection behaviour when a conspecific egg was used in the experiment. A stuffed collared dove Streptopelia decaocto, accompanied by its call, was used as a control, and did not cause any increased rejection. Great reed warblers were more aggressive towards the cuckoo than to the dove dummy. When the cuckoo eggs in naturally parasitised clutches were exchanged with artificial cuckoo eggs, we observed no increase in the rejection rate. We conclude that great reed warblers in our heavily parasitised population are capable of detecting brood parasitism in their clutch by identifying the parasitic egg. The efficiency of this identification depends mainly on the mimicry of the foreign egg. The sight of the cuckoo at the nest may increase rejection rate by stimulus summation, and this conditional effect is mainly affected by the degree of mimicry of the parasitic egg. [source]


    Noninvasive Assessment of Cardiac Resynchronization Therapy for Congestive Heart Failure Using Myocardial Strain and Left Ventricular Peak Power as Parameters of Myocardial Synchrony and Function

    Resynchronization Therapy for Heart Failure.Introduction: Although invasive studies have shown that cardiac resynchronization therapy by biventricular pacing improves left ventricular (LV) function in selected heart failure patients, it is impractical to apply such techniques in the clinical setting. The aim of this study was to assess the acute effects of cardiac resynchronization therapy by noninvasive techniques. Methods and Results: Twenty-two patients enrolled in the InSync trial (age 64 ± 9 years, 18 men and 4 women; all with ejection fraction <35% and QRS >130 msec) were studied 1 to 12 months after pacemaker implantation during pacing, and while ventricular pacing was inhibited. Regional myocardial strains of the interventricular septum, LV free wall, and right ventricular free wall were derived from color Doppler tissue echocardiography. Peak power index was calculated as a product of simultaneously recorded noninvasive blood pressure and pulse-wave (PW) Doppler velocity of the LV outflow tract. The Z ratio (sum of LV ejection and filling times divided by RR interval) and tei index were calculated from PW Doppler data. During pacing, overall regional strain improved (P = 0.01), while the LV strain coefficient of variation decreased from 2.7 ± 2.4 to 1.3 ± 0.7 (P = 0.009). Additionally, peak power index improved from 84 ± 24 to 94 ± 27 cm· mmHg/sec (P = 0.004). The Z ratio increased from 0.71 ± 0.08 to 0.78 ± 0.07 (P = 0.0005), while the tei index decreased from 0.86 ± 0.33 to 0.59 ± 0.16 (P = 0.0002). Conclusion: Using novel noninvasive indices, we demonstrated that cardiac resynchronization therapy improves LV performance. [source]


    Effects of canopy heterogeneity, seed abscission and inertia on wind-driven dispersal kernels of tree seeds

    Summary 1Understanding seed dispersal by wind and, in particular, long-distance dispersal (LDD) is needed for management of plant populations and communities, especially in response to changes in climate, land use and natural habitats. Numerical models designed to explore complex, nonlinear atmospheric processes are essential tools for understanding the fundamental mechanisms involved in seed dispersal. Yet, thus far, nearly all such models have not explicitly accounted for the spatial heterogeneity that is a typical feature of all ecosystems. 2The recently developed Regional Atmospheric Modelling System (RAMS)-based Forest Large Eddy Simulation (RAFLES) is used here to explore how within-stand canopy heterogeneity impacts LDD. RAFLES resolves microscale canopy heterogeneity such as small gaps and variable tree heights, and it simulates their impacts on turbulence inside and above the canopy in the atmospheric boundary layer (ABL). For that purpose, an Eulerian,Lagrangian module of seed dispersal is added to RAFLES to simulate seed trajectories. 3Particular attention is paid to the sensitivity of statistical attributes of the dispersal kernels (i.e. mean, mode, variance, tail) to key simplifications common to all seed dispersal models, such as horizontal homogeneity in the canopy and flow field, and the tight coupling between air parcel trajectories and seed trajectories (i.e. neglecting seed inertia). These attributes appear to be sensitive to various factors operating at scales ranging from the seed scale to the ABL scale. 4Simulations with RAFLES show that LDD is characterized by a dispersal kernel with a ,tail', asymptotically approaching a power law decay of ,3/2 (mainly occurring for lighter seeds at high wind speeds). This is consistent with asymptotic predictions from analytical models. The wind speed threshold at which seed abscission occurs, set-up to be twice the standard deviation of the vertical wind speed, is shown to affect short-distance dispersal, but has no significant impact on LDD. Ignoring the effects of seed inertia on the seed trajectory calculations has a minor effect on short-distance dispersal and no effect on the probability of seed uplift. Thus, it has no significant impact on LDD. 5Synthesis. Tree-scale canopy heterogeneity affects the turbulence characteristics inside and above the canopy and, consequently, this affects dispersal kernel statistics. A key finding from this study is that ejection is enhanced above the shorter trees of the canopy. Seeds dispersed above shorter trees have a higher probability of experiencing LDD while their short-distance dispersal remains practically the same. At inter-annual time scales, such interactions could affect species composition. [source]


    Dynamic assembly of chromatin complexes during cellular senescence: implications for the growth arrest of human melanocytic nevi

    Summary The retinoblastoma (RB)/p16INK4a pathway regulates senescence of human melanocytes in culture and oncogene-induced senescence of melanocytic nevi in vivo. This senescence response is likely due to chromatin modifications because RB complexes from senescent melanocytes contain increased levels of histone deacetylase (HDAC) activity and tethered HDAC1. Here we show that HDAC1 is prominently detected in p16INK4a -positive, senescent intradermal melanocytic nevi but not in proliferating, recurrent nevus cells that localize to the epidermal/dermal junction. To assess the role of HDAC1 in the senescence of melanocytes and nevi, we used tetracycline-based inducible expression systems in cultured melanocytic cells. We found that HDAC1 drives a sequential and cooperative activity of chromatin remodeling effectors, including transient recruitment of Brahma (Brm1) into RB/HDAC1 mega-complexes, formation of heterochromatin protein 1, (HP1,)/SUV39H1 foci, methylation of H3-K9, stable association of RB with chromatin and significant global heterochromatinization. These chromatin changes coincide with expression of typical markers of senescence, including the senescent-associated ,-galactosidase marker. Notably, formation of RB/HP1, foci and early tethering of RB to chromatin depends on intact Brm1 ATPase activity. As cells reached senescence, ejection of Brm1 from chromatin coincided with its dissociation from HP1,/RB and relocalization to protein complexes of lower molecular weight. These results provide new insights into the role of the RB pathway in regulating cellular senescence and implicate HDAC1 as a likely mediator of early chromatin remodeling events. [source]


    UV laser-induced desorption mechanism analyzed through two-layer alkali halide samples

    Abstract Time of flight-mass spectrometry (TOF-MS) is used to analyze positive and negative desorbed ions generated by UV laser ablation of several alkali (X) halide (Y) salts. Most of the observed desorbed cluster ions have the structure (XY)nX+ or (XY)nY,. Their desorption yields decrease as exp(,kn), where k , 2 for both series, suggesting that the neutral component (XY)n plays the dominant role in the desorption process. Mass spectrum measurements were performed for compound samples in which two salts (out of CsI, RbI, KBr, KCl and KI) are homogeneously mixed or disposed in two superposed layers. The detection of small new ion species and large cluster ions of the original salts supports the scenario that the uppermost layers are completely atomized while deep layers are emitted colder and fragmented: It is proposed that ns-pulsed laser induced desorption of ionic salts occurs via two sequential mechanisms: (1) ejection of cations and anions in the hot plume, followed by recombination into new cluster ions and (2) ejection of relatively cold preformed species originated from deep layers or from periphery of the irradiated region.     Evaluation of relative DNA binding affinities of anthrapyrazoles by electrospray ionization mass spectrometry

    Abstract Binding interactions of a new series of anthrapyrazoles (APs) with DNA were evaluated by electrospray ionization mass spectrometry (ESI-MS). Relative binding affinities were estimated from the ESI-MS data based on the fraction of bound DNA for DNA/anthrapyrazole mixtures, and they show a correlation to the shift in melting point of the DNA measured from a previous study. Minimal sequence specificity was observed for the series of anthrapyrazoles. Upon collisionally activated dissociation of the duplex/anthrapyrazole complexes, typically ejection of the ligand was the dominant pathway for most of the complexes. However, for complexes containing AP2 or mitoxantrone, strand separation with the ligand remaining on one of the single strands was observed, indicative of a different binding mode or stronger binding.     Influence on mass-selective ion ejection of the phase difference between the drive r.f. and the axial modulation potentials

    Abstract The phase difference between the drive r.f. and the axial modulation potential is known to influence significantly the mass shift, and all commercial ion trap mass spectrometers use a fixed value for this difference. However, although this important parameter is partly responsible for the good precision achievable today in most commercial ion traps, little discussion on the variation of the phase difference between the drive r.f. and the axial modulation potential has appeared in the literature. We present here an examination of the influence of a low-level axial modulation potential superimposed by capacitive coupling between the electrodes. Low-level axial modulation potentials are used for certain analytical scans such as reverse scan or slow scan speeds. Such low-level potentials help to prevent deterioration of mass resolution due to, for example, the dissociation of the ions during their resonant ejection from the ion trap. Reverse and forward scans are used to illustrate the mass shift and change in resolution, caused by a change in the phase difference between the drive r.f. potential applied to the ring electrode and the axial modulation potential applied on an end-cap electrode, in electrospray ionization mass spectra.     Behavioral and electrophysiological effects of 5-HT in globus pallidus of 6-hydroxydopamine lesioned rats

    Abstract Anatomical studies have shown that the globus pallidus receives abundant 5-hydroxytryptamine (5-HT) innervations from raphe nuclei. 5-HT may occupy an important position in the modulation of motor function through its affect on the activity of globus pallidus. In the present study, intrapallidal microinjection of 5-HT (0.1 mM) alone did not induce any motor behavior or postural asymmetry in the unilateral 6-hydroxydopamine (6-OHDA)-lesioned rats. However, when infused concomitantly with a low dose of 3, 4-dihydroxyphenylalanine (L-DOPA, 3 mg/kg i.p.), which itself can induce modest contralateral rotational behavior, 5-HT significantly potentiated the number of contralateral rotations. To elucidate the cellular mechanism, in vivo extracellular recordings were performed to examine the effects of 5-HT on globus pallidus neurons. In normal rats, the predominant effect of micropressure ejection of 5-HT on pallidal neurons was excitation. In 6-OHDA-lesioned rats, although 5-HT increased the firing rate in most pallidal neurons, 5-HT-induced inhibitory effects was stronger than that on the unlesioned side as well as normal rats. Furthermore, 5-HT1B receptors are mainly involved in 5-HT-induced excitation while 5-HT1A receptors are involved in 5-HT-induced inhibition. The results suggest that 5-HT may potentiate the antiparkinsonian effect of L-DOPA through modulating the activity of globus pallidus. © 2009 Wiley-Liss, Inc. [source]

    In the calculations, we also have a very special angular distribution for the slow electrons observed in the case of Ti:Sapphire laser pulse. The appearance of slow electrons is clearly demonstrated at the angularlyresolved 2D momentum distribution which low-energy part is presented in this figure. The observed 2D low-momentum distribution seems to be very consistent with the total momentum distribution: the angular directions of preferable electron ejection can be evidently seen. At other directions, the electron ejection is suppressed. The appearance of such directions of predominant or suppressed electron ejection results from the interference of the different waves in electron wave packet during its recollision with the parent ion.     A friction energy approach to quantifying lubrication under fretting sliding

    Abstract The problem of a proper lubrication under low-speed small oscillatory movement can be a decisive factor for the reliability of various components. There is a need to characterise the lubricious behaviour of the interface under oil-bath fretting wear conditions for ball bearing applications. Fast and reliable methods to quantify this behaviour for broad range of mechanical conditions are proposed and validated. Pure sliding reciprocation induces mixed lubrication mode. It was found that transient film profiles depend on the non-Newtonian response of the oils and the type of motion. Running-in period has a crucial importance for the tribofilm formation, and is a result of the interplay of the oil-sliding surfaces interface and is directly connected with the total energy dissipated from the contact region. The stability of structured tribofilm in steady-state period relies on the balance between the competitive processes: replenishment of the oil to the contact and ejection of the oil pending the oscillatory movement. The phenomenon of starvation was observed when the system was moved away from dynamical equilibrium and the growth of the dissipated energy was spotted.     39Ar- 40Ar dating of the Zagami Martian shergottite and implications for magma origin of excess 40Ar

    1992, 1999). Like several shergottites, Zagami contains excess 40Ar relative to its formation age. To understand the origin of this excess 40Ar, we made 39Ar- 40Ar analyses on plagioclase and pyroxene minerals from two phases representing different stages in the magma evolution. Surprisingly, all these separates show similar concentrations of excess 40Ar, ,1 × 10,6 cm3/g. We present arguments against this excess 40Ar having been introduced from the Martian atmosphere as impact glass. We also present evidence against excess 40Ar being a partially degassed residue from a basalt that actually formed ,4 Gyr ago. We utilize our experimental data on Ar diffusion in Zagami and evidence that it was shock-heated to only ,70 °C, and we assume this heating occurred during an ejection from Mars ,3 Myr ago. With these constraints, thermal considerations necessitates either that its ejected mass was impossibly large, or that its shock-heating temperature was an order of magnitude higher than that measured. We suggest that this excess 40Ar was inherited from the Zagami magma, and that it was introduced into the magma either by degassing of a larger volume of material or by early assimilation of old, K-rich crustal material. Similar concentrations of excess 40Ar in the analyzed separates imply that this magma maintained a relatively constant 40Ar concentration throughout its crystallization. This likely occurred through volatile degassing as the magma rose toward the surface and lithostatic pressure was released. These concepts have implications for excess 40Ar in other shergottites.     The effects of nebula surface density profile and giant-planet eccentricities on planetary accretion in the inner solar system

    Two surface density profiles are examined: a decaying profile with , , 1/a, where a is orbital semi-major axis, and a peaked profile in which , increases for a < 2 AU and decreases for a > 2 AU. The peaked profiles are generated by models of coagulation in an initially hot nebula. Models with initial ej,s = 0.05 (the current value) and 0.1 are considered. Simulations using the decaying profile with ej,s = 0.1 produce systems most like the observed planets in terms of mass-weighted mean a and the absence of a planet in the asteroid belt. Simulations with doubled , produce planets roughly twice as massive as the nominal case. Most initial embryos are removed in each simulation via ejection from the solar system or collision with the Sun. The asteroid belt is almost entirely cleared on a timescale of 10,100 Ma that depends sensitively on ej,s. Most initial mass with a < 2 AU survives, with the degree of mass loss increasing with a. Mass loss from the terrestrial region occurs on a timescale that is long compared to the mass loss time for the asteroid belt. Substantial radial mixing of material occurs in all simulations, but is greater in simulations with initital ej,s = 0.05. The degree of mixing is equivalent to a feeding zone of half width 1.5 and 0.9 AU for an Earth mass planet at 1 AU for the cases ej,s = 0.05 and 0.1, respectively. In simulations with ej,s = 0.05, roughly one-third and 5,10% of the mass contained in final terrestrial planets originated in the region a > 2.5 AU for the decaying and peaked profiles, respectively. In the case ej,s = 0.1, the median mass accreted from a > 2.5 AU is zero for both profiles. [source]


    Petrology and chemistry of the new shergottite Dar al Gani 476

    The meteorite is classified as a basaltic shergottite and is only the 13th martian meteorite known to date. It has a porphyritic texture consisting of a fine-grained groundmass and larger olivines. The groundmass consists of pyroxene and feldspathic glass. Minor phases are oxides and sulfides as well as phosphates. The presence of olivine, orthopyroxene, and chromite is a feature that DaG 476 has in common with lithology A of Elephant Moraine (EET) A79001. However, in DaG 476, these phases appear to be early phenocrysts rather than xenocrysts. Shock features, such as twinning, mosaicism, and impact-melt pockets, are ubiquitous. Terrestrial weathering was severe and led to formation of carbonate veins following grain boundaries and cracks. With a molar MgO/(MgO + FeO) of 0.68, DaG 476 is the most magnesian member among the basaltic shergottites. Compositions of augite and pigeonite and some of the bulk element concentrations are intermediate between those of lherzolitic and basaltic shergottites. However, major elements, such as Fe and Ti, as well as LREE concentrations are considerably lower than in other shergottites. Noble gas concentrations are low and dominated by the mantle component previously found in Chassigny. A component, similar to that representing martian atmosphere, is virtually absent. The ejection age of 1.35 ± 0.10 Ma is older than that of EETA79001 and could possibly mark a distinct ejection. Dar al Gani 476 is classified as a basaltic shergottite based on its mineralogy. It has a fine-grained groundmass consisting of clinopyroxene, pigeonite and augite, feldspathic glass and chromite, Ti-chromite, ilmenite, sulfides, and whitlockite. Isolated olivine and single chromite grains occur in the groundmass. Orthopyroxene forms cores of some pigeonite grains. Shock-features, such as shock-twinning, mosaicism, cracks, and impact-melt pockets, are abundant. Severe weathering in the Sahara led to significant formation of carbonate veins crosscutting the entire meteorite. Dar al Gani 476 is distinct from other known shergottites. Chemically, it is the most magnesian member among known basaltic shergottites and intermediate in composition for most trace and major elements between Iherzolitic and basaltic shergottites. Unique are the very low bulk REE element abundances. The CI-normalized abundances of LREEs are even lower than those of Iherzolitic shergottites. The overall abundance pattern, however, is similar to that of QUE 94201. Textural evidence indicates that orthopyroxene, as well as olivine and chromite, crystallized as phenocrysts from a magma similar in composition to that of bulk DaG 476. Whether such a magma composition can be a shergottite parent melt or was formed by impact melting needs to be explored further. At this time, it cannot entirely be ruled out that these phases represent relics of disaggregated xenoliths that were incorporated and partially assimilated by a basaltic melt, although the texture does not support this possibility. Trapped noble gas concentrations are low and dominated by a Chassigny-like mantle component. Virtually no martian atmosphere was trapped in DaG 476 whole-rock splits. The exposure age of 1.26 ± 0.09 Ma is younger than that of most shergottites and closer to that of EETA79001. The ejection age of 1.35 ± 0.1 Ma could mark another distinct impact event. [source]


    FliL is essential for swarming: motor rotation in absence of FliL fractures the flagellar rod in swarmer cells of Salmonella enterica

    Summary fliL is the first gene in a flagellar operon that specifies members of the switch complex and type III export system in Salmonella enterica and Escherichia coli, but no function has been ascribed to this gene thus far. Here we report that a fliL mutant is slightly impaired for swimming but completely defective in swarming in both organisms, and have studied this phenotype further in S. enterica. We have found that on swarm agar, mutant cells release or ,eject' their flagellar filaments. The released filaments are attached to the hook and part of the rod structure; we have identified the distal rod protein FlgG but not the proximal rod protein FlgF in these filaments. Rod fracture was not observed if flagellar rotation was prevented by removal of proteins that supply proton flow through the motor. Based on these and other results, we propose that motors experience a higher torque on swarm agar owing to an increased proton motive force, and that FliL allows the rod to withstand the increased torsional stress. The flagella-release phenotype of the S. enterica fliL mutant has a bearing on FliL-dependent flagellar ejection during the swimmer- to stalk-cell transition in the developmental cycle of Caulobacter crescentus.
